2. Effects of Glucomannan Feed Additives on Large White Turkey Female Performance, Gastrointestinal Microbial Population, and Blood Corticosterone Levels in a Simulated Transport Challenge

Turkeys may experience stress due to conditions of transport, heat, and in some cases, feed and water restrictions. This may include experiencing conditions present when female turkeys are transported from the brooder house to the grow-out house, typically at 4 to 5 wk of age. This study’s objective was to determine the effect of glucomannan feed additives on performance and corticosterone response in Large White commercial females after an application of simulated conditions which birds might experience during transportation at 4 to 5 wk of age. The transport simulation consisted of reducing the space per bird from 0.198 m2 /bird to 0.028m2 /bird, with no food and water, and exposed to heat lamps, for no more than 18 h. Fourteen hundred and forty Large White female turkeys were randomly assigned to 48 concrete, pine shavings covered floor pens. The experimental design was a completely randomized block design with a one-factor arrangement of 2 sources of glucomannans (medium-chain and long-chain) with two inclusion levels of each (0.02 and 0.20% of the diet). These treatments were compared to two control treatments (no glucomannans), however, one with no simulated transport conditions and one exposed to simulated transport conditions. Bird’s performance, bursa, spleen, and blood corticosterone levels were analyzed in SAS 9.4 in a mixed model. No effects on growth performance of birds fed 0.02% glucomannans were observed. Birds fed glucomannans at 0.20% in the diet had a higher corticosterone level after the transport simulation was applied when compared to other treatments. However, birds fed medium-chain glucomannans at 0.20% had an improved performance after the transport simulation compared to birds fed the other glucomannan treatments. All birds recuperated their performance by d 45. No differences were found in the spleen and bursa weights at 45 d of age. Both glucomannans reduced the stress impact on intestinal bacteria by controlling the overgrowth of Lactobacillus and Fusobacteria while maintaining Bacteroidetes. In conclusion, glucomannans can be used in diets with no change in performance or health at 45 d of age while preserving intestinal microbiota from the negative effects of stress. At 45d, all birds recuperated from the simulated transport conditions applied at 29 d. Thus, the effects of glucomannans on turkey performance and health should be further investigated.

Five experiments were performed, with iron (10 mg); iron (10 mg) plus vitamin A (450, 900 and 1800 µg), and placebo. After an 8-hour fast, basal (T0) blood samples were collected: basal (T0), 2 hours (T1), and 4 hours (T2) after the ingestion of the compounds to be studied. Iron was determined by inductively coupled plasma mass spectrometry. Serum ferritin was determined by an immunometric method, ie, by chemoluminescent enzyme immunoassay. Plasma retinol was measured by high-pressure liquid chromatography. Serum curves and the sum of the area under the curve adjusted to the mixed effects linear model were determined (P < 0.05).Results: Vitamin A at the doses of 450 and 900 µg had a stimulating effect, which, however, did not differ significantly from that of experiment 1 in which iron was used alone. At the dose of 1800 µg, vitamin A had a negative effect on iron absorption.Conclusion: High doses of vitamin A may cause lower serum iron levels, whereas a low dose favors iron absorption.Keywords: iron absorption, serum iron, vitamin A, oral iron, oral supplement

9. Biologia Populacional e padrões de coloração do caranguejo terrestre, Cardisoma guanhumi (Latreille 1828) (Crustacea: Gecarcinidae) no Nordeste do Brasil

The objective of this study was to analyze the population biology and color patterns of Cardisoma guanhumi Latreille, 1828 in a mangrove area in Natal, Rio Grande do Norte, Brazil. Crabs were collected monthly between February 2010 and January 2012 and totaled 1,837 individuals. Sex ratios were similar between males and females in the first year and differed in the second. Sex ratios by size class differed statistically in the extremes of the distribution, with an abundance of males in the large size classes. There was no difference (p > 0.05) in carapace width between males and females in the first year, but in the second year, males were larger than females (p = 0.003), showing the importance of considering interannual variation in such studies. Both sexes presented a unimodal distribution. The size of first maturing of the females was 6.00 cm. Asymptotic carapace width for males was 9.4 cm and females for 9.2 cm, using the Wetherall method. There was no defined modal progression, which made the determination of the Von Bertalanffy growth parameter K impossible. Juveniles were found throughout all months of the year, demonstrating that there was no defined period of recruiting, although slightly fewer juveniles were found in the dry season. Four color patterns were observed, the three first patterns were the same for both sexes, while the fourth pattern was observed in females only. The four color patterns identified could be clearly related to the stage of development (size) and sexual maturity of the animal. O objetivo deste artigo foi analisar a biologia populacional e os padrões de coloração da carapaça de Cardisoma guanhumi Latreille, 1828 em uma área de manguezal em Natal, Rio Grande do Norte, Brasil. Os caranguejos foram coletados mensalmente entre fevereiro de 2010 e janeiro de 2012 e totalizaram 1837 caranguejos. A proporção sexual foi semelhante para machos e fêmeas (1:1) no primeiro ano e diferiu no segundo ano. A proporção sexual por classe de tamanho diferiu estatisticamente nas classes extremas da distribuição, com abundância de machos nas superiores. Não houve diferença (p ≥ 0,05) entre as larguras de carapaça de machos e fêmeas no primeiro ano, porém no segundo os machos foram maiores que as fêmeas (p = 0,003), revelando à importância de se considerar as variações interanuais nestes estudos. Ambos os sexos apresentaram distribuição unimodal. O tamanho de primeira maturação sexual das fêmeas foi 6,00 cm. A largura de carapaça assintótica para machos foi de 9,4 cm e para as fêmeas de 9,2 cm, usando o método Wetherall. Não houve progressão modal definida, o que impossibilitou a determinação do parâmetro de crescimento k de Von Bertalanffy. Foram encontrados juvenis ao longo de todos os meses do ano, mostrando não haver um período de recrutamento definido, embora a abundância de juvenis tenha sido menor na estação seca. Foram observados quatro padrões de coloração, três para ambos os sexos e um quarto padrão observado apenas para as fêmeas. Os quatro padrões de coloração da carapaça estão claramente relacionados ao estágio de desenvolvimento (tamanho) e à maturidade sexual do animal.

