Abstract: European forests are threatened by increasing numbers of invasive pests and pathogens. Over the past century, Lecanosticta acicola, a foliar pathogen predominantly of Pinus spp., has expanded its range globally, and is increasing in impact. Lecanosticta acicola causes brown spot needle blight, resulting in premature defoliation, reduced growth, and mortality in some hosts. Originating from southern regions of North American, it devastated forests in the USA's southern states in the early twentieth century, and in 1942 was discovered in Spain. Derived from Euphresco project 'Brownspotrisk,' this study aimed to establish the current distribution of Lecanosticta species, and assess the risks of L. acicola to European forests. Pathogen reports from the literature, and new/ unpublished survey data were combined into an open-access geo-database (http://www.portalofforestpathology.com), and used to visualise the pathogen's range, infer its climatic tolerance, and update its host range. Lecanosticta species have now been recorded in 44 countries, mostly in the northern hemisphere. The type species, L. acicola, has increased its range in recent years, and is present in 24 out of the 26 European countries where data were available. Other species of Lecanosticta are largely restricted to Mexico and Central America, and recently Colombia. The geo-database records demonstrate that L. acicola tolerates a wide range of climates across the northern hemisphere, and indicate its potential to colonise Pinus spp. forests across large swathes of the Europe. Preliminary analyses suggest L. acicola could affect 62% of global Pinus species area by the end of this century, under climate change predictions. Abstract: Non-native pests, climate change, and their interactions are likely to alter relationships between trees and tree-associated organisms with consequences for forest health. To understand and predict such changes, factors structuring tree-associated communities need to be determined. Here, we analysed the data consisting of records of insects and fungi collected from dormant twigs from 155 tree species at 51 botanical gardens or arboreta in 32 countries. Generalized dissimilarity models revealed similar relative importance of studied climatic, host-related and geographic factors on differences in tree-associated communities. Mean annual temperature, phylogenetic distance between hosts and geographic distance between locations were the major drivers of dissimilarities. The increasing importance of high temperatures on differences in studied communities indicate that climate change could affect tree-associated organisms directly and indirectly through host range shifts. Insect and fungal communities were more similar between closely related vs. distant hosts suggesting that host range shifts may facilitate the emergence of new pests. Moreover, dissimilarities among tree-associated communities increased with geographic distance indicating that human-mediated transport may serve as a pathway of the introductions of new pests. The results of this study highlight the need to limit the establishment of tree pests and increase the resilience of forest ecosystems to changes in climate.

Abstract: International trade in plants and climate change are two of the main factors causing damaging tree pests (i.e. fungi and insects) to spread into new areas. To mitigate these risks, a large-scale assessment of tree-associated fungi and insects is needed. We present records of endophytic fungi and insects in twigs of 17 angiosperm and gymnosperm genera, from 51 locations in 32 countries worldwide. Endophytic fungi were characterized by high-throughput sequencing of 352 samples from 145 tree species in 28 countries. Insects were reared from 227 samples of 109 tree species in 18 countries and sorted into taxonomic orders and feeding guilds. Herbivorous insects were grouped into morphospecies and were identified using molecular and morphological approaches. This dataset reveals the diversity of tree-associated taxa, as it contains 12,721 fungal Amplicon Sequence Variants and 208 herbivorous insect morphospecies, sampled across broad geographic and climatic gradients and for many tree species. This dataset will facilitate applied and fundamental studies on the distribution of fungal endophytes and insects in trees.

