Abstract The application of pesticides may adversely impact a variety of non-target organisms. The use of side-effect-free herbal remedies to protect against the toxicity of harmful pesticides such as etoxazole has gained attention in recent times. The current study aimed to reveal the potential mitigating efficacy of Achillea millefolium L. extract against etoxazole toxicity in Allium cepa L. A. cepa bulbs in the control group were applied with tap water, while bulbs in the treatment groups were applied with etoxazole at dose of 0.5 m/L and two different doses of A. millefolium extract (200 mg/L and 400 mg/L). The impact of the treatments on certain parameters was evaluated. The molecular docking analysis was employed to investigate the potential interactions of etoxazole with DNA species, DNA topoisomerases, tubulin proteins, glutamate-1-semialdehyde aminotransferase, and protochlorophyllide reductase. The phenolic profile of A. millefolium was assessed. Etoxazole exposure reduced rooting percentage, root length, weight gain, mitotic index, and levels of chlorophyll a and chlorophyll b. Conversely, etoxazole treatment led to an increase in chromosomal aberrations and micronuclei occurrence. The most frequently observed chromosomal aberrations induced by etoxazole, which serve as bioindicators of genotoxicity, were fragment, vagrant chromosome, sticky chromosome, unequal chromatin distribution, bridge, reverse polarization, and vacuolated nucleus. The levels of malondialdehyde and antioxidant enzyme (superoxide dismutase and catalase) activities were also elevated. Epidermis cell damage, flattened cell nucleus, thickened cortex cell wall, and thickened conduction tissue were the meristematic cell disorders triggered by etoxazole. Molecular docking studies showed that etoxazole can interact directly with DNA, tubulins, and the enzymes mentioned above. A. millefolium extract was found to contain a substantial quantity of phenolic compounds. A. millefolium extract, when co-administered with etoxazole, attenuated all toxic effects of etoxazole dose-dependently. In conclusion, A. millefolium may potentially serve as a reliable pharmacological shield against the toxicity of pesticides in non-target organisms.

Abstract The growth of industrialization growth the risk of vanadium (V) contamination. The objective of this study was to examine the impact of 200 µg L− 1 VCI3 -induced toxicity as well as the potential protective effect of 187.5 mg L− 1 and 375 mg L− 1 Hypericum perforatum (H. perforatum) extracts against this toxicity on the Allium cepa (A. cepa) model organism. For this purpose, a series of investigations were conducted on the growth physiology alterations (germination percentage, root elongation, weight gain), cytogenetic alterations (mitotic index, micronucleus, chromosomal aberrations), biochemical alterations (malondialdehyde, superoxide dismutase, catalase) and defects in meristematic tissue in A. cepa. In addition, the phenolic compound content of H. perforatum extract was determined by the LC/MS-MS method. V application negatively affected all the investigated parameters and caused a serious phytotoxic and genotoxic effect as well as oxidative stress in A. cepa. Conversely, no statistical difference was observed between the parameters of the groups treated with H. perforatum extract and those of the control group. The administration of H. perforatum extract combined with V resulted in a notable enhancement in germination percentage, root elongation, weight gain, mitotic index value, chlorophyll a level and chlorophyll b level. Additionally, it led to a reduction in micronucleus and chromosomal aberrations frequencies, as well as meristematic tissue defects. Furthermore, LC/MS-MS analysis demonstrated that H. perforatum extract contains phenolic compounds, including catechin, epicatechin, quercetin, oleuropein and rutin, which confer antioxidant properties to the extract. The study provided clear evidence that H. perforatum extract attenuates the toxic effects of V in A. cepa, which can be attributed to its high content of bioactive phenols. The findings of the study indicate that H. perforatum extract may serve as a protective natural agent for daily use against heavy metal toxicity.

