Abstract The receptor-binding site of influenza A virus hemagglutinin partially overlaps with major antigenic sites and constantly evolves. In this study, we observe that mutations G186D and D190N in the hemagglutinin receptor-binding site have coevolved in two recent human H3N2 clades. X-ray crystallography results show that these mutations coordinately drive the evolution of the hemagglutinin receptor binding mode. Epistasis between G186D and D190N is further demonstrated by glycan binding and thermostability analyses. Immunization and neutralization experiments using mouse and human samples indicate that the evolution of receptor binding mode is accompanied by a change in antigenicity. Besides, combinatorial mutagenesis reveals that G186D and D190N, along with other natural mutations in recent H3N2 strains, alter the compatibility with a common egg-adaptive mutation in seasonal influenza vaccines. Overall, our findings elucidate the role of epistasis in shaping the recent evolution of human H3N2 hemagglutinin and substantiate the high evolvability of its receptor-binding mode.

Incorporation of irreversible steps in pathway design enhances the overall thermodynamic favorability and often leads to better bioconversion yield given functional enzymes. Using this concept, here we constructed the first non-natural itaconate biosynthesis pathway driven by thioester hydrolysis. Itaconate is a commercially valuable platform chemical with wide applications in the synthetic polymer industry. Production of itaconate has long relied on the decarboxylation of TCA cycle intermediate cis-aconitate as the only biosynthetic route. Inspired by nature's design of itaconate detoxification, here we engineered a novel itaconate producing pathway orthogonal to native metabolism with no requirement of auxotrophic knock-out. The reversed degradation pathway initiates with pyruvate and acetyl-CoA condensation forming (S)-citramalyl-CoA, followed by its dehydration and isomerization into itaconyl-CoA then hydrolysis into itaconate. Phenylacetyl-CoA thioesterase (PaaI) from Escherichia coli was identified via screening to deliver the highest itaconate formation efficiency when coupled to the reversible activity of citramalate lyase and itaconyl-CoA hydratase. The preference of PaaI towards itaconyl-CoA hydrolysis over acetyl-CoA and (S)-citramalyl-CoA also minimized the inevitable precursor loss due to enzyme promiscuity. With acetate recycling, acetyl-CoA conservation, and condition optimization, we achieved a final itaconate titer of 1 g/L using the thioesterase driven pathway, which is a significant improvement compared to the original degradation pathway based on CoA transferase. This study illustrates the significance of thermodynamic favorability as a design principle in pathway engineering.

In order to further extend understanding of the heat transfer characteristics under an enclosed area, the influence of backpressure on a single impingement jet with small nozzle-to-plate spacing (H/D) is studied. Particle image velocimetry (PIV) technique and a commercial fluid dynamics solver (CFD) are respectively employed to investigate the fluid dynamics of the impinging jet, with a Reynolds number (Re) of 3462–6125, at a small nozzle-to-plate spacing (H/D) of 0.25–1.25. Experimental data are in well agreement with numerical simulation data. The findings indicate that the H/D ratio significantly influences the formation of backpressure in the flow field. The backpressure then influences the boundary layer velocity on the impingement surface, in turn affecting the impingement surface's heat transfer characteristics. In addition, a turning point in the horizontal velocity curve of H/D = 0.4 is found for an entire range of jet heights, which is independent of the Reynolds number (Re).

Abstract Einstein–Podolsky–Rosen (EPR) steering is a quantum effect based on quantum entanglement and it is the key resource for building quantum networks because of its useful properties. Based on the criterion for genuine multipartite EPR steering, the genuine quadripartite EPR steering is confirmed and it can be generated by a spontaneous parametric down-conversion cascaded process with two sum-frequency generations in an optical superlattice. This occurs either below the oscillation threshold and without oscillation threshold. The influence of the parameters of cascaded nonlinear process on the quadripartite EPR steering among signal, idler, and two sum-frequency beams are also discussed. Choosing appropriate nonlinear parameters can achieve good quadripartite quantum steering. This scheme of the generation of genuine quadripartite EPR steering has potential applications in quantum communication and computing.

Paddy rice is the second-largest grain crop in China and plays an important role in ensuring global food security. However, there is no high-resolution map of rice covering all of China. This study developed a new rice-mapping method by combining optical and synthetic aperture radar (SAR) images in cloudy areas based on the time-weighted dynamic time warping (TWDTW) method and produced distribution maps of single-season rice in 21 provincial administrative regions of China from 2017 to 2022 at a 10 or 20 m resolution. The accuracy was examined using 108 195 survey samples and county-level statistical data. On average, the user's, producer's, and overall accuracy values over all investigated provincial administrative regions were 73.08 %, 82.81 %, and 85.23 %, respectively. Compared with the statistical data from 2017 to 2019, the distribution maps explained 83 % of the spatial variation of county-level planting areas on average. Summary: The ability of the human immune system to generate antibodies to any given antigen can be strongly influenced by immunoglobulin V-gene allelic polymorphisms. However, previous studies have provided only limited examples. Therefore, the prevalence of this phenomenon has been unclear. By analyzing >1,000 publicly available antibody-antigen structures, we show that many V-gene allelic polymorphisms in antibody paratopes are determinants for antibody binding activity. Biolayer interferometry experiments further demonstrate that paratope allelic polymorphisms on both heavy and light chains often abolish antibody binding. We also illustrate the importance of minor V-gene allelic polymorphisms with low frequency in several broadly neutralizing antibodies to severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and influenza virus. Overall, this study not only highlights the pervasive impact of V-gene allelic polymorphisms on antibody binding but also provides mechanistic insights into the variability of antibody repertoires across individuals, which in turn have important implications for vaccine development and antibody discovery.

