Soil compaction is one of the primary threats to soil degradation in Europe; however, data to guide grassland farmers on how to avoid traffic induced soil compaction is limited. In grassland systems, soil moisture regimes are measured by daily soil moisture deficit (SMD) values and, when coupled with soil physical indicators could help safeguard the soil physical quality (SPQ). The objective of this study is to investigate how soil physical quality changes across different induced traffic compaction events at targeted SMD. A field study at Johnstown Castle Beef Farm (Wexford, Ireland) investigated the severity of soil physical changes caused by machine trafficking across different targeted soil moisture regimes. A tractor and a fully loaded slurry tanker trafficked moderately drained soil plots at SMD targets of 10 (dry (D)), 0 (moist (M)) and – 10 (wet (W)) mm. Compaction events simulated four passes across one year of grassland management: at the time of the first silage cut, in April; after the first cut silage harvest, in June; before the slurry spreading opening season, in October; and at the beginning of the slurry spreading period, in January. Soil bulk density (BD) samples were taken in the middle of the tyre marks at different depths (0-10, 10-20 and 20-30 cm). To examine indirect soil physical quality of various treatments, soil physical data was used to calculate the S value (Si) using the SawCal model. Resistance to insecticides used to control pests is an issue of increasing concern for agriculture. The grain aphid, Sitobion avenae, is a pest of cereals and grasses worldwide, and one of growing concern due to the evolution of resistance to certain insecticides. Resistance confers benefits to insects by enabling them to survive exposure to insecticide compounds; however, the mutations conferring resistance may also penalise the insect in pesticide-free environments due to fitness costs associated with the new phenotype. Here we tested the hypothesis of a reproductive penalty linked to the knockdown resistance mutation (kdr) to pyrethroid insecticides. The mutation occurs predominantly in a single SA3 clone. To date, only heterozygous-resistant forms (kdr-SR) have been detected in populations in Ireland and the UK, and this suggests that a fitness penalty may preclude the formation of both male and female heterozygous-resistant sexual forms. By designing an experiment which included a resistant and a non-resistant clone, we were able to simulate reduced daylight and temperature conditions which, in nature, trigger sexual reproduction and therefore study the responses of each clone. This allowed us to detect the switch from asexual females to sexual females and males and report on the conditions associated with the production of sexual forms. The results showed that both aphid clones were able to produce sexual forms with no difference in the onset of sexual reproduction, although reproductive strategies differed between clones. The later onset of male forms in the SA3 clone may decrease the likelihood of mating interactions to create fully resistant (kdr-RR) genotypes and this may constitute a fitness penalty due to pyrethroid resistance.

Background: Early intramuscular administration of SARS-CoV-2-neutralising monoclonal antibody combination, tixagevimab–cilgavimab, to non-hospitalised adults with mild to moderate COVID-19 has potential to prevent disease progression. We aimed to evaluate the safety and efficacy of tixagevimab–cilgavimab in preventing progression to severe COVID-19 or death. Methods: TACKLE is an ongoing, phase 3, randomised, double-blind, placebo-controlled study conducted at 95 sites in the USA, Latin America, Europe, and Japan. Eligible participants were non-hospitalised adults aged 18 years or older with a laboratory-confirmed SARS-CoV-2 infection (determined by RT-PCR or an antigen test) from any respiratory tract specimen collected 3 days or less before enrolment and who had not received a COVID-19 vaccination. A WHO Clinical Progression Scale score from more than 1 to less than 4 was required for inclusion and participants had to receive the study drug 7 days or less from self-reported onset of mild to moderate COVID-19 symptoms or measured fever. Participants were randomly assigned (1:1) to receive either a single tixagevimab–cilgavimab 600 mg dose (two consecutive 3 mL intramuscular injections, one each of 300 mg tixagevimab and 300 mg cilgavimab) or placebo. Randomisation was stratified (using central blocked randomisation with randomly varying block sizes) by time from symptom onset, and high-risk versus low-risk of progression to severe