Prenatal and postnatal growth of rats conceived and reared at different G-intensities from 1.0G (earth gravity) to a maximum of 2.03G were compared. Prenatal growth was not generally impaired but the lung/body mass ratio of 22-day old fetuses at 2.03G was decreased significantly compared to 1.06 controls. Survival of neonatal rats was substantially reduced at 1.71G and 2.03G. Postnatal growth was decreased at the higher G-intensities and showed smaller or no effects at the lower G-intensities. Comparisons of organ/body mass ratios of hyper-G and 1.0G rats at 9 wks of age showed relatively few differences at the lower G-intensities. Postnatal growth of mice at 2.03G was suppressed during the neonatal period but recovered later so that after 9 wks the body mass of females reached and of males approached controls. Results of this preliminary study clearly show the influence of body mass in scaling the effects of hyper-G on the growth and development of and between different animal species.

Effects of hypergravity, simulated by chronic centrifugation, followed by a return to normal G (deceleration) on the immune system of rats were investigated. Two groups of male rats (28 days at 2.1 G, and 3.1 G) were compared to the control group (1.0 G). The animals were immunized by i.p. injections of sheep red blood cells on days 29, 42, and 57, and bled on days 36, 47, and 62. While the centrifuged rats ate and gainedsignificantly less than the control rats, the antibody titers and the organ/body mass ratios for the adrenal glands, kidneys, lungs, heart, and thymus were unaffected by gravity exposures, as were the values of the hematocrit and the white blood cell counts. It is concluded that deceleration does not adversely affect these particular aspects of the immune system.

Acute initial exposure to hypergravity (HG) was previously found to induce prolonged diestrous in rats, which was followed by return to normal estrous cycling upon more prolonged exposure to continuous HG. Bromergocryptine was found to prevent this prolonged diestrous. In this study it is found that in female rats 20 h of 3.14 G exposure (D-1 1200 h until D-2 0800 h) can induce prolactin surge at D-2 1600 h. Shorter exposure time (8 h), or exposure during a different part of the estrous cycle (19 h: from D-1 0700 h until D-2 0200 h) could not elicit this prolactin surge. Similar exposure of male rats of HG did not alter significantly their prolactin levels. It is possible that the hypothalamus of male and female rats responds differently to stimulation by HG.

The broad objective of this research program is to determine the role which gravity plays in the growth and development of mammalian animals. Current studies are focused on the effects of graded hypergravitatinal field intensities on mice, rats and other small sized laboratory animals using the chronic centrifugation technique. They include studies on reproduction and prenatal and postnatel growth and development. Among the important questions addressed are: (1) what stage or stages in animal development are affected by hypergravity and what are the effects? (2) is there a minimum or critical body size for hypergravity to produce a significant effect on growth and development? (3) are there field intensity thresholds for the preceding questions? From analysis of the body masses at birth of rats conceived and allowed to undergo gestation under 2.1G and under normal gravity (1G), it was found that there was no significant difference between the two groups. Futhermore, their growth rates postnatally were the same until they reached a body mass of approximately 50 grams when the 2.1G group showed a significantly slower rate. Results from these studies support the conclusion that prenatal as well as the early postnatal stages of growth and development of the rat are refractory to hyper-G.

The effect of glucogenesis on the plasma glucose and liver glycogen of rats exposed to hyper-G stress is investigated. Twelve male Sprague-Dawley rats are injected with C-14 lactate, alanine, of glycerol, and six of the rats are exposed to 3.1 G for 0.25, 0.50, and 1.0 hr. The plasma glucose and liver glycogen of the centrifuged and noncentrifuged rats are analyzed. A significant increase in the C-14 incorporation of the substrate into the plasma glucose and liver glycogen is observed in the centrifuged rats. The injection of 5-methoxyindole-2-carboxylic acid, a gluconeogenesis inhibitor, results in a blocked increase in plasma glucose and liver glycogen. The role of epinephrine on the hyperglycemic and liver glycogen responses of centrifuged rats is studied. It is concluded that the initial increase in plasma glucose and liver glycogen in rats exposed to hyper-G stress is the result of an increased rate of gluconeogenesis.

The present investigation is concerned with possible relations of the hyperglycemic response of rats exposed to hyper-G stress to (1) alterations in blood levels of the glucoregulatory hormones and gluconeogenic substrates, and (2) changes in insulin response on muscle glucose uptake. Male Sprague-Dawley rats weighing 250-300 g were used in the study. The results of the experiments indicate that the initial rapid rise in blood glucose of rats exposed to hyper-G stress is mediated by increases in circulating catecholamines and glucagon, both potent stimulators of hepatic gluconeogenesis. Lactate, derived from epinephrine stimulation of muscle glycogenolysis, appears to be a major precursor for the initial rise in blood glucose. The inhibition of the insulin-stimulated glucose uptake by muscle tissues may be a factor in the observed sustained hyperglycemia.

