Abstract In acute skin injury, healing is impaired by the excessive release of reactive oxygen species (ROS). Melanin, an efficient scavenger of radical species in the skin, performs a key role in ROS scavenging in response to UV radiation and is upregulated in response to toxic insult. In a chemical injury model in mice, we demonstrate that the topical application of synthetic melanin particles (SMPs) significantly decreases edema, reduces eschar detachment time, and increases the rate of wound area reduction compared to vehicle controls. Furthermore, these results were replicated in a UV-injury model. Immune array analysis shows downregulated gene expression in apoptotic and inflammatory signaling pathways consistent with histological reduction in apoptosis. Mechanistically, synthetic melanin intervention increases superoxide dismutase (SOD) activity, decreases Mmp9 expression, and suppresses ERK1/2 phosphorylation. Furthermore, we observed that the application of SMPs caused increased populations of anti-inflammatory immune cells to accumulate in the skin, mirroring their decrease from splenic populations. To enhance antioxidant capacity, an engineered biomimetic High Surface Area SMP was deployed, exhibiting increased wound healing efficiency. Finally, in human skin explants, SMP intervention significantly decreased the damage caused by chemical injury. Therefore, SMPs are promising and effective candidates as topical therapies for accelerated wound healing, including via pathways validated in human skin.

Hydrogen is an essential gas to multiple industrial processes, and due to its high gravimetric density, it promises a large potential as a clean energy source. The risks of hydrogen at low pressures, however, have deterred substantial progress in improving hydrogen storage technology. Metal-organic frameworks (MOFs) are crystalline porous materials that have emerged as excellent gas adsorbents, and their gas storage properties can be tuned using a variety of synthetic methods. Herein, we leveraged the tunability and porosity of MOFs to introduce defect engineering as a method to improve hydrogen storage technology at low pressure regimes in two zirconium-based MOFs (Zr-MOFs), UiO-66 and NU-403, which feature ideal pore apertures (7 Å) for studying the confinement effect and ease of defect engineering. By reducing the number of defects in these Zr-MOFs, and thereby decreasing the quantity of larger pores, we can induce a structural confinement effect that increases the selectivity of hydrogen adsorption. A combination of thermogravimetric analysis (TGA) and nuclear magnetic resonance (NMR) spectroscopic analysis enabled quantification of the defect levels, confirming that each sample exhibits a distinct level of defectiveness. Gas adsorption measurements revealed that the adsorption of hydrogen is greatly enhanced in samples with fewer defects, while the calculated isosteric heats of enthalpy (Qst) indicate that there are no open metal sites in these MOFs that could be the cause. Overall, we can conclude that defect engineering for pore tailoring is a viable strategy to enhance hydrogen adsorption at low-pressure regimes.

Here, the authors describe a colloidal crystal engineering strategy to prepare hierarchical structures from metal–organic framework nanoparticles and DNA which retain permanent porosity and catalytic activity.

We demonstrate a microporous metal–organic framework NU-400 based on a 2,7-disubstituted pyrene linker as a highly efficient photosensitizer for generating singlet oxygen and subsequent oxidative degradation of chemical warfare agents (CWAs). The high activity of NU-400 permits photocatalytic conversion of the 2-chloroethyl ethyl sulfide (CEES) mustard gas simulant into a benign sulfoxide derivative, in air, with less than 15 minutes’ half-life. This is a considerable improvement to NU-1000, based on a 1,3,6,8-tetrasubstituted pyrene unit, demonstrating how variation of the substitution pattern of a metal–organic framework linker permits modification of its photoactive behavior.

Summary: Enzymes are promising catalysts with high selectivity and activity under mild reaction conditions. However, their practical application has largely been hindered by their high cost and poor stability. Metal-organic frameworks (MOFs) as host materials show potential in protecting proteins against denaturing conditions, but a systematic study investigating the stabilizing mechanism is still lacking. In this study, we stabilized enzyme cytochrome c (cyt c) by encapsulating it in a hierarchical mesoporous zirconium-based MOF, NU-1000 against denaturing organic solvents. Cyt c@NU-1000 showed a significantly enhanced activity compared to the native enzyme, and the composite retained this enhanced activity after treatment with five denaturing organic solvents. Moreover, the composite was recyclable without activity loss for at least three cycles. Our cyt c@NU-1000 model system demonstrates that enzyme@MOF composites prepared via post-synthetic encapsulation offer a promising route to overcome the challenges of enzyme stability and recyclability that impede the widespread adoption of biocatalysis.