COVID-19. Participants, investigators, and sponsor staff involved in the treatment or clinical evaluation and monitoring of the participants were masked to treatment-group assignments. The primary endpoints were severe COVID-19 or death from any cause through to day 29, and safety. This study is registered with ClinicalTrials.gov, NCT04723394. Findings: Between Jan 28, 2021, and July 22, 2021, 1014 participants were enrolled, of whom 910 were randomly assigned to a treatment group (456 to receive tixagevimab–cilgavimab and 454 to receive placebo). The mean age of participants was 46·1 years (SD 15·2). Severe COVID-19 or death occurred in 18 (4%) of 407 participants in the tixagevimab–cilgavimab group versus 37 (9%) of 415 participants in the placebo group (relative risk reduction 50·5% [95% CI 14·6–71·3]; p=0·0096). The absolute risk reduction was 4·5% (95% CI 1·1–8·0; p Interpretation: A single intramuscular tixagevimab–cilgavimab dose provided statistically and clinically significant protection against progression to severe COVID-19 or death versus placebo in unvaccinated individuals and safety was favourable. Treating mild to moderate COVID-19 earlier in the disease course with tixagevimab–cilgavimab might lead to more favourable outcomes.

In addition, the economic burden of the disease by assessing COPD-related healthcare costs was described.Patients and Methods: Based on a German claims dataset, continuously insured individuals (04/2014-03/2019) aged 40 years or older with at least two incident pulmonologist’s diagnoses or one inpatient diagnosis of COPD (ICD-10-GM code J44.-; no respective diagnosis in a 12-month baseline period) were selected. Treatment patterns after incident diagnosis considering inhaled maintenance therapies identified by ATC codes (outpatient prescriptions) were analyzed. Prescription patterns were compared with recommendations of German COPD treatment guidelines. Severe exacerbations were assessed as hospitalizations with main diagnosis ICD-10-GM code J44.1. COPD-associated costs from the perspective of the health insurance fund AOK PLUS were calculated per patient-year (PY).Results: The sample comprised 17,464 incident COPD patients with a mean age of 71.5 years. 58.9% were male and the mean Charlson-Comorbidity-Index was 5.3. During follow-up (median: 2.0 years), 57.1% of the patients received at least one prescription of an inhaled maintenance therapy, whereas 42.9% did not. Among treated patients, 35.2% started their treatment with LABA/LAMA, 25.3% with LAMA monotherapy, 16.2% with LABA/ICS, and 7.8% with LABA/LAMA/ICS therapy. Within four weeks after initial diagnosis, ICS-containing therapies were prescribed in 14.1% of patients. Of all patients with a prescribed triple therapy, 68.9% had no corresponding exacerbation history documented. On average, 0.16 severe exacerbations and 0.19 COPD-related hospitalizations were observed per PY during available follow-up. Direct COPD-related costs were 3,693 €/PY, with COPD-related hospitalizations being responsible for about 79.2% of these costs.Conclusion: Long-acting bronchodilators are the mainstay of pharmacological treatment of incident COPD patients in Germany, in line with guideline recommendations. Stable Isotope Ratio Analysis (SIRA) is commonly used for the authentication of dairy commodities, providing evidence to support the geographical origin and production background of products. We set out to optimize methods for the isolation of a common constituent (casein) from three dairy commodities, which would permit easier inter- and intra-commodity comparisons following SIRA.Three published methods for isolation of protein (from cheese, milk, and butter) were adapted to yield protein (casein) fractions from commercial cheddar cheese, whole milk powder (WMP) and butter samples with a high degree of purity for subsequent SIRA. The casein fractions isolated underwent elemental analysis (H, C and N), protein determination, and some underwent SIRA for H, C, N, O, and S. Two-way analysis of variance and Tukey post-hoc comparisons tested differences between methods.For each product, an optimised casein isolation method was chosen based on the C/N ratio and protein content (%). An optimum solvent lipid extraction (petroleum spirit:diethyl ether (2:1)) and casein precipitation method was chosen following consistent results for cheddar cheese casein for all methods. A final solvent lipid extraction (heptane:isopropanol (3:2)) was necessary for WMP and butter casein extraction. δCasein of high purity, for subsequent SIRA, can be isolated from cheddar cheese, WMP and butter following modifications of previously published methods.

