Multivariate techniques better fit the anatomy of complex neuropsychiatric disorders which are characterized not by alterations in a single region, but rather by variations across distributed brain networks. Here, we used principal component analysis (PCA) to identify patterns of covariance across brain regions and relate them to clinical and demographic variables in a large generalizable dataset of individuals with bipolar disorders and controls. We then compared performance of PCA and clustering on identical sample to identify which methodology was better in capturing links between brain and clinical measures. Using data from the ENIGMA-BD working group, we investigated T1-weighted structural MRI data from 2436 participants with BD and healthy controls, and applied PCA to cortical thickness and surface area measures. We then studied the association of principal components with clinical and demographic variables using mixed regression models. We compared the PCA model with our prior clustering analyses of the same data and also tested it in a replication sample of 327 participants with BD or schizophrenia and healthy controls. The first principal component, which indexed a greater cortical thickness across all 68 cortical regions, was negatively associated with BD, BMI, antipsychotic medications, and age and was positively associated with Li treatment. PCA demonstrated superior goodness of fit to clustering when predicting diagnosis and BMI. Moreover, applying the PCA model to the replication sample yielded significant differences in cortical thickness between healthy controls and individuals with BD or schizophrenia. Cortical thickness in the same widespread regional network as determined by PCA was negatively associated with different clinical and demographic variables, including diagnosis, age, BMI, and treatment with antipsychotic medications or lithium. PCA outperformed clustering and provided an easy-to-use and interpret method to study multivariate associations between brain structure and system-level variables. PRACTITIONER POINTS: In this study of 2770 Individuals, we confirmed that cortical thickness in widespread regional networks as determined by principal component analysis (PCA) was negatively associated with relevant clinical and demographic variables, including diagnosis, age, BMI, and treatment with antipsychotic medications or lithium. Significant associations of many different system-level variables with the same brain network suggest a lack of one-to-one mapping of individual clinical and demographic factors to specific patterns of brain changes. PCA outperformed clustering analysis in the same data set when predicting group or BMI, providing a superior method for studying multivariate associations between brain structure and system-level variables.

Abstract The incorporation of digital pathology in clinical practice will require the training of pathologists in digital skills. Our study aimed to assess the reliability among pathologists in determining tumor percentage in whole slide images (WSI) of non-small cell lung cancer (NSCLC) using digital image analysis, and study how the results correlate with the molecular findings. Pathologists from nine centers were trained to quantify epithelial tumor cells, tumor-associated stromal cells, and non-neoplastic cells from NSCLC WSI using QuPath. Then, we conducted two consecutive ring trials. In the first trial, analyzing four WSI, reliability between pathologists in the assessment of tumor cell percentage was poor (intraclass correlation coefficient (ICC) 0.09). After performing the first ring trial pathologists received feedback. The second trial, comprising 10 WSI with paired next-generation sequencing results, also showed poor reliability (ICC 0.24). Cases near the recommended 20% visual threshold for molecular techniques exhibited higher values with digital analysis. In the second ring trial reliability slightly improved and human errors were reduced from 5.6% to 1.25%. Most discrepancies arose from subjective tasks, such as the annotation process, suggesting potential improvement with future artificial intelligence solutions.

Abstract This study examines the presence of bisphenol A (BPA), S (BPS), F (BPF), and M (BPM) in various recycled plastics readily available on the market (LDPE, HDPE, PET, and PP), in light of European Food Safety Authority (EFSA) limits. Twenty samples of different origin are analyzed, cleaning treatments are applied, and the migration potential of these bisphenols into food is studied. BPM is absent in all samples, but a post-consumer recycled LDPE sample reveals high bisphenol concentrations, raising concerns, reaching 8540 ng/g, 370 ng/g, and 29 ng/g of BPA, BPS, and BPF, respectively. Migration tests show substantial migration of these contaminants into food simulants. Using a cleaning treatment with polyethylene glycol (PEG 400) reduces BPA in LDPE, HDPE, PP, and PET samples by 95%, 99%, 97% and 28%, respectively, highlighting the importance of cleaning treatments across various polymers in plastic recycling. These findings not only protect food safety but addressing environmental challenges associated with plastic recycling.