Abstract Trifloxystrobin (TFS) is a widely used strobilurin class fungicide. Ginkgo biloba L. has gained popularity due to its recognized medicinal and antioxidant properties. The aim of this study was to determine whether Ginkgo biloba L. extract (Gbex) has a protective role against TFS-induced phytotoxicity, genotoxicity and oxidative damage in A. cepa. Different groups were formed from Allium cepa L. bulbs subjected to tap water (control), 200 mg/L Gbex (Gbex1), 400 mg/L Gbex (Gbex2), 0.8 g/L TFS solution (TFS), 200 mg/L Gbex + 0.8 g/L TFS (TFS + Gbex1) and 400 mg/L Gbex + 0.8 g/L TFS (TFS + Gbex2), respectively. The phenolic composition of Gbex and alterations in the morphological, physiological, biochemical, genotoxicity and anatomical parameters were evaluated. Rutin, protocatechuic acid, catechin, gallic acid, taxifolin, p-coumaric acid, caffeic acid, epicatechin, syringic acid and quercetin were the most prevalent phenolic substances in Gbex. Rooting percentage, root elongation, weight gain, chlorophyll a and chlorophyll b decreased by approximately 50%, 85%, 77%, 55% and 70%, respectively, as a result of TFS treatment compared to the control. In the TFS group, the mitotic index fell by 28% compared to the control group, but chromosomal abnormalities, micronuclei frequency and tail DNA percentage increased. Fragment, vagrant chromosome, sticky chromosome, uneven chromatin distribution, bridge, vacuole-containing nucleus, reverse polarization and irregular mitosis were the chromosomal abnormalities observed in the TFS group. The levels of proline (2.17-fold) and malondialdehyde (2.71-fold), as well as the activities of catalase (2.75-fold) and superoxide dismutase (2.03-fold) were increased by TFS in comparison to the control. TFS-provoked meristematic disorders were damaged epidermis and cortex cells, flattened cell nucleus and thickened cortex cell wall. Gbex combined with TFS relieved all these TFS-induced stress signs in a dose-dependent manner. This investigation showed that Gbex can play protective role in A. cepa against the phytotoxicity, genotoxicity and oxidative damage caused by TFS. The results demonstrated that Gbex had this antioxidant and antigenotoxic potential owing to its high phenolic content.

Hop stunt viroid (HSVd) infects various plants such as citrus, hop, almond, grapevine, pear, plum, peach, mulberry, fig, and pistachio. Medlar trees in an orchard in Malatya province of Türkiye were surveyed for the presence of HSVd in 2021. Twenty leaf and flower samples were collected and tested by RT-PCR methods using pathogen-specific primers. HSVd was found in five of the twenty medlar samples showing novel sequence variations. Two of the five HSVd variations were chosen at random and registered in GenBank. Both Turkish HSVd isolates had genomes that were 300 nucleotides long. The complete genome sequence of these variations was compared to isolates in GenBank. The nucleotide sequences of HSVd isolates exhibited 89.7-100% similarity with HSVd isolates found in various crops worldwide. Analysing the alignment of multiple sequences and conducting phylogenetic analyses revealed that identified HSVd variants clustered with citrus Türkiye (MZ995256), citrus Italy (KC584022), citrus Iran (GQ260203) and citrus Japan (X06719) isolates with 100% similarity rate and citrus China (FJ716172) and citrus Spain (AF213503) isolates with 99.5% and 98.0% similarity rates, respectively. To our knowledge, this is the first report of medlar serving as a natural host for HSVd. HSVd infection in medlar could be a problem in the future, and additional study is needed. The infection appears to be latent, but it might be a source of infection for susceptible plants.

Abstract Although the antioxidant properties of Melissa officinalis extract (Mox) are widely known, little work has focused on its protective capacity against heavy metal stress. The primary objective of this study was to determine the potential of Mox to mitigate manganese (II) chloride (MnCI2)-induced cyto-genotoxicity using the Allium and comet assays. Physiological, genotoxic, biochemical and anatomical parameters as well as the phenolic composition of Mox were examined in Allium cepa (L.). Application of 1000 µM MnCl2 reduced the rooting percentage, root elongation, weight gain, mitotic index and levels of chlorophyll a and chlorophyll b pigments compared to the control group. However, it increased micronuclei formation, chromosomal abnormality frequencies, tail DNA percentage, proline amount, lipid peroxidation level and meristematic damage severity. The activities of superoxide dismutase and catalase also increased. Chromosomal aberrations induced by MnCl2 were fragment, sticky chromosome, vagrant chromosome, unequal distribution of chromatin and bridge. Application of 250 mg/L Mox and 500 mg/L Mox along with MnCl2 significantly alleviated adverse effects dose dependently. The antioxidant activity bestowed by the phenolic compounds in Mox assisted the organism to combat MnCl2 toxicity. Consequently, Mox exerted remarkable protection against MnCl2 toxicity and it needs to be investigated further as a potential therapeutic option.

