2. Real-Time Cattle Interaction Recognition via Triple-stream Network

Abstract

In stockbreeding of beef cattle, computer vision-based approaches have been widely employed to monitor cattle conditions (e.g. the physical, physiology, and health). To this end, the accurate and effective recognition of cattle action is a prerequisite. Generally, most existing models are confined to individual behavior that uses video-based methods to extract spatial-temporal features for recognizing the individual actions of each cattle. However, there is sociality among cattle and their interaction usually reflects important conditions, e.g. estrus, and also video-based method neglects the real-time capability of the model. Based on this, we tackle the challenging task of real-time recognizing interactions between cattle in a single frame in this paper. The pipeline of our method includes two main modules: Cattle Localization Network and Interaction Recognition Network. At every moment, cattle localization network outputs high-quality interaction proposals from every detected cattle and feeds them into the interaction recognition network with a triple-stream architecture. Such a triple-stream network allows us to fuse different features relevant to recognizing interactions. Specifically, the three kinds of features are a visual feature that extracts the appearance representation of interaction proposals, a geometric feature that reflects the spatial relationship between cattle, and a semantic feature that captures our prior knowledge of the relationship between the individual action and interaction of cattle. In addition, to solve the problem of insufficient quantity of labeled data, we pre-train the model based on self-supervised learning. Abstract

Chronic active Epstein–Barr virus (CAEBV) infection is a rare disease, mainly affecting children, typically characterized by persistent infectious mononucleosis (IM)-like symptoms. We describe an adult case of CAEBV without IM-like symptoms, which was indistinguishable from autoimmune hepatitis (AIH). A 60-year-old woman with liver damage was diagnosed with AIH (International Diagnostic Score: 16 points). She had been treated with prednisolone for three years; however, her transaminases had never normalized. She was admitted for another liver biopsy due to repeated high fevers and worsening of her liver damage over two months. Her EBV-DNA copy number was 2.9 × 10(4) copies/μg DNA, and EBV-encoded small RNA1-positive lymphocytic infiltration was observed in both the present and previously collected (three years ago) liver tissue samples. This case implies that hepatic involvement in a CAEBV without IM-like symptoms is difficult to distinguish from AIH and may be misdiagnosed. In some steroid resistant AIH cases, evaluating for CAEBV may be valuable.

Abstract

The esophageal epithelium is a prototypical stratified squamous epithelium that exhibits an exquisite equilibrium between proliferation and differentiation. After basal cells proliferate, they migrate outward toward the luminal surface, undergo differentiation, and eventually slough due to apoptosis. The identification and characterization of stem cells responsible for the maintenance of the esophageal epithelium remains elusive. Here, we employed Hoechst dye extrusion and BrdU label--retaining assays to identify in mice a potential esophageal stem cell population that localizes to the basal cell compartment. The self-renewing capacity of this population was characterized using a clonogenic assay and a 3D organotypic culture model. The putative esophageal stem cells were also capable of epithelial reconstitution in vivo in direct esophageal epithelial injury models. In both the 3D organotypic culture and direct mucosal injury models, the putative stem cells gave rise to undifferentiated and differentiated cells. Abstract

The epidermal growth factor receptor (EGFR) activates several signaling cascades in response to epidermal growth factor stimulation. One of these signaling events involves tyrosine phosphorylation of signal transducer and activator of transcription (STAT), whereas another involves activation of the phosphatidylinositol 3-OH kinase pathway. Two possibilities for STAT activation exist: a janus kinase (JAK)-dependent and a JAK-independent mechanism. Herein, we demonstrate that EGFR overexpression in primary esophageal keratinocytes activates STAT in a JAK-dependent fashion with the functional consequence of enhanced cell migration, which can be abolished by use of a JAK-specific inhibitor, AG-490. We determined the mechanisms underlying the signal transduction pathway responsible for increased cell migration. Stimulation of EGFR induces Tyr701 phosphorylation of STAT 1 and initiates complex formation of STAT 1 and STAT3 with JAK1 and JAK2. Thereafter, the STATs translocate to the nucleus within 15 min. In addition, we found that activation of this signaling pathway results in matrix metalloproteinase-1 (MMP-1) activity. By contrast, Akt activation does not impact the EGFR-STATs-JAKs complex formation and nuclear translocation of the STATs with subsequent MMP-1 activity, although Akt activation may contribute to cell migration through an independent mechanism. Abstract

Previous studies have indicated that some leptin gene polymorphisms were associated with economically important traits in cattle breeds. However, polymorphisms in the leptin gene have not been reported thus far in Japanese Black cattle. Here, we aimed to identify the leptin gene polymorphisms which are associated with carcass traits and fatty acid composition in Japanese Black cattle. We sequenced the full‐length CDS of leptin gene for eight Japanese Black cattle. Sequence comparison revealed eight SNPs. Three of these were predicted to cause amino acid substitutions: Y7F, R25C, and A80V. Then, we genotyped these SNPs in two populations (JB1 with 560 animals and JB2 with 450 animals) and investigated the effects on the traits.