The role of the tail in the altered thermoregulation of rats during acute exposure to hypergravity was investigated, using groups of rats of two ages: 55 days (young) and 138 days (old). Rectal and foot temperature changes were measured in intact and tailless rats subjected to 1 h centrifugation of 2.8 G, with preceding (1 h) and following (1-3 h) 1 G periods. At 22 C, the loss of body heat from the tail per se does not measurably contribute to the hypothermia induced by hypergravity. However, the heat loss from the feet was greater in the tailless rats than in the intact rats from the young group of animals, although there was no significant difference between the tailless and intact rats in the old animal group. It is concluded that the inhibition of heat production is a significant factor in the hypothermia of centrifuged tailless rats, as it has been previously shown to be in the intact animals.

In a recent study of the effects of hypergravity (HG) on the reproductive system of the rat, it was found that the estrous cycle is perturbed in a pseudopregnancy-like pattern upon first exposure of animals to HG. This can be prevented by previous exposure to HG or by administration of bromergo cryptin. Adapted rats can mate and deliver in HG. However, little is known of the condition of the individual pregnant rat, fetuses, and newborn pups in this environment. There are also questions regarding the effects of HG on the number of fetuses, the mortality rate in different HG levels, and the effects on production and secretion of PRL and TSH. The present investigation is concerned with such questions, taking into account possible approaches to avoid or minimize the lethal effects of HG. The obtained results suggest that peripartum plasma PRL levels and thyroid hormone levels are critical for the survival of the litter.

The immune response in rats exposed to simulated hypergravity (2.1 G and 3.1 G) by chronic centrifugation was assessed. Rats were immunized with sheep red blood cells (SRBC), either on the day of initial exposure to hypergravity (hyper-G), or after being centrifuged for 28 d and remaining on the centrifuge thereafter. Pair-fed and ad libitum fed noncentrifuged controls were used. Although there were some alterations in leukocyte counts, hyper-G did not systematically affect the primary or secondary anti-SRBC response, hematocrits, or the sizes of the liver, spleen, kidneys, thymus, or adrenal glands. The immune system is thus remarkably homeostatic under hypergravity conditions which do affect other physiologic parameters.

The requirements to and capabilities of a Space Station biological facility centrifuge are discussed on the basis of an assessment of the objectives and subjects of future microgravity biological experiments. It is argued that the facility should be capable of both acute and extended chronic exposure of test subjects and biological materials to altered-g loading. In addition, the experimental approaches and equipment for microgravity studies on a Space Station are outlined. Finally, the engineering requirements of such a centrifuge are examined, with consideration of radial gravity gradients, size, and physical access to animals.

Animal hypergravitational effects on metabolism and thermoregulation was studied. The two major problem areas investigated are: initial and short-term exposure effects, and chronic, long-term exposure effects involving developmental and adaptational effects. Investigations focused on: (1) quantifying changes in thermoregulation with graded G-intensities in rats; (2) further delineating the effects of duration on gluconeogenesis, gluconeogenic hormones and substrates, and glucose homeostasis; and (3) reproduction and neonatal survival rates under different G-intensities.

This experiment was designed to measure the immune response in acutely stressed and chronically centrifuged hyper-G-adapted male rats. Rats were exposed to 2.1 and 3.1 G. Acutely stressed animals were injected with sheep red blood cells (SRBC) on the day of inital exposure to hyper-G, and were chronically centrifuged for 10 to 15 days after immunization. Hyper-G-adapted rats were chronically centrifuged for 28 days prior to antigen injection and for 21 days after injection. Booster injections were given and serum samples taken at intervals from 3 to 9 days after the initial and booster injections. Antigen dose, injected ip, ranged between 1.35 x 10 to the 6th and 1.38 x 10 to the 9th SRBC/100 g. body weight. Pair-fed and ad libitum fed noncentrifuged controls were used. No significant differences in anti-SRBC antibody titers were found between centrifuged and control animals, although there were some changes in WBC counts and a significant increase in adrenal-gland size in acutely stressed animals.

