A Gram-negative, non-motile bacterium КMM 3653T was isolated from a sediment sample from the Sea of Japan seashore, Russia. On the basis of the 16S rRNA gene sequence analysis the strain КMM 3653T was positioned within the family Rhodobacteraceae (class Alphaproteobacteria) forming a distinct lineage with the highest gene sequence similarities to the members of the genera Pacificibacter (95.2–94.7%) and Nioella (95.1–94.5%), respectively. According to the phylogenomic tree based on 400 conserved protein sequences, strain КMM 3653T was placed in the cluster comprising Vannielia litorea, Nioella nitratireducens, Litoreibacter albidus and Pseudoruegeria aquimaris as a separate lineage adjacent to V. litorea KCTC 32083T. The average nucleotide identity values between strain КMM 3653T and V. litorea KCTC 32083T, N. nitratireducens KCTC 32417T, L. albidus KMM 3851T, and P. aquimaris CECT 7680T were 71.1, 70.3, 69.6, and 71.0%, respectively. Strain КMM 3653T contained Q-10 as the predominant ubiquinone and C18:1ω7c as the major fatty acid followed by C16:0. The polar lipids were phosphatidylcholine, phosphatidylglycerol, diphosphatidylglycerol, an unidentified phospholipid, two unidentified aminolipids, and five unidentified lipids. The DNA G+C content of 61.8% was calculated from the genome sequence. Based on the phylogenetic evidence and distinctive phenotypic characteristics, we proposed strain KMM 3653T (= KCTC 82575T) to be classified as a novel genus and species Harenicola maris gen. nov., sp. nov.

An aerobic, Gram-negative, non-pigmented non-motile bacterium designed КMM 8518T was isolated from a seawater sampled from the Sea of Japan seashore. Strain КMM 8518T grew at 7–42 °C and in the presence of 1–7% NaCl. The phylogenetic analyses based on 16S rRNA gene and whole-genome sequences placed the novel strain КMM 8518T into the genus Thalassobius as a separate lineage. Strain КMM 8518T shared the highest 16S rRNA gene sequence similarity of 98% to Thalassobius gelatinovorus KCTC 22092T and similarity values of ≤ 97% to other recognized Thalassobius species. The average nucleotide identity and digital DNA–DNA hybridization values between strain КMM 8518T and T. gelatinovorus KCTC 22092T were 79.6% and 23.5%, respectively. The major respiratory quinone was ubiquinone-10. The major fatty acid was C18:1ω7c followed by 11-methyl C18:1ω7c. Polar lipids comprised phosphatidylcholine, phosphatidylglycerol, diphosphatidylglycerol, an unidentified aminolipid, an unidentified phospholipid, and three unidentified lipids. The DNA G+C content of 62.7% was calculated from genome sequence analysis. Based on the phylogenetic analyses and distinctive phenotypic characteristics, the marine bacterium КMM 8518T is concluded to represent a novel species of the genus Thalassobius for which the name Thalassobius aquimarinus sp. nov. is proposed. The type strain of the species is strain KMM 8518T (= KCTC 82576T).

An ability of marine macroalgae Ulva lactuca (L.) to adapt membrane lipid matrix to elevated seawater temperature depending on rate of warm acclimation was studied. Plants were collected in the Peter the Great Gulf of the Sea of Japan in winter at 4°С and acclimated to summer temperature (20°С) at different rates: 16 or 2°С/day. The fatty acid composition and the crystal – liquid crystal thermal transition of glycolipids (monogalactosyl diacylglycerol, digalactosyl diacylglycerol, sulfoquinovosyl diacylglycerol) and betaine lipid 1,2-diacylglycero-O-4'-(N,N,N-tri-methyl)-homoserine (DGTS) from U. lactuca were investigated by GLC, HPLC-MS and differential scanning calorimetry, respectively. The dependence of the results of temperature acclimation on its rate was shown. Thus, rapid warm acclimation induced the decrease of the ratios of n-3/n-6 polyunsaturated fatty acids and unsaturated/saturated fatty acids, as well as the unsaturation index of fatty acid composition of lipids, which corresponded to seasonal changes. However, this trend did not retain, or even reverse changes occurred at lower warming rate. Redistribution of the composition of molecular forms of glycolipids in acclimated algae led not only to maintain the peak maximum temperature of thermal transition (Tmax), but also to phase separation of glycolipids. This contributed to the fit of the thermogram profiles to those of summer samples. The calorimetric transition of DGTS was most effectively changed: even with rapid acclimation, the Tmax value of the summer sample was reached. In general, U. lactuca showed the ability to adapt lipid matrix of membranes at different rates of warming seawater.

