Abstract

Background: Although long-term management of psoriasis is paramount, this approach is challenging in clinical practice. In the recent PSO-LONG trial, a fixed-dose combination of betamethasone dipropionate (BD) and calcipotriol (Cal) foam applied twice a week on non-consecutive days for 52 weeks (proactive treatment) reduced the risk of relapse. However, the role of Cal/BD foam in the long-term management of psoriasis needs further clarifications. The ProActive Management (PAM) program, a nationwide Italian project, aims at reaching a consensus on the role of proactive management of psoriasis. Methods: A steering committee generated some statements through the nominal group technique (NGT). The statements were voted by an expert panel in an adapted Delphi voting process. Results: Eighteen statements were proposed, and the majority of them (14/18) reached a consensus during the Delphi voting. The need to provide long-term proactive topical treatment to reduce the risk of relapse for the treatment of challenging diseases sites or in patients where phototherapy or systemic therapies are contraindicated/ineffective was widely recognized. A consensus was reached about the possibility to associate the proactive treatment with systemic and biological therapies, without the need for dose intensification, thus favoring a prolonged remission. Moreover, the proactive treatment was recognized as more effective than weekend therapy in increasing time free from relapses. Approaches to improve adherence, on the other hand, need further investigation. Conclusions: The inclusion in guidelines of a proactive strategy among the effective treatment options will be a fundamental step in the evolution of a mild-moderate psoriasis therapeutic approach.

Abstract

Background: Psoriasis (Ps) is a chronic systemic autoimmune disease associated with pruritus in 64–98% of patients. However, few modestly sized studies assess factors associated with psoriatic pruritus. Objective: To investigate factors associated with Ps pruritus intensity. Methods: Psoriasis patients 18years or older seen in one of 155 centres in Italy between September 2005 and 2009 were identified from the Italian PsoCare registry. Patients without cutaneous psoriasis and those with missed information on pruritus were excluded. Results: We identified 10802 patients, with a mean age 48.8±14.3years. Mild itch was present in 33.2% of patients, moderate in 34.4%, severe in 18.7% and very severe in 13.7%. Higher itch intensity was associated with female gender, lower educational attainment compared to university degree, pustular psoriasis, psoriasis on the head, face, palmoplantar areas, folds and genitalia, more severe disease, disease duration

Abstract

La recente restrizione dell'utilizzo di formulati a base di rame, principale strumento per la difesa dalla peronospora della vite in agricoltura biologica, ha intensificato gli studi su possibili strategie alternative. Il presente studio è stato condotto su viti in vaso trattate alla radice con un formulato a base di T. asperellum e T. gamsii e alla foglia con Silicio in forma di soluzione di gel di silice. Le piantine sono state poi inoculate con sospensione di sporangi di P. viticola, valutando, a livello fogliare, sia l'esito dell'infezione sia la produzione di acido jasmonico. In campo si è operato su due vigneti e 3 anni di osservazione, rilevando la diffusione e la gravità della malattia su piante trattate con Trichoderma e Silicio. I dati ottenuti su viti in vaso mostrano un incremento di acido jasmonico nelle piante trattate, maggiore nella combinazione dei due prodotti, cui corrisponde una riduzione della gravità del sintomo fogliare. Una certa riduzione della gravità della malattia emerge anche dalle prove in vigneto. Abstract

Among grapevine trunk diseases-related fungi, the basidiomycete Fomitiporia mediterranea (Fmed) is responsible for wood-decay in Vitis vinifera, showing an intermediate behaviour between brownand white-rot fungi. Nowadays, it is widely accepted that, in brown-rot fungi, wood decay is the result of synergic enzymatic and non-enzymatic degrading action. In the first one, enzymatic action, laccases and manganese peroxidases (MnP) play a major role in catalysing lignin degradation, splitting C?-C?, ?-aryl, C1-C? and aromatic C-C bonds. In the second one, non- enzymatic action, the lignin/cellulose degradation is attributed to the non-enzymatic Fenton/Haber Weiss pathway, generating hydroxyl radicals within the wood cell, also thanks to Fe oxidation. Because low-molecular weight (LMW) iron-binding compounds mediate this wood degradation, the mechanism has been named "chelator-mediated Fenton" (CMF) reaction. Since it is still not clear if Fmed has both mechanisms of lignin/cellulose degradation (enzymatic and CMF reactions), these two pathways were evaluated screening 13 Fmed strains, assaying: i) laccase and MnP activity; ii) the strains capability to produce LMW iron-binding compounds, ferrous-iron (Fe2+) accumulation and the consequent production of hydroxyl radicals.

