G-quadruplexes (G4s) are helical four-stranded structures forming from guanine-rich nucleic acid sequences, which are thought to play a role in cancer development and malignant transformation. Most current studies focus on G4 monomers, yet under suitable and biologically relevant conditions G4s undergo multimerization. Here, we address the structural, conformational and energetic features of G4 multimers formed in solutions by the human telomere sequence. A novel multi-technique approach is used combining Small Angle X-ray Scattering (SAXS) and circular dichroism experiments with coarse-grained simulations, to provide quantitative information about large-scale structural features and the stability of G4 multimers. The latter show a significant polydispersity with an exponential distribution of contour lengths, suggesting a step-growth polymerization. On increasing DNA concentration, the strength of G4 stacking interaction increases, as well as the number of the units in the aggregates, with dimers and trimers as the most probable forms. At the same time, a variation of G4 conformation is observed. Our findings provide a quantitative picture of human telomere G4 multimers, which must be accounted for to achieve a rational design of anticancer drugs targeting DNA structures.

Poly(N-isopropylacrylamide) (PNIPAM) is a synthetic polymer that is widely studied for its thermoresponsive character. However, recent works also reported evidence of a low temperature (protein-like) dynamical transition around 225 K in concentrated PNIPAM suspensions, independently of the polymer architecture, i.e., both for linear chains and for microgels. In this work, we investigate water-polymer interactions by extensive differential scanning calorimetry (DSC) measurements of both systems, in order to understand the effect of the different topological structures on the solution behaviour, in particular regarding crystallization and melting processes. In addition, we compare protiated and deuterated microgels, in both water and deuterated water. The DSC results are complemented by dynamic light scattering experiments, which confirm that the selective isotopic substitution differently affects the solution behaviour. Our findings highlight the important role played by the polymer architecture on the solution behaviour: indeed, microgels turn out to be more efficient confining agents, able to avoid water crystallization in a wider concentration range with respect to linear chains. Altogether, the present data will be valuable to interpret future low-temperature investigations of PNIPAM dispersions, particularly by neutron scattering experiments., Comment: 33 pages, 10 figures

Combining elastic incoherent neutron scattering and differential scanning calorimetry, we investigate the occurrence of the volume phase transition (VPT) in very concentrated PNIPAM microgel suspensions, from a polymer weight fraction of 30~wt\% up to dry conditions. Although samples are arrested at the macroscopic scale, atomic degrees of freedom are equilibrated and can be probed in a reproducible way. A clear signature of the VPT is present as a sharp drop of the mean square displacement of PNIPAM hydrogen atoms obtained by neutron scattering. As a function of concentration, the VPT gets smoother as dry conditions are approached whereas the VPT temperature shows a minimum at about 43~wt\%. This behavior is qualitatively confirmed by calorimetry measurements. Molecular dynamics simulations are employed to complement experimental results and gain further insights into the nature of the VPT, confirming that it involves the formation of an attractive gel state between the microgels. Overall, these results provide evidence that the VPT in PNIPAM-based systems can be detected at different time- and length-scales as well as in overcrowded conditions.

Combining elastic incoherent neutron scattering experiments at different resolutions with molecular dynamics simulations, we report the observation of a protein-like dynamical transition in linear chains of Poly(N-isopropylacrylamide). We identify the onset of the transition at a temperature $T_d$ of about 225~K. Thanks to a novel global fit procedure, we find quantitative agreement between measured and calculated polymer mean-squared displacements at all temperatures and time resolutions. Our results confirm the generality of the dynamical transition in macromolecular systems in aqueous environments, independently of the internal polymer topology.

We present the concept of a novel time-focusing technique for neutron spectrometers, which allows to disentangle time-focusing from beam divergence. The core of this approach is a double rotating-crystal monochromator that can be used to extract a larger wavelength band from a white beam, thus providing a higher flux at the sample compared to standard time-of-flight instruments, yet preserving energy resolution and beam collimation. The performances of a spectrometer based on this approach are quantitatively discussed in terms of possible incident wavelengths, flux at the sample and $(Q,E)$-resolution. Analytical estimates suggest flux gains of about one order of magnitude at comparable resolutions in comparison to conventional time-of-flight spectrometers. Moreover, the double monochromator configuration natively shifts the sample away from the source line-of-sight, thus significantly improving the signal-to-noise ratio. The latter, in combination with a system that does not increase the beam divergence, brings the further advantage of a cleaner access to the low-$Q$ region, which is recognized to be of fundamental interest for magnetism and for disordered materials, from glasses to biological systems.

The long debated protein dynamical transition was recently found also in non-biological macromolecules, such as poly-N-isopropylacrylamide (PNIPAM) microgels. Here, by using atomistic molecular dynamics simulations, we report a description of the molecular origin of the dynamical transition in these systems. We show that PNIPAM and water dynamics below the dynamical transition temperature Td are dominated by methyl group rotations and hydrogen bonding, respectively. By comparing with bulk water, we unambiguously identify PNIPAM-water hydrogen bonding as the main responsible for the occurrence of the transition. The observed phenomenology thus crucially depends on the water-macromolecule coupling, being relevant to a wide class of hydrated systems, independently from the biological function.