Accumulation of heavy metals and their effect on the composition of fatty acids and molecular species of phosphatidylcholine (PC) and phosphatidylethanolamine (PE) has been studied in the molluskan organs and tissues of the Gray mussel from clean and polluted water areas of Peter the Great Bay of the Sea of Japan. The total load of heavy metals accumulated in the mussel was evaluated with the coefficient of total concentration (ΣCC), which turned out to be extremely high for all the organs studied in mollusks from a polluted site. Cu and Pb accounted for the main share in the total load. It was found that accumulation of heavy metals in the kidneys, gills, and muscles had no effect on the composition of fatty acids, molecular species, and forms of basic membrane lipids of PC and PE. It has been shown that high resistance of membrane lipids of the Gray mussel to chronic anthropogenic pollution with heavy metals allows the mussel to function under conditions of contamination.

The cold-water gorgonian coral Paragorgia arborea is considered as a foundation species of deep-sea ecosystems in the northern Atlantic and Pacific oceans. To advance lipidomic studies of deep-sea corals, molecular species compositions of diacylglycerol ethers (DAGE), which are specific storage lipids of corals, and structural glycerophospholipids (GPL) including ethanolamine, choline, inositol and serine GPL (PE, PC, PI, and PS, respectively) were analyzed in P. arborea by HPLC and tandem mass spectrometry. In DAGE molecules, alkyl groups (16:0, 14:0, and 18:1), polyunsaturated fatty acids (PUFA), and monounsaturated FA are mainly substituted the glycerol moiety at position sn-1, sn-2, and sn-3, respectively. The ether form (1-O-alkyl-2-acyl) predominates in PE and PC, while PI is comprised of the 1,2-diacyl form. Both ether and diacyl forms were observed in PS. At position sn-2, C20 PUFA are mainly attached to PC, but C24 PUFA, soft coral chemotaxonomic markers, concentrate in PS, PI, and PE. A comparison of non-polar parts of molecules has shown that DAGE, ether PE, and ether PC can originate from one set of 1-O-alkyl-2-acyl-sn-glycerols. Ether PE may be converted to ether PS by the base-exchange reaction. A diacylglycerol unit generated from phosphatidic acid can be a precursor for diacyl PS, PC, and PI. Thus, a lipidomic approach has confirmed the difference in biosynthetic origins between ether and diacyl lipids of deep-sea gorgonians.

For the first time search for tetrodotoxin (TTX) and its analogues in the extracts of nemerteans using HPLC-MS/MS was performed. TTX analogues were detected in two nemertean species in addition to TTX: 7 analogues were detected in the extract of Cephalothrix simula , 3 analogues - in the extract 11-norTTX of Kulikovia manchenkoi . Presence of 5-deoxyTTX, 11-deoxyTTX, 5,6,11-trideoxyTTX and −6(R)-ol in nemerteans was shown for the first time.

The composition of phospholipids (PLs), fatty acids (FAs), molecular species of major membrane lipids phosphatidylcholine (PC) and phosphatidylethanolamine (PE) as well as the cholesterol (CL) level in the gills and liver of the plain sculpin Myxocephalus jaok were analyzed at different habitat temperatures (18, 9, 0°C). Polar lipids and cholesterol were shown to be actively involved in adaptation of the plain sculpin to changes in environmental temperature. A decrease in temperature evoked multidirectional changes in the level of monoenoic (MUFA) and polyenoic (PUFA) FAs, ω-3 PUFA, etheric PLs, and in the unsaturation index (UI) of FAs in PC and PE of th e plain sculpin organs. Changes in the composition of PL molecular forms were unidirectional in all organs but showed some organ specificity. Thus, PC showed an increase in the total percentage of SFA/PUFA and MUFA/PUFA containing predominantly 20:5, 22:5 and 22:6 of PUFA and a decrease in the percentage of SFA/MUFA and PUFA/PUFA as well as in the level of alkylacyl forms of PC. PE showed an increase in the percentage of MUFA/PUFA and a decrease in that of SFA/PUFA and PUFA/PUFA as well as in the level of alkenylacyl forms of PE. Despite a close FA composition of PC and PE, the repertoire of their molecular forms differed in an organ- and temperature-dependent manner. Molecular mechanisms of thermal adaptation in the plain sculpin organs were traced more distinctly at the level of PC and PE molecular forms rather than in their FA spectrum.