11. Rainfall and temperature influence expression of foliar symptoms of grapevine leaf stripe disease (esca complex) in vineyards

Abstract

Two vineyards in the Abruzzo Region, central Italy, affected by grapevine leaf stripe disease (GLSD), were monitored for incidence and severity of foliar symptoms of the disease for 21 consecutive years (1994 to 2014). Each year, rainfall and temperature were recorded. Correlations between symptom incidence or severity and rainfall or temperature of different periods of the year or single months were assessed. July rainfall and temperature were highly correlated with incidence and severity of leaf symptoms. The vineyards were characterized by high longevity, vigour and yields, and a particular dynamic of GLSD. In the period following 2005, the year of peak vine mortality, there was a decrease of the expression of foliar symptoms, which was not correlated with both of the climate parameters. Greater mortality occurred for vines showing severe symptoms at the first appearance of GLSD than for vines with low severity

Abstract

SummaryBackground The nationwide prevalence of latent tuberculosis infection (LTBI) in Italian patients with psoriasis has never been investigated. Objectives To estimate the nationwide prevalence of LTBI in Italian patients with psoriasis who are candidates for systemic treatment. Methods Data were obtained from the Psocare Registry on those patients (n = 4946) with age > 18 years, systemic treatment at entry specified and tuberculin skin test (TST) performed according to the Mantoux method. LTBI diagnosis was based on a positive TST result in the absence of any clinical, radiological or microbiological evidence of active tuberculosis. Results Latent tuberculosis infection was diagnosed in 8·3% of patients with psoriasis (409 of 4946). Abstract

Downy mildew is a serious disease in organic viticulture as only copper is allowed for its control. Despite this already great difficulty the European Community, and is probably soon going to decrease the allowed quantity to 3 kg ha−1 year−1. Thus, an improvement in copper efficiency, for example enhancing plant response to the disease, is urgently needed. Two days after infection by Plasmopara viticola, resistants produce a peak of methyl-jasmonate higher than the sensitive grapevines. Jasmonate production could be triggered in plants applying Trichoderma or silicon. In this frame Trichoderma, silicon or their combination were tested on potted vines to evaluate the ability of treatments to induce, far from the time of application, plant response such as production of methyl-jasmonate and reduction of the leaf symptoms. The study was carried out analyzing the expression of 13-lipooxygenases LOXA and LOXO, the 9-LOX and the coronatine insensitive protein 1 (COI1) as genes involved in the synthesis of the jasmonic acid and in the plant response to this hormone. The LOXO activation precede or is contemporary to methyl-jasmonate peak and is probably involved in its synthesis, while the activation of LOXA follows the peak, as a responsive gene. Trichoderma activate 9-LOX response while silicon activates the COI1. Since the two compounds activate different pathways, they may integrate each to other. The higher effect observed for the combination against artificial infection seems to confirm this hypothesis.

Abstract

Several elicitors, stimulating induced resistance mechanisms, have potential in preventing or mitigating pathogen infections. Some of these compounds, triggering the production of jasmonic acid (JA), a precursor of herbivore-induced plant volatiles, could also play a central role in indirect resistance to pest species, by improving beneficial arthropod performance, and necrotrophic pathogens. In the current work, Trichoderma gamsii / T. asperellum and silica gel treatments – alone and in combination – were studied to evaluate the plant defence mechanism on grapevines (Vitis vinifera L.) by laboratory and field trials. JA production level was measured before and after Plasmopara viticola infection on potted vines. JA production induced by silica gel was higher than that caused by Trichoderma before infection. In Trichoderma -treated plants, JA production increased after P. viticola inoculation. In vineyard field trials, Mymaridae (Hymenoptera: Chalcidoidea) showed higher captures in transparent sticky traps on silica gel-treated plants, in comparison with control. On the other hand, no significant attraction was detected for Ichneumonoidea and other Chalcidoidea in silica gel and T. gamsii / T. asperellum -treated plants. Abstract

