1. Combined UV and IR ozone profile retrieval from TROPOMI and CrIS measurements

Abstract

Vertical ozone profiles from combined spectral measurements in the ultraviolet and infrared spectral range were retrieved by using data from the TROPOspheric Monitoring Instrument on the Sentinel-5 Precursor (TROPOMI/S5P) and the Cross-track Infrared Sounder on the Suomi National Polar-orbiting Partnership (CrIS/Suomi-NPP), which are flying in loose formation 3 min apart in the same orbit. A previous study of ozone profiles retrieved exclusively from TROPOMI UV spectra showed that the vertical resolution in the troposphere is clearly limited (Mettig et al., 2021). The vertical resolution and the vertical extent of the ozone profiles is improved by combining both wavelength ranges compared to retrievals limited to UV or IR spectral data only. The combined retrieval particularly improves the accuracy of the retrieved tropospheric ozone and to a lesser degree stratospheric ozone up to 30 km. An increase in the degrees of freedom (DOF) by 1 DOF was found in the UV + IR retrieval compared to the UV-only retrieval. Compared to previous publications, which investigated combinations of UV and IR observations from the Ozone Monitoring Instrument and Tropospheric Emission Spectrometer (OMI and TES) and Global Ozone Monitoring Experiment version 2 and Infrared Atmospheric Sounding Interferometer (GOME-2 and IASI) pairs, the degree of freedom is lower, which is attributed to the reduced spectral resolution of CrIS compared to TES or IASI. Tropospheric lidar and ozonesondes were used to validate the ozone profiles and tropospheric ozone content (TOC). In their comparison with tropospheric lidars, both ozone profiles and TOCs show smaller biases for the retrieved data from the combined UV + IR observation than from the UV observations alone. For the ozone profiles below 10 km, the mean differences are around ±10 % and the mean TOC varies around ±3 DU. We show that TOCs from the combined retrieval agree better with ozonesonde results at northern latitudes than the UV-only and IR-only retrievals and also have lower scatter. In the tropics, the IR-only retrieval shows the best agrement with TOCs derived from ozonesondes. While in general the TOCs show good agreement with ozonesonde data, the profiles have a positive bias of around 30 % between 10 and 15 km. The reason is probably a positive stratospheric bias from the IR retrieval. The comparison of the UV + IR and UV ozone profiles up to 30 km with the Microwave Limb Sounder (MLS) demonstrates the improvement of the UV + IR profile in the stratosphere above 18 km. In comparison to the UV-only approach the retrieval shows improvements of up to 10 % depending on latitude but can also show worse results in some regions and latitudes.

Abstract

Abstract Background Being exposed to crises during pregnancy can affect maternal health through stress exposure, which can in return impact neonatal health. We investigated temporal trends in neonatal outcomes in Switzerland between 2007 and 2022 and their variations depending on exposure to the economic crisis of 2008, the flu pandemic of 2009, heatwaves (2015 and 2018) and the COVID-19 pandemic. Methods Using individual cross-sectional data encompassing all births occurring in Switzerland at the monthly level (2007-2022), we analysed changes in birth weight and in the rates of preterm birth (PTB) and stillbirth through time with generalized additive models. We assessed whether the intensity or length of crisis exposure was associated with variations in these outcomes. Furthermore, we explored effects of exposure depending on trimesters of pregnancy. Results Over 1.2 million singleton births were included in our analyses. While birth weight and the rate of stillbirth have remained stable since 2007, the rate of PTB has declined by one percentage point. Exposure to the crises led to different results, but effect sizes were overall small. Exposure to COVID-19, irrespective of the pregnancy trimester, was associated with a higher birth weight (+12 grams [95% confidence interval (CI) 5.5 to 17.9 grams]). Being exposed to COVID-19 during the last trimester was associated with an increased risk of stillbirth (odds ratio 1.24 [95%CI 1.02 to 1.50]). Exposure to the 2008 economic crisis during pregnancy was not associated with any changes in neonatal health outcomes, while heatwave effect was difficult to interpret. Conclusion Overall, maternal and neonatal health demonstrated resilience to the economic crisis and to the COVID-19 pandemic in a high-income country like Switzerland. However, the effect of exposure to the COVID-19 pandemic is dual, and the negative impact of maternal infection on pregnancy is well-documented. Stress exposure and economic constraint may also have had adverse effects among the most vulnerable subgroups of Switzerland. To investigate better the impact of heatwave exposure on neonatal health, weekly or daily-level data is needed, instead of monthly-level data.

