The Interstellar Boundary Explorer (IBEX) has been observing the outer heliosphere and its interactions with the very local interstellar medium (VLISM) via measurements of energetic neutral atoms (ENAs) for over 14 yr. We discovered the IBEX Ribbon—a structure completely unanticipated by any prior theory or model—that almost certainly resides beyond the heliopause in the VLISM. We also characterized the other major source of heliospheric ENAs, the globally distributed flux (GDF), produced largely in the heliosheath between the termination shock and heliopause. In this study, we make three major new contributions. First, we validate, provide, and analyze the most recent 3 yr of IBEX-Hi (0.5–6 keV FWHM) data (2020–2022) for the first time. Second, we link these observations to the prior 11 yr of observations, exploring long-term variations. Finally, we provide the first IBEX team-validated Ribbon/GDF separation scheme and separated maps. Because of the uncertainty in separating different line-of-sight integrated sources, we provide not just best guess (median) maps, but also maps with upper and lower reasonable values of Ribbon and GDF fluxes, along with bounding fluxes that add the uncertainties to the upper and lower values. This allows theories and models to be compared with a range of possible values that the IBEX team believes are consistent with data. These observations, along with the reanalysis of the prior 11 yr of IBEX-Hi data, provide new insights and even further develop our detailed understanding of the heliosphere’s interaction with the local interstellar medium unlocked by IBEX.

Taxonomy underpins biological research because names are needed for comparative analysis, conservation status, and public communication. Despite this, many species remain undescribed and are therefore vulnerable and unprotected, particularly in the tropics. Neotropical Staphylinidae beetles (Coleoptera) are among the insect groups most likely to contain significant unknown and/or cryptic diversity. Here we used an integrative taxonomic framework to conduct a preliminary review of one particularly diverse genus of Staphylinidae (Anotylus) in Área de Conservaciόn Guanacaste, northwestern Costa Rica. We began by DNA barcoding novel collections and using Barcode Index Numbers (BINs) as an estimate of taxonomic diversity; we found 18 provisional new species. We augmented this genetic analysis with a morphometric analysis of adult morphological characters and found that we could differentiate most provisional species by external morphology as well as by elevation of collection. All the most abundant species could be differentiated from each other by differences in body size. One BIN included slight (~1%) genetic variation that corresponded with some morphological differentiation suggesting the existence of two species within a DNA BIN. Our results support the efficacy of DNA barcoding collections of tropical insects as an effective biodiversity estimator, one that can be used as a primer for integrative taxonomic studies using BINs as species hypotheses.

Three new genera are described: Michener (Proteropinae), Bioalfa (Rogadinae), and Hermosomastax (Rogadinae). Keys are given for the New World genera of the following braconid subfamilies: Agathidinae, Braconinae, Cheloninae, Homolobinae, Hormiinae, Ichneutinae, Macrocentrinae, Orgilinae, Proteropinae, Rhysipolinae, and Rogadinae. In these subfamilies 416 species are described or redescribed. Most of the species have been reared and all but 13 are new to science. A consensus sequence of the COI barcodes possessed by each species is employed to diagnose the species, and this approach is justified in the introduction. Most descriptions consist of a lateral or dorsal image of the holotype, a diagnostic COI consensus barcode, the Barcode Index Number (BIN) code with a link to the Barcode of Life Database (BOLD), and the holotype specimen information required by the International Code of Zoological Nomenclature. Two new Costa Rican species of the braconid parasitoid wasp subfamily Cardiochilinae, Heteropteron kidonoi Dabek & Whitfield and Heteropteron hasegawai Dabek & Whitfield, are described and illustrated from dry forest in the Area de Conservacion Guanacastae, along with data on rearing from their hosts. Heteropteron kidonoi is a solitary endoparasitoid of Stenoma cathosiota (Lepidoptera: Depressariidae) on Roupala montana (Proteaceae), while H. hasegawai is a solitary endoparasitoid of Carthara abrupta (Lepidoptera: Pyralidae) on the same host plant, but typically at slightly higher elevation localities. Diagnostic characters are provided to distinguish these two new species from each other, and also from the three previously decsribed species of Heteropteron. Heteropteron kidonoi and H. hasegawai are the first species of Heteropteron to have any host data, and also are the first to be reported in Costa Rica.