Summary: Creating crystalline porous materials with large pores is typically challenging due to undesired interpenetration, staggered stacking, or weakened framework stability. Here, we report a pore size expansion strategy by “shape-matching” intermolecular π-π stacking interactions in a series of two-dimensional (2D) hydrogen-bonded organic frameworks (HOFs), HOF-10x (x = 0,1,2), self-assembled from pyrene-based tectons with systematic elongation of π-conjugated molecular arms. This strategy successfully avoids interpenetration or staggered stacking and expands the pore size of HOF materials to access mesoporous HOF-102, which features a surface area of ∼2,500 m2/g and the largest pore volume (1.3 cm3/g) to date among all reported HOFs. More importantly, HOF-102 shows significantly enhanced thermal and chemical stability as evidenced by powder X-ray diffraction and N2 isotherms after treatments in challenging conditions. Such stability enables the easy fabrication of a HOF-102/fiber composite for the efficient photochemical detoxification of a mustard gas simulant.

Summary: Recent efforts have examined metal-organic frameworks (MOFs) as sorbents for removing pollutants and trace chemicals, such as pharmaceuticals, from aqueous media. To date, few studies have investigated the energetics of interactions between toxic species and MOF-based sorbents. To this end, we elected to use isothermal titration calorimetry (ITC) to quantify the thermodynamic parameters associated with the adsorption of uremic toxins, namely p-cresyl sulfate and indoxyl sulfate, onto zirconium-based MOFs. NU-1000, a mesoporous MOF comprising a pyrene-based linker, possesses two distinct adsorption sites that display dramatically different enthalpic and entropic parameters. ITC measurements further reveal that a concerted interaction that embodies both favorable enthalpic and entropic contributions is critical for designing a high-capacity MOF-based adsorbent.

Direct removal of 99TcO4 − from highly radioactive and acidic nuclear waste solutions is beneficial for uranium and plutonium recovery and radioactive pollution control but this represents a huge challenge. Here the authors show a cationic polymeric network with high 99TcO4 − sorption capability and stability.

The emergence of thousands of metal–organic frameworks (MOFs) has created the challenge of finding promising structures for particular applications. Here, the authors present a tool for computer-aided material discovery where a large number of MOFs are screened, with the top-ranked structure synthesized for oxygen storage applications.

Comprised of high valent metal centers connected via oxide bonds, polyoxometalates (POMs) have a rich history in redox reactions. These discrete metal oxide clusters often are immobilized on heterogeneous supports to increase stability and allow for recyclability for catalytic reactions. One such support is the metal-organic framework (MOF), NU-1000. When POMs are installed into NU-1000, they are first sited in the mesoporous channel. Upon application of heat and removal of some physisorbed water, the POMs migrate to the microporous channel, where some active sites are blocked by the MOF linkers, resulting in lowered catalytic activity. To restrict this movement from the mesopore, we report here the use of two MOFs, naphthalene dicarboxylate-modified NU-1000 (NU-1000-NDC) and NU-1008, as supports for the Keggin-type polyoxometalate (POM), H3PW12O40. Upon the application of heat, these frameworks were found to hinder or prevent the movement of the POM between channels by blocking the aperture(s) connecting the mesoporous and microporous channels. The composite POM@MOF materials were used for the oxidation of a mustard gas simulant, 2-cholorethyl ethyl sulfide, using H2O2 as the oxidant. The POM@MOF catalysts exhibit enhanced reactivity when compared to the POM or MOF alone, more than doubling the initial rates. The activity trend in PW12@NU-1000-NDC matched that in PW12@NU-1000, where the scCO2-activated sample poised the POM in the mesopores to give greater substrate accessibility and enhance the reaction rate compared to the heated sample where the POMs are poised in the micropores. Contrary to these observed trends, PW12@NU-1008 prohibits POM migration and performs superior when water has been removed at elevated temperatures as the active sites are more accessible in the mesoporous channel.