Abstract

The Match technique was used to determine chemically induced ozone loss inside the stratospheric vortex during the Arctic winter 2002/2003. From end of November 2002, which is the earliest start of a Match campaign ever, until end of March 2003 approximately 800 ozonesondes were launched from 34 stations in the Arctic and mid latitudes. Ozone loss rates were quantified from the beginning of December until mid-March in the vertical region of 400–550 K potential temperature. In accordance with the occurrence of a large area of conditions favourable for the formation of polar stratospheric clouds in December ozone destruction rates varied between 10–15 ppbv/day depending on height. Maximum loss rates around 35 ppbv/day were reached during late January. Afterwards ozone loss rates decreased until mid-March when the final warming of the vortex began. In the period from 2 December 2002 to 16 March 2003 the accumulated ozone loss reduced the partial ozone column of 400–500 K potential temperature by 56±4 DU. This value is in good agreement with that inferred from the empirical relation of ozone loss against the volume of potential polar stratospheric clouds within the northern hemisphere. The sensitivity of the results on recent improvements of the approach has been tested.

Abstract

A total ozone depletion of 68±7 Dobson units between 380 and 525K from 10 December 2002 to 10 March 2003 is derived from ozone sonde data by the vortex-average method, taking into account both diabatic descent of the air masses and transport of air into the vortex. When the vortex is divided into three equal-area regions, the results are 85±9DU for the collar region (closest to the edge), 52±5DU for the vortex centre and 68±7DU for the middle region in between centre and collar. Our results compare well with other studies: We find good agreement with ozone loss deduced from SAOZ data, with results inferred from POAM III observations and with results from tracer-tracer correlations using HF as the long-lived tracer. We find a higher ozone loss than that deduced by tracer-tracer correlations using CH4. We have made a careful comparison with Match results: The results were recalculated using a common time period, vortex edge definition and height interval. The two methods generally compare very well, except at the 475K level which exhibits an unexplained discrepancy.

Abstract

Ozone measurements from ozonesondes, AROTAL, DIAL, and POAM III instruments during the SOLVE-2/VINTERSOL period are composited in a time-varying, flow-following quasi-conservative (PV-θ) coordinate space; the resulting composites from each instrument are mapped onto the other instruments' locations and times. The mapped data are then used to intercompare data from the different instruments. Overall, the four ozone data sets are found to be in good agreement. AROTAL shows somewhat lower values below 16 km, and DIAL has a positive bias at the upper limits of its altitude range. These intercomparisons are consistent with those obtained from more conventional near-coincident profiles, where available. Although the PV-θ mapping technique entails larger uncertainties of individual profile differences compared to direct near-coincident comparisons, the ability to include much larger numbers of comparisons can make this technique advantageous.

Abstract

Background The International epidemiology Databases to Evaluate AIDS conducts research in several regions, including in Southern Africa. We assessed authorship inequalities for the Southern African region, which is led by South African and Swiss investigators.Methods We analysed authorships of publications from 2007 to 2020 by gender, country income group, time and citation impact. We used 2020 World Bank categories to define income groups and the relative citation ratio (RCR) to assess citation impact. Authorship parasitism was defined as articles without authors from the countries where the study was conducted. A regression model examined the probability of different authorship positions.Results We included 313 articles. Of the 1064 contributing authors, 547 (51.4%) were women, and 223 (21.0%) were from 32 low-income/lower middle-income countries (LLMICs), 269 (25.3%) were from 13 upper middle-income countries and 572 (53.8%) were from 25 high-income countries (HICs). Most articles (150/157, 95.5%) reporting data from Southern Africa included authors from all participating countries. Women were more likely to be the first author than men (OR 1.74; 95% CI 1.06 to 2.83) but less likely to be last authors (OR 0.63; 95% CI 0.40 to 0.99). Compared with HIC, LLMIC authors were less likely to publish as first (OR 0.21; 95% CI 0.11 to 0.41) or last author (OR 0.20; 95% CI 0.09 to 0.42). The proportion of women and LLMIC first and last authors increased over time. The RCR tended to be higher, indicating greater impact, if first or last authors were from HIC (p=0.06).Conclusions This analysis of a global health collaboration co-led by South African and Swiss investigators showed little evidence of authorship parasitism. There were stark inequalities in authorship position, with women occupying more first and men more last author positions and researchers from LLMIC being ‘stuck in the middle’ on the byline. Global health research collaborations should monitor, analyse and address authorship inequalities.

