1. Optimizing reaction and transport fluxes in temperature gradient-driven chemical reaction-diffusion systems

Abstract: Temperature gradients represent energy sources that can be harvested to generate steady reaction or transport fluxes. Technological developments could lead to the transfer of free energy from heat sources and sinks to chemical systems for the purpose of extraction, thermal batteries, or nonequilibrium synthesis. We present a theoretical study of 1D chemical systems subjected to temperature gradients. A complete theoretical framework describes the behavior of the system induced by various temperature profiles. An exact mathematical derivation was established for a simple two-compartment model and was generalized to arbitrary reaction-diffusion systems based on numerical models. An experimental system was eventually scaled and tuned to optimize either nonequilibrium chemical transport or reaction. The relevant parameters for this description were identified; they focused on the system symmetry for chemical reaction and transport. Nonequilibrium thermodynamic approaches lead to a description analogous to electric circuits. Temperature gradients lead to the onset of a steady chemical force, which maintains steady reaction-diffusion fluxes moderated by chemical resistance. The system activity was then assessed using the entropy production rate as a measure of its dissipated power. The chemical characteristics of the system can be tuned for general optimization of the nonequilibrium state or for the specific optimization of either transport or reaction processes. The shape of the temperature gradient can be tailored to precisely control the spatial localization of active processes, targeting either precise spatial localization or propagation over large areas. The resulting temperature-driven chemical system can in turn be used to drive secondary processes into either nonequilibrium reaction fluxes or concentration gradients., Comment: Maint text: 17 pages, 5 figures, 2 tables. Abstract: PLATO (PLAnetary Transits and Oscillations of stars) is ESA's M3 mission designed to detect and characterise extrasolar planets and perform asteroseismic monitoring of a large number of stars. PLATO will detect small planets (down to <2 R_(Earth)) around bright stars (<11 mag), including terrestrial planets in the habitable zone of solar-like stars. With the complement of radial velocity observations from the ground, planets will be characterised for their radius, mass, and age with high accuracy (5 %, 10 %, 10 % for an Earth-Sun combination respectively). PLATO will provide us with a large-scale catalogue of well-characterised small planets up to intermediate orbital periods, relevant for a meaningful comparison to planet formation theories and to better understand planet evolution. It will make possible comparative exoplanetology to place our Solar System planets in a broader context. In parallel, PLATO will study (host) stars using asteroseismology, allowing us to determine the stellar properties with high accuracy, substantially enhancing our knowledge of stellar structure and evolution. The payload instrument consists of 26 cameras with 12cm aperture each. For at least four years, the mission will perform high-precision photometric measurements. Here we review the science objectives, present PLATO's target samples and fields, provide an overview of expected core science performance as well as a description of the instrument and the mission profile at the beginning of the serial production of the flight cameras. PLATO is scheduled for a launch date end 2026. This overview therefore provides a summary of the mission to the community in preparation of the upcoming operational phases.

Abstract: There is a growing demand for molecules of natural origin for biocontrol and biostimulation, given the current trend away from synthetic chemical products. Leachates extracted from plantain stems were obtained after biodegradation of the plant material. To characterize the leachate, quantitative determinations of nitrogen, carbon, phosphorus, and cations (K+, Ca2+, Mg2+, Na+), Q2/4, Q2/6, and Q4/6 absorbance ratios, and metabolomic analysis were carried out. The potential role of plantain leachates as fungicide, elicitor of plant defense, and/or plant biostimulant was evaluated by agar well diffusion method, phenotypic, molecular, and imaging approaches. The plant extracts induced a slight inhibition of fungal growth of an aggressive strain of Colletotrichum gloeosporioides, which causes anthracnose. Organic compounds such as cinnamic, ellagic, quinic, and fulvic acids and indole alkaloid such as ellipticine, along with some minerals such as potassium, calcium, and phosphorus, may be responsible for the inhibition of fungal growth. In addition, jasmonic, benzoic, and salicylic acids, which are known to play a role in plant defense and as biostimulants in tomato, were detected in leachate extract. Indeed, foliar application of banana leachate induced overexpression of LOXD, PPOD, and Worky70-80 genes, which are involved in phenylpropanoid metabolism, jasmonic acid biosynthesis, and salicylic acid metabolism, respectively. Leachate also activated root growth in tomato seedlings. However, the main impact of the leachate was observed on mature plants, where it caused a reduction in leaf area and fresh weight, the remodeling of stem cell wall glycopolymers, and an increase in the expression of proline dehydrogenase.

