Rational manipulation and assembly of discrete colloidal particles into architected superstructures have enabled several applications in materials science and nanotechnology. Optical manipulation techniques, typically operated in fluid media, facilitate the precise arrangement of colloidal particles into superstructures by using focused laser beams. However, as the optical energy is turned off, the inherent Brownian motion of the particles in fluid media impedes the retention and reconfiguration of such superstructures. Overcoming this fundamental limitation, we present on-demand, three-dimensional (3D) optical manipulation of colloidal particles in a phase-change solid medium made of surfactant bilayers. Unlike liquid crystal media, the lack of fluid flow within the bilayer media enables the assembly and retention of colloids for diverse spatial configurations. By utilizing the optically controlled temperature-dependent interactions between the particles and their surrounding media, we experimentally exhibit the holonomic microscale control of diverse particles for repeatable, reconfigurable, and controlled colloidal arrangements in 3D. Finally, we demonstrate tunable light-matter interactions between the particles and 2D materials by successfully manipulating and retaining these particles at fixed distances from the 2D material layers. Our experimental results demonstrate that the particles can be retained for over 120 days without any change in their relative positions or degradation in the bilayers. With the capability of arranging particles in 3D configurations with long-term stability, our platform pushes the frontiers of optical manipulation for distinct applications such as metamaterial fabrication, information storage, and security.

We study the hydrodynamic coupling of neighboring micro-beads placed in a multiple optical trap setup allowing us to precisely control the degree of coupling and directly measure time-dependent trajectories of entrained beads. We performed measurements on configurations with increasing complexity starting with a pair of entrained beads moving in one dimension, then in two dimensions, and finally a triplet of beads moving in two dimensions. The average experimental trajectories of a probe bead compare well with the theoretical computation, illustrating the role of viscous coupling and setting timescales for probe bead relaxation. The findings also provide direct experimental corroborations of hydrodynamic coupling at large, micrometer spatial scales and long, millisecond timescales, of relevance to, e.g., microfluidic device design and hydrodynamic-assisted colloidal assembly, improving the capability of optical tweezers, and understanding the coupling between micrometer-scale objects within a living cell. Phonon lasers, as mechanical analogues of optical lasers, are unique tools for not only fundamental studies of the emerging field of phononics but also diverse applications such as deep-ocean monitoring, force sensing, and biomedical ultrasonics. Recently, nonlinear phonon-lasing effects were observed in an opto-levitated micro-sphere, i.e., the spontaneous emerging of weak signals of high-order phonon harmonics in the phonon lasing regime. However, both the strengths and the quality factors of the emerging phonon harmonics are very poor, thus severely hindering their potential applications in making and utilizing nonlinear phonon-laser devices. Here we show that, by applying a single-colour electronic injection to this levitated system, giant enhancement can be achieved for all higher-order phonon harmonics, with more than 3 orders enhanced brightness and 5 orders narrowed linewidth. Such an electronically-enhanced phonon laser is also far more stable, with frequency stability extended from a dozen of minutes to over 1 h. More importantly, higher-order phonon correlations, as an essential lasing feature, are confirmed to be enhanced by the electronic injection as well, which as far as we know, has not been reported in previous works using this technique. This work, providing much stronger and better-quality signals of coherent phonon harmonics, is a key step towards controlling and utilizing nonlinear phonon lasers for applications such as phonon frequency combs, broadband phonon sensors, and ultrasonic bio-medical diagnosis. Advances in physics have been significantly driven by state‐of‐the‐art technology, and in photonics and X‐ray science this calls for the ability to manipulate the characteristics of optical beams. Orbital angular momentum (OAM) beams hold substantial promise in various domains such as ultra‐high‐capacity optical communication, rotating body detection, optical tweezers, laser processing, super‐resolution imaging etc. Hence, the advancement of OAM beam‐generation technology and the enhancement of its technical proficiency and characterization capabilities are of paramount importance. These endeavours will not only facilitate the use of OAM beams in the aforementioned sectors but also extend the scope of applications in diverse fields related to OAM beams. At the FERMI Free‐Electron Laser (Trieste, Italy), OAM beams are generated either by tailoring the emission process on the undulator side or, in most cases, by coupling a spiral zone plate (SZP) in tandem with the refocusing Kirkpatrick–Baez active optic system (KAOS). To provide a robust and reproducible workflow to users, a Hartmann wavefront sensor (WFS) is used for both optics tuning and beam characterization. KAOS is capable of delivering both tightly focused and broad spots, with independent control over vertical and horizontal magnification. This study explores a novel non‐conventional 'near collimation' operational mode aimed at generating beams with OAM that employs the use of a lithographically manufactured SZP to achieve this goal. The article evaluates the mirror's performance through Hartmann wavefront sensing, offers a discussion of data analysis methodologies, and provides a quantitative analysis of these results with ptychographic reconstructions. Red blood cells (RBCs) are the most abundant human blood cells. RBC aggregation and deformation strongly determine blood viscosity which impacts hemorheology and microcirculation. In turn, RBC properties depend on different endogenous and exogenous factors. One such factor is nitric oxide (NO), which is mainly produced by endothelial cells (EC) from L-arginine amino acid in the circulatory system. Since the mechanisms of the RBC-endothelium interplay are not clear up to date and considering its possible clinical importance, the aims of this study are to investigate in vitro: (1) The effect of L-arginine induced NO on RBC aggregation and adhesion to endothelium; (2) the NO effect on RBC aggregation and deformation induced by L-arginine and sodium nitroprusside without the presence of endothelium in the samples. The RBC aggregation and adhesion to a monolayer of EC were studied using optical tweezers (OT). The RBC deformability and aggregation without endothelium in the samples were studied using the flow chamber method and Myrenne aggregometer. We confirmed that NO increases deformability and decreases aggregation of RBCs. We showed that the soluble guanylate cyclase pathway appears to be the only NO signaling pathway involved. In the samples with the endothelium, the "bell-shaped" dependence of RBC