Drought is one of the most serious problems for human societies and ecosystems. Climate change has begun to affect the whole world, but regional climate change can have different effects. Precipitation is frequently used to determine drought; besides, temperature has started to be used in recent years. The effect of temperature on the occurrence of drought was investigated for the Marmara region, located in the northeast of Turkey. The spatiotemporal variability of drought in the Marmara region was analyzed by using the standardized precipitation index (SPI) and standardized precipitation evapotranspiration index (SPEI) for 3-, 6-, and 12-month series. Results show that SPEI droughts occur with longer duration and increased magnitude, which can be attributed to the rise in potential evapotranspiration rates with an increase in temperature. The correlation between SPI and SPEI is strong for the same time series, and the number of extreme drought values for SPI is higher than SPEI, while the number of severe and moderate drought values is lower compared to SPEI. The years 1989, 1990, 2001, 2007, and 2014 were characterized as the highest drought years throughout the region. The study concludes that temperature should be considered in future studies. HIGHLIGHTS This study evaluates drought in terms of short- and long-time periods in the Marmara region.; The drought has been analyzed temporally and spatially.; The study gives an idea about the importance of considering the temperature in the determination of drought.; The drought duration and intensity were analyzed according to the short- and long-time periods.; Standardized precipitation and evapotranspiration index droughts were found to last longer and be more severe than standardized precipitation index.;

Surveys were conducted in Malatya and Elazığ provinces of Eastern Türkiye between 2018–2019 to detect the presence and distribution of Deformed wing virus (DWV), Acute bee paralysis virus (ABPV) and Black queen cell virus (BQCV) infecting honey bees and Varroa mites. Thirty Varroa mites feeding on honey bees and 147 honey bees were collected from twenty-five apiaries of Malatya and fifteen apiaries of Elazığ. The collected samples were subjected to reverse transcriptase-polymerase chain reaction. All tested viruses were found to be present in the samples singly or as mixed infections. The prevalence of the viruses were 23.81%, 12.93%, and 10.20% for DWV, ABPV and BQCV, respectively. Five out of thirty Varroa mites tested for the viruses were found to be infected only with DWV. Randomly chosen sequences of each detected virus’s partial polyprotein gene region were registered in GenBank under the accession numbers OP805878, OP805879, OP805880, OP805887, OP805888, OP805889, OP805890, OP805891 (DWV), OP805881, OP805882, OP805883, OP805884 (ABPV) and OP805885, OP805886 (BQCV). The phylogenetic tree of the viral isolates were compared with the world isolates. DWV isolates were found to be closely related to the UK, Lebanon, Türkiye, France, Germany and Israel isolates, whereas ABPV isolates were related to Türkiye, South Africa, Slovenia, Serbia, France, Hungary, Syria and USA isolates and BQCV isolates were related to China, South Korea, Czech Republic, France, Hungary, Poland, South Africa, Australia and Sweden isolates. The presence of DWV, ABPV and BQCV infections in Malatya and Elazığ provinces of Türkiye is revealed for the first time in this study.

Abstract The excessive use of metaldehyde in agriculture to combat mollusks endangers both the environment and non-target organisms. The aim of this study is to investigate the toxicity caused by metaldehyde in Allium cepa with the help of physiological, cytogenetic, biochemical and anatomical parameters. Also, DNA fragmentation caused by metaldehyde in root tip cells was measured by the "Comet Assay" method. The control group was germinated with tap water and the application groups were germinated with 20 mg/L metaldehyde, 40 mg/L metaldehyde, 100 mg/L metaldehyde and 200 mg/L metaldehyde for 72 h. The results of the physiological parameters showed that metaldehyde had a growth-limiting effect in A. cepa, depending on the application dose. According to root elongation levels, the EC50 (effective concentration) value for metaldehyde was 60.6 mg/L in A. cepa. As the treatment dose increased, the incidence of micronucleus and chromosomal aberrations gradually increased while mitotic index decreased. Metaldehyde exposure induced damages such as sticky chromosome, fragment, unequal distribution of chromatin, reverse polarization, bridge, and multipolar anaphase. In addition, metaldehyde caused cell damage in epidermis and cortex, thickening of the cortex cell wall and flattened cell nucleus in root meristem. Increasing doses of metaldehyde application also increased malondialdehyde levels, superoxide dismutase and catalase activities. As a result, it has been determined that the toxicity of metaldehyde in plants is versatile and the A. cepa test material is a suitable biological indicator to determine this toxicity.