Einstein–Podolsky–Rosen (EPR) steering is different from quantum entanglement because of its unique asymmetry. Multipartite asymmetric EPR steering can break through one-to-one monogamy steering and achieve one to many one-way steering. That is to say, the state of one part can steer the other rest parts simultaneously, while the other parts cannot steer this part. Here, a scheme is proposed to generate one to many one-way steering by optical parametric amplification cascaded with a sum-frequency generation process. One to many non-monogamy EPR steering, such as one to two and one to three one-way steering are demonstrated based on the criterion for asymmetric EPR steering. It is also find that different kinds of asymmetric EPR steering can be obtained by choosing different nonlinear parameters. This non-monogamous one to many quantum control has potential application in constructing quantum networks and realizing one-way quantum computing.

To elucidate the molecular chemical compositions, volatility–polarity distributions, and influencing factors of Chinese cooking emissions, a comprehensive cooking emission experiment was conducted. Volatile organic compounds (VOCs), intermediate volatility, and semi-volatile organic compounds (I/SVOCs) from cooking fumes were analysed by a thermal desorption comprehensive two-dimensional gas chromatography coupled with quadrupole mass spectrometer (TD-GC × GC-qMS). Emissions from four typical Chinese dishes, i.e. fried chicken, Kung Pao chicken, pan-fried tofu, and stir-fried cabbage were investigated to illustrate the impact of cooking style and material. Fumes of chicken fried with corn, peanut, soybean, and sunflower oils were investigated to demonstrate the influence of cooking oil. A total of 201 chemicals were quantified. Kung Pao chicken emitted more pollutants than other dishes due to its rather intense cooking method. Aromatics and oxygenated compounds were extensively detected among meat-related cooking fumes, while a vegetable-related profile was observed in the emissions of stir-fried cabbage. Ozone formation potential (OFP) was dominated by chemicals in the VOC range. Of the secondary organic aerosol (SOA) estimation, 10.2 %–32.0 % could be explained by S/IVOCs. Pixel-based partial least squares discriminant analysis (PLS-DA) and multiway principal component analysis (MPCA) were utilized for sample classification and component identification. The results indicated that the oil factor explained more variance of chemical compositions than the cooking style factor. MPCA results emphasize the importance of the unsaturated fatty acid-alkadienal-volatile products mechanism (oil autoxidation) accelerated by the cooking and heating procedure.

In our earlier works, we have identified rate-limiting steps in the dark-to-light transition of PSII. By measuring chlorophyll a fluorescence transients elicited by single-turnover saturating flashes (STSFs) we have shown that in diuron-treated samples an STSF generates only F1 (< Fm) fluorescence level, and to produce the maximum (Fm) level, additional excitations are required, which, however, can only be effective if sufficiently long Δτ waiting times are allowed between the excitations. Biological variations in the half-rise time (Δτ1/2) of the fluorescence increment suggest that it may be sensitive to the physicochemical environment of PSII. Here, we investigated the influence of the lipidic environment on Δτ1/2 of PSII core complexes of Thermosynechococcus vulcanus. We found that while non-native lipids had no noticeable effects, thylakoid membrane lipids considerably shortened the Δτ1/2, from ~ 1 ms to ~ 0.2 ms. The importance of the presence of native lipids was confirmed by obtaining similarly short Δτ1/2 values in the whole T. vulcanus cells and isolated pea thylakoid membranes. Minor, lipid-dependent reorganizations were also observed by steady-state and time-resolved spectroscopic measurements. These data show that the processes beyond the dark-to-light transition of PSII depend significantly on the lipid matrix of the reaction center.

Objective: Obesity and nutrient oversupply increase mammalian target of rapamycin (mTOR) signaling in multiple cell types and organs, contributing to the onset of insulin resistance and complications of metabolic disease. However, it remains unclear when and where mTOR activation mediates these effects, limiting options for therapeutic intervention. The objective of this study was to isolate the role of constitutive mTOR activation in Nav1.8-expressing peripheral neurons in the onset of diet-induced obesity, bone loss, and metabolic disease. Methods: In humans, loss of function mutations in tuberous sclerosis complex 2 (TSC2) lead to maximal constitutive activation of mTOR. To mirror this in mice, we bred Nav1.8-Cre with TSC2fl/fl animals to conditionally delete TSC2 in Nav1.8-expressing neurons. Male and female mice were studied from 4- to 34-weeks of age and a subset of animals were fed a high-fat diet (HFD) for 24-weeks. Assays of metabolism, body composition, bone morphology, and behavior were performed. Results: By lineage tracing, Nav1.8-Cre targeted peripheral sensory neurons, a subpopulation of postganglionic sympathetics, and several regions of the brain. Conditional knockout of TSC2 in Nav1.8-expressing neurons (Nav1.8-TSC2KO) selectively upregulated neuronal mTORC1 signaling. Male, but not female, Nav1.8-TSC2KO mice had a 4–10% decrease in body size at baseline. When challenged with HFD, both male and female Nav1.8-TSC2KO mice resisted diet-induced gains in body mass. However, this did not protect against HFD-induced metabolic dysfunction and bone loss. In addition, despite not gaining weight, Nav1.8-TSC2KO mice fed HFD still developed high body fat, a unique phenotype previously referred to as ‘normal weight obesity’. Nav1.8-TSC2KO mice also had signs of chronic itch, mild increases in anxiety-like behavior, and sex-specific alterations in HFD-induced fat distribution that led to enhanced visceral obesity in males and preferential deposition of subcutaneous fat in females. Conclusions: Knockout of TSC2 in Nav1.8+ neurons increases itch- and anxiety-like behaviors and substantially modifies fat storage and metabolic responses to HFD. Though this prevents HFD-induced weight gain, it masks depot-specific fat expansion and persistent detrimental effects on metabolic health and peripheral organs such as bone, mimicking the ‘normal weight obesity’ phenotype that is of growing concern. This supports a mechanism by which increased neuronal mTOR signaling can predispose to altered adipose tissue distribution, adipose tissue expansion, impaired peripheral metabolism, and detrimental changes to skeletal health with HFD – despite resistance to weight gain.