The descriptive taxonomic study reported here is focused on Glyptapanteles, a species-rich genus of hymenopteran parasitoid wasps. The species were found within the framework of two independent long-term Neotropical caterpillar rearing projects: northwestern Costa Rica (Área de Conservación Guanacaste, ACG) and eastern Andes, Ecuador (centered on Yanayacu Biological Station, YBS). One hundred thirty-six new species of Glyptapanteles Ashmead are described and all of them are authored by Arias-Penna. None of them was recorded in both countries; thus, 78 are from Costa Rica and the remaining 58 from Ecuador. Before this revision, the number of Neotropical described Glyptapanteles did not reach double digits. Reasonable boundaries among species were generated by integrating three datasets: Cytochrome Oxidase I (COI) gene sequencing data, natural history (host records), and external morphological characters. Each species description is accompanied by images and known geographical distribution. Characteristics such as shape, ornamentation, and location of spun Glyptapanteles cocoons were imaged as well. Host-parasitoid associations and food plants are also here published for the first time. A total of 88 species within 84 genera in 15 Lepidoptera families was encountered as hosts in the field. With respect to food plants, these wild-caught parasitized caterpillars were reared on leaves of 147 species within 118 genera in 60 families. The majority of Glyptapanteles species appeared to be relatively specialized on one family of Lepidoptera or even on some much lower level of taxonomic refinement. Those herbivores in turn are highly food-plant specialized, and once caterpillars were collected, early instars (1–3) yielded more parasitoids than later instars. Glyptapanteles jimmilleri Arias-Penna, sp. nov. is the first egg-larval parasitoid recorded within the genus, though there may be many more since such natural history requires a more focused collection of eggs. The rate of hyperparasitoidism within the genus was approximately 4% and was represented by Mesochorus spp. (Ichneumonidae). A single case of multiparasitoidism was reported, Copidosoma floridanum Ashmead (Encyrtidae) and Glyptapanteles ilarisaaksjarvi Arias-Penna, sp. nov. both parasitoid species emerged from the caterpillar of Noctuidae: Condica cupienta (Cramer). Bodyguard behavior was observed in two Glyptapanteles species: G. howelldalyi Arias-Penna, sp. nov. and G. paulhansoni Arias-Penna, sp. nov. A dichotomous key for all the new species is provided. The numerous species described here, and an equal number already reared but not formally described, signal a far greater Glyptapanteles species richness in the Neotropics than suggested by the few described previously.

Here we elucidate and justify a DNA barcode approach to insect species description that can be applied to name tens of thousands of species of Ichneumonoidea and many other species-rich taxa. Each description consists of a lateral habitus image of the specimen, a COI barcode diagnosis, and the holotype specimen information required by the International Code of Zoological Nomenclature. We believe this approach, or a slight modification of it, will be useful for many other underdescribed hyperdiverse taxa, especially in the tropics. Due to the extreme species-richness of the Ichneumonoidea, the very low percentage of described species, and the lack of detailed biological information for most described species, the standard taxonomic approach is inefficient and overwhelmingly time consuming. A DNA barcode-based approach to initial description will provide a solid foundation of species hypotheses from which more comprehensive descriptions can be developed as other data, time, and budgets permit. Here we elucidate this view and detailed methodology that can generally be applied to species-rich underdescribed taxa. A real example is given by describing species in two genera, Hemichoma and Zelomorpha, reared from the Área de Conservación Guanacaste in northwestern Costa Rica. The generic type species Zelomorpha arizonensis is given a DNA barcode diagnosis and the following new species are described: Zelomorpha angelsolisi, Zelomorpha bobandersoni, Zelomorpha danjohnsoni, Zelomorpha donwindsori, Zelomorpha effugia, Zelomorpha johnchemsaki, Zelomorpha kellyanneae, Zelomorpha larrykirkendalli, Zelomorpha mariyavladmirovnae, Zelomorpha mikeiviei, Zelomorpha myricagaleae, Zelomorpha noahjaneae, Zelomorpha paulgoldsteini, Zelomorpha terryerwini, Zelomorpha willsflowersi, Hemichoma donwhiteheadi, Hemichoma frankhovorei, and Hemichoma johnkingsolveri.

