Hypoxic preconditioning (HP) is a stem cell preconditioning modality designed to augment the therapeutic effects of mesenchymal stem cells (MSCs). Although autophagy is expected to play a role in HP, very little is known regarding the relationship between HP and autophagy.The adipose-derived stem cell (ASC)-secretome obtained under normoxia (NCM) and ASC-secretome obtained under HP (HCM) were obtained by culturing ASCs for 24 h under normoxic (21% partial pressure of OHP appears to induce ASCs to release a secretome with enhanced anti-apoptotic effects by promoting the autophagic process of ASCs.

Apoptotic cells are rapidly engulfed and removed by phagocytes after displaying cell surface eat-me signals. Among many phospholipids, only phosphatidylserine (PS) is known to act as an eat-me signal on apoptotic cells. Using unbiased proteomics, we identified externalized phosphatidylinositides (PIPs) as apoptotic eat-me signals recognized by CD14+phagocytes. Exofacial PIPs on the surfaces of early and late-apoptotic cells were observed in patches and blebs using anti-PI(3,4,5)P3antibody, AKT- and PLCδ PH-domains, and CD14 protein. Phagocytosis of apoptotic cells was blocked either by masking exofacial PIPs or by CD14 knockout in phagocytes. We further confirmed that exofacial PIP+thymocytes increased dramatically after in vivo irradiation and that exofacial PIP+cells represented more significant populations in tissues ofCd14−/−than WT mice, especially after induction of apoptosis. Our findings reveal exofacial PIPs to be previously unknown cell death signals recognized by CD14+phagocytes.

Cell reversal is observed when a current load is applied to the polymer electrolyte membrane fuel cell under fuel starvation conditions. Cell reversal causes severe corrosion (or oxidation) of the carbon support in the anode, which leads to a decrease in overall fuel cell performance. To suppress the corrosion reaction of carbon under cell reversal conditions and to increase the durability of fuel cells, studies on anode additives are being conducted. However, studies on the effect of additives on catalysts with different platinum contents have not been conducted. In this study, 20 wt%, 40 wt%, 60 wt% commercial Pt/C catalyst was applied to the anode, and 50 cycles of cell reversal were performed. Furthermore, the performance change with and without IrO2 as an additive was observed and its effect was assessed. Changes in the morphologies of the electrodes before and after cell reversal tests were also observed using a transmission electron microscope and a scanning electron microscope. The higher the platinum content of the catalyst, the more resistant to cell reversal. In addition, the addition of IrO2 to the anode effectively prevents performance degradation due to cell reversal.

A substantial amount of research effort has been directed toward the development of Pt-based catalysts with higher performance and durability than conventional polycrystalline Pt nanoparticles to achieve high-power and innovative energy conversion systems. Currently, attention has been paid toward expanding the electrochemically active surface area (ECSA) of catalysts and increase their intrinsic activity in the oxygen reduction reaction (ORR). However, despite innumerable efforts having been carried out to explore this possibility, most of these achievements have focused on the rotating disk electrode (RDE) in half-cells, and relatively few results have been adaptable to membrane electrode assemblies (MEAs) in full-cells, which is the actual operating condition of fuel cells. Thus, it is uncertain whether these advanced catalysts can be used as a substitute in practical fuel cell applications, and an improvement in the catalytic performance in real-life fuel cells is still necessary. Therefore, from a more practical and industrial point of view, the goal of this review is to compare the ORR catalyst performance and durability in half- and full-cells, providing a differentiated approach to the durability concerns in half- and full-cells, and share new perspectives for strategic designs used to induce additional performance in full-cell devices.

