CIBER (Center for Biomedical Network Research; Centro de Investigación Biomédica En Red) is a public national consortium created in 2006 under the umbrella of the Spanish National Institute of Health Carlos III (ISCIII). This innovative research structure comprises 11 different specific areas dedicated to the main public health priorities in the National Health System. CIBERER, the thematic area of CIBER focused on rare diseases (RDs) currently consists of 75 research groups belonging to universities, research centers, and hospitals of the entire country. CIBERER's mission is to be a center prioritizing and favoring collaboration and cooperation between biomedical and clinical research groups, with special emphasis on the aspects of genetic, molecular, biochemical, and cellular research of RDs. This research is the basis for providing new tools for the diagnosis and therapy of low-prevalence diseases, in line with the International Rare Diseases Research Consortium (IRDiRC) objectives, thus favoring translational research between the scientific environment of the laboratory and the clinical setting of health centers. In this article, we intend to review CIBERER's 15-year journey and summarize the main results obtained in terms of internationalization, scientific production, contributions toward the discovery of new therapies and novel genes associated to diseases, cooperation with patients' associations and many other topics related to RD research.

Lafora disease (LD) is a fatal rare type of progressive myoclonus epilepsy that appears during early adolescence. The disease is caused by mutations in EPM2A or EPM2B genes, which encode laforin, a glucan phosphatase, and malin, an E3-ubiquitin ligase, respectively. Although the exact roles of laforin and malin are still not well understood, it is known that they work as a complex in which laforin recruits targets that will be ubiquitinated by malin. Recently, we suggested that the type of epilepsy that accompanies LD could be due to deficiencies in the function of the astrocytic glutamate transporter GLT-1. We described that astrocytes from LD mouse models presented decreased levels of GLT-1 at the plasma membrane, leading to increased levels of glutamate in the brain parenchyma. In this work, we present evidence indicating that in the absence of a functional laforin/malin complex (as in LD cellular models) there is an alteration in the ubiquitination of GLT-1, which could be the cause of the reduction in the levels of GLT-1 at the plasma membrane. On the contrary, overexpression of the laforin/malin complex promotes the retention of GLT-1 at the plasma membrane. This retention may be due to the direct ubiquitination of GLT-1 and/or to an opposite effect of this complex on the dynamics of the Nedd4.2-mediated endocytosis of the transporter. This work, therefore, presents new pieces of evidence on the regulation of GLT-1 by the laforin/malin complex, highlighting its value as a therapeutic target for the amelioration of the type of epilepsy that accompanies LD.

[ES] La enfermedad de Lafora es una epilepsia mioclónica progresiva causada por mutaciones en los genes que codifican las proteínas laforina o malina. La mutación de una de las dos proteínas impide que se forme el complejo funcional y a causa de ello se acumulan inclusiones de glucógeno insoluble, conocidas como cuerpos de Lafora. Otra patología descrita en los pacientes de Lafora es la alteración en la homeostasis del transportador de glutamato EAAT2 de astrocitos (GLT1 en ratón). Este transportador se localiza normalmente en la membrana plasmática de estas células para internalizar el glutamato (neurotransmisor) y detener la señal de excitación de las neuronas. Esta localización está altamente regulada por procesos de ubiquitinación/desubiquitinación. En la ruta de internalización del transportador de glutamato interviene una E3 ubiquitín ligasa denominada Nedd4.2. Para poder unirse al transportador de glutamato y ubiquitinarlo, precisa de proteínas adaptadoras denominadas arrestinas. Todavía no hay mucha información sobre la función de las arrestinas pero se han descrito como adaptadores para las E3 ubiquitín ligasas en la endocitosis de transportadores de membrana. En la enfermedad de Lafora, el nivel de transportadores de glutamato en la membrana plasmática de los astrocitos