Lophomyra Schaus, 1911 (Noctuidae) is the smaller of two noctuid genera originally described by Schaus that include species recently associated with ferns (Pteridophyta), in this case Polypodiaceae, as larval food plants. Following an examination of type material and reared specimens accompanied by DNA barcode data, Lophomyra is revised to include L. tacita Schaus, 1911, L. santista (Jones, 1914), and L. commixta (Schaus, 1914), comb. n., the last of which is transferred from Chytonidia Schaus, 1914 (= Leucosigma Druce, 1908). Lophomyra is characterized based on adult and larval morphology, especially that of the male genitalia. Structures associated with the valvae are discussed with reference to dissected and in situ images. Larvae of L. commixta and L. tacita are described from images, and the recorded food plants of both species are discussed in the context of known New World noctuid pteridivores.

Chytonidia Schaus, 1914, is one of two noctuine genera originally described by Schaus that includes species recently found to feed on fern foliage (Pteridophyta) as larvae. By examining museum specimens, including type material and reared specimens accompanied by DNA barcode data, Chytonidia Schaus, 1914, syn. n. is synonymized with Leucosigma Druce, 1908, all currently recognized species are re-described, including males of three species described from female holotypes, and three new species are described: Leucosigma solisae Goldstein, sp. n., Leucosigma poolei Goldstein, sp. n., and L. schausi Goldstein, sp. n. Images of adults and, where available, larvae as well as dissected genitalia are presented, with a key to adults.

Abstract Background Although high-throughput sequencers (HTS) have largely displaced their Sanger counterparts, the short read lengths and high error rates of most platforms constrain their utility for amplicon sequencing. The present study tests the capacity of single molecule, real-time (SMRT) sequencing implemented on the SEQUEL platform to overcome these limitations, employing 658 bp amplicons of the mitochondrial cytochrome c oxidase I gene as a model system. Results By examining templates from more than 5000 species and 20,000 specimens, the performance of SMRT sequencing was tested with amplicons showing wide variation in GC composition and varied sequence attributes. SMRT and Sanger sequences were very similar, but SMRT sequencing provided more complete coverage, especially for amplicons with homopolymer tracts. Because it can characterize amplicon pools from 10,000 DNA extracts in a single run, the SEQUEL can reduce greatly reduce sequencing costs in comparison to first (Sanger) and second generation platforms (Illumina, Ion). Conclusions SMRT analysis generates high-fidelity sequences from amplicons with varying GC content and is resilient to homopolymer tracts. Analytical costs are low, substantially less than those for first or second generation sequencers. When implemented on the SEQUEL platform, SMRT analysis enables massive amplicon characterization because each instrument can recover sequences from more than 5 million DNA extracts a year.