I microrganismi benefici utilizzati in agricoltura biologica possono instaurare interazioni con la pianta ospite in grado di proteggerla dalle malattie e di migliorarne la crescita e l’assorbimento di soluti e acqua. Quest’ultimo aspetto potrebbe peraltro divenire importante a causa di una prevedibile progressiva limitazione della disponibilità di acqua. Piante di lattuga sono state allevate in vaso, in ambiente controllato per temperatura, intensità luminosa e fotoperiodo. Le piante sono state trattate con formulati commerciali a base di Trichoderma sp., di Glomus sp., e di loro combinazioni anche con ulteriori microrganismi benefici. Per ciascun formulato, le applicazioni sono state realizzate secondo il seguente schema: alla semina; alla semina e al trapianto; alla semina, al trapianto e durante la crescita in vaso. Il consumo d’acqua è stato registrato mediante l’ausilio di appositi sensori per il contenuto volumetrico d’acqua nel suolo (VWC). Da parte nostra abbiamo inteso valutare contestualmente e in funzione del numero e del posizionamento delle applicazioni: l’attività dei prodotti contro Rhizoctonia solani inoculata al terreno successivamente e il loro effetto sull’utilizzo dell’acqua da parte della pianta. I dati ottenuti mostrano una maggiore attività dei prodotti quando sono stati applicati alla semina e al trapianto. La maggior parte dei microrganismi saggiati ha inoltre evidenziato un consumo d’acqua minore per unità di peso prodotto. I formulati a base di Trichoderma mostrano la migliore combinazione tra i parametri in prova.

Abstract

Background: Some studies have shown that switching patients from one tumor necrosis factor (TNF)-alfa inhibitor to another may be beneficial when they have an inadequate response or an adverse event. Objective: We sought to assess the variables predicting the efficacy of the second TNF-alfa inhibitor in patients discontinuing the first TNF-alfa inhibitor. Methods: Data from all 5423 consecutive patients starting TNF-alfa inhibitor therapy for psoriasis between September 2005 and September 2010 who were included in the Italian Psocare registry were analyzed. Results: In 105 patients who switched to a second TNF-alfa inhibitor who had complete follow-up data, 75% improvement in the Psoriasis Area Severity Index score (PASI 75) was reached by 29% after 16 weeks and by 45.6% after 24 weeks. Patients who switched because of secondary loss of efficacy (loss of initial PASI 75 response) or adverse events/intolerance were more likely to reach PASI 75 than those who switched as a result of primary inefficacy (PASI 75 never achieved) (hazard ratio 2.7, 95% confidence interval 1.3-5.5 vs hazard ratio 2.0, 95% confidence interval 1.0-3.9 and 1, respectively). Limitations: There was a small number of patients with complete follow-up data. Conclusion: PASI 75 response in patients who switched from one antie-TNF-alfa agent to another was significantly reduced in patients who showed primary inefficacy of the first antie-TNF-alfa.

Abstract

Esca is the most severe and widespread grapevine wood disease. Although foliar symptom expression is not correlated with vine wood deterioration because of the intermittent nature of esca leaf symptom, the loss of production is associated with foliar symptom expression being diseased asymptomatic vines able to produce grapes with similar characteristics to grapes from healthy vines. Foliar symptoms seem to be associated with transport of toxins from infected wood to leaves. A reduction in esca symptom severity was observed in many-year trials applying commercial formulations of fosetyl Al either before or soon after the appearance of esca in vineyards properly managed and following the plan for downy mildew control. Moreover, a reduction of the average length of internal necrosis following inoculations with vascular esca pathogens and treated by fosetyl Al was noticed in potted vines. Investigations were carried out in order to explain the activity of fosetyl Al towards esca and associated pathogens. Physiological parameters of potted plants were measured: net photosynthetic rates of infected untreated plants were depressed by 20-50% for 24-60 hours after treatment compared to infected untreated ones. Foliar total proteins were extracted and tested by spectrophotometric analyses. In infected treated leaves chitobiosidase and glucosaminidase activities increased from 3 to 9 days after treatment, glucanase activity was enhanced at the third day and lypoxigenase activity increased over the time at pH 6.5, compared to untreated leaves. No differences in trans-resveratrol and ε-viniferin contents were found in the wood of treated and untreated vines. Results were discussed.