This study analyses self-presentation practices and profiles among Spanish teenagers on Instagram and TikTok. Both of these online spaces prioritise and promote visual publications, are structured to allow feedback on self-presentation, and offer the user filters both to control self-image and to target specific audiences. Three research questions guided the methodological process for the twofold analysis of self-presentation practices on social networks: an exploratory factor analysis to identify latent factors among these practices; and a descriptive analysis of the profiles identified by gender and age. Results indicate that adolescents' self-presentation practices were related to three different factors: social validation; authenticity; and image control. One of the most outstanding results is that self-presentation practices could be less guided by social feedback, since the number of followers or likes was irrelevant for most adolescents, and that adolescents increasingly tend to be guided by innovative predispositions of truthfulness. In turn, conclusions suggest that teens need to be equipped with suitable self-representation practices for safe and sustainable identity narratives on social networks, since the global COVID-19 pandemic has exponentially increased both the usage and the time spent on social networking sites, enlarging the availability of spaces for adolescents to express themselves and build their identities through different self-representation practices.

Cancer is a global health issue, increasingly prevalent and a leading cause of mortality. Despite extensive research, conventional treatments remain aggressive, often damaging healthy cells, and exhibit limited efficacy. Addressing drug resistance and enhancing treatment effectiveness are critical challenges in advancing cancer therapy. This review examines the potential of natural plant compounds, particularly phytochemicals and their derivatives, in developing novel anticancer agents. These metabolites have a long history in traditional medicine, with 42% of molecules approved for cancer treatment between 1981 and 2019 being either natural products or derivatives. The Cactaceae family, which comprises more than 1,500 species, represents a largely untapped source of potentially useful chemopreventive and anticancer agents. Although more than 3,000 plants and their derivatives have contributed to chemotherapeutic development, cactus species have received limited attention until recently. Emerging evidence highlights the anticancer potential of fruits, stems, and cladodes from various cactus species. This review provides a comprehensive and current overview of experimental studies on Cactaceae in cancer research, aiming to pave the way for the development of innovative, natural cancer therapeutics and contribute to the ongoing battle against this formidable disease.

Objective To determine the prevalence of gingival recessions in students of the faculty of odontology at Universidad de Los Andes and the factor associated with its presence. Methods In this cross-sectional study, a randomized stratified sampling was used to select the students. The sample included 311 undergraduate students evaluated between March and December 2022. The year spectrum was from 17 to 29 years old. Two calibrated examiners intra e interoperator completed a clinical evaluation with a periodontal probe, where the purpose was to diagnose gingival recessions and identify if there were any related factors such as smoking, braces, bruxism, marginal insertion frenulum, fine phenotype, and malposition. The total prevalence was described with each trust interval of 95%. The association between categorical variables was analyzed with the chi-square test, and the t-student test of the Mann-Whitney test analyzed the difference between continuous variables and recession prevalence. Results 93,56% of the students presented at least one gingival recession. The highest prevalence related to teeth occurred in pieces 1.6, 3.4, 2.6, and 4.1, and the most severe were 3.3, 4.3, and 1.3. The most associated factor related to recessions was orthodontics, with a prevalence of 75,2%. Bruxism showed a positive association with the number of recessions, with a prevalence of 60,7%. Conclusions Buccal gingival recessions were very prevalent in undergraduate students at the dental school of Universidad de los Andes during 2022, and they were strongly associated with the history of orthodontics.

Abstract Following a request from the European Commission, EFSA was asked to deliver a scientific opinion on Loigolactobacillus coryniformis DSM 34345 when used as a technological additive to improve ensiling of fresh plant material. The additive is intended for use with all fresh plant material for all animal species at a proposed minimum concentration of 1 × 108 colony forming units (CFU)/kg fresh plant material. The bacterial species L. coryniformis is considered by EFSA to be suitable for the qualified presumption of safety approach to safety assessment. The identity of the strain was established and no acquired antimicrobial resistance genes of concern were detected. Therefore, the FEEDAP Panel concluded that the use of the strain as a silage additive is considered safe for all the animal species, for consumers of products from animals fed the treated silage and for the environment. Regarding user safety, the additive containing Loigolactobacillus coryniformis DSM 34345 should be considered as a potential skin and respiratory sensitiser, and any exposure through skin and respiratory tract is considered a risk. One preparation was shown not to be irritant to skin or eyes. However, the Panel cannot assess the irritation potential of other possible preparations. The FEEDAP Panel concluded that Loigolactobacillus coryniformis DSM 34345 has the potential to improve the production of silages prepared from all fresh plant materials at a minimum concentration of 1 × 108 CFU/kg fresh material.