A low-temperature dynamical transition has been reported in several proteins. We provide the first observation of a `protein-like' dynamical transition in nonbiological aqueous environments. To this aim we exploit the popular colloidal system of poly-N-isopropylacrylamide (PNIPAM) microgels, extending their investigation to unprecedentedly high concentrations. Owing to the heterogeneous architecture of the microgels, water crystallization is avoided in concentrated samples, allowing us to monitor atomic dynamics at low temperatures. By elastic incoherent neutron scattering and molecular dynamics simulations, we find that a dynamical transition occurs at a temperature $T_d\sim250$~K, independently from PNIPAM mass fraction. However, the transition is smeared out on approaching dry conditions. The quantitative agreement between experiments and simulations provides evidence that the transition occurs simultaneously for PNIPAM and water dynamics. The similarity of these results with hydrated protein powders suggests that the dynamical transition is a generic feature in complex macromolecular systems, independently from their biological function.

A large area ring-imaging Cherenkov detector has been designed to provide clean hadron identification capability in the momentum range from 3 GeV/c up to 8 GeV/c for the CLAS12 experiments at the upgraded 12 GeV continuous electron beam accelerator facility of Jefferson Laboratory. The adopted solution foresees a novel hybrid optics design based on aerogel radiator, composite mirrors and high-packed and high-segmented photon detectors. Cherenkov light will either be imaged directly (forward tracks) or after two mirror reflections (large angle tracks). We report here the results of the tests of a large scale prototype of the RICH detector performed with the hadron beam of the CERN T9 experimental hall for the direct detection configuration. The tests demonstrated that the proposed design provides the required pion-to-kaon rejection factor of 1:500 in the whole momentum range., Comment: 15 pages, 23 figures, to appear on EPJA

The NESSiE Collaboration has been setup to undertake a conclusive experiment to clarify the {\em muon--neutrino disappearance} measurements at short baselines in order to put severe constraints to models with more than the three--standard neutrinos. To this aim the current FNAL--Booster neutrino beam for a Short--Baseline experiment was carefully evaluated by considering the use of magnetic spectrometers at two sites, near and far ones. The detector locations were studied, together with the achievable performances of two OPERA--like spectrometers. The study was constrained by the availability of existing hardware and a time--schedule compatible with the undergoing project of multi--site Liquid--Argon detectors at FNAL. The settled physics case and the kind of proposed experiment on the Booster neutrino beam would definitively clarify the existing tension between the $\nu_{\mu}$ disappearance and the $\nu_e$ appearance/disappearance at the eV mass scale. In the context of neutrino oscillations the measurement of $\nu_{\mu}$ disappearance is a robust and fast approach to either reject or discover new neutrino states at the eV mass scale. We discuss an experimental program able to extend by more than one order of magnitude (for neutrino disappearance) and by almost one order of magnitude (for antineutrino disappearance) the present range of sensitivity for the mixing angle between standard and sterile neutrinos. Experimental tests of Bell-type inequalities distinguishing between quantum mechanics and local realistic theories remain of considerable interest if performed on massive particles, for which no conclusive result has yet been obtained. Only two-particle experiments may specifically test the concept of spatial nonlocality in quantum theory, whereas single-particle experiments may generally test the concept of quantum noncontextuality. Here we have performed the first Bell-type experiment with a beam of thermal-neutron pairs in the singlet state of spin, as originally suggested by J. S. Bell. These measurements confirm the quantum-theoretical predictions, in agreement with the results of the well-known polarization experiments carried out on optical photons years ago.

This report describes the scientific aims and potentials as well as the preliminary technical design of IRIDE, an innovative tool for multi-disciplinary investigations in a wide field of scientific, technological and industrial applications. IRIDE will be a high intensity 'particle factory', based on a combination of a high duty cycle radio-frequency superconducting electron linac and of high energy lasers. Conceived to provide unique research possibilities for particle physics, for condensed matter physics, chemistry and material science, for structural biology and industrial applications, IRIDE will open completely new research possibilities and advance our knowledge in many branches of science and technology. IRIDE will contribute to open new avenues of discoveries and to address most important riddles: What does matter consist of? What is the structure of proteins that have a fundamental role in life processes? What can we learn from protein structure to improve the treatment of diseases and to design more efficient drugs? But also how does an electronic chip behave under the effect of radiations? How can the heat flow in a large heat exchanger be optimized? The scientific potential of IRIDE is far reaching and justifies the construction of such a large facility in Italy in synergy with the national research institutes and companies and in the framework of the European and international research. It will impact also on R&D work for ILC, FEL, and will be complementarity to other large scale accelerator projects. The dynamical structure factor of solid $^{15}$N$_{2}$ in the $\alpha$ phase ($T<35$K) is measured at the IN4 time-of-flight spectrometer at the Institut Laue Langevin, and the potential performance of this substance as a UCN converter is assessed. The cross-section to down-scatter neutrons to ultra-cold neutron energies is determined as a function of incident energy, as well as the up-scattering mean free path. The UCN production cross-section is found to be approximately 20% of that of deuterium. However, UCN with energy 181 neV have an up-scattering mean free path of 46 cm at $T=5.9$ K, which is $\sim20$ times larger than deuterium. Therefore, a large volume $\alpha-^{15}$N$_{2}$ source may produce an improved UCN density if sufficient isotopic purity can be achieved., Comment: 7 pages, 6 figures