Thirty two new species of Lytopylus (Agathidinae) are described with image plates for each species: Lytopylus alejandromasisi sp. n., Lytopylus alfredomainieri sp. n., Lytopylus anamariamongeae sp. n., Lytopylus angelagonzalezae sp. n., Lytopylus cesarmorai sp. n., Lytopylus eddysanchezi sp. n., Lytopylus eliethcantillanoae sp. n., Lytopylus ericchapmani sp. n., Lytopylus gahyunae sp. n., Lytopylus gisukae sp. n., Lytopylus guillermopereirai sp. n., Lytopylus gustavoindunii sp. n., Lytopylus hartmanguidoi sp. n., Lytopylus hernanbravoi sp. n., Lytopylus hokwoni sp. n., Lytopylus ivanniasandovalae sp. n., Lytopylus johanvalerioi sp. n., Lytopylus josecortesi sp. n., Lytopylus luisgaritai sp. n., Lytopylus mariamartachavarriae sp. n., Lytopylus miguelviquezi sp. n., Lytopylus motohasegawai sp. n., Lytopylus okchunae sp. n., Lytopylus pablocobbi sp. n., Lytopylus robertofernandezi sp. n., Lytopylus rogerblancoi sp. n., Lytopylus salvadorlopezi sp. n., Lytopylus sangyeoni sp. n., Lytopylus sarahmeierottoae sp. n., Lytopylus sergiobermudezi sp. n., Lytopylus sigifredomarini sp. n., and Lytopylus youngcheae sp. n. A dichotomous key and a link to an electronic, interactive key are included. All specimens were reared from Lepidoptera larvae collected in Area de Conservación Guanacaste (ACG) and all are associated with ecological information including host caterpillar, collection date, eclosion date, caterpillar food plant, and locality. Neighbor-joining and maximum likelihood analyses of the barcode region of the mitochondrial cytochrome c oxidase subunit I gene (COI DNA barcode) were conducted to aid in species delimitation.

90 species of Euplectrus are treated: 55 newly described, all from Area de Conservación Guanacaste (ACG), and 35 previously described species, of which 20 occur in ACG. Three of the previously described species (E. brasiliensis Ashmead, E. hircinus (Say), E. ronnai (Brèthes)) have unknown status, owing to missing or severely damaged type material. The new species, all authored by C. Hansson, are: Euplectrus alejandrovalerioi, E. alexsmithi, E. alvarowillei, E. andybennetti, E. andydeansi, E. annettewalkerae, E. billbrowni, E. bobwhartoni, E. carlosarmientoi, E. carlrettenmeyeri, E. charlesmicheneri, E. charlesporteri, E. chrisdarlingi, E. chrisgrinteri, E. corriemoreauae, E. daveroubiki, E. davesmithi, E. davidwahli, E. dianariasae, E. donquickei, E. eowilsoni, E. garygibsoni, E. gavinbroadi, E. gerarddelvarei, E. henrytownesi, E. howelldalyi, E. hugokonsi, E. iangauldi, E. jacklonginoi, E. jesusugaldei, E. jimwhitfieldi, E. jjrodriguezae, E. johnheratyi, E. johnlasallei, E. johnnoyesi, E. josefernandezi, E. lubomirmasneri, E. markshawi, E. mikegatesi, E. mikeschauffi, E. mikesharkeyi, E. ninazitaniae, E. pammitchellae, E. paulhansoni, E. paulheberti, E. paulhurdi, E. philwardi, E. robbinthorpi, E. ronaldzunigai, E. roysnellingi, E. scottshawi, E. sondrawardae, E. sydneycameronae, E. victoriapookae, E. wonyoungchoi. The species are described or redescribed, and thoroughly and uniformly illustrated, and included in two identification keys, one for females and one for males. Lectotypes are designated for eight species: Euplectrus catocalae Howard (♂), E. junctus Gahan (♀), E. leucotrophis Howard (♂), E. marginatus Ashmead (♀), E. pachyscaphus Girault (♀), E. platyhypenae Howard (♂), E. semimarginatus Girault (♀), Heteroscapus ronnai Brèthes (♂). One synonym is established: Euplectrus walteri Schauff is a junior synonym of E. testaceipes (Cameron). Brief image notes and host records are provided on the natural history of the wasps as well as the details of their morphology. Hosts are known for 74 Euplectrus species.

