Abstract

Recent biomarker innovations hold potential for transforming diagnosis, prognostic modeling, and precision therapeutic targeting of traumatic brain injury (TBI). However, many biomarkers, including brain imaging, genomics, and proteomics, involve vast quantities of high-throughput and high-content data. Management, curation, analysis, and evidence synthesis of these data are not trivial tasks. In this review, we discuss data management concepts and statistical and data sharing strategies when dealing with biomarker data in the context of TBI research. We propose that application of biomarkers involves three distinct steps-discovery, evaluation, and evidence synthesis. First, complex/big data has to be reduced to useful data elements at the stage of biomarker discovery. Second, inferential statistical approaches must be applied to these biomarker data elements for assessment of biomarker clinical utility and validity. Last, synthesis of relevant research is required to support practice guidelines and enable health decisions informed by the highest quality, up-to-date evidence available. We focus our discussion around recent experiences from the International Traumatic Brain Injury Research (InTBIR) initiative, with a specific focus on four major clinical projects (Transforming Research and Clinical Knowledge in TBI, Collaborative European NeuroTrauma Effectiveness Research in TBI, Collaborative Research on Acute Traumatic Brain Injury in Intensive Care Medicine in Europe, and Approaches and Decisions in Acute Pediatric TBI Trial), which are currently enrolling subjects in North America and Europe. We discuss common data elements, data collection efforts, data-sharing opportunities, and challenges, as well as examine the statistical techniques required to realize successful adoption and use of biomarkers in the clinic as a foundation for precision medicine in TBI.

20. Detection of polystyrene sphere translocations using resizable elastomeric nanopores

Abstract

Resizable elastomeric nanopores have been used to measure pulses of ionic current caused by carboxylated polystyrene spheres of diameter 200 nm and 800 nm. The nanopores represent a novel technology which enables nanoscale resizing of a pore by macroscopic actuation of an elastomeric membrane. Three different pores were employed with variable applied strain, transmembrane potential, particle concentration and sphere radius. Theory describing current pulse magnitude has been extended to conical pore geometry. A consistent method for interpretation of data close to the noise threshold has been introduced, and experimental data has been used to compare several methods for efficient, non-destructive calculation of pore dimensions. The most effective models emphasize the absolute pulse size, which is predominantly determined by the opening radius at the narrowest part of the roughly conical pores, rather than the profile along the entire pore length. Experiments were carried out in a regime for which both electro-osmotic and electrophoretic transport are significant.

Abstract

Current diagnostic methods for Johne's disease in cattle allow reliable detection of infections with Mycobacterium avium ssp. paratuberculosis (MAP) not before animals are 2 years of age. Applying a flow cytometry-based approach (FCA) to quantify a MAP-specific interferon-gamma (IFN-γ) response in T cell subsets, the present study sought to monitor the kinetics of the cell-mediated immune response in experimentally infected calves. Six MAP-negative calves and six calves, orally inoculated with MAP at 10 days of age, were sampled every 4 weeks for 52 weeks post-inoculation (wpi). Peripheral blood mononuclear cells (PBMC) were stimulated with either purified protein derivatives (PPD) or whole cell sonicates derived from MAP (WCSj), M. avium ssp. avium or M. phlei for 6 days followed by labeling of intracellular IFN-γ in CD4+ and CD8+ T cells. No antigen-specific IFN-γ production was detectable in CD8+ cells throughout and the responses of CD4+ cells of MAP-infected and control calves were similar up to 12 wpi. However, the mean fluorescence intensity (MFI) for the detection of IFN-γ in CD4+ cells after WCSj antigen stimulation allowed for a differentiation of animal groups from 16 wpi onwards. This approach had a superior sensitivity (87.8%) and specificity (86.8%) to detect infected animals from 16 wpi onwards, i.e., in an early infection stage, as compared to the IFN-γ release assay (IGRA). Quantification of specific IFN-γ production at the level of individual CD4+ cells may serve, therefore, as a valuable tool to identify MAP-infected juvenile cattle. [ABSTRACT FROM AUTHOR]

36. Effects of Atlantification and changing sea‐ice dynamics on zooplankton community structure and carbon flux between 2000 and 2016 in the eastern Fram Strait

Abstract

The collection of zooplankton swimmers and sinkers in time‐series sediment traps provides unique insight into year‐round and interannual trends in zooplankton population dynamics. These samples are particularly valuable in remote and difficult to access areas such as the Arctic Ocean, where samples from the ice‐covered season are rare. In the present study, we investigated zooplankton composition based on swimmers and sinkers collected by sediment traps at water depths of 180–280, 800–1320, and 2320–2550 m, over a period of 16 yr (2000–2016) at the Long‐Term Ecological Research observatory HAUSGARTEN located in the eastern Fram Strait (79°N, 4°E). The time‐series data showed seasonal and interannual trends within the dominant zooplankton groups including copepoda, foraminifera, ostracoda, amphipoda, pteropoda, and chaetognatha. Amphipoda and copepoda dominated the abundance of swimmers while pteropoda and foraminifera were the most important sinkers. Although the seasonal occurrence of these groups was relatively consistent between years, there were notable interannual variations in abundance, suggesting the influence of various environmental conditions such as sea‐ice dynamic and lateral advection of water masses, for example, meltwater and Atlantic water. Statistical analyses revealed a correlation between the Arctic dipole climatic index and sea‐ice dynamics (i.e., ice coverage and concentration), as well as the importance of the distance from the ice edge on swimmer composition patterns and carbon export.
