Aims: This study aims to describe a new method that may be used as a supplement to evaluate humeral rotational alignment during intramedullary nail (IMN) insertion using the profile of the perpendicular peak of the greater tuberosity and its relation to the transepicondylar axis. We called this angle the greater tuberosity version angle (GTVA). Methods: This study analyzed 506 cadaveric humeri of adult patients. All humeri were CT scanned using 0.625 × 0.625 × 0.625 mm cubic voxels. The images acquired were used to generate 3D surface models of the humerus. Next, 3D landmarks were automatically calculated on each 3D bone using custom-written C++ software. The anatomical landmarks analyzed were the transepicondylar axis, the humerus anatomical axis, and the peak of the perpendicular axis of the greater tuberosity. Lastly, the angle between the transepicondylar axis and the greater tuberosity axis was calculated and defined as the GTVA. Results: The value of GTVA was 20.9° (SD 4.7°) (95% CI 20.47° to 21.3°). Results of analysis of variance revealed that females had a statistically significant larger angle of 21.95° (SD 4.49°) compared to males, which were found to be 20.49° (SD 4.8°) (p = 0.001). Conclusion: This study identified a consistent relationship between palpable anatomical landmarks, enhancing IMN accuracy by utilizing 3D CT scans and replicating a 20.9° angle from the greater tuberosity to the transepicondylar axis. Using this angle as a secondary reference may help mitigate the complications associated with malrotation of the humerus following IMN. However, future trials are needed for clinical validation. Abstract Background Plant-parasitic root-knot nematode (Meloidogyne incognita) causes global yield loss in agri- and horticultural crops. Nematode management options rely on chemical method. However, only a handful of nematicides are commercially available. Resistance breeding efforts are not sustainable because R gene sources are limited and nematodes have developed resistance-breaking populations against the commercially available Mi-1.2 gene-expressing tomatoes. RNAi crops that manage nematode infection are yet to be commercialized because of the regulatory hurdles associated with transgenic crops. The deployment of the CRISPR/Cas9 system to improve nematode tolerance (by knocking out the susceptibility factors) in plants has emerged as a feasible alternative lately. Results In the present study, a M. incognita-responsive susceptibility (S) gene, amino acid permease (AAP6), was characterized from the model plant Arabidodpsis thaliana by generating the AtAAP6 overexpression line, followed by performing the GUS reporter assay by fusing the promoter of AtAAP6 with the β-glucuronidase (GUS) gene. Upon challenge inoculation with M. incognita, overexpression lines supported greater nematode multiplication, and AtAAP6 expression was inducible to the early stage of nematode infection. Next, using CRISPR/Cas9, AtAAP6 was selectively knocked out without incurring any growth penalty in the host plant. The ‘Cas9-free’ homozygous T3 line was challenge inoculated with M. incognita, and CRISPR-edited A. thaliana plants exhibited considerably reduced susceptibility to nematode infection compared to the non-edited plants. Additionally, host defense response genes were unaltered between edited and non-edited plants, implicating the direct role of AtAAP6 towards nematode susceptibility. Conclusion The present findings enrich the existing literature on CRISPR/Cas9 research in plant-nematode interactions, which is quite limited currently while compared with the other plant-pathogen interaction systems.

The taxonomy of king cobras (Ophiophagus) was reevaluated using qualitative, mensural and meristic characters, based on 148 entire and five skeletal specimens, and supported by a recent molecular phylogenetic analysis. We provide nomenclatural synopses of both the genus and species-series nomina. We restrict the concept of Ophiophagus hannah s. str. to populations from eastern Pakistan, northern and eastern India, the Andaman Islands, Indo-Burma and Indo-China, south to central Thailand. The nomen Ophiophagus bungarus (Schlegel, 1837) comb. nov. is revived for the populations inhabiting the Sunda Shelf area, including the Malay Peninsula, the Greater Sunda Islands and parts of the southern Philippines. We describe two new species, Ophiophagus kaalinga Gowri Shankar, Das & Ganesh sp. nov. endemic to the Western Ghats of south-western India and Ophiophagus salvatana Gowri Shankar, Das & Wüster sp. nov., inhabiting the island of Luzon in northern Philippines. For the purpose of nomenclatural stability, we designate a neotype for Hamadryas hannah Cantor, 1836. A dichotomous identification key to the four species recognized here is provided.

Multidrug-resistant yeast Candida auris is a serious threat to public health with documented survival in various hospital niches. The dynamics of this survival benefit and its trade off with drug resistance are still unknown for this pathogen. In this study we investigate the oxidative stress response (OSR) in fluconazole-resistant C. auris and compare its relative fitness with fluconazole-susceptible strains. A total of 351 C. auris clinical isolates (61 fluconazole-susceptible and 290 fluconazole-resistant) were screened for stress tolerance by spot assay and 95.08 % fluconazole-susceptible isolates were hyper-resistant to oxidative stress while majority (94.5 %) fluconazole-resistant isolates had lower oxidative tolerance. Expression of Hog1 and Cta1 gene transcript levels and cellular catalase levels were significantly higher in fluconazole-susceptible isolates and a corresponding higher intracellular reactive oxygen species level (iROS) was accumulated in the fluconazole-resistant isolates. Biofilm formation and cell viability under oxidative stress revealed higher biofilm formation and better viability in fluconazole-susceptible isolates. Fluconazole-resistant isolates had higher basal cell wall chitin. On comparison of virulence, the % cytotoxicity in A549 cell line was higher in fluconazole-susceptible isolates and the median survival of the infected larvae in G. mellonella infection model was higher in fluconazole-resistant (5; IQR:4.5–5 days) vs. fluconazole-susceptible C. auris (2; IQR:1.5–2.5 days). All organisms evolve with changes in their environmental conditions, to ensure an optimal balance between proliferation and survival. Development of tolerance to a certain kind of stress example antifungal exposure in yeast can leads to a compensatory decrease in tolerance for other stresses. This study provides useful insights into the comparative fitness and antifungal susceptibility trade off in C. auris. We report a negative association between H2O2 tolerance and fluconazole susceptibility. Using in-vitro cell cytotoxicity and in-vivo survival assays we also demonstrate the higher virulence potential of fluconazole-susceptible C. auris isolates corroborating the negative correlation between susceptibility and pathogen survival or virulence. These findings could also be translated to clinical practice by investigating the possibility of using molecules targeting stress response and fitness regulating pathways for management of this serious infection.

Introduction: The main goal of the root-end filling material is to create a hermetic seal to protect against microbes and their by-products. Excellent biocompatibility and sealing ability are characteristics of MTA developed by Torabinejad et al. This study aimed to compare the sealing ability of different type MTA as root-end filling material using dye penetration technique. Material and Method: One-twenty (N = 120) extracted human maxillary anterior teeth were decontaminated, cleaned, and decoronated. Endodontic treatment and root-end resection were done. Then root-end cavity was prepared and filled with tested materials (N = 30). A calibrated stereomicroscope was used to evaluate linear measurement. All data were tabulated and statistically analyzed with a level of significance set at P < .05. Result: This order of increasing microleaks was observed: MTA Angelus < MTA Plus < PRO-Root MTA < Control group. There was a statistically significant difference in mean microleakage in MTA Angelus and MTA Plus groups (P = 0.040). MTA Angelus shows the least microleakage among all the bioceramic material groups. Conclusion: Although the sealing ability of MTA Angelus is superior to MTA Plus, PRO-Root MTA. MTA Plus, PRO-Root MTA could be considered an acceptable alternative to MTA Angelus in peri-radicular surgeries.

Abstract Background Root-knot nematode Meloidogyne graminicola has emerged as a major threat in rice agroecosystems owing to climate change-induced changes in cultivation practices. Synthetic nematicides are continually being withdrawn from the nematode management toolbox because of their ill effects on the environment. A sustainable strategy would be to develop novel nematicides or resistant plants that would target nematode sensory perception, which is a key step in the host finding biology of plant-parasitic nematodes (PPNs). However, compared to the extensive literature on the free-living nematode Caenorhabditis elegans, negligible research has been performed on PPN chemosensory biology. Results The present study characterizes the five chemosensory genes (Mg-odr-7, Mg-tax-4, Mg-tax-4.1, Mg-osm-9, and Mg-ocr-2) from M. graminicola that are putatively associated with nematode host-finding biology. All the genes were highly transcribed in the early life stages, and RNA interference (RNAi)-induced downregulation of each candidate gene perturbed the normal behavioural phenotypes of M. graminicola, as determined by examining the tracking pattern of juveniles on Pluronic gel medium, attraction to and penetration in rice root tip, and developmental progression in rice root. In addition, a detrimental effect on nematode chemotaxis towards different volatile and nonvolatile organic compounds and host root exudates was documented. Conclusion Our findings enrich the existing literature on PPN chemosensory biology and can supplement future research aimed at identifying a comprehensive chemosensory signal transduction pathway in PPNs.