The purpose of this review is to outline our understanding of the nature, mechanism and physiological significance of light-induced reversible reorganizations in closed Type II reaction centre (RC) complexes. In the so-called ‘closed' state, purple bacterial RC (bRC) and photosystem II (PSII) RC complexes are incapable of generating additional stable charge separation. Yet, upon continued excitation they display well-discernible changes in their photophysical and photochemical parameters. Substantial stabilization of their charge-separated states has been thoroughly documented—uncovering light-induced reorganizations in closed RCs and revealing their physiological importance in gradually optimizing the operation of the photosynthetic machinery during the dark-to-light transition. A range of subtle light-induced conformational changes has indeed been detected experimentally in different laboratories using different bRC and PSII-containing preparations. In general, the presently available data strongly suggest similar structural dynamics of closed bRC and PSII RC complexes, and similar physical mechanisms, in which dielectric relaxation processes and structural memory effects of proteins are proposed to play important roles.

In the present work, we conducted experiments of secondary organic aerosol (SOA) formation from urban cooking and vehicular sources to characterize the mass spectral features of primary organic aerosol (POA) and SOA using an high-resolution time-of-flight aerosol mass spectrometer (HR-ToF-AMS). Our results showed that the cooking styles have a greater impact on aged COA (cooking organic aerosol) mass spectra than oxidation conditions. However, the oxidation conditions affect the aged HOA (hydrocarbon-like OA) spectra more significantly than vehicle operating conditions. In our study, we use mass spectra similarity analysis and positive matrix factorization (PMF) analysis to establish the POA and SOA mass spectra of these two sources. These mass spectra are used as source constraints in a multilinear engine (ME-2) model to apportion the OA (organic aerosol) sources in the atmosphere. Compared with the traditional ambient PMF results, the improved ME-2 model can better quantify the contribution of POA and SOA from cooking and vehicular sources. Our work, for the first time, establishes the vehicle and cooking SOA source profiles, and can be further used in the OA source apportionment in the ambient atmosphere.

Vehicle exhaust and cooking emissions are closely related to the daily life of city dwellers. Here, we defined the secondary organic aerosols (SOAs) derived from vehicle exhaust and cooking emissions as “urban-lifestyle SOAs” and simulated their formation using a Gothenburg potential aerosol mass reactor (Go:PAM). The vehicle exhaust and cooking emissions were separately simulated, and their samples were defined as “vehicle group” and “cooking group”, respectively. After samples had been aged under 0.3–5.5 d of equivalent photochemical age, these two urban-lifestyle SOAs showed markedly distinct features in the SOA mass growth potential, oxidation pathways, and mass spectra. The SOA/POA (primary organic aerosol) mass ratios of vehicle groups (107) were 44 times larger than those of cooking groups (2.38) at about 2 d of equivalent photochemical age, according to the measurement of scanning mobility particle sizer (SMPS). A high-resolution time-of-flight aerosol mass spectrometer was used to perform a deeper analysis. It revealed that organics from the vehicle may undergo the alcohol and/or peroxide and carboxylic acid oxidation pathway to produce abundant less and more oxidized oxygenated OAs (LO-OOAs and MO-OOAs), and only a few primary hydrocarbon-like organic aerosols (HOAs) remain unaged. In contrast, organics from cooking may undergo the alcohol and/or peroxide oxidation pathway to produce moderate LO-OOAs, and comparable primary cooking organic aerosols (COAs) remain unaged. Our findings provide an insight into atmospheric contributions and chemical evolutions for urban-lifestyle SOAs, which could greatly influence the air quality and health risk assessments in urban areas.

Cyanobacteria can convert CO2 to chemicals such as 2,3-butanediol (2,3-BDO), rendering them promising for renewable production and carbon neutralization, but their applications are limited by low titers. To enhance cyanobacterial 2,3-BDO production, we developed a combinatorial CRISPR interference (CRISPRi) library strategy. We integrated the 2,3-BDO pathway genes and a CRISPRi library into the cyanobacterium PCC7942 using the orthogonal CRISPR system to overexpress pathway genes and attenuate genes that inhibit 2,3-BDO formation. The combinatorial CRISPRi library strategy allowed us to inhibit fbp, pdh, ppc, and sps (which catalyzes the synthesis of fructose-6-phosphate, acetyl-coenzyme A, oxaloacetate, and sucrose, respectively) at different levels, thereby allowing for rapid screening of a strain that enhances 2,3-BDO production by almost 2-fold to 1583.8 mg/L. Coupled with a statistical model, we elucidated that differentially inhibiting all the four genes enhances 2,3-BDO synthesis to varying degrees. fbp and pdh suppression exerted more profound effects on 2,3-BDO production than ppc and sps suppression, and these four genes can be repressed simultaneously without mutual interference. The CRISPRi library approach paves a new avenue to combinatorial metabolic engineering of cyanobacteria.