BACKGROUND: Molecular characterization in pediatric papillary thyroid cancer (PTC), distinct from adult PTC, is important for developing molecularly targeted therapies for progressive radioiodine-refractory ((131)I-refractory) PTC. METHODS: PTC samples from 106 pediatric patients (age range: 4.3–19.8 years; n = 84 girls, n = 22 boys) who were admitted to SNUH (January 1983–March 2020) were available for genomic profiling. Previous transcriptomic data from 125 adult PTC samples were used for comparison. RESULTS: We identified genetic drivers in 80 tumors: 31 with fusion oncogenes (RET in 21 patients, ALK in 6 patients, and NTRK1/3 in 4 patients); 47 with point mutations (BRAF(V600E) in 41 patients, TERT(C228T) in 2 patients [1 of whom had a coexisting BRAF(V600E)], and DICER1 variants in 5 patients); and 2 with amplifications. Background: Di-2-ethylhexyl phthalate (DEHP) is known to disrupt thyroid hormonal status. However, the underlying molecular mechanism of this disruption is unclear. Therefore, we investigated the direct effects of DEHP on the thyroid gland.Methods: DEHP (vehicle, 50 mg/kg, and 500 mg/kg) was administered to Sprague-Dawley rats for 2 weeks. The expression of the thyroid hormone synthesis pathway in rat thyroid tissues was analyzed through RNA sequencing analysis, quantitative reverse transcription-polymerase chain reaction (RT-PCR), and immunohistochemical (IHC) staining. DEHP was treated to FRTL-5 rat thyroid cells, and an RT-PCR analysis was performed. A reporter gene assay containing the promoter of thyroid stimulating hormone receptor (TSHR) in Nthy-ori 3-1 human thyroid cells was constructed, and luciferase activity was determined.Results: After DEHP treatment, the free thyroxine (T4) and total T4 levels in rats significantly decreased. RNA sequencing analysis of rat thyroid tissues showed little difference between vehicle and DEHP groups. In the RT-PCR analysis, Tshr expression was significantly lower in both DEHP groups (50 and 500 mg/kg) compared to that in the vehicle group, and IHC staining showed that TSHR expression in the 50 mg/kg DEHP group significantly decreased. DEHP treatment to FRTL-5 cells significantly down-regulated Tshr expression. DEHP treatment also reduced luciferase activity in a reporter gene assay for TSHR.Conclusion: Although overall genetic changes in the thyroid hormone synthesis pathway are not clear, DEHP exposure could significantly down-regulate Tshr expression in thyroid glands. Down-regulation of Tshr gene appears to be one of potential mechanisms of thyroid disruption by DEHP exposure.

An ideal polymer electrolyte membrane fuel cell (PEMFC) is one that continuously generates electricity as long as hydrogen and oxygen (or air) are supplied to its anode and cathode, respectively. However, internal and/or external conditions could bring about the degradation of its electrodes, which are composed of nanoparticle catalysts. Particularly, when the hydrogen supply to the anode is disrupted, a reverse voltage is generated. This phenomenon, which seriously degrades the anode catalyst, is referred to as cell reversal. To prevent its occurrence, iridium oxide (IrO2) particles were added to the anode in the membrane-electrode assembly of the PEMFC single-cells. After 100 cell reversal cycles, the single-cell voltage profiles of the anode with Pt/C only and the anodes with Pt/C and various IrO2 contents were obtained. Additionally, the cell reversal-induced degradation phenomenon was also confirmed electrochemically and physically, and the use of anodes with various IrO2 contents was also discussed.

As the importance of additional capacity beyond the theoretical limitation of lithium-ion batteries has been recognized, extensive research into effectively utilizing the extra lithium accommodation is being conducted. One of the most effective strategies to increase capacity is the use of catalytic materials well-known to promote electrochemical reactivity. Herein, we adopt platinum (Pt) metal as an electrode material to take advantage of dual charge storage properties: (i) the Li–Pt alloying reaction and (ii) additional Li storage reaction induced by catalytic properties. The prepared Pt electrode yields an initial capacity of ∼863 mA h g−1 during the 1st cycle and a reversible capacity of ∼600 mA h g−1, which is much larger compared to the previously reported capacity of 137 mA h g−1. Additionally, the complex Li-ion storage in Pt metal involving alloying reaction and catalytic effect induced charge storage reactions shows high reversibility, realizing a stable electrochemical performance upon cycling. This study holds great promise for understanding the origin of additional capacities from the catalytic effect of alloying type anodes and could be a trendsetter for designing high-capacity anode materials for next-generation lithium rechargeable batteries.