10. Prospective evaluation of glutamine and phospholipids levels in first degree relatives of patients with Type 1 Diabetes from a multiethnic population

- Abstract
BACKGROUND: A dysregulation in the metabolism of lipids may be an early marker of autoimmunity in Type 1 Diabetes (T1D). It would be of general importance to identify metabolic patterns that would predict the risk for T1D later in life. The aim of this study was to perform a prospective evaluation of glutamine and phospholipids levels in Brazilian first degree relatives (FDR) of patients with T1D in a mean interval of 5 years. FINDINGS: Brazilian FDR (n = 30) of patients with T1D were evaluated and blood was sampled to measure the levels of glutamine and phospholipids in the fasting serum by quantitative colorimetric method. The tests were repeated after a mean interval of 5 years and compared to a control group (n = 20). The FDR presented lower levels of phospholipids than controls (p = 0.028), but not of glutamine (p = 0.075). Phospholipids levels decreased over time (p = 0.028) in FDR and were associated with Glutamic acid decarboxylase autoantibody (GADA) titers (p = 0.045), autoantibody positivity (p = 0.008) and PTPN22 polymorphisms (p = 0.014). CONCLUSIONS: In this Brazilian multiethnic population, there was a significant decrease in phospholipids levels in FDR in patients with T1D during a 5-year prospective follow-up, as well as a significant association between these metabolite, GADA and PTPN22 polymorphisms. For Glutamine no difference was found. These findings suggest that a dysregulation in the metabolism of lipids may precede the onset of the autoimmunity in T1D.

The main changes in the buccal and maxillofacial region are modifications of the facial bones (leading to the loss of the mandibular angle due to micrognathia), deep palate, premature and/or delayed tooth eruption, hypopneumatization of the maxillary sinuses, chin hypoplasia, hypercementosis, enamel hypoplasia, hyperdontia, incomplete dentin calcification, root hypoplasia, pulp chamber obliteration, and increased free functional space. This paper aims to report a case of a patient with PYCD, in which are highlighted the oral manifestations of this syndrome and present an option of oral rehabilitative treatment through a removable overdenture made on natural tooth of the patient. The technique of dental coating through overdenture is a simple, viable and conservative alternative for oral rehabilitation of patients with large maxillomandibular discrepancies, as in cases of pycnodysostosis, especially in the presence of increased free space – characteristic of patients with this syndrome. The result was quite satisfactory both aesthetically and functionally and provided a better life quality for the patient.Keywords: malocclusion, pycnodysostosis, oral rehabilitation

Vitamin A deficiency is one of the major nutritional problems in the world, most common in developing countries. Food fortification is a recognised approach to supply vitamins and minerals to needed populations. Vegetable cooking oils were previously suggested by us as a carrier for vitamin A fortification. Fortification of cooking oil with beta-carotene could also be a strategy to prevent vitamin A deficiency. The objective of this article is to start studies on the use of cooking soya oil as a vehicle for synthetic carotene, to evaluate its stability to heat treatment, and to test its bioavailability and bioconversion to vitamin A in rats. Batches of carotene-fortified soybean oil were prepared, containing 2, 4 and 8 RE/g of diet. Some of them were heated to test its stability. At 100 degrees C there was no loss of carotene, at higher temperature carotene retention was 65%. The bioavailability and bioconversion of beta-carotene added to soybean oil was measured through feeding nursing rats and their pups method. Weight gain was good and plasma vitamin A increased significantly in all groups. Liver vitamin A values of rats fed diets with fortified soybean oil heated at 100 degrees C was similar to the 4 RE non-heated fortified oil group (0.72 +/- 0.06 and 0.64 +/- 0.08 mumol/g, respectively). Heated at 170 degrees C the liver total vitamin A value was reduced (0.45 +/- 0.04 mumol/g), but kept bioavailable vitamin A equivalent to 2 RE (0.47 +/- 0.09 mumol/g). Bioconversion of beta-carotene to vitamin A was validated by the plasma and liver findings. beta-carotene added to soybean oil showed good stability to heat and its bioconversion to vitamin A was shown in rat assays. beta-carotene mixed well with edible soybean oil and the fortified cooking oil showed potential as a carrier to be used for the prevention of vitamin A deficiency.

Odontoameloblastoma (OA) is a very rare mixed odontogenic neoplasm, characterized by the simultaneous occurrence of an ameloblastoma and a compound or complex odontoma in the same tumor mass. To date, less than 50 cases of OA and/or ameloblastic odontoma have been reported in the English dental literature. This neoplasm was called ameloblastic odontoma. The term OA was included in the 1971 WHO classification. In this study, we present two cases of OA, which we hope will contribute to the awareness and knowledge of surgeons regarding the existence of this odontogenic tumor so that patients having it may be treated and followed-up properly.