In the present work, we reported the synthesis and characterization [single crystal X-ray diffraction technique, spectroscopic, etc.] of two new Ni(II) and Zn(II) coordination compounds, viz. [Ni(2,6-PDC)2]2[Ni(en)2(H2O)2]2[Ni(en)(H2O)4]·4H2O (1) and [Zn(2,6-PDC)(Hdmpz)2] (2) (where 2,6-PDC = 2,6-pyridinedicarboxylate, en = ethylene-1,2-diamine, and Hdmpz = 3,5-dimethyl pyrazole). Compound 1 is found to crystallize as a multicomponent Ni(II) compound with five discrete complex moieties, whereas compound 2 is isolated as a mononuclear Zn(II) compound. A deep analysis of the crystal structure of 1 unfolds unusual dual enclathration of guest complex cationic moieties within the supramolecular host cavity stabilized by anion–π, π-stacking, N–H⋯O, C–H⋯O, and O–H⋯O hydrogen bonding interactions. Again, the crystal structure of compound 2 is stabilized by the presence of unconventional C–H⋯π(chelate ring) interactions along with C–H⋯O, C–H⋯N hydrogen bonding, π-stacking, and C–H⋯π(pyridyl) interactions. These non-covalent interactions were further studied theoretically using density functional theory (DFT) calculations, molecular electrostatic potential (MEP) surfaces, non-covalent interaction (NCI) plot index, and quantum theory of atoms in molecules (QTAIM) computational tools. The computational study displays that π-stacking or H bonds greatly tune the directionality of compound 1, although non-directional electrostatic forces dominate energetically. For compound 2, a combined QTAIM/NCI plot analysis confirms the presence of unconventional C–H⋯π(chelate ring) interactions along with other weak interactions obtained from the crystal structure analysis. Further, the individual energy contributions of these weak yet significant non-covalent interactions have also been determined computationally.

Chronic wounds are a major health problem because of delayed healing, causing hardships for the patient. The infection present in these wounds plays a role in delayed wound healing. Silver wound dressings have been used for decades, beginning in the 1960s with silver sulfadiazine for infection prevention for burn wounds. Since that time, there has been a large number of commercial silver dressings that have obtained FDA clearance. In this review, we examine the literature involving in vitro and in vivo (both animal and human clinical) studies with commercial silver dressings and attempt to glean the important characteristics of these dressings in treating infected wounds. The primary presentation of the literature is in the form of detailed tables. The narrative part of the review focuses on the different types of silver dressings, including the supporting matrix, the release characteristics of the silver into the surroundings, and their toxicity. Though there are many clinical studies of chronic and burn wounds using silver dressings that we discuss, it is difficult to compare the performances of the dressings directly because of the differences in the study protocols. We conclude that silver dressings can assist in wound healing, although it is difficult to provide general treatment guidelines. From a wound dressing point of view, future studies will need to focus on new delivery systems for silver, as well as the type of matrix in which the silver is deposited. Clearly, adding other actives to enhance the antimicrobial activity, including the disruption of mature biofilms is of interest. From a clinical point of view, the focus needs to be on the wound healing characteristics, and thus randomized control trials will provide more confidence in the results. The application of different wound dressings for specific wounds needs to be clarified, along with the application protocols. It is most likely that no single silver-based dressing can be used for all wounds.

Abstract Background Plant-parasitic root-knot nematodes cause immense yield declines in crop plants that ultimately obviate global food security. They maintain an intimate relationship with their host plants and hijack the host metabolic machinery to their own advantage. The existing resistance breeding strategies utilizing RNAi and resistance (R) genes might not be particularly effective. Alternatively, knocking out the susceptibility (S) genes in crop plants appears to be a feasible approach, as the induced mutations in S genes are likely to be long-lasting and may confer broad-spectrum resistance. This could be facilitated by the use of CRISPR/Cas9-based genome editing technology that precisely edits the gene of interest using customizable guide RNAs (gRNAs) and Cas9 endonuclease. Results Initially, we characterized the nematode-responsive S gene HIPP27 from Arabidopsis thaliana by generating HIPP27 overexpression lines, which were inoculated with Meloidogyne incognita. Next, two gRNAs (corresponding to the HIPP27 gene) were artificially synthesized using laboratory protocols, sequentially cloned into a Cas9 editor plasmid, mobilized into Agrobacterium tumefaciens strain GV3101, and transformed into Arabidopsis plants using the floral dip method. Apart from 1–3 bp deletions and 1 bp insertions adjacent to the PAM site, a long deletion of approximately 161 bp was documented in the T0 generation. Phenotypic analysis of homozygous, ‘transgene-free’ T2 plants revealed reduced nematode infection compared to wild-type plants. Additionally, no growth impairment was observed in gene-edited plants. Conclusion Our results suggest that the loss of function of HIPP27 in A. thaliana by CRISPR/Cas9-induced mutagenesis can improve host resistance to M. incognita.

Total elbow arthroplasty revision rates have been increasing over time due to the increasing use of the procedure with the accompanying increase in complications. The most common complications that typically require revision surgery include aseptic loosening, periprosthetic fractures, infection, and component failure. The associated instability has an overall revision rate reported to be as high as 13%. One important factor when performing a revision surgery is bone quality and bone loss; this represents a challenge during the clinical decision-making process. Currently, there are several strategies used to address bone loss such as arthrodesis, resection arthroplasty, impaction grafting, allograft-prosthetic composite reconstruction, and custom prostheses. The aim of this review article is to provide a comprehensive review of the current strategies to improve diagnosis of failed total elbow arthroplasty and improve management and outcomes of this patient population.

Root-knot nematode Meloidogyne graminicola is one of the most destructive plant parasites in upland as well as direct seeded rice. As an integral part of nematode biology, host finding behavior involves perceiving and responding to different chemical cues originating from the rhizosphere. A sustainable management tactic may include retardation of nematode chemoreception that would impair them to detect and discriminate the host stimuli. Deciphering the molecular basis of nematode chemoreception is vital to identify chokepoints for chemical or genetic interventions. However, compared to the well-characterized chemoreception mechanism in model nematode Caenorhabditis elegans, plant nematode chemoreception is yet underexplored. Herein, the full-length cDNA sequences of two chemotaxis-related genes (Mg-odr-1 and Mg-odr-3) were cloned from M. graminicola. Both the genes were markedly upregulated in the early developmental stages of M. graminicola suggesting their involvement in host finding processes. RNAi-induced independent knockdown of Mg-odr-1 and Mg-odr-3 caused behavioral aberration in second-stage juveniles of M. graminicola which in turn perturbed the nematodes' host finding ability and parasitic success inside rice roots. Additionally, nematodes’ chemotactic response to different host root exudates, volatile and nonvolatile compounds was affected. Our results demonstrating the role of specific chemosensory genes in modulating M. graminicola host seeking behavior can enrich the existing knowledge of plant nematode chemoreception mechanism, and these genes can be targeted for novel nematicide development or in planta RNAi screens.

Regenerated silk fibroin (RSF) protein comprising both hydrophilic and hydrophobic chain segments offer great potential for interfacial interaction and stabilization of organic nanoparticles (NPs) in aqueous medium. In this work, aqueous dispersion of poly(3-hexylthiophene):phenyl-C61-butyric acid methyl ester (P3HT:PCBM) NPs of 1:1 weight ratio were prepared using RSF as surfactant for the first time via the mini-emulsion method. The size, morphology, internal structure, interface and hierarchical organization of RSF-stabilized P3HT:PCBM NPs was investigated using UV–visible spectroscopy, contrast-variation small-angle neutron scattering (CV-SANS) and ultra-small-angle neutron scattering (CV-USANS). The P3HT:PCBM NPs were established to be stabilized in the matrix of RSF colloidal microparticles in water. Unlike the commonly used surfactant sodium dodecyl sulfate which produces core–shell (PCBM–P3HT) NPs, the RSF molecules generated P3HT:PCBM NPs with internal structure that resemble phase-separated solvent-cast films. The as-prepared dispersion has good film-forming ability and has been demonstrated for fabrication of composite films comprising graphene oxide.

Abstract Background Di(2-ethylhexyl) phthalate (DEHP) is a widely detected plasticizer and a priority pollutant of utmost concern for its adverse impact on humans, wildlife and the environment. To eliminate such toxic burden, biological processes are the most promising ways to combat rampant environmental insults under eco-friendly conditions. The present study investigated the biochemical and molecular assessment of the catabolic potential of Mycolicibacterium sp. strain MBM in the assimilation of estrogenic DEHP. Results A detailed biochemical study revealed an initial hydrolytic pathway of degradation for DEHP followed by the assimilation of hydrolyzed phthalic acid and 2-ethylhexanol to TCA cycle intermediates. Besides the inducible nature of DEHP-catabolic enzymes, strain MBM can efficiently utilize various low- and high-molecular-weight phthalate diesters and can grow under moderately halotolerant conditions. Whole genome sequence analysis exhibited a genome size of 6.2 Mb with a GC content of 66.51% containing 6,878 coding sequences, including multiple genes, annotated as relevant to the catabolism of phthalic acid esters (PAEs). Substantiating the annotated genes through transcriptome assessment followed by RT-qPCR analysis, the possible roles of upregulated genes/gene clusters in the metabolism of DEHP were revealed, reinforcing the biochemical pathway of degradation at the molecular level. Conclusions A detailed co-relation of biochemical, genomic, transcriptomic and RT-qPCR analyses highlights the PAE-degrading catabolic machineries in strain MBM. Further, due to functional attributes in the salinity range of both freshwater and seawater, strain MBM may find use as a suitable candidate in the bioremediation of PAEs.