está reducido y esto podría ser debido a un cambio en el patrón de ubiquitinación del transportador por parte del complejo laforina-malina. Este trabajo estudia uno de los componentes de la familia de las ¿-arrestinas, ARRDC3, dado que se ha definido que a nivel transcripcional su expresión en astrocitos es muy elevada y este tipo celular parece ser fundamental en la etiopatología de la enfermedad de Lafora. Nuestros resultados apuntan que ARRDC3 puede estar implicada en la homeostasis del transportador EAAT2 ya que hemos demostrado su interacción con los dominios WW de Nedd4.2. Sin embargo, no se ha observado interacción entre ARRDC3 y el complejo laforina-malina tanto utilizando el sistem, [EN] Lafora disease is a progressive myoclonic epilepsy caused by mutations in the genes that encode laforin or malin. The mutation of one of the two proteins prevents the formation of the functional complex and, as a result, insoluble glycogen inclusions, known as Lafora bodies, accumulate. Another pathology described in Lafora patients is the altered homeostasis of the astrocitic glutamate transporter EAAT2 (GLT-1 in mouse). This transporter is normally located at the plasma membrane of these cells to internalize glutamate (neurotransmitter) and to stop the excitation of the neurons. This location is highly regulated by ubiquitination/ deubiquitination processes. An E3 ubiquitin ligase called Nedd4.2 intervenes in the internalization pathway of the glutamate transporter. In order to bind to the glutamate transporter and to ubiquitinate it, it needs adaptor proteins called arrestins. There is still not much information about the function of arrestins but they have been described as E3 ubiquitin ligases adaptors in the endocytosis of membrane transporters. In Lafora disease, the level of glutamate transporters at the plasma membrane of astrocytes is reduced and this could be due to a change in the ubiquitination pattern of the transporter by the laforin-malin complex. In this paper we study one of the ¿-arrestin family component, ARRDC3, since it has been defined that its expression at the transcriptional level in astrocytes is elevated and this cell type seems to be fundamental in the etiopathology of Lafora disease. Our results suggest that ARRDC3 may be involved in the homeostasis of the EAAT2 transporter since we have demonstrated its interaction with the WW domains of Nedd4.2. However, no interaction between ARRDC3 and the laforin-malin complex has been observed either using the yeast two hybrid system or immunoaffinity techniques in cell cultures. Also, preliminary results seem to indicate that there is no ubiquitination of ARRDC3 by the complex.

Lafora disease (LD) is a fatal rare neurodegenerative disorder characterized by epilepsy, neurodegeneration and insoluble polyglucosan accumulation in brain and other peripheral tissues. Although in the last two decades we have increased our knowledge on the molecular basis underlying the pathophysiology of LD, only a small part of the research in LD has paid attention to the mechanisms triggering one of the most lethal features of the disease: epilepsy. Recent studies in our laboratory suggested that a dysfunction in the activity of the mouse astrocytic glutamate transporter 1 (GLT-1) could contribute to epilepsy in LD. In this work, we present new in vivo evidence of a GLT-1 dysfunction, contributing to increased levels of extracellular glutamate in the hippocampus of a mouse model of Lafora disease (Epm2b-/-, lacking the E3-ubiquitin ligase malin). According to our results, Epm2b-/- mice showed an increased neuronal activity, as assessed by c-fos expression, in the hippocampus, an area directly correlated to epileptogenesis. This brain area presented lesser ability to remove synaptic glutamate after local GLT-1 blockade with dihydrokainate (DHK), in comparison to Epm2b+/+ animals, suggesting that these animals have a compromised glutamate clearance when a challenging condition was presented. These results correlate with a hippocampal upregulation of the minor isoform of the Glt-1 gene, named Glt-1b, which has been associated with compensatory mechanisms activated in response to neuronal stress. In conclusion, the hippocampus of Epm2b-/- mice presents an in vivo impairment in glutamate uptake which could contribute to epileptogenesis.

