4. Dynamics of Golgi matrix proteins in planta

Abstract

The plant Golgi apparatus plays a central role in the secretory pathway, being the site of protein glycosylation, sorting and the synthesis of cell wall polysaccharides and glycolipids. Golgi matrix proteins are involved in the formation and maintenance of the Golgi stack and putative plant homologues have been identified, but their general function and role in plant Golgi biogenesis still is unknown. To study Golgi biogenesis, the Golgi was disrupted using both the drug Brefeldin A (BFA) and a mutated GTP-Iocked form of the small GTPase Sarlp, blocking protein transport from the endoplasmic reticulum to the Golgi. As prolonged expression of Sarl-GTP is toxic to cells, the gene was inserted into a dexamethasone-inducible promoter system, allowing controlled expression and Golgi disruption. The system was characterised by live cell imaging of fluorescent Golgi and ER exit site markers and at the ultrastructural level by electron microscopy. A novel method to reverse the dexamethasone-inducib'e system in tobacco using the glucocorticoid antagonist RU486 was established. The subcellular location of the Golgi matrix proteins GFP-AtCASP, GFP-golgin-84, AtGRIP-GFP and GFP-TMF and other Golgi markers fused to fluorescent proteins was studied during Golgi disruption and reformation. After disruption of Golgi membranes, the trans-Golgi matrix proteins GFP-TMF and AtGRIP-GFP were redistributed into the cytoplasm. The cis-Golgi matrix proteins GFP-AtCASP and GFP-golgin-84 labelled not only the ER and cytoplasm respectively, but also remnant punctate structures. Those remnants were found to co-locate with the ER exit site marker Sarl-GTP-YFP, indicating a close relationship between the cis-Golgi matrix and ER exit sites. After BFA washout, cis-Golgi matrix proteins labelled the reforming Golgi before transGolgi matrix proteins. Interactions between Golgi matrix proteins and regulatory proteins were visualised in vivo in tobacco leaf epidermal cells using fluorescence lifetime Imagmg (FUM) combined with fluorescence resonance energy transfer (FRET).

23. Living on the edge: The role of Atgolgin-84A at the plant ER-Golgi interface

Abstract

The plant Golgi apparatus is responsible for the processing of proteins received from the endoplasmic reticulum (ER) and their distribution to multiple destinations within the cell. Golgi matrix components, such as golgins, have been identified and suggested to function as putative tethering factors to mediate the physical connections between Golgi bodies and the ER network. Golgins are proteins anchored to the Golgi membrane by the C-terminus either through transmembrane domains (TMDs) or interaction with small regulatory GTPases. The golgin N-terminus contains long coiled coil domains which consist of a number of α-helices wrapped around each other to form a structure similar to a rope being made from several strands, reaching into the cytoplasm. In animal cells golgins are also implicated in specific recognition of cargo at the Golgi. Here we investigate the plant golgin Atgolgin-84A for its subcellular localisation and potential role as a tethering factor at the ER-Golgi interface. For this, fluorescent fusions of Atgolgin-84A and an Atgolgin-84A truncation lacking the coiled-coil domains (Atgolgin-84AΔ1-557) were transiently expressed in tobacco leaf epidermal cells and imaged using high-resolution confocal microscopy. We show that Atgolgin-84A localises to a pre-cis-Golgi compartment that is also labelled by one of the COPII proteins as well as by the tether protein AtCASP. Upon overexpression of Atgolgin-84A or its deletion mutant, transport between the ER and Golgi bodies is impaired and cargo proteins are redirected to the vacuole.

32. Sex matters: The effects of biological sex on adipose tissue biology and energy metabolism

Abstract

Adipose tissue is a complex and multi-faceted organ. It responds dynamically to internal and external stimuli, depending on the developmental stage and activity of the organism. The most common functional subunits of adipose tissue, white and brown adipocytes, regulate and respond to endocrine processes, which then determine metabolic rate as well as adipose tissue functions. While the molecular aspects of white and brown adipose biology have become clearer in the recent past, much less is known about sex-specific differences in regulation and deposition of adipose tissue, and the specific role of the so-called pink adipocytes during lactation in females. This review summarises the current understanding of adipose tissue dynamics with a focus on sex-specific differences in adipose tissue energy metabolism and endocrine functions, focussing on mammalian model organisms as well as human-derived data. In females, pink adipocytes trans-differentiate during pregnancy from subcutaneous white adipocytes and are responsible for milk-secretion in mammary glands. Overlooking biological sex variation may ultimately hamper clinical treatments of many aspects of metabolic disorders.