aggregation force on L-arginine concentration was observed, which improves our knowledge about the process of NO production by endothelium. Additionally, data related to L-arginine accumulation by endothelium were obtained: Necessity of the presence of extracellular L-arginine stated by other authors was put under question. In our study, NO decreased the RBC-endothelium adhesion, however, the tendency appeared to be weak and was not confirmed in another set of experiments. Mutation in oncogene KRas plays a crucial role in the occurrence and progression of numerous malignant tumors. Malignancy involves changes in cell mechanics for extensive cellular deformation during metastatic dissemination. We hypothesize that oncogene KRas mutations are intrinsic to alterations in cellular mechanics that promote malignant tumor generation and progression. Here, we demonstrate the use of optical tweezers coupled with a confocal fluorescence imaging system and gene interference technique to reveal that the mutant KRas protein can be transported between homogeneous and heterogeneous tumor cells by tunneling nanotubes (TNTs), resulting in a significant reduction of membrane tension and acceleration of membrane phospholipid flow in the recipient cells. Simultaneously, the changes in membrane mechanical properties of the tumor cells also enhance the metastatic and invasive ability of the tumors, which further contribute to the deterioration of the tumors. This finding helps to clarify the association between oncogene mutations and changes in the mechanical properties of tumor cells, which provides a theoretical basis for the development of cancer treatment strategies. Here, we present a laser confocal fluorescence system integrated with optical tweezers to observe the transfer of mutant KRasG12D protein from mutant cells to wild-type cells through TNTs. Malignancy involves changes in cell mechanics for extensive cellular deformation during metastatic dissemination. Our results demonstrate a significant decrease in membrane tension and an increase in membrane phospholipid flow in recipient cells. These alterations in mechanical properties augment the migration and invasive capabilities of tumor cells, contributing to tumor malignancy. Our findings propose that cellular mechanical properties could serve as new markers for tumor development, and targeting membrane tension may hold potential as a therapeutic strategy. We report on a method of measuring the stiffness in photophoretic force tweezers in air by using an experimental configuration with two counter-propagating hollow beams. By setting the optical traps of both beams in the same focal plane of the camera, we are able to record the flight procedure of a trapped particle into the other trap after the initial trapping beam is switched off. Then, the stiffness of optical trap can be obtained by fitting the relationship of particle position vs time during the flight procedure. It is found that the measurement is tolerant to the distance between the traps and laser power variation. In addition, the measured stiffness is demonstrated applicable in determining the size of aerosol particles. The reliable method provided in this work is not only useful in studying the physical behavior of signal particle under atmosphere condition but also meaningful in the applications of aerosol studies, such as in situ aerosol characterization and the local surrounding environment sensing. Templated DNA repair that occurs during homologous recombination and replication stress relies on RAD51. RAD51 activity is positively regulated by BRCA2 and the RAD51 paralogs. The Shu complex is a RAD51 paralog-containing complex consisting of SWSAP1, SWS1, and SPIDR. We demonstrate that SWSAP1-SWS1 binds RAD51, maintains RAD51 filament stability, and enables strand exchange. Using single-molecule confocal fluorescence microscopy combined with optical tweezers, we show that SWSAP1-SWS1 decorates RAD51 filaments proficient for homologous recombination. We also find SWSAP1-SWS1 enhances RPA diffusion on ssDNA. Importantly, we show human sgSWSAP1 and sgSWS1 knockout cells are sensitive to pharmacological inhibition of PARP and APE1. Lastly, we identify cancer variants in SWSAP1 that alter Shu complex formation. Together, we show that SWSAP1-SWS1 stimulates RAD51-dependent high-fidelity repair and may be an important new cancer therapeutic target. The human Shu complex promotes homologous recombination by regulating RAD51. Here the authors reveal that the Shu complex proteins, SWSAP1-SWS1, decorate the RAD51 filament on ssDNA and facilitate its strand exchange reaction by stimulating RPA diffusion on ssDNA. It is well established that the interaction of light with matter generally imparts mechanical forces and associated torques. Where laser light is involved, such effects can be deployed for the microscale and nanoscale manipulation of matter, giving rise to notable applications in wide areas of physics, chemistry, and the life sciences. The diversity of applications is enriched not only by the complexity of the constitution of matter, but also by the structure of the laser light itself, according to its polarisation, amplitude, and phase. Usually, optical tweezers for trapping atoms or nanoparticles are based on the focusing of a Gaussian laser beam (GB). The optical trap is characterised by its longitudinal stability (LS), expressed as the ratio of the backward axial gradient and the forward scattering forces. Replacing the GB with a LGp0 beam (one central peak surrounded by p rings) does not improve the LS because the on-axis intensity distribution is the same whatever the mode order p. However, it has been recently demonstrated that a restructured LGp0 beam can improve greatly the LS. In this paper, we consider the restructuring of a LGp0 beam when passing through a simple binary diffracting optical element called a circular π-plate (CPP). For a particular radius of the dephasing zone of the CPP, it is found that the LS is multiplied by a factor corresponding to a relative increase of about 220% to 320%. [ABSTRACT FROM AUTHOR]

Optical tweezers exploit light–matter interactions to trap particles ranging from single atoms to micrometer-sized eukaryotic cells. For this reason, optical tweezers are a ubiquitous tool in physics, biology, and nanotechnology. Recently, the use of deep learning has started to enhance optical tweezers by improving their design, calibration, and real-time control as well as the tracking and analysis of the trapped objects, often outperforming classical methods thanks to the higher computational speed and versatility of deep learning. In this perspective, we show how cutting-edge deep learning approaches can remarkably improve optical tweezers, and explore the exciting, new future possibilities enabled by this dynamic synergy. Quantum computation and quantum communication are expected to provide users with capabilities inaccessible by classical physics. However, scalability to larger systems with many qubits is challenging. One solution is to develop a quantum network consisting of small-scale quantum registers containing computation qubits that are reversibly interfaced to communication qubits. In this study, we report on a register that uses both optical tweezers and optical lattices to deterministically assemble a two-dimensional array of atoms