Como certos fungos e leveduras podem residir na pele, a positividade em culturas de escamas ungueais deve ser interpretada obrigatoriamente em concordância com os respectivos dados clínicos, exames diretos das amostras, quantificação das colônias isoladas em relação aos pontos de inoculação e, principalmente, deve a positividade ser mantida após a repetição dos cultivos. A invasão da unha por fungos não dermatofíticos (nondermatophytic molds, NDM) em diferentes estudos varia de 1,45% a 17,6%. OBJETIVOS: Os objetivos do trabalho são mostrar a ocorrência e realçar a importância dos fungos emergentes como causadores de onicomicose. MÉTODOS: As unhas dos pacientes atendidos nos consultórios dos dermatologistas foram examinadas, e, caso houvesse suspeita clínica de onicomicose, coletavam-se amostras que eram enviadas para exame micológico no laboratório. RESULTADOS: Neste estudo, foram avaliados 2.271 pacientes e diagnosticada onicomicose em 400 deles, sendo 264 com acometimento das unhas do pé, e 136 das unhas da mão. O agente etiológico foi confirmado pelo exame micológico direto e crescimento em cultura. A onicomicose por fungos emergentes representou 4,5% de todas as infecções de unha. Foram detectadas as leveduras do gênero Candida (49%) como agentes etiológicos mais freqüentes de onicomicoses nas unhas das mãos e em mulheres. Em contraste, os dermatófitos foram os mais freqüentes de todas as onicomicoses dos pés, no total de 186 (46,5%). CONCLUSÃO: Ao contrário da rotina diagnóstica hoje utilizada, concluiu-se que o diagnóstico correto do agente etiológico da onicomicose não dermatofítica deve obedecer a determinados critérios, dos quais se destaca o conjunto formado pela positividade no exame direto, na cultura, com comprovação na repetição do exame.

Introdução: O conceito de prótese intraluminal foi introduzido por CARREL, em 1912. Em 1978, DUREAU & ABLASA descreveram os primeiros casos de dissecção aguda de aorta tratados com o emprego da prótese intraluminal. Esta prótese foi utilizada nos anos 80 por vários grupos com bons resultados. Vários relatos de complicações (migração, embolias, estenoses, formação de pseudo-aneurismas) fizeram com que os grupos cirúrgicos perdessem o entusiasmo por esta técnica. Objetivo: Avaliar a anastomose empregando a prótese intraluminal sem sutura no tratamento da dissecção aguda de aorta tipo A. Casuística e Métodos: Sessenta e quatro pacientes com diagnóstico de dissecção aguda de aorta do tipo A foram operados em nosso Serviço. Em todos os casos empregamos o anel intraluminal como técnica de sutura. A idade média dos pacientes era de 56,3 anos, sendo 57 (89%) do sexo masculino. Todos os pacientes eram portadores de hipertensão arterial sistêmica. Em 26 pacientes utilizamos somente a anastomose intraluminal, com o anel proximal acima dos óstios coronários de modo a ressuspender a valva aórtica e o anel distal posicionado junto à origem do tronco braquiocefálico. O tempo médio de pinçamento da aorta nestes pacientes foi de 9 minutos e o tempo médio de CEC de 26 minutos. Somente em 1 caso houve rotura da ligadura sobre o anel proximal, quando realizamos uma operação radical (Bentall- DeBonno), mantendo o anel distal. Em 23 pacientes utilizamos a ressuspensão da valva aórtica e o preparo da raiz da aorta com duplo retalho de Dacron, empregando o anel intraluminal na anastomose distal. Abordamos o arco aórtico em 8 pacientes. A troca da valva aórtica foi necessária em apenas 6 pacientes. A mortalidade global foi de 10,3%, sendo que nos pacientes onde foi possível o tratamento com o duplo anel não houve nenhum óbito. Na avaliação pós-operatória destes pacientes foi realizado ecocardiograma e aortografia. Não observamos formação de pseudo-aneurisma, migração e o gradiente máximo encontrado foi de 16 mmHg. Conclusão: O emprego da prótese intraluminal para o tratamento da dissecção aguda de aorta do tipo A nos proporciona uma anastomose rápida, segura e hem2ostática. Na avaliação pós-operatória e a médio prazo não observamos qualquer complicação relacionada ao emprego desta técnica.

O objetivo deste trabalho foi avaliar o efeito do uso de isolante térmico de telhado instalado como forro sob a cobertura no desempenho de frangos alojados em diferentes densidades (10, 16 e 22 aves/m²) e nas características da cama de aviário em galpão convencional. O experimento foi realizado durante o verão, utilizando 3328 aves da linhagem Hubbard, distribuídas em um delineamento inteiramente casualizado, em um esquema fatorial 3x2 (densidade e sexo), com quatro repetições dentro de cada ambiente. Para a comparação dos ambientes, utilizou-se a análise conjunta. Durante o período experimental, registraram-se dados de temperatura ambiente em vários pontos do galpão e em diversos horários, umidade relativa e carga térmica radiante. O teor de nitrogênio e matéria seca e a temperatura da cama foram determinados. No ambiente com isolante térmico, as temperaturas ambientais médias e a amplitude térmica diária foram menores e a umidade relativa do ar, maior. O desempenho das aves foi melhor no ambiente com isolante térmico, no qual as aves tiveram maior consumo de ração, maior ganho de peso, melhor conversão alimentar, menor mortalidade e maior produção por área. A temperatura retal das aves do ambiente sem isolante térmico foi mais elevada. À medida que se elevou a densidade, a mortalidade aumentou, porém a produção por área foi maior. O uso do isolante não influenciou a produção de cama, mas diminuiu a porcentagem de nitrogênio na cama. Com o aumento da densidade, a produção de cama elevou-se, pelo maior volume de excretas, mas não influenciou sua porcentagem de nitrogênio. O uso do isolante térmico permitiu aumento da densidade de criação dos frangos.