Marine invertebrates are a paraphyletic group that comprises more than 90% of all marine animal species. Lipids form the structural basis of cell membranes, are utilized as an energy reserve by all marine invertebrates, and are, therefore, considered important indicators of their ecology and biochemistry. The nutritional value of commercial invertebrates directly depends on their lipid composition. The lipid classes and fatty acids of marine invertebrates have been studied in detail, but data on their lipidomes (the profiles of all lipid molecules) remain very limited. To date, lipidomes or their parts are known only for a few species of mollusks, coral polyps, ascidians, jellyfish, sea anemones, sponges, sea stars, sea urchins, sea cucumbers, crabs, copepods, shrimp, and squid. This paper reviews various features of the lipid molecular species of these animals. The results of the application of the lipidomic approach in ecology, embryology, physiology, lipid biosynthesis, and in studies on the nutritional value of marine invertebrates are also discussed. The possible applications of lipidomics in the study of marine invertebrates are considered.

Increasing global temperatures are expected to increase the risk of extinction of various species due to acceleration in the pace of shifting climate zones. Nevertheless, there is no information on the physicochemical properties of membrane lipids that enable the adaptation of the algae to different climatic zones. The present work aimed to compare fatty acid composition and thermal transitions of membrane lipids from green macroalgae Ulva lactuca harvested in the Sea of Japan and the Adriatic Sea in summer. U. lactuca inhabiting the Adriatic Sea had bleached parts of thalli which were completely devoid of chloroplast glycolipids. The adaptation to a warmer climatic zone was also accompanied by a significant decrease in the ratio between unsaturated and saturated fatty acids (UFA/SFA) of membrane lipids, especially in bleached thalli. Hence, bleaching of algae is probably associated with the significant decrease of the UFA/SFA ratio in glycolipids. The decreasing ratio of n-3/n-6 polyunsaturated fatty acids (PUFAs) was observed in extra-plastidial lipids and only in the major glycolipid, non-lamellar monogalactosyldiacylglycerol. The opposite thermotropic behavior of non-lamellar and lamellar glycolipids can contribute to maintenance of the highly dynamic structure of thylakoid membranes of algae in response to the increasing temperatures of climatic zones.

This study examines the fatty radical (FR) composition and heat-induced crystalline to liquidcrystalline phase transitions of phosphatidylcholine (PC) and phosphatidylethanolamine (PE) from the gills, hepatopancreas, gonads, and muscle of the tanner crab Chionoecetes bairdi, which was collected in the summer at a near-bottom water temperature of 2.8°C. The location of the PC and PE thermograms below 2.8°C indicates the functionally optimal liquid crystalline state of the membrane lipid matrix. The proximity of the thermogram profiles of PC and PE from the different organs and tissues of C. bairdi and significant overlapping of the temperature areas of transitions (symbatic behavior) correlate with a similar composition of major FR and their total parameters in PC and PE. The obtained data point to the effective adaptation of the bairdi crab to low water temperatures and to the need for adaptive changes in the FR composition or change of habitat with increasing temperature. The thermotropic behavior of muscle PC, in which the greater part of the thermogram is in the temperature range from 2.8 to 32°C, suggests a potential for the tanner crab to adapt to increased temperatures.

To study the adjuvant activity and to optimize tubular immunostimulating complexes (TI complexes) with respect to the soluble antigen, an investigation was performed on cucumariosides and their fractions, i.e., individual triterpene glycosides that were obtained from the holothurian Cucumaria japonica, as well as on phospholipids from the sea urchin Strongylocentrotus intermedius and the starfish Distolasterias nippon included in the composition of TI complexes in order to assess their effects on the immunogenicity of human serum albumin (HSA). Generally, TI complexes showed a moderate adjuvant activity with respect to HSA; this activity can be adjusted by substitution of a cucumarioside and/or a polar lipid in the composition of the TI complex. TI complexes based on monogalactosyldyacylglycerol (MGDG) from the sea alga Ulva lactuca and cucumariosides A2-2 and A4-4 maximally stimulated anti-HSA antibody production in mice. Substitution of MGDG for phosphatidylcholine (PC) from the starfish D. nippon with an increased (compared with other investigated phospholipids) ratio of n-3/n-6 polyunsaturated fatty acids did not result in a change in the immunostimulating activity of the TI complex. However, the use of other phospholipids led to a decrease in the adjuvant activity of the TI complex, compared with that of the TI complex based on PC from D. nippon and cucumarioside A2-2. The greatest fluctuations in the contents of cytokines IL-6, IL-12, IFN-γ and GM-CSF depending on the composition of the TI complexes indicate the possibility of regulation of the T-cell immune response. The values of hematological parameters were normal.