A new experiment with an intense ~2 GeV neutrino beam at CERN SPS is proposed in order to definitely clarify the possible existence of additional neutrino states, as pointed out by neutrino calibration source experiments, reactor and accelerator experiments and measure the corresponding oscillation parameters. The experiment is based on two identical LAr-TPCs complemented by magnetized spectrometers detecting electron and muon neutrino events at Far and Near positions, 1600 m and 300 m from the proton target, respectively. The ICARUS T600 detector, the largest LAr-TPC ever built with a size of about 600 ton of imaging mass, now running in the LNGS underground laboratory, will be moved at the CERN Far position. An additional 1/4 of the T600 detector (T150) will be constructed and located in the Near position. Two large area spectrometers will be placed downstream of the two LAr-TPC detectors to perform charge identification and muon momentum measurements from sub-GeV to several GeV energy range, greatly complementing the physics capabilities. This document describes the technical layout and the expected performance of the Straw Tube Tracker (STT), the main tracking detector of the PANDA target spectrometer. The STT encloses a Micro-Vertex-Detector (MVD) for the inner tracking and is followed in beam direction by a set of GEM-stations. The tasks of the STT are the measurement of the particle momentum from the reconstructed trajectory and the measurement of the specific energy-loss for a particle identification. Dedicated simulations with full analysis studies of certain proton-antiproton reactions, identified as being benchmark tests for the whole PANDA scientific program, have been performed to test the STT layout and performance. This proposal describes an experimental search for sterile neutrinos beyond the Standard Model with a new CERN-SPS neutrino beam. The experiment is based on two identical LAr-TPC's followed by magnetized spectrometers, observing the electron and muon neutrino events at 1600 and 300 m from the proton target. This project will exploit the ICARUS T600, moved from LNGS to the CERN "Far" position. An additional 1/4 of the T600 detector will be constructed and located in the "Near" position. Two spectrometers will be placed downstream of the two LAr-TPC detectors to greatly complement the physics capabilities. Spectrometers will exploit a classical dipole magnetic field with iron slabs, and a new concept air-magnet, to perform charge identification and muon momentum measurements in a wide energy range over a large transverse area. In the two positions, the radial and energy spectra of the nu_e beam are practically identical. Comparing the two detectors, in absence of oscillations, all cross sections and experimental biases cancel out, and the two experimentally observed event distributions must be identical. Any difference of the event distributions at the locations of the two detectors might be attributed to the possible existence of {\nu}-oscillations, presumably due to additional neutrinos with a mixing angle sin^2(2theta_new) and a larger mass difference Delta_m^2_new. The superior quality of the LAr imaging TPC, in particular its unique electron-pi_zero discrimination allows full rejection of backgrounds and offers a lossless nu_e detection capability. The determination of the muon charge with the spectrometers allows the full separation of nu_mu from anti-nu_mu and therefore controlling systematics from muon mis-identification largely at high momenta., Comment: Experiment proposal

Tensions in several phenomenological models grew with experimental results on neutrino/antineutrino oscillations at Short-Baseline (SBL) and with the recent, carefully recomputed, antineutrino fluxes from nuclear reactors. At a refurbished SBL CERN-PS facility an experiment aimed to address the open issues has been proposed [1], based on the technology of imaging in ultra-pure cryogenic Liquid Argon (LAr). Motivated by this scenario a detailed study of the physics case was performed. We tackled specific physics models and we optimized the neutrino beam through a full simulation. Experimental aspects not fully covered by the LAr detection, i.e. the measurements of the lepton charge on event-by-event basis and their energy over a wide range, were also investigated. Indeed the muon leptons from Charged Current (CC) (anti-)neutrino interactions play an important role in disentangling different phenomenological scenarios provided their charge state is determined. Also, the study of muon appearance/disappearance can benefit of the large statistics of CC muon events from the primary neutrino beam. Results of our study are reported in detail in this proposal. We aim to design, construct and install two Spectrometers at "NEAR" and "FAR" sites of the SBL CERN-PS, compatible with the already proposed LAr detectors. We describe an experiment confirming the evidence of the antibunching effect on a beam of non interacting thermal neutrons. The comparison between the results recorded with a high energy-resolution source of neutrons and those recorded with a broad energy-resolution source enables us to clarify the role played by the beam coherence in the occurrence of the antibunching effect., Comment: 4 pages, 3 figures
