Seit der Einführung des Kfz-Katalysators werden die Platingruppenelemente Platin, Palladium und Rhodium vermehrt in die Umwelt eingetragen und gelangen u.a. über Straßenabflussgewässer in aquatische Ökosysteme. Die vorliegende Arbeit untersuchte die Effekte verschiedener Konzentrationen des löslichen Platinchlorids (0, 0.1, 10, 50, 100 und 200 µg/L PtCl2) auf verschiedene Endpunkte während der frühen Individualentwicklung der beiden Süßwasserorganismen Zebrabärbling (Danio rerio) und Paradies-Schnecke (Marisa cornuarietis). Bei beiden Organismen wurden die Bioakkumulation von Platin, Effekte auf die Embryonalentwicklung, die Induktion von Stressproteinen (Hsp70) und histopathologische Gewebe- und Organveränderungen untersucht sowie gentoxische Untersuchungen durchgeführt. Es zeigte sich, dass beide Organismen Platin stark anreicherten. Bei D. rerio wurden Bioakkumulationsfaktoren (BAF) von 5-55 und bei M. cornuarietis sogar BAF von 218-714 gefunden. Die Embryotests ergaben für beide Organismen eine Verzögerung der Herzschlagrate und der Schlupfrate. Bei M. cornuarietis war weiterhin die Augenentwicklung verzögert sowie das Gewicht reduziert. Histopathologische Untersuchungen ergaben bei D. rerio Effekte von Platin auf Leber und Darm, bei M. cornuarietis zeigten die Epidermis, die Mitteldarmdrüse und die Kieme eine Reaktion auf die Platinexposition. Bei den Stressproteinuntersuchungen wurde bei D. rerio eine erhöhte Hsp70-Induktion nur bei der höchsten getesteten Konzentration von 100 µg/L PtCl2 gefunden, wogegen bei M. cornuarietis keine erhöhten Hsp70 Werte bei allen untersuchten Pt-Konzentrationen gefunden wurden. Anhand der Einzelzell-Gelelektrophorese (Comet Assay) traten bei M. cornuarietis schon ab 1 µg/L PtCl2 DNA-Schäden auf, wogegen sich bei D. rerio für die untersuchten Pt-Konzentrationen kein gentoxischer Effekt zeigte. Die Untersuchungen ergaben, dass Pt auch bereits bei umweltrelevanter Konzentration Effekte auf die Embryonalentwicklung der beiden untersuchten Organismen ausübte und somit von ökotoxikologischer Relevanz ist. Weiterhin wurde in dieser Arbeit erstmals eine durch einen Schadstoff induzierte Schaleninternalisierung bei M. cornuarietis nachgewiesen, welche im Laufe der Evolution mehrfach und unabhängig voneinander bei verschiedenen Molluskentaxa auftrat. Eine Exposition von 100-200 µg/L PtCl2 während der Embryonalentwicklung verhinderte die Bildung einer äußeren Schale und durch die Färbung des schalensekretierenden Mantelrandes mittels des Chromogens Diaminobenzidin sowie Rasterelektronenmikroskopie und synchrotroner Röntgenstrahl-Phasenkontrast-Mikrotomographie konnte beobachtet werden, dass das schalensekretierende Gewebe nicht wie üblich als Mantelrand den Eingeweidesack überwuchs, sondern in der ursprünglich embryonalen Position verblieb, so dass das Schalenmaterial gedrungen und tütenförmig ins Innere der Schnecke wuchs. Im Gegensatz zum archetypischen Bauplan prosobrancher Schnecken verblieb die Kieme dabei posterior und die Torsion wurde von der Bildung einer Mantelhöhle entkoppelt. Diese durch Platin induzierte Bauplanveränderung wurde durch verschiedene Methoden eingehender untersucht. Dabei zeigte sich anhand von Pulsexpositionsversuchen, dass das sensitive „Fenster“ während der Embryonalentwicklung, in welchem Pt die Ausbildung einer äußeren Schale verhindert, an Tag 4 und 5 nach der Befruchtung lag. Die durch Pt induzierte Bauplanveränderung trat nicht nur bei der prosobranchen Paradies-Schnecke, sondern auch bei der pulmonaten Posthorn-Schnecke P. corneus auf. Weitere getestete Metalle ergaben, dass neben Platin einzig hohe Konzentrationen des Metalls Lithium (2,5 und 3 mg/L LiCl), in einzelnen Fällen Schnecken ohne äußere Gehäuse induzierten. Durch die Zugabe einer äquimolaren Konzentration von Ca2+ zur Platinlösung konnte dieser Effekt großteils verhindert werden. Da Analyseverfahren ergaben, dass nicht, wie erwartet, durch die Calciumzugabe weniger Pt akkumuliert wurde, liegt es