[EN] Lafora progressive myoclonic epilepsy (LD) is a rare disease characterized by neurodegeneration, epilepsy and theaccumulation of polyglucosans in different tissues, known as Lafora bodies. At the moment, there is a limited knowledge on the molecular bases of the disease and there is no available treatment, which leads to the death of patients within 10 years after the onset of the first symptoms. Until now, mutations in two genes that could explain the basis of the disease in 92% of the cases: EPM2A, encoding phosphatase dual laforin, and EPM2B, encoding malin, an E3-ubiquitin ligase. Both proteins interact forming a complex able to ubiquitinate different substrates. Recently, the involvement of the complex in several pathways has been demonstrated. However, there has been little advance in the understanding of the molecular bases underlying one of the most important features of the disease: epilepsy. For this reason, the aim of this thesis was to attempt to understand this process. The most part of the studies analyzing the epilepsy in LD focused on the analysis of the neural systems. However, recent studies suggest that an impairment of the astrocytes due to existing neuroinflamation could contribute to the triggering and/or the development of epilepsy and other neurological diseases. For this reason, our efforts have been focused on the study of the main excitatory neurotransmitter homeostasis, glutamate, in cellular and animal models of LD., The mentioned homeostasis depends on the correct functioning of the astrocytic cells and their deregulation contributes significantly to an increase in neural hyperexcitability and death. In this work, an alteration of the main astrocytic glutamate transporter, GLT-1, has been identified. This transporter is responsible of 90% of extracellular glutamate uptake. Therefore, a decrease in the activity of GLT-1 leads to an increase in synaptic glutamate, contributing to the hyperexcitability. Primary astrocytes from cortex of mice deficient in Epm2a or Epm2b genes, models of LD, show a decrease in the amount of GLT-1 located at the cell surface, which is correlated to a lower capacity of these cells to take glutamate up. This change in the subcellular localization is regulated by changes in the ubiquitination of the transporter, which is increased in the presence of overexpressed laforin-malin complex. Finally, we demonstrated that mouse models for LD present a reduced capacity to uptake glutamate compared to the control group, using in vivo microdialysis. Hence this thesis presents evidences of a deregulation in the homeostasis of the glutamate transporter GLT-1 in animals and cells models for LD and suggests that this alteration could contribute to epilepsy in LD., [ES] La epilepsia mioclónica progresiva de tipo Lafora (LD) es una enfermedad rara caracterizada por neurodegeneración, epilepsia y acumulación de poliglucosanos en diferentes tejidos, denominados cuerpos de Lafora. Por el momento, existe un conocimiento limitado de las bases moleculares de la enfermedad y todavía no se dispone de un tratamiento adecuado, lo que lleva a la muerte de los pacientes 10 años después del debut de los primeros síntomas.Hasta la fecha se han identificado mutaciones en dos genes que explicarían la base de la enfermedad en un 92% de los casos: EPM2A, que codifica la fosfatasa dual laforina, y EPM2B, que codifica una E3-ubicuitina ligasa. Ambas proteínas interaccionan formando un complejo capaz de ubicuitinar diferentes sustratos.Recientemente se ha demostrado la afectación de varias vías en la enfermedad de Lafora. Hasta la fecha, la mayor parte de estudios realizados analizando la epilepsia en la enfermedad de Lafora se han centrado en analizar los sistemas neurales. No obstante, estudios recientes sugieren que una afectación de los astrocitos debido a la neuroinflamación existente podría contribuir al desarrollo de la epilepsia. Por ese motivo, nuestros esfuerzos han ido dirigidos al estudio de la homeostasis del principal neurotransmisor excitatorio, el glutamato, en modelos celulares y animales de la enfermedad de Lafora. Dicha homeostasis depende del correcto funcionamiento de las células astrocitarias y su desregulación contribuye significativamente a un aumento de la hiperexcitabilidad y la muerte neuronal., En este trabajo se ha identificado una desregulación del principal transportador de glutamato astrocitario, GLT 1. Dicho transportador es el responsable de la captación del 90% del glutamato extracelular, por lo que una disminución de la actividad de GLT-1 provoca un aumento del glutamato sináptico, favoreciendo la hiperexcitabilidad. Astrocitos primarios procedentes de corteza de ratón deficientes en el gen Epm2a o Epm2b muestran una disminución de la cantidad de GLT-1 expresada en la superficie celular, que se correlaciona con una menor capacidad de estas células para captar glutamato. Este cambio en la localización subcelular está regulado por cambios en la ubicuitinación del transportador, que se encuentra aumentada en presencia de laforina-malina sobreexpresada. Por último, se ha demostrado que los ratones modelo para LD presentan una capacidad disminuida para captar glutamato respecto a los controles, utilizando microdiálisis in vivo.Por lo tanto, esta tesis presenta evidencias de una desregulación de la homeostasis del transportador de glutamato GLT-1 en células y animales modelo para LD y sugiere que esta alteración podría contribuir a la epilepsia presente en LD.