33. The role of RTNLB3 in endoplasmic reticulum structure and cytokinesis in planta

Abstract

Reticulons are a large family of ER membrane shaping proteins that are ubiquitous to eukaryotic cells. Plants possess the highest number of different reticulon proteins, which contain a conserved reticulon homology domain that has been associated with their ER curvature inducing properties. Individual reticulon family members are suspected to play other roles in cell function, although most remain unknown. RTNLB3 is a reticulon that has been identified in a plasmodesmatal proteomic screen and could play a role in constricting the ER membrane into the highly curved rod of the desmotubule, forming the core of these structures within plant cell walls. The role of the ER in cytokinesis and the importance of the regulation of ER structure during this process has not been previously investigated in detail. This study demonstrates that RTNLB3 becomes highly concentrated at the cell plate during its formation and that over-expression of a non-functional truncated form of the protein interferes with cell wall formation. Over-expression of truncated RTNLB3 results in an increased proportion of cisternal ER within the ER network. Stable over-expression of the construct proved lethal to BY-2 cell lines and Arabidopsis thaliana plants. Analysis of meristematic root tip cells of Arabidopsis plants stably transformed with an inducible truncated RTNLB3 construct, and subsequent induction prior to analysis, resulted in disruption to cell plate formation. RTNLB3 therefore may play a direct role in the formation of the cell plate; the loss of regulation of ER membrane curvature within the network prevents successful completion of cytokinesis; or a combination of these factors. The lethal phenotypes observed within this study, and the abnormal ultrastructure of cells identified as a result of over-expression of a mutant form of RTNLB3, demonstrate the importance of this ER shaping protein and ER structure in the process of plant cytokinesis.

36. Interactions between the plant Golgi apparatus and the cytoskeleton

Abstract

In animal cells, the relationship between the Golgi apparatus and cytoskeleton has been well characterised but not much is known in plants. The functions of the Golgi apparatus are conserved amongst eukaryotes. It is one of the main stations in the secretory pathway and is involved in protein processing and sorting to different destinations. In plants, it is also involved in trafficking and positioning of cell wall components. In tobacco epidermal cells, fluorescent labelling with Golgi marker proteins has shown that the Golgi apparatus is made of hundreds of individual units scattered in the cortical cytoplasm and moving on the actin cytoskeleton. The contribution of actin filaments to Golgi body motility in plant has been extensively described, but this actin-centric view has recently been challenged. Emerging evidence suggests that microtubules may contribute to short distance movement and ‘fine tuning’ of Golgi body displacement. Moreover, proteomic studies linking the actin- cytoskeleton to microtubules have demonstrated that these two components of the cytoskeleton are closely related and a role of the microtubules in Golgi movement cannot be excluded. In this thesis, automated tracking of Golgi bodies was used to understand and quantify the contribution of actin filaments and microtubules to the organelle dynamics. The tracking technique is also used to assess how the labelling of the cytoskeleton, with a novel fluorescent nanoprobe, affects the dynamics and stability of the actin filaments and the movement of Golgi bodies; FRAP analysis (fluorescent recovery after photo-bleaching) was also used to investigate the binding properties of the fluorescent nanoprobe to the actin filaments. The nanoprobe was compared with another cytoskeletal marker, Lifeact-GFP, to evaluate their suitability for studying the organelle’s motility in relation to the actin-cytoskeleton. Abstract

Background: A nationwide Movement Control Order (MCO) was enforced in Malaysia on 18 March 2020 in view of the global COVID-19 pandemic. Malaysia implemented various public health measures and later raced against time to administer COVID-19 vaccines when they became available. As a result of various public health measures to curb the spread of the virus, people in Malaysia faced unprecedented circumstances and new challenges. This study addressed the knowledge gap in our understanding the experiences, coping strategies and perspectives of the people in Malaysia about infection countermeasures by investigating their experiences during the COVID-19 pandemic., Methods: A sequential mixed method approach was used to conduct an online survey and in-depth interviews among residents in Malaysia. A total of 827 respondents participated in the online survey from 1st May to 30th June 2020. Nineteen in-depth interviews were conducted online and by phone with key informants and members of the public, who were selected through maximum variation purposive sampling between 2nd May 2020 to 20th December 2021. The semi-structured interviews employed a phenomenological approach and transcripts were analysed using thematic analysis. The survey data were analysed using descriptive statistics in Stata 15.0., Results: The survey reflected significant economic impacts of the pandemic, the maximum number of days that people could cope during the MCO, and their coping strategies, which generally entailed changes in lifestyle. The internet and social media were vital platforms to mitigate against the impact of public health measures. Thematic analysis of the interview data revealed participant experiences and perceptions of COVID-19 and public health measures in four main themes: (1) work and business; (2) emotional impact (3) coping with change and (4) the COVID-19 vaccine., Conclusions: This study provides insights into the experiences, coping strategies and perspectives of people in Malaysia living through the first-ever MCO during the COVID-19 pandemic.