in an optical cavity. Harnessing a single atom-addressing beam, we stimulate the emission of a photon from each atom and demonstrate multiplexed atom-photon entanglement with a generation-to detection efficiency approaching 90%. AbstractThe widespread use of polymer flooding has resulted in a large number of produced liquids consisting of crude oil emulsions and oily wastewater. Efficient flocculants are required to treat these produced liquids till the oily wastewater satisfies the requirements for reinjection formation in order to maintain the regular manufacturing process. In this work, an unsaturated polyether macromonomer (PEP-30) was prepared by ring-opening polymerization using isobutenol as a starting material. Subsequently, a novel flocculant (PMAP) was obtained by copolymerization of dimethylamino propylmethacrylamide (DMAPMA) and PEP-30. For the produced liquids with different crude oil emulsion content, the oil removal rate of PMAP at a dosage of 150 mg/L was more than 75%, and the oil removal rate and versatility were better than those of conventional cationic flocculants. Furthermore, the findings of the flocculation mechanism investigation demonstrated that PMAP had a low interfacial elastic modulus and a large interfacial adsorption mass. Specifically, the optical tweezers were used to detect the interaction force between oil droplets. PMAP effectively strengthened the attraction between oil droplets, extending their duration in contact during flocculation and promoting their coalescence. [ABSTRACT FROM AUTHOR]

Förster resonance energy transfer (FRET) is ubiquitous in optical processes in the natural world. A methodology is proposed that uses an optical force to control its efficiency without contact in an aqueous solution of a thermo‐responsive polymer, polyvinyl methyl ether (PVME). Focusing irradiation of a near infrared laser beam into the solution results in the formation and trapping of a single polymer droplet. The polymer concentration in the droplet is controllable by changing the optical force from the laser light is shown. The polarity inside the droplet decreases with increasing the optical force. When small amounts of dye molecules, D (energy donor) and A (energy accepter), are dissolved in the polymer solution, D and A are absorbed (extracted) into the droplet. The concentrations of D and A are controllable by the optical force. Based on this mechanism, FRET between D and A is induced successfully, and can control the FRET efficiency. Finally, the modulation of fluorescence color of the droplet from blue, green, yellow, to an orange color is demosntrated simply by changing the optical force. Optical tweezers enable noncontact trapping of microscale objects using light. It is not known how tightly it is possible to three-dimensionally (3D) trap microparticles with a given photon budget. Reaching this elusive limit would enable maximally stiff particle trapping for precision measurements on the nanoscale and photon-efficient tweezing of light-sensitive objects. Here, we customize the shape of light fields to suit specific particles, with the aim of optimizing trapping stiffness in 3D. We show, theoretically, that the confinement volume of microspheres held in sculpted optical traps can be reduced by one to two orders of magnitude. Experimentally, we use a wavefront shaping-inspired strategy to passively suppress the Brownian fluctuations of microspheres in every direction concurrently, demonstrating order-of-magnitude reductions in their confinement volumes. The chiral light field has attracted great attention owing to its interaction with chiral matter. The generation of chiral light fields with rich structures has become crucial as it can expand application scenarios. Herein, we introduce a chiral optical vortex lattice. As a whole, the optical vortex lattice has a chiral intensity distribution, with each spiral arm having sub-vortices (chiral phase). By using an expansion factor to adjust the involute of a circular lattice, this helical optical vortex lattice can be continuously varied from a circular lattice. The chirality of intensity and phase can be controlled independently. Furthermore, the optical tweezers using the lattice demonstrate the capability of sub-vortices to manipulate particle movement, with the chiral intensity determining the trajectory of particle motion. The scattering of x linearly polarized non-diffracting Lommel beam by a perfect electromagnetic conductor (PEMC) sphere is analyzed within the generalized Lorenz–Mie theory (GLMT) framework. The electromagnetic fields of the incident Lommel beams are expressed by employing spherical vector wave functions (SVWFs) and beam shape coefficients (BSCs). These BSCs are mainly considered in the expression of shaped beams in the GLMT with slight beam axicon angle. The scattered electromagnetic fields are constructed using unknown scattering field coefficients (SFCs) and SVWFs. These undetermined SFCs are calculated employing boundary conditions (BCs) at the PEMC surface. The expressions of the far-region/far-zone scattering intensity (FSI) are given and computed. The influences of Lommel beam configuration parameters like as beam order, beam half cone angle, beam asymmetry parameter, beam center coordinates positions, and PEMC sphere size parameter in addition to PEMC scalar admittance on the FSI are analyzed and discussed. The experience of this research work will be quite helpful in analyzing optical radiation force and torque, electromagnetic scattering, light manipulation, antenna engineering domain, optical tweezers, particle interaction, and trapping. Vector Lommel beams are a class of non-diffracting laser beams, characterized by their ability to propagate without any changes in their intensity profile. Understanding, the effect of the polarization state on the interaction between these beams and particles is of significant interest. In this context, we use the polarization states that obey Maxwell's equations to investigate the scattering of vector Lommel beams by a dielectric homogeneous spherical particle within the framework of generalized Lorenz-Mie theory (GLMT). The quadrature method is employed to derive closed-form expressions for the beam shape coefficients of both linearly and circularly polarized Lommel beams. These coefficients are then used to compute the differential scattering cross section of the scattered intensity of vector Lommel beams by spherical particles. The results of this study indicate that the scattered intensity is heavenly influenced by the polarization states of vector Lommel beams. Additionally, these findings offer significant benefits for applications across various fields, including the scattering of non-diffracting beams by objects, laser light scattering, optical tweezers, and the trapping and manipulation of particles through the optical forces exerted by such beams. Microrheology, the study of material flow at micron scales, has advanced significantly since Robert Brown's discovery of Brownian motion in 1827. Mason and Weitz's seminal work in 1995 established the foundation for microrheology techniques, enabling the measurement of viscoelastic properties of complex fluids using light-scattering particles. However, existing techniques face limitations in exploring very slow dynamics, crucial for understanding biological systems. Here, we