The present study was carried out to evaluate the effect of supplementation of exogenous fibrolytic enzymes (EFE) on voluntary feed intake, availability of different nutrients and growth performance in weaned crossbred calves. Fifteen weaned healthy female Jersey crossbred calves (weight 79.73±3.46 kg; age 234.5±11.33 days) were divided equally into three groups viz., Control (T0), Treatment-1 (T1) and Treatment-2 (T2). Growth trial was conducted on these calves for 90 days. Animals in each treatment group were fed ad lib. total mixed ration (TMR) (CP 12%, TDN 65%) individually for 90 days. Calves under control (T0) group were fed ad lib. TMR (without EFE). Calves under T1 and T2 were fed ad lib. TMR supplemented with EFE cellulase and xylanase @ 8000 and 16000 IU/kg DM of TMR and with EFE cellulase and xylanase @ 12000 and 24000 IU/kg DM of TMR, respectively. Significantly greater DMI per unit body weight (kg/100 kg BW and g/kg W0.75) was recorded in enzyme supplemented groups compared to control group; however, difference between T1 and T2 was non-significant. Addition of EFE in the TMR diet of calves increased TDN and DCP intake (g/kg W0.75) in T1 and T2 than T0 . Supplementation of EFE also enhanced TDN (%), DCP (%), DNDF (%), DE (Mcal/kg of feed) and ME (Mcal/kg of feed) value of the diets under T1 and T2 than T0. Average daily weight gain (g/d/calf) was estimated greater in T1 and T2 than T0. The feed conversion efficiency was similar among three treatments. Whereas, plasma total protein and albumin concentration increased significantly due to supplementation of EFE in the diet of calves. Therefore, it may be concluded that supplementation of exogenous fibrolytic enzymes (cellulase and xylanase @ 8000 and 16000 IU/kg TMR DM) may boost growth potential of finisher Jersey crossbred calves with enhanced intake of nutrients.

Environmental heat stress is one of the restrictive factors for optimum production of dairy cows. Present study was conducted to assess alteration in cardinal physiological responses, heat tolerance indices and milk yield in Jersey crossbred cows at three levels of thermal humidity index (THI), viz. THI-1 (80). THI levels significantly influenced rectal temperature (RT) and respiration rate (RR), but not the pulse rate. With increase in THI level 1 to 3, the RT and RR increased significantly by 0.75°F and 10.70 counts/min during morning and 0.92°F and 12.5 counts/min during afternoon, respectively. The respective enhancement between THI-2 to THI-3 was 0.48°F and 7.06/min in morning and 0.58°F and 7.45/min in afternoon. Among the cardinal physiological responses, breathing rate was the most diurnal variable parameter and it increased by 22-29% from 8:00 am to 2:00 pm with enhancement of stress levels fromTHI-1 to THI-3. Measures of heat tolerance indices revealed that cows were comfortable at THI-1 and discomfort levels were prominent at THI > 80. Low yielders (80. In case of high yielders (>10 kg/day) increase in THI level 1 to 2, DMY did not drop significantly; in contrast, with increase of THI-2 to THI-3, the DMY declined. It was concluded that enhanced breathing rate was the most prominent bio-indicator of natural heat stress in Jersey crossbred cows and at THI>80 production decline due to natural heat stress was nonsignificant in low yielders, whereas highly significant in high yielders (more than 21%). It was recommended that at THI >80, inside of cow shed needs immediate amelioration measures to diminish production losses and high yielders (>10 kg/day) warrant more attention than low yielders during heat stress.

Introduction: Covid-19 pandemic has been a challenge for healthcare system; the doctors in public and private setups are at the center of this challenge. Public and private doctors differ in personality and some occupational aspects. Do these differences reflect in their response to Covid? Aim: To study the difference in anxiety, depression, burnout, and professional fulfillment between doctors in public and private setup during Covid-19 second wave and to assess their coping strategies. Methods: A cross-sectional Internet-based observational study was conducted using Stanford PFI, GAD-7, PHQ-2, BFI-10, and brief COPE questionnaires. Results: A total of 114 public and 37 private doctors participated in the study. Doctors in private were older in age and deferred in personality profile. Significantly, more private doctors screened for anxiety disorder. 62.2% of private and 41.2% public doctors felt professionally fulfilled. Burnout and depression were not significantly different between groups. Active coping, acceptance, and planning coping were significantly more used by private doctors. Conclusion: There are differences how doctors in different setups respond to Covid-19, and there is need to understand these factors.

A study was conducted to investigate the effects of chemical treatment in characterizing the tensile strength, durability, and application of untreated/treated coir geotextile in low-volume roads. Two woven and two nonwoven coir geotextiles were used in this study. Sodium hydroxide and potassium permanganate were the chemicals used to treat the coir geotextiles. The results show that the tensile strength of both the woven coir geotextiles treated with potassium permanganate was lower as compared to both the untreated woven coir geotextiles. The tensile strength of both the nonwoven coir geotextiles treated with sodium hydroxide was higher as compared to both the untreated nonwoven coir geotextiles. Further, the tensile strength of both the woven as well as nonwoven coir geotextiles treated with potassium permanganate was higher in comparison to the one treated with sodium hydroxide. The tensile strength of both the woven as well as nonwoven coir geotextiles first treated with sodium hydroxide followed by potassium permanganate was higher as compared to both the woven as well as nonwoven coir geotextiles treated with sodium hydroxide and potassium permanganate individually. The percentage reduction in tensile strength due to deterioration in the cross-machine direction for both the woven and nonwoven coir geotextiles was higher as compared to the direction of the machine. The results of monotonic load tests show that the models reinforced with untreated/treated woven and nonwoven coir geotextiles show an increase in the bearing pressure at greater deformation. Untreated woven/treated nonwoven coir geotextiles placed at the interface of the sand layer overlying silty sand can be useful in the low-volume roads.

The paper presents the bearing ratio behavior of sand overlying clay with two woven and two non-woven treated coir geotextiles at the interface. The chemicals such as p-aminophenol, sodium periodate and sodium hydroxide were used to treat the coir geotextiles. The results indicated that the unsoaked/soaked bearing ratio of the sand overlying clay with woven coir geotextiles at the interface decreased after the chemical treatment whereas the trend was reverse in case non-woven geotextiles. The unsoaked bearing ratio of the sand overlying clay with the treated/untreated woven and non-woven coir geotextiles at the interface was higher than the soaked bearing ratio. The C: O ratio both for the woven and the non-woven coir geotextiles increased after the chemical treatment. The emissions of Si increased for the woven coir geotextiles whereas it decreased for the non-woven coir geotextiles after the chemical treatment. The chemical treatment results in the modification of the surface of both the untreated woven and non-woven coir geotextiles.

The present study was carried out to evaluate the effect of supplementation of exogenous fibrolytic enzymes (EFE) to the Total Mixed Ration (TMR) at two different levels on intake, nutrient digestibility and nutrient availability in Black Bengal kids. A digestibility trial was conducted on 15 post-weaned Black Bengal kids divided into three groups. Control (T0) group was fed ad lib. TMR was prepared from concentrate mixture and green fodder @ 40: 60 on DM basis. T1 and T2 groups were supplemented with EFE cellulase and xylanase @ 8000 and 16000 IU/kg TMR DM and 12000 and 24000 IU/kg TMR DM, respectively. The study revealed significantly higher digestibility of nutrients, i.e. DM, OM, EE, NDF, ADF, hemicellulose, cellulose and total carbohydrates in both enzyme supplemented groups than the control group. However, the digestibility of CP, cellulose and hemicellulose was significantly higher only in T2 group than the control. There was no significant difference between two enzyme supplemented groups in terms of digestibility of different nutrients except for CP which was significantly higher in T2 than T1. The difference in voluntary intake of DM and OM were non-significant but the intake of CP, DCP and TDN were higher in enzyme supplemented groups. Similarly, TDN, DE and ME content of the diet were also higher in two enzyme supplemented groups than control. However, there was no significant difference between T1 and T2 in terms of nutrient availability and intake. Based on the present study, supplementation of EFE @ cellulase 8000 and xylanase 16000 IU/ kg DM was found to be optimum for improving the nutrient digestibility and availability in Black Bengal kids, which may further improve the productive and reproductive performance of the animals.