These β-D-glucans differ from each other by their branching rate, which is possibly responsible for their biological activities. In the present study, we characterized a laminaran fraction extracted from Laminaria hyperborea and named LAM2 using sugar composition and structural analyses (NMR). Then, we evaluated its activity as a potential plant elicitor in vitro on tomato seedlings using gene expression analysis and cell wall immunofluorescence labeling. Our study showed that LAM2 isolated from L. hyperborea is a succinylated laminaran which significantly enhanced the plant defense of tomato seedlings and induced cell wall modifications, suggesting a higher elicitor activity than the laminaran standard extracted from Laminaria digitata.

Abstract: The root extracellular trap (RET) consists of root-associated, cap-derived cells (root AC-DCs) and their mucilaginous secretions, and forms a structure around the root tip that protects against biotic and abiotic stresses. However, there is little information concerning the changes undergone by the RET during droughts, especially for tree species. Morphological and immunocytochemical approaches were used to study the RET of black poplar (Populus nigra L.) seedlings grown in vitro under optimal conditions (on agar-gelled medium) or when polyethylene glycol-mediated (PEG6000—infused agar-gelled medium) was used to mimic drought conditions through osmotic stress. Under optimal conditions, the root cap released three populations of individual AC-DC morphotypes, with a very low proportion of spherical morphotypes, and equivalent proportions of intermediate and elongated morphotypes. Immunolabeling experiments using anti-glycan antibodies specific to cell wall polysaccharide and arabinogalactan protein (AGP) epitopes revealed the presence of homogalacturonan (HG), galactan chains of rhamnogalacturonan-I (RG-I), and AGPs in root AC-DC cell walls. The data also showed the presence of xylogalacturonan (XGA), xylan, AGPs, and low levels of arabinans in the mucilage. The findings also showed that under osmotic stress conditions, both the number of AC-DCs (spherical and intermediate morphotypes) and the total quantity of mucilage per root tip increased, whereas the mucilage was devoid of the epitopes associated with the polysaccharides RG-I, XGA, xylan, and AGPs. Osmotic stress also led to reduced root growth and increased root expression of the P5CS2 gene, which is involved in proline biosynthesis and cellular osmolarity maintenance (or preservation) in aerial parts. Together, our findings show that the RET is a dynamic structure that undergoes pronounced structural and molecular remodeling, which might contribute to the survival of the root tip under osmotic conditions.

Recent works in this field introduce the concept of recycling, rather than the traditional open-flow system described by Frank. This approach has been criticized by Blackmond et al. They claimed that such systems are thermodynamically impossible, except in the cases where non-microreversible reactions are introduced, like in photochemical reactions, or under the influence of physical actions (e.g. by crystal crushing). This point of view reveals misunderstandings about this model of a recycled system, overlooks the possibility of energy exchanges that could take place in prebiotic systems, and leads the authors to unawarely remove the activation reaction and energy source from their "non-equilibrium" models. The classical approaches through the computation of the right null space leads to the decomposition of the network into complex ``cycles'' of reactions concerned with all metabolites. We show how, departing from the left null space computation, the flux analysis can be decoupled into linear fluxes and single loops, allowing a more refine qualitative analysis as a function of the antagonisms and connections among these local fluxes. While the roles of polysaccharides and glycoproteins secreted by root AC-DC have started to be elucidated, how the low-molecular-weight exudates of the RET contribute to root defense is poorly known. In order to better understand the RET and its defense response, the transcriptomes, proteomes and metabolomes of roots, root AC-DC and mucilage of soybean (Glycine max (L.) Merr, var. Castetis) upon elicitation with the peptide PEP-13 were investigated. This peptide is derived from the pathogenic oomycete Phytophthora sojae. In this study, the root and the RET responses to elicitation were dissected and sequenced using transcriptional, proteomic and metabolomic approaches. The major finding is increased synthesis and secretion of specialized metabolites upon induced defense activation following PEP-13 peptide elicitation. This study provides novel findings related to the pivotal role of the root extracellular trap in root defense.