present a proof of concept for a novel microrheology technique called "Optical Halo", which utilises a ring-shaped Bessel beam created by optical tweezers to overcome existing limitations. Through numerical simulations and theoretical analysis, we demonstrate the efficacy of the Optical Halo in probing viscoelastic properties across a wide frequency range, including low-frequency regimes inaccessible to conventional methods. The study aims to determine how chitosan impacts pectin hydrogel's ability to attach peritoneal leukocytes, activate complement, induce hemolysis, and adsorb blood proteins. The hydrogels PEC-Chi0, PEC-Chi25, PEC-Chi50, and PEC-Chi75 were prepared by placing a mixture solution of 4% pectin and 4% chitosan in a ratio of 4:0, 3:1, 2:2, and 1:3 in a solution of 1.0 M CaCl2. Chitosan was found to modify the mechanical properties of pectin–calcium hydrogels, such as hardness and cohesiveness-to-adhesiveness ratio. Chitosan in the pectin–calcium hydrogel caused pH-sensitive swelling in Hanks' solution. The PEC-Chi75 hydrogel was shown to adsorb serum proteins at pH 7.4 to a greater extent than other hydrogels. PEC-Chi75's strong adsorption capacity was related to lower peritoneal leukocyte adherence to its surface when compared to other hydrogels, showing improved biocompatibility. Using the optical tweezers approach, it was shown that the force of interaction between pectin–chitosan hydrogels and plasma proteins increased from 10 to 24 pN with increasing chitosan content from 0 to 75%. Thus, the properties of pectin–calcium hydrogel, which determine interactions with body tissues after implantation, are improved by the addition of chitosan, making pectin–chitosan hydrogel a promising candidate for smart biomaterial development. Trapping and manipulating micro-objects and achieving high-precision measurements of tiny forces and displacements are of paramount importance in both physical and biological research. While conventional optical tweezers rely on tightly focused beams generated by bulky microscope systems, the emergence of flat lenses, particularly metalenses, has revolutionized miniature optical tweezers applications. In contrast to traditional objectives, the metalenses can be seamlessly integrated into sample chambers, facilitating flat-optics-based light manipulation. In this study, we propose an experimentally realized transmissive and polarization-insensitive water-immersion metalens, constructed using adaptive nano-antennas. This metalens boasts an ultra-high numerical aperture of 1.28 and achieves a remarkable focusing efficiency of approximately 50 % at a wavelength of 532 nm. Employing this metalens, we successfully demonstrate stable optical trapping, achieving lateral trapping stiffness exceeding 500 pN/(μm W). This stiffness magnitude aligns with that of conventional objectives and surpasses the performance of previously reported flat lenses. Furthermore, our bead steering experiment showcases a lateral manipulation range exceeding 2 μm, including a region of around 0.5 μm exhibiting minimal changes in stiffness for smoothly optical manipulation. We believe that this metalens paves the way for flat-optics-based optical tweezers, simplifying and enhancing optical trapping and manipulation processes, attributing ease of use, reliability, high performance, and compatibility with prevalent optical tweezers applications, including single-molecule and single-cell experiments. An experimental and computational optical pump-probe model is constructed, which utilizes two ultrafast pump pulses within the repetition period of a mode-locked laser to generate electron spin polarization. This report focuses on the effects of resonant spin amplification induced by an infinite train of the two-pump pulses. The first pump pulse is used to generate ordinary resonant spin amplification spectra, while the second pump pulse is used to manipulate the generated spectra. This model gives control of the accumulation of spin polarized electrons along a magnetic field by selecting the temporal separation of the two-pump pulses. The computational model accurately predicts and agrees with the experimental results, which shows manipulation of resonant spin peaks that are no longer entirely dependent on the external magnetic field. This two-pump model and the associated manipulations of resonant spin peaks can be used as a platform to construct and conceptualize resonant spin amplification-based optospintronic devices and applications. Membrane tension is thought to be a long-range integrator of cell physiology. Membrane tension has been proposed to enable cell polarity during migration through front-back coordination and long-range protrusion competition. These roles necessitate effective tension transmission across the cell. However, conflicting observations have left the field divided as to whether cell membranes support or resist tension propagation. This discrepancy likely originates from the use of exogenous forces that may not accurately mimic endogenous forces. We overcome this complication by leveraging optogenetics to directly control localized actin-based protrusions or actomyosin contractions while simultaneously monitoring the propagation of membrane tension using dual-trap optical tweezers. Surprisingly, actin-driven protrusions and actomyosin contractions both elicit rapid global membrane tension propagation, whereas forces applied to cell membranes alone do not. We present a simple unifying mechanical model in which mechanical forces that engage the actin cortex drive rapid, robust membrane tension propagation through long-range membrane flows.

Mechanobiology is an emerging multidisciplinary field that involves the study of the mechanisms by which biological organisms sense and respond to mechanical stimuli. In recent years, this field has seen significant advancements through the application of microfluidics and optical manipulation. Microfluidics enables precise control of channel content and flow with great precision, while optical trapping allow for manipulation of microscopic objects. Combining these disciplines offers new opportunities for studying biological phenomena with reduced scale experiments. However, challenges remain in coordinating microfluidics with optical manipulation within confined spaces, in particular when working with biological entities. To address these limitations, an integrated approach is proposed, using 3D optical manipulation setup, microfluidics and deep learning image recognition to estimate forces experienced by optically trapped objects. By analyzing the bead’s displacement within the flow, forces are quantified using a deep learning algorithm. Experimental results demonstrate force variations based on the position within the chip, revealing the potential for improved understanding of biological mechanisms through characterization of local forces. Optical tweezers are a means to manipulate objects with light. Optical tweezers can obtain a nanometer space, piconewton force, and a millisecond resolution; thus, they are excellently suited for studying biological processes from the single-cell to the single-molecule level. Optical tweezers can screen and manipulate tumor cells, study single molecule, and monitor cancer treatments. Thus, optical tweezers will promote the progress of cancer research and treatments.