O tratamento cirúrgico das doenças da aorta é geralmente acompanhado de altas taxas de morbimortalidade. O paciente que tem média de idade avançada geralmente é apresentado ao cirurgião em estado grave, com má condição nutricional, muitas vezes com alterações em vários órgãos e sistemas causados pela própria doença aguda. A correção convencional exige técnicas coadjuvantes complexas e agressivas como a circulação extracorpórea prolongada, hipotermia profunda, parada circulatória total além de prolongados tempos de pinçamento de aorta. Na tentativa de reduzir a agressão cirúrgica no paciente já intensamente debilitado pela própria doença, desenvolvemos, em 1988, um anel intraluminal (1, 2) que tem medidas projetadas e experimentadas para facilitar a sua manipulação e anastomose, proporcionando uma diminuição acentuada no tempo de operação, tempo de CEC, tempo de pinçamento de aorta, excluindo muitas vezes a circulação extracorpórea e a hipotermia, obtendo uma anastomose fácil, rápida, segura e hemostática. Entre março de 1988 e janeiro de 1999, 432 pacientes foram submetidos a tratamento cirúrgico de dissecções ou aneurismas da aorta em nosso Serviço. Em 328 pacientes empregamos o anel intraluminal como técnica de anastomose. Usamos 489 anéis. Cento e vinte e cinco pacientes eram portadores de dissecção aguda de aorta do tipo A, 29 eram portadores de dissecção aguda de aorta do tipo B, 81 de aneurisma de aorta ascendente, 8 de aneurisma de arco aórtico, 28 pacientes eram portadores de aneurisma de aorta torácica descendente, 17 eram portadores de aneurisma toracoabdominal e 40 pacientes de aneurisma de aorta abdominal infra-renal. A mortalidade global foi de 13,41%. O seguimento ambulatorial destes pacientes variou de 11 anos a 25 dias. A curva actuarial de sobrevivência em 11 anos mostra 67,3%. Em nenhum caso observamos as complicações descritas na literatura, como embolia, formação de pseudo- aneurisma, roturas ou estenose. Concluindo, em nosso Serviço, o emprego de anel intraluminal facilita o ato cirúrgico, reduz o tempo de anastomose e o sangramento, proporcionando facilidade técnica, redução da mortalidade e boa perspectiva de sobrevida a longo prazo.

33. Application of 1-octanol in the extraction and GC-FID analysis of volatile organic compounds produced in biogas and biohydrogen processes.

Information about the volatile organic compounds generated in biogas and hydrogen production bioreactors is essential to elucidate the metabolic routes and varying yields of CH
4 and H2 processes. In this work, the determination of 12 compounds (acetone, methanol, ethanol, 1-propanol, 1-butanol, and acetic, propanoic, butyric, isovaleric, valeric, caproic, and lactic acids) was performed by gas chromatography, after a vortex-assisted liquid-liquid microextraction (VALLME) procedure using 1.2 mL of crude sample and 400 µL of 1-octanol. Optimization of the separation process was performed, considering the solvent viscosity. The analytical curves were validated using ANOVA, demonstrating satisfactory precision and accuracy. Selectivity was confirmed by GC-MS analysis, which allowed the detection of glycerol, 1,3-propanediol, and 1,3-butanediol in some samples. Methanol levels exceeded the upper limit of quantification, with acetic acid and ethanol being the predominant compounds in the analyzed reactors. An additional investigation was conducted to assess potential interferences for lactic acid. - Abstract
GH acts in numerous organs expressing the GH receptor (GHR), including the brain. However, the mechanisms behind the brain's permeability to GH and how this hormone accesses different brain regions remain unclear. It is well-known that an acute GH administration induces phosphorylation of the signal transducer and activator of transcription 5 (pSTAT5) in the mouse brain. Thus, the pattern of pSTAT5 immunoreactive cells was analyzed at different time points after IP or intracerebroventricular GH injections. After a systemic GH injection, the first cells expressing pSTAT5 were those near circumventricular organs, such as arcuate nucleus neurons adjacent to the median eminence. Both systemic and central GH injections induced a medial-to-lateral pattern of pSTAT5 immunoreactivity over time because GH-responsive cells were initially observed in periventricular areas and were progressively detected in lateral brain structures. Very few choroid plexus cells exhibited GH-induced pSTAT5. Additionally, Ghr mRNA was poorly expressed in the mouse choroid plexus. In contrast, some tanycytes lining the floor of the third ventricle expressed Ghr mRNA and exhibited GH-induced pSTAT5. The transport of radiolabeled GH into the hypothalamus did not differ between wild-type and dwarf Ghr knockout mice, indicating that GH transport into the mouse brain is GHR independent. Also, single-photon emission computed tomography confirmed that radiolabeled GH rapidly reaches the ventral part of the tuberal hypothalamus. Commercially available insecticides present acute toxicity to the health of fish and other aquatic organisms, which may impair the local aquaculture. This study evaluated the gonadal morphology of freshwater fish exposed to pyriproxyfen and fenthion. Forty-five juvenile male Nile tilapias (Oreochromis niloticus) were divided into control, pyriproxyfen-exposed (0.01 g/L), and fenthion-exposed (0.001 g/L) groups. They were evaluated in three moments (30, 60, and 90 days). The variables analyzed were the gonadosomatic index (GSI), weight to length ratio, seminiferous tubules morphometry (diameter and height), tissue damage, and immunohistochemical analysis for caspase-3, tumor necrosis factor alpha (TNF-α), and vascular endothelial growth factor (VEGF). Pyriproxyfen and fenthion injured the seminiferous tubule tissue, and the damage progressed according to the exposure time. In addition, the GSI gradually reduced over time in all groups compared with the first moment (30 days), while caspase-3, TNF-α, and VEGF values increased only in the fenthion-exposed group. Therefore, pyriproxyfen and fenthion changed the gonadal morphology of male Oreochromis niloticus, which may affect their reproduction in the wild or captivity., (© 2024 Fisheries Society of the British Isles.)