The Neotropical genus of parasitoid wasps Sendaphne (Hymenoptera, Braconidae, Microgastrinae) is revised and the following six new species are described, all authored by Fernández-Triana and Whitfield: anitae, bennetti, broadi, dianariaspennae, penteadodiasae, and rogerblancoi. The greatest species richness is found in northern South America, but the genus extends north to 23° N in Mexico. Most species have been collected in rainforest below altitudes of 900 m, with only a few species found in cloud forests up to 1900 m. Nothing is known of the host caterpillars for these parasitoid wasps.

We discuss 45 Costa Rican species of Ethmia Hübner, 1819, including 23 previously described: E. delliella (Fernald), E. bittenella (Busck), E. festiva Busck, E. scythropa Walsingham, E. perpulchra Walsingham, E. terpnota Walsingham, E. elutella Busck, E. janzeni Powell, E. ungulatella Busck, E. exornata (Zeller), E. phylacis Walsingham, E. mnesicosma Meyrick, E. chemsaki Powell, E. baliostola Walsingham, E. duckworthi Powell, E. sandra Powell, E. nigritaenia Powell, E. catapeltica Meyrick, E. lichyi Powell, E. transversella Busck, E. similatella Busck, E. hammella Busck, E. linda Busck, and 22 new species: E. blaineorum, E. millerorum, E. dianemillerae, E. adrianforsythi, E. stephenrumseyi, E. berndkerni, E. dimauraorum, E. billalleni, E. ehakernae, E. helenmillerae, E. johnpringlei, E. laphamorum, E. petersterlingi, E. lesliesaulae, E. turnerorum, E. normgershenzi, E. nicholsonorum, E. hendersonorum, E. randyjonesi, E. randycurtisi, E. miriamschulmanae and E. tilneyorum. We illustrate all species and their male and female genitalia, along with distribution maps of Costa Rican localities. Immature stages are illustrated for 11 species, and food plants are listed when known. Gesneriaceae is added as a new food plant family record for Ethmia. CO1 nucleotide sequences (“DNA barcodes”) were obtained for 41 of the species.

Bacillus thuringiensis (Bt) synthesizes crystalline inclusions that are toxic to caterpillars (Lepidoptera) and other orders of invertebrates. Materials associated with 37 caterpillars from 16 species, collected while feeding on 15 different species of host plants in dry, cloud and rain forests located in the Área de Conservación Guanacaste in northwestern Costa Rica, were examined for the presence of Bt. From a total of 101 derived samples, 25 Bt isolates were cultured: 56% from host plant leaves, 8% from caterpillar guts and 36% from caterpillar fecal pellets. Bt was isolated from at least one sample in 38% of the systems constituted by the food plant, gut and fecal pellets corresponding to a single caterpillar. Four different morphologies of crystalline inclusions were observed, with bipyramidal and irregular crystal morphologies being the most prevalent. Rev. Biol. Trop. 54(2): 265-271. Su presencia fue examinada en materiales asociados a 37 orugas de mariposas de 16 especies, las cuales fueron colectadas mientras se alimentaban en 15 especies diferentes de plantas hospederas en bosques secos, nubosos y húmedos localizados dentro del Área de Conservación Guanacaste (ACG) en el noroeste de Costa Rica. A partir de un total de 101 muestras se obtuvo 25 aislamientos de Bt: 56% a partir de material foliar de las plantas hospederas, 8% a partir del contenido intestinal de las larvas y 36% a partir de sus excrementos. Esta bacteria fue cultivada a partir de al menos uno de los 3 diferentes tipos de muestra asociados a una oruga particular (planta hospedera, intestino, excremento) en 38% de los casos posibles. En la colección de aislamientos obtenida se observaron cuatro morfologías de inclusiones cristalinas, siendo aquellas bipiramidales e irregulares las más prevalentes.