ABSTRACT Phthalate diesters are extensively used as plasticizers in manufacturing plastic materials; however, because of their estrogenic properties, these chemicals have emerged as a global threat to human health. The present study investigated the course of degradation of a widely used plasticizer, benzyl butyl phthalate (BBP), by the bacterium PAE-6, belonging to the genus Rhodococcus. The metabolism of BBP, possessing structurally dissimilar side chains, was evaluated biochemically using a combination of respirometric, chromatographic, enzymatic, and mass-spectrometric analyses, depicting pathways of degradation. Consequently, the biochemical observations were corroborated by identifying possible catabolic genes from whole-genome analysis, and the involvement of inducible specific esterases and other degradative enzymes was validated by transcriptomic, reverse transcription-quantitative PCR (RT-qPCR) and proteomic analyses. Nonetheless, phthalic acid (PA), an intermediate of BBP, could not be efficiently metabolized by strain PAE-6, although the genome contains a PA-degrading gene cluster. This deficiency of complete degradation of BBP by strain PAE-6 was effectively managed by using a coculture of strains PAE-6 and PAE-2. The latter was identified as a Paenarthrobacter strain which can efficiently utilize PA. Based on sequence analysis of the PA-degrading gene cluster in strain PAE-6, it appeared that the alpha subunit of the multicomponent phthalate 3,4-dioxygenase harbors a number of altered residues in the multiple sequence alignment of homologous subunits, which may play a role(s) in poor turnover of PA. IMPORTANCE Benzyl butyl phthalate (BBP), an estrogenic, high-molecular-weight phthalic acid diester, is an extensively used plasticizer throughout the world. Due to its structural rigidity and hydrophobic nature, BBP gets adsorbed on sediments and largely escapes the biotic and abiotic degradative processes of the ecosystem. In the present study, a potent BBP-degrading bacterial strain belonging to the genus Rhodococcus was isolated that can also assimilate a number of other phthalate diesters of environmental concern. Various biochemical and multi-omics analyses revealed that the strain harbors all the required catabolic machinery for the degradation of the plasticizer and elucidated the inducible regulation of the associated catabolic genes and gene clusters.

IntroductionMillets are nutritionally superior and climate-resilient short-duration crops and hold a prominent place in cropping sequences around the world. They have immense potential to grow in a marginal environment due to diverse adaptive mechanisms.MethodsAn experiment was conducted in an organic production system in the North Eastern Himalayan foothills of India for 3 consecutive years by evaluating high-yielding varieties (HYVs) of different millets, viz., finger millet, foxtail millet, little millet, barnyard millet, proso millet, and browntop millet, along with local landraces of finger millets (Sikkim-1 and Sikkim-2; Nagaland-1 and Nagaland-2) to identify stable, high-yielding, and nutritionally superior genotypes suited for the region.ResultsAmong the various millets, finger millet, followed by little millet and foxtail millet, proved their superiority in terms of productivity (ranging between 1.16 and 1.43 Mg ha−1) compared to other millets. Among different varieties of finger millets, cv. VL Mandua 352 recorded the highest average grain yield (1.43 Mg ha−1) followed by local landraces, Nagaland-2 (1.31 Mg ha−1) and Sikkim-1 (1.25 Mg ha−1). Root traits such as total root length, root volume, average diameter of roots, and root surface area were significantly higher in finger millet landraces Nagaland-1, Nagaland-2, and Sikkim-1 compared to the rest of the millet genotypes. The different millets were found to be rich sources of protein as recorded in foxtail millet cv. SiA 3088 (12.3%), proso millet cv. TNAU 145 (11.5%), and finger millet landraces, Sikkim-1 and Nagaland-2 (8.7% each). Finger millet landrace Sikkim-2 recorded the highest omega-6 content (1.16%), followed by barnyard millet cv. VL 207 (1.09%). Barnyard millet cv. VL 207 recorded the highest polyunsaturated fatty acid (PUFA) content (1.23%), followed by foxtail millet cv. SiA 3088 (1.09%). The local finger millet landraces Sikkim-1 and Sikkim-2 recorded the highest levels of histidine (0.41%) and tryptophan (0.12%), respectively. Sikkim-1 and Nagaland-2 recorded the highest level of thiamine (0.32%) compared to the HYVs.ConclusionThese findings indicate that finger millet has great potential in the organic production system of the North Eastern Himalayan Region (NEHR) of India, and apart from HYVs like VL Mandua 352, local landraces, viz., Nagaland-2 and Sikkim-1, should also be promoted for ensuring food and nutritional security in this fragile ecosystem.

Bioplastic films comprising both plant- and animal-derived proteins have the potential to integrate the optimal characteristics inherent to the specific domain, which offers enormous potential to develop polymer alternatives to petroleum-based plastic. Herein, we present a facile strategy to develop hybrid films comprised of both wheat gluten and wool keratin proteins for the first time, employing a ruthenium-based photocrosslinking strategy. This approach addresses the demand for sustainable materials, reducing the environmental impact by using proteins from renewable and biodegradable sources. Gluten film was fabricated from an alcohol–water mixture soluble fraction, largely comprised of gliadin proteins. Co-crosslinking hydrolyzed low-molecular-weight keratin with gluten enhanced its hydrophilic properties and enabled the tuning of its physicochemical properties. Furthermore, the hierarchical structure of the fabricated films was studied using neutron scattering techniques, which revealed the presence of both hydrophobic and hydrophilic nanodomains, gliadin nanoclusters, and interconnected micropores in the matrix. The films exhibited a largely (>40%) β-sheet secondary structure, with diminishing gliadin aggregate intensity and increasing micropore size (from 1.2 to 2.2 µm) with an increase in keratin content. The hybrid films displayed improved molecular chain mobility, as evidenced by the decrease in the glass-transition temperature from ~179.7 °C to ~173.5 °C. Amongst the fabricated films, the G14K6 hybrid sample showed superior water uptake (6.80% after 30 days) compared to the pristine G20 sample (1.04%). The suitability of the developed system for multilayer 3D printing has also been demonstrated, with the 10-layer 3D-printed film exhibiting >92% accuracy, which has the potential for use in packaging, agricultural, and biomedical applications.

The origin of high-energy galactic cosmic rays is yet to be understood, but some galactic cosmic-ray accelerators can accelerate cosmic rays up to PeV energies. The high-energy cosmic rays are expected to interact with the surrounding material or radiation, resulting in the production of gamma-rays and neutrinos. To optimize for the detection of such associated production of gamma-rays and neutrinos for a given source morphology and spectrum, a multimessenger analysis that combines gamma-rays and neutrinos is required. In this study, we use the Multi-Mission Maximum Likelihood framework with IceCube Maximum Likelihood Analysis software and HAWC Accelerated Likelihood to search for a correlation between 22 known gamma-ray sources from the third HAWC gamma-ray catalog and 14 yr of IceCube track-like data. No significant neutrino emission from the direction of the HAWC sources was found. We report the best-fit gamma-ray model and 90% CL neutrino flux limit from the 22 sources. From the neutrino flux limit, we conclude that, for five of the sources, the gamma-ray emission observed by HAWC cannot be produced purely from hadronic interactions. We report the limit for the fraction of gamma-rays produced by hadronic interactions for these five sources.

Active galactic nuclei (AGN) are prime candidate sources of the high-energy, astrophysical neutrinos detected by IceCube. This is demonstrated by the real-time multimessenger detection of the blazar TXS 0506+056 and the recent evidence of neutrino emission from NGC 1068 from a separate time-averaged study. However, the production mechanism of the astrophysical neutrinos in AGN is not well established, which can be resolved via correlation studies with photon observations. For neutrinos produced due to photohadronic interactions in AGN, in addition to a correlation of neutrinos with high-energy photons, there would also be a correlation of neutrinos with photons emitted at radio wavelengths. In this work, we perform an in-depth stacking study of the correlation between 15 GHz radio observations of AGN reported in the MOJAVE XV catalog, and 10 yr of neutrino data from IceCube. We also use a time-dependent approach, which improves the statistical power of the stacking analysis. No significant correlation was found for both analyses, and upper limits are reported. When compared to the IceCube diffuse flux, at 100 TeV and for a spectral index of 2.5, the upper limits derived are ∼3% and ∼9% for the time-averaged and time-dependent cases, respectively.

Abstract Background Due to the prolonged usage of Bt-based biopesticides and Bt-transgenic crops worldwide, insects are continually developing resistance against Cry toxins. This resistance may occur if any mechanistic step in the insecticidal process is disrupted possibly because of the alteration in Cry-receptor binding affinity due to mutation in receptor genes. Compared to other lepidopteran insects, Cry receptor-related research has made asymmetric progress in the model insect Galleria mellonella. Results Present study describes the molecular characterization and functional analysis of five Cry toxin receptor-related genes (prohibitin, GLTP, α-amylase, ADAM and UDP-GT) and a gut repair gene (arylphorin) from the gut tissues of G. mellonella. Protein–protein docking analysis revealed that Cry1AcF putatively binds with all the five candidate proteins, suggesting their receptor-like function. These receptor-like genes were significantly overexpressed in the gut tissues of fourth-instar G. mellonella larvae upon early exposure to a sub-lethal dose of Cry1AcF toxin. However, targeted knockdown (by using bacterially-expressed dsRNAs) of these genes led to variable effect on insect susceptibility to Cry1AcF toxin. Insects pre-treated with prohibitin and α-amylase dsRNA exhibited significant reduction in Cry1AcF-induced mortality, suggesting their probable role as Cry receptor. By contrast, insects pre-treated with GLTP, ADAM and UDP-GT dsRNA exhibited no significant decline in mortality. This maybe explained by the possibility of RNAi feedback regulation (as few of the receptors belong to multigene family) or redundant role of GLTP, ADAM and UDP-GT in Cry intoxication process. Conclusion Since the laboratory culture of G. mellonella develop Bt resistance quite rapidly, findings of the current investigation may provide some useful information for future Cry receptor-related research in the model insect.

