ABSTRACT Pest regulation may be favoured in crop fields adjacent to semi‐natural vegetation such as fragments of the Atlantic Forest in Southeastern Brazil. Therefore, the objective of this study was to analyse the dynamics of mortality factors for eggs and larvae of Diatraea saccharalis (Lepidoptera: Crambidae) in sugarcane adjacent to two types of forest fragments (Plateau and Gallery) at two distances from these fragments during two growing seasons. In each area (seven sugarcane plots), two 50 m long transects were established, parallel and spaced at 5 and 100 m from the field edge, and cohorts of the insect at each stage of development were monitored. The collected data were used to construct an ecological life table and determine the main mortality factors. Natural mortality factors that could not be characterised were grouped as unknown. In both seasons and stages of development observed, there was no difference in mortality between the two types of forest fragments (Plateau and Gallery) or between the two distances from the forest. For the egg stage, the mortality factors were classified as inviable due to infertility, predation, parasitism, desiccation and dislodgement. In both seasons, there was a significant difference in the natural mortality factors of D. saccharalis, with predation distinct from the others. Ants (Hymenoptera: Formicidae) and green lacewings (Neuroptera: Chrysopidae) were the most frequent predatory agents and occurred at both distances and types of forest fragments. For the larval stage, the mortality factors were classified as predation, parasitism, drowning and unknown, with the unknown group differing from the others. Unknown factors might have included predation, larval movement towards other plants and displacement caused by wind after hatching. Overall, predation is a key mortality component of D. saccharalis suppression. Therefore, agricultural practices focused on the conservation of natural enemies such as predators may provide more sustainable management. [ABSTRACT FROM AUTHOR]

Purpose: Hepatic fibrosis develops as a response to chronic liver injury, resulting in the formation of fibrous scars. This process is initiated and driven by collagen-producing activated myofibroblasts which reportedly express high levels of platelet derived growth factor receptor-β (PDGFRβ). We therefore regard PDGFRβ as an anchor for diagnosis and therapy. The Fibrobody® SP02SP26-ABD is a biparatopic VHH-construct targeting PDGFRβ. Here, we explore its potential as a theranostic vector for liver fibrosis. Methods: Specificity, cross-species binding, and cellular uptake of SP02SP26-ABD was assessed using human, mouse and rat PDGFRβ ectodomains and PDGFRβ-expressing cells. Cellular uptake by PDGFRβ-expressing cells was also evaluated by equipping the Fibrobody® with auristatinF and reading out in vitro cytotoxicity. The validity of PDGFRβ as a marker for active fibrosis was confirmed in human liver samples and 3 mouse models of liver fibrosis (DDC, CCl4, CDA-HFD) through immunohistochemistry and RT-PCR. After radiolabeling of DFO*-SP02SP26-ABD with 89Zr, its in vivo targeting ability was assessed in healthy mice and mice with liver fibrosis by PET-CT imaging, ex vivo biodistribution and autoradiography. Results: SP02SP26-ABD shows similar nanomolar affinity for human, mouse and rat PDGFRβ. Cellular uptake and hence subnanomolar cytotoxic potency of auristatinF-conjugated SP02SP26-ABD was observed in PDGFRβ-expressing cell lines. Immunohistochemistry of mouse and human fibrotic livers confirmed co-localization of PDGFRβ with markers of active fibrosis. In all three liver fibrosis models, PET-CT imaging and biodistribution analysis of [89Zr]Zr-SP02SP26-ABD revealed increased PDGFRβ-specific uptake in fibrotic livers. In the DDC model, liver uptake was 12.15 ± 0.45, 15.07 ± 0.90, 20.23 ± 1.34, and 20.93 ± 4.35%ID/g after 1,2,3 and 4 weeks of fibrogenesis, respectively, compared to 7.56 ± 0.85%ID/g in healthy mice. Autoradiography revealed preferential uptake in the fibrotic (PDGFRβ-expressing) periportal areas. Conclusion: The anti-PDGFRβ Fibrobody® SP02SP26-ABD shows selective and high-degree targeting of activated myofibroblasts in liver fibrosis, and qualifies as a vector for diagnostic and therapeutic purposes. This article aims to discuss the quality of early childhood education and care in Brazil and the process of infants' and toddlers' "Inserimento" to daycare. The theoretical framework of our arguments is based on the notion of quality of child education as a negotiated concept, and by the Italian concept of "Inserimento." The methodology is based on the Mosaic Approach, a participative method to listen to young children. As part of the methodology, we participated in the "inserimento" process in five municipalities in southeastern Brazil, from 2016 and 2020. Data analysis was based on the pedagogy of listening and on studies on gender and racial relationships. The results show how laws, normative documents, family expectations, planning and routine of childhood education, available toys, space, babies, families, and professionals creates a complex mosaic of discourses about what constitutes the quality of childhood education in the babies' first days in daycare centers.
