Adrenocorticotropin (ACTH), melanotropins (MSHs), growth hormone (GH) and gonadotropin (GTH) have been identified or cloned from the pituitary gland of sea lampreys (Petromyzon marinus). The present study was designed to gain insights into the functional significance of these hormones through a description of changes in the occurrence and distribution of cells immunoreactive to their antibodies at several different stages of the sea lamprey life cycle. ACTH-like cells and MSH-like cells were distributed in the rostral pars distalis and the pars intermedia, respectively, throughout the life cycle from ammocoetes (larvae) to pre-spawning adults. A large number of ACTH-like cells were observed during the pre-spawning period when animals may experience the highest stressful conditions. On the other hand, the number of MSH-like cells increased markedly during metamorphosis, in accordance with the completion of eye development. A small number of GH-like cells were present in the proximal pars distalis during the larval and metamorphic phases, but the number of cells increased markedly during the parasitic period, which corresponded well with the rapid somatic growth. GTH-like cells were not observed in the pituitary during the larval and metamorphic phases, but were present in the proximal pars distalis of immediately post-metamorphosed animals. Since there was a high accumulation of GTH-like cells in pre-spawning adults, these cells appeared to be involved in gonadotropic functions. The results of changing immunoreactivity during the lamprey life cycle suggest that lamprey adenohypophysial hormones, ACTH, MSH, GH and GTH, may possess biological functions similar to those of more advanced gnathostome vertebrates. Given that lampreys represent the most ancient group of vertebrates, it is most likely that these hormones have been conserved for their functions throughout vertebrate evolution.

Cystathionine gamma-synthase, the first committed enzyme of methionine biosynthesis in higher plants, is encoded by the CGS1 gene in Arabidopsis thaliana. We have shown previously that the stability of the CGS1 mRNA is negatively regulated in response to methionine application [Chiba, Y., Ishikawa, M., Kijima, F., Tyson, R. H., Kim, J., Yamamoto, A., Nambara, E., Leustek, T., Wallsgrove, R. M. & Naito, S. (1999) Science 286, 1371-1374]. To determine whether methionine itself is the effector of the CGS1 exon 1-mediated posttranscriptional regulation, we carried out transfection experiments. The results suggested that, rather than methionine, S-adenosyl-L-methionine (AdoMet), or one of its metabolites, acts as the effector of this regulation. To further identify the actual effector, we exploited the wheat germ in vitro translation system. The effects of various metabolites and analogs of AdoMet were tested by using RNA carrying a CGS1 exon 1-reporter fusion. These tests identified AdoMet as the effector of this regulation. S-adenosyl-L-ethionine, an analog of AdoMet, also had effector activity. A. thaliana mto1 mutants, which are deficient in this regulation, showed a much reduced response to AdoMet in vitro, with a leaky allele showing a less reduced response. RNA translated in vitro in the presence of AdoMet contained a 5'-truncated RNA species, similar to the one that we previously suggested was an in vivo degradation intermediate of CGS1 mRNA. Together, the results show that the basic reactions of CGS1 exon 1-mediated posttranscriptional regulation occur in the wheat germ in vitro translation system, and that AdoMet acts as the effector.

Cystathionine gamma-synthase (CGS) catalyzes the first committed step of Met biosynthesis in plants. We have previously shown that expression of the gene for CGS is feedback-regulated at the level of mRNA stability, and that the amino acid sequence encoded by the first exon of the CGS gene itself is responsible for the regulation (Chiba, Y., Ishikawa, M., Kijima, F., Tyson, R. H., Kim, J., Yamamoto, A., Nambara, E., Leustek, T., Wallsgrove, R. M., and Naito, S. (1999) Science 286, 1371-1374). To identify the functional region within CGS exon 1, deletion analysis was performed. The results showed that the 41-amino acid region of exon 1 highly conserved among plants is necessary and sufficient for the regulation. Analyses of in vivo and in vitro generated mutations that abolish the regulation identified the functionally important amino acids as 11-13 residues within this conserved region. The importance of these residues was confirmed by deletion analysis within the conserved region. These studies identified the functional region of CGS exon 1 required for the posttranscriptional autoregulation of the CGS gene as (A)RRNCSNIGVAQ(I), with uncertainty of the first and last residues. This sequence is almost perfectly conserved among CGS sequences of higher plants but cannot be found elsewhere in the public databases.