[ES] La degradación intracelular de proteínas es un proceso catabólico clave para la supervivencia celular frente a cambios ambientales. Este proceso degradativo tiene como función principal la eliminación de proteínas mal plegadas, que pueden ser tóxicas al acumularse, o la eliminación de proteínas que no son útiles en ciertas situaciones, para conseguir una rápida adaptación al medio. The main function of protein degradation is the removal of unfolded protein, which accumulation may be toxic, or useless proteins, providing a quickly environmental adaptation. The products of that degradation are amino acids that are reused for the synthesis of new proteins or are metabolized for energy obtaining. There are two protein degradation mechanisms: lysosomal-dependent pathway and lysosomal-independent pathway. One of the main mechanisms in the lysosomal dependent pathway is macroautophagy (called autophagy here after to simplify). Autophagy consists in formation of a double membrane that will close sequestering cytoplasmic portions. These double membrane structures, called autophagosomes, will ultimately fuse with endosomes and lysosomes forming autolysosomes. Lysosomal enzymes, which contain autolysosomes, will degrade the engulfed material. Autophagy is a highly regulated process. Upon nutrient deprivation, such as amino acids, autophagy is induced to provide necessary requirements to cell survival. On the other hand, presence of amino acids suppresses autophagy. However, glucose, that is other nutrient, regulates the process in an opposite way. It has been demonstrated that addition of glucose to mammalian cells after a starvation period induces autophagy. This induction, surprisingly, is mTOR and AMPK-independent and p38 MAPK-dependent. p38 MAPK translocates to the nucleus in the presence of glucose suggesting a role in the regulation of gene expression. The main objective of this work is to determine p38 MAPK regulated-genes that may be implicated in autophagy induction by glucose. To this end, we analysed a PCR Array data where MEFs cells p38+/+ and p38-/- in the presence or absence of glucose were compared, and we selected four candidate genes: Rgs19, Bcl2, Ifna4 y Akt1. La supervivencia celular frente a los cambios ambientales requiere el mantenimiento de un equilibrio dinámico entre la síntesis y la degradación de proteínas. La degradación de proteínas, además de regular diferentes procesos celulares, tiene como función principal la eliminación de productos que no son útiles para la célula en determinadas situaciones o cuya acumulación puede ser tóxica. Los productos de esta degradación, es decir los aminoácidos, son reutilizados para la síntesis de nuevas moléculas o son metabolizados para la obtención de energía. La alteración de esta proteólisis intracelular puede llevar a la acumulación en el citoplasma de orgánulos defectuosos o de moléculas que se pueden agrupar en agregados insolubles y que pueden así desencadenar diferentes patologías. Aunque se ha avanzado bastante durante los últimos años en los conocimientos sobre la degradación intracelular de proteínas y de sus principales mecanismos, existen bastantes detalles moleculares todavía desconocidos. Por este motivo es necesario aportar nueva información sobre estos procesos que además podría ser relevante para identificar nuevas dianas terapéuticas y desarrollar tratamientos más eficaces para las enfermedades derivadas de alteraciones en los mismos. La degradación de proteínas ocurre por diferentes mecanismos que pueden clasificarse generalmente en dependientes o no de unos orgánulos citoplásmicos, los lisosomas. Vectores recombinantes basados en el virus vaccinia modificado de Ankara (MVA) como vacunas contra la leishmaniasis. Los vectores de la invención contienen secuencias codificantes de la proteína LACK, preferentemente insertadas en el locus de hemaglutinina del virus y bajo el control de un promotor que permite su expresión a lo largo del ciclo de infección del virus. Son vectores seguros, estables, que dan lugar a una potente respuesta inmune que confiere protección frente a la leishmaniasis, por lo que son buenos candidatos para ser utilizados en vacunación frente a esta enfermedad, especialmente en seres humanos, así como en el reservorio animal más importante de esta antropozoonosis, los perros.

[EN]The invention relates to recombinant vectors based on the Modified Vaccinia Ankara (MVA) virus as vaccines against leishmaniasis. The inventive vectors contain sequences encoding the LACK protein, which are preferably inserted into the hemagglutinin locus of the virus under the control of a promoter which enables the expression of same throughout the infection cycle of the virus.