Tweezers based on optical, electric, magnetic, and acoustic fields have shown great potential for contactless object manipulation. However, current tweezers designed for manipulating millimeter‐sized objects such as droplets, particles, and small animals exhibit limitations in translation resolution, range, and path complexity. Here, a novel acoustic vortex tweezers system is introduced, which leverages a unique airborne acoustic vortex end effector integrated with a three‐degree‐of‐freedom (DoF) linear motion stage, for enabling contactless, multi‐mode, programmable manipulation of millimeter‐sized objects. The acoustic vortex end effector utilizes a cascaded circular acoustic array, which is portable and battery‐powered, to generate an acoustic vortex with a ring‐shaped energy pattern. The vortex applies acoustic radiation forces to trap and spin an object at its center, simultaneously protecting this object by repelling other materials away with its high‐energy ring. Moreover, The vortex tweezers system facilitates contactless, multi‐mode, programmable object surfing, as demonstrated in experiments involving trapping, repelling, and spinning particles, translating particles along complex paths, guiding particles around barriers, translating and rotating droplets containing zebrafish larvae, and merging droplets. With these capabilities, It is anticipated that the tweezers system will become a valuable tool for the automated, contactless handling of droplets, particles, and bio‐samples in biomedical and biochemical research. Structured vortex light with orbital angular momentum (OAM) shows great promise for high-bandwidth optical communications, quantum information and computing, optical tweezers, microscopy, astronomy, among others. Generating, controlling, and detecting of vortex light by all-electrical means is at the heart of next generation nanophotonic platforms. However, on-chip electrical photodetection of structured vortex light remains challenging. Here, we propose an on-chip photodetector based on 2D broadband thermoelectric material (PdSe2) with a well-designed spin-Hall couplers to directly characterize angular momentum modes of vortex structured light. Photothermoelectric responses in the PdSe2 nanoflake, excited by the focusing surface plasmons, show a magnitude proportional to the total angular momentum modes of the infrared vortex beams, thereby achieving direct detection of spin and orbital angular momentum, as well as the chirality and ellipticity of scalar vortex lights. Our works provide a promising strategy for developing on-chip angular momentum optoelectronic devices, which play a key role in the next-generation high-capacity optical communications, quantum information and computing, imaging, and other photonic systems. Background: Raman tweezers spectroscopy (RTS) is a powerful tool that combines optical tweezers and Raman spectroscopy to study single living cells. RTS has become increasingly popular in biomedical and clinical research due to its high molecular specificity and sensitivity, which enable the study of cell viability, cell deformation, cell-protein, cell-nanoparticle, cell-cell interaction, etc. In transfusion medicine, RTS can give valuable insights into the storage lesions and effects of various preservatives and intravenous fluids on blood cells. Summary: By analyzing the Raman spectra of individual blood cells, RTS can detect changes in the cellular blood components which can be used to monitor the quality of blood products during storage and transfusion. The present review article highlights the principle and clinical applications of RTS in transfusion medicine. The HIV-1 nucleocapsid protein (NC) is a multifunctional viral protein necessary for HIV-1 replication. Recent studies have demonstrated that reverse transcription (RT) completes in the intact viral capsid, and the timing of RT and uncoating are correlated. How the small viral core stably contains the ~10 kbp double stranded (ds) DNA product of RT, and the role of NC in this process, are not well understood. We showed previously that NC binds and saturates dsDNA in a non-specific electrostatic binding mode that triggers uniform DNA self-attraction, condensing dsDNA into a tight globule against extending forces up to 10 pN. In this study, we use optical tweezers and atomic force microscopy to characterize the role of NC's basic residues in dsDNA condensation. Basic residue mutations of NC lead to defective interaction with the dsDNA substrate, with the constant force plateau condensation observed with wild-type (WT) NC missing or diminished. These results suggest that NC's high positive charge is essential to its dsDNA condensing activity, and electrostatic interactions involving NC's basic residues are responsible in large part for the conformation, size, and stability of the dsDNA-protein complex inside the viral core. We observe DNA re-solubilization and charge reversal in the presence of excess NC, consistent with the electrostatic nature of NC-induced DNA condensation. Previous studies of HIV-1 replication in the presence of the same cationic residue mutations in NC showed significant defects in both single- and multiple-round viral infectivity. Although NC participates in many stages of viral replication, our results are consistent with the hypothesis that cationic residue mutations inhibit genomic DNA condensation, resulting in increased premature capsid uncoating and contributing to viral replication defects. The antibacterial effect of nanoparticles is mainly studied on the ensembles of the bacteria. In contrast, the optical tweezer technique allows the investigation of similar effects on individual bacterium. E. coli is a self-propelled micro-swimmer and ATP-driven active microorganism. In this work, an optical tweezer is employed to examine the mechanical properties of E. coli incubated with ZnO and Ag nanoparticles (NP) in the growth medium. ZnO and Ag NP with a concentration of 10 µg/ml were dispersed in growth medium during active log-growth phase of E. coli. This E. coli-NP incubation is further continued for 12 h. The E. coli after incubation for 2 h, 6 h and 12 h were separately studied by the optical tweezer for their mechanical property. The IR laser (λ = 975 nm; power = 100 mW) was used for trapping the individual cells and estimated trapping force, trapping stiffness and corner frequency. The optical trapping force on E. coli incubated in nanoparticle suspension shows linear decreases with incubation time. This work brings the importance of optical trapping force measurement in probing the antibacterial stress due to nanoparticles on the individual bacterium. [ABSTRACT FROM AUTHOR]

In this paper, we discuss a technique for selectively loading a particle into a magneto-gravitational trap using the sublimation of camphor to release particles from a tungsten probe tip directly into the trapping region. This sublimation-activated release (SAR) loading technique makes use of micropositioners with tungsten probe tips, as well as the relatively fast rate of sublimation of camphor at room temperature, to selectively load particles having diameters ranging from 8 to 100 μm or more. The advantages of this method include its ability to selectively load unique particles or particles in limited supply, its low loss compared to alternative techniques, the low speed of the particle when released, and the versatility of its design, which allows for loading into traps with complex geometries. The focusing characteristics of the Bessel–Gaussian (BG) beam, characterized by a tailored space-variant quadratic (SQ) phase, are explored based on the principles of vector diffraction theory. The effects of different parameters on the optical field distribution and the optical gradient force distribution are discussed in detail. The results show that the two intensity peaks will move toward the central region and merge into a single intensity peak when the beam parameter less than 3.2 and will separate along the X-axis when the