1. Screening of an Alcohol Tolerant and High-yield L-lactic Acid Strain and Optimization of Culture Medium

Abstract

In order to increase the yield of L-lactic acid and reduce the production cost of L-lactic acid, a strain of Lactobacillus rhamnosus AK-0779 with alcohol tolerance and high yield of L-lactic acid was screened and domesticated. Corn distiller's grains were used as the nitrogen source for the fermentation medium of strain AK-0779 instead of part of yeast powder. On the basis of single factor experiments, three factors and three levels response surface optimization experiments were carried out on the addition of glucose, yeast powder and corn distiller's grains. The results showed that the optimal fermentation medium was glucose 9.80%, corn distiller's grains 0.98%, yeast powder 1.72%, L-lactic acid yield 78.91 g/L and sugar acid conversion rate 80.52%. There was no significant difference in the yield of L-lactic acid compared with yeast powder, which was 82.36 g/L, indicating that corn distiller's grains could effectively replace part of yeast powder as the nitrogen source of fermentation medium and reduce the production cost of L-lactic acid.

Abstract

A previous study found that a biocontrol bacterium, Bacillus pumilus HR10, inhibited the Sphaeropsis shoot blight disease of pine, and the fermentation broth of HR10 strain contained protein antifungal substances. The optimal formulation of the fermentation medium for the antagonistic substance of B. pumilus HR10 was finally obtained by single-factor test, Packett-Burman test, steepest ascent test and Box-Behnken Design (BBD) response surface test, and the best formulation of the fermentation medium for the antagonistic substance of B. pumilus HR10 was 12 g/L corn meal, 15 g/L beef extract and 13 g/L magnesium sulfate, with a predicted bacterial inhibition rate of 89%. The fermentation filtrate of B. pumilus HR10 cultured with the optimized medium formulation was verified to have an inhibition rate of (87.04 ± 3.2) % on the growth of Sphaeropsis sapinea by three replicate tests. The antagonistic crude protein of B. pumilus HR10 were further isolated and identified using HiTrap Capto Q strong Ion-Exchange Chromatography and LC–MS-MS, and it was speculated that glycoside hydrolase (Ghy), beta-glucanase (Beta), arabinogalactan endonuclease β-1,4-galactanase (Arab), and immunosuppressant A (ImA) are proteins with antagonistic activity against S. sapinea in the B. pumilus HR10. Abstract

The present study aimed to perform the acidic adaptive evolution on a ε-PL-producing strain of Streptomyces diastatochromogenes T17 to increase its ε-PL production, verify the acid resistance abilities of the evolutionary strains and optimize the fermentation medium. The results showed that the dry cell weights of three evolutionary strains were significantly higher than that of the original strain after 4 days of fermentation (initial pH of 4.0). Besides, the better mycelium growth performance and spore maturity at plates with different pH values (5.0, 4.5 and 4.0) also proved that the acid resistance abilities of the evolutionary strains were stronger than that of the original strain. The ε-PL production of evolutionary S. diastatochromogenes AE 56 after fermentation medium optimization reached 1.08 g/L, increasing 12.5%. [ABSTRACT FROM AUTHOR]

Abstract

• Strain CGMCC 17276 is a high-yield BC-producing novel species of Komagataeibacter. • bcs II and bcs III operon showed high transcriptional expression under static and agitated conditions. • The highest yield of BC production reached using glycerol as single carbon source. • Waste glycerol was successfully used as a sustainable strategy for BC production. Bacterial cellulose (BC) has received considerable attention as an environment-friendly, biodegradable nanomaterial. In this study, the strain Komagataeibacter sp. nov. CGMCC 17276, which showed rapid cell growth and high BC-production ability, was isolated and classified into a novel species in the Komagataeibacter genus. Four BC synthase operons were annotated using whole-genome analysis, partially explaining the high BC yield of strain CGMCC 17276. Operons bcs Ⅱ and bcs Ⅲ showed high transcriptional levels under static and agitated culture conditions, indicating their importance in BC synthesis. Of the eight suitable carbon sources identified by whole-genome analysis, the highest BC production was achieved using glycerol as a single carbon source. Finally, waste glycerol was successfully used as an eco-friendly and sustainable strategy for BC production. This study provides valuable insights into the mechanism of BC synthesis, genetic structure of BC-producing strains, and industrialization of BC production using an eco-friendly and low-cost strategy. Abstract