Abstract The current study was undertaken to assess the attenuating potential of lycopene against Dithane toxicity in Allium cepa L. roots. A. cepa bulbs were arranged in 6 groups. The control group was treated with tap water while the other groups were treated with 215 mg/L lycopene, 430 mg/L lycopene, 500 mg/L Dithane, 500 mg/L Dithane + 215 mg/L lycopene and 500 mg/L Dithane + 430 mg/L lycopene, respectively. When the treatments were completed, growth inhibition, biochemical, genotoxicity and meristematic cell injury analyses were performed. Lycopene did not cause any toxic effect when applied alone. While rooting percentage, root elongation, weight gain and mitotic index (MI) decreased in response to Dithane exposure, the frequency of micronucleus (MN) and chromosomal abnormalities (CAs) in addition to malondialdehyde (MDA) level and the catalytic activities of superoxide dismutase (SOD) and catalase (CAT) increased. Dithane promoted fragment, sticky chromosome, vagrant chromosome, unequal distribution of chromatin, bridge, nucleus bud and reverse polarization formation in meristem cells. Dithane also provoked meristematic cell injuries, including indistinct appearance of vascular tissue, epidermis cell damage and flattened cell nucleus. Lycopene mitigated all damage types, depending on the lycopene dose applied with Dithane. Hence, the data analysis revealed that lycopene provides exceptional antioxidant protection against the fungicide Dithane, which has devastating toxic potential.

Abstract Etoxazole is among the systemic pesticides with acaricidal and insecticidal characteristics. This paper reports the first evaluation of the toxic effects of Etoxazole on Allium cepa L. Etoxazole solutions were applied to three groups formed from A. cepa bulbs at 0.125 mL/L, 0.25 mL/L and 0.5 mL/L doses, respectively. The control group was treated with tap water throughout the experimental period. The toxic effects of Etoxazole became more apparent as the dose of Etoxazole was increased. The growth-limiting effect was most pronounced in the highest dose group with approximately 29%, 70% and 58.5% reductions in germination percentage, root elongation and weight gain, respectively. The genotoxic effect of Etoxazole was most severe in the 0.5 mL/L dose group. In this group, the mitotic index decreased by 30% compared to the control group, while the micronucleus frequency increased to 45.3 ± 3.74. The most observed aberrations were fragment, vagrant chromosome, sticky chromosome, unequal distribution of chromatin, bridge, reverse polarization and nucleus with vacuoles. The malondialdehyde level showed a gradual increase with increasing Etoxazole doses and reached 2.7 times that of the control group in the 0.5 mL/L Etoxazole applied group. Catalase and Superoxide dismutase activities increased in the groups exposed to 0.125 mL/L and 0.25 mL/L Etoxazole with dose dependence and decreased abruptly in the group treated with 0.5 mL/L Etoxazole. Etoxazole triggered meristematic cell damages, such as epidermis cell damage, thickening of cortex cell walls, flattened cell nucleus and indistinct transmission tissue. Considering the versatile toxicity induced by Etoxazole, we announce that this chemical has the potential to cause serious damage to non-target organisms. It should be noted that the higher the dose of exposure, the more severe the level of damage. This study will be an important reminder to limit the indiscriminate use of this highly risky agrochemical.

Abstract Trifloxystrobin (TFS) is a strobilurin-type fungicide that should be investigated due to its risks to non-targeted organisms. The goal of this study was to assess the susceptibility of Allium cepa L. to TFS in a multi-pronged approach. For 72 h, 0.2 g/L, 0.4 g/L and 0.8 g/L doses of TFS were administered to A. cepa bulbs and the control group was treated with tap water. The toxic effects of TFS were tested, considering physiological, cytogenetic, biochemical and anatomical analyses. TFS delayed growth by reducing the rooting ratio, root elongation and weight increase. Following TFS treatments, mitotic index (MI) scores decreased, while the formation of micronucleus (MN) and chromosomal aberrations (CAs) ascended. CAs types induced by TFS were listed according to their frequency as fragment, vagrant chromosome, sticky chromosome, uneven distribution of chromatin, bridge, nucleus with vacuoles, reverse polarization and irregular mitosis. TFS provoked an increment in superoxide dismutase (SOD) and catalase (CAT) enzyme activities as well as an accumulation of malondialdehyde (MDA). Meristematic cells of A. cepa roots treated with TFS had various anatomical damages, including damaged epidermis, flattened cell nucleus, damaged cortex and thickness in the cortex cell wall. All damages arising from TFS treatments exhibited dose-dependency. The findings of the present study revealed the serious toxicity of TFS in a non-targeted plant. It should not be neglected to evaluate the potential hazards of TFS with different toxicity tests.