nahe, dass Ca2+ platinsensitive Signaltransduktionswege stabilisiert. Es konnte gezeigt werden, dass minimale Einflüsse auf das Entwicklungsprogramm während der Embryogenese zu nachhaltigen Veränderungen des Bauplans führen können. Following the introduction of automobile catalysts, the platinum group elements platinum (Pt), palladium (Pd), and rhodium (Rh) have been emitted into the environment and enter the aquatic ecosystem via runoff rainwater. The present study determined the effects of different concentrations of platinum chloride (0, 0.1, 10, 50, 100 und 200 µg/L PtCl2) on various endpoints during the early development of the freshwater organisms zebrafish (Danio rerio) and ramshorn snail (Marisa cornuarietis). In both organisms bioaccumulation of platinum and effects on the embryonic development were determined, the 70 kD stress protein response was measured and histopathological alterations and genotoxic effects were determined. Platinum was found to be accumulated in both organisms. Bioaccumulation factors were in the range of 5–55 for D. rerio and of 218–724 for M. cornuarietis. During embryonic development, Pt was shown to decrease the heart rate of both organisms and caused a delayed hatch of the embryos. Furthermore, a retardation of the eye development and a loss of weight due to platinum exposure was observed in M. cornuarietis. Histopathological investigations revealed effects of platinum on gut and liver of D. rerio and effects on epidermis, hepatopancreas and gill of M. cornuarietis. Stressprotein responses revealed an increased induction only at the highest tested concentration of 100 µg/L PtCl2 in D. rerio but no proteotoxic effect of platinum on M. cornuarietis. Using the single cell gel electrophoresis (comet assay) genotoxic effects of platinum were recorded already at 1 µg/L PtCl2 and higher concentrations in M. cornuarietis. However, in D. rerio no genotoxic effects of the investigated Pt concentrations were recorded. The present work shows that platinum exerts effects of already environmentally relevant concentration on early stages of both tested organisms and is therefore of ecotoxicological relevance. Furthermore, this work presents first fundings of artificially induced shell internalization in M. cornuarietis, which evolved in several molluscan taxa during the evolution. Exposure to 100-200 µg/L PtCl2 during the embryonic development prevented the formation of an external shell. Using diamino-benzidine staining of the mantle edge, scanning electron microscopy and synchrotron X-ray phase contrast holotomography, the shell secreting mantle tissue was observed not, as usual, to overgrow the visceral sac thus leading to an external shell, but remained at the original embryonic position and invaginated into the snails body. Different to the archetypal bodyplan of prosobranch snails the gill remained posterior to the heart at the hind part of the viszeral sac and torsion was uncoupled of the formation of the mantle cavity. Pulse-exposure experiments revealed days 4 and 5 postfertilization to be the most susceptible to Pt action on shell formation. The shell internalization due to platinum exposure was not restricted to the prosobranch snail M. cornuarietis but could also be observed in the pulmonate snail Planorbarius corneus. Further tested metalls revealed that except for platinum also high concentrations of lithium (2,5 und 3 mg/L LiCl) occasionally induced snails without external shell. So far, the only known way to partly protect embryos from the action of Pt is to supplement the Pt solution with equimolar concentrations of bivalent calcium ions. Since Ca supplements did not diminish Pt uptake, we conclude that Pt likely interacts with Ca signalling pathways involved in the positional system, which may be stabilized by increasing intracellular levels of Ca. This work demonstrates that selective alteration of embryonic key processes can result in fundamental changes of an existing body plan.