To improve understanding of real-world asthma treatment and inform physician education, we evaluated regional variation in asthma prevalence and oral corticosteroid (OCS) use across Germany. We developed a machine learning gradient boosted tree model with IMS® Disease Analyzer electronic medical records, which cover 3% of German patients. This model had a 91% accuracy in predicting the presence of asthma and chronic obstructive pulmonary disease. We applied the model to the IMS® Longitudinal Prescription database, with 82% national coverage, to classify patients receiving treatment for airflow obstruction from October 2017–September 2018 in 63 regions in Germany. Of 2.4 million individuals under statutory health insurance with predicted high OCS use for asthma (defined as 1 tablet per day for the duration of therapy), 13.7%, 18.7%, 36.5%, 29.4%, and 1.7% were categorised as receiving Global Initiative for Asthma (GINA) Steps 1, 2, 3, 4, and 5 treatment, respectively. Despite the availability of biologicals in recent years, studies show patients with inadequately controlled severe asthma in real life. The aim of the current study was to characterize and estimate the number of patients with NVL/GINA level 4 or 5 asthma and signs of poor control in Germany.Patients and Methods: In 2021, we retrospectively analyzed data collected during 2019 using the IQVIA™ LRx and IQVIA™ Disease Analyzer databases which contain anonymized longitudinal data covering approximately 80% of statutory health insurance (GKV) prescriptions in Germany with most relevant information about prescriptions, basic patient demographics or location of the prescriber; the IQVIA™ Disease Analyzer anonymized electronic medical records from a representative sample of office-based GPs and specialists. An expert committee of pulmonologists from different hospitals and expert practices supported the study. Asthma patients treated according to NVL/GINA 4/5 who used SABAs frequently (≥ 3 on days with no ICS-containing prescriptions/year) and/or received prescriptions for oral corticosteroids (OCS) (score of ≥ 2/year, a pulmonologist prescription scored 1.0, GP 0.75) were classified as severe, uncontrolled asthma.Results: In 2019, 3.4 million patients received at least two prescriptions of respiratory medications and 2.4 million patients on maintenance respiratory treatment have asthma. A total of 625,000 asthma patients were treated according to NVL/GINA step 4 or 5. Among these, 54,000 were uncontrolled according to the pre-defined OCS and/or SABA use, which corresponds to approximately 15% of patients in certain regions.Conclusion: In 2019, approximately 54,000 patients in Germany treated according to NVL/GINA step 4/5 had evidence suggestive for poor asthma control, up to 15% of patients in certain regions. Management of patients with respiratory disorders, such as asthma or chronic obstructive pulmonary disease (COPD), became challenging during the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) pandemic due to infection prevention measures. To maintain care, a remote monitoring program was initiated, comprising a smartphone app and a Bluetooth spirometry device.To assess patient- and physician-related experience with remote monitoring.Structured questionnaires were developed to rate experiences from the patient or physician perspective on six-level Likert scales. Interactions between patients and physicians via the digital platform and overall utilization was analyzed.A total of 745 patients with asthma, COPD, post-coronavirus disease 2019 (COVID-19) and other respiratory diseases were enrolled from 31 centers in Germany. Mean follow-up was 49.4 ± 12.6 weeks. Each participant submitted on average 289 measurements. Patient-reported experience with the remote monitoring program was positive, with the highest satisfaction reported for "Experience with home measurement" (1.4 ± 0.5; 99% positive), followed by "Communication/interaction" (1.8 ± 0.9; 83% positive) and "Overall satisfaction with program" (1.8 ± 0.8; 87% positive). In all, 70% reported subjective quality of life improvements related to participation in the program. Physician satisfaction with the program was also high with a mean rating of 2.2 ± 1.2.App-based remote monitoring was successfully implemented in routine care during the SARS-CoV‑2 pandemic and demonstrated potential for improvements in care. Patient-relevant experience was positive in all dimensions and remote monitoring was well accepted. Physicians who participated in the program also expressed positive experiences, as demonstrated