Desde 1994 hasta 1997, 1800 muestras de artrópodos correspondientes a 642 especies distribuidas en 21 órdenes y 95 familias fueron recolectadas en 21 localidades protegidas del Área de Conservación Guanacaste, para ser estudiadas como fuente potencial de nuevos medicamentos. Las localidades con más recolectas fueron Santa María (231 spp.; 421 muestras), Santa Rosa (110 spp.; 172 muestras), Cacao (98 spp.; 203 muestras) y Pitilla (67 spp.; 79 muestras), siendo la mayor cantidad recolectadas durante la estación lluviosa. El 49.72% de las muestras recolectadas correspondió al orden Lepidoptera, 15.75% a Coleoptera, 13.33% a Hymenoptera, 11.43% a Orthoptera, 6.75% a Hemiptera, 3.20% a Homoptera y 7.89% a otros grupos. Además, un 54.26% de las muestras correspondía a individuos adultos, 19.90% a larvas, 11.20% a estiércol, 6.46% a pupas, 6.12% a prepupas, 2.07% a ninfas y 3.74% a otros estadios. Un 44.38% de las muestras se recolectó mediante recolecta manual, seguido por crianza de insectos (25.73%), trampa de luz (18.80%), diferentes tipos de redes (10.52%) y otros métodos (0.16%). La alta diversidad de los trópicos y la gran abundancia de artrópodos hace de este grupo una fuente potencial de productos farmacéuticos. Sin embargo, se debe evitar la recolecta intensiva en la misma localidad durante años consecutivos (sobre todo hembras), minimizar la interferencia en procesos vitales de los organismos y la dinámica de sus poblaciones. Además se debe recolectar la menor cantidad posible de individuos y escoger la época del año adecuada, para lograr el reestablecimiento natural de las poblacionesThis study describes the results and collection practices for obtaining arthropod samples to be studied as potential sources of new medicines in a bioprospecting effort. From 1994 to 1998, 1800 arthropod samples of 6-10 g were collected in 21 sites of the Área de Conservación Guancaste (A.C.G) in Northwestern Costa Rica. The samples corresponded to 642 species distributed in 21 orders and 95 families. Most of the collections were obtained in the rainy season and in the tropical rainforest and dry forest of the ACG. Samples were obtained from a diversity of arthropod orders: 49.72% of the samples collected corresponded to Lepidoptera, 15.75% to Coleoptera, 13.33% to Hymenoptera, 11.43% to Orthoptera, 6.75% to Hemiptera, 3.20% to Homoptera and 7.89% to other groups. Different life stages per arthropod species were obtained in most samples, 54.26% of them were adults, 19.90% corresponded to larvae, 6.46% to pupae, 6.12% to pre-pupae, 2.07% to nymphs and 3.74% to other stages. Other materials associated to insects like frass represented 11.20% of the samples collected. Several collecting methods were explored, based on the possibility of accessing the necessary amount of material causing the less impact. Most of the samples were obtained by manual collection (44.38%), followed by insects breeding (25.73%), light traps (18.80%), different types of nets (10.52%) and other methods (0.16%). In general, collecting methods and practices excluded the use of solvents, mixing different species or life stages in the same bag, which might have introduced undesirable effects in the screening systems for new compounds. Based on the possibility of finding new chemicals in similar samples associated to one arthropod species, the collecting strategy included the generation of several samples from same species, separated according to differences in life stages, collecting sites, ecosystems, seasons, feeding materials or behavioral aspects. This strategy allowed the generation a larger number of samples submitted to bioassays in different areas of pharmaceutical research