Abstract: Fusarium circinatum, the causal agent of pine pitch canker (PPC), is currently one of the most important threats of Pinus spp. globally. This pathogen is known in many pine-growing regions, including natural and planted forests, and can affect all life stages of trees, from emerging seedlings to mature trees. Despite the importance of PPC, the global distribution of F. circinatum is poorly documented, and this problem is also true of the hosts within countries that are affected. The aim of this study was to review the global distribution of F. circinatum, with a particular focus on Europe. We considered (1) the current and historical pathogen records, both positive and negative, based on confirmed reports from Europe and globally; (2) the genetic diversity and population structure of the pathogen; (3) the current distribution of PPC in Europe, comparing published models of predicted disease distribution; and (4) host susceptibility by reviewing literature and generating a comprehensive list of known hosts for the fungus. These data were collated from 41 countries and used to compile a specially constructed geo-database. A review of 6297 observation records showed that F. circinatum and the symptoms it causes on conifers occurred in 14 countries, including four in Europe, and is absent in 28 countries. Field observations and experimental data from 138 host species revealed 106 susceptible host species including 85 Pinus species, 6 non-pine tree species and 15 grass and herb species. Our data confirm that susceptibility to F. circinatum varies between different host species, tree ages and environmental characteristics. Knowledge on the geographic distribution, host range and the relative susceptibility of different hosts is essential for disease management, mitigation and containment strategies. Abstract: Pines are major components of native forests and plantations in Europe, where they have both economic significance and an important ecological role. Diseases of pines are mainly caused by fungal and oomycete pathogens, and can significantly reduce the survival, vigor, and yield of both individual trees and entire stands or plantations. Pine pitch canker (PPC), caused by Fusarium circinatum (Nirenberg and O'Donnell), is among the most devastating pine diseases in the world, and is an example of an emergent invasive disease in Europe. The effects of microbial interactions on plant health, as well as the possible roles plant microbiomes may have in disease expression, have been the focus of several recent studies. Here, we describe the possible effects of co-infection with pathogenic fungi and oomycetes with F. circinatum on the health of pine seedlings and mature plants, in an attempt to expand our understanding of the role that biotic interactions may play in the future of PPC disease in European nurseries and forests. The available information on pine pathogens that are able to co-occur with F. circinatum in Europe is here reviewed and interpreted to theoretically predict the effects of such co-occurrences on pine survival, growth, and yield. Beside the awareness that F. circinatum may co-occurr on pines with other pathogens, an additional outcome from this review is an updating of the literature, including the so-called grey literature, to document the geographical distribution of the relevant pathogens and to facilitate differential diagnoses, particularly in nurseries, where some of them may cause symptoms similar to those induced by F. circinatum. An early and accurate diagnosis of F. circinatum, a pathogen that has been recently introduced and that is currently regulated in Europe, is essential to prevent its introduction and spread in plantings and forests.

Abstract: [EN] Fusarium circinatum is a harmful pathogenic fungus mostly attacking Pinus species and also Pseudotsuga menziesii, causing cankers in trees of all ages, damping-off in seedlings, and mortality in cuttings and mother plants for clonal production. This fungus is listed as a quarantine pest in several parts of the world and the trade of potentially contaminated pine material such as cuttings, seedlings or seeds is restricted in order to prevent its spread to disease-free areas. Inspection of plant material often relies on DNA testing and several conventional or real-time PCR based tests targeting F. circinatum are available in the literature. In this work, an international collaborative study joined 23 partners to assess the transferability and the performance of nine molecular protocols, using a wide panel of DNA from 71 representative strains of F. circinatum and