Abstract

We validate the Ozone Monitoring Instrument (OMI) Ozone Profile (PROFOZ) product from October 2004 through December 2014 retrieved by the Smithsonian Astrophysical Observatory (SAO) algorithm against ozonesonde observations. We also evaluate the effects of OMI row anomaly (RA) on the retrieval by dividing the dataset into before and after the occurrence of serious OMI RA, i.e., pre-RA (2004–2008) and post-RA (2009–2014). The retrieval shows good agreement with ozonesondes in the tropics and midlatitudes and for pressure ∼ 50 hPa after applying OMI averaging kernels to ozonesonde data. The MBs of the stratospheric ozone column (SOC, the ozone column from the tropopause pressure to the ozonesonde burst pressure) are within 2 % with SDs of ∼ 50 hPa. The SOC MBs increase up to 3 % with SDs as great as 6 % and the TOC SDs increase up to 30 %. The comparison generally degrades at larger solar zenith angles (SZA) due to weaker signals and additional sources of error, leading to worse performance at high latitudes and during the midlatitude winter. Agreement also degrades with increasing cloudiness for pressure > ∼ 100 hPa and varies with cross-track position, especially with large MBs and SDs at extreme off-nadir positions. In the tropics and midlatitudes, the post-RA comparison is considerably worse with larger SDs reaching 2 % in the stratosphere and 8 % in the troposphere and up to 6 % in TOC. There are systematic differences that vary with latitude compared to the pre-RA comparison. The retrieval comparison demonstrates good long-term stability during the pre-RA period but exhibits a statistically significant trend of 0.14–0.7 % year−1 for pressure

From end of November 2002, which is the earliest start of a Match campaign ever, until end of March 2003 approximately 800 ozonesondes were launched from 34 stations in the Arctic and mid latitudes. Ozone loss rates were quantified from the beginning of December until mid-March in the vertical region of 400–550 K potential temperature. In accordance with the occurrence of a large area of conditions favourable for the formation of polar stratospheric clouds in December ozone destruction rates varied between 10–15 ppbv/day depending on height. Maximum loss rates around 35 ppbv/day were reached during late January. Afterwards ozone loss rates decreased until mid-March when the final warming of the vortex began. In the period from 2 December 2002 to 16 March 2003 the accumulated ozone loss reduced the partial ozone column of 400–500 K potential temperature by 56±4 DU. This value is in good agreement with that inferred from the empirical relation of ozone loss against the volume of potential polar stratospheric clouds within the northern hemisphere. The sensitivity of the results on recent improvements of the approach has been tested.

Abstract

A total ozone depletion of 68±7 Dobson units between 380 and 525K from 10 December 2002 to 10 March 2003 is derived from ozone sonde data by the vortex-average method, taking into account both diabatic descent of the air masses and transport of air into the vortex. When the vortex is divided into three equal-area regions, the results are 85±9DU for the collar region (closest to the edge), 52±5DU for the vortex centre and 68±7DU for the middle region in between centre and collar. Our results compare well with other studies: We find good agreement with ozone loss deduced from SAOZ data, with results inferred from POAMIII observations and with results from tracer-tracer correlations using HF as the long-lived tracer. We find a higher ozone loss than that deduced by tracer-tracer correlations using CH4. We have made a careful comparison with Match results: The results were recalculated using a common time period, vortex edge definition and height interval. The two methods generally compare very well, except at the 475K level which exhibits an unexplained discrepancy.