A variety of methods have been developed for accurate and systematic evaluation of chemical genotoxicity. Ceric ammonium nitrate (CAN) and 1,3-propane sultone (1,3-PS) have been extensively applied in industrial fields. Although 1,3-PS, but not CAN, has been reported as a potent carcinogen, systematic assessment of the genotoxic properties of these chemicals has not been conducted. The purpose of this study was to establish a decision tree for evaluating genotoxicity based on the good laboratory practices (GLP) system using 1,3-PS and CAN as test chemicals. In vitro studies were performed including the bacterial reverse mutation assay, chromosomal aberration assay, and micronucleus assay. We conducted in vivo studies using a combined micronucleus and alkaline comet (MN-CMT) assay and the Pig-a gene mutation assay, which is a promising method for detecting gene mutations in vivo. CAN showed negative responses in all in vitro genotoxicity assays and the in vivo combined MN-CMT assay. Meanwhile, 1,3-PS had positive results in all in vitro and in vivo genotoxicity assays. In this study, we confirmed the genotoxicity of 1,3-PS and CAN using both in vitro and in vivo assays. We propose a decision tree for evaluating chemical-induced genotoxicity.

To explore extracellular vesicle microRNAs (EV miRNAs) and their target mRNAs in relation to diabetic kidney disease (DKD), we performed paired plasma and urinary EV small RNA sequencing (n = 18) in patients with type 2 diabetes and DKD (n = 5) and healthy subjects (n = 4) and metabolic network analyses using our own miRNA and public mRNA datasets. We found 13 common differentially expressed EV miRNAs in both fluids and 17 target mRNAs, including RRM2, NT5E, and UGDH. Because succinate dehydrogenase B was suggested to interact with proteins encoded by these three genes, we measured urinary succinate and adenosine in a validation study (n = 194). These two urinary metabolite concentrations were associated with DKD progression. In addition, renal expressions of NT5E and UGDH proteins were increased in db/db mice with DKD compared to control mice. In conclusion, we profiled DKD-related EV miRNAs in plasma and urine samples and found their relevant target pathways.

Isocitrate dehydrogenase 2 (IDH2) is an NADP+-dependent enzyme that catalyzes the oxidative decarboxylation of isocitrate to α-ketoglutarate in the mitochondrial matrix, and is critical for the production of NADPH to limit the accumulation of mitochondrial reactive oxygen species (ROS). Here, we showed that high-fat diet (HFD) feeding resulted in accelerated weight gain in the IDH2KO mice due to a reduction in whole-body energy expenditure. Moreover, the levels of NADP+, NADPH, NAD+, and NADH were significantly decreased in the brown adipose tissue (BAT) of the HFD-fed IDH2KO animals, accompanied by decreased mitochondrial function and reduced expression of key genes involved in mitochondrial biogenesis, energy expenditure, and ROS resolution. Interestingly, these changes were partially reversed when the antioxidant butylated hydroxyanisole was added to the HFD. These observations reveal a crucial role for IDH2 in limiting ROS-dependent mitochondrial damage when BAT metabolism is normally enhanced to limit weight gain in response to dietary caloric overload., Obesity: Possible protective enzyme identified An enzyme that limits the build-up of reactive oxygen species (ROS) in fat cells protects mice against metabolic stress during a high-fat diet. Calorie overload leads to high levels of damaging ROS in the mitochondria of brown fat cells. This can disrupt processes that regulate energy expenditure and glucose metabolism. A team led by Seung-Soon Im at Keimyung University, Daegu, South Korea, and Timothy F. Osborne at Johns Hopkins University, St. Petersburg, USA, examined the role of an enzyme called isocitrate dehydrogenase 2 (IDH2), which is known to regulate the build-up of mitochondrial ROS. In mice fed a high-fat diet, those without IDH2 experienced accelerated weight gain, triggered by increased ROS levels and decreased mitochondrial function. A dose of an antioxidant in the diet reduced this effect, suggesting that patients with obesity may benefit from antioxidant supplements.