To investigate the composition, variation, and sources of nitrated phenols (NPs) in the winter of Beijing, gas-phase NPs were measured by a chemical ionization long time-of-flight mass spectrometer (CI-LToF-MS). A box model was applied to simulate the secondary formation process of NPs. In addition, the primary sources of NPs were resolved by a non-negative matrix factorization (NMF) model. Our results showed that secondary formation contributed 38 %, 9 %, 5 %, 17 %, and almost 100 % of the nitrophenol (NP), methyl-nitrophenol (MNP), dinitrophenol (DNP), methyl-dinitrophenol (MDNP or DNOC), and dimethyl-nitrophenol (DMNP) concentrations. The phenol–OH reaction was the predominant loss pathway (46.7 %) during the heavy pollution episode, which produced the phenoxy radical (C6H5O). The phenoxy radical consequently reacted with NO2 and produced nitrophenol. By estimating the primarily emitted phenol from the ratio of phenol/CO from freshly emitted vehicle exhaust, this study proposed that oxidation of primary phenol contributes much more nitrophenol (37 %) than that from benzene oxidation ( %) in the winter of Beijing. The latter pathway was widely used in models and might lead to great uncertainties. The source apportionment results by NMF indicated the importance of combustion sources (>50 %) to the gas-phase NPs. The industry source contributed 30 % and 9 % to DNP and MDNP, respectively, which was non-negligible. The concentration weighted trajectory (CWT) analysis demonstrated that regional transport from provinces that surround the Yellow and Bohai seas contributed more primary NPs to Beijing. Both primary sources and secondary formation at either local or regional scale should be considered when making control policies of NPs.

In the present work, we performed chassis dynamometer experiments to investigate the emissions and secondary organic aerosol (SOA) formation potential of intermediate-volatility organic compounds (IVOCs) from an on-road Chinese gasoline vehicle. High IVOC emission factors (EFs) and distinct volatility distribution were recognized. The IVOC EFs for the China V vehicle ranged from 12.1 to 226.3 mg per kilogram fuel, with a median value of 83.7 mg per kilogram fuel, which was higher than that from US vehicles. Besides, a large discrepancy in volatility distribution and chemical composition of IVOCs from Chinese gasoline vehicle exhaust was discovered, with larger contributions of B14–B16 compounds (retention time bins corresponding to C14-C16 n-alkanes) and a higher percentage of n-alkanes. Further we investigated the possible reasons that influence the IVOC EFs and volatility distribution and found that fuel type, starting mode, operating cycles and acceleration rates did have an impact on the IVOC EF. When using E10 (ethanol volume ratio of 10 %, v/v) as fuel, the IVOC EF of the tested vehicle was lower than that using commercial China standard V fuel. The average IVOC-to-THC (total hydrocarbon) ratios for gasoline-fueled and E10-fueled gasoline vehicles were 0.07±0.01 and 0.11±0.02, respectively. Cold-start operation had higher IVOC EFs than hot-start operation. The China Light-Duty Vehicle Test Cycle (CLTC) produced 70 % higher IVOCs than those from the Worldwide Harmonized Light Vehicles Test Cycle (WLTC). We found that the tested vehicle emitted more IVOCs at lower acceleration rates, which leads to high EFs under CLTC. The only factor that may influence the volatility distribution and compound composition is the engine aftertreatment system, which has compound and volatility selectivity in exhaust purification. These distinct characteristics in EFs and volatility may result in higher SOA formation potential in China. Using published yield data and a surrogate equivalent method, we estimated SOA formation under different OA (organic aerosol) loading and NOx conditions. Results showed that under low- and high-NOx conditions at different OA loadings, IVOCs contributed more than 80 % of the predicted SOA. Furthermore, we built up a parameterization method to simply estimate the vehicular SOA based on our bottom-up measurement of VOCs (volatile organic compounds) and IVOCs, which would provide another dimension of information when considering the vehicular contribution to the ambient OA. Our results indicate that vehicular IVOCs contribute significantly to SOA, implying the importance of reducing IVOCs when making air pollution controlling policies in urban areas of China.

