Abstract Objectives The red panda is currently the only surviving member of the Ailuridae family in the Caniformia suborder. In this study, we provide data on anatomical, morphometric, histological and histochemical examination of the orbital region, eyelids, orbital gland, and eye tunics in two adult males Ailurus fulgens fulgens from the Wroclaw Zoological Garden, Poland. Methods The study involved morphometric analysis of the eyeball and selected accessory organs of the eye, along with analysis of the bony orbit, including its morphometry, macroscopic, and microscopic evaluation. Microscopic evaluation encompassed histological and histochemical staining, with the former involving hematoxylin & eosin (H&E), Movat pentachrome, picro-Mallory trichrome, Fontana-Masson, and the latter including PAS, AB pH 1.0, AB pH 2.5; AB pH 2.5/PAS, and HDI. Results The upper (UE) and lower (LE) eyelids presented well-developed tarsal glands, sebaceous glands, and a characteristic simple alveolar gland (producing a mucous secretion). The palpebral part of the lacrimal gland was present. A single lymphoid follicle was observed only in the upper eyelids. The superficial gland of the third eyelid (SGTE) was a multilobar acinar complex that produces mucous secretion and is contained within the interlobular ducts of numerous aggregates of lymph nodes. The third eyelid (TE) was T-shaped and composed of hyaline tissue, containing CALT. The lacrimal gland (LG) also revealed a multilobar acinar complex that produced mucous secretion, with a single lymphoid follicle. The cornea consisted of 4 layers, as Bowman’s membrane was absent. The Vogt palisades composed of 7–10 layers of epithelial cells were demonstrated. The pupil was horizontally ovoid at rest (post-mortem). The sphincter pupil and the dilator pupil were well developed. Macroscopically, the tapetum lucidum appeared as a milky, non-opalescent crescent. Histologically, the choroidal tapetum lucidum cellulosum consisted of 5 to 9 layers of loosely packed oval cells. The retina showed a composition similar to that of terrestrial nocturnal carnivores. Conclusions The results of our research indicate that the anatomical features of the eye and orbital region in the red panda share similarities with those described in the Musteloidea clade, as well as the Canidae and Ursidae families.

Background: The orbital glands, viz. lacrimal gland, superficial and deep gland of third eyelid (LG, SGT and HG), are important for normal eye functions. These glands have different functions in various animals. The information about the enzyme histochemical nature of prenatal orbital glands in Indian buffalo seems to be unavailable. Therefore, the study was planned on orbital glands of six full term recently died fetuses from animals with dystocia. Methods: The frozen sections of all these glands were subjected to standard localization protocols for Alkaline Phosphatase (AKPase), Glucose 6 phosphatase (G-6-Pase), Lactate dehydrogenase (LDH), Succinate dehydrogenase (SDH), Glucose 6 phosphate dehydrogenase (G-6-PD), Nicotinamide Adenine Dinucleotide Hydrogen Diaphorase (NADHD), Nicotinamide Adenine Dinucleotide Phosphate Hydrogen diaphorase (NADPHD), Dihydroxy phenylalanine oxidase (DOPA-O), Tyrosinase, non-specific esterase (NSE) and Carbonic anhydrase (CAse). Results: The results revealed a mixed spectrum of reaction for the above enzymes in LG, SGT and HG which ranged from moderate (for LDH in SGT) to intense (for most of the enzymes in all three glands). However, DOPA-O, Tyrosinase and CAse did not show any reaction. From the present study, it can be postulated that the orbital glands of fetus have a high activity of metabolism as it has many developmental and functional activities which were mediated with the higher activity of the enzymes involved. G. Kingsley Noble reported the possibility of orbital gland involvement in reproduction of spelerpine salamanders, particularly Eurycea, through rudimentary gross examination of the heads of male and female salamanders. In his report, Noble provided details from only one taxon of salamander, E. bislineata. To examine the variability of orbital glands with secondary sexual function in Eurycea, orbital glands of E. longicauda were assessed for sexual dimorphism and a distinct seasonal secretory cycle that mirrored the reproductive cycle. Orbital glands were found not to be sexually dimorphic in E. longicauda and possessed constant secretory activity in males and females throughout the year. Known secondary sexual glands (mental glands, caudal courtship glands, and cloacal glands) were sexually dimorphic between males and females and possessed a secretory cycle that could be correlated with events of the reproductive cycle, particularly mating activity. Thus, no evidence was found for orbital gland involvement in reproduction in E. longicauda. Orbital glands of both male and female E. longicauda are morphologically identical and produce copious lipoproteins throughout the year. Orbital gland secretions are potentially involved in lubrication of the eye and the prevention of fluid evaporation from the eye as described for lipoprotein secretions in other vertebrate lineages. Nasolacrimal ducts are a terrestrial vertebrate adaptation and appear to have co-evolved with orbital glands. Although plethodontid salamanders possess orbital glands, a recent study concluded that plethodontid salamanders lack nasolacrimal ducts. Functionally, the absence of nasolacrimal ducts closes the route for orbital gland secretion passage into the nasal and vomeronasal organ cavities. Orbital glands have been implicated in enhancement of vomeronasal function so loss could have important implications for communication. Multiple older studies depict or discuss nasolacrimal ducts in plethodontid salamanders. Interestingly, the only consensus between recent and older literature is that Desmognathus lacks nasolacrimal ducts. To determine if plethodontid salamanders truly lack nasolacrimal ducts, we sectioned plethodontid salamander heads for general histological examination of species from the majority of the plethodontid tribes. From our representative sample, we found only two species that completely lacked nasolacrimal ducts (Desmognathus fuscus and Eurycea tynerensis) and one species that possessed nasolacrimal ducts that ended blindly before reaching the nasal cavities (E. spelaea). Bayesian ancestral state reconstruction resulted in the presence of nasolacrimal ducts on the branch leading to Plethodontidae and both subfamilies within Plethodontidae, with two independent losses in Desmognathus and Eurycea.