Abstract: This study presents a novel three-dimensional (3D) tool “3D in vitro choice” for chemotaxis assays with cyst nematodes. The original 3D in vitro choice was customized through digital printing. Freshly hatched second stage juveniles (J2s) of the cyst nematode Globodera pallida were used as the nematode model to illustrate chemo-orientation behavior in the 3D system. The efficiency and reliability of the 3D in vitro choice were validated with 2% Phytagel as navigation medium, in three biological assays and using tomato root exudates or potato root border cells and their associated mucilage as a positive attractant as compared with water. For each biological assay, J2s were hatched from the same population of a single generation glasshouse-cultured cysts. This novel easy to use and low-cost 3d-device could be a useful replacement to Petri dishes assays in nematode behavioral studies due to the ease of deposition of nematodes and test substances, coupled with its distinctive zones that allow for precision in choice making by the nematodes.

Abstract: Although Phaeodactylum tricornutum is gaining importance in plant molecular farming for the production of high-value molecules such as monoclonal antibodies, little is currently known about key cell metabolism occurring in this diatom such as protein glycosylation. For example, incorporation of fucose residues in the glycans N-linked to protein in P. tricornutum is questionable. Indeed, such epitope has previously been found on N-glycans of endogenous glycoproteins in P. tricornutum. Meanwhile, the potential immunogenicity of the α(1,3)-fucose epitope present on plant-derived biopharmaceuticals is still a matter of debate. In this paper, we have studied molecular actors potentially involved in the fucosylation of the glycoproteins in P. tricornutum. Based on sequence similarities, we have identified a putative P. tricornutum GDP-L-fucose transporter and three fucosyltransferase (FuT) candidates. The putative P. tricornutum GDP-L-fucose transporter coding sequence was expressed in the Chinese Hamster Ovary (CHO)-gmt5 mutant lacking its endogenous GDP-L-fucose transporter activity. We show that the P. tricornutum transporter is able to rescue the fucosylation of proteins in this CHO-gmt5 mutant cell line, thus demonstrating the functional activity of the diatom transporter and its appropriate Golgi localization. In addition, we overexpressed one of the three FuT candidates, namely the FuT54599, in P. tricornutum and investigated its localization within Golgi stacks of the diatom. Our findings show that overexpression of the FuT54599 leads to a significant increase of the α(1,3)-fucosylation of the diatom endogenous glycoproteins.

In the second part of this study, we investigated if exDNA release was modified during an elicitation with PEP-13 (a peptide elicitor from oomycete genus Phytophthora). Abstract: Abstract Living organisms perform and control complex behaviours by using webs of chemical reactions organized in precise networks. This powerful system concept, which is at the very core of biology, has recently become a new foundation for bioengineering. Remarkably, however, it is still extremely difficult to rationally create such network architectures in artificial, non‐living and well‐controlled settings. We introduce here a method for such a purpose, on the basis of standard DNA biochemistry. This approach is demonstrated by assembling de novo an efficient chemical oscillator: we encode the wiring of the corresponding network in the sequence of small DNA templates and obtain the predicted dynamics. Our results show that the rational cascading of standard elements opens the possibility to implement complex behaviours in vitro. Because of the simple and well‐controlled environment, the corresponding chemical network is easily amenable to quantitative mathematical analysis. These synthetic systems may thus accelerate our understanding of the underlying principles of biological dynamic modules.

Abstract: International audience; The Radio and Plasma Waves (RPW) instrument on the ESA Solar Orbiter mission is designed to measure in situ magnetic and electric fields and waves from the continuum up to several hundred kHz. The RPW also observes solar and heliospheric radio emissions up to 16 MHz. It was switched on and its antennae were successfully deployed two days after the launch of Solar Orbiter on February 10, 2020. Since then, the instrument has acquired enough data to make it possible to assess its performance and the electromagnetic disturbances it experiences. In this article, we assess its scientific performance and present the first RPW observations. In particular, we focus on a statistical analysis of the first observations of interplanetary dust by the instrument’s Thermal Noise Receiver. We also review the electro-magnetic disturbances that RPW suffers, especially those which potential users of the instrument data should be aware of before starting their research work.

Abstract: International audience; Solar Orbiter's in situ coordination working group met frequently during the development of the mission with the goal of ensuring that its in situ payload has the necessary level of coordination to maximise science return. Here we present the results of that work, namely how the design of each of the in situ instruments (EPD, MAG, RPW, SWA) was guided by the need for coordination, the importance of time synchronisation, and how science operations will be conducted in a coordinated way. We discuss the mechanisms by which instrument sampling schemes are aligned such that complementary measurements will be made simultaneously by different instruments, and how burst modes are scheduled to allow a maximum overlap of burst intervals between the four instruments (telemetry constraints mean different instruments can spend different amounts of time in burst mode). We also explain how onboard autonomy, inter-instrument communication, and selective data downlink will be used to maximise the number of transient events that will be studied using high-resolution modes of all the instruments. Finally, we briefly address coordination between Solar Orbiter's in situ payload and other missions.

