7. Antistaphylococcal Triazole-Based Molecular Hybrids: Design, Synthesis and Activity.

Abstract

Background: In the era of resistance, the design and search for new "small" molecules with a narrow spectrum of activity that target a protein or enzyme specific to a certain bacterium with high selectivity and minimal side effects remains an urgent problem of medicinal chemistry. In this regard, we developed and successfully implemented a strategy for the search for new hybrid molecules, namely, the not broadly known [2-(3-R-1H-[1,2,4]-triazol-5-yl)phenyl]amines. They can act as "building blocks" and allow for the introduction of certain structural motifs into the desired final products in order to enhance the antistaphylococcal effect. Methods: The "one-pot" synthesis of the latter is based on the conversion of substituted 4-hydrazinoquinazolines or substituted 2-aminobenzonitriles and carboxylic acid derivatives to the target products. The possible molecular mechanism of the synthesized compounds (DNA gyrase inhibitors) was investigated and discussed using molecular docking, and their further study for antistaphylococcal activity was substantiated. Results: A significant part of the obtained compounds showed high antibacterial activity against Staphylococcus aureus (MIC: 10.1–62.4 µM) and 5-bromo-2-(3-(furan-3-yl)-1H-1,2,4-triazol-5-yl)aniline and 5-fluoro-2-(3-(thiophen-3-yl)-1H-1,2,4-triazol-5-yl)aniline, with MICs of 5.2 and 6.1 µM, respectively, approaching the strength of the effect of the reference drug, "Ciprofloxacin" (MIC: 4.7 µM). The conducted SAR and ADME analyses confirm the prospects of the further structural modification of these compounds. The obtained [2-(3-R-1H-[1,2,4]-triazol-5-yl)phenyl]amines reveal significant antimicrobial activity and deserve further structural modification and detailed study as effective antistaphylococcal agents. The SAR analysis revealed that the presence of a cycloalkyl or electron-rich heterocyclic fragment in the third position of the triazole ring was essential for the antibacterial activity of the obtained compounds. At the same time, the introduction of a methyl group into the aniline moiety led to an enhancement of activity. The introduction of halogen into the aniline fragment has an ambiguous effect on the level of antistaphylococcal activity and depends on the nature of the substituent in the third position. Conclusions: Obtained [2-(3-R-1H-[1,2,4]-triazol-5-yl)phenyl]amines reveal significant antistaphylococcal activity and deserve for further detailed study as effective antibacterial agents. Abstract

This work is dedicated to the 85th anniversary of the birthday of an outstanding scientist, professor, doctor of chemical sciences, honored worker of science and technology of Ukraine Lilia Ivanovna Kas’yan. L.I. Kasyan graduated from Odesa State University (1960). In 1965, she graduated from the Department of Organic Chemistry of Dnipropetrovsk University (1965). In 1970, she defended her candidate’s thesis (PhD equivalent) «Synthesis and study of some epoxides of the bicyclo[2.2.1]heptane family», and in 1990 she defended her doctoral thesis (D.Sc. habilitation) «Epoxides of the alicyclic family. Stereochemistry, reactivity, and methods of synthesis». Since 1991, he has been a professor at the Department of Organic Chemistry at the Dnipropetrovsk State University (now Oles Honchar Dnipro National University). Her scientific works concerned the chemistry of epoxides of the alicyclic family, the study of their reactivity and the development of methods of synthesis. She headed the scientific school for the study of epoxy compounds. Based on her fundamental research many epoxy-containing compositions have been developed for chemical industry. A recognized specialist in the synthesis, stereochemistry and spectroscopy of cage-like compounds, including chemical agents with biological (pesticide and pharmacological) activity. Author of more than 500 scientific papers, including 4 books (monographs). She paid great attention to the creative education of young people, for a long time she headed the work with the student scientific society of the Department of Chemistry. More than 20 student scientific works performed under her supervision received the highest awards at regional and state contests, 17 PhD theses were defended., Стаття присвячена 85-річчю від дня народження відомої вченої, професора, доктора хімічних наук, заслуженого діяча науки і техніки України Лілії Іванівни Кас’ян. Наводяться короткі біографічні відомості про вчену. 24. SEARCH FOR NEW TYROSINE KINASE INHIBITORS AMONG 2-(3-R-1H-1,2,4-TRIAZOL-5-YL)ANILINES AS POTENTIAL ANTITUMOR AGENTS USING MOLECULAR DOCKING.