Indonesia was known as a producer and exporter of cacao, but nowadays Indonesia had an issue in cacao production caused by microorganisms (Phytophtora palmivora) that capable of reducing the amount of cacao production each year. This study follows up a previous study that had found compounds that are actively against at Phytophtora palmivora from Aspergillus oryzae metabolites. In this study the compound will be produced in greater numbers to optimize fermentation medium. The first stage was conducted to determine the profile of the active compound and to analyze the best carbon, nitrogen source for Aspergillus oryzae fermentation medium. The second stage was designed by using Response Surface Method (RSM) with Central Composite Design (CCD) to optimize two variables in the fermentation medium, glucose concentration (the best carbon source) and the concentration of peptone (best nitrogen source). The expected response was the most active compounds percentage of the area. Data analysis was performed by Design Expert DX 7.1.5 program. The first stage of this study presumed that Tenuazonic acid compounds with a molecular weight of 198.1126 gram.mol was an antifungal compound produced by Aspergillus oryzae fungus to prevent Phythopthora palmivora fungus pathogens cacao pods. In addition the results are that glucose was the best carbon source (mean % relative area of antifungal compounds: 85.39%) and that peptone was selected as the best source of nitrogen (mean % relative area of antifungal compounds: 91.07%). The second stage showed the optimum conditions of fermentation medium antifungal Aspergillus oryzae was the composition of glucose 35.25 g/L and peptone 16.7 g/L. The combination of both compositions would yield a mean response of laboratory data % relative area of antifungal compounds 91.2217% and the estimated value (prediction) by Design Expert models 92.2936%. Difference in the estimated values (the response of the model) to the value of the results of response in laboratory experiments at 1.1614%, then it could be concluded that the model used was appropriate and able to explain the applied experimental data.ABSTRAKIndonesia dikenal sebagai produsen dan eksportir buah kakao, tetapi saat ini Indonesia memiliki masalah dalam produksi buah kakao dikarenakan adanya mikroorganisme penyebab penyakit (Phytophtora palmivora) yang menurunkan jumlah produksi buah kakao tiap tahunnya. Penelitian ini melanjutkan penelitian sebelumnya yang telah menemukan senyawa aktif yang bersifat antijamur terhadap Phytophtora palmivora dari metabolit Aspergillus oryzae. Pada penelitian ini senyawa tersebut akan diproduksi dalam jumlah yang lebih banyak melalui optimasi media fermentasi. Penelitian tahap I dilakukan untuk mengetahui profil senyawa aktif yang dominan dan mendapatkan sumber karbon, nitrogen yang terbaik untuk media fermentasi Aspergillus oryzae. Penelitian tahap II dirancang menggunakan Response Surface Method (RSM) dengan Central Composite Design (CCD) untuk mengoptimasi 2 variabel dalam media fermentasi yaitu konsentrasi glukosa (sumber karbon terbaik) dan konsentrasi pepton (sumber nitrogen terbaik). Respon yang diinginkan adalah persentase luas area senyawa aktif yang paling besar. Analisis data dilakukan dengan program Design Expert D.X. 7.1.5. Tahap I penelitian ini menduga bahwa senyawa Tenuazonic acid dengan bobot molekul 198.1126 gram.mol-1 adalah senyawa antijamur yang dihasilkan jamur Aspergillus oryzae untuk jamur patogen buah kakao Phythopthora palmivora. Selain itu, diperoleh hasil bahwa glukosa merupakan sumber karbon terbaik (rerata % relatif luas area senyawa antijamur: 85.39%) dan pepton dipilih sebagai sumber nitrogen yang terbaik (rerata % relatif luas area senyawa antijamur: 91,07%). Penelitian tahap II menghasilkan kondisi optimum media fermentasi antijamur dari A. oryzae dengan komposisi glukosa sebesar 35,25 gram/L dan pepton sebesar 16,7 gram/L. Kombinasi kedua komposisi tersebut akan menghasilkan data laboratorium respon rerata % relatif luas area senyawa antijamur sebesar 91,2217% dan nilai dugaan (prediksi) oleh model Design Expert sebesar 92,2936%. Perbedaan nilai dugaan (respon dari model) dengan nilai respon hasil percobaan di laboratorium sebesar 1,1614% sehingga dapat disimpulkan bahwa model yang digunakan telah sesuai dan mampu menjelaskan data percobaan yang digunakan.