Abstract: The Radio and Plasma Waves (RPW) instrument on the ESA Solar Orbiter mission is described in this paper. This instrument is designed to measure in-situ magnetic and electric fields and waves from the continuous to a few hundreds of kHz. RPW will also observe solar radio emissions up to 16 MHz. The RPW instrument is of primary importance to the Solar Orbiter mission and science requirements since it is essential to answer three of the four mission overarching science objectives. In addition RPW will exchange on-board data with the other in-situ instruments in order to process algorithms for interplanetary shocks and type III langmuir waves detections. Abstract: Abstract Background In eukaryotic cells, the membrane compartments that constitute the exocytic pathway are traversed by a constant flow of lipids and proteins. This is particularly true for the endoplasmic reticulum (ER), the main "gateway of the secretory pathway", where biosynthesis of sterols, lipids, membrane-bound and soluble proteins, and glycoproteins occurs. Maintenance of the resident proteins in this compartment implies they have to be distinguished from the secretory cargo. To this end, they must possess specific ER localization determinants to prevent their exit from the ER, and/or to interact with receptors responsible for their retrieval from the Golgi apparatus. Very few information is available about the signal(s) involved in the retention of membrane type II protein in the ER but it is generally accepted that sorting of ER type II cargo membrane proteins depends on motifs mainly located in their cytosolic tails. Results Here, using Arabidopsis glucosidase I as a model, we have identified two types of signals sufficient for the location of a type II membrane protein in the ER. A first signal is located in the luminal domain, while a second signal corresponds to a short amino acid sequence located in the cytosolic tail of the membrane protein. The cytosolic tail contains at its N-terminal end four arginine residues constitutive of three di-arginine motifs (RR, RXR or RXXR) independently sufficient to confer ER localization. Interestingly, when only one di-arginine motif is present, fusion proteins are located both in the ER and in mobile punctate structures, distinct but close to Golgi bodies. Soluble and membrane ER protein markers are excluded from these punctate structures, which also do not colocalize with an ER-exit-site marker. It is hypothesized they correspond to sites involved in Golgi to ER retrotransport. Conclusion Altogether, these results clearly show that cytosolic and luminal signals responsible for ER retention could coexist in a same type II membrane protein. These data also suggest that both retrieval and retention mechanisms govern protein residency in the ER membrane. We hypothesized that mobile punctate structures not yet described at the ER/Golgi interface and tentatively named GERES, could be involved in retrieval mechanisms from the Golgi to the ER.

Abstract: A pre-restoration diagnosis revealed a high amount of pyrite in the wood of the Lyon Saint-Georges 4 shipwreck (end of the second century). The occurrence of this phase is supposed to result from the microbiologically influenced corrosion of the iron fasteners. So, all the nails and metallic elements were removed from the remains before treatment and the wreck was consolidated by polyethylene glycol impregnation coupled to a specific desalination process. Treated and non-treated samples extracted from the wreck were studied in order to identify the iron/sulfur-containing compounds present in the wood before and after treatment and figure out its effect. Sample analyses relied on an original approach combining magnetic characterization methods and more common elemental and structural analysis methods. The results showed that the treatment was effective in removing soluble salts. However, a large amount of unstable iron sulfides remained inside the wood. [ABSTRACT FROM AUTHOR]

Abstract: PLATO 2.0 has been selected by ESA as the third medium-class Mission (M3) of the Cosmic Vision Program. Its Payload is conceived for the discovery of new transiting exoplanets on the disk of their parent stars and for the study of planetary system formation and evolution as well as to answer fundamental questions concerning the existence of other planetary systems like our own, including the presence of potentially habitable new worlds. The PLATO Payload design is based on the adoption of four sets of short focal length telescopes having a large field of view in order to exploit a large sky coverage and to reach, at the same time, the needed photometry accuracy and signalto- noise ratio (S/N) within a few tens of seconds of exposure time. The large amount of data produced by the telescope is collected and processed by means of the Payload’s Data Processing System (DPS) composed by many processing electronics units. This paper gives an overview of the PLATO 2.0 DPS, mainly focusing on the architecture and processing capabilities of its Instrument Control Unit (ICU), the electronic subsystem acting as the main interface between the Payload (P/L) and the Spacecraft (S/C).