Abstract

The present work is devoted to the in silico study of 2-(3-R-1H-1,2,4-triazol-5-yl)anilines as potential inhibitors of EGFR (epidermal growth factor receptor) and RET (rearranged during transfection)-, which play a significant role in regulating the physiological cycle of non-small cell lung cancer. The well-known docking software AutoDock Vina was used for the study. The seven studied compounds and two standard drugs (vandetanib and gefitinib) were docked to the crystal structures of EGFR and RET proteins. It was found that among the newly investigated substances, 2-(3-(indolyl-2)-1H-1,2,4-triazol-5-yl)aniline (a5) has the highest affinity towards EGFR and RET with the binding energy of --9.7 and --8.7 kcal/mol, respectively. Visualization of the molecular docking results of this compound using the Discovery Studio software showed that it is characterized by similar to standard ligands location in the active sites of the enzymes, stable hydrogen bonds and π-stacking interactions, which are provided by the presence of indole and aniline fragments in the molecule. Thus, we have identified a new effective ligand that can be used as a "base" molecule for further fragment-oriented design using molecular hybridization methodology (fragment fusion, coupling or extension methods) or structural modification by introducing "pharmacophore" groups into the molecule. Summarizing the above screening results, we can say that 2-(3-R-1H-1,2,4-triazol-5- yl)anilines require further careful consideration as effective tyrosine kinase inhibitors for the search for promising anticancer agents for the treatment of non-small cell lung cancer. Abstract

Present review is critical analysis of recently published data devoted to the search of the novel anti-inflammatory agents among substituted and condensed quinazolines. It has been shown that anti-inflammatory activity is typical for various quinazoline-containing compounds including substituted quinazolin-4-ones, quinazolin-4-amines, condensed and spiro-condensed quinazolines. The features of target compounds synthesis were systematized and discussed. It was shown that synthesis of quinazoline-containing anti-inflammatory agents in most of cases based on modification of anthranilic acid derivatives, substituted 2-aminobenzonitriles or isatoic anhydride. Particular attention was paid to the discussion of 2-azaheterylanilines as original initial compounds for synthesis of condensed quinazolines with promising anti-inflammatory activity. The mechanisms of quinazolines biological activity have been presented and discussed as well. It has been shown that described compounds can effect on COX- 1, COX-2, LOX-15, NO, PGE2, IL-1b, IL-6, TNF-α and other typical for anti-inflammatory agents' biomolecular targets. For some series of compounds, the structure-biological activity relationships were discussed. The data presented in review substantiates the reasonability of the search of new highly effective and low-toxic drugs with antiinflammatory activity among quinazoline containing compounds. Abstract

Tetrazoles are found broad applications in numerous fields such as in medicine, biochemistry, pharmacology, and in industry. Considering our continued interest in new azaheterocycles based on β-ketosulfones in this work a threecomponent heterocyclization of dihydro-2H-thiopyran-3(4H)-one-1,1-dioxide, 1H-tetrazol-5-amine, and aromatic aldehydes under microwave irradiation was studied. Regardless of the reaction conditions, 5-aryl-7,8,9,10- tetrahydro-5H-tetrazolo[1,5-a]thiopyrano[3,2-d]pyrimidine 6,6-dioxides were isolated as sole reaction products in good to excellent yields. To the best of our knowledge mentioned azaheterocycles are novel and previously not reported heterocyclic ensemble. Proposed structures were confirmed by spectral methods. Considering druglikeness we can conclude that compounds match parameters for Lipinski, Ghose, Veber, Egan and Muegge rules, and also correspond to the V class of acute toxicity. In silico screening of the biological profile of new derivatives showed adequate ADMET properties along with high (60 % and more) probability levels of activity against such pathogens/diseases as Candida albicans, Alphis gossypii, Tripomastigote Chagas, Tcruzi amastigota, Tcruzi epimastigota etc. [ABSTRACT FROM AUTHOR]