Growth hormone (GH), prolactin (PRL) and somatolactin (SL) are members of a pituitary hormone family that are believed to have evolved from a common ancestral gene by duplication and subsequent divergence. Since these hormones are found both in bony fish and cartilaginous fish, their ancestral form(s) should be present in the Agnatha. Thus, although there is no convincing evidence that the lamprey pituitary secretes GH or PRL, GH- and/or PRL-like immunoreactivity was examined in the pituitary of adult sea lampreys (Petromyzon marinus), using antibodies to GHs, PRLs and SL of mammalian and/or fish origins. Our initial attempt with ordinary immunohistochemical procedures failed to detect any positive reactions in the lamprey pituitary. Following the hydrated autoclave pretreatment of the sections, anti-salmon GH, anti-salmon PRL and anti-blue shark GH gave positive reactions in most cells distributed in the dorsal half of the proximal pars distalis. These results suggest that the material immunoreactive to those antibodies is related, to some extent, to GH/PRL, but enhancement of immunoreactivity to reveal this by the hydrated autoclave pretreatment of sections is needed due to low crossreactivity. The similarity of the topographic distributions within the pituitary between lampreys and teleosts suggests that lamprey GH/PRL-like cells are GH cells of the lamprey.

In the lamprey, adrenocorticotropin (ACTH) and melanotropins (MSHs) are produced from two distinct precursors, proopiocortin (POC) and proopiomelanotropin (POM). Both POC and POM have been suggested to be glycoproteins. The present study aimed to demonstrate glycoconjugates in ACTH and MSH cells in the pituitary of adult sea lampreys (Petromyzon marinus) by means of a lectin histochemistry. A total of 19 kinds of lectins were tested. ACTH cells were distributed in both the rostral pars distalis and the proximal pars distalis, and were stained positively with N-acetylglucosamine binding lectins (i.e., succinylated wheat germ agglutinin), N-acetylgalactosamine binding lectins (i.e., soybean agglutinin), D-mannose binding lectins (i.e., Lens culinaris agglutinin), and D-galactose binding lectins (i.e., Erythrina cristagall lectin). MSH cells were distributed in the pars intermedia, and were stained with N-acetylgalactosamine binding lectins (i.e., Dolichos biflorus agglutinin), D-mannose binding lectin (Pisum sativum agglutinin) and D-galactose binding lectins (i.e., peanut agglutinin). These results suggested that ACTH and MSH cells produce different types of glycoconjugates which may be attributed to the difference in glycoconjugate moieties between the precursor proteins, POC and POM.

Adenohypophysial cell types in the pituitary of adult sea lampreys, Petromyzon marinus, was localized by means of immunocytochemical and lectin cytochemical techniques. At least four types of adenohypophysial hormone cells are present in the pituitary of adult sea lampreys. The first type of cell is ACTH-like and occupies most parts of the rostral pars distalis (RPD), but a few scattered ACTH-like cells are also present in the proximal pars distalis (PPD). The second type of cell is MSH-like and occupies the whole pars intermedia. The third type of cell is GH/PRL-like and occupies the dorsal half of the PPD. These GH/PRL-like cells were initially detected by heterologous immunocytochemistry using antibodies to salmon GH, salmon PRL and blue shark GH, after hydrated autoclave pretreatment of sections. Later, by use of an antiserum raised against a synthetic peptide corresponding to the partial sequence of lamprey GH/PRL, the same cells as those containing GH/PRL-like immunoreactivity were stained positively. Similarity of the topographic distributions between lamprey GH/PRL-like cells and gnathostome fish GH cells in the pituitary suggests that GH/PRL-like cells in the lamprey may be GH cells. The last type of cell is GTH-like and occupies the ventral half of the PPD. Although GTH has not yet been isolated from the lamprey pituitary, our immunocytochemical data suggest that GTH-like material in the sea lamprey pituitary is more closely related to mammalian-like LH, rather than to FSH or TSH. These four types of adenohypophysial cells occupy most parts of the lamprey adenohypophysis and indeed there is little room for TSH or PRL cells. Thus, the present study further suggests that GH and LH-like GTH are ancestral forms of GH/PRL/SL family and glycoprotein hormones, respectively.