Polycyclic aromatic hydrocarbons (PAHs) are toxic compounds in the atmosphere and have adverse effects on public health, especially through the inhalation of particulate matter (PM). At present, there is limited understanding of the size distribution of particulate-bound PAHs and their health risks on a continental scale. In this study, we carried out a PM campaign from October 2012 to September 2013 at 12 sampling sites simultaneously, including urban, suburban and remote sites in different regions of China. Size-segregated PAHs and typical tracers of coal combustion (picene), biomass burning (levoglucosan) and vehicle exhaust (hopanes) were measured. The annual averages of the 24 total measured PAHs (∑24PAHs) and benzo[a]pyrene (BaP) carcinogenic equivalent concentration (BaPeq) ranged from 7.56 to 205 ng/m3 with a mean of 53.5 ng/m3 and from 0.21 to 22.2 ng/m3 with a mean of 5.02 ng/m3, respectively. At all the sites, ∑24PAHs and BaPeq were dominant in the ultrafine particles with aerodynamic diameter < 1.1 µm, followed by those in the size ranges of 1.1–3.3 µm and > 3.3 µm. Compared with southern China, northern China witnessed much higher ∑24PAHs (87.36 vs. 17.56 ng/m3), BaPeq (8.48 vs. 1.34 ng/m3) and PAHs' inhalation cancer risk (7.4 × 10−4 vs. 1.2 × 10−4). Nationwide increases in both PAH levels and inhalation cancer risk occurred in winter. The unfavorable meteorological conditions and enhanced emissions of coal combustion and biomass burning together led to severe PAHs' pollution and high cancer risk in the atmosphere of northern China, especially during winter. Coal combustion is the major source of BaPeq in all size particles at most sampling sites. Our results suggested that the reduction of coal and biofuel consumption in the residential sector could be crucial and effective in lowering PAH concentrations and their inhalation cancer risk in China.

The concerted effort for bioproduction of higher alcohols and other commodity chemicals has yielded a consortium of metabolic engineering techniques to identify targets to enhance performance of engineered microbial strains. Here, we demonstrate the use of metabolomics as a tool to systematically identify targets for improved production phenotypes in Escherichia coli. Gas chromatography/mass spectrometry (GC/MS) and ion-pair LC-MS/MS were performed to investigate metabolic perturbations in various 1-propanol producing strains. Two initial strains were compared that differ in the expression of the citramalate and threonine pathways, which hold a synergistic relationship to maximize production yields. While this results in increased productivity, no change in titer was observed when the threonine pathway was overexpressed beyond native levels. Metabolomics revealed accumulation of upstream byproducts, norvaline and 2-aminobutyrate, both of which are derived from 2-ketobutyrate (2KB). Eliminating the competing pathway by gene knockouts or improving flux through overexpression of glycolysis gene effectively increased the intracellular 2KB pool. However, the increase in 2KB intracellular concentration yielded decreased production titers, indicating toxicity caused by 2KB and an insufficient turnover rate of 2KB to 1-propanol. Optimization of alcohol dehydrogenase YqhD activity using an ribosome binding site (RBS) library improved 1-propanol titer (g/L) and yield (g/g of glucose) by 38 and 29% in 72 h compared to the base strain, respectively. This study demonstrates the use of metabolomics as a powerful tool to aid systematic strain improvement for metabolically engineered organisms.

Satellite-based models have been widely used to simulate vegetation gross primary production (GPP) at the site, regional, or global scales in recent years. However, accurately reproducing the interannual variations in GPP remains a major challenge, and the long-term changes in GPP remain highly uncertain. In this study, we generated a long-term global GPP dataset at 0.05∘ latitude by 0.05∘ longitude and 8 d interval by revising a light use efficiency model (i.e., EC-LUE model). In the revised EC-LUE model, we integrated the regulations of several major environmental variables: atmospheric CO2 concentration, radiation components, and atmospheric vapor pressure deficit (VPD). These environmental variables showed substantial long-term changes, which could greatly impact the global vegetation productivity. Eddy covariance (EC) measurements at 95 towers from the FLUXNET2015 dataset, covering nine major ecosystem types around the globe, were used to calibrate and validate the model. In general, the revised EC-LUE model could effectively reproduce the spatial, seasonal, and annual variations in the tower-estimated GPP at most sites. The revised EC-LUE model could explain 71 % of the spatial variations in annual GPP over 95 sites. At more than 95 % of the sites, the correlation coefficients (R2) of seasonal changes between tower-estimated and model-simulated GPP are larger than 0.5. Particularly, the revised EC-LUE model improved the model performance in reproducing the interannual variations in GPP, and the averaged R2 between annual mean tower-estimated and model-simulated GPP is 0.44 over all 55 sites with observations longer than 5 years, which is significantly higher than those of the original EC-LUE model (R2=0.36) and other LUE models (R2 ranged from 0.06 to 0.30 with an average value of 0.16). At the global scale, GPP derived from light use efficiency models, machine learning models, and process-based biophysical models shows substantial differences in magnitude and interannual variations. The revised EC-LUE model quantified the mean global GPP from 1982 to 2017 as 106.2±2.9 Pg C yr−1 with the trend 0.15 Pg C yr−1. Sensitivity analysis indicated that GPP simulated by the revised EC-LUE model was sensitive to atmospheric CO2 concentration, VPD, and radiation. Over the period of 1982–2017, the CO2 fertilization effect on the global GPP (0.22±0.07 Pg C yr−1) could be partly offset by increased VPD (-0.17±0.06 Pg C yr−1). The long-term changes in the environmental variables could be well reflected in global GPP. Overall, the revised EC-LUE model is able to provide a reliable long-term estimate of global GPP. Biosynthesis of itaconic acid occurs through decarboxylation of the TCA cycle intermediate cis-aconitate. Engineering of efficient itaconate producers often requires elimination of the highly active isocitrate dehydrogenase to conserve cis-aconitate, leading to 2-ketoglutarate auxotrophy in the producing strains. Supplementation of glutamate or complex protein hydrolysate then becomes necessary, often in large quantities, to support the high cell density desired during itaconate fermentation and adds to the production cost. Here, we present an alternative approach to overcome the glutamate auxotrophy in itaconate producers by synthetically introducing the Weimberg pathway from Burkholderia xenovorans for 2-ketoglutarate biosynthesis. Because of its independence from natural carbohydrate assimilation pathways in Escherichia coli, the Weimberg pathway is able to provide 2-ketoglutarate using xylose without compromising the carbon flux toward itaconate. With xylose concentration carefully tuned to minimize excess 2-ketoglutarate flux in the stationary phase, the final strain accumulated 20 g/L of itaconate in minimal medium from 18 g/L of xylose and 45 g/L of glycerol. Necessity of the recombinant Weimberg pathway for growth also allowed us to maintain multi-copy plasmids carrying in operon the itaconate-producing genes without addition of antibiotics. Use of the Weimberg pathway for growth restoration is applicable to other production systems with disrupted 2-ketoglutarate synthesis.