Abstract

At the present time one of the main remediation technologies for such environmental pollutant as 2,4-dinitrotoluene (DNT) is advanced oxidation processes (AOPs). Since hydroxyl radical is the most common active species for AOPs, in particular for Fenton oxidation, the study modeled mechanism of interaction between DNT and hydroxyl radical at SMD(Pauling)/M06-2X/6-31+G(d,p) level. Computed results allow to suggest the most energetically favourable pathway for the process. DNT decomposition consists of sequential hydrogen abstractions and hydroxyl attachments passing through 2,4-dinitrobenzyl alcohol, 2,4-dinitrobenzaldehyde, and 2,4-dinitrobenzoic acid. Further replacement of nitro- and carboxyl groups by hydroxyl leads to 2,4-dihydroxybenzoic acid and 2,4-dinitrophenol, respectively. Reaction intermediates and products are experimentally confirmed. Mostly of reaction steps have low energy barriers, some steps are diffusion controlled. The whole process is highly exothermic., В настоящее время одной из основных технологий для рекультивации такого загрязнителя окружающей среды, как 2,4-динитротолуола (DNT), являются прогрессивные процессы окисления (AOPs). Так как гидроксильный радикал является наиболее распространенным активным агентом для AOPs, в частности для окисления Фентона, механизм взаимодействия между DNT и гидроксильным радикалом смоделирован на SMD(Pauling)/M06-2X/6-31+G(d,p) теоретическом уровне. Рассчитанные результаты позволяют предположить наиболее энергетически выгодный путь для процесса. Разложение DNT состоит из последовательных отщеплений водорода и присоединений гидроксила, проходящих через2,4-динитробензиловый спирт, 2,4-динитробензойный альдегид и 2,4-динитробензойную кислоту. Дальнейшая замена нитро- и карбоксильных групп гидроксилом приводит к 2,4-дигидроксибензойной кислоте и 2,4-динитрофенолу соответственно. Промежуточные и конечные продукты реакции подтверждены экспериментально. Большинство стадий реакции имеют низкие энергетические барьеры, некоторые стадии контролируются диффузией. Весь реакционный процесс является сильно экзотермическим., У даний час однією з основних технологій для рекультивації такого забруднювача навколишнього середовища, як 2,4-динітротолуена (DNT), є прогресивні процеси окислення (AOPs). Оскільки гідроксильний радикал є найбільш поширеним активним агентом для AOPs, зокрема для окислення Фентона, механізм взаємодії між DNT і гідроксильним радикалом змодельований на SMD(Pauling)/M06-2X/6-31+G(d,p) теоретичному рівні. Розраховані результати дозволяють припустити найбільш енергетично вигідний шлях для процесу. Розкладання DNT складається з послідовних відщеплень Гідрогену і приєднань гідроксилу, що проходять через 2,4-динітробензиловий спирт, 2,4-динітробензойний альдегід і 2,4-динітробензойну кислоту. Подальша заміна нітро- і карбоксильної груп гідроксилом приводить до 2,4-дигідроксибензойної кислоти і 2,4-динітрофенолу відповідно. Проміжні та кінцеві продукти реакції підтверджені експериментально. Більшість стадій реакції мають низькі енергетичні бар'єри, деякі стадії контролюються дифузією. Весь реакційний процес є сильно екзотермічним.

Abstract

Alkaline hydrolysis is an effective method to destroy such the pollutant as 2,4,6-trinitrotoluene (TNT) in solution and in well-mixed soil. The mechanism of hydrolytic transformation of polynegative complex, which is one of the products of early stages of TNT hydrolysis, was theoretically investigated at the SMD(Pauling)/M06-2X/6-31+G(d,p) level under alkali condition. The studied process consists of more than twenty steps and includes a six-membered cycle cleavage and sequenced [1,3]-hydrogen migration and C-C bond rupture. The highest energy barrier is observed for interaction of nitromethanide with hydroxide. The most exothermic steps are C–C bonds breaking. As a result final products such as formate, acetate, ammonium, and nitrogen are formed., Лужний гідроліз – ефективний метод знищення такого забруднювача навколишнього середовища, як 2,4,6-тринітротолуол (ТНТ) у розчині і ґрунті. Механізм гідролітичного перетворення полізарядного комплексу, який є одним із продуктів ранніх стадій гідролізу TНT, теоретично досліджений у наближенні SMD(Pauling)/M06-2X/6-31+G(d,p) за лужних умов. Вивчений процес передбачає більше ніж двадцять кроків і включає розщеплення шестичленного циклу і послідовні [1,3]-міграцію Гідрогену і розрив C-C зв’язків. Найвищий енергетичний бар’єр спостерігається за взаємодії нітрометаніду з гідроксидом. Найбільш екзотермічні кроки реакції – це розрив C–C зв’язків. У результаті реакції гідролізу утворюються такі кінцеві продукти, як форміат, ацетат, амоніак і азот., Щелочной гидролиз является эффективным методом нейтрализации такого загрязнителя окружающей среды, как 2,4,6-тринитротолуол (ТНТ) в растворе и грунте. Abstract