Abstract

Forced labour is a serious crime that currently affects thousands of people across the UK -- and the number of cases is growing. JRF has supported research into the nature, scale and scope of forced labour in the UK since 2010. As the UK Government, Northern Ireland Assembly and Scottish Parliament consider new legislation to tackle the issue, this round-up draws together JRF's programme of research, highlighting the most significant findings and key recommendations.Key points:The growth of forced labour has coincided with changes in the nature of the UK's labour market. Increasing casualisation of jobs and longer supply chains within big companies have led to greater potential for workers to be exploited. The government's light-touch approach to workforce regulation, weak enforcement of labour standards and immigration policies that exclude people from formal employment also make workers more vulnerable.Forced labour can take many forms, and is not limited to immigrant workers or those who are working in the UK illegally. Interviews with those affected reveal different types of exploitation and the research explains why workers in some industries are particularly prone to it.Improved regulation, enforcement and protection for those affected is needed, and this document recommends ways it can be provided. It stresses that forced labour will only be eradicated through greater joined-up working by the government, which must address the causes, not just the symptoms.

Abstract

We estimate the tropospheric column ozone using a forward trajectory model to increase the horizontal resolution of the Aura Microwave Limb Sounder (MLS) derived stratospheric column ozone. Subtracting the MLS stratospheric column from Ozone Monitoring Instrument total column measurements gives the trajectory enhanced tropospheric ozone residual (TTOR). Because of different tropopause definitions, we validate the basic residual technique by computing the 200-hPa-to-surface column and comparing it to the same product from ozonesondes and Tropospheric Emission Spectrometer measurements. Comparisons show good agreement in the tropics and reasonable agreement at middle latitudes, but there is a persistent low bias in the TTOR that may be due to a slight high bias in MLS stratospheric column. With the improved stratospheric column resolution, we note a strong correlation of extratropical tropospheric ozone column anomalies with probable troposphere-stratosphere exchange events or folds. The folds can be identified by their colocation with strong horizontal tropopause gradients. TTOR anomalies due to folds may be mistaken for pollution events since folds often occur in the Atlantic and Pacific pollution corridors. We also compare the 200-hPa-to-surface column with Global Modeling Initiative chemical model estimates of the same quantity. While the tropical comparisons are good, we note that chemical model variations in 200-hPa-to-surface column at middle latitudes are much smaller than seen in the TTOR.

Abstract

International audience; The Arctic polar vortex exhibited widespread regions of low temperatures during the winter of 2005, resulting in significant ozone depletion by chlorine and bromine species. We show that chemical loss of column ozone (ΔO3) and the volume of Arctic vortex air cold enough to support the existence of polar stratospheric clouds (VPSC) both exceed levels found for any other Arctic winter during the past 40 years. Cold conditions and ozone loss in the lowermost Arctic stratosphere (e.g., between potential temperatures of 360 to 400 K) were particularly unusual compared to previous years. Measurements indicate ΔO3 = 121 +/- 20 DU and that ΔO3 versus VPSC lies along an extension of the compact, near linear relation observed for previous Arctic winters. The maximum value of VPSC during five to ten year intervals exhibits a steady, monotonic increase over the past four decades, indicating that the coldest Arctic winters have become significantly colder, and hence are more conducive to ozone depletion by anthropogenic halogens.

Abstract

Ozone measurements from ozonesondes, AROTAL, DIAL, and POAM III instruments during the SOLVE-2/VINTERSOL period are composited in a time-varying, flow-following quasi-conservative (PV-θ) coordinate space; the resulting composites from each instrument are mapped onto the other instruments' locations and times. The mapped data are then used to intercompare data from the different instruments. Overall, the four ozone data sets are found to be in good agreement. AROTAL shows somewhat lower values below 16 km, and DIAL has a positive bias at the upper limits of its altitude range. These intercomparisons are consistent with those obtained from more conventional near-coincident profiles, where available. Although the PV-θ mapping technique entails larger uncertainties of individual profile differences compared to direct near-coincident comparisons, the ability to include much larger numbers of comparisons can make this technique advantageous.