related Fusarium species. Diagnostic sensitivity, specificity and accuracy of the nine protocols all reached values >80%, and the diagnostic specificity was the only parameter differing significantly between protocols. The rates of false positives and of false negatives were computed and only the false positive rates differed significantly, ranging from 3.0% to 17.3%. The difference between protocols for some of the performance values were mainly due to cross-reactions with DNA from non-target species, which were either not tested or documented in the original articles. Considering that participating laboratories were free to use their own reagents and equipment, this study demonstrated that the diagnostic protocols for F. circinatum were not easily transferable to end-users. More generally, our results suggest that the use of protocols using conventional or real-time PCR outside their initial development and validation conditions should require careful characterization of the performance data prior to use under modified conditions (i.e. reagents and equipment). Abstract: The present study was carried out during 2008 in order to determine the presence of Annosum root and butt rot on living Abies bornmülleriana and Abies cilicicatrees in 3 different stands located in Bolu and Antalya (Akseki) provinces. Totally, 255 wood samples, taken from fresh stump surfaces of fir trees just after felling, were carried to laboratory and examined for the existence of the anamorph stage of Heterobasidion(Spiniger meineckellus (Olson) Stalpers) species. Disease incidence was 28.2% and 18.8% on the pure and mixed stands of A. bornmülleriana in Bolu, respectively, and 11.5% on the pure A. cilicica stand in Akseki. It is concluded that 78.5 % of discs cut from A. cilicicawere contaminated by airborne spores of the fungus., Bu çalışma 2008 yılında, Annosum kök ve alt gövde çürüklüğünün Abies bornmülleriana ve Abies cilicica meşcereleri üzerinde yoğunluğunu belirlemek üzere gerçekleştirilmiştir. Bu amaçla, Bolu ve Antalya (Akseki) illerinde, 3 meşcerede arazi sörveyleri yapılmıştır. Göknar ağaçlarından alınan disk ve disk parçalarından oluşan 255 örnek, laboratuar koşullarında Heterobasidion türlerinin eşeysiz döneminin (Spiniger meineckellus (Olson) Stalpers) varlığı açısından incelenmiştir. Elde edilen sonuçlara göre; A. bornmülleriana’ nın saf ve karışık meşcerelerinde hastalık yoğunluğu sırasıyla % 28.2 ve % 18.8, A. cilicica meşceresinde ise bu oran % 11.5 olarak bulunmuştur. Bunun yanında, A. cilicica’ dan kesilen disklerin % 78,5’inde görülen bulaşmanın havadan ulaşan sporlar aracılığı ile olduğu sonucuna varılmıştır.

Abstract: Fungal colonisation originating from endophytic thalli in wood of healthy Taurus fir (Abies cilicica) was studied. Bore cores were taken from living trees during field surveys. The bore cores were cut into 0.5 cm long fragments, washed with 70% (v/v) ethanol, flamed and placed into petri dishes containing malt extract, agar and streptomycin. The plates were incubated at 25°C for 4 weeks. The outgrowing mycelia were pure-cultured and identified based on amplification and sequencing of the internal transcribed spacer (ITS) region of their nuclear ribosomal DNA. Totally 12 different fungal species were isolated from Abies cilicica wood cores. Five of them belong to Basidiomycetes. The present study is the first investigation of endophytic wood fungi occurring in fir in Turkey using DNA-based methods. Therefore the study is expected to provide new information of the pioneering colonizers of wood tissues in stems of living Taurus firs

Abstract: In this study, the proportion of Abies bornmülleriana trees infected by Heterobasidion spp. was investigated in Kastamonu Province in different types of stands. One 1 cm-thick disc was taken from each of 100 freshly cut trees, washed under a running tap, placed into plastic bags and incubated in growth chamber at 24 C° for 7 days. The area occupied by the conidial stage of Heterobasidion annosum s.l. was determined under a stereomicroscope using transparent film placed onto the upper surface of each disc. During this investigation conidia of Heterobasidion annosum s.l. were taken with a needle and placed onto agar plates. The obtained isolates were identified with pairing tests and DNA-based methods.All 36 isolates responded as Heterobasidion abietinum to the tester isolates. Identification based on PCR amplification with MJF – MJR and KJF-KFR primers gave the same result. 34% (34 out of 100) of the discs taken from the fir forests were found to be infected with H. abietinum. The characteristics of the colonized patches on the discs indicated that the H. abietinum colonies originated from stem infections of the trees. The proportion of the disc area covered by the conidial stage of the fungus was 80% in one of the discs, 6-10% in 6 discs, 1-5% in 18 discs, and 0-1 % in 11 discs. 18. Susceptibility of Pinus nigra and Cedrus libani to Turkish Gremmeniella abietina isolates