beam parameter exceeds 3.2. Moreover, an optical trap appears in the focal area, and multiplied small-intensity peaks will be observed due to adjusting the fractional topological charge number. Besides, gradient force evolution differs for the variation of beam parameter and topological charge number. Novel patterns of gradient force emerge as a result, such as force rings, banded gradient force, square array gradient force. Semiflexible fiber gels such as collagen and fibrin have unique nonlinear mechanical properties that play an important role in tissue morphogenesis, wound healing, and cancer metastasis. Optical tweezers microrheology has greatly contributed to the understanding of the mechanics of fibrous gels at the microscale, including its heterogeneity and anisotropy. However, the explicit relationship between micromechanical properties and gel deformation has been largely overlooked. We introduce a unique gel-stretching apparatus and employ it to study the relationship between microscale strain and stiffening in fibrin and collagen gels, focusing on the development of anisotropy in the gel. We find that gels stretched by as much as 15 % stiffen significantly both in parallel and perpendicular to the stretching axis, and that the parallel axis is 2–3 times stiffer than the transverse axis. We also measure the stiffening and anisotropy along bands of aligned fibers created by aggregates of cancer cells, and find similar effects as in gels stretched with the tensile apparatus. Our results illustrate that the extracellular microenvironment is highly sensitive to deformation, with implications for tissue homeostasis and pathology. The inherent fibrous architecture of the extracellular matrix (ECM) gives rise to unique strain-stiffening mechanics. The micromechanics of fibrous networks has been studied extensively, but the deformations involved in its stiffening at the microscale were not quantified. Here we introduce an apparatus that enables measuring the deformations in the gel as it is being stretched while simultaneously using optical tweezers to measure its microscale anisotropic stiffness. We reveal that fibrin and collagen both stiffen dramatically already at ∼10 % deformation, accompanied by the emergence of significant, yet moderate anisotropy. We measure similar stiffening and anisotropy in the matrix remodeled by the tensile apparatus to those found between cancer cell aggregates. Our results emphasize that small strains are enough to introduce substantial stiffening and anisotropy. These have been shown to result in directional cell migration and enhanced force propagation, and possibly control processes like morphogenesis and cancer metastasis. The Sec61 translocon allows the translocation of secretory preproteins from the cytosol to the endoplasmic reticulum lumen during polypeptide biosynthesis. These proteins possess an N‐terminal signal peptide (SP) which docks at the translocon. SP mutations can abolish translocation and cause diseases, suggesting an essential role for this SP/Sec61 interaction. However, a detailed biophysical characterization of this binding is still missing. Here, optical tweezers force spectroscopy was used to characterize the kinetic parameters of the dissociation process between Sec61 and the SP of prepro‐alpha‐factor. The unbinding parameters including off‐rate constant and distance to the transition state were obtained by fitting rupture force data to Dudko–Hummer–Szabo models. Interestingly, the translocation inhibitor mycolactone increases the off‐rate and accelerates the SP/Sec61 dissociation, while also weakening the interaction. Whereas the translocation deficient mutant containing a single point mutation in the SP abolished the specificity of the SP/Sec61 binding, resulting in an unstable interaction. In conclusion, we characterize quantitatively the dissociation process between the signal peptide and the translocon, and how the unbinding parameters are modified by a translocation inhibitor. The self-organization of cells relies on the profound complexity of protein-protein interactions. Challenges in directly observing these events have hindered progress toward understanding their diverse behaviors. One notable example is the interaction between molecular motors and cytoskeletal systems that combine to perform a variety of cellular functions. In this work, we leverage theory and experiments to identify and quantify the rate-limiting mechanism of the initial association between a cargo-bound kinesin motor and a microtubule track. Recent advances in optical tweezers provide binding times for several lengths of kinesin motors trapped at varying distances from a microtubule, empowering the investigation of competing models. We first explore a diffusion-limited model of binding. Through Brownian dynamics simulations and simulation-based inference, we find this simple diffusion model fails to explain the experimental binding times, but an extended model that accounts for the ADP state of the molecular motor agrees closely with the data, even under the scrutiny of penalizing for additional model complexity. We provide quantification of both kinetic rates and biophysical parameters underlying the proposed binding process. Our model suggests that a typical binding event is limited by ADP state rather than physical search. Lastly, we predict how these association rates can be modulated in distinct ways through variation of environmental concentrations and physical properties. Author summary: Cytoskeletal-motor assemblies self-organize to achieve cellular functions ranging from delivering intracellular cargoes to generating forces in mitosis. Advancements in single-molecule experiments have revealed immense detail about motor detachment and stepping, but relatively little regarding attachment. Newly available binding times for individual kinesin motors allow for the evaluation of mechanistic models of the process. We find that a model limited by both diffusive search and a weak-to-strong transition from ADP release best explains the data. The coupled chemo-mechanical nature of this interaction can be modulated more richly than either separately, possibly explaining the diversity and regulation observed in cells. Hierarchical micro/nanostructures have garnered considerable attention for their capabilities in light modulation, but the flexible fabrication of designed optical functional structures at both micro and nano scales remains challenging. Here, a polarization‐modulated patterned laser ablation method complemented by flowing liquid is proposed to fabricate hierarchical microgrooves featuring tunable cross‐sections and engraved surface nanostructures with controllable periods and orientations. The liquid‐assisted ablation counters the shielding effect of ablation debris through laser‐induced microjets, ensuring accurate control of the microgroove's shape by modulating the laser pattern in the focal plane. Simultaneously, the absence of debris also permits the consistent formation of laser‐induced periodic surface structures (LIPSS) across the microgrooves. The LIPSS's period and orientation can be finely adjusted by manipulating the pulse energy and polarization within the patterned laser spot, facilitating the adaptable creation of hierarchical micro/nanostructures for optical application needs. As a demonstration, blazed gratings featuring orientation‐customized LIPSS are fabricated, which exhibit polarization‐dependent diffraction efficiency. The laser fabrication technique offers a highly versatile solution for sculpturing shape‐controllable hierarchical gratings on hard‐to‐machine materials, paving the way for the swift production of customized optical elements. Single‐molecule optoelectronics offers opportunities for advancing integrated photonics and electronics, which also serves as a tool to elucidate the underlying mechanism of light‐matter interaction. Plasmonics, which plays pivotal role in the interaction of photons and matter, have became an emerging area. A comprehensive