Abstract

During Arctic winters with a cold, stable stratospheric circulation, reactions on the surface of polar stratospheric clouds (PSCs) lead to elevated abundances of chlorine monoxide (ClO) that, in the presence of sunlight, destroy ozone. Here we show that PSCs were more widespread during the 1999/2000 Arctic winter than for any other Arctic winter in the past two decades. We have used three fundamentally different approaches to derive the degree of chemical ozone loss from ozonesonde, balloon, aircraft, and satellite instruments. We show that the ozone losses derived from these different instruments and approaches agree very well, resulting in a high level of confidence in the results. Chemical processes led to a 70% reduction of ozone for a region similar to1 km thick of the lower stratosphere, the largest degree of local loss ever reported for the Arctic. The Match analysis of ozonesonde data shows that the accumulated chemical loss of ozone inside the Arctic vortex totaled 117 +/- 14 Dobson units (DU) by the end of winter. This loss, combined with dynamical redistribution of air parcels, resulted in a 88 +/- 13 DU reduction in total column ozone compared to the amount that would have been present in the absence of any chemical loss. The chemical loss of ozone throughout the winter was nearly balanced by dynamical resupply of ozone to the vortex, resulting in a relatively constant value of total ozone of 340 +/- 50 DU between early January and late March. This observation of nearly constant total ozone in the Arctic vortex is in contrast to the increase of total column ozone between January and March that is observed during most years.

Abstract

Ozone observations made by the Airborne Raman Ozone, Temperature, and Aerosol Lidar(AROTEL) and Differential Absorption Lidar (DIAL) on board the NASA DC-8 aircraft, the NOAAin situ instrument on board the NASA ER-2 aircraft, and Third European Stratospheric Experimenton Ozone 2000 (THESEO 2000) ozonesondes are analyzed by applying a quasi-conservativecoordinate mapping technique. Measurements from the late winter/early spring SAGE III OzoneLoss and Validation Experiment (SOLVE) period (January through March 2000) are incorporatedinto a time-varying composite field in a potential vorticity-potential temperature coordinate space;ozone loss rates are calculated both with and without diabatic effects. The average loss rate frommid-January to mid-March near the 450 K isentropic surface in the polar vortex is found to beapproximately 0.03 ppmv/d.

Abstract

We present validation studies of MLS version 2.2 upper tropospheric and stratospheric ozone profiles using ozonesonde and lidar data as well as climatological data. Ozone measurements from over 60 ozonesonde stations worldwide and three lidar stations are compared with coincident MLS data. The MLS ozone stratospheric data between 150 and 3 hPa agree well with ozonesonde measurements, within 8% for the global average. MLS values at 215 hPa are biased high compared to ozonesondes by A`20% at middle to high latitude, although there is a lot of variability in this altitude region. Comparisons between MLS and ground-based lidar measurements from Mauna Loa, Hawaii, from the Table Mountain Facility, California, and from the Observatoire de Haute-Provence, France, give very good agreement, within A`5%, for the stratospheric values. The comparisons between MLS and the Table Mountain Facility tropospheric ozone lidar show that MLS data are biased high by A`30% at 215 hPa, consistent with that indicated by the ozonesonde data. We obtain better global average agreement between MLS and ozonesonde partial column values down to 215 hPa, although the average MLS values at low to middle latitudes are higher than the ozonesonde values by up to a few percent. MLS v2.2 ozone data agree better than the MLS v1.5 data with ozonesonde and lidar measurements. MLS tropical data show the wave one longitudinal pattern in the upper troposphere, with similarities to the average distribution from ozonesondes. High upper tropospheric ozone values are also observed by MLS in the tropical Pacific from June to November.