Abstract Background Strides have been made in Bangladesh to promote the utilisation of biological control agents (BCAs), however farmer utilisation remains sub-optimal. The establishment of local BCA production hubs, although touted as a panacea to this problem, has no proven business case. This study makes the case for a non-linear business model. Methods Qualitative and quantitative data from maize growing areas in Bangladesh was collected via telephone interviews from key informants representing four key stakeholders—national research institute, regional research stations, farmer producer organisations and agro-dealers. Results Farmer uptake of BCAs in Bangladesh for FAW management is hindered by several factors—lack of BCAs availability in local markets, negative farmer and agro-dealer perceptions, poor input industry linkages for the supply of BCAs products to agro-dealers and inadequate institutional finances for capacity building of and technical support by research scientists and extension agents. Given these challenges to BCAs uptake, an innovation systems-based business model that links researchers, extensionists, agro-dealers and farmer producer organizations in a non-linear pathway is proposed for Bangladesh. This translates into the establishment of local BCA production hubs owner-managed by farm entrepreneurs, with scientists providing them with nucleus culture, while extension services provide technical support for quality assurance. The interaction between all stakeholders is non-linear with all actors intellectually consulted and engaged, with technical capacity on BCAs available for any actor requiring it. Multi-disciplinary research, that takes into account feedback from stakeholders, complements the process thus generating robust and relevant knowledge for feedbacking to improve the business model, capacity building initiatives and farmer engagement. Conclusions Mentoring and capacity building leveraged via engagement of research institutions; and demonstration of technology use and guidance utilising extension services and agro-dealer networks, will promote the utilisation of BCAs for FAW management and enable local farm entrepreneurs to meet the increased demand via establishment of local BCA production hubs.

The objective of this study was to evaluate the effect of dietary supplementation of Da qing shu (Ficus hookeri) leaf meal as herbal feed additive on nutrients utilization and growth performance of growing crossbred calves. Ten Jersey male cross-bred calves (~ 4 months of age) were divided equally into two groups (G1 and G2) and were fed ad lib. under stall feeding on a paddy straw based mixed ration consisting of 50% paddy straw and 50% concentrate mixture for a duration of 140 days. Two types (C1 and C2) of iso-nitrogenous concentrate mixtures were prepared. Four per cent (w/w) wheat bran in concentrate mixture (C2) of test group (G2) was replaced by sun dried ground Ficus hookeri leaf meal. Experimental calves of test group (G2) were fed Ficus hookeri leaf meal @ 2% of the diet. Daily dry matter intake (g/d) was similar among the calves of two experimental groups. Apparent digestibility of DM, OM, NDF, ADF and cellulose improved in the Ficus hookeri leaf meal fed calves (G2). DCP value of the ration was similar while, TDN value of the ration was higher in the calves of Ficus hookeri leaf meal supplemented group (G2). However, plane of nutrition among the calves of both experimental groups was similar. Average finishing body weight, daily body weight gain, feed conversion efficiency and blood glucose level were higher in Ficus hookeri leaf fed calves (G2). Therefore, it may be concluded from the results of the study that dietary supplementation of Da qing shu (Ficus hookeri) leaf meal as herbal feed additive @ 2% of total mixed diet significantly improved the fibre digestibility, average daily body weight gain and feed conversion efficiency in growing Jersey male crossbred calves.

Soil contaminated with hexavalent chromium seriously threatens the environment and human health. The use of FeS, which has a high redox activity and excellent reduction capacity, limits its application in soil remediation due to its premature surface oxidation and massive aggregation. To prevent premature surface oxidation and agglomeration, cetyltrimethylammonium bromide-supported nano-ferrous sulfide (CTAB-nFeS) was chemically synthesized and used for immobilizing Cr(VI) in contaminated soil. In order to evaluate the role of CTAB stabilization of nFeS and interaction mechanisms were investigated by XPS, FTIR, XRD, and FESEM. Batch experiments showed a complete reduction of Cr(VI) within 3 h with only 235% excess of CTAB-nFeS at a soil pH of 8 compared to days as reported in the literature with alternative FeS forms. The reduction kinetic data could be satisfactorily fitted into the second-order rate model. The rate constant linearly depends on the soil-to-water ratio, but its logarithmic form is linear in the given pH range. The oxidation-reduction potential increases with decreasing initial pH, thus positively impacting the reduction process. XPS analysis revealed the reduction process as multi-steps (reduction, adsorption, and co-precipitation). Ecological studies showed improved plant growth and earthworm survival rate in the remediated soil. Medium-term stability experiments suggested a significant decrease in TCLP leachate concentration of Cr after CTAB-nFeS treatment and remained stable for 60 d. Overall results of our study suggested a sustainable, feasible, and effective strategy for in-situ remediation of Cr(VI)-contaminated soil using CTAB-nFeS at natural pH.

Pest profiles in today’s global food production system are continually affected by climate change and extreme weather. Under varying climatic conditions, plant-parasitic nematodes (PPNs) cause substantial economic damage to a wide variety of agricultural and horticultural commodities. In parallel, their herbivory also accredit to diverse ecosystem services such as nutrient cycling, allocation and turnover of plant biomass, shaping of vegetation community, and alteration of rhizospheric microorganism consortium by modifying the root exudation pattern. Thus PPNs, together with the vast majority of free-living nematodes, act as ecological drivers. Because of direct exposure to the open environment, PPN biology and physiology are largely governed by environmental factors including temperature, precipitation, humidity, atmospheric and soil carbon dioxide level, and weather extremes. The negative effects of climate change such as global warming, elevated CO2, altered precipitation and the weather extremes including heat waves, droughts, floods, wildfires and storms greatly influence the biogeographic range, distribution, abundance, survival, fitness, reproduction, and parasitic potential of the PPNs. Changes in these biological and ecological parameters associated to the PPNs exert huge impact on agriculture. Yet, depending on how adaptable the species are according to their geo-spatial distribution, the consequences of climate change include both positive and negative effects on the PPN communities. While assorting the effects of climate change as a whole, it can be estimated that the changing environmental factors, on one hand, will aggravate the PPN damage by aiding to abundance, distribution, reproduction, generation, plant growth and reduced plant defense, but the phenomena like sex reversal, entering cryptobiosis, and reduced survival should act in counter direction. This seemingly creates a contraposition effect, where assessing any confluent trend is difficult. However, as the climate change effects will differ according to space and time it is apprehensible that the PPNs will react and adapt according to their location and species specificity. Nevertheless, the bio-ecological shifts in the PPNs will necessitate tweaking their management practices from the agri-horticultural perspective. In this regard, we must aim for a ‘climate-smart’ package that will take care of the food production, pest prevention and environment protection. Integrated nematode management involving precise monitoring and modeling-based studies of population dynamics in relation to climatic fluctuations with escalated reliance on biocontrol, host resistance, and other safer approaches like crop rotation, crop scheduling, cover cropping, biofumigation, use of farmyard manure (FYM) would surely prove to be viable options. Although the novel nematicidal molecules are target-specific and relatively less harmful to the environment, their application should not be promoted following the global aim to reduce pesticide usage in future agriculture. Thus, having a reliable risk assessment with scenario planning, the adaptive management strategies must be designed to cope with the impending situation and satisfy the farmers’ need.

The main goal of treatment for chronically unreduced elbow dislocations is to restore a stable, concentric joint and regain a satisfactory arc of motion. Due to the conflicting goals of restoring elbow stability and regaining a good arc of motion, the treatment of chronic elbow dislocation remains a challenge for even the experienced orthopedic surgeon. The standard treatment of these dislocations consists of open reduction, V-Y muscleplasty of the triceps, and temporary arthrodesis or cast immobilization. However, prolonged postoperative immobilization may result in elbow stiffness, which significantly limits the functional outcome. We present our surgical technique with a focus on restoring stable reduction such that early motion can be instituted and complications of prolonged immobilization can be avoided. From position to wound closure, surgical steps are presented in detail, with pearls for practice and a discussion on chronic elbow dislocation. The internal joint stabilizer is a safe and effective implant that complements the management of chronic elbow dislocations. This reproducible surgical technique allows for stability and early mobility while having the added benefit of circumventing complications associated with prolonged immobilization and hinged external fixation. Understanding the surgical indications, as well as the nuances of the surgical technique utilizing the internal joint stabilizer, is critical in order to improve patient outcomes and avoid complications.