Natural minerals in soil can inhibit the growth of bacteria that protect organic carbon from decay. However, the mechanism inhibiting the bacterial growth remains poorly understood. Here, using a series of cultivation experiments and biological, chemical and synchrotron-based spectral analyses, we showed that kaolinite, hematite, goethite and ferrihydrite had a significant inhibitory effect on the growth of the model bacteria Pseudomonas brassicacearum J12, which was more prominent with a concentration of 25 mg mL−1 than it was with either 10 or 5 mg mL−1. In contrast, montmorillonite promoted the growth of J12. Compared to Al-containing minerals, Fe(III)-containing minerals produced more hydroxyl radical (HO⚫) that has high efficiency for the inhibition of J12. Moreover, a significant positive correlation between HO⚫ radical and Fe(II) was found, suggesting that Fe(II) contributes to the generation of HO⚫. Furthermore, both micro X-ray fluorescence and X-ray photoelectron spectroscopies indicated that surface Fe(III) was reduced to Fe(II), which can produce HO⚫ through the well-known Fenton reaction series. Together, these findings indicate that the reduced surface Fe(II) derived from Fe(III)-containing minerals inhibits the growth of Pseudomonas brassicacearum J12 via a free-radical mechanism, which may serve as a ubiquitous mechanism between iron minerals and all of the heterotrophic bacteria in view of taxonomically and ecologically diverse heterotrophic bacteria from terrestrial environments as a vast source of superoxide.

Bowel obstructions can have a variety of causes, including impacted feces, adhesions, volvulus, non-internal hernias, and in rare cases internal hernias. We report a 63-year-old woman who presented to the emergency department with severe abdominal pain, nausea, vomiting, and obstructive symptoms that had started 12 hours earlier. A computed tomographic scan of the abdomen and pelvis showed a right internal hernia with a cecal bascule traversing through the foramen of Winslow, concerning for a closed-loop obstruction. The patient underwent an exploratory laparotomy with cecal bascule reduction and cecopexy. Given the increased mortality risk if undiagnosed, it is important to remain aware of internal hernias. Patient outcomes are markedly improved with early diagnosis and surgical intervention.

We theoretically investigate the Andreev reflection of the massive pseudospin-1 Dirac fermions including the + U -type, the − U -type, and the S _z -type mass terms, corresponding to the flat band locating at the top, the bottom, and the center of the band gap, respectively. For the ± U -type fermions, it is found that the Andreev reflection probability at the oblique incidence can be even larger than that at the normal incidence. For the retro-reflection, such an oblique enhancement occurs in the n -doped + U -type ( p -doped − U -type) massive fermion systems. While for the specular reflection, the enhancement occurs in the n -doped − U -type ( p -doped + U -type) systems. For the S _z -type massive fermions, an ideal Andreev reflection with all-angle unit efficiency is predicted in an undoped junction with the incident energy equal to the superconducting gap.

The synthesis of l-ascorbyl myristate through esterification of l-ascorbic acid and myristic acid catalyzed by lipase was studied. The effect of different parameters including lipase, molecular sieve 4 Å, organic solvent, and tetrabutyl ammonium bromide (TBAB) was evaluated. The results showed excellent yield in the presence of Novozyme435 as catalyst, 10 % (w/w of l-ascorbic acid) of 4 Å molecular sieve and 10 % (w/w of l-ascorbic acid) of TBAB as additives in 1,4-dioxane. Furthermore, response surface methodology (RSM) and 5-level-4-factor central composite design (CCD) were employed to evaluate the effects of parameters on the yield of l-ascorbyl myristate. According to the analysis result of ridge max, the optimal synthesis conditions were predicted, as follows: reaction time 27.1 h, temperature 54.1 °C, enzyme amount 25.9 % (% w/w of l-ascorbic acid), substrate molar ratio 5, and the optimal actual yield was 85.0 %.

Abstract We develop an analytical theory of the low-frequency ac quantum spin Hall (QSH) effect based upon the scattering matrix formalism. It is shown that the ac QSH effect can be interpreted as a bulk quantum pumping effect. When the electron spin is conserved, the integer-quantized ac spin Hall conductivity can be linked to the winding numbers of the reflection matrices in the electrodes, which also equal to the bulk spin Chern numbers of the QSH material. Furthermore, a possible experimental scheme by using ferromagnetic metals as electrodes is proposed to detect the topological ac spin current by electrical means.