Adsorption of hexahydro-1,3,5-trinitro-1,3,5-triazine (RDX) and octahydro-1,3,5,7-tetranitro-1,3,5,7-tetrazocine (HMX) on (001) surface of α-quartzwas studiedat the M05/tzvp level using cluster approximation. Hydrogen bonds between nitramines and silica surface were analyzed by atoms in molecules (AIM) method. Electron attachment causes significant change in geometry of adsorbed complexes. Redox properties of adsorbed RDX and HMX were compared with those of gas-phase and hydrated species by calculation of the ionization potential, electron affinity, oxidation and reduction Gibbs free energies, oxidation and reduction potentials. Calculations show that adsorbed RDX and HMX have lower ability to undergo redox transformations than hydrated ones., Вивчено адсорбцію гексагідро-1,3,5-тринітро-1,3,5-триазина (RDX) і октагідроциклоокта-1,3,5,7-тетранітро-1,3,5,7-тетразоцина (HMX) на (001) поверхні альфа-кварцу методом М05/tzvp з використанням кластерного наближення. Методом «атоми в молекулі» (AIM) проаналізовано водневі зв'язки між нітроамінами і поверхнею кремнезему. Приєднання електрону викликає значні зміни в геометрії адсорбованих комплексів. Проведено порівняльний аналіз окисно-відновних властивостей RDX і HMX у газовій фазі, у водному розчині та у адсорбованому стані. Розраховані потенціали іонізації, спорідненість до електрона, вільні енергії Гіббса процесів окислення і відновлення, потенціали окислення і відновлення. Розрахунки показують, що адсорбовані RDX і HMX мають більш низьку здатність зазнавати окислювально-відновних перетворень, ніж гідратовані., Изучена адсорбция гексагидро-1,3,5-тринитро-1,3,5-триазина (RDX) и октагидроциклоокта-1,3,5,7-тетра-нитро-1,3,5,7-тетразоцина (HMX) на (001) поверхности альфа-кварца методом М05/tzvp с использованием кластерного приближения. Рассчитаны потенциалы ионизации, сродство к электрону, свободные энергии Гиббса процессов окисления и восстановления, потенциалы окисления и восстановления. Расчеты показывают, что адсорбированные RDX и HMX имеют более низкую способность подвергаться окислительно-восстановительным превращениям, чем гидратированные.

Abstract

The stereochemical aspects of epoxidation of norbornene and its 7-syn-substituted derivatives by performic acid were investigated. Geometry and thermodynamic parameters of transition states and prereactive complexes were computed at the UBHandHLYP/6-31G(d) level of theory. It is shown that the transition states have a pronounced biradical character and a nearly coplanar orientation of the C=C bond and the molecule of performic acid. Transition state analysis revealed that, in the case of the syn-7-hydroxy derivative, the preference for the exo-approach of the oxidant can be explained by the stabilization of transition state with hydrogen bonding. In contrast, a chlorine atom or a methyl group at the 7-syn position facilitated the formation of endo-epoxides due to steric repulsion between the substituent and the oxidant., Вивчено стереохімічні аспекти епоксидування норбонена та його 7-syn-заміщених похідних пероксимурашиною кислотою. Геометричні та термодинамічні параметри перехідних станів та передреакціцних комплексів розраховані у наближенні UBHandHLYP/6-31G(d). Показано, що перехідні стані мають бірадикальний характер та близьку до копланарності орієнтацію зв'язку C=C та молекули пероксикислоти. Аналіз перехідних станів свидчіть про те, що, у випадку syn-7-гидрокси похідного, переважна екзо-атака окиснювача пояснюється стабілізацією перехідного стану за рахунок водневих зв'язків. Наявність в 7-syn положенні атома хлору або метильної групи, навпаки, призводить до утворення ендо-епоксиду внаслідок стеричного відштовхування між замісником та окиснювачем., Изучены стереохимические аспекты эпоксидирования норбонена и его 7-syn-замещенных производных пероксимуравьиной кислотой. Геометрические и термодинамические параметры переходных состояний и предреакционных комплексов рассчитаны в приближении UBHandHLYP/6-31G(d). Показано, что переходные состояния имеют бирадикальный характер и близкую к копланарности ориентацию связи C=C и молекулы пероксикислоты. Анализ переходных состояний свидетельствует о том, что в случае syn-7-гидрокси производного, преимущественная экзо-атака окислителя объясняется стабилизацией переходного состояния за счет водородных связей. Наличие в 7-syn положении атома хлора или метильной группы, напротив, приводит к образованию эндо-эпоксида вследствие стерического отталкивания между заместителем и окислителем.