understanding of the plasmonic excitation and modulation mechanisms within single‐molecule junctions (SMJs) lays the foundation for optoelectronic devices. Consequently, this review primarily concentrates on illuminating the fundamental principles of plasmonics within SMJs, delving into their research methods and modulation factors of plasmon‐exciton. Moreover, we underscore the interaction phenomena within SMJs, including the enhancement of molecular fluorescence by plasmonics, Fano resonance and Rabi splitting caused by the interaction of plasmon‐exciton. Finally, by emphasizing the potential applications of plasmonics within SMJs, such as their roles in optical tweezers, single‐photon sources, super‐resolution imaging, and chemical reactions, we elucidate the future prospects and current challenges in this domain. Myriad DNA-binding proteins undergo dynamic assembly, translocation, and conformational changes while on DNA or alter the physical configuration of the DNA substrate to control its metabolism. It is now possible to directly observe these activities—often central to the protein function—thanks to the advent of single-molecule fluorescence- and force-based techniques. In particular, the integration of fluorescence detection and force manipulation has unlocked multidimensional measurements of protein–DNA interactions and yielded unprecedented mechanistic insights into the biomolecular processes that orchestrate cellular life. In this review, we first introduce the different experimental geometries developed for single-molecule correlative force and fluorescence microscopy, with a focus on optical tweezers as the manipulation technique. We then describe the utility of these integrative platforms for imaging protein dynamics on DNA and chromatin, as well as their unique capabilities in generating complex DNA configurations and uncovering force-dependent protein behaviors. Controlling ultracold atoms with laser light has greatly advanced quantum science. The wavelength of light sets a typical length scale for most experiments to the order of 500 nanometers (nm) or greater. In this work, we implemented a super-resolution technique that localizes and arranges atoms on a sub- 50-nm scale, without any fundamental limit in resolution. We demonstrate this technique by creating a bilayer of dysprosium atoms and observing dipolar interactions between two physically separated layers through interlayer sympathetic cooling and coupled collective excitations. At 50-nm distance, dipolar interactions are 1000 times stronger than at 500 nm. For two atoms in optical tweezers, this should enable purely magnetic dipolar gates with kilohertz speed. In summer fracturing operation, microorganisms such as bacteria can cause the guar gum fracturing fluids to spoil and lose their effectiveness. Bactericides have thus to be used in the fracturing fluids to kill the bacteria. Presently there is no technical means of evaluating the performance of the bactericides. In this study a method based on optical fiber tweezer (OFT) is presented to evaluate the performance of the bactericides. The saprophytic bacteria are first captured before and after adding a bactericide into a fracturing fluid with a conical OFT, the movement pattern of the bacteria is then analyzed at microscopic scale, and this process can be used to invert the effect of the bactericides. Experimental results show that this method can be used to effectively evaluate the activity of the saprophytic bacteria. After adding bactericides into a fracturing fluid, the cells of the saprophytic bacteria become inactive and the movement distance of the saprophytic bacteria is greatly reduced to about only 0.8 – 0.9 μm. For saprophytic bacteria without contacting with bactericides, the movement distance is generally about 12.1 μm. The study proves that the OFT method is effective in evaluating the performance of bactericides, and it provides a reliable theoretical base for the direct evaluation of the performance of bactericides. Optical Bessel beams are used in numerous applications like fluorescence microscopy, material processing and optical trapping. These applications require Bessel beams having a central core with defined full width at half maximum and a defined axial length. Often, the side lobes of Bessel beams, which are associated with their non-diffracting properties, can interfere with the experimental process. We theoretically describe and practically verify the performance of a new refractive optical system to generate zoomable annular ring intensities. The ability to zoom the output ring diameter allows for flexibly choosing the Bessel beam parameters. Secondly, we introduce the use of a Michelson interferometer for destructively interfering Bessel beam side lobes in one direction. If two Bessel beams of zeroth order and first kind are coherently superposed with a small shift with respect to each other, their side lobes are enhanced in one direction and cancelled in the other direction. We suggest that applications like light-sheet microscopy can exploit the axis of destructive interference to improve their contrast. [ABSTRACT FROM AUTHOR]

Pathological processes often change the mechanical properties of cells. Increased rigidity could be a marker of cellular malfunction. Erythrocytes are a type of cell that deforms to squeeze through tiny capillaries; changes in their rigidity can dramatically affect their functionality. Furthermore, differences in the homeostatic elasticity of the cell can be used as a tool for diagnosis and even for choosing the adequate treatment for some illnesses. More accurate types of equipment needed to study biomechanical phenomena at the single-cell level are very costly and thus out of reach for many laboratories around the world. This study presents a simple and low-cost technique to study the rigidity of red blood cells (RBCs) through the application of electric fields in a hand-made microfluidic chamber that uses a capacitor principle. As RBCs are deformed with the application of voltage, cells are observed under a light microscope. From mechanical force vs. deformation data, the elastic constant of the cells is determined. The results obtained with the capacitor-based method were compared with those obtained using optical tweezers, finding good agreement. In addition, P. falciparum-infected erythrocytes were tested with the electric field applicator. Our technique provides a simple means of testing the mechanical properties of individual cells. A kind of surface plasmon waveguide composed of two nested cylindrical dielectric parallel nanowire pairs coated with graphene was designed and studied. The dependence of the mode characteristics and the normalized gradient force of the lowest two modes supported by the waveguide on the parameters involved were analyzed by using the multipole method. To ensure rigor, the finite element method was employed to verify the accuracy of the multipole method, thus confirming its results. The results show that the multipole method is a powerful tool for handling this type of waveguide. The real part of the effective refractive index, the propagation length, the figure of merit, and the normalized gradient force can be significantly affected by the operating wavelength, the Fermi energy of graphene, the waveguide geometric parameters, and the refractive index of the inner dielectric nanowire. Due to the employment of nested dielectric nanowire pairs coated with graphene, this waveguide structure exhibits significant gradient force that surpasses 100 nN·μm−1·mW−1. The observed phenomena can be attributed to the interaction of the field with graphene. Predicting the unknown from the first-person perspective is expected as a necessary step toward machine intelligence, which is essential for practical applications including autonomous driving and robotics. As a human-level task, egocentric action anticipation aims at predicting an unknown action seconds before it is performed from the first-person viewpoint. Egocentric