Abstract

The effect of different soil treatments, including i) commercial formulations of Trichoderma harzianum strain T22 (Rootshield), ii) a mixture of bacteria and fungi (Ekoprop) and iii) silicon, on physiological and morphological parameters of potted kiwifruit plants artificially inoculated with Fomitiporia mediterranea (Fom) and Phaeoacremonium aleophilum (Pal), has been investigated in an experiment performed in 2005 and 2006. Glucanase and chitinase activity of leaf proteins, shoot dry weight, leaf chlorophyll content and root apparatus development have been detected. Glucanase activity, determined in 2006, was increased by Ekoprop and Rootshield, Fom inoculation, Rootshield + Pal inoculation in July and by all treatments + Fom, except for silicon in April, and reduced by silicon treatment applied singly. Chitinase activity, detected in 2006, was increased by all single treatments and inoculations, by Rootshield + Pal in April, by silicon + Pal in June and by Rootshield + Fom in April. In 2005, all treatments increased shoot dry weight, whereas silicon caused a slightly decrease in 2006. In general, the chlorophyll content was not influenced by biological treatments, while the root apparatus was enhanced by the treatments.

Abstract

OBJECTIVE: To evaluate variations in laboratory parameters and diagnoses of selected clinical conditions up to 16 weeks after starting a new systemic psoriasis treatment for Psocare Registry enrollees. DESIGN: Prospective cohort study. SETTING: Italian public referral centres for psoriasis treatment. PATIENTS: First-time recipients (n = 10,539) of continuous systemic psoriasis treatment for at least 16 weeks. MAIN OUTCOME MEASURE: Mean variations in (weeks 8 and 16) and proportions of patients reaching a clinically meaningful increase in serum levels (week 16) of total and low-density lipoprotein cholesterol, triglycerides, aspartate amino transferase, alanine amino transferase and creatinine, as well as week-16 cumulative incidences of new diagnoses of diabetes mellitus and arterial hypertension. RESULTS: Mean cholesterol and triglyceride levels significantly increased in patients treated with acitretin or cyclosporine. Mean triglyceride levels also increased in efalizumab- and etanercept-treated patients. Mean transaminase values increased in methotrexate-treated patients, and mean aspartate amino transferase levels increased in infliximab-treated patients. The average serum creatinine value increased in cyclosporine-treated patients. Acitretin and cyclosporine were associated with risk of hypercholesterolaemia (odds ratios 1.51 and 1.34) and acitretin with risk of hypertriglyceridaemia (odds ratio 1.43). Methotrexate and infliximab were associated with risk of more than doubling the upper normal aspartate amino transferase (odds ratios 2.06 and 1.87) and alanine amino transferase (odds ratios 2.38 and 1.74) values. The relative risk of developing arterial hypertension and diabetes was increased for patients receiving cyclosporine (odds ratios 3.31 and 2.88). CONCLUSION: Systemic treatments for psoriasis resulted in heterogeneous effects on the parameters analysed.