In the sea lamprey, Petromyzon marinus, two forms of GnRH, lamprey GnRH-I and -III, have been demonstrated to be neurohormones regulating the pituitary-gonadal axis. The objective of the present study was to determine the distribution of lamprey GnRH-III in the brains of adult sea lampreys and to compare it to the distribution of lamprey GnRH-I. For this purpose, two kinds of immunostaining were employed: one was a single immunostaining by one of two GnRH antibodies using two successive sections; the other was double immunostaining of a single section. A dense accumulation of neuronal cells immunoreactive (ir) to antisera against either lamprey GnRH-I or -III was found in the arc-shaped preoptico-anterior hypothalamic area. Additional smaller numbers of irGnRH cells were found in the periventricular zone of the posterior hypothalamus. In the above-mentioned locations, the distribution of both irGnRH-I and -III cells was intermixed and very similar, but the cells exhibiting GnRH-III immunoreactivity were distinctly different from those exhibiting GnRH-I immunoreactivity. The relative numbers of irGnRH-III cells were larger than those of irGnRH-I cells in the preoptico-anterior hypothalamic area, and more than 90% of GnRH cells in the posterior hypothalamus were irGnRH-III cells. Both irGnRH-I and -III cells projected their fibers primarily into the neurohypophysis. The relative densities of the accumulated irGnRH-III fibers were similar to those of irGnRH-I fibers in the anterior neurohypophysis but higher than those of irGnRH-I fibers in the posterior neurohypophysis. The present study provides further immunocytochemical data to the already compelling physiological evidence that indicates that both lamprey GnRH-I and -III act through the hypothalamic-pituitary-gonadal axis to modulate reproductive processes in the sea lamprey., (Copyright 2000 Academic Press.)

In lampreys, although gonadotropin (GTH) has not yet been isolated from the pituitary gland, the presence of GTH has been strongly suggested. To detect possible GTH in the sea lamprey (Petromyzon marinus) pituitary, two different cytochemical probes were tested: One was the use of antibodies to GTHs, and the other was the use of lectin-screening kits for demonstration of glycoconjugate in hormonal molecules. GTH-like immunoreactivity was found in cells distributed in the ventral half of the proximal pars distalis. These cells were stained intensely by all four lots of anti-ovine LH including LHbeta, and were stained moderately or weakly by several other antibodies to LH-related GTHs, such as human LHbeta, hCGbeta, amphibian LH, and sturgeon GTH IIbeta. On the other hand, there were no positive reactions in the sea lamprey pituitary using the antibodies to FSH-related GTHs, thyrotropin (TSH), or pituitary glycoprotein hormones of teleost origin. Thus, GTH-like material in the sea lamprey pituitary seems to be more closely related to mammalian-like LH, rather than to FSH or TSH, as far as immunocytochemical determinants. A total of 21 kinds of lectins was tested. Among those, GTH-positive cells were also stained positively by concanavalin A and Vicia villosa agglutinin. Thus, the present study demonstrates colocalization of LH-like immunoreactivity and glycoconjugate in cells in the ventral half of the proximal pars distalis of the sea lamprey pituitary. Cytotoxicities of twenty-seven fungal naphtho-gamma-pyrone derivatives to KB cells were examined. Chaetochromins A-D (1-4) and ustilaginoidins D (10) and E (11) exhibited strong effects and the structure-activity relationships are discussed. The antitumor effects in vivo of some of these compounds were examined but, due to their toxicity and to their marginal activity, development as antitumor agents was abandoned. Deoxyribonucleic acid, ribonucleic acid and protein synthesis in KB cells were equally inhibited by chaetochromin A (1), cephalochromin (5), and ustilaginoidin A (7) in parallel with the cytotoxicity of the drugs. Thus it was suggested that other still unknown mechanism(s) might induce cytotoxic lesion, resulting in the inhibition of the macromolecule syntheses.