Background: Expandable magnetic rods and intramedullary nails are being used in a number of innovative ways, including limb length discrepancy and scoliosis correction. However, recently, the full complement of these devices has been further explored, with the utilization of their compressive capacity to improve fracture healing. The purpose of the present study was to report on early results of compressive magnetic intramedullary nailing for humeral shaft delayed unions and nonunions. Methods: This retrospective case series was completed at a level 1 trauma center, with adult patients who underwent compressive intramedullary nailing from 2017 to 2021 for humeral shaft nonunion or delayed union. The primary indication for this procedure was nonunion in the setting of previous conventional fixation, but a subset of patients with atrophic nonunions and risk factors for recalcitrant nonunion were also included. Results: Fourteen patients, with a mean age of 51 ± 17 years, underwent compressive magnetic intramedullary nailing. Nine patients had previously underwent surgery, 6 of which had undergone multiple prior procedures. Five others were initially treated nonoperatively and underwent surgery 4.1 ± 2.9 months out from injury. Ten patients went on to union at a mean of 2.9 ± 2.4 months. One patient experienced hardware failure with nail cut-out at 2 weeks, and one required revision surgery for a wound infection. Three other patients were lost to follow-up, one of which was deceased for reasons unrelated to surgery. Conclusion: Compressive magnetic intramedullary nails are a viable solution for complex humeral shaft nonunions, particularly in the setting of previously well-fixed fractures and those at risk of recalcitrant nonunion. However, comparative and prospective studies looking at union rates and secondary procedures are needed to more clearly define their role in treatment and assure their safety, given recent concerns regarding osteolysis at the nail modular junction.

Abstract Background Septic arthritis of the native shoulder is traditionally diagnosed with the same strategies as knee or hip septic arthritis. However, septic arthritis of the shoulder is frequently a missed or delayed diagnosis. Reliance on aspiration and serum markers has been called into question recently. The purpose of this study was to conduct a systematic review investigating the value of joint aspiration and serum markers in the diagnosis of native shoulder joint sepsis. Methods PubMed/MEDLINE, Scopus, and the Cochrane Library were used in the systematic literature search from January 1, 1960, through January 23, 2021. The primary outcome was to report on the synovial white cell count of patients with native shoulder sepsis. Descriptive statistics using percentages, means, and intraclass correlation coefficient (ICC) values were used to summarize the results. Results Thirty-one studies, including 25 case series, one case-control, and five cohort studies with a total of 7434 native shoulder joints, were included. There was no standardized approach to diagnosing septic arthritis of the shoulder. Only 10 studies (32%) reported on synovial white cell count with the majority yielding aspiration counts greater than 50,000 cells/mm3, although one study was as low as 30,000 cells/mm3. Conclusions The diagnosis of native shoulder joint sepsis lacks uniformity. Methods used to evaluate shoulder sepsis are heterogeneous and may lead to delays or misdiagnosis with devastating sequelae. Synovial white cell count is underutilized and may also present with a lower value than expected, which is likely related to the time interval between symptom onset and diagnosis.

Thirty-two weaned male Barbari kids (av. age, 144.65 days; weight, 9.43±0.24 kg), reared under intensive system of feeding, were divided equally into four groups (T1, T2, T3 and T4) to investigate the effect of supplementation of different levels of concentrate mixture on voluntary intake, growth performance, carcass traits and quality. All kids were fed with Bengal gram straw ad lib. + green fodder ad lib. as basal ration and supplemented with barley grain @ 0.7% of body weight in T1 and concentrate mixture @ 0.7, 1.4, and 2.1% of body weight in T2, T3, and T4, respectively. Dry Matter Intake (DMI, g)/kg W0.75 increased significantly in T1 and T4 than T2 and T3. Average daily weight gain (ADG) (g/d/kid) was enhanced significantly with each gradual increase in concentrate level. Slaughter weight and empty body weight were greatest in T4 compared with other treatments. Significantly greater dressing percentage and yield of separated lean meat was observed in kids under T3 and T4 than other two groups. But, meat:bone ratio and separated carcass fat yield was significantly higher only in kids under T4. Meat protein content (%) increased in T2, T3, and T4 as compared to control T1. Meat fat content increased linearly due to a gradual increase in the level of concentrate mixture supplementation to the finisher kids. Whereas, meat moisture and ash per cent remained unaltered due to different levels of concentrate supplementation to kids. Therefore, it may be concluded that supplementation of concentrate mixture @ 2.1% of the body weight in the roughage-based diet increased DMI in finisher Barbari kids with greater energy and protein values, which attributed to enhanced ADG in such kids by 408.3% over the control (supplemented with barley grain only), and also improved feed conversion ratio, carcass yield, carcass traits, and meat quality.

Background A multitude of Cry toxins (secreted by Bacillus thuringiensis or Bt) has been deployed globally either via transgenic mean or bio-pesticidal formulations in order to manage insect pests. However, Bt resistance development in insects is emerging as a major concern. To avoid this problem, multiple gene pyramiding or protein-engineered chimeric toxin-based strategy has been analyzed. Methods In the present study, one such chimeric toxin Cry1AcF (contain the swapped domains of Cry1Ac and Cry1F) was used to investigate its in vivo pathogenesis process in lepidopteran pests Spodoptera frugiperda and S. litura. A number of biochemical and molecular analysis were performed. Results Oral ingestion of Cry1AcF caused greater toxicity in S. frugiperda than S. litura with larvae displaying increased hemolymph melanization. Histopathology of the midgut transverse sections exhibited Cry1AcF-induced extensive gut damage in both the test insects followed by cytotoxicity in terms of reduced hemocyte numbers and viability. Elevated hemolymph phenoloxidase activity indicated the immune-stimulatory nature of Cry1AcF. In order to analyze the role of gut receptor proteins in Cry1AcF intoxication in test insects, we performed RNAi-mediated silencing using bacterially-expressed dsRNAs of individual receptor-encoding genes including CAD, ABCC2, ALP1 and APN. Target-specific induced downregulation of receptor mRNAs differentially altered the insect susceptibility to Cry1AcF toxin in our study. The susceptibility of ALP1 and APN dsRNA pre-treated S. frugiperda was considerably decreased when treated with Cry1AcF in LD50 and LD90 doses, whereas susceptibility of CAD and ABCC2 dsRNA pre-treated S. litura was significantly reduced when ingested with Cry1AcF in different doses. CAD/ABCC2-silenced S. frugiperda and ALP1/APN-silenced S. litura were vulnerable to Cry1AcF alike of control larvae. In conclusion, our results indicate ALP1/APN and CAD/ABCC2 as the functional receptor for Cry1AcF toxicity in S. frugiperda and S. litura, respectively.

Photorhabdus bacteria secrete a repertoire of protein toxins that can kill the host insect. Among them, toxin complex (Tc) proteins have gained significant attention due to their wider conservation across the different bacterial genera. In our laboratory, a C-terminal domain of TcaB protein was characterized from P. akhurstii bacterium that conferred the potent oral insecticidal effect on Galleria mellonella. However, the role of insect gut receptors in the TcaB intoxication process was yet to be investigated. In the current study, we examined the transcription of candidate midgut receptors in TcaB-infected larvae and subsequently cloned a cadherin-like gene, GmCAD, from G. mellonella. GmCAD was highly transcribed in the fourth-instar larval stage and specifically in the midgut tissues. Our ligand blot and binding ELISA assays indicated that TcaB binds to the truncated peptides from the GmCAD transmembrane-proximal region with greater affinity than that from the transmembrane-distal region. Oral administration of bacterially expressed GmCAD dsRNA in G. mellonella severely attenuated the expression of target mRNA, which in turn alleviated the negative effect of TcaB on insect survival (TcaB-induced mortality in CAD dsRNA pretreated larvae reduced by 72–83% compared to control), implying the association of GmCAD in the TcaB intoxication process. Present findings form a basis of future research related to the insect gut receptor interactions with Photorhabdus toxins.

Advances made in synthesis and analytical techniques has allowed the exploration and mimicry of natural materials. Resilin-mimetics have emerged from this advance as a biomaterial with a range of potential applications. Here, the authors review the history and current research on resilin-mimetics, providing a future perspective.

In most of the developing nations, fast paced urbanisation is going on. This has changed the spatial patterns of Land Use Land Cover (LULC) and Land Surface Temperature (LST) over time. Continual studies are required in this context to know these phenomena more clearly. This study is carried out to analyse the spatio-temporal changes in LULC, urbanisation and LST in the metropolitan cities of India namely Delhi and Bengaluru. Landsat images of TM and OLI sensors are taken for the years 2001, 2010 and 2020. The LULC layer is obtained through supervised image classification. Concentric circles at the interval of 2 km are drawn from the centroid of the study areas which are used to compute Shannon's entropy through zonal analysis of the reclassified LULC layer. The thermal band of the Landsat is used for the computation of LST. The results of both the study areas revealed 1) decline in the open land, vegetation and water body; 2) rampant growth of built-up and urban area which become more compact over the years; and 3) spread of the higher LST zones.