We used classical culture techniques to explore gut bacteria and changes associated with dietary change in the highly polyphagous, tropical caterpillar Automeris zugana (Saturniidae). Fifty-five third instar wild-caught sibs feeding on Annona purpurea (Annonaceae) in the Área de Conservación Guanacaste (ACG) in northwestern Costa Rica were divided into eight groups. Each of seven groups was reared to the ultimate instar on another species of food plant normally used by A. zugana. Some pupae were also analyzed for the presence of bacteria. Aerobic bacterial cultures were obtained from all 33 caterpillar guts and the eight pupae inventoried. There was no clear pattern in species composition of cultivated bacteria among the eight diets, and each caterpillar on a given food plant carried only a small fraction of the total set of species isolated from the set of caterpillars feeding on that food plant. Taken as a whole, the larvae and pupae contained 22 species of cultivable bacteria in 12 genera. Enterobacter, present in 81.8% of the samples, was the genus most frequently isolated from the caterpillars, followed by Micrococcus and Bacillus. Bacillus thuringiensis was isolated from 30.3% of the dissected caterpillars, but found in caterpillars feeding on only half of the species of food plantsUsamos técnicas de cultivo clásicas para explorar las bacterias del intestino y los cambios asociados con el cambio dietético en la, altamente polífaga, oruga tropical Automeris zugana (Saturniidae). Se capturaron 55 individuos hermanos, silvestres, de tercer estadio, alimentándose de Annona purpurea (Annonaceae) en el Área de Conservación Guanacaste (ACG) en el noroeste de Costa Rica y se dividieron en ocho grupos. Siete grupos fueron criados hasta el último estadio en otra especie de planta a la normalmente usada por A. zugana. Algunas pupas también fueron analizadas para ver la presencia de bacterias. Se obtuvieron cultivos de bacterias aeróbicas de los 33 intestinos de las orugas y de las ocho pupas muestreadas. No hubo un patrón claro en la composición de especies de las bacterias cultivadas entre las ocho dietas y cada oruga en una planta dada representó solo una pequeña fracción del conjunto total de especies aisladas de orugas alimentándose de esa planta. Tomándolas como un solo grupo, las larvas y pupas tuvieron 22 especies de bacterias cultivables en 12 géneros. Enterobacter, presente en 81.8% de las muestras, fue el género aislado de las orugas con más frecuencia, seguido por Micrococcus y Bacillus. Bacillus thuringiensis fue aislado de 30.3% de las orugas disectadas, pero encontrado en orugas alimentándose de solo la mitad de las especies de plantas

Wolbachia is a genus of bacterial endosymbionts that impacts the breeding systems of their hosts. Wolbachia can confuse the patterns of mitochondrial variation, including DNA barcodes, because it influences the pathways through which mitochondria are inherited. We examined the extent to which these endosymbionts are detected in routine DNA barcoding, assessed their impact upon the insect sequence divergence and identification accuracy, and considered the variation present in Wolbachia COI. Using both standard PCR assays (Wolbachia surface coding protein--wsp), and bacterial COI fragments we found evidence of Wolbachia in insect total genomic extracts created for DNA barcoding library construction. When >2 million insect COI trace files were examined on the Barcode of Life Datasystem (BOLD) Wolbachia COI was present in 0.16% of the cases. It is possible to generate Wolbachia COI using standard insect primers; however, that amplicon was never confused with the COI of the host. Wolbachia alleles recovered were predominantly Supergroup A and were broadly distributed geographically and phylogenetically. We conclude that the presence of the Wolbachia DNA in total genomic extracts made from insects is unlikely to compromise the accuracy of the DNA barcode library; in fact, the ability to query this DNA library (the database and the extracts) for endosymbionts is one of the ancillary benefits of such a large scale endeavor--which we provide several examples. It is our conclusion that regular assays for Wolbachia presence and type can, and should, be adopted by large scale insect barcoding initiatives. While COI is one of the five multi-locus sequence typing (MLST) genes used for categorizing Wolbachia, there is limited overlap with the eukaryotic DNA barcode region.