Abstract

Theoretical investigations of the conformational properties and 1H NMR chemical shifts for N-methyl-4-tolyl-1-(4-bromonaphthyl)amine and N-phenyl-1-(4-bromonaphthyl)amine are reported. The calculations were performed at the DFT level (PBE1PBE functional) using magnetically consistent 6-31G## and STO##-3Gmag basis sets. Conformational properties of the amines were studied using potential energy surface scanning. Chemical shifts were calculated using the GIAO and CSGT methods and averaged in proportion to the population of the corresponding conformations. Solvent effects (CDCl3) were accounted via PCM method. The obtained results allowed to assign the 1H NMR signals for the naphthalene moiety, which could not be done based on the experimental data alone., Представлены результаты теоретического исследования конформационных свойств и химических сдвигов ядер в спектрах 1Н ЯМР N-метил-4-толил-1-(4-бромнафтил)амина и N-фенил-1-(4-бромнафтил)амина. Расчеты проведены в приближении теории функционала плотности (функционал PBE1PBE) с использованием адаптированных для расчета магнитных свойств наборов базисных функций 6-31G## и STO##-3Gmag. Конформационные свойства аминов рассчитаны путем сканирования поверхности потенциальной энергии. Величины химических сдвигов рассчитаны методами GIAO и CSGT как средние величины, пропорционально заселенности соответствующих конформеров. Влияние растворителя, используемого при записи ЯМР спектров (CDCl3), учитывалось методом PCM. Полученные результаты позволили выполнить отнесение сигналов в спектрах к соответствующим ядрам, в частности, для нафталинового фрагмента, что не представлялось возможным сделать на основании только экспериментальных спектральных данных., Наведені результати теоретичного дослідження конформаційних властивостей та хімічних зсувів ядер в спектрах 1Н ЯМР N-метил-4-толіл-1-(4-бромонафтил)-аміну та N-феніл-1-(4-бромонафтил)-аміну. Розрахунки проведені в наближенні теорії функціоналу густини (функціонал PBE1PBE) з використанням адаптованих для розрахунку магнітних властивостей наборів базисних функцій 6-31G## и STO##-3Gmag. Конформаційні властивості амінів розраховані шляхом сканування поверхні потенційної енергії. Значення хімічних зсувів розраховані методами GIAO и CSGT як середні значення, пропорційно заселеності відповідних конформерів. Вплив розчинника який використовувався під час запису спектрів ЯМР (CDCl3) враховувався методом PCM. Отримані результати дозволили виконати віднесення сигналів в спектрах до відповідних ядер, зокрема, для нафталінового фрагменту, що було неможливо зробити лише на основі експериментальних спектральних даних.

Abstract

At the present time one of the main remediation technologies for such environmental pollutant as 2,4-dinitrotoluene (DNT) is advanced oxidation processes (AOPs). Since hydroxyl radical is the most common active species for AOPs, in particular for Fenton oxidation, the study modeled mechanism of interaction between DNT and hydroxyl radical at SMD(Pauling)/M06-2X/6-31+G(d,p) level. Computed results allow to suggest the most energetically favourable pathway for the process. DNT decomposition consists of sequential hydrogen abstractions and hydroxyl attachments passing through 2,4-dinitrobenzyl alcohol, 2,4-dinitrobenzaldehyde, and 2,4-dinitrobenzoic acid. Further replacement of nitro- and carboxyl groups by hydroxyl leads to 2,4-dihydroxybenzoic acid and 2,4-dinitrophenol, respectively. Reaction intermediates and products are experimentally confirmed. Mostly of reaction steps have low energy barriers, some steps are diffusion controlled. The whole process is highly exothermic., У даний час однією з основних технологій для рекультивації такого забруднювача навколишнього середовища, як 2,4-динітротолуена (DNT), є прогресивні процеси окислення (AOPs). Оскільки гідроксильний радикал є найбільш поширеним активним агентом для AOPs, зокрема для окислення Фентона, механізм взаємодії між DNT і гідроксильним радикалом змодельований на SMD(Pauling)/M06-2X/6-31+G(d,p) теоретичному рівні. Розраховані результати дозволяють припустити найбільш енергетично вигідний шлях для процесу. Розкладання DNT складається з послідовних відщеплень Гідрогену і приєднань гідроксилу, що проходять через 2,4-динітробензиловий спирт, 2,4-динітробензойний альдегід і 2,4-динітробензойну кислоту. Подальша заміна нітро- і карбоксильної груп гідроксилом приводить до 2,4-дигідроксибензойної кислоти і 2,4-динітрофенолу відповідно. Проміжні та кінцеві продукти реакції підтверджені експериментально. Більшість стадій реакції мають низькі енергетичні бар'єри, деякі стадії контролюються дифузією.