Abstract: Gremmeniella abietina causes shoot dieback and stem cankers on conifers throughout Northern hemisphere. The aim of this study was to investigate the virulence of Turkish G. abietina isolates in a field experiment. The lower branches of 15-20-year-old P. nigra and C. libani in a plantation site at 1,050 m a.s.l. in Isparta were inoculated at 1-2-month intervals during September-January. Five isolates obtained from high altitude mountainous forests were used. Each isolate was inoculated into two branches per tree and repeated ten times on both tree species at each inoculation date. The branches were sampled at the end of February, and in August, and lesion lengths in the inner bark measured. The mean lesion length on P. nigra and C libani were 10.6 ± 0.8 and 3.8 ± 0.2 mm in February and 17.6 ± 1.4 and 7.8 ± 0.8 mm in August, respectively. Differences in the mean lesion length between the isolates were small. Nevertheless, there were significant differences between the isolates on P. nigra in November and January inoculations, and on C. libani at all four inoculation times. The mean lesion lengths for all isolates at both sampling dates was the highest (p < 0.01) in December inoculations for both P. nigra (22.0 ± 1.9 February; 32.9 ± 2.9 August) and C. libani (5.6 ± 0.7; 11.3 ± 1.2). There was no difference between the September and January inoculations on P. nigra, despite the almost six-fold difference in incubation period. During the December inoculations, the trees were most likely in winter dormancy, i.e. unable to defend themselves, which would explain the large lesions., La Gremmeniella abietina causa la muerte regresiva de brotes y cancros sobre coníferas en todo el hemisferio norte. El objetivo de este estudio fue investigar la virulencia de aislados turcos de G. abietina en un experimento de campo. Se inocularon las ramas más bajas de P. nigra y C. libani de 15 a 20 años de edad en un sitio de plantación a 1.050 m snm en Isparta a intervalos de 1-2 meses entre septiembre y enero utilizando cinco aislamientos obtenidos de bosques de las zonas montañosas altas. Cada aislado se inoculó en dos ramas por árbol y se repitieron diez veces en las dos especies en cada fecha de inoculación. Se tomaron muestras de las ramas al final del mes de febrero, y en agosto, y se midieron la longitud de la lesión en la corteza interna. La longitud media de la lesión en P. nigra y C. libani fueron 10,6 ± 0,8 y 3,8 ± 0,2 mm en febrero y 17,6 ± 1,4 y 7,8 ± 0,8 mm en agosto, respectivamente. Las diferencias en la longitud de la lesión media entre los aislados eran pequeñas. Sin embargo, hubo diferencias significativas entre los aislamientos de P. nigra en inoculaciones de noviembre y enero, y en C. libani en los cuatro tiempos de inoculación. La longitud media de la lesión para todos los aislamientos en ambas fechas de muestreo fue la más alta (p < 0,01) en las inoculaciones de diciembre tanto para P. nigra (22,0 ± 1,9 de febrero, 32,9 ± 2,9 de agosto) como para C. libani (5,6 ± 0,7; 11,3 ± 1,2). Abstract: The efficacy of 30% aqueous urea solution, borax powder and spore suspensions of Phlebiopsis gigantea and Trichoderma harzianum against establishment of artificially inoculated Heterobasidion abietinum on Abies cilicica was tested both in the field and in a simulated stump treatment experiment carried out in a growth chamber. In the field, in each of the four selected stands 20 fresh stumps per control agent were treated and 20 stumps were left as untreated controls. In two of the stands, the treatments were applied in June and in the two others in November. Stumps were sampled twice, at 6- and 12-months after treatment. In the growth chamber, 10 log pieces per treatment and control were used, and sampling performed after 6-week incubation. Results of the urea and borax treatments were consistent between the experiments; the mean efficacies were 98.8 and 99.4% in the growth chamber, and 80.291.3 and 89.490.1% in the two samplings of the field experiment, respectively. Despite the high efficacies of the P. gigantea (85.9%) and T. harzianum (97.5%) treatments in the growth chamber, efficacies of these biological control agents in the field were 47.149.2 and 61.365.5%, respectively.