Abstract

We validate the Ozone Monitoring Instrument (OMI) ozone-profile (PROFOZ) product from October 2004 through December 2014 retrieved by the Smithsonian Astrophysical Observatory (SAO) algorithm against ozonesonde observations. We also evaluate the effects of OMI Row anomaly (RA) on the retrieval by dividing the data set into before and after the occurrence of serious OMI RA, i.e., pre-RA (2004-2008) and post-RA (2009-2014). The retrieval shows good agreement with ozonesondes in the tropics and mid-latitudes and for pressure < ~50 hPa in the high latitudes. It demonstrates clear improvement over the a priori down to the lower troposphere in the tropics and down to an average of ~550 (300) hPa at middle (high latitudes). In the tropics and mid-latitudes, the profile mean biases (MBs) are less than 6%, and the standard deviations (SDs) range from 5-10% for pressure < ~50 hPa to less than 18% (27%) in the tropics (midlatitudes) for pressure > ~50 hPa after applying OMI averaging kernels to ozonesonde data. The MBs of the stratospheric ozone column (SOC) are within 2% with SDs of < 5% and the MBs of the tropospheric ozone column (TOC) are within 6% with SDs of 15%. In the high latitudes, the profile MBs are within 10% with SDs of 5-15% for pressure < ~50 hPa, but increase to 30% with SDs as great as 40% for pressure > ~50 hPa. The SOC MBs increase up to 3% with SDs as great as 6% and the TOC SDs increase up to 30%. The comparison generally degrades at larger solarzenith angles (SZA) due to weaker signals and additional sources of error, leading to worse performance at high latitudes and during the mid-latitude winter. Agreement also degrades with increasing cloudiness for pressure > ~100 hPa and varies with cross-track position, especially with large MBs and SDs at extreme off-nadir positions. In the tropics and mid-latitudes, the post-RA comparison is considerably worse with larger SDs reaching 2% in the stratosphere and 8% in the troposphere and up to 6% in TOC. There are systematic differences that vary with latitude compared to the pre-RA comparison. The retrieval comparison demonstrates good long-term stability during the pre-RA period, but exhibits a statistically significant trend of 0.14-0.7%/year for pressure < ~ 80 hPa, 0.7 DU/year in SOC and -0.33 DU/year in TOC during the post-RA period. 21. The effect of the COVID-19 lockdown on mental health care use in South Africa: an interrupted time-series analysis

AbstractAimsThe coronavirus disease 2019 (COVID-19) pandemic and ensuing restrictions have negatively affected the mental health and well-being of the general population, and there is increasing evidence suggesting that lockdowns have led to a disruption of health services. In March 2020, South Africa introduced a lockdown in response to the COVID-19 pandemic, entailing the suspension of all non-essential activities and a complete ban of tobacco and alcohol sales. We studied the effect of the lockdown on mental health care utilisation rates in private-sector care in South Africa.MethodsWe conducted an interrupted time-series analysis using insurance claims from 1 January 2017 to 1 June 2020 of beneficiaries 18 years or older from a large private sector medical insurance scheme. We calculated weekly outpatient consultation and hospital admission rates for organic mental disorders, substance use disorders, serious mental disorders, depression, anxiety, other mental disorders, any mental disorder and alcohol withdrawal syndrome. We calculated adjusted odds ratios (OR) for the effect of the lockdown on weekly outpatient consultation and hospital admission rates and the weekly change in rates during the lockdown until 1 June 2020.Results710 367 persons were followed up for a median of 153 weeks. Hospital admission rates (OR 0.38; 95% confidence interval (CI) 0.33–0.44) and outpatient consultation rates (OR 0.74; 95% CI 0.63–0.87) for any mental disorder decreased substantially after the introduction of the lockdown and did not recover to pre-lockdown levels by 1 June 2020. Health care utilisation rates for alcohol withdrawal syndrome doubled after the introduction of the lockdown, but the statistical uncertainty around the estimates was large (OR 2.24; 95% CI 0.69–7.24).ConclusionsMental health care utilisation rates for inpatient and outpatient services decreased substantially after the introduction of the lockdown. Hospital admissions and outpatient consultations for alcohol withdrawal syndrome increased after the introduction of the lockdown, but statistical uncertainty precludes strong conclusions about a potential unintended effect of the alcohol sales ban. Governments should integrate strategies for ensuring access and continuity of essential mental health services during lockdowns in pandemic preparedness planning.