Food contamination by mycotoxigenic fungi is one of the principal factors that cause food loss and economic losses in the food industry. The objective of this work was to incorporate the essential oil from Corymbia citriodora Hook and its constituents citronellal and β-citronellol into poly(lactic acid) nanofibers; to characterize the nanofibers by scanning electron microscopy (SEM), Fourier transform infrared spectroscopy and differential scanning calorimetry; to evaluate the antifungal activity by the fumigation method; to evaluate the antimycotoxigenic activity against Aspergillus carbonarius, Aspergillus ochraceus, Aspergillus westerdijkiae, Aspergillus flavus, and Aspergillus parasiticus; and to evaluate the morphology of these microorganisms. All the nanofibers had a regular, smooth, and continuous morphology. FTIR analyses confirmed that the active ingredients were incorporated into the polymer matrix. All samples exhibited antifungal and ochratoxigenic inhibitory activities of up to 100% and 99%, respectively, with the best results observed for (PLA + 30 wt% β-citronellol) nanofibers and (PLA + 30 wt% citronellal) nanofibers. However, 100% inhibition of the production of aflatoxin B1 and B2 was not observed. The images obtained by SEM indicated that the nanofibers caused damage to the hyphae, caused a decrease in the production of spores, and caused deformation, rupture, and non-formation of the conid head, might be an alternative for the control of mycotoxigenic fungi., (© The Author(s) 2024. Fresh produce have a more limited shelf life than processed ones. Their sensory attributes such as appearance and surface texture are important features in consumer perception and liking. The decomposition of fresh produce, which is caused by enzymes, chemical reactions, and microbial infections, often caused by Colletotrichum species, is inevitable. However, it can be slowed down. Several materials have been developed for this purpose, with an emphasis on active coatings using nanomaterials. In this study, the protective effects of a zein coating containing chitosan nanowhiskers (CSW) for the maintenance of fruit quality were investigated using guava (Psidium guajava L.) as a model fruit. CSW were previously characterized, and their antifungal effects against distinct Colletotrichum species (Colletotrichum asianum, Colletotrichum tropicale, Colletotrichum gloeosporioides, and Colletotrichum brevisporum) were proven. Coatings were characterized by thermogravimetric analysis, optical profilometry, and mechanical properties. Total soluble solids, pH, mass loss, and visual inspection of uncoated and coated guava fruits were also verified during 9 days. Results show that CSW length and aspect ratio decreased for longer extraction times. A similar behavior was found for x-ray diffraction in which peak intensity decreases under the same conditions. CSW degradation (ca. 250-400°C) also depends on extraction time in which more crystalline whiskers are the most thermally stable ones. The addition of CSW did not significantly (p < 0.05) modify the homogeneity and continuity of coating but prevented microbial growth assuring fruit quality during storage. In summary, coatings protected guava fruits from post-harvest spoilage while preserving quality and extending shelf life. Objective: to verify sepsis incidence among young adults admitted to intensive care due to COVID-19 and to analyze its association with demographic, clinical and outcome variables., Methods: a quantitative, longitudinal, retrospective and analytical study, consisting of 58 adults aged 20 to 40 years in intensive care for SARS-CoV-2. It was carried out in a university hospital, from March 2020 to December 2021, with data collected from electronic medical records., Results: sepsis incidence was 65%. Sepsis was associated with acute kidney injury, use of vasoactive drugs and mechanical ventilation, being admitted to the emergency room, severity according to the Simplified Acute Physiology Score III and bacterial pulmonary co-infection, the latter being the most frequent etiology for sepsis., Conclusions: there was a high sepsis incidence, with 42% of deaths, which points to the importance of investing in preventive measures, especially in relation to bacterial pulmonary coinfections.