Abstract: Summary: Ceratocystis platani was detected at several locations in the European side of Istanbul, causing severe dieback and mortality, mainly on Platanus × acerifolia imported from Italy approximately 160 years ago, but also on Platanus orientalis. In the work reported here, the causal agent of the dieback and mortality was identified based on morphological characteristics in culture and internal transcribed spacer (ITS) sequencing, confirming the presence of C. platani in Turkey. All analysed C. platani isolates from Istanbul shared the same ITS sequence with European C. platani strains available in the NCBI, but differed from other species previously ascribed to the Ceratocystis fimbriata species complex. Pathogenicity of C. platani was proven by inoculation onto 25‐cm‐long and 4–9 mm diam. P. orientalis twigs and the causal agent reisolated from symptomatic tissues. In a survey of 976 Platanus in streets and parks of Istanbul, 26.5% of trees showed symptoms of C. platani infections, whilst 5.6% were dead. Abstract: The ability of two strains of the antibiotic-producing bacillus Aneurinibacillus migulanus to control Dothistroma septosporum infection of pine was tested in artificial inoculation experiments on 2-year-old Pinus contorta plants. Disease severity (% needles with conidiomata) on control plants was 5.8 ± 1.7%, compared with 6.2 ± 2.0% on plants treated with A. migulanus NCTC 7096. In contrast, treatment with A. migulanus Nagano reduced Dothistroma needle blight ( DNB) severity to 1.1 ± 0.4%. The conidial density and percentage germination of D. septosporum on needles were also greatly reduced on seedlings treated with A. migulanus Nagano compared with the A. migulanus NCTC 7096 and control treatments. Abstract: Herpotrichia pinetorum, Gremmenia infestans and Gremmeniella abietina were inoculated onto 2-year-old Anatolian black pine ( Pinus nigra subsp. pallasiana) and Taurus cedar ( Cedrus libani) seedlings planted in a high mountain forest (1800 m a.s.l) in south-western Turkey, to determine the effects of these fungi during winter. In June, 8 months after inoculation, 39.9% of experimental plants were dead and 20.4% of the surviving plants failed to flush. Gremmeniella abietina and H. pinetorum caused the most fatalities. Prevention of new shoot formation on surviving plants, however, was mainly an effect of G. abietina infections, although many surviving plants inoculated with G. infestans or H. pinetorum also failed to flush. All three pathogens had the potential to severely damage young plants of P. nigra subsp. pallasiana and C. libani growing at high elevations near to forests with heavy inoculum loads. The implications of this finding for P. nigra afforestations at high altitudes in Turkey are discussed. The bore cores were cut into 0.5 cm long fragments, washed with 70% (v/v) ethanol, flamed and placed into petri dishes containing malt extract, agar and streptomycin. The plates were incubated at 25°C for 4 weeks. The outgrowing mycelia were pure-cultured and identified based on amplification and sequencing of the internal transcribed spacer (ITS) region of their nuclear ribosomal DNA. Totally 12 different fungal species were isolated from Abies cilicica wood cores. Five of them belong to Basidiomycetes. The present study is the first investigation of endophytic wood fungi occurring in fir in Turkey using DNA-based methods. Abstract: In this study, the proportion of Abies bornmülleriana trees infected by Heterobasidion spp. was investigated in Kastamonu Province in different types of stands. One 1 cm-thick disc was taken from each of 100 freshly cut trees, washed under a running tap, placed into plastic bags and incubated in growth chamber at 24 C° for 7 days. The area occupied by the conidial stage of Heterobasidion annosum s.l. was determined under a stereomicroscope using transparent film placed onto the upper surface of each disc. During this investigation conidia of Heterobasidion annosum s.l. were taken with a needle and placed onto agar plates. The obtained isolates were identified with pairing tests and DNA-based methods. All 36 isolates responded as Heterobasidion abietinum to the tester isolates. Identification based on PCR amplification with MJF - MJR and KJF-KFR primers gave the same result. 34% (34 out of 100) of the discs taken from the fir forests were found to be infected with H. abietinum. The characteristics of the colonized patches on the discs indicated that the H. abietinum colonies originated from stem infections of the trees. The proportion of the disc area covered by the conidial stage of the fungus was 80% in one of the discs, 6-10% in 6 discs, 1-5% in 18 discs, and 0-1 % in 11 discs. Abstract: Non-native pests, climate change, and their interactions are likely to alter relationships between trees and tree-associated organisms with consequences for forest health. To understand and predict such changes, factors structuring tree-associated communities need to be determined. Here, we analysed the data consisting of records of insects and fungi collected from dormant twigs from 155 tree species at 51 botanical gardens or arboreta in 32 countries. Generalized dissimilarity models revealed similar relative importance of studied