BACKGROUND: The many components of conservation through biodiversity development of a large complex tropical wildland, Area de Conservacion Guanacaste (ACG), thrive on knowing what is its biodiversity and natural history. For 32 years a growing team of Costa Rican parataxonomists has conducted biodiversity inventory of ACG caterpillars, their food plants, and their parasitoids. In 2003, DNA barcoding was added to the inventory process. METHODOLOGY/PRINCIPAL FINDINGS: We describe some of the salient consequences for the parataxonomists of barcoding becoming part of a field biodiversity inventory process that has centuries of tradition. From the barcoding results, the parataxonomists, as well as other downstream users, gain a more fine-scale and greater understanding of the specimens they find, rear, photograph, database and deliver. The parataxonomists also need to adjust to collecting more specimens of what appear to be the "same species"--cryptic species that cannot be distinguished by eye or even food plant alone--while having to work with the name changes and taxonomic uncertainty that comes with discovering that what looked like one species may be many. CONCLUSIONS/SIGNIFICANCE: These career parataxonomists, despite their lack of formal higher education, have proven very capable of absorbing and working around the additional complexity and requirements for accuracy and detail that are generated by adding barcoding to the field base of the ACG inventory. In the process, they have also gained a greater understanding of the fine details of phylogeny, relatedness, evolution, and species-packing in their own tropical complex ecosytems. There is no reason to view DNA barcoding as incompatible in any way with tropical biodiversity inventory as conducted by parataxonomists. Their year-round on-site inventory effort lends itself well to the sampling patterns and sample sizes needed to build a thorough barcode library. Furthermore, the biological understanding that comes with barcoding increases the scientific penetrance of biodiversity information, DNA understanding, evolution, and ecology into the communities in which the parataxonomists and their families are resident.

DNA barcoding is an effective approach for species identification and for discovery of new and/or cryptic species. Sanger sequencing technology is the method of choice for obtaining standard 650 bp cytochrome c oxidase subunit I (COI) barcodes. However, DNA degradation/fragmentation makes it difficult to obtain a full-length barcode from old specimens. Mini-barcodes of 130 bp from the standard barcode region have been shown to be effective for accurate identification in many animal groups and may be readily obtained from museum samples. Here we demonstrate the application of an alternative sequencing technology, the four-enzymes single-specimen pyrosequencing, in rapid, cost-effective mini-barcode analysis. We were able to generate sequences of up to 100 bp from mini-barcode fragments of COI in 135 fresh and 50 old Lepidoptera specimens (ranging from 53-97 year-old). The sequences obtained using pyrosequencing were of high quality and we were able to robustly match all the tested pyro-sequenced samples to their respective Sanger-sequenced standard barcode sequences, where available. Simplicity of the protocol and instrumentation coupled with higher speed and lower cost per sequence than Sanger sequencing makes this approach potentially useful in efforts to link standard barcode sequences from unidentified specimens to known museum specimens with only short DNA fragments.

BACKGROUND: An intense, 30-year, ongoing biodiversity inventory of Lepidoptera, together with their food plants and parasitoids, is centered on the rearing of wild-caught caterpillars in the 120,000 terrestrial hectares of dry, rain, and cloud forest of Area de Conservacion Guanacaste (ACG) in northwestern Costa Rica. Since 2003, DNA barcoding of all species has aided their identification and discovery. We summarize the process and results for a large set of the species of two speciose subfamilies of ACG skipper butterflies (Hesperiidae) and emphasize the effectiveness of barcoding these species (which are often difficult and time-consuming to identify). METHODOLOGY/PRINCIPAL FINDINGS: Adults are DNA barcoded by the Biodiversity Institute of Ontario, Guelph, Canada; and they are identified by correlating the resulting COI barcode information with more traditional information such as food plant, facies, genitalia, microlocation within ACG, caterpillar traits, etc. This process has found about 303 morphologically defined species of eudamine and pyrgine Hesperiidae breeding in ACG (about 25% of the ACG butterfly fauna) and another 44 units indicated by distinct barcodes (n = 9,094), which may be additional species and therefore may represent as much as a 13% increase. All but the members of one complex can be identified by their DNA barcodes. CONCLUSIONS/SIGNIFICANCE: Addition of DNA barcoding to the methodology greatly improved the inventory, both through faster (hence cheaper) accurate identification of the species that are distinguishable without barcoding, as well as those that require it, and through the revelation of species "hidden" within what have long been viewed as single species. Barcoding increased the recognition of species-level specialization. It would be no more appropriate to ignore barcode data in a species inventory than it would be to ignore adult genitalia variation or caterpillar ecology.