Abstract Some anti‐mycobacterial drugs are known to cause QT interval prolongation, potentially leading to life‐threatening ventricular arrhythmia. However, the highest leprosy and tuberculosis burden occurs in settings where electrocardiographic monitoring is challenging. The feasibility and accuracy of alternative strategies, such as the use of automated measurements or a mobile electrocardiogram (mECG) device, have not been evaluated in this context. As part of the phase II randomized controlled BE‐PEOPLE trial evaluating the safety of bedaquiline‐enhanced post‐exposure prophylaxis (bedaquiline and rifampicin, BE‐PEP, versus rifampicin, SDR‐PEP) for leprosy, all participants had corrected QT intervals (QTc) measured at baseline and on the day after receiving post‐exposure prophylaxis. The accuracy of mECG measurements as well as automated 12L‐ECG measurements was evaluated. In total, 635 mECGs from 323 participants were recorded, of which 616 (97%) were of sufficient quality for QTc measurement. Este artículo analiza dos novelas acerca del archipiélago de Chiloé ubicado en el sur de Chile: Gente en la isla (1938) de Rubén Azócar y Huipampa, tierra de sonámbulos (1944), de Nicasio Tangol. Las novelas incluyen rasgos que son típicos de las novelas de comienzo del siglo veinte en la literatura chilena, aunque al presente han recibido escasos comentarios críticos. Consideramos aquí la contribución de estos textos a la construcción literaria del archipiélago de Chiloé como un territorio distante dentro de la nación chilena. Dialogando con debates recientes acerca de la literatura regionalista en Latinoamérica, con especial atención a temas de género, proponemos la figura de la ‘mirada masculina’ con el fin de comparar los enfoques narrativos de estas dos novelas. Mostraremos cómo, desplegando diferentes articulaciones de esta mirada masculina, Azócar y Tangol describen una tierra distintivamente feminizada, abordando en el proceso problemas históricos relativos a la integración de Chiloé a la nación chilena.

The journey of Brazilian female Paralympians transcends mere statistical increases in women's participation. Behind the modest athlete growth lies the reality of women who are doubly marginalized by the intersection of gender and disability in an arena tailored for able-bodied men. Our study aimed to catalyze critical discourses surrounding the historical trajectory of Paralympic women's sports. Through a comprehensive documentary analysis based on the Brazilian Paralympic Committee's official documents from 1976 to 2021, we sought to shed light on this complex scenario. Numerically, Brazil's representation comprised 229 women who, predominantly, had physical impairments and engaged in individual sports. In addition to a sporting legacy deeply entrenched in physical rehabilitation with limited opportunities for team-based sports, we observed negative influences stemming from ableist and sexist narratives. A thorough investigation into Paralympic milestones revealed a multitude of social barriers and highlighted the significant impact of societal changes in reshaping athletic opportunities and challenging traditional stereotypes. La generación fotovoltaica se presenta hoy en día como una alternativa ventajosa desde el punto de vista ambiental frente a las fuentes convencionales de energía y es una opción para el sector agropecuario poder disminuir los gastos debido a la facturación de electricidad. Un sistema fotovoltaico conectado a la red en La Empresa Agropecuaria Horquita permitirá reducir el consumo de electricidad desde la empresa eléctrica. En este artículo se describe una propuesta de sistema fotovoltaico conectado a red que consta de 1014 paneles de 270 Wp con 13 inversores. La capacidad total del sistema es de 273 kWp y es capaz de generar 346,4 MWh/año. Se simuló el desempeño del sistema utilizando el software profesional PVSyst 7.1 para comprobar los resultados del cálculo manual además de obtener información adicional sobre el desempeño del sistema en el tiempo. La propuesta es factible económicamente arrojando un VPN de 1,7 MMCUP con un periodo de recuperación de 8 años a partir de la disminución de la facturación de electricidad en 640 840 CUP/año. Sin embargo, se detectó que este tipo de proyecto no es muy atractivo para aprovechar fondos de oportunidad si se tiene en cuenta que la TIR es muy baja (4%) en comparación con la tasa de descuento empleada. El proyecto tiene como ventaja adicional que permite la reducción de emisiones de CO2 a la atmósfera estimándose emisiones evitadas de 242,5 t de CO2/año. ABSTRAC Photovoltaic generation is presented today as an environmentally advantageous alternative to conventional energy sources and is an option for the agricultural sector to reduce the expenditure incurred due to electricity billing. A photovoltaic system connected to the grid at the Horquita Agricultural Enterprise allow reducing electricity consumption from the electrical company. This paper details a proposal for a connected to grid photovoltaic system that consists of 1014 panels with 270 Wp connected to 13 inverters. The total capacity of the system is 273 kWp and it is capable of generating 346,4 MWh/year. The performance of the proposed system was simulated using the professional software PVSyst 7.1 and the results of the manual calculation were verified in addition to obtaining other information about its performance. The proposal is feasible from an economic point of view, yielding a positive NPV of 1,7 MMCUP and a payback period of 8 years from the decrease in electricity billing by 640,840 CUP/year. However, it was detected that this type of project is not very attractive to take advantage of opportunity funds if it is taken into account that the IRR is very low (4%) compared to the discount rate used. The additional advantage of the project is that it allows the reduction of CO2 emissions into the atmosphere, estimating avoided emissions of 242.5 t of CO2/year. Abstract Huntington’s disease (HD) is caused by an aberrant expansion of CAG repeats in the HTT gene that mainly affects basal ganglia. Although striatal dysfunction has been widely studied in HD mouse models, other brain areas can also be relevant to the pathology. In this sense, we have special interest on the retina as this is the most exposed part of the central nervous system that enable health monitoring of patients using noninvasive techniques. To establish the retina as an appropriate tissue for HD studies, we need to correlate the retinal alterations with those in the inner brain, i.e., striatum. We confirmed the malfunction of the transgenic R6/1 retinas, which underwent a rearrangement of their transcriptome as extensive as in the striatum. Although tissue-enriched genes were downregulated in both areas, a neuroinflammation signature was only clearly induced in the R6/1 retina in which the observed glial activation was reminiscent of the situation in HD patient’s brains. The retinal neuroinflammation was confirmed in the slow progressive knock-in zQ175 strain. Overall, these results demonstrated the suitability of the mouse retina as a research model for HD and its associated glial activation.