climatic, host-related and geographic factors on differences in tree-associated communities. Mean annual temperature, phylogenetic distance between hosts and geographic distance between locations were the major drivers of dissimilarities. The increasing importance of high temperatures on differences in studied communities indicate that climate change could affect tree-associated organisms directly and indirectly through host range shifts. Insect and fungal communities were more similar between closely related vs. distant hosts suggesting that host range shifts may facilitate the emergence of new pests. Moreover, dissimilarities among tree-associated communities increased with geographic distance indicating that human-mediated transport may serve as a pathway of the introductions of new pests. Abstract: Lecanosticta acicola is a pine needle pathogen causing brown spot needle blight that results in premature needle shedding with considerable damage described in North America, Europe, and Asia. Microsatellite and mating type markers were used to study the population genetics, migration history, and reproduction mode of the pathogen, based on a collection of 650 isolates from 27 countries and 26 hosts across the range of L. acicola. The presence of L. acicola in Georgia was confirmed in this study. Migration analyses indicate there have been several introduction events from North America into Europe. However, some of the source populations still appear to remain unknown. The populations in Croatia and western Asia appear to originate from genetically similar populations in North America. Intercontinental movement of the pathogen was reflected in an identical haplotype occurring on two continents, in North America (Canada) and Europe (Germany). Several shared haplotypes between European populations further suggests more local pathogen movement between countries. Moreover, migration analyses indicate that the populations in northern Europe originate from more established populations in central Europe. Overall, the highest genetic diversity was observed in south-eastern USA. In Europe, the highest diversity was observed in France, where the presence of both known pathogen lineages was recorded. Less than half of the observed populations contained mating types in equal proportions. Abstract: International trade in plants and climate change are two of the main factors causing damaging tree pests (i.e. fungi and insects) to spread into new areas. To mitigate these risks, a large-scale assessment of tree-associated fungi and insects is needed. We present records of endophytic fungi and insects in twigs of 17 angiosperm and gymnosperm genera, from 51 locations in 32 countries worldwide. Endophytic fungi were characterized by high-throughput sequencing of 352 samples from 145 tree species in 28 countries. Insects were reared from 227 samples of 109 tree species in 18 countries and sorted into taxonomic orders and feeding guilds. Herbivorous insects were grouped into morphospecies and were identified using molecular and morphological approaches. This dataset reveals the diversity of tree-associated taxa, as it contains 12,721 fungal Amplicon Sequence Variants and 208 herbivorous insect morphospecies, sampled across broad geographic and climatic gradients and for many tree species. Abstract: Pest introductions via trade in tree seed may result from a lack of adequate survey and validation protocols. Developing better diagnostic protocols to identify potentially harmful pests and pathogens in forest tree seed is of critical importance. High-throughput sequencing-based barcoding and metabarcoding provide effective tools for screening potentially harmful organisms in various plant materials, including seeds. However, the sample size needed to detect the total microorganism diversity of a community is a major challenge in microbiome studies. In this work, we examined how increasing sample size (ranging between 100 and 1000 seeds) influences diversity of fungal communities detected by high throughput sequencing in Pinus sylvestris seeds. Our results showed that as sample size increased, fungal alpha diversity also increased. Beta-diversity estimators detected significant differences between the mycobiota from different samples. However, taxonomic and functional diversity were not correlated with sample size. In addition, we found that increasing the number of PCR replicates resulted in a higher abundance of plant pathogens. We concluded that for the purpose of screening for potentially harmful pathogens using HTS, greater efforts should be made to increase the sample size and replicates when testing tree seed., (© 2022. Abstract: Dothistroma needle blight (DNB) is one of the most damaging foliage diseases of pine in plantations and natural forests worldwide and is caused by two closely related fungi: Dothistroma septosporum and D. pini, which are virtually impossible to differentiate from each other based on morphology. Although diagnosis of DNB based on symptoms is relatively reliable in the later stages of the disease when fruit bodies (conidiomata) are formed, for diagnosis in the early stages, as well as identification of the causal agent at species level, molecular methods are required. In addition, reliable