Aqueous synthesis of metal−organic frameworks (MOFs) at room temperature offers many advantages such as reduction in the generation of toxic byproducts and operation costs, as well as increased safety in the material’s production. Functional group-bearing MOFs have received growing attention compared to nonfunctionalized analogues due to enhanced adsorption properties of the former in many cases. Here, we report an aqueous solution-based synthesis of a robust zirconium MOF, UiO-66-NO2, at room temperature. We evaluated the phase purity, porosity, thermal stability, particle morphology and size of the resulting material. High uptake, as well as near complete recyclability of water and ethanol vapor isotherms at room temperature supports the potential of UiO-66-NO2 as a solid adsorbent in adsorption-based cooling applications or water harvesting systems.

As a C1 feedstock, CO2 has the potential to be uniquely highly economical in both a chemical and a financial sense. In particular, the highly atom-economical acid-catalyzed cycloaddition of CO2 to epoxides to yield cyclic organic carbonates (OCs), a functionality having many important industrial applications, is an attractive reaction for the utilization of CO2 as a chemical feedstock. Metal–organic frameworks (MOFs) are promising candidates in catalysis as they are a class of crystalline, porous and functional materials with remarkable properties including great surface area, high stability, open channels and permanent porosity. MOFs structure tunability and their affinity for CO2, makes them great catalysts for the formation of OCs using CO2 and epoxides. In this review, we examine MOF-based catalytic materials for the cycloaddition of carbon dioxide to epoxides. Catalysts are grouped based on the location of catalytic sites, i.e., at the struts, nodes, defect sites, or some combination thereof. Additionally, important features of each catalyst system are critically discussed.

Reaction of styrene oxide with sodium cyanoborohydride and a catalytic amount of Hf-NU-1000 yields the anti-Markovnikov product, 2-phenylethanol, with over 98% regioselectivity. On the other hand, propylene oxide is ring opened in a Markovnikov fashion to form 2-propanol with 95% regioselectivity. Both styrene oxide and propylene oxide failed to react with sodium cyanoborohydride without the addition of Hf-NU-1000 indicative of the crucial role of Hf-NU-1000 as a catalyst in this reaction. To the best of our knowledge, this is the first report of the use of a metal-organic framework material as a catalyst for ring-opening of epoxides with hydrides.

Perovskite-containing solar cells were fabricated in a two-step procedure in which PbI2 is deposited via spin-coating and subsequently converted to the CH3NH3PbI3 perovskite by dipping in a solution of CH3NH3I. By varying the dipping time from 5 s to 2 h, we observe that the device performance shows an unexpectedly remarkable trend. At dipping times below 15 min the current density and voltage of the device are enhanced from 10.1 mA/cm2 and 933 mV (5 s) to 15.1 mA/cm2 and 1036 mV (15 min). However, upon further conversion, the current density decreases to 9.7 mA/cm2 and 846 mV after 2 h. Based on X-ray diffraction data, we determined that remnant PbI2 is always present in these devices. Work function and dark current measurements showed that the remnant PbI2 has a beneficial effect and acts as a blocking layer between the TiO2 semiconductor and the perovskite itself reducing the probability of back electron transfer (charge recombination). Furthermore, we find that increased dipping time leads to an increase in the size of perovskite crystals at the perovskite-hole-transporting material interface. Overall, approximately 15 min dipping time (∼2% unconverted PbI2) is necessary for achieving optimal device efficiency.

A two-component undergraduate laboratory experience has been developed by students in a senior level capstone course. The first component is a 3 h laboratory experience dedicated to the rapid synthesis of a metal-organic framework (MOF-808) in aqueous solution using readily available reagents and equipment. During the second component, MOF-808 was characterized via a suite of instruments: powder X-ray diffraction (PXRD), thermal gravimetric analysis (TGA), and diffuse reflectance infrared Fourier transform spectroscopy (DRIFTS). In addition, quantitative [superscript 19]F{[superscript 1]H} NMR spectroscopy was utilized to quantify the amount of perfluorobutanesulfonate (PFBS), one example of a poly- or perfluoroalkyl substance (aka PFAS), adsorbed from solution. The two 3 h laboratory experiences were subsequently deployed in a foundation level inorganic chemistry course. This two-component, multi-instrument lab experience provides students an opportunity to synthesize a modern porous solid and utilize it in an emerging application of MOF science.