BACKGROUND: Recently, the exclusive use of mesenchymal stem cell (MSC)-secreted molecules, named as the secretome, have been evaluated for overcoming the limitations of cell-based therapy while maintaining its advantages. AIM: To improve cell-free therapy by adding disease-specificity through stimulation of MSCs using disease-causing materials. METHODS: We collected the secretory materials (named as inducers) released from AML12 hepatocytes that had been pretreated with thioacetamide (TAA) and generated the TAA-induced secretome (TAA-isecretome) after stimulating adipose-derived stem cells with the inducers. The TAA-isecretome was intravenously administered to mice with TAA-induced hepatic failure and those with partial hepatectomy. RESULTS: TAA-isecretome infusion showed higher therapeutic potential in terms of (1) restoring disorganized hepatic tissue to normal tissue; (2) inhibiting proinflammatory cytokines (interleukin-6 and tumor necrosis factor-α); and (3) reducing abnormally elevated liver enzymes (aspartate aminotransferase and alanine aminotransferase) compared to the naïve secretome infusion in mice with TAA-induced hepatic failure. However, the TAA-isecretome showed inferior therapeutic potential for restoring hepatic function in partially hepatectomized mice. Proteomic analysis of TAA-isecretome identified that antioxidant processes were the most predominant enriched biological networks of the proteins exclusively identified in the TAA-isecretome. In addition, peroxiredoxin-1, a potent antioxidant protein, was found to be one of representative components of TAA-isecretome and played a central role in the protection of TAA-induced hepatic injury. CONCLUSION: Appropriate stimulation of adipose-derived stem cells with TAA led to the production of a secretome enriched with proteins, especially peroxiredoxin-1, with higher antioxidant activity. Our results suggest that appropriate stimulation of MSCs with pathogenic agents can lead to the production of a secretome specialized for protecting against the pathogen. This approach is expected to open a new way of developing various specific therapeutics based on the high plasticity and responsiveness of MSCs.

Anion conducting polymers (ACPs) are essential materials for alkaline electrochemical energy technology such as anion-exchange membrane fuel cells (AEMFCs) and water electrolysers (AEMWEs). The aforementioned polymers are promising alternatives for proton exchange membrane-based systems due to the possibility of using platinum group metal-free electrocatalysts. However, there are still no reliable ACPs possessing the desired performance and stability, which is a major challenge for developing alkaline energy systems. Herein, we highlight an anion-exchange membrane and ionomer based on quaternised poly-carbazole (QPC-TMA) with a rigid ether-free and curved backbone structure comprised of carbazole monomers. The developed ACP exhibits excellent ionic conductivity, as well as chemical and mechanical stability. Moreover, the AEMFC using QPC-TMA shows excellent performance (1.61 W cm−2) compared with the other best-performing AEMFCs. In addition, the AEMWE using QPC-TMA demonstrates outstanding stability and state-of-the-art performance (3.5 A cm−2 at 1.9 V), which is the first report of an AEMWE that outperforms the best-performing proton-exchange membrane water electrolysers.