This study examines the crystal-liquid crystal phase transitions of the major phospholipids, phosphatidylcholine (PC) and phosphatidylethanolamine (PE), from muscle tissue of marine fish living at temperatures of 0–4.1°C (the Pacific cod Gadus macrocephalus, banded Irish lord Hemilepidotus gilberti, Pacific halibut Hippoglossus hippoglossus, black edged sculpin Gymnocanthus herzensteini, dark colored flounder Pleuronectes obscurus, and plain sculpin Myoxocephalus jaok), as well as of fish living at 14 and 18°C (Pacific redfin Tribolodon brandti). The PC and PE phase-transition thermograms of all the investigated species displayed specific profiles. The largest share of the thermogram area at temperatures higher than those of the habitat was found for the PC (28–40%) and PE (47–82%) of the black-edged sculpin, dark-colored flounder, and the plain sculpin, which have reduced physiological activity at low temperatures. In the Pacific cod, banded Irish lord, and the Pacific redfin, this parameter was much lower, 0–18% (PC) and 0–39% (PE). The thermotropic behavior PC and PE was symbate in all fish, except for the cod and the plain sculpin. The transition enthalpy of PC in all the investigated species was 2.8 times higher than that of PE. To interpret the varied PC and PE thermogram profiles of fish with similar fatty-acid compositions, the data on the composition of the molecular species of these phospholipids appeared to be the most informative. This study suggests that each fish species has its own strategy for thermal adaptation, which is realized through a certain set of phospholipid molecular species.

Individual variations in the phospholipid compositions of the organs of marine invertebrates were investigated in the representatives of three phyla: Arthropoda [decapods: Paralithodes camtschaticus (Tilesius, 1815) and Erimacrus isenbeckii (Brandt, 1848)]; Echinodermata [starfishes: Distolasterias nipon (Doderlein, 1902) and Evasterias echinosoma Fisher, 1926]; and Tunicata [ascidians: Halocynthia aurantium (Pallas, 1787), H. roretzi (Drasche, 1884), and Styela clava Herdman, 1881]. The specificity of individual phospholipid variation was shown to be related to the systematic status and functional properties of the organs of marine invertebrates. Phosphatidylcholine and phosphatidylethanolamine were the least variable. Low variation of phospholipid composition was found in the liver, legs, and gills of decapods; in the liver of starfishes; and in the gonads and skin-muscular and gill sacs of ascidians. The contents of ceramide aminoethylphosphonate (CAEP) and phosphatidylglycerol (PG) showed the highest variation. These phospholipids were found but not in all individuals or organs studied. After 7 days of starvation, CAEP completely disappeared from the organs of the animals; thus, we suggest its exogenous origin.

The phospholipid compositions of organs and tissues were determined in representatives of two phyla of marine invertebrates: Echinodermata [Asteroidea: Aphelasterias japonica (Bell, 1881), Evasterias echinosoma Fisher, 1926, Distolasterias nipon (Doderlein, 1902), Asterias amurensis Lutken, 1871; Echinoidea: Strongylocentrotus intermedius (A. Agassiz, 1863); Holothurioidea: Cucumaria frondosa japonica (Semper, 1868), Eupentacta fraudatrix (Djakonov et Baranova, 1958), Apostichopus japonicus (Selenka, 1867)], and Tunicata [Ascidia: Halocynthia aurantium (Pallas, 1787), H. roretzi (Drasche, 1884), and Styela clava (Herdman, 1881)]. The specificity of phospholipid distribution was shown to be related to the taxonomic position of marine invertebrates and the functional properties of their organs and tissues. Ceramide aminoethylphosphonate was found only in the digestive organs of all holothurians and the starfishes D. nipon and A. amurensis, suggesting its exogenous origin. Phosphatidylglycerol was found in all organs and tissues of the holothurians C. frondosa japonica and E. fraudatrix, as well as in the gastrointestinal tract of ascidians; its origin is unclear.