Abstract The surface states of three-dimensional topological insulators possess the unique property of spin-momentum interlocking. This property gives rise to the interesting inverse Edelstein effect (IEE), in which an applied spin bias μ is converted to a measurable charge voltage difference V. We develop a semiclassical theory for the IEE of the surface states of Bi2Se3 thin films, which is applicable from the ballistic regime to diffusive regime. We find that the efficiency of the spin-charge conversion, defined as γ = V/μ, exhibits a universal dependence on the ratio between sample size and electron mean free path. The efficiency increases from γ = π/4 in the ballistic limit to γ = π in the diffusive limit, suggesting that sufficient strength of impurity scattering is favorable for the IEE.

The research on the mixed rare earth-iron based permanent magnet compounds by doping rare earth elements in Nd-Fe-B or Pr-Fe-B permanent magnet has been a hot topic in recent years. In this paper, La doped Pr-Fe-B ((LaxPr1-x)3Fe14B, x=0, 0.3, 0.6, (La, Pr)-Fe-B) ribbons were prepared by vacuum arc melting technique and subsequently melt-spinning method at the optimal quenching speed. The microstructure, phase distribution and magnetic properties of the nanocomposite magnets were investigated. It is found that the samples present 2:14:1 main phase, La2O3 phase and minor α-Fe phase. No excess amorphous phase is observed, indicating that 20 m/s is an optimization of preparation condition to fabricate the (La, Pr)-Fe-B ribbons in this work. The EBSD results reveal that La-rich phases distribute mostly over the grain boundary. The substitution of La for Pr in Pr-Fe-B has some influence on the coercivity, but little on the saturation magnetization. This is thought to be related with the agglomeration of rare earth-rich intermetallic phase. We hope that this work can provide new ideas in the exploration of mixed rare earth-iron based permanent magnet compounds, which possess a cost advantage and a promising potential for practical applications.

Polycrystalline MnCoGe0.99In0.01 with magnetostructural transition temperature (Tmstr) around 330 K has been prepared by arc-melting technique, and the pressure-tuned magnetostructural transition as well as the magnetocaloric effect (MCE) has been investigated. The experimental results indicate that a pressure (P) smaller than 0.53 GPa can shift Tmstr to lower temperature at a considerable rate of 119 K/GPa with the coupled nature of magnetostructural transition unchanged. However, as P reaches 0.53 GPa, the martensitic structural transition temperature (TM) further shifts to 254 K while the magnetic transition temperature of austenitic phase (TCA) occurs at around 282 K, denoting the decoupling of magnetostructural transition. Further increasing P to 0.87 GPa leads the further shift of TM to a lower temperature while the TCA keeps nearly unchanged. Therefore, the entropy change (ΔS) of the MnCoGe0.99In0.01 under different magnetic fields can be tailored by adjusting the hydrostatic pressure.

The photosynthetic cyanobacterium Synechococcus elongatus PCC7942 has recently gained great attention for its ability to directly convert CO2 into renewable chemicals upon genetic engineering. Thus, it is of great interest to increase the growth speed and lower the medium requirement for cultivating this cyanobacterium. The cultivation medium of Synechococcus elongatus PCC7942 has been developed, which consists of many inorganic and metal ingredients with a specific composition, known as the BG-11 medium. In this work, we analyzed the concentration effect of each ingredient and identified the absolutely essential components in BG-11 medium for cyanobacteria growth using the concentration gradient generator chip (CGGC) fabricated by MEMS technology. As shown by our results, removal of the individual component sodium nitrate, potassium phosphate, or magnesium sulfate from the BG-11 medium led to severe growth inhibition of Synechococcus elongatus PCC7942. Contrary to our expectation, increasing concentration of the crucial ingredients showed either insignificant or negative impact on cell growth. Overall, standard growth could be achieved without supplementation of ethylenediaminetetraacetic acid (EDTA) disodium, sodium carbonate, or sodium citrate to the culture medium. Further improvement of the CGGC-based microfluidic system based on this preliminary study may broaden its application range to analyze more complicated correlations.

The shortage of energy and water is a critical issue in various industrial sectors especially in chemical engineering industry. Using the system integrated method can greatly reduce the fresh water and public utility consumption. The integration of water network and heat exchange network can merely optimize the usage of water and energy respectively, but the optimization of heat-integrated water networks can reduce not only the water consumption but also the energy and heat exchangers. This paper introduced the regeneration of wastewater into a single contaminant heat-integrated water networks, and improved the temperature- concentration graphical method. The optimization here presented the design method for heat-integrated water networks considering the regeneration recycling of wastewater and the regeneration reuse of wastewater. Compared to the earlier method, this method reduces the consumption of fresh water from 90 kg/s to 20 kg/s, and it also reduce the minimum heating utility consumption from 4,165 kW to 1,929 kW. The improved temperature-concentration graphical method of wastewater regeneration recycling system is better than that of wastewater regeneration reuse system. Considering the regeneration of the wastewater to optimize the heat- integrated water network, the consumption of fresh water and minimum heating utility decreases greatly and the result of water-saving and energy-saving is obvious.

Shale gas is an unconventional gas existing in organic-rich shale formations. The world is in a new energy revolution due to abundant shale gas reserve and thus its conversion and utilization has been a hot research topic in the field of chemical industry. Similar to natural gas, the primary component of shale gas is methane, with a little other alkane especially ethane, carbon dioxide and nitrogen. Through separation and conversion, the stream mainly contains methane can be the substitute of natural gas while that mainly contains ethane can be applied to produce ethylene by cracking reaction. So estimating the energy consumption for the aforementioned separation and conversion is of great significance to the application of shale gas. This paper carries on the research by establishing a shale gas separation and conversion process and performing a simulation by Aspen Plus and Aspen Hysys software. The total consumption is obtained by the simulation and heuristic energy integration is considered in this paper. The data indicates that this design is an energy saving process. The result of this paper can be a reference to practical shale gas separation and conversion energy consumption in chemical industries.