The paper presents the bearing ratio behavior of sand overlying silty sandy with two woven and two non-woven treated coir geotextiles at the interface under unsoaked and soaked condition. The chemicals such as sodium hydroxide and potassium permanganate were used to treat the coir geotextiles. The coir geotextiles were placed at two different locations (Designated as Series A and Series B). The results indicated that the bearing ratio of the sand overlying silty sand with untreated/ treated woven/non-woven coir geotextiles at the interface was higher in the unsoaked condition in comparison to soaked condition for the Series A and Series B. The improvement in the bearing ratio of the sand overlying silty sand with untreated/ treated woven/non-woven coir geotextiles at the interface in the unsoaked/soaked condition was higher for the Series A in comparison to the Series B. The bearing ratio of the sand overlying silty sand increased when the woven/non-woven coir geotextiles at the interface were treated with sodium hydroxide. The improvement in the bearing ratio of the sand overlying silty sand with potassium permanganate treated woven/non-woven coir geotextiles at the interface was higher in comparison to the sodium hydroxide treated woven/non-woven coir geotextiles. The highest improvement in the bearing ratio was achieved when both the woven/non-woven coir geotextiles were treated with sodium hydroxide followed by potassium permanganate.

Abstract Computer-aided detection of brain lesions from volumetric magnetic resonance imaging (MRI) is in demand for fast and automatic diagnosis of neural diseases. The template-matching technique can provide satisfactory outcome for automatic localization of brain lesions; however, finding the optimal template size that maximizes similarity of the template and the lesion remains challenging. This increases the complexity of the algorithm and the requirement for computational resources, while processing large MRI volumes with three-dimensional (3D) templates. Hence, reducing the computational complexity of template matching is needed. In this paper, we first propose a mathematical framework for computing the normalized cross-correlation coefficient (NCCC) as the similarity measure between the MRI volume and approximated 3D Gaussian template with linear time complexity, $${\mathbf{\mathcal{O}}}\left( {{\varvec{a}}_{{{\varvec{max}}}} {\varvec{N}}} \right)$$ O a max N , as opposed to the conventional fast Fourier transform (FFT) based approach with the complexity $${\mathbf{\mathcal{O}}}\left( {{\varvec{a}}_{{{\varvec{max}}}} {\varvec{N}}\log {\varvec{N}}} \right)$$ O a max N log N , where $${\varvec{N}}$$ N is the number of voxels in the image and $${\varvec{a}}_{{{\varvec{max}}}}$$ a max is the number of tried template radii. We then propose a mathematical formulation to analytically estimate the optimal template radius for each voxel in the image and compute the NCCC with the location-dependent optimal radius, reducing the complexity to $${\mathbf{\mathcal{O}}}\left( {\varvec{N}} \right)$$ O N . We test our methods on one synthetic and two real multiple-sclerosis databases, and compare their performances in lesion detection with FFT and a state-of-the-art lesion prediction algorithm. We demonstrate through our experiments the efficiency of the proposed methods for brain lesion detection and their comparable performance with existing techniques.

Targeted protein degradation is a rapidly advancing and expanding therapeutic approach. Drugs that degrade GSPT1 via the CRL4CRBN ubiquitin ligase are a new class of cancer therapy in active clinical development with evidence of activity against acute myeloid leukemia in early-phase trials. However, other than activation of the integrated stress response, the downstream effects of GSPT1 degradation leading to cell death are largely undefined, and no murine models are available to study these agents. We identified the domains of GSPT1 essential for cell survival and show that GSPT1 degradation leads to impaired translation termination, activation of the integrated stress response pathway, and TP53-independent cell death. CRISPR/Cas9 screens implicated decreased translation initiation as protective following GSPT1 degradation, suggesting that cells with higher levels of translation are more susceptible to the effects of GSPT1 degradation. We defined 2 Crbn amino acids that prevent Gspt1 degradation in mice, generated a knockin mouse with alteration of these residues, and demonstrated the efficacy of GSPT1-degrading drugs in vivo with relative sparing of numbers and function of long-term hematopoietic stem cells. Our results provide a mechanistic basis for the use of GSPT1 degraders for the treatment of cancer, including TP53-mutant acute myeloid leukemia.

In this work, regenerated silk fibroin (RSF)/reduced graphene oxide (rGO) hybrid ink system was fabricated in aqueous phase, extrusion printed and photocrosslinked to hydrogels with tunable physicochemical properties and cell-gel interactions. Aqueous isopropyl alcohol was used to disperse the rGO into RSF matrix, where the RSF molecules showed relatively increased radius of gyration with IPA resulting in increased pore size and water uptake capacity of the photocrosslinked hydrogels. Incorporation of IPA along with increase in rGO content systematically decreased the viscosity, contact angle and printing accuracy of RSF/rGO hybrid inks, where the ink with highest rGO content exhibited relatively loose network structure at printing shear rate. However, no secondary structural change and fibrillogenesis was observed under printing shear stress, making it a suitable system for bioprinting silk hydrogel scaffolds. In addition, increase in rGO content systematically increased the crosslink density, β-sheet content, and mechanical properties of RSF/rGO hybrid hydrogels, whereas decreased the micropore size and water uptake capacity. Moreover, with increase in rGO content, the hybrid and bioprinted hydrogels showed good biocompatibility with marginal difference in cell viability on hybrid and bioprinted gels. The developed hydrogel systems could be potentially applied for tissue engineering and other functional applications.

Coluber fasciolatus Shaw, 1802 (today Argyrogena fasciolata auctt.) is the name used for a widespread terrestrial colubrid snake species inhabiting subtropical and tropical dry deciduous/thorn forests of South Asia from Pakistan to India, with unconfirmed records of distribution in Nepal, Bangladesh and Myanmar and a single doubtful record from the northern tip of Sri Lanka. During the past 200 years, A. fasciolata (common name Banded racer) has been placed in different genera, i.e. Tyria Fitzinger, 1826, Zamenis Wagler, 1830, Coryphodon Duméril, Bibron and Duméril, 1854, and Coluber Linnaeus, 1758 where it primarly remained until the mid 1960s and exceptionally until the year 2011. Three subsequently introduced names, viz. Coluber hebe Daudin, 1803, Coluber curvirostris Cantor, 1839, and Argyrogena rostrata Werner, 1924 were synonymized with C. fasciolatus shortly after its description. Based on a combination of characters including body pattern, external morphology and osteological differences Wilson (1967) reviewed the taxon fasciolatus Shaw and considered it as generically distinguishable, removed it from the then heterogeneous and undefined collective genus Coluber and assigned it to the resurrected genus Argyrogena Werner, 1924. Shaw’s (1802) description of C. fasciolatus was based exclusively on the information of Russell’s “Nooni Paragoodoo” published in 1796 in his “Account of Indian Serpents, collected on the coast of Coromandel; […]”. Our analysis of the original data and the depicted type specimen in Russell (1796) revealed that the name fasciolata was initially established for a species distinct from that currently known as the “Banded racer”, and that Russell’s data have been used simultaneously but unwittingly, for more than 150 years, as original source for two valid species from two different genera. Specimens of Banded racer found in the southeastern part of peninsular India are morphologically and genetically distinct from populations of the rest of the distribution area. These populations from central and southern Tamil Nadu state represent a different species, consequently described as a new species herein. Furthermore, examination of specimens of the Banded racer from different populations across its entire range, including the type specimen of the genus Argyrogena (A. rostrata Werner), reveals a similarity in morphology with the genus Platyceps Blyth, 1860. This was further supported by molecular data which demonstrates that the genus Argyrogena is nested within Platyceps.

Two Na(I) coordination polymers, namely, {Na(BA)2(μ-H2O)2}n{adp}n (1) and {[Na2(μ-BA)(μ-fum)(μ-H2O)4](BA)}n (2) (where, BA = boric acid, adp = adipic acid, fum = fumarate),were prepared and characterized using elemental analysis, TGA, FT-IR, and single-crystal X-ray diffraction techniques. Various unconventional supramolecular interactions, i.e., CH∙∙∙HC and parallel CO∙∙∙CO interactions, stabilize the layered assembly of compound 1. Interesting dual enclathration of BA molecules within the supramolecular host cavities formed by O-H∙∙∙O and C-H∙∙∙C interactions stabilizes the crystal structure of compound 2. The H-bonding interactions in 1 and 2 were further studied theoretically using the quantum theory of atoms in molecules (QTAIM) and the noncovalent interaction plot (NCI Plot) computational tools. The energy of the H-bonds was estimated using the potential energy density at the bond critical points. Theoretical calculations confirmed the presence of O-H∙∙∙O H-bonding interactions in both compounds, forming structure-guiding R22(8) synthons relevant for the stability of the compounds.