Important changes in glucose transporter (GLUT) expression should be expected if the glucose influx plays a pivotal role in fuelling or connecting metabolic pathways that are upregulated in response to exercise. The aim was to assess GLUT4, 8, and 12 dynamics in response to training and acute exercise. Methods: Sixteen untrained Standardbred mares (3-4 year) performed an incremental SET at the start and end of 8 weeks harness training. M. pectoralis (PM) and M. vastus lateralis (VL) muscle biopsies were taken before and after each SET, allowing for comparing rest and acute samples in untrained (UT) and trained (T) condition using Western Blot for GLUT quantification and Image Pro v.10 for Blot analysis. Data were normalized against GAPDH. Basal GLUT-levels of PM versus VL were analysed with the Wilcoxon matched-pairs signed rank test. The effect of acute exercise or training was assessed using the Friedman test with a post hoc Dunn's. Results: Basal GLUT4 and GLUT12 protein expression were significantly higher in the VL compared to the PM (P
GLUT4 = 0.031 and PGLUT12 = 0.002). Training had no effect on basal GLUT4 expression, neither in the VL ( p > 0.9999), nor the PM ( p > 0.9999). However, acute exercise in trained condition significantly decreased GLUT4 expression in the VL ( p = 0.0148). Neither training nor acute exercise significantly changed total GLUT8 protein expression. Training significantly decreased total GLUT12 protein expression in rest biopsies, only visible in the VL ( p = 0.0359). This decrease was even more prominent in the VL after acute exercise in trained condition (PVL = 0.0025). Conclusion: The important changes seen in GLUT12 expression downregulation, both in response to training and acute exercise in the horse, the downregulation of GLUT4 expression after acute exercise in trained condition and the lack of differential shifts in GLUT8 expression in any of the studied conditions, questions the importance of glucose as substrate to fuel training and exercise in healthy horses. Free formaldehyde is a carcinogen whose emission reduction in particleboard has been studied recently to mitigate this environmental and human health problem. One alternative to reduce the emission of formaldehyde in particleboards is by using adhesives produced from natural sources. Cardanol-formaldehyde is an environmentally friendly adhesive made with cashew nut liquid, a byproduct from the cashew chain. This work aimed to produce particleboard using cardanol-formaldehyde in place of urea. In addition, different proportions of bean straw wastes were used to replace pine wood. The combination of eco-friendly adhesive and lignocellulosic waste particles could result in a product that meets market demands while being environmentally nonaggressive. Cardanol-formaldehyde promoted a higher modulus of elasticity (MOE) (1172 MPa) and modulus of rupture (MOR) (4.39 MPa) about panels glued with urea-formaldehyde, which presented a MOE of 764 MPa and MOR of 2.45 MPa. Furthermore, the cardanol-formaldehyde adhesive promoted a 93% reduction in formaldehyde emission, with a reduction from 16.76 to 1.09 mg/100 g oven-dry board for particleboards produced with cardanol-formaldehyde, indicating potential as an adhesive in the particleboard industry., (© 2023. 41. Molecular Cloning and AlphaFold Modeling of Thyrotropin (ag-TSH) From the Amazonian Fish Pirarucu ( Arapaima gigas ).

Arapaima gigas , known as Pirarucu in Brazil, is one of the largest freshwater fish in the world. Some individuals could reach 3 m in length and weight up to 200 kg. Due to extinction risks and its economic value, the species has been a focus for preservation and reproduction studies. Thyrotropin (TSH) is a glycoprotein hormone formed by 2 subunits α and β whose main activity is related to the synthesis of thyroid hormones (THs)-T3 and T4. In this work, we present a combination of bioinformatics tools to identify Arapaima gigas βTSH (ag-βTSH), modeling its molecular structure and express the recombinant heterodimer form in mammalian cells. Using the combination of computational biology, based on genome-related information, in silico molecular cloning and modeling led to confirm results of the ag-βTSH sequence by reverse transcriptase-polymerase chain reaction (RT-PCR) and transient expression in human embryonic kidney (HEK293F) cells. Molecular cloning of ag-βTSH retrieved 146 amino acids with a signal peptide of 21 amino acid residues and 6 disulfide bonds. The sequence has a similarity to 39 fish species, ranging between 43.1% and 81.6%, whose domains are extremely conserved, such as cystine knot motif and N-glycosylation site. The Arapaima gigas thyrotropin (ag-TSH) model, solved by AlphaFold, was used in molecular dynamics simulations with Scleropages formosus receptor, providing similar values of free energy ΔG
bind and ΔGPMF in comparison with Homo sapiens model. The recombinant expression in HEK293F cells reached a yield of 25 mg/L, characterized via chromatographic and physical-chemical techniques. Context: The reactivity of graphene oxide (GO) with amines is related to the ring-opening of the epoxy groups in its basal surface, as addressed experimentally. Therefore, discussing the hydroxyl/epoxy ratio for GO is relevant to improve the characterization of such material. As the adsorption of Methylene Blue into GO is related to a graphene derivative's oxidation degree (OD), we combined adsorption experimental information and theoretical data to estimate the hydroxyl/epoxy ratio. The theoretical data were compared to the experimental adsorption of Methylene Blue and Indigo Carmine into GO synthesized in our department. Our results show GO systems with hydroxyl/epoxy ratios equal to 0.7, 0.8, and 0.9 are the most representative in which the most coherent model corresponds to OH/EP=0.8 for our GO previously synthesized., Methods: The GO-MODEL software was developed in the present work to obtain cartesian coordinates of GO systems. We investigated 204 systems comprising models with 486 carbon atoms with the GFN2-xTB semiempirical quantum method. In a previous work, the common gonadotrophic hormone α-subunit (ag-GTHα), the ag-FSH β- and ag-LH β-subunit cDNAs, were isolated and characterized by our research group from A. gigas pituitaries, while a preliminary synthesis of ag-FSH was also carried out in human embryonic kidney 293 (HEK293) cells. In the present work, the cDNA sequence encoding the ag-growth hormone (ag-GH) has also been isolated from the same giant Arapaimidae Amazonian fish. The ag-GH consists of 208 amino acids with a putative 23 amino acid signal peptide and a 185 amino acid mature peptide. The highest identity, based on the amino acid sequences, was found with the Elopiformes (82.0%), followed by Anguilliformes (79.7%) and Acipenseriformes (74.5%). The identity with the corresponding human GH (hGH) amino acid sequence is remarkable (44.8%), and the two disulfide bonds present in both sequences were perfectly conserved. Three-dimensional (3D) models of ag-GH, in comparison with hGH, were generated using the threading modeling method followed by molecular dynamics. Our simulations suggest that the two proteins have similar structural properties without major conformational changes under the simulated conditions, even though they are separated from each other by a >100 Myr evolutionary period (1 Myr = 1 million years). The sequence found will be used for the biotechnological synthesis of ag-GH while the ag-GH cDNA obtained will be utilized for preliminary Gene Therapy studies.

