Abstract

Developmental control of cell proliferation is crucial, and abnormal principal cell proliferation may contribute to cystogenesis in polycystic kidney disease. This study investigates roles of cAMP and its primary effector, cAMP-dependent protein kinase (protein kinase A; PKA), in control of cell proliferation in filter-grown noncystic (NC) and cystic (CY)-derived principal cell cultures. These cultures had similar cAMP pathway characteristics upstream of PKA subunit distribution but differed in predicted PKA subtype distribution. Functionally, cultures were proliferative before polarization, with constitutively higher proliferation in CY cultures. NC cultures achieved levels similar to those of CY cultures on pharmacological manipulation of cAMP production or PKA activation or inhibition of PKA subtype I activity. Inhibition of overall PKA activity, or of PKA subtype II anchoring, diminished cAMP/PKA-mediated proliferation in NC cultures but had no effect on CY cultures. Polarized CY monolayers remained proliferative, but NC monolayers lost responsiveness. No large proliferation changes resulted from treatments of polarized cultures; however, polarized NC and CY cultures differed in poststimulation handling of PKA catalytic and type II regulatory subunits. Our results support PKA subtype regulation of prepolarization proliferation in NC principal cells and altered regulation of PKA in CY cells and suggest that differences at or downstream of PKA can contribute to altered proliferation in a developmental renal disease. epidermal growth factor receptor; polycystic kidney disease; protein kinase subtypes; immunolocalization

Abstract

Tesis (Tecnólogo Médico, Especialidad en Oftalmología) La Organización Mundial de la Salud (OMS) considera Baja Visión a pacientes con una Agudeza Visual (AV) entre 0.3 y 0.05 y/o con un Campo Visual (CV) inferior a 20°, la ceguera se considera aquella visión menor de 0,05 y el término de Ceguera Legal considera aquella visión menor a 0,1 o que independientemente de que su visión sea mejor, tenga un CV inferior a 10°. Para estos conceptos se considera el mejor ojo y con la mejor corrección. Las principales causas de baja visión y ceguera asociadas a la edad, pueden ser: Cataratas, Degeneración macular, Retinopatía diabética, Miopía magna, Glaucoma, entre otras. Como consecuencia de estas patologías es preciso incorporar la rehabilitación visual (en los casos que se estimen convenientes), para la obtención del máximo aprovechamiento del resto visual, potenciando la autonomía personal y su integración social. La organización de la Salud en Chile está a cargo del Sistema Nacional de Servicios de Salud (SNSS) mientras que, existen entidades con una organización independiente que tienen sus propios beneficios de Salud como el Sistema de Salud de las Fuerzas Armadas y de Orden. El objetivo principal de este estudio es analizar la presencia de baja visión y ceguera en pacientes mayores de 65 años, beneficiarios del sistema de salud de las Fuerzas Armada de Chile (Hospital Naval Almirante Nef), en el período comprendido entre Enero del 2010 hasta Junio del 2014, para su inserción en programas de rehabilitación visual. Se realizó un estudio estadístico de la población mayor a 65 años con un "n" muestra! de 318 pacientes para una población total de 1.600 pacientes de ambos sexos, con un nivel de confianza del 95%. En la población analizada según su AV, existe un 12% del total de pacientes mayores a 65 años que presentan baja visión, cuya principal causa fue catarata con una prevalencia de un 42%. Existe un 1% del total de pacientes mayores a 65 años que presentan ceguera, cuya principal causa fue glaucoma. Por último, hay un 87% y un 70% de pacientes estudiados con baja visión que según AV y CV respectivamente, no pertenece a un programa de rehabilitación visual. Finalmente a partir de la investigación se concluye que, existen muy pocos estudios a nivel nacional acerca de la prevalencia de baja visión, ceguera y ceguera legal, que permitan estimar la realidad de la discapacidad visual. Por lo que, los datos que se obtuvieron de la población estudiada, no posibilitan hacer un parangón preciso. Cabe la necesidad para futuras investigaciones abarcar una gran red hospitalaria para dar un mayor sustento a los datos, así considerar la importancia de implementar planes y estrategias de rehabilitación visual que cubra las necesidades de la población, con el fin de que puedan optar a una mejor calidad de vida.

29. Na+−K+−Cl−co-transport in the intestine of a marine teleost

Abstract

A key element in the absorption of salt and water by various epithelia is the presence, in the apical or brush border membrane, of a mechanism for directly coupling the inward movements of Na+and Cl−(ref. 1). In this way the downhill gradient for Na+from lumen to cell, which is maintained by the cell's Na+/K+pump, can provide the energy for cellular accumulation and transepithelial transport of Cl−. A requirement for K+at the serosal surface of the epithelium has long been accepted as essential to the operation of the Na+/K+pump and therefore to salt absorption. Until very recently, however, the effect of luminal K+on NaCl absorption has not been explored. We show here that luminal K+stimulates NaCl absorption in the intestine of a marine teleost, the winter flounder, Pseudopleuronectes americanus. Furthermore, K+uptake across the brush border membrane depends on the presence of both Na+and Cl−in the bathing medium and is inhibited by furosemide, which also inhibits the coupled uptake of Na+and Cl−(ref. 2). Thus, flounder intestine seems to have a Na+−K+−Cl−co-transport system similar to that demonstrated in erythrocytes3–5, cultured tumour cells6, cultured kidney cells7and squid giant axon8. Electrophysiological studies of the thick ascending limb of Henlé's loop in mammalian kidney9and the diluting segment in amphibian kidney10also indicate that luminal K+is required for salt absorption. Co-transport of Na+, K+and Cl−may therefore be characteristic of a variety of cells and tissues, including salt-absorbing epithelia.