28. A possible positive interaction between tebuconazole and electrolysed water as a wood penetrating formulation in propagation material against Phaeomoniella chlamydospora

Abstract

Research on trunk diseases has shown that infections in the nursery are not only due to basal and graft union infections of the cuttings via the wounds but also to systemic infection from mother vines. To reduce cane infections the application of a highly penetrating fungicide, tebuconazole (Folicur®SE), provided in a special - devoted to wood treatments - formulation was tested at hydration. Cut ends of cuttings were sealed by wax, immersed in a fungicide suspension, and penetration into the wood of tebuconazole verified by HPLC analysis. To further improve both penetration into the wood and efficiency of the funcidide, tebuconazole (very active in reducing Phaeomoniella chlamydospora mycelium growth but not so efficient on spore germination) was also combined with electrolyzed water (EAW), very active in inhibiting spore germination, and previously studied for a possible use in hydration. The HPLC data showed that, even if systemic fungicide as triazoles can not be transported in the sap of cuttings as it happens in vines in the field, still the passive absorption of Folicur®SE in non active tissue was high enough to be at the active concentration in all wood parts after soaking, even after 3 months from treatment. Different trials were carried out in a commercial nursery on cuttings artificially or naturally infected with P. chlamydospora. The use of the Folicur®SE formulation without or, even more with EAW, provided a strong reduction of the infections in naturally and artificially infected cuttings.

Abstract

An experimental formulation based on copper oxychloride and gluconates, which is able to penetrate vine foliar tissue, was tested for wood treatment against some of the fungi involved in the diseases included in the esca disease complex. In vitro trials were carried out to: i) test the effect of the formulation on the mycelial growth rate of Phaeomoniella chlamydospora (Pch), Phaeoacremonium aleophilum (Pal) and Fomitiporia mediterranea (Fmed), and on the conidial germination of Pch and Pal; and ii) conduct a preliminary investigation of the presence of the copper formulation on the ability of Pal to produce toxic metabolites. Trials were also conducted on potted vines to i) measure copper penetration into the trunk of potted vines using atomic absorption spectrophotometry; and ii) test the effect of the copper formulation on colonization of the fungus in trunks. Finally, the copper formulation was field-tested in vineyards showing leaf stripe. The implications of the tests in the control of the esca disease complex are discussed.

37. Trans-resveratrol and leaf symptom expression in Esca of grapevine

Abstract

Esca of grapevine is a serious disease in some aspects still lacking, including the nature of foliar symptoms. Leaves collected at different plant growth stage from esca symptomatic vineyards cv. Trebbiano d'Abruzzo showed the presence of higher concentrations of trans-resveratrol, the major stilbene produced in grapevine, compared to leaves from healthy plants. The increased levels of trans-resveratrol were supposed to be associated with the host-plant defence response, thought it also raises new questions on a possible role of the stilbene in the foliar symptom development. In 2009, symptomatic leaves were collected from the above mentioned vineyards, and were divided into 4 classes calculating symptom severity as a percentage of chlorosis and necrosis on the total leaf area. For each class, leaves were collected at pre-bunch closure, post-veraison and harvest, and the level of trans-resveratrol was determined. The results showed higher concentrations of trans-resveratrol in symptomatic leaves collected at pre-bunch closure compared to what observed at the other growth stages. These results are in agreement with our previous findings and the stronger response of leaves might be correlated with the leaf functioning at the different plant growth stages; moreover, the increasing of concentrations of trans-resveratrol with the increasing of the severity of leaf symptoms seems to exclude an involvement of the stilbene in the formation of symptoms. Further investigations on cut leaves inoculated with pathogen culture filtrates and/or trans-resveratrol may lead to a better understanding of the relationship between the stilbene and the foliar symptom expression.

40. Laboratory assays on non-enzymatic processes associated with iron and esca tracheomycotic fungi

Abstract

The tracheomycotic fungi Phaeomoniella chlamydospora and Phaeoacremonium aleophilum are considered the main pathogens causing esca of grapevine, a widely spread wood disease that affects vine yield and longevity. These pathogens can produce toxins and extracellular enzymes in a complex infection process, not yet fully understood. The possibility that fungi employ also non-enzymatic means in such process was investigated: biological tests in the laboratory were carried out to examine the role of iron in promoting non-enzymatic processes with the production of radicals. P. chlamydospora and P. aleophilum produced siderophores, and reduced ferric iron; although both fungi lack of specific enzymatic activity, they were able to degrade crystalline cellulose, but only in the presence of iron. It was therefore hypothesized that the activation of iron-dependent cellulose-degrading mechanisms that supplement the degrading activity of the enzymes, may have taken place with the production of hydroxyl radicals; this hypothesis was confirmed by a specific spectrophotometric test as capacity of the radicals to break down 2-deoxi-D-ribose. The present study demonstrated that P. chlamydospora and P. aleophilum can activate iron-dependent non-enzymatic mechanisms, amplifying their pathogenic capacity. These findings, if confirmed by further study, may furnish a new way to explain the nature of the vine/pathogen relationship in term of wood degradation and/or foliar symptoms formation, leading to a better understanding of the esca complex infection process.

