Abstract

As a highly sensitive vibrational imaging method, stimulated Raman scattering (SRS) microscopy is finding various applications, while its theoretical treatment seems still under development. Here we present a theoretical estimation of spontaneous Raman signal and SRS signal from Raman scattering cross section, irrespective of the numerical aperture of the objective lens. We confirm a reasonable agreement between our theory with a recently proposed treatment based on the stimulated Raman scattering cross section. 2. Standalone gradient measurement of matrix norm for programmable unitary converters

Abstract

Programmable unitary converters are powerful tools for realizing unitary transformations, advancing fields of computing and communication. The accuracy of these unitary transformations is crucial for maintaining high fidelity in such applications. However, various physical artifacts can impair the accuracy of the synthesized transformations. A commonly employed approach uses the system's gradient to restore accuracy. Matrix norm is used to define error between matrices, and minimization of this norm using the gradient restores the accuracy. Although this gradient can indeed be physically measured using external equipment, it leads to a rather complex optical system. In this study, we propose a standalone method for measuring matrix norm gradients, where `standalone' means that no additional optical equipment is needed. This method is based on the mathematical fact that the central difference, which is generally used for the approximation of differentiation, can yield exact differentiation for any unitary converters. Furthermore, we introduce a new matrix distance that is suitable for optimizing unitary converters which use intensity detectors at the output. This distance also yields the exact differentiation with the central difference. 3. Iterative configuration of programmable unitary converter based on few layer redundant multi-plane light conversion

Abstract

Programmable unitary photonic devices are emerging as promising tools to implement unitary transformation for quantum information processing, machine learning, and optical communication. These devices typically use a rectangular mesh of Mach-Zehnder interferometers (MZIs), which has a clear mathematical structure and can be configured deterministically. However, this mesh architecture is sensitive to fabrication errors, and the correction techniques are still under investigation. In contrast, the multi-plane light conversion (MPLC) architecture is more robust against fabrication errors, but a deterministic method for configuring the converter has not yet been developed due to its complex mathematical structure. In this work, we propose a fast iterative configuration method for MPLC, following the mathematical review of the matrix distance and proposal of a new norm. We show through numerical simulations that adding a few redundant layers significantly improves the convergence of the MPLC architecture, making it a practical and attractive option. We also consider the effects of finite resolution and crosstalk in phase shifters in our simulations. In addition, we propose a phase-insensitive distance suited for applications using only intensity detections. Our method demonstrates orders of magnitude better accuracy and a 20-fold speed-up compared to previous approaches.

Abstract

We demonstrate high-throughput label-free single-cell image cytometry and image-based classification of Euglena gracilis (a microalgal species) under different culture conditions. We perform it with our high-throughput optofluidic image cytometer composed of a time-stretch microscope with 780-nm resolution and 75-Hz line rate, and an inertial-focusing microfluidic device. By analyzing a large number of single-cell images from the image cytometer, we identify differences in morphological and intracellular phenotypes between E. gracilis cell groups and statistically classify them under various culture conditions including nitrogen deficiency for lipid induction. Our method holds promise for real-time evaluation of culture techniques for E. gracilis and possibly other microalgae in a non-invasive manner.

Abstract

Plants can rapidly respond to different stresses by activating multiple signaling and defense pathways. The ability to directly visualize and quantify these pathways in real time using bioorthogonal probes would have practical applications, including characterizing plant responses to both abiotic and biotic stress. Fluorescence-based labels are widely used for tagging of small biomolecules but are relatively bulky and with potential effects on their endogenous localization and metabolism. This work describes the use of deuterium- and alkyne-derived fatty acid Raman probes to visualize and track the real-time response of plants to abiotic stress within the roots. Relative quantification of the respective signals could be used to track their localization and overall real-time responses in their fatty acid pools due to drought and heat stress without labor-intensive isolation procedures. Their overall usability and low toxicity suggest that Raman probes have great untapped potential in the field of plant bioengineering.

Abstract

The advent of image-activated cell sorting and imaging-based cell picking has advanced our knowledge and exploitation of biological systems in the last decade. Unfortunately, they generally rely on fluorescent labeling for cellular phenotyping, an indirect measure of the molecular landscape in the cell, which has critical limitations. Here we demonstrate Raman image-activated cell sorting by directly probing chemically specific intracellular molecular vibrations via ultrafast multicolor stimulated Raman scattering (SRS) microscopy for cellular phenotyping. Specifically, the technology enables real-time SRS-image-based sorting of single live cells with a throughput of up to ~100 events per second without the need for fluorescent labeling. To show the broad utility of the technology, we show its applicability to diverse cell types and sizes. The technology is highly versatile and holds promise for numerous applications that are previously difficult or undesirable with fluorescence-based technologies.