In light of limited existing knowledge, we aimed to examine the connection between disclosure facilitators and delays in the context of forensic interviews. We examined interview records from 403 children under 18 suspected of sexual abuse at a child advocacy center. The forensic interview records, family interview forms, and psychiatric examination reports in the files were retrospectively examined. Frequency, mean, and chi-square tests were implemented to analyze the data. The reasons for facilitating and delaying the disclosure of abuse were evaluated using a descriptive analysis method. Of the cases, 79.4% were girls, and 74% were in the 12–18 age group. It was determined that the presence of witnesses, evidence, and messages (16.37%) and discomfort with the suspect's/abuser's behavior (11.66%) facilitated the disclosure of abuse. On the other hand, reporting of abuse was delayed due to the suspect's/abuser's threats (18.85%) and the victim's fear, embarrassment and avoidance of her/his family (10.66%). The characteristics of abuse and disclosure reasons varied based on age groups and the intimacy level of the suspect. It has been observed that pre-adolescent children may face cognitive obstacles in disclosing abuse. [ABSTRACT FROM AUTHOR]

Turkey is known as a top producer and exporter of apricots, both fresh and dried. More than half of Turkey's apricot production is provided by Malatya province. There are many pests and diseases affecting apricots in addition to climatic factors such as frost and hail. Partial shoot drying and sudden drying in apricot orchards have increased in recent years. In this study, it was aimed to determine the chemical control possibilities of Neoscytalidium dimidiatum (Penz.) Crous &Slippers, which is one of the fungal agents that cause partial branch drying and sudden drying in apricot orchards in Malatya province by in vitro studies. For this purpose, Kale 4-C isolate of N. dimidiatum collected from Malatya in a previous study was used. Mycelial growth of N. dimidiatum Kale 4-C isolate in PDA were observed in 0 (control), 0.01, 0.03, 0.1, 1, 3, 10, 30 ve 100 μg/mL concentrations of Azoxystrobin 250g/L, Trifloxystrobin %50, Tebuconazole %25, Floupyram 200 g/L+Tebuconazole 200 g/L, Cyprodinil+Fludioxonil %37.5+25, %70 Thiophanate-Methyl and 400 g/L Phosphorous acide fungicides applications. As a result of the study, Floupyram 200g/L+Tebuconazole 200g/L ve Cyprodinil+Fludioxonil %37.5+25 fungicides effectively inhibited the mycelial growth of the fungus.

Purpose: The aim of this study was to determine the levels of pain, depression, somatization and disability, the relationship among them, and the affecting factors in older individuals who applied to the physical therapy unit. Materials and Methods: This descriptive cross-sectional study included 95 volunteer patients aged 65 years and older who applied to the physical therapy and rehabilitation department of a training-research hospital between January 2018 and January 2019. Data were collected by the Interview Form, the Charlson Comorbidity Index, the Geriatric Pain Measure, the Geriatric Depression Scale, the Somatosensory Amplification Scale, and the World Health Organization Disability Assessment Schedule-II. Results: The comorbidity index was Grade 3 in 56.8% of the participants, 36.8% of them were high probability to have depression, the overall pain score was moderate, and the total disability was moderate-mild. Pain, depression, somatization and disability scores were found to be higher in those with poor general health perception. A significant positive relationship was found between pain and depression, between amplification/ somatization score and pain and depression scores. In addition, the total score of disability was found to have a significant positive correlation with the comorbidity index, and the depression scores. Conclusion: Pain, somatization, depression, and disability in the older patients were correlated and affected by various sociodemographic and clinical characteristics. These results may guide the planning of health services offered to older adults.

Turkey is one of noteworthy countries for both fruit genetic resources and amount of fruit production in the world. Mulberry is cultivated throughout Turkey, most commonly in Central, Northeast and Southeast Anatolia. Mulberry has a great market potential thanks to its fresh consumption and usage of processed food products. In June 2019, a disease was observed causing deaths in shoots and branches on mulberry trees in Malatya province of Turkey. The causative agent of the disease was identified as Neoscytalidium novaehollandiae according to morphological characteristics and sequencing of TEF 1-α gene (Accession no. MT362602 and MT362603), ITS (Accession no. MT195554 and MT195555) and LSU (Accession no. MT195552 and MT195553). Based on the concatenated sequences of the ITS, TEF 1-α, and LSU, a phylogenetic tree was built using Bayesian analysis. Reactions of nine mulberry cultivars against the disease (‘Ulukale’, ‘Ayaş’, ‘Ichinose’, ‘Poser’, ‘Kenmochi’, ‘Arapgir’, ‘Sarı aşı’, ‘Horum’ and ‘Istanbul’) inoculated with Malatya isolate of N. novaehollandiae were evaluated under growth chamber conditions. All-mulberry cultivars artificially inoculated with N. novaehollandiae isolate exhibited severe necrosis symptoms on woody tissues of tested plants. It was confirmed that N. novaehollandiae is a fungal pathogen associated with dieback and canker on mulberry trees in Turkey for the first time. New mulberry plantations could be endangered by this emerging new disease.
