Abstract

Oral immunotherapy (OIT) is an effective approach to controlling food allergy. Although the detailed molecular and cellular mechanisms of OIT are unknown currently, they must be understood to advance the treatment of allergic diseases in general. To elucidate the mechanisms of OIT, especially during the immunological transition from desensitization to allergy regulation, we generated a clinical OIT murine model and used it to examine immunological events of OIT. We found that in mice that completed OIT successfully, desensitized mast cells (MCs) showed functionally beneficial alterations, such as increased induction of regulatory cytokines and enhanced expansion of regulatory T cells. Importantly, these regulatory-T-cell-mediated inhibitions of allergic responses were dramatically decreased in mice lacking OIT-induced desensitized MC. Collectively, these findings show that the desensitization process modulates the activation of MCs, leading directly to enhanced induction of regulatory-T-cell expansion and promotion of clinical allergic unresponsiveness. Our results suggest that efficiently inducing regulatory MCs is a novel strategy for the treatment of allergic disease.

Abstract

Critical to the function of mast cells in immune responses including allergy is their production of lipid mediators, among which only omega-6 ([omega]-6) arachidonate-derived eicosanoids have been well characterized. Here, by employing comprehensive lipidomics, we identify omega-3 ([omega]-3) fatty acid epoxides as new mast cell-derived lipid mediators and show that they are produced by PAF-AH2, an oxidized-phospholipid-selective phospholipase A2. Genetic or pharmacological deletion of PAF-AH2 reduced the steady-state production of [omega]-3 epoxides, leading to attenuated mast cell activation and anaphylaxis following Fc[var epsilon]RI cross-linking. Mechanistically, the [omega]-3 epoxides promote IgE-mediated activation of mast cells by downregulating Srcin1, a Src-inhibitory protein that counteracts Fc[var epsilon]RI signaling, through a pathway involving PPARg. 14. Immunopharmacological studies on human mast cells and basophils

Abstract

The aims of this project were first to investigate the extent to which drugs used for the treatment of allergic diseases may have an effect on the early phase of IgE-dependent mast cell activation. Second, as IL-3, some eosinophil granule proteins and histamine-releasing factors are considered to initiate basophil activation in the late phase of allergic reaction, the effect of these agents on histamine release from human mast cells was investigated. Third, the production of cytokines by human mast cells was investigated. High concentrations of sodium cromoglycate, nedocromil sodium and antihistamines were required to significantly inhibit IgE-dependent release of histamine and PGD₂ from mast cells. There was some heterogeneity observed for mast cells from different human tissues in the degree of pharmacological control by these agents. In contrast, nanomolar concentrations of β-adrenoceptor agonists consistently inhibited histamine and PGD_2 release. IL-3, eosinophil granule proteins and peripheral blood mononuclear cell-derived histamine-releasing factors neither induced nor primed IgE-dependent histamine release from human mast cells. As a part of these studies, the mechanisms of activation of basophils by IL-3 were investigated. Histamine release by IL-3 required both extracellular Ca^2+ and the presence of membrane IgE, but IL-3 priming did not. The activation of stem cell factor-primed mast cells via Fc_6RI, induced expression of IL-4 and IL-5 mRNA at time points ranging from 2 to 24 h and 2 to 72 h, respectively. In conclusion, (1) antiallergic drugs are weak inhibitors of histamine and PGD_2 release from mast cells in vitro; (2) several basophil priming factors are without effect on the functional activity of mast cells; (3) human mast cells may contribute to the chronicity of tissue inflammation by the production of cytokines.

Abstract

Carcinoembryonic antigen-related cell adhesion molecule 1 (CEACAM1) is expressed in a number of tumor cell types. The immunoreceptor tyrosine-based inhibitory motif (ITIM)-containing isoforms of this molecule which possess a long cytoplasmic tail (CEACAM1-L) generally play inhibitory roles in cell function by interacting with Src homology 2 domain-containing tyrosine phosphatase (SHP)-1 and/or SHP-2. Src family kinases (SFKs) are also known to bind to and phosphorylate CEACAM1-L isoforms. Here, we report that CEACAM1 was uniquely expressed at high levels in both human neoplastic mast cells (mastocytosis) and medullary thyroid carcinoma cell (MTC) lines, when compared with their expression in nonneoplastic mast cells or nonneoplastic C cells. This expression was mainly derived from CEACAM1-L isoforms based upon assessment of CEACAM1 mRNA expression. CEACAM1 knockdown upregulated cell growth of HMC1.2 cells harboring KIT mutations detected in clinical mastocytosis, whereas downregulated the growth of TT cells harboring RET mutations detected in clinical MTCs. Immunoblotting, ELISA and immunoprecipitaion analysis showed that activated SHP-1 is preferentially associated with CEACAM1 in HMC1.2 cells harboring KIT mutations, whereas Src family kinases (SFKs) are preferentially associated with CEACAM1 in TT cells harboring RET mutations. These studies suggest that the dominantly interacting proteins SHP1 or SFK determine whether CEACAM1-L displays a positive or negative role in tumor cells.