Coffee (Coffea L.) is one of the main crops produced globally. Its contamination by the fungus Hemileia vastatrix Berkeley and Broome has been economically detrimental for producers. The objective of this work was to extract and characterize the essential oils from Eucalyptus citriodora Hook, Eucalyptus camaldulensis Dehn and Eucalyptus grandis Hill ex Maiden, produce and characterize nanoparticles containing these essential oils and evaluate the in vivo and in vitro antifungal activity of free and nanoencapsulated essential oils. The principal constituent of the essential oil from E. citriodora was citronellal; that from E. grandis was α-pinene; and that from E. camaldulensis was 1,8-cineol. The in vitro antifungal activity against the fungus H. vastatrix was 100% at a concentration of 1000 μl l
-1 for all the oils and nanoparticles containing these natural products. The sizes of the nanoparticles produced with the essential oils from E. citriodora, E. camaldulensis and E. grandis were 402·13 nm, 275·33 nm and 328·5 nm, respectively, with surface charges of -11·8 mV, -9·24 mV and - 6·76 mV, respectively. Fourier transform infrared analyses proved that the encapsulation of essential oils occurred in the polymeric matrix of poly(ε-caprolactone). The incorporation of essential oils into biodegradable poly(ε-caprolactone) nanoparticles increased their efficiency as biofungicides in the fight against coffee rust, decreasing the severity of the disease by up to 90·75% after treatment with the nanoparticles containing the essential oil from E. grandis., (© 2022 Society for Applied Microbiology.)- Published