by a high level of interaction with the platform and positive evaluations of effects from the program.HINTERGRUND: Das Management von Patienten mit respiratorischen Erkrankungen ist seit Beginn der Severe-acute-respiratory-syndrome-coronavirus-2(SARS-CoV-2)-Pandemie durch Maßnahmen zur Infektionsprävention teilweise erschwert. Um die Versorgung zu gewährleisten, wurde ein digitales Versorgungsprogramm implementiert, mit dem Krankheitsverläufe von Patienten per Smartphone-App und Bluetooth-Spirometer überwacht werden können.Ermittlung der Erfahrungen von Patienten und Ärzten mit der digitalen Versorgung.Zur Analyse der Erfahrungen wurden strukturierte Fragebögen entwickelt, mit denen verschiedene Dimensionen aus Sicht von Patienten und Ärzten bewertet wurden. Nutzung und Interaktion wurden quantitativ erfasst.In das Programm wurden 745 Patienten mit Asthma, chronisch-obstruktiver Lungenerkrankung, Zustand nach „coronavirus disease 2019“ (COVID-19) sowie anderen Lungenerkrankungen eingeschlossen. Im mittleren Nachsorgezeitraum von 49,4 ± 12,6 Wochen erfolgten durchschnittlich 289 Messungen. Aus Patientenperspektive wurde das digitale Programm positiv bewertet: Die höchste Bewertung wurde für „Erfahrung mit der häuslichen Messung“ ermittelt (1,4 ± 0,5; 99 % positiv), gefolgt von „Kommunikation/Interaktion“ (1,8 ± 0,9; 83 % positiv). Ein Anteil von 70 % gab eine subjektive Verbesserung der Lebensqualität durch die Teilnahme am Programm an. Die Bewertung durch die Ärzte war mit einer mittleren Bewertung von 2,2 ± 1,2 ebenfalls positiv.Das App-basierte digitale Versorgungsprogramm konnte sinnvoll in die Routineversorgung während der SARS-CoV-2-Pandemie integriert werden und hat das Potenzial, die Versorgung auch darüber hinaus zu unterstützen. Patientenrelevante Erfahrungen sind in allen Dimensionen positiv und die digitale Versorgung wurde gut akzeptiert. Vonseiten der teilnehmenden Ärzte wird das Programm positiv bewertet, was sich an der hohen Interaktion mit der Plattform und den positiven Bewertungen der Effekte zeigt.

Soil organisms drive major ecosystem functions by mineralising carbon and releasing nutrients during decomposition processes, which supports plant growth, aboveground biodiversity and, ultimately, human nutrition. Soil ecologists often operate with functional groups to infer the effects of individual taxa on ecosystem functions and services. Simultaneous assessment of the functional roles of multiple taxa is possible using food-web reconstructions, but our knowledge of the feeding habits of many taxa is insufficient and often based on limited evidence. Over the last two decades, molecular, biochemical and isotopic tools have improved our understanding of the feeding habits of various soil organisms, yet this knowledge is still to be synthesised into a common functional framework. Here, we provide a comprehensive review of the feeding habits of consumers in soil, including protists, micro-, meso- and macrofauna (invertebrates), and soil-associated vertebrates. We have integrated existing functional group classifications with findings gained with novel methods and compiled an overarching classification across taxa focusing on key universal traits such as food resource preferences, body masses, microhabitat specialisation, protection and hunting mechanisms. Our summary highlights various strands of evidence that many functional groups commonly used in soil ecology and food-web models are feeding on multiple types of food resources. In many cases, omnivory is observed down to the species level of taxonomic resolution, challenging realism of traditional soil food-web models based on distinct resource-based energy channels. Novel methods, such as stable isotope, fatty acid and DNA gut content analyses, have revealed previously hidden facets of trophic relationships of soil consumers, such as food assimilation, multichannel feeding across trophic levels, hidden trophic niche differentiation and the importance of alternative food/prey, as well as energy transfers across ecosystem compartments. Wider adoption of such tools and the development of open interoperable platforms that assemble morphological, ecological and trophic data as traits of soil taxa will enable the refinement and expansion of the multifunctional classification of consumers in soil. The compiled multifunctional classification of soil-associated consumers will serve as a reference for ecologists working with biodiversity changes and biodiversity–ecosystem functioning relationships, making soil food-web research more accessible and reproducible.