The role of gluconeogenesis in the altered carbohydrate metabolism in rats exposed to hyper-G stress is investigated. The blood levels of the substrates and hormones involved in gluconeogenesis were determined in rats exposed to 3.1 G for various time periods (0.25 to 24 hr). It is found that hyper-G stressed rats showed an immediate increase in plasma glucose at the onset of centrifugation which persisted throughout all the exposure periods. A substantial part of the initial rise in blood glucose is attributed to an increased rate of gluconeogenesis. An increase in liver glycogen deposition was observed in centrifuged rats as early as 0.50 hr exposure time, with progressively larger amounts accumulated as the exposure time was extended to 24 hr. It is concluded that the increase in gluconeogenic activity of hyper-G stressed rats is due to an increase in the mobilization of gluconeogenic substrates from perpheral tissues to the liver as a result of increases in circulating catecholamines and glucagon.

Several experiments concerned with the exposure of animals to acute or chronic centrifugation are described. The effects of hypergravity particularly discussed include the decreased growth rate and body weight, increased metabolic rate, skeletal deformation, and loss of body fat.

It was found that the rate of aging of 17 month old rats which had been exposed to 3.14 times normal gravity in an animal centrifuge for 8 months was larger than that of the controls as determined by the apparently elevated lipofuscin content in heart and kidney, reduced numbers and increased size of mitochondria of heart tissue, and inferior liver mitochondria respiration. Steady-state food intake per day per kg body weight, which is presumably proportional to rate of living or specific basal metabolic expenditure, was found to be about 18 percent higher than in the controls after an initial 2 month adaptation period. Although half of the centrifuged animals lived only a little shorter than the controls (average about 343 vs. 364 days on the average, statistically nonsignificant), the remaining half (longest survivors) lived on the centrifuge an average of 520 days (range 483-572) compared to an average of 574 days (range 502-615) for the controls, computed from the onset of centrifugation, or 11 percent shorter. These findings indicate that a moderate increase of the level of basal metabolism of young adult rats adapted to hypergravity compared to controls in normal gravity is accompanied by a roughly similar increase in the rate of organ aging and reduction of survival, in agreement with Pearl's (1928) rate of living theory of aging, previously experimentally demonstrated only in poikilotherms.

A review of studies investigating the thermal response of rats exposed to hypergravic fields well below maximum tolerance levels is presented. It is concluded that several lines of evidence indicate that the neural switching network for temperature regulation and cardiovascular channeling of blood flow is transiently affected during the first hour a rat is exposed to hypergravity. Moreover, even after one hour of exposure, when the core temperature has fallen several degrees, shivering and nonshivering thermogenesis are not fully activated. Only after prolonged exposure to hypergravic fields do heat production mechanisms recover sufficiently to bring the core temperature back to a normal level. Thus, the data indicate a more rapid recovery of effector mechanisms for heat loss than for heat production.

A study is described calling into question the hypothesis that an increased physical load during chronic centrifugation contributes to the body composition changes observed in centrifuged rats. Considering fat-free and fat-free dry masses of the total body and carcass, it is seen that centrifugation combined with either wheel running or restraint reduced these masses to approximately 85% of the respective 1-g values, that is, the same result with either sedentary rats or rats running several hundred meters per day. It is pointed out that if an effect of a centrifugation-induced load is present but hidden by opposing factors, an analysis of variance should reveal it as an interaction between acceleration and other variables; however, no such interactions are found here. Tables are included emphasizing the pervasive influence of chronic centrifugation after only 12 days exposure.

Rats induced into a hypermetabolic state by exposure to chronic (7 mo) centrifugation at 4.15 g exhibited increased glucose uptake at lower plasma insulin levels than weight-matched control animals following oral glucose administration. In order to determine the insulin sensitivity of specific tissues, the effect of exogenous insulin on glucose uptake by isolated perfused livers and hindlim skeletal muscle from rats adapted to chronic centrifugation for one year was compared with perfused tissue from 2.5 mo-old noncentrifuged control animals of equal body weight. Metabolic glucose clearance by skeletal muscle from hypergravic rats did not prove significantly greater than control muscle when perfused in the absence of insulin (10.6 vs 8.1 microliters/min-g-muscle), but was twice as fast (23.0 vs 9.5) at perfusate insulin levels of 35 micro-U/ml. Conversely, glucose uptake by hypergravic livers was significantly decreased (P is less than 0.001) compared with control livers (10.3 vs 27.8) at perfusate insulin levels of 40 micro-U/ml. Results suggest that skeletal muscle rather than liver is primarily responsible for the enhanced sensitivity to insulin and the increased energy expenditure observed in rats subjected to hypergravity.