Abstract: CoRoT is a space telescope dedicated to stellar seismology and the search for extrasolar planets. The mission is led by CNES in association with French laboratories and has a large international participation: the European Space Agency (ESA), Austria, Belgium and Germany contribute to the payload, and Spain and Brazil contribute to the ground segment. Development of the spacecraft, which is based on a PROTEUS low earth orbit recurrent platform, commenced in October 2000 and the satellite was launched on December 27th 2006. The instrument and platform characteristics prior to launch have been described in ESA publication (SP-1306) . In the present paper we detail the behaviour in flight, based on raw and corrected data. Five runs have been completed since January 2007. The data used here are essentially those acquired during the commissioning phase and from a long run which lasted 146 days, these enable us to give a complete overview of the instrument and platform behaviour for all environmental conditions. The ground based data processing is not described in detail, the most important method being published elsewhere. It is shown that the performance specifications are easily satisfied when the environmental conditions are favourable. Most of the perturbations, and consequently data corrections, are related to Low Earth Orbit (LEO) perturbations: high energy particles inside the South Atlantic Anomaly (SAA), eclipses and temperature variations, and line of sight fluctuations due to the attitude control system. Abstract: Aims. We report the discovery of very shallow ($\Delta F/F \approx 3.4\times 10^{-4}$), periodic dips in the light curve of an active $V = 11.7$G9V star observed by the CoRoT satellite, which we interpret as caused by a transiting companion. We describe the 3-colour CoRoT data and complementary ground-based observations that support the planetary nature of the companion.

45. 2‐Hydroxyazobenzenes to Tailor pH Pulses and Oscillations with Light

Abstract: This paper evaluates the 2‐hydroxyazobenzene platform for tailoring proton concentration pulses and oscillations with monochromatic light. The easily prepared 2‐hydroxyazobenzenes exhibit large absorptions in the near‐UV range. Photoisomerization was investigated by UV/Vis absorption, 1H NMR spectroscopy, and steady‐state fluorescence emission. In the whole investigated series, the transstereoisomer of the 2‐hydroxyazobenzene motif provides the corresponding cisderivative with an action cross section in the 103M−1cm−1range. At the same time, photoisomerization is accompanied by a significant pKdrop of the phenol group. According to the phenyl‐substituent pattern, cis‐to‐transthermal back‐isomerization can be tuned in the 10 ms–100 s range. Up to 2 units of reversible pH drops or pH oscillations on the 10 s timescale have been obtained by appropriately tailoring single‐wavelength illumination of 2‐hydroxyazobenzene solutions.

46. Emergence of homochirality in far‐from‐equilibrium systems: Mechanisms and role in prebiotic chemistry

Abstract: Since the model proposed by Frank (Frank FC, Biochem Biophys Acta 1953;11:459–463), several alternative models have been developed to explain how an asymmetric non‐racemic steady state can be reached by a chirally symmetric chemical reactive system. This paper explains how a stable non‐racemic regime can be obtained as a symmetry breaking occurring in a far‐from‐equilibrium reactive system initiated with an initial imbalance. Departing from the variations around the original Frank's model that are commonly described in the literature, i.e. open‐flow systems of direct autocatalytic reactions, we discuss recent developments emphasizing both an active recycling of components and an autocatalytic network of simple reactions. We will present our APED model as the most natural realization of such thermodynamic openness and non‐equilibrium, of recycling and of network autocatalysis, each of these in prebiotic conditions. The different experimental and theoretical models in the literature will be classified according to mechanism. 47. Prebiotic synthesis of sequential peptides on the Hadean beach by a molecular engine working with nitrogen oxides as energy sources

Abstract: Addressing the still open question of the prebiotic origin of sequential macromolecules (peptides, nucleic acids) on the primitive Earth, we describe a molecular engine (the primary pump), which works at ambient temperature and continuously generates, elongates and complexifies sequential peptides. This new scenario is based on a cyclic reaction sequence, whose keystep is the activation of amino acids into their N‐carboxyanhydrides (NCA) through nitrosation by NOx. This process could have taken place on tidal beaches; it requires a buffered ocean, emerged land and a nitrosating atmosphere. With the help of geochemical studies and computer simulations of atmosphere photochemistry, we show that the primitive Earth during the Hadean may have satisfied all these requirements.© 2001 Society of Chemical Industry.