Tezepelumab is a human monoclonal antibody that blocks the activity of thymic stromal lymphopoietin. SOURCE evaluated the oral corticosteroid-sparing effect of tezepelumab in adults with oral corticosteroid-dependent asthma.We conducted this phase 3, multicentre, randomised, double-blind, placebo-controlled study across 60 sites in seven countries. Participants aged 18-80 years with physician-diagnosed asthma, who had been receiving medium-dose or high-dose inhaled corticosteroids and had at least one asthma exacerbation in the 12 months before screening were eligible. Patients who were receiving medium-dose inhaled corticosteroids must have had their dose increased to a high dose for at least 3 months before screening. After an oral corticosteroid optimisation phase of up to 8 weeks, participants were randomly assigned according to a computer-generated fixed block randomisation sequence to receive tezepelumab 210 mg or placebo subcutaneously every 4 weeks during a 48 week treatment period (4 week induction phase, 36 week oral corticosteroid reduction phase, and 8 week maintenance phase). Randomisation was stratified by region. Participants, investigators, and site staff were masked to treatment assignment. The primary endpoint was the categorised percentage reduction from baseline in daily oral corticosteroid dose at week 48 without the loss of asthma control. Efficacy and safety endpoints were assessed in all participants who received at least one dose of study drug. This study is registered with ClinicalTrials.gov, NCT03406078.Between March 5, 2018, and Sept 27, 2019, 150 participants were randomly assigned to receive tezepelumab 210 mg (n=74) or placebo (n=76). The cumulative odds of achieving a category of greater percentage reduction in an oral corticosteroid dose for daily maintenance at week 48 were similar with tezepelumab or placebo in the overall population (odds ratio [OR] 1·28 [95% CI 0·69-2·35], p=0·43; the primary endpoint was not met). The cumulative odds were higher with tezepelumab than with placebo in participants with baseline blood eosinophil counts of at least 150 cells per μL (2·58 [1·16-5·75]), but not in participants with counts below 150 cells per μL (0·40 [0·14-1·13]). Tezepelumab was well tolerated, with no safety concerns identified. 53 (72%) of 74 tezepelumab-assigned participants and 65 (86%) of 76 placebo-assigned participants reported an adverse event. Serious adverse events were reported in 12 (16%) participants in the tezepelumab group and 16 (21%) participants in the placebo group.We did not observe a significant improvement in oral corticosteroid dose reduction with tezepelumab versus placebo in the overall population of this oral corticosteroid-sparing study, although an improvement was observed in participants with baseline blood eosinophil counts of at least 150 cells per μL.AstraZeneca and Amgen.