BACKGROUND Recently, the exclusive use of mesenchymal stem cell (MSC)-secreted molecules, called secretome, rather than cells, has been evaluated for overcoming the limitations of cell-based therapy, while maintaining its advantages. However, the use of naïve secretome may not fully satisfy the specificity of each disease. Therefore, it appears to be more advantageous to use the functionally reinforced secretome through a series of processes involving physico-chemical adjustments or genetic manipulation rather than to the use naïve secretome. AIM To determine the therapeutic potential of the secretome released from miR-122-transfected adipose-derived stromal cells (ASCs). METHODS We collected secretory materials released from ASCs that had been transfected with antifibrotic miR-122 (MCM) and compared their antifibrotic effects with those of the naïve secretome (CM). MCM and CM were intravenously administered to the mouse model of thioacetamide-induced liver fibrosis, and their therapeutic potentials were compared. RESULTS MCM infusion provided higher therapeutic potential in terms of: (A) Reducing collagen content in the liver; (B) Inhibiting proinflammatory cytokines; and (C) Reducing abnormally elevated liver enzymes than the infusion of the naïve secretome. The proteomic analysis of MCM also indicated that the contents of antifibrotic proteins were significantly elevated compared to those in the naïve secretome. CONCLUSION We could, thus, conclude that the secretome released from miR-122-transfected ASCs has higher antifibrotic and anti-inflammatory properties than the naïve secretome. Because miR-122 transfection into ASCs provides a specific way of potentiating the antifibrotic properties of ASC secretome, it could be considered as an enhanced method for reinforcing secretome effectiveness.

BACKGROUND The use of methyl-tertiary butyl ether (MTBE) to dissolve gallstones has been limited due to concerns over its toxicity and the widespread recognition of the safety of laparoscopic cholecystectomy. The adverse effects of MTBE are largely attributed to its low boiling point, resulting in a tendency to evaporate. Therefore, if there is a material with a higher boiling point and similar or higher dissolubility than MTBE, it is expected to be an attractive alternative to MTBE. AIM To determine whether tert-amyl ethyl ether (TAEE), an MTBE analogue with a relatively higher boiling point (102 °C), could be used as an alternative to MTBE in terms of gallstone dissolubility and toxicity. METHODS The in vitro dissolubility of MTBE and TAEE was determined by measuring the dry weights of human gallstones at predetermined time intervals after placing them in glass containers with either of the two solvents. The in vivo dissolubility was determined by comparing the weights of solvent-treated gallstones and control (dimethyl sulfoxide)-treated gallstones, after the direct infusion of each solvent into the gallbladder in both hamster models with cholesterol and pigmented gallstones. RESULTS The in vitro results demonstrated a 24 h TAEE-dissolubility of 76.7%, 56.5% and 38.75% for cholesterol, mixed, and pigmented gallstones, respectively, which represented a 1.2-, 1.4-, and 1.3-fold increase in dissolubility compared to that of MTBE. In the in vitro experiment, the 24 h-dissolubility of TAEE was 71.7% and 63.0% for cholesterol and pigmented gallstones, respectively, which represented a 1.4- and 1.9-fold increase in dissolubility compared to that of MTBE. In addition, the results of the cell viability assay and western blot analysis indicated that TAEE had a lower toxicity towards gallbladder epithelial cells than MTBE. CONCLUSION We demonstrated that TAEE has higher gallstone dissolubility properties and safety than those of MTBE. As such, TAEE could present an attractive alternative to MTBE if our findings regarding its efficacy and safety can be consistently reproduced in further subclinical and clinical studies.