IκB kinase ∊ (IKK∊, IKBKE) is a key regulator of innate immunity and a breast cancer oncogene, amplified in ∼30% of breast cancers, that promotes malignant transformation through NF-κB activation. Here, we show that IKK∊ is modified and regulated by K63-linked polyubiquitination at lysine 30 and lysine 401. Tumor necrosis factor alpha and interleukin-1β stimulation induces IKK∊ K63-linked polyubiquitination over baseline levels in both macrophages and breast cancer cell lines, and this modification is essential for IKK∊ kinase activity, IKK∊-mediated NF-κB activation, and IKK∊-induced malignant transformation. Disruption of K63-linked ubiquitination of IKK∊ does not affect its overall structure but impairs the recruitment of canonical NF-κB proteins. A cIAP1/cIAP2/TRAF2 E3 ligase complex binds to and ubiquitinates IKK∊. Altogether, these observations demonstrate that K63-linked polyubiquitination regulates IKK∊ activity in both inflammatory and oncogenic contexts and suggests an alternative approach to targeting this breast cancer oncogene.

Background/aims: The optimal intervention for Boerhaave perforation has not been determined. Options include surgical repair with/without a pedicled muscle flap, T tube placement, esophageal resection or diversion, or an endoscopic approach. All management strategies require adequate drainage and nutritional support. Our aim was to evaluate outcomes following Boerhaave perforation treated with surgery, endoscopic therapy, or both. Patients and methods: We performed a 10-year review of our prospectively maintained databases of adult patients with Boerhaave perforations. We documented clinical presentation, extent of injury, primary intervention, “salvage” treatment (any treatment for persistent leak), and outcome. Results were analyzed using the Fisher’s exact and Kruskal – Wallis tests. Results: Between October 2004 and October 2014, 235 patients presented with esophageal leak/fistula with 17 Boerhaave perforations. Median age was 68 years. Median length of perforation was 1.25 cm (range 0.8 – 5 cm). Four patients presented with systemic sepsis (two treated with palliative stent and two surgically). Primary endotherapy was performed for eight (50 %) and primary surgery for eight (50 %) patients. Two endotherapy patients required multiple stents. Median stent duration was 61 days (range 56 – 76). “Salvage” intervention was required in 2/8 (25 %) endotherapy patients and 1/8 (13 %) surgery patient (stent). All patients healed without resection/reconstruction. There were no deaths in the surgically treated group and two in the endotherapy group (stented with palliative intent due to poor systemic condition). Readmission within 30 days occurred in 3/6 of alive endotherapy patients (50 %) and 0/8 surgery patients. Re-intervention within 30 days was required for one endotherapy patient. Conclusion: Endoscopic repair of Boerhaave perforations can be useful in carefully selected patients without evidence of systemic sepsis. Endoscopic therapy such as stenting is particularly valuable as a “salvage” intervention. The benefits of endoscopic therapy and esophageal preservation are offset against an increased risk of readmission in patients primarily treated endoscopically.

The MYLK gene encodes the multifunctional enzyme, myosin light chain kinase (MLCK), involved in isoform-specific non-muscle and smooth muscle contraction and regulation of vascular permeability during inflammation. Three MYLK SNPs (P21H, S147P, V261A) alter the N-terminal amino acid sequence of the non-muscle isoform of MLCK (nmMLCK) and are highly associated with susceptibility to acute lung injury (ALI) and asthma, especially in individuals of African descent. To understand the functional effects of SNP associations, we examined the N-terminal segments of nmMLCK by 1H-15N heteronuclear single quantum correlation (HSQC) spectroscopy, a 2-D NMR technique, and by in silico molecular modeling. Both NMR analysis and molecular modeling indicated SNP localization to loops that connect the immunoglobulin-like domains of nmMLCK, consistent with minimal structural changes evoked by these SNPs. Molecular modeling analysis identified protein-protein interaction motifs adversely affected by these MYLK SNPs including binding by the scaffold protein 14-3-3, results confirmed by immunoprecipitation and western blot studies. These structure-function studies suggest novel mechanisms for nmMLCK regulation, which may confirm MYLK as a candidate gene in inflammatory lung disease and advance knowledge of the genetic underpinning of lung-related health disparities.

We propose an Andreev interferometer, based on a branched Y-junction, to detect the finite momentum pairing in Fulde–Ferrell (FF) superconductors. In this interferometer, the oscillation of subgap conductance is a unique function of phase difference between the two channels of the Y-junction, which is determined by the phase modulation of the order parameter in the FF superconductors. This interferometer has the potential not only to determine the magnitude but also the direction of the momentum of Cooper pairs in the FF superconductor. The possible applications of the interferometer in the identification of the finite momentum pairing in non-centrosymmetric superconductors are also discussed.

Topological phase transitions in a three-dimensional (3D) topological insulator (TI) with an exchange field of strength g are studied by calculating spin Chern numbers C ^± ( k _z ) with momentum k _z as a parameter. When | g | exceeds a critical value g _C , a transition of the 3D TI into a Weyl semimetal occurs, where two Weyl points appear as critical points separating k _z regions with different first Chern numbers.