43. Protection of pruning wounds on kiwifruit by Trichoderma harzianum and effects on the host plant

Abstract

Decay of kiwifruit (Actinidia deliciosa var. deliciosa) is a recently discovered chronic wood disease, widespread in Italy, which causes reduced productivity and longevity of vineyards. The disease is highly dangerous, is usually underestimated and is difficult to eradicate once present. Different pathogenic fungi, above all Phaeoacremonium aleophilum and Fomitiporia mediterranea, are known to cause the disease. The main route for infection in plants appears to be through pruning wounds. In order to develop low-impact strategies for sustainable agriculture, we investigated the use of Trichoderma harzianum as a potential defence agent on summer and winter pruning wounds, to prevent disease development. During the summer, 1-year-old shoots from potted plants were cut and immediately treated with a T. harzianum suspension; the formation of healing callus on wounds was followed, and a better developed callus, together with a shorter callusing period, were observed in treated plants compared to the control. The physiological mechanisms involved in wound healing were investigated by measuring the concentration of the growth promoter hormone indole-3-acetic acid (IAA) in the pruned shoots, using gas chromatography-mass spectrometry (GC-MS). Changes in the level of endogenous free IAA were observed during the first 8 days after pruning, suggesting a possible implication in callus formation. Furthermore, the response of winter pruning wounds, treated with Trichoderma, to artificial infection with Phaeoacremonium aleophilum was investigated, through re-isolation of the pathogen from inoculated shoots. The results are discussed.

45. Effects of Trichoderma applications on vines grown in organic nursery

Abstract

A two-year trial was conducted to evaluate the effects of applications of commercial formulations of the fungus Trichoderma on graftlings of grapevine (Vitis vinifera L.) in a commercial nursery where plants were grown under organic management. Treatments were carried out at callusing, rooting and callusing+rooting. Effects on the host-plant morpho-physiological characteristics were observed and depended on the type and timing of Trichoderma application. Treatment at rooting was the most effective whilst application at callusing and combination of treatments gave controversial results. The most noticeable effect of application of Trichoderma was an increase of quantitative-qualitative characteristics of the root system, with a consistent development of root hairs. Compared to untreated plants the percentage of certifiable plants treated at rooting was higher.

47. Effects of biostimulants on esca

Abstract

Biostimulants are compounds that influence plant physiological processes producing faster growing and healthier plants and enhanced stress tolerance. The esca complex is correlated with host-plant physiology and can be also considered a stress-related disease. Efficient control methods in the vineyard are most lacking, especially increasing age of plant and incidence of the disease. The activity of some biostimulants was investigated in many-year trials in order to assess effects on both the incidence and severity of leaf symptoms in the vineyard and the development of Phaeomoniella chlamydospora artificially inoculated in potted vines. Field trials were carried out in 5 15-20-year-old vineyards for 4-7 consecutive years. Three-four treatments for each biostimulants were set up based on the Companies indications. Potted plants were treated, trunk inoculated with P. chlamydospora, and treated for the 2 following consecutive years. In field trials, treatments did not provide a reduction of foliar symptoms; in some cases the annual and cumulated incidence increased. Treated vines that showed foliar symptoms in a certain year of investigation will have showed foliar symptoms during the following years more frequently compared to untreated vines. On the other hand the percentage of mortality decreased in treated vineyards. After 3 years of treatmens, two of the tested biostimulants reduced the development of the internal necrosis caused by P. chlamydospora. The percentage of fungal re-isolation from infected treated vines was generally lower than untreated ones. Prospects for biostimulants as a means to be considered in esca control strategies are discussed.