Background Clinically, liver fibrosis and cholestasis are two major disease entities, ultimately leading to hepatic failure. Although autophagy plays a substantial role in the pathogenesis of these diseases, its precise mechanism has not been determined yet. Materials and methods Mouse models of liver fibrosis or cholestasis were obtained after the serial administration of thioacetamide (TAA) or surgical bile duct ligation (BDL), respectively. Then, after obtaining liver specimens at specific time points, we compared the expression of makers related to apoptosis (cleaved caspases), inflammation (CD68), necrosis (high-mobility group box 1), phospho-c-Jun N-terminal kinase (p-JNK), and autophagy (microtubule-associated protein light chain 3B and p62) in the fibrotic or cholestatic mouse livers, by polymerase chain reaction, Western blot analysis, immunohistochemistry, and immunofluorescence. Results Although cholestatic livers exhibited the tendency of progressively increasing the expression of most apoptosis-related markers (cleaved caspases), it was not prominent when it was compared with the tendency found in the livers of TAA-treated mice. Contrastingly, the necrosis-related factor (high-mobility group box 1) was significantly increased in the livers of BDL mice over time, reaching their peak values on day 7 after BDL. In addition, the inflammation-related factor (CD68) was highly expressed in BDL mice compared with TAA-treated mice over time. Autophagy marker studies indicated that autophagy was upregulated in fibrotic livers, whereas it was downregulated in cholestatic livers. We also observed mild to moderate activation of p-JNK in the livers of TAA-treated mice, whereas significantly higher p-JNK activation was detected in the livers of BDL mice. Conclusions Unlike TAA-treated mice, BDL mice exhibited higher expression of the markers related with inflammation and necrosis, especially including p-JNK, while maintaining low levels of autophagic process. Therefore, obstructive cholestasis is characterized by higher p-JNK activation, which could be related with marked necrotic cell death resulting from extensive inflammation and little chance of compensatory autophagy.

Formic acid is a chemical with a simple molecular structure containing hydrogen. This liquid at room temperature is easy to handle and has a low toxicity, and is thus in the spotlight as a fuel. In particular, formic acid is an excellent fuel candidate because it can be operated at low temperatures when applied as a fuel in fuel cells with a high theoretical open-circuit voltage (1.48 V). However, it has a drawback in that the electrode catalyst is deactivated due to the generation of CO intermediates when formic acid is oxidized during cell operation. Therefore, to prevent this, an irreversibly adsorbed Bi on Pt catalyst is applied to a direct formic acid fuel cell (DFAFC) anode because it is easy to synthesize and economical. Physical analyses such as transmission electron microscopy (TEM), X-ray diffraction (XRD), and thermogravimetric analysis (TGA) were conducted, and electrochemical evaluations were performed through half-cell and single-cell level tests. The results revealed that the formic acid oxidation reaction activity of the Bi-modified Pt/C was 13 times higher than that of the conventional catalyst at 0.58 V. Further, a DFAFC stack was fabricated using the Bi-modified Pt/C, which yielded a power of 300 W. These results suggest that a simple synthesis method can be applied to fabricating industrially available DFAFC stacks.

Enhanced mass transport in polymer electrolyte membrane fuel cells (PEMFCs) is required for achieving high performance because concentration losses dominate cell performance. In particular, the flow field is crucial for mass transport. Recently, metal foam has been proposed as an alternative flow field owing to its three-dimensional pores, high porosity, and enhanced electrical conductivity. Here, we inspect the microstructure of various copper foams and investigate its effect as a flow field on PEMFCs. The PEMFCs with the optimized foam flow field deliver the highest performance reported to date. A large contact area and small ribs of the optimized foam flow field are advantageous for mass transfer and ohmic resistance. In addition, the internally generated pressure increases the partial pressure of the reactant, which leads to increased performance. This foam flow field has a significant potential for achieving high cell performance by enhancing the electrochemical reaction of the catalyst.

In this study, an iridium oxide (IrO2) inverse-opal membrane-electrode assembly (inverse-opal MEA) was fabricated via the decal-transfer method for an anode in polymer-electrolyte membrane water electrolysis (PEMWE) to decrease the loading of the noble catalyst. Electrodeposition parameters including current and total number of cycles were investigated to achieve the IrO2 inverse-opal electrode. The inverse-opal MEA with ultra-low loading exhibited outstanding performance that exceeded or was comparable to that obtained in other PEMWE studies. Additionally, it exhibited higher performance and lower ohmic and charge-transfer resistance when compared with that of commercial IrO2. Furthermore, the performance corresponded to the highest mass activity reported to date since the loading in the inverse-opal MEA was ultra-low. This was because the inverse-opal structure improved electron transfer owing to the interconnected pores and increased the surface area due to high porosity, thereby leading to the enhanced utilization of the catalyst.