Abstract

ABSTRACTThe ability to identify bacterial pathogens at the subspecies level in clinical diagnostics is currently limited. We investigated whether splitting Escherichia colispecies into clonal groups (clonotypes) predicts antimicrobial susceptibility or clinical outcome. A total of 1,679 extraintestinal E. coliisolates (collected from 2010 to 2012) were collected from one German and 5 U.S. clinical microbiology laboratories. Clonotype identity was determined by fumCand fimH(CH) sequencing. The associations of clonotype with antimicrobial susceptibility and clinical variables were evaluated. CH typing divided the isolates into >200 CH clonotypes, with 93% of the isolates belonging to clonotypes with =2 isolates. Antimicrobial susceptibility varied substantially among clonotypes but was consistent across different locations. Clonotype-guided antimicrobial selection significantly reduced “drug-bug” mismatch compared to that which occurs with the use of conventional empirical therapy. With trimethoprim-sulfamethoxazole and fluoroquinolones, the drug-bug mismatch was predicted to decrease 62% and 78%, respectively. Recurrent or persistent urinary tract infection and clinical sepsis were significantly correlated with specific clonotypes, especially with CH40-30 (also known as H30), a recently described clonotype within sequence type 131 (ST131). We were able to clonotype directly from patient urine samples within 1 to 3 h of obtaining the specimen. In E. coli, subspecies-level identification by clonotyping can be used to significantly improve empirical predictions of antimicrobial susceptibility and clinical outcomes in a timely manner.

Homogenized data series of total ozone measurements taken by the regularly and well calibrated Dobson and Brewer spectrophotometers at Hradec Králové (Czech) and the data from the re-analyses ERA-40 and ERA-Interim were merged and compared to investigate differences between the particular data sets originated in Central Europe, the Northern Hemisphere (NH) mid-latitudes. The Dobson-to-Brewer transfer function and the algorithm for approximation of the data from the re-analyses were developed, tested and applied for creation of instrumentally consistent and completed total ozone data series of the 50-yr period 1961–2010 of observations. This correction has reduced the well-known seasonal differences between Dobson and Brewer data below the 1% calibration limit of the spectrophotometers. Incorporation of the ERA-40 and ERA-Interim total ozone data on days with missing measurements significantly improved completeness and reliability of the data series mainly in the first two decades of the period concerned. Consistent behaviour of the original and corrected/merged data sets was found in the pre-ozone-hole period (1961–1985). In the post-Pinatubo (1994–2010) era the data series show seasonal differences that can introduce uncertainty in estimation of ozone recovery mainly in the winter-spring season when the effect of the Montreal Protocol and its Amendments is expected. All the data sets confirm substantial depletion of ozone also in the summer months that gives rise to the question about its origin. The merged and completed data series of total ozone will be further analyzed to quantify chemical ozone losses and contribution of natural atmospheric processes to the ozone depletion over the region. This case study points out the importance of selection and evaluation of the quality and consistency of the input data sets used in estimation of long-term ozone changes including recovery of the ozone layer over the selected areas. 26. Longitudinal study revealing motor, cognitive and behavioral decline in a transgenic minipig model of Huntington's disease

Abstract

Huntington's disease (HD) is an inherited devastating neurodegenerative disease with no known cure to date. Several therapeutic treatments for HD are in development, but their safety, tolerability and efficacy need to be tested before translation to bedside. The monogenetic nature of this disorder has enabled the generation of transgenic animal models carrying a mutant huntingtin (mHTT) gene causing HD. A large animal model reflecting disease progression in humans would be beneficial for testing the potential therapeutic approaches. Progression of the motor, cognitive and behavioral phenotype was monitored in transgenic Huntington's disease minipigs (TgHD) expressing the N-terminal part of human mHTT. New tests were established to investigate physical activity by telemetry, and to explore the stress-induced behavioral and cognitive changes in minipigs. The longitudinal study revealed significant differences between 6- to 8-year-old TgHD animals and their wild-type (WT) controls in a majority of the tests. The telemetric study showed increased physical activity of 4.6- to 6.5-year-old TgHD boars compared to their WT counterparts during the lunch period as well as in the afternoon. Abstract

