The present study aims to indirectly evaluate the occurrence of suction and its effects in laboratory tests of a chemically stabilized soft soil. To achieve these goals, a series of unconfined compressive strength (UCS) and triaxial tests were performed, with and without the application of cyclic loading and with and without membrane during the cyclic loading stage, to evaluate the occurrence of suction and its effect on the mechanical behavior of a chemically stabilized soft soil. The results show that during the cyclic loading stage there is an occurrence of suction which influences both the permanent axial strain during the cyclic loading and the maximum strength of the stabilized soil. During the cyclic loading stage, the occurrence of suction promoted the increase of the plastic deformation. In terms of strength, it is observed an occurrence of suction in the stabilized soil which may be related with the increase of the strength. It is observed that suction can be avoided by saturation of the sample, verified in triaxial tests, or by using a membrane during the cyclic UCS test.

Age is a prominent risk factor for cardiometabolic disease, often leading to heart structural and functional changes. However, precise molecular mechanisms underlying cardiac remodeling and dysfunction exclusively resulting from physiological aging remain elusive. Previous research demonstrated age‐related functional alterations in baboons, analogous to humans. The goal of this study is to identify early cardiac molecular alterations preceding functional adaptations, shedding light on the regulation of age‐associated changes. Unbiased transcriptomics of left ventricle samples are performed from female baboons aged 7.5–22.1 years (human equivalent ≈30–88 years). Weighted‐gene correlation network and pathway enrichment analyses are performed, with histological validation. Modules of transcripts negatively correlated with age implicated declined metabolism‐oxidative phosphorylation, tricarboxylic acid cycle, glycolysis, and fatty‐acid β‐oxidation. Transcripts positively correlated with age suggested a metabolic shift toward glucose‐dependent anabolic pathways, including hexosamine biosynthetic pathway (HBP). This shift is associated with increased glycosaminoglycan synthesis, modification, precursor synthesis via HBP, and extracellular matrix accumulation, verified histologically. Upregulated extracellular matrix‐induced signaling coincided with glycosaminoglycan accumulation, followed by cardiac hypertrophy‐related pathways. Overall, these findings revealed a transcriptional shift in metabolism favoring glycosaminoglycan accumulation through HBP before cardiac hypertrophy. Simple Summary: Sperm cells are specialized cells that are highly susceptible to different kinds of damage, impairing the success of in vitro fertilization and, consequently, of assisted reproductive techniques (ART). Oxidative stress plays a significant role in this context causing DNA injury, reduced motility, and compromising the fertilization ability of sperm. The use of antioxidants aims to mitigate the oxidative stress, protect sperm cells from damage, and enhance their fertilization potential. Urolithin A (UA) is a natural compound with proven anti-aging and antioxidant effects in various cells, but its effect on bovine sperm has not been tested yet. The objective of this study was to investigate the impact of UA on bovine sperm function and fertilization rate, thus contributing to animal reproductive development. The results showed that UA improved sperm motility quality and reduced oxidative stress levels in a dose-dependent manner. The use of UA as an antioxidant therapy may have a significant impact on the advancement of reproductive biotechnologies. Reactive oxygen species (ROS) play a critical role in the functional competence of sperm cells. Conversely, excessive generation of ROS can impair sperm function, including their fertilization ability. Urolithin A (UA), a gut bacteria-derived metabolite produced from the transformation of ellagitannins, with anti-aging and antioxidant properties, was investigated for the first time in bovine sperm cells in the present study. Firstly, different doses of UA (0, 1, and 10 μM; 8–16 sessions) were used during the capacitation process of frozen-thawed bovine sperm. Sperm motility was assessed using optical microscopy and CASA. Sperm vitality (eosin-nigrosin), ROS, and ATP levels, as well as mitochondrial membrane potential (JC1) and oxygen consumption were evaluated. A second experiment to test the effect of different doses of UA (0, 1, and 10 μM; 9 sessions) in both the capacitation medium, as above, and the fertilization medium, was also implemented. The embryonic development and quality were evaluated. UA, at a concentration of 1 μM, significantly improved sperm movement quality (p < 0.03). There was a trend towards an increase in the oxygen consumption rate (OCR) of capacitated sperm with 1 μM and 10 μM UA supplementation. Moreover, an increase in ATP levels (p < 0.01) was observed, accompanied by a reduction in ROS levels at the higher UA concentration. These results suggest that UA may enhance spermatozoa mitochondrial function, modifying their metabolic activity while reducing the oxidative stress. Also, the number of produced embryos appears to be positively affected by UA supplementation, although differences between the bulls may have mitigated this effect. In conclusion, presented results further support previous findings indicating the potential therapeutic value of UA for addressing reproductive sub/infertility problems and improving ART outcomes. In addition, our results also reinforce the important bull effect on ART and that male sperm bioenergetic parameters should be used to predict spermatozoa functionality and developmental potential. When a soil is subjected to cyclic loading, there are changes in the material's geomechanical behaviour that need to be characterised before safely designing any future projects. In terms of cyclic loading, it is important to characterise not only the failure of the soil but also its behaviour before failure, in particular the yield point and the elastic behaviour of the material. This study examines the effects of the number of loading cycles on the behaviour of a chemically stabilised soft soil with a particular focus on the yield surface. To this end, a series of triaxial tests were performed on specimens, previously or not subjected to a different number of loading cycles (1000–100 000). The results were analysed in terms of the evolution of accumulated permanent axial strain, the yield surface and stress–strain behaviour. It was observed that an increase in the number of loading cycles promoted: an increase in the permanent axial strain, an increase in the undrained resilient modulus, a shrinkage of the yield surface but its shape is maintained, and there is a small increase in the peak strength of the stabilised soil explained by the strain-hardening effect induced by the cyclic loading. Robust data management establishes credibility, fostering trust within the scientific community and benefiting researchers' careers. In experimental biomedicine, comprehensive data management is vital due to the typically intricate protocols, extensive metadata, and large datasets. Low-throughput experiments, in particular, require careful management to address variations and errors in protocols and raw data quality. Transparent and accountable research practices rely on accurate documentation of procedures, data collection, and analysis methods. Proper data management ensures long-term preservation and accessibility of valuable datasets. Well-managed data can be revisited, contributing to cumulative knowledge and potential new discoveries. Publicly funded research has an added responsibility for transparency, resource allocation, and avoiding redundancy. Meeting funding agency expectations increasingly requires rigorous methodologies, adherence to standards, comprehensive documentation, and widespread sharing of data, code, and other auxiliary resources. This review provides critical insights into raw and processed data, metadata, high-throughput versus low-throughput datasets, a common language for documentation, experimental and reporting guidelines, efficient data management systems, sharing practices, and relevant repositories.