Plant microbiota is usually enriched with bacteria producers of secondary metabolites and represents a valuable source of novel species and compounds. Here, we analyzed the diversity of culturable root-associated bacteria of the medicinal native plant Baccharis trimera (Carqueja) and screened promising isolates for their antimicrobial properties. The rhizobacteria were isolated from the endosphere and rhizosphere of B. trimera from Ponta Grossa and Ortigueira localities and identified by sequencing and restriction analysis of the 16S rDNA. The most promising isolates were screened for antifungal activities and the production of siderophores and biosurfactants. B. trimera presented a diverse community of rhizobacteria, constituted of 26 families and 41 genera, with a predominance of Streptomyces and Bacillus genera, followed by Paenibacillus, Staphylococcus, Methylobacterium, Rhizobium, Tardiphaga, Paraburkholderia, Burkholderia, and Pseudomonas. The more abundant genera were represented by different species, showing a high diversity of the microbiota associated to B. trimera. Some of these isolates potentially represent novel species and deserve further examination. The communities were influenced by both the edaphic properties of the sampling locations and the plant niches. Approximately one-third of the rhizobacteria exhibited antifungal activity against Sclerotinia sclerotiorum and Colletotrichum gloeosporioides, and a high proportion of isolates produced siderophores (25%) and biosurfactants (42%). The most promising isolates were members of the Streptomyces genus. The survey of B. trimera returned a diverse community of culturable rhizobacteria and identified potential candidates for the development of plant growth-promoting and protection products, reinforcing the need for more comprehensive investigations of the microbiota of Brazilian native plants and habitats., (© 2022. Essential oils encapsulated in a polymeric matrix can be used as an alternative method to control fungi and mycotoxins. The essential oils were extracted by hydrodistillation and characterized by gas chromatography. The nanofibres were produced from poly (acid lactic) (PLA) containing essential oils by the Solution Blow Spinning method. The antifungal and antimicotoxygenic properties were evaluated against Aspergillus ochraceus and Aspergillus westerdijkiae by the fumigation method. Terpinen-4-ol (20·23%), sabinene (20·18%), 1·8-cineole (16·69%) and γ-terpinene (11·03%) were the principal compounds present in the essential oil from Alpinia speciosa, whereas citral (97·67%) was dominant from Cymbopogon flexuosus. Microscopy images showed that the addition of essential oils caused an increase in the diameter of the nanofibres. The infrared spectroscopy results indicated the presence of essential oils in the PLA nanofibres. Differential scanning calorimetry curves also indicated the existence of interactions between the essential oils and polymeric macromolecules through their plasticizing action. The hydrophobic character of nanofibres was revealed by the contact angle technique. An antifungal effect was observed, the mycelial growths (3·25-100%) and the synthesis of ochratoxin A (25·94-100%) were inhibited by the presence of the nanofibres. Poly(lactic acid) (PLA) nanofibres containing different proportions of the essential oils from Ocimum basilicum L. and Ocimum gratissimum L. were prepared by solution blow spinning method. The essential oils were extracted by hydrodistillation and characterized by gas chromatography. MEV, contact angle, DSC and FTIR were used to characterize the nanofibres. The effect of bioative nanofibres on the growth of the fungus and on the production of ochratoxin A were evaluated using the fumigation test. Linalool, 1·8-cineole and camphor were the principal components of the essential oil from O. basilicum, and eugenol was the principal constituent in the oil from O. gratissimum. An increase in the average diameter of the nanofibres was observed with the addition of the essential oils. The essential oils acted as a plasticizer, resulting in a reduction in the crystallinity of the PLA. The encapsulation of essential oils in PLA nanofibres was verified by FTIR. An effective antifungal and antimicotoxygenic activity against Aspergillus ochraceus and Aspergillus westerdjikiae was observed for the bioative nanofibres. Herpetic dermatitis and oral recurrent herpes (ORH) are among the most common human infections. Antiviral drugs such as acyclovir (ACV) are used in the standard treatment for ORH. Despite its therapeutic efficacy, ACV is continuously and repetitively administered in high doses. In this sense, the development of controlled release drug delivery systems such as core-shell fibers have a great potential in the treatment of ORH. In this work, poly(lactic acid)/poly(ethylene glycol) (PLA/PEG) fibers were produced by solution blow spinning (SBS) for the controlled release of ACV encapsulated in the core. PLA/PEG nanofibers containing four different blend ratios (100:0, 90:10, 80:20 and 70:30 wt%) without or with 10 wt% ACV were characterized by scanning electron microscopy (SEM), thermogravimetry (TG) and differential scanning calorimetry (DSC). The ACV release profile for 21 days was accessed by UV-Vis spectroscopy. Static water contact angles of the spun fiber mats were measured by the sessile drop method to evaluate fiber wettability upon contact with skin for transdermal release. Cytotoxicity and antiviral efficacy against Herpes simplex viruses (HSV-1) were evaluated using Vero cells. ACV addition did not impact on morphology, but slightly improved thermal stability of the fibers. Addition of hydrophilic PEG in PLA/PEG blends, however, increased drug release as confirmed by contact angle measurements and release profile. The in vitro tests showed the effectiveness of the drug delivery systems developed in reducing HSV-1 viral titer, which is related to the judicious combination of polymers used in the fibrous mats, in addition to not being cytotoxic to Vero cells. These results show the great potential of PLA/PEG solution blow-spun fibers in the controlled release of ACV to develop practical devices for the treatment of cold sores, while favoring the aesthetic appearance by covering them with a soft tissue patch (fibrous mats)., (Copyright © 2022 Elsevier B.V. 50. Profiling the Aerobic Window of Horses in Response to Training by Means of a Modified Lactate Minimum Speed Test: Flatten the Curve.

There is a great need for objective external training load prescription and performance capacity evaluation in equestrian disciplines. Therefore, reliable standardised exercise tests (SETs) are needed. Classic SETs require maximum intensities with associated risks to deduce training loads from pre-described cut-off values. The lactate minimum speed (LMS) test could be a valuable alternative. Our aim was to compare new performance parameters of a modified LMS-test with those of an incremental SET, to assess the effect of training on LMS-test parameters and curve-shape, and to identify the optimal mathematical approach for LMS-curve parameters. Six untrained standardbred mares (3-4 years) performed a SET and LMS-test at the start and end of the 8-week harness training. The SET-protocol contains 5 increments (4 km/h; 3 min/step). The LMS-test started with a 3-min trot at 36-40 km/h [until blood lactate (BL) > 5 mmol/L] followed by 8 incremental steps (2 km/h; 3 min/step). The maximum lactate steady state estimation (MLSS) entailed >10 km run at the LMS and 110% LMS. The GPS, heartrate (Polar
® ), and blood lactate (BL) were monitored and plotted. Curve-parameters (R core team, 3.6.0) were (SET) VLa1 .5/2/4 and (LMS-test) area under the curve (AUC>/ ), LMS and Aerobic Window (AW) via angular vs. threshold method. Statistics for comparison: a paired t -test was applied, except for LMS: paired Wilcoxon test; ( p < 0.05). The Pearson correlation ( r > 0.80), Bland-Altman method, and ordinary least products (OLP) regression analyses were determined for test-correlation and concordance. Training induced a significant increase in VLa 1 .5/2/4 . The width of the AW increased significantly while the AUC> LMS and LMS decreased post-training (flattening U-curve). The LMS BL steady-state is reached earlier and maintained longer after training. BLmax was significantly lower for LMS vs. SET. The 40° angular method is the optimal approach. The correlation between LMS and VMLSS was significantly better compared to the SET. The VLa4 is unreliable for equine aerobic capacity assessment. The LMS-test allows more reliable individual performance capacity assessment at lower speed and BL compared to SETs. The LMS-test protocol can be further adapted, especially post-training; however, inducing modest hyperlactatemia prior to the incremental LMS-stages and omitting inclusion of a per-test recovery contributes to its robustness.