Two multi-component coordination compounds of Mn(II), viz. [Mn(H2O)6](2-Mepy)2(4-NO2bz)2·2H2O (1) and [Mn(H2O)6][Mn(2,3-PDCH)3]2 (2) (where, 2-Mepy = 2-methylpyridine, 4-NO2bz = 4-nitrobenzoate, 2,3-PDC = 2,3-pyridinedicarboxylate), have been synthesized and characterized using elemental, spectroscopic (FT-IR and electronic), TGA and single-crystal X-ray diffraction analyses. Complex 1 is a co-crystal hydrate of Mn(II) involving uncoordinated 2-Mepy, 4-NO2bz and water molecules; while compound 2 is a multi-component molecular complex salt of Mn(II) comprising cationic [Mn(H2O)6]2+ and anionic [Mn(2,3-PDCH)3]−complex moieties. The uncoordinated 2-Mepy and 4-NO2bz moieties of 1 are involved in lone-pair (l.p)-π and C–H⋯π interactions which stabilize the layered assembly of the compound. The crystal structure of compound 2 has been previously reported. However, we have explored the unusual enclathration of complex cationic moieties within the supramolecular host cavities formed by the molecular assembly of complex anionic moieties. The supramolecular assemblies obtained in the crystal structure have been further studied theoretically using DFT calculations, quantum theory of atoms-in-molecules (QTAIM) and non-covalent interaction plot (NCI plot) computational tools. Theoretical studies reveal that the combination of π-staking interactions (l.p-π, π-π and C–H···π) have more structure-guiding roles compared to the H-bonds. The large binding energy of π-stacking interactions in 2 is due to the antiparallel orientation of aromatic rings and their coordination to the metal centers, thereby increasing the contribution of the dipole–dipole interactions.

Purpose: The people of Northeast India comprise mostly of Mongoloid descent with characteristic craniofacial features. The purpose of this study was to evaluate the normal orbital structures with MRI and determine normative data in the Northeast Indian pediatric population. Methods: MRI images of the 302 orbits of 151 pediatric patients below 16 years of age were retrospectively evaluated to measure the various orbital structures. Both axial and coronal MRI images were utilized for various orbital measurements. An independent sample t-test was done to compare various orbital data according to the sex and side. Linear regression was also done. Results: The mean age of the pediatric population was 9.64 ± 1.47 years with a male: Female ratio of 1.13:1. The mean interzygomatic line was 88.4 ± 9.09 mm and the interorbital line was 22.18 ± 3.62 mm. The mean thicknesses of medial, lateral, superior, and inferior recti muscles were 2.58 ± 0.46, 2.34 ± 0.42, 2.16 ± 0.40, and 2.53 ± 0.49 mm in males and 2.41 ± 0.41, 2.08 ± 0.34, 2.08 ± 0.46, and 2.46 ± 0.49 mm in females. The mean horizontal orbital, vertical orbital diameters, orbital index, mean volume of eyeball, and orbital cavity were 30.27 ± 2.97,3 7.06 ± 3.57, 122.58 ± 7.39, 4.63 ± 0.84, and 15.29 ± 3.52 in males, while 29.16 ± 3.23, 34.96 ± 3.99,119.96 ± 7.31, 4.49 ± 0.87, and 14.65 ± 3.47 in females, respectively. With an increase in age, the interzygomatic line (r = 0.883, r2 = 0.780; P < 0.0005), anterior medial interorbital line (r = 0.808, r2 = 0.652; P < 0.0005), mean volume of eyeball (r = 0.915, r2 = 0.838; P < 0.0005), orbital cavity (r = 0.924, r2 = 0.854; P < 0.0005), and distance between the optic nerve entry site (r = 0.829, r2 = 0.687; P < 0.0005) were increased. Conclusion: This study provides normative data of various orbital structures in a pediatric population and these data likely to be useful for diagnosing various pediatric orbital disorders and helps in the planning of various surgical procedures of orbits.

Abstract Host–pathogen interaction is one of the most powerful determinants involved in coevolutionary processes covering a broad range of biological phenomena at molecular, cellular, organismal and/or population level. The present study explored host–pathogen interaction from the perspective of human–bacteria protein–protein interaction based on large-scale interspecific and intraspecific interactome data for human and three pathogenic bacterial species, Bacillus anthracis, Francisella tularensis and Yersinia pestis. The network features revealed a preferential enrichment of intraspecific hubs and bottlenecks for both human and bacterial pathogens in the interspecific human–bacteria interaction. Analyses unveiled that these bacterial pathogens interact mostly with human party-hubs that may enable them to affect desired functional modules, leading to pathogenesis. Structural features of pathogen-interacting human proteins indicated an abundance of protein domains, providing opportunities for interspecific domain-domain interactions. Moreover, these interactions do not always occur with high-affinity, as we observed that bacteria-interacting human proteins are rich in protein-disorder content, which correlates positively with the number of interacting pathogen proteins, facilitating low-affinity interspecific interactions. Furthermore, functional analyses of pathogen-interacting human proteins revealed an enrichment in regulation of processes like metabolism, immune system, cellular localization and transport apart from divulging functional competence to bind enzyme/protein, nucleic acids and cell adhesion molecules, necessary for host-microbial cross-talk.

Background: The coronavirus-19 (COVID-19) pandemic has affected millions of people across the world since early 2020. Besides the large number of case fatalities, this virus has produced significant health-related sequelae involving multiple systems of the body. As with previous coronavirus infections, this was also found to be associated with various neuropsychiatric symptoms. Psychosis has been uncommon, and the few reported cases across the world have forwarded association with either raised inflammatory markers or the consequences of social isolation. Materials and Methods: This is a retrospective descriptive study of 12 patients, who were admitted with COVID-19 infection and psychosis, between March 2020 and December 2020. Cases of head injury, any neurological or metabolic illnesses, and substance use disorders were excluded. Results: Cases with psychosis formed only 0.19% of all cases of COVID-19 admissions. All of them were young male and employed. Most of them had abrupt onset of psychosis with confusion, delusions, hallucinations, agitation, and sleep disturbances. Investigations including inflammatory markers (C-reactive protein) and computerized tomography scans were largely normal. Medications used were mainly benzodiazepines and antipsychotics. Most of the cases resolved within the second week, and follow-up after a month did not elicit any residual symptoms in majority. Diagnosis was acute and transient psychotic disorder (about 75%), bipolar affective disorder (2 cases), and schizophrenia (one). Conclusions: The major findings included nonreactive inflammatory markers, quick resolution of symptoms, requirement of low doses of antipsychotic drugs, and no long-term sequelae.

The rise of drug-resistant tuberculosis poses a major risk to public health. Statins, which inhibit both cholesterol biosynthesis and protein prenylation branches of the mevalonate pathway, increase anti-tubercular antibiotic efficacy in animal models. However, the underlying molecular mechanisms are unknown. In this study, we used an in vitro macrophage infection model to investigate simvastatin's anti-tubercular activity by systematically inhibiting each branch of the mevalonate pathway and evaluating the effects of the branch-specific inhibitors on mycobacterial growth. The anti-tubercular activity of simvastatin used at clinically relevant doses specifically targeted the cholesterol biosynthetic branch rather than the prenylation branches of the mevalonate pathway. Using Western blot analysis and AMP/ATP measurements, we found that simvastatin treatment blocked activation of mechanistic target of rapamycin complex 1 (mTORC1), activated AMP-activated protein kinase (AMPK) through increased intracellular AMP:ATP ratios, and favored nuclear translocation of transcription factor EB (TFEB). These mechanisms all induce autophagy, which is anti-mycobacterial. The biological effects of simvastatin on the AMPK-mTORC1-TFEB-autophagy axis were reversed by adding exogenous cholesterol to the cells. Our data demonstrate that the anti-tubercular activity of simvastatin requires inhibiting cholesterol biosynthesis, reveal novel links between cholesterol homeostasis, the AMPK-mTORC1-TFEB axis, and Mycobacterium tuberculosis infection control, and uncover new anti-tubercular therapy targets.

Aim: The present study was conducted to record the prevalence of multidrug-resistant (MDR), extended-spectrum β-lactamases (ESBLs) producing Escherichia coli from pig population of organized and unorganized farms of Mizoram and to record the presence of ESBLs, non-ESBLs, and integrons. Materials and Methods: Fecal samples were collected from pigs under organized (n=40) and unorganized (n=58) farms of Mizoram. Samples were processed for isolation and identification of E. coli by conventional techniques, BD Phoenix™ automated bacterial system, and polymerase chain reaction (PCR) based confirmatory test. All the isolates were subjected to antimicrobial sensitivity test by disk diffusion assay and ESBLs production by double-disk synergy test (DDST). The ESBLs producing isolates were subjected to PCR for determination of ESBLs genes and all the isolates were screened for non-ESBLs genes and integrons by PCR. Results: A total of 258 E. coli was isolated and identified from organized (n=120) and unorganized farms (n=138). Majority of the E. coli isolates exhibited high level of resistance against amoxicillin (Ax) (81.78%), cefalexin (85.42%), co-trimoxazole (50.78%), sulfafurazole (69.38%), tetracycline (65.89%), and trimethoprim (TR) (51.94%).