and sensitive diagnostics before sporulation is a prerequisite for early detection to minimize accidental introductions of disease through movement of infected plant materials, especially seedlings. While amplification and sequencing of the ITS region of the rDNA alone is not reliable to differentiate the two species, conventional PCR (cPCR) using species-specific primers or mating type-specific primers and quantitative PCR (qPCR) are widely used and accepted molecular methods to identify and differentiate the DNB pathogens, either from cultures or directly from needles., (© 2022. Abstract: Dothistroma septosporum , the primary causal agent of Dothistroma needle blight, is one of the most significant foliar pathogens of pine worldwide. Its wide host and environmental ranges have led to its global success as a pathogen and severe economic damage to pine forests in many regions. This comprehensive global population study elucidated the historical migration pathways of the pathogen to reveal the Eurasian origin of the fungus. When over 3800 isolates were examined, three major population clusters were revealed: North America, Western Europe, and Eastern Europe, with distinct subclusters in the highly diverse Eastern European cluster. Modeling of historical scenarios using approximate Bayesian computation revealed the North American cluster was derived from an ancestral population in Eurasia. The Northeastern European subcluster was shown to be ancestral to all other European clusters and subclusters. The Turkish subcluster diverged first, followed by the Central European subcluster, then the Western European cluster, which has subsequently spread to much of the Southern Hemisphere. All clusters and subclusters contained both mating-types of the fungus, indicating the potential for sexual reproduction, although asexual reproduction remained the primary mode of reproduction. The study strongly suggests the native range of D. septosporum to be in Eastern Europe (i.e., the Baltic and Western Russia) and Western Asia.

Abstract: Fusarium circinatum is a harmful pathogenic fungus mostly attacking Pinus species and also Pseudotsuga menziesii, causing cankers in trees of all ages, damping-off in seedlings, and mortality in cuttings and mother plants for clonal production. This fungus is listed as a quarantine pest in several parts of the world and the trade of potentially contaminated pine material such as cuttings, seedlings or seeds is restricted in order to prevent its spread to disease-free areas. Inspection of plant material often relies on DNA testing and several conventional or real-time PCR based tests targeting F. circinatum are available in the literature. In this work, an international collaborative study joined 23 partners to assess the transferability and the performance of nine molecular protocols, using a wide panel of DNA from 71 representative strains of F. circinatum and related Fusarium species. Diagnostic sensitivity, specificity and accuracy of the nine protocols all reached values >80%, and the diagnostic specificity was the only parameter differing significantly between protocols. The rates of false positives and of false negatives were computed and only the false positive rates differed significantly, ranging from 3.0% to 17.3%. The difference between protocols for some of the performance values were mainly due to cross-reactions with DNA from non-target species, which were either not tested or documented in the original articles. Considering that participating laboratories were free to use their own reagents and equipment, this study demonstrated that the diagnostic protocols for F. circinatum were not easily transferable to end-users. More generally, our results suggest that the use of protocols using conventional or real-time PCR outside their initial development and validation conditions should require careful characterization of the performance data prior to use under modified conditions (i.e. reagents and equipment). Suggestions to improve the transfer are proposed.

39. Invasive forest pathogens in Europe: Cross-country variation in public awareness but consistency in policy acceptability.

Abstract: Political action can reduce introductions of diseases caused by invasive forest pathogens (IPs) and public support is important for effective prevention. The public's awareness of IP problems and the acceptability of policies aiming to combat these pathogens were surveyed in nine European countries (N = 3469). Although awareness of specific diseases (e.g., ash dieback) varied, problem awareness and policy acceptability were similar across countries. The public was positive towards policies for informational measures and stricter standards for plant production, but less positive towards restricting public access to protected areas. Multilevel models, including individual and country level variables, revealed that media exposure was positively associated with awareness of IP problems, and strengthened the link between problem awareness and policy acceptability. Results suggest that learning about IPs through the media and recognizing the associated problems increase policy acceptability. Overall, the study elaborates on the anthropogenic dimension of diseases caused by IPs.