Abstract Objective To evaluate the relationship between oral health status, self-perception of oral health, and depression. Methods This cross-sectional study included 2953 individuals that were ≥ 18 years of age and participated in the Chilean National Health Survey (NHS), 2016–2017. Information on oral, dental, and mental health, and the presence or absence of depressive symptoms was collected. Secondary data analysis was carried out using STATA and included logistic regression models adjusted for sex, age, and educational level. The analyses factored in the expansion weights to estimate representative prevalences of the entire population. Results Participants experiencing frequent dental or prosthesis-related discomfort while speaking (OR: 1.57; 95% CI: 1.01–2.43) were related with exhibiting suspected depression. Removable upper denture users were at a higher risk of exhibiting suspected (OR: 2.04; 95% CI: 1.11–3.74) than those not using them. Participants diagnosed with depression in the past 12 months had a similar number of teeth (median = 24) compared to those without depression (median = 25) (OR: 0.99; 95% CI: 0.96–1.02). Conclusion Experiencing dental or prosthesis-related difficulties in speaking is related to suspected depression or a diagnosis of depression. These findings highlight the importance of developing comprehensive healthcare approaches that consider mental health in the context of oral health.

ABSTRACTLeprosy is an infectious neglected tropical disease, which can cause irreversible disabilities if not diagnosed in time. Colombia continues to show high rates of leprosy-related disability, mainly due to a delay in diagnosis. Limited knowledge is available that explains this delay, therefore our study aimed to explore the perceptions and experiences of leprosy health professionals with the delay in leprosy diagnosis in the Cesar and Valle del Cauca departments, Colombia. Nine semi-structured expert interviews with leprosy health professionals were conducted in May-June 2023 in Colombia. Thematic analysis was performed to analyse the interview results. Our analysis highlighted that the main reasons for delay at the health system-level included accessibility issues to obtain a diagnosis, lack of expertise by health staff, and barriers related to the organisation of the care pathway. Individual – and community-level factors included a lack of leprosy awareness among the general population and leprosy-related stigma. Diagnostic delay consists of a fluid interplay of various factors. Structural changes within the health system, such as organising integral leprosy care centres and highlighting leprosy in the medical curriculum, as well as awareness-related interventions among the general population, might help reducing diagnostic delays.

Abstract Following a request from the European Commission, EFSA was asked to deliver a scientific opinion on the safety of sepiolite as a technological feed additive for all animal species. In 2022, the Panel on Additives and Products or Substances used in Animal Feed (FEEDAP) delivered an Opinion on the safety and efficacy of the same additive. The Panel concluded that sepiolite used as a feed additive is safe for the consumers and the environment, and efficacious as a thickener‐suspending agent, binder and anticaking agent in feed for all animal species under the proposed conditions of use. The additive was not considered an eye or skin irritant. However, it was considered a respiratory irritant, a respiratory and dermal sensitiser; owing to the dusting potential and its silica content, the additive was considered a risk by inhalation. Regarding the target species, in the previous Opinion, the Panel concluded on the safety of the additive for dairy ruminants. However, no conclusion could be drawn for all other species/categories. Based on the tolerance studies in chickens for fattening, weaned piglets and trout evaluated in the current assessment, and the one in dairy cows previously assessed, the Panel concluded that the inclusion of sepiolite at the maximum recommended level of 20,000 mg/kg complete feed is safe for all animal species.