actions are usually provided as verb-noun pairs; however, predicting the unknown action may be trapped in insufficient training data for all possible combinations. Therefore, it is crucial for intelligent systems to use limited known verb-noun pairs to predict new combinations of actions that have never appeared, which is known as compositional generalization. In this article, we are the first to explore the egocentric compositional action anticipation problem, which is more in line with real-world settings but neglected by existing studies. Whereas prediction results are prone to suffer from semantic bias considering the distinct difference between training and test distributions, we further introduce a general and flexible adaptive semantic debiasing framework that is compatible with different deep neural networks. To capture and mitigate semantic bias, we can imagine one counterfactual situation where no visual representations have been observed and only semantic patterns of observation are used to predict the next action. Instead of the traditional counterfactual analysis scheme that reduces semantic bias in a mindless way, we devise a novel counterfactual analysis scheme to adaptively amplify or penalize the effect of semantic experience by considering the discrepancy both among categories and among examples. We also demonstrate that the traditional counterfactual analysis scheme is a special case of the devised adaptive counterfactual analysis scheme. Single particle analysis is essential for a better understanding of the particle transformation process and to predict its environmental impact. In this study, we developed an aerosol optical tweezer (AOT) Raman spectroscopy system to investigate the phase state and morphology of suspended aerosol droplets in real time. The system comprises four modules: optical trapping, reaction, illumination and imaging, and detection. The optical trapping module utilizes a 532 nm laser and a 100 × oil immersion objective to stably trap aerosol droplets within 30 s. The reaction module allows us to adjust relative humidity (RH) and introduce reaction gases into the droplet levitation chamber, facilitating experiments to study liquid–liquid phase transitions. The illumination and imaging module employs a high-speed camera to monitor the trapped droplets, while the detector module records Raman scattering light. We trapped sodium chloride (NaCl) and 3-methyl glutaric acid (3-MGA) mixed droplets to examine RH-dependent morphology changes. Liquid–liquid phase separation (LLPS) occurred when RH was decreased. Additionally, we introduced ozone and limonene/pinene to generate secondary organic aerosol (SOA) particles in situ, which collided with the trapped droplet and dissolved in it. To determine the trapped droplet's characteristics, we utilized an open-source program based on Mie theory to retrieve diameter and refractive index from the observed whispering gallery modes (WGMs) in Raman spectra. It is found that mixed droplets formed core–shell morphology when RH was decreased, and the RH dependence of the droplets' phase transitions generated by different SOA precursors varied. Manipulating thermo-convective fluid flow induced by plasmonic nanostructures under light illumination has garnered significant attention in various fields, such as biomedical sensing, particle trapping, and drug delivery. However, achieving symmetric optical manipulation of fluid flow encounters challenges in certain applications due to the inherent temporal and spatial symmetry in the energy transfer process. Here, a design of plasmonic nanostructures is proposed to achieve a platform for the asymmetric manipulation of thermally induced fluid flow in an optofluidic environment. The difference in fluid flow rate between forward and backward directions is due to the combined effect of the local asymmetry of the heat transfer in multilayer plasmonic nanostructure and nonreciprocity. The nonreciprocity originates from the violation of time-symmetry due to the temperature gradient-induced convection. We show that the asymmetric convective flow can also be achieved when the size of the plasmonic structure enlarges from nanometer to micrometer, and it can be used for efficient particle separation or transportation in microfluidic systems. This study showcases an experimental demonstration of near‐field optical trapping and dynamic manipulation of an individual extracellular vesicle. This is accomplished through the utilization of a plasmonic dielectric nanoantenna designed to support an optical anapole state—a non‐radiating optical state resulting from the destructive interference between electric and toroidal dipoles in the far‐field, leading to robust near‐field enhancement. To further enhance the field intensity associated with the optical anapole state, a plasmonic mirror is incorporated, thereby boosting trapping capabilities. In addition to demonstrating near‐field optical trapping, the study achieves dynamic manipulation of extracellular vesicles by harnessing the thermoelectric effect. This effect is induced in the presence of an ionic surfactant, cetyltrimethylammonium chloride, combined with plasmonic heating. Furthermore, the thermoelectric effect improves trapping stability by a deep trapping potential. In summary, the hybrid plasmonic‐dielectric trapping platform offers a versatile approach for actively transporting, stably trapping, and dynamically manipulating individual extracellular vesicles. In diverse realms of research, such as holographic optical tweezer mechanical measurements, colloidal particle motion state examinations, cell tracking, and drug delivery, the localization and analysis of particle motion command paramount significance. Algorithms ranging from conventional numerical methods to advanced deep-learning networks mark substantial strides in the sphere of particle orientation analysis. However, the need for datasets has hindered the application of deep learning in particle tracking. In this work, we elucidated an efficacious methodology pivoted toward generating synthetic datasets conducive to this domain that resonates with robustness and precision when applied to real-world data of tracking 3D particles. We developed a 3D real-time particle positioning network based on the CenterNet network. After conducting experiments, our network has achieved a horizontal positioning error of 0.0478 μm and a z -axis positioning error of 0.1990 μm. It shows the capability to handle real-time tracking of particles, diverse in dimensions, near the focal plane with high precision. High-order cylindrical vector beams possess flexible spatial polarization and exhibit new effects and phenomena that can expand the functionality and enhance the capability of optical systems. However, building a general analytical model for highly focused beams with different polarization orders remains a challenge. Here, we elaborately develop the vector theory of high-order cylindrical vector beams in a high numerical aperture focusing system and achieve the vectorial diffraction integrals for describing the tight focusing field with the space-variant distribution of polarization orders within the framework of Richards–Wolf diffraction theory. The analytical formulae include the exact three Cartesian components of electric and magnetic distributions in the tightly focused region. Additionally, utilizing the analytical formulae, we can achieve the gradient force, scattering force, and curl-spin force exerted on Rayleigh particles trapped by high-order cylindrical vector beams. These results are crucial for improving the design and engineering of the tightly focused field by modulating the polarization orders of high-order cylindrical vector beams, particularly for applications such as optical tweezers and optical manipulation. This theoretical analysis also extends to the calculation of complicated optical vortex vector fields and the design of diffractive optical elements with high diffraction efficiency and resolution.