Harvesting whole-tree biomass for biofuel combustion intensifies removal of nutrients from the ecosystem. This can be partly abated by applying ash from the combustion back to the system, as the ash is rich in nutrients. Ash is very alkaline and ash application raises soil pH, which in turn can stimulate microbial activity and thus decomposition and mineralization. Our aim was to test if ash induced decomposition activity was associated with enhanced turnover of recalcitrant, i.e. relatively old, organic pools. Two experiments were conducted in the same coniferous plantation after the application of 0, 3, 4.5 and 6 t ash ha −1, and 0, 3, 9, 15 and 30 t ash ha −1, respectively. We used natural abundance of 15N in mosses, mites and ectomycorrhizal fungi 26 months after ash application, as well as temporal variation in δ 15N values of ectomycorrhizal fungi, as an indicator of decomposition of recalcitrant organic matter in the first experiment. Furthermore, in the second experiment we used measurements of extracellular manganese peroxidase activity almost 4 years after ash application as an indication of potential decomposition of lignin, an important component of recalcitrant organic matter. The δ 15N signature increased significantly for ectomycorrhizal fungi, dead moss, Nothroid and Gamasida mites, and manganese peroxidase activity tended to increase, with increasing ash doses. This suggests that ash application stimulates turnover of recalcitrant organic matter, which can increase the available pool of nitrogen in the system. This will potentially enhance the fertilizer value of ash. However, the δ 15N in ectomycorrhizal fungi tended to peak at 18 months after ash application, before decreasing, suggesting that the turnover of recalcitrant organic matter is reduced again with time.

Determination of the ratios of natural stable isotopes (13C/12C and 15N/14N) in unfed Ixodes ricinus nymphs and adults, which, in their previous stage, fed on captive wild rodents (Apodemus sylvaticus and Myodes glareolus), wild birds (Parus major and Cyanistes caeruleus) or domestic ruminants (Ovis aries and Bos taurus), demonstrated that it is possible to identify each host category with confidence. First, the tickblood spacing, which is the difference between values obtained from ticks and the blood of hosts that they had fed on in the previous stage, was consistent (152 spacings investigated from 15 host individuals in total). Second, potential confounding factors (tick age and sex) did not affect the discriminatory power of the isotope patterns, nor did different rearing conditions (room temperature vs. 4 °C) or the duration of development (maximum of 430 days). The findings that the tickblood isotope spacings, across a diverse range of hosts, were similar and predictable, and that confounders had little or no effect on this, strongly support the usage of the isotope approach. Because each of the host categories has a different role in the population dynamics of I. ricinus and in tick‐borne pathogen ecology, the method described here has great potential for the clarification of tick and tick‐borne pathogen ecology in the field.

The quality of a soil is its ability to deliver functions providing ecosystem services, human health and well-being. Soil physical quality (SPQ) values use different parts of the soil water retention curve (SWRC) to calculate SPQ. For example, the plant available water capacity (PAWC) method is the difference in water content between permanent wilting point and field capacity. The S-index uses the slope of the SWRC at its inflection point and the relative air-water energy (AWr) is the integral of “dry” divided by the “wet” area of the SWRC. Increasing demand for soil monitoring policies calls for reliable and sensitive soil quality indicators (SQIs). The objectives of the study were to assess the sensitivity and applicability of SPQ indicators using direct and indirect data inputs. The indirect approach provided sufficient data complexity for the PAWC and S-index values, but the more complex AWr required the direct approach. PAWC and S-index values were identified as static SPQ indicators. The values obtained from these approaches should be used to form baseline static datasets and therefore have an indicative role only. The AWr value was identified as a dynamic SPQ indicator and provided required sensitivity to pick up temporal changes in SPQ. This indicator could be used at multiple scales and could even guide grassland management in terms of SPQ. Higher SWRC data resolution will require more complex hydraulic models to fit and will ultimately improve the accuracy of soil hydraulic data and improve the sensitivity of AWr as a SPQ indicator.