International audience; Chemical ozone loss rates inside the Arctic polar vortex were determined in early 1998 and early 1999 by using the Match technique based on coordinated ozonesonde measurements. These two winters provide the only opportunities in recent years to investigate chemical ozone loss in a warm Arctic vortex under threshold conditions, i.e., where the preconditions for chlorine activation, and hence ozone destruction, only occurred occasionally. In 1998, results were obtained in January and February between 410 and 520 K. The overall ozone loss was observed to be largely insignificant, with the exception of late February, when those air parcels exposed to temperatures below 195 K were affected by chemical ozone loss. In 1999, results are confined to the 475 K isentropic level, where no significant ozone loss was observed. Average temperatures were some 8°-10° higher than those in 1995, 1996, and 1997, when substantial chemical ozone loss occurred. The results underline the strong dependence of the chemical ozone loss on the stratospheric temperatures. This study shows that enhanced chlorine alone does not provide a sufficient condition for ozone loss. The evolution of stratospheric temperatures over the next decade will be the determining factor for the amount of wintertime chemical ozone loss in the Arctic stratosphere.

Abstract

Summary: Background: Drug resistance threatens global tuberculosis control. We aimed to examine mortality in patients with tuberculosis from high-burden countries, according to concordance or discordance of results from drug susceptibility testing done locally and whole-genome sequencing (WGS). Methods: In this multicentre cohort study, we collected pulmonary Mycobacterium tuberculosis isolates and clinical data from individuals with tuberculosis from antiretroviral therapy programmes and tuberculosis clinics in Côte d'Ivoire, Democratic Republic of the Congo, Kenya, Nigeria, Peru, South Africa, and Thailand, stratified by HIV status and drug resistance. Sites tested drug susceptibility using routinely available methods. WGS was done on Illumina HiSeq 2500 in the USA and Switzerland, and TBprofiler was used to analyse the genomes. We included individuals aged 16 years or older with pulmonary tuberculosis (bacteriologically confirmed or clinically diagnosed). We analysed mortality in multivariable logistic regression models adjusted for sex, age, HIV status, history of tuberculosis, and sputum positivity. Findings: Between Sept 1, 2014, and July 4, 2016, of 634 patients included in our previous analysis, we included 582 patients with tuberculosis (median age 33 years [IQR 27–43], 225 [39%] women, and 247 [42%] HIV-positive). Based on WGS, 339 (58%) isolates were pan-susceptible, 35 (6%) monoresistant, 146 (25%) multidrug-resistant, and 24 (4%) pre-extensively drug-resistant (pre-XDR) or XDR. The analysis of mortality was based on 530 patients; 63 (12%) died and 77 (15%) patients received inappropriate treatment. Mortality ranged from 6% (18 of 310) in patients with pan-susceptible tuberculosis to 39% (nine of 23) in patients with pre-XDR or XDR tuberculosis. The adjusted odds ratio for mortality was 4·92 (95% CI 2·47–9·78) among undertreated patients, compared with appropriately treated patients. Interpretation: In seven countries with a high burden of tuberculosis, we observed discrepancies between drug resistance patterns obtained locally and WGS. The underdiagnosis of drug resistance resulted in inappropriate treatment and higher mortality. WGS can provide accurate and detailed drug resistance information required to improve the outcomes of drug-resistant tuberculosis in high-burden settings. Our results support WHO's call for point-of-care tests based on WGS. Abstract

Huntington's disease (HD) is an inherited devastating neurodegenerative disease with no known cure to date. Several therapeutic treatments for HD are in development, but their safety, tolerability and efficacy need to be tested before translation to bedside. The monogenetic nature of this disorder has enabled the generation of transgenic animal models carrying a mutant huntingtin (mHTT) gene causing HD. A large animal model reflecting disease progression in humans would be beneficial for testing the potential therapeutic approaches. Progression of the motor, cognitive and behavioral phenotype was monitored in transgenic Huntington's disease minipigs (TgHD) expressing the N-terminal part of human mHTT. New tests were established to investigate physical activity by telemetry, and to explore the stress-induced behavioral and cognitive changes in minipigs. The longitudinal study revealed significant differences between 6- to 8-year-old TgHD animals and their wild-type (WT) controls in a majority of the tests. The telemetric study showed increased physical activity of 4.6- to 6.5-year-old TgHD boars compared to their WT counterparts during the lunch period as well as in the afternoon. Our phenotypic study indicates progression in adult TgHD minipigs and therefore this model could be suitable for longstanding preclinical studies of HD.