The development of cryopreservation methods for embryonic cells and larvae of sea animals offers a great potential for marine biotechnology. Larval cells of bivalves and sea urchins were frozen to -196 degrees C using traditional cryoprotectants (Me(2)SO and trehalose) and the cryoprotective mixture developed by us. In addition to Me(2)SO and trehalose, this mixture contained an exogenous lipid extract from mussel tissues and antioxidants. A positive effect of antioxidants (alpha-tocopherol acetate, ascorbic acid or echinochrome, the quinoid pigment of sea urchins) on cell viability became significant only in the presence of exogenous lipids. Antioxidants added to cryoprotective mixtures did not reveal visible cryoprotective activity when used separately. To better understand the mechanism of the protective effect of exogenous lipids on cell membranes of sea animals, a comparative analysis of the fatty acid (FA) composition of total lipids in larval cells before and after freezing was carried out using a gas-liquid chromatography. The results indicate that freezing-thawing has direct effects on the FA composition of major lipid classes in marine invertebrate cells, and these effects can vary depending on the provenance of the cells. We have found that (I) both cell viability and the FA profile of cell lipids after cryopreservation depend on the cryoprotectants used; (II) an amount of saturated, monoenic and polyenic FAs changes significantly after cryopreservation. We assume that the addition of the exogenous lipid extract in form of liposomes could promote a renewal of disturbance areas and prevent from membrane damages during freezing-thawing.

The fatty acid (FA) composition of the main membrane phospholipids, phosphatidylcholine (PC) and phosphatidylethanolamine (PE), was investigated in the muscle, gills, and liver of the small-scaled redfin Tribolodon brandti (Dybowskii, 1872) at different temperatures under natural and experimental conditions. It was established that a water temperature decrease in the natural habitat was accompanied by an increase in polyunsaturated fatty acid contents and the unsaturation index, mainly at the expense of FAs of the ω3 series (20:5ω3 and 22:6ω3), and by a decrease in saturated fatty acid levels in PC and PE. A similar, but less pronounced tendency was revealed in experiments with a rapid lowering of water temperature. These findings suggest the weak adaptation ability of the small-scaled redfin to a drastic shift of environmental temperature. Temperature changes produced the greatest alterations in the FA composition of phospholipids in the liver and the smallest changes occurred in muscle tissue.

The phospholipid composition of organs and tissues was investigated in representatives of five phyla of marine invertebrates: Annelida (Chaetopterus variopedatus, Serpula vermicularis), Echiuroidea (Urechis unicinctus), Sipunculoidea (Phascolosoma japonicum), Mollusca (Gastropoda: Tectonatica janthostoma, Neptunea polycostata; Bivalvia: Mactra sulcataria, Peronidia venuloza, Patinopecten yessoensis, Crenomytilus grayanus; Cephalopoda: Octopus conispadiceus, Todarodes pacificus), and Arthropoda (Paralythodes camtschatica, Erimacrus isenbekii, Hemigrapsus sanguineus, Pinixa rathbunii). The specificity of phospholipid distribution was shown to be related to the taxonomic position of marine invertebrates and functional properties of their organs and tissues. In Echiuroidea, Sipunculoidea, and Arthropoda, ceramide aminoethylphosphonate was found only in the digestive organs. This suggests an exogenous origin of this phospholipid.

The study deals with the lipid and fatty acid compositions of the muscles, gills and liver of marine fishes inhabiting cold waters (0.5–6°C) and caught in Peter the Great Bay (3 species) and Vostok Bay (2 species), as well as with the fatty acid compositions of the main phospholipids in the muscle tissues of fish from Olyutorskii Bay (4 species). The average content of phosphatidylcholine was about 60% in muscles and in the liver and 53.8% of the sum of all phospholipids in gills. The phosphatidylethanolamine content was on the average 24.3, 25.1 and 22.3% in muscles, liver and gills, respectively. Increased contents of phosphatidylserine and sphingomyeline were recorded in the gills. The mean (S.D.) molar ratios of cholesterol/phospholipids were 0.20, 0.32, and 0.58 in the muscles, liver, and gills, respectively. It was established that phosphatidylcholine has a higher content of saturated fatty acids, whereas phosphatidylethanolamine was richer in monoenic acids and docosahexaenoic acid (DHA). It was noted that the level of polyenic fatty acids was increasing and the level of monoenic and saturated acids was decreasing in the series from gills-liver-muscles. The species with a more active mode of life were distinguished by an increased content of docosahexaenoic acid.
