An abstract on the study relating to the interesting features and host interaction of pyrenochaeta lycopersici genome assembly is presented.

Pyrenochaeta lycopersici (Gerlach and Schneider 1964), a soilinhabiting ascomycete, is the causal agent of an important disease of tomato and other solanaceous crops worldwide, named corky root rot (CRR). Yield lossess up to 40-70% have been reported in certain years. The fungus attacks the host roots, inducing necrotic lesions that spread throughout the entire system. A first draft of P. lycopersici genome assembly was obtained from short sequence reads using a paired-end Illumina platform. The total assembly, obtained with Velvet, was 56 Mb with a N50 of 70 kb. An in silico prediction using GeneMarkES allowed the identification about 15,000 genes with an average length of 1,500-1,700 bases. In addition to the fungal genome, RNAs from tomato roots extracted eight days post infection with a P. lycopersici isolate and RNAs from mycelium grown in absence of contact with the host plant, have been sequenced. Analysis of RNA-seq data with Cufflinks identified more than 15,000 putative transcripts 80% of which overlap the in silico predictions. Putative transcripts were functionally annotated using the fungal protein database of UniProt. Results, although preliminary, show that de novo assembly of a large size genome as that of a filamentous fungus may be successfully obtained by paired-end sequencing and the resulting data can be used for investigating the fungal lifestyle during pathogenesis and for comparative studies with other important fungal pathogens. This study evaluates the quality and readability of informed consent documents generated by AI platforms ChatGPT-4 and Bard Gemini Advanced compared to those written by a first-year oral surgery resident for common oral surgery procedures. The evaluation, conducted by 18 experienced oral and maxillofacial surgeons, assessed consents for accuracy, completeness, readability, and overall quality. ChatGPT-4 consistently outperformed both Bard and human-written consents. ChatGPT-4 consents had a median accuracy score of 4 [IQR 4-4], compared to Bard's 3 [IQR 3-4] and human's 4 [IQR 3-4]. Completeness scores were higher for ChatGPT-4 (4 [IQR 4-5]) than Bard (3 [IQR 3-4]) and human (4 [IQR 3-4]). Readability was also superior for ChatGPT-4, with a median score of 4 [IQR 4-5] compared to Bard and human consents, both at 4 [IQR 4-4] and 4 [IQR 3-4], respectively. The Gunning Fog Index for ChatGPT-4 was 17.2 [IQR 16.5-18.2], better than Bard's 23.1 [IQR 20.5-24.7] and the human consents' 20 [IQR 19.2-20.9]. Overall, ChatGPT-4's consents received the highest quality ratings, underscoring AI's potential in enhancing patient communication and the informed consent process. Despite their diverse assumptions, clinical psychology approaches share the goal of mental health promotion. The literature highlights their usefulness, but also some issues related to their effectiveness, such as their difficulties in monitoring psychological change. The elective strategy for activating and managing psychological change is the clinical question. But how do different types of questions foster psychological change? This work tries to answer this issue by studying therapist-patient interactions with a ML model for text analysis. The goal was to investigate how psychological change occurs thanks to different types of questions, and to see if the ML model recognized this difference in analyzing patients' answers to therapists' clinical questions. The experimental dataset of 14,567 texts was divided based on two different question purposes, splitting answers in two categories: those elicited by questions asking patients to start describing their clinical situation, or those from asking them to detail how they evaluate their situation and mental health condition. The hypothesis that these categories are distinguishable by the model was confirmed by the results, which corroborate the different valences of the questions. These results foreshadow the possibility to train ML and AI models to suggest clinical questions to therapists based on patients' answers, allowing the increase of clinicians' knowledge, techniques, and skills.

Brucella abortus is an important zoonotic pathogen that infects cattle and buffaloes. In Italy the application of eradication programs combined with vaccination has greatly contributed to reduce the incidence of brucellosis. However, despite the eradication programs brucellosis continue to persist with a high endemicity in some areas of Italy including the province of Caserta. In the present study the genomes of 44 B. abortus strains isolated from different outbreak cases that affected the province of Caserta were sequenced to characterize the genetic diversity of the Brucella strains circulating during the period from 2017 to 2022. The relatedness among these isolates was compared to 52 publicly available genomes of Italian B. abortus isolates. Background: Auditory verbal hallucinations (AVHs) are a significant symptom of various psychological conditions, often stigmatized and misunderstood. Moving beyond traditional psychological, psychotherapeutic and psychiatric approaches, recent research shifts focus on understanding AVHs through community perspectives and the resulting stigmatization. This research approach is crucial for better support and understanding of AVHs, however it still suffers from the lack of a rigorous and shared methodology for studying and reducing stigma., Methods: Our study, part of the Italian "PsicoVoice" project, aims to investigate community discourses on AVHs, in order to observe whether and to what extent they are drivers of stigmatisation processes. Engaging 268 participants with direct (hearers) and indirect (such as relatives and professionals) experiences of AVHs, the research analyzes a corpus of 54,320 instances using MADIT: a text analysis methodology which is both qualitative and quantitative. MADIT allows for an innovative examination of the rhetorical-argumentative structures within narratives, producing an index for measuring the narratives' practical impact on people' interactions around AVHs., Results: The analysis revealed that the overall community discourses are predominantly shaped by absolute and personal belief-driven modalities. This way of conveying sense, even with non-necessarily-judgmental words, contributes to a stigmatizing environment for individuals with AVHs, cementing a static representation dominated by personal opinions and reducing the potential for more nuanced, diverse interactions about AVHs., Conclusion: The study's findings underscore the importance of addressing the narrative structures within community discourses. By intervening in these narratives, there is potential to shift towards a less stigmatizing social construction of AVHs. Background: ADHD is the most common childhood neurodevelopmental disorder. The symptomatology makes the management of ADHD particularly demanding in school, so teachers' training programs have been widely implemented. Nevertheless, these interventions could lead teachers to concentrate on the dysfunctional elements of these students, exposing them to the risk of stigmatisation. Conceptualising stigma and inclusion as narrative processes, the present study observed how teacher ADHD training texts, endorsed by the Italian government, impact on the inclusion process of students., Methods: The research analysed a corpus of N = 31,261 text occurrences and focused on three areas: (1) ADHD as a clinical condition; (2) the impact of ADHD characteristics in the scholastic setting; (3) interventions to manage ADHD criticalities in school settings. To observe the interactive processes fostered by the narratives under scrutiny, we used Dialogic Science and MADIT methodology, since they allow us to measure the language use modalities through an index: the Dialogical Weight (dW). The value of dW ranges between 0.1 (min) and 0.9 (max) and is linked to the potential outcomes of inclusion for students with ADHD. A low dW accounts for narratives entrenched in personal beliefs presented as absolute truths, undermining inclusion of students with ADHD. In contrast, high dW signals language interaction relying on sharable elements, able to foster social unity and diminish stigma., Results: The results yielded a critical discursive configuration, both in general and for the three distinct areas. We measured an overall Dialogical Weight of 0.4dW and, for the three areas (1) = 0.3dW; (2) = 0.3dW; (3) = 0.4dW. The analysed text does not maximise the triggering of inclusive interactions, as they rely on individual references and present one's narrative as the sole plausible perspective: reinforcing already existing positions and exposing to the risk of stereotyping of the pupils., Conclusions: The study highlighted how the ADHD training materials analysed, focusing on a purely informational and clinical approach, lose in effectiveness with respect to generating inclusive school settings.