It is recognised that the establishment and function of soil biota is critical for successful mine residue rehabilitation. Bauxite residues are alkaline, saline and sodic and, whilst methods for establishing vegetation are well studied, little is known about key soil fauna such as earthworms. At a bauxite residue disposal area in Ireland, a 12-year-old rehabilitated residue was examined for evidence of earthworm populations. Five species of earthworm, dominated by Allolobophora chlorotica, were recorded in the rehabilitated residue representing the endogeic, epigeic and epi-anecic ecological groups. To further understand the potential for rehabilitated residues to support earthworm communities, a series of exposure tests was conducted. Whilst unamended residues (pH 10.2, EC 0.629 mS cm−1, ESP 54) was hostile to A. chlorotica survival, 100% survival was observed after 90 days for gypsum and organic-amended residue at salinity of up to 2.9 mS cm−1, possibly due to calcium becoming the dominant cation. Survival of earthworms at salinities higher than anticipated tolerance levels suggests that specific ion dominance plays a role in earthworm survival in saline soils. Percent mass change was negatively correlated with pH, EC and sodium content of the residues. Residue from the 12-year-old site also supported the anecic species Aporrectodea longa over 100 days. Background Grazed grassland management regimes can have various effects on soil fauna. For example, effects on earthworms can be negative through compaction induced by grazing animals, or positive mediated by increases in sward productivity and cattle dung pats providing a food source. Knowledge gaps exist in relation to the behaviour of different earthworm species i.e. their movement towards and aggregation under dung pats, the legacy effects of pats and the spatial area of recruitment. The present study addressed these knowledge gaps in field experiments, over 2 years, using natural and simulated dung pats on two permanent, intensively grazed pastures in Ireland. Results Dung pats strongly affected spatial earthworm distribution, with up to four times more earthworms aggregating beneath pats, than in the control locations away from pats. In these earthworm communities comprising 11 species, temporally different aggregation and dispersal patterns were observed, including absence of individual species from control locations, but no clear successional responses. Epigeic species in general, but also certain species of the anecic and endogeic groups were aggregating under dung. Sampling after complete dung pat disappearance (27 weeks after application) suggested an absence of a dung pat legacy effect on earthworm communities. Based on species distributions, the maximum size of the recruitment area from which earthworms moved to pats was estimated to be 3.8 m2 per dung pat. Since actual grazing over 6 weeks would result in the deposition of about 300 dung pats per ha, it is estimated that a surface area of 1140 m2 or about 11% of the total grazing area can be influenced by dung pats in a given grazing period. Conclusions This study showed that the presence of dung pats in pastures creates temporary hot spots in spatial earthworm species distribution, which changes over time. The findings highlight the importance of considering dung pats, temporally and spatially, when sampling earthworms in grazed pastures. Published comparisons of grazed and cut grasslands probably reached incorrect conclusions by ignoring or deliberately avoiding dung pats. Furthermore, the observed intense aggregation of earthworms beneath dung pats suggests that earthworm functions need to be assessed separately at these hot spots. Rare studies investigated influence of neonicotinoid insecticides on the whole soil biota including non-target invertebrates and microorganisms. And less is known about the consequent intervention on soil C processes. This study aimed to decipher Collembola-fungi-bacteria interactive effects on pathways of maize C translocation, combining isotopic tracer analysis of relevant compartments with high-throughput sequencing for bacterial and fungal genetic profiles. Dinotefuran was applied at 0 or 100 μg kg−1 (a simulating residual dosage) to microcosms containing soils, Collembola and 13C labelled maize. Dinotefuran drastically reduced the density and maize-derived biomass C of Collembola, while intensifying antagonistic associations between soil organisms, with flourishing growth of Ascomycota and Actinobacteria, e.g., Streptomyces. This led to higher soil organic C (SOC) mineralization (elevated by 9.8–10.5%) across soils, attributing to the shift in microbial taxonomic and functional guild, e.g., with the increased abundance of genes aligned to cytochrome P450. Maize decomposition was controlled by Collembola that primarily fed on maize, via grazing behavior that facilitated labile maize C preferred decomposers, e.g., Xanthomonadaceae. These findings elucidate the influence of minute dinotefuran on intra-linkages between biomes (Collembola, fungi and bacteria), and highlight such legacy effects on maize and SOC mineralization.