Abstract Following a request from the European Commission, EFSA was asked to deliver a scientific opinion on the safety and efficacy of Saccharomyces cerevisiae CNCM I‐1079 as a zootechnical feed additive for dogs and all other Canidae. The additive is intended for use in feed for dogs and all other Canidae at a proposed minimum inclusion level of 1 × 109 CFU per kg of complete feed. Saccharomyces cerevisiae is considered by EFSA to be suitable for the qualified presumption of safety approach to safety assessment. Since the identity of the active agent has been clearly established and no concerns are expected from other components of the product, the additive is considered safe for the target species. Since the additive is intended to be used only in feed for dogs and other non‐food‐producing animals, an assessment of the safety for the consumer and the environment is not needed. The non‐coated form of the additive was shown to be non‐irritant to skin and eyes. No conclusion can be drawn on the eye irritation potential of the coated form of the additive due to the lack of data. The additive in both forms, should be considered a skin and respiratory sensitiser and any exposure through skin and respiratory tract is considered a risk. The Panel was not in the position to conclude on the efficacy of Saccharomyces cerevisiae CNCM I‐1079 at the proposed conditions of use.

Abstract This guidance document is intended to assist the applicant in preparing and presenting an application, as foreseen in Article 7.6 of Regulation (EC) No 1831/2003, for the authorisation of additives for use in animal nutrition. It specifically covers the assessment of the efficacy of feed additives.

Abstract Following a request from the European Commission, EFSA was asked to deliver a scientific opinion on the safety and efficacy of the alpha‐amylase (Ronozyme® HiStarch CT/L) produced with a genetically modified strain of Bacillus licheniformis (DSM 34315) as a zootechnical feed additive for chickens for fattening, turkeys for fattening and minor growing poultry species. The additive is available in two forms, a coated thermotolerant granulate formulation and a liquid formulation. The production strain and its DNA were not detected in an intermediate concentrated product representative of the final formulations. The Panel on Additives and Products or Substances used in Animal Feed (FEEDAP) concluded that the final product did not trigger safety concerns with regard to the genetic modification. The Panel concluded that Ronozyme® HiStarch CT/L is safe for chickens for fattening, turkeys for fattening and minor growing poultry species at the recommended inclusion level of 80 KNU/kg complete feed. The use of Ronozyme® HiStarch CT/L in animal nutrition under the proposed conditions of use is safe for the consumers and the environment. The additive in any form was shown to be non‐irritant to the skin and the solid form was shown to be non‐irritant to the eyes. No conclusions could be drawn on the potential of the liquid form to be irritant to the eyes or on the potential of both forms of the additive to be dermal sensitisers due to lack of data. Owing to the proteinaceous nature of the active substance, the additive in either form was considered to be a respiratory sensitiser and any exposure by inhalation is considered a risk. In the absence of data, the Panel was not in the position to conclude on the efficacy of Ronozyme® HiStarch CT/L for chickens for fattening, turkeys for fattening and minor growing poultry species.

Abstract Following a request from the European Commission, EFSA was asked to deliver a scientific opinion on the safety and efficacy of a feed additive consisting of Quillaja saponaria powder and Yucca schidigera powder (Magni‐Phi®) for all avian species (to slaughter age/weight, or to the point of lay) and ornamental birds, as a zootechnical additive (digestibility enhancer and other zootechnical additives). The EFSA Panel on Additives and Products or Substances used in Animal Feed (FEEDAP) concluded that the additive is safe for chickens for fattening at the level of 250 mg/kg complete feed with a margin of safety of 20 assuming that the additive contains 3.58% of saponins. This conclusion was extrapolated to all growing poultry species and ornamental birds. The Panel concluded that the use of the feed additive in animal nutrition at 250 mg/kg complete feed is of no concern for the safety for the consumer and the environment. The Panel also concluded that the additive is not irritant to skin, but irritant to the eyes and to the respiratory system. Due to the lack of data, the FEEDAP Panel could not conclude on the skin sensitisation potential of the additive. The FEEDAP Panel was not in the position to conclude on the efficacy of the additive for all poultry species and ornamental birds.
