Throughout history, Turkish coffee has been the most widely consumed type of coffee in Turkey. Therefore, the goal of this study was to determine the amount of ochratoxin A (OTA) present in Turkish coffee and to analyze any potential health hazards. A total of 41 Turkish coffees were collected and analyzed for OTA activity using a competitive enzyme immunoassay (ELISA). Furthermore, dietary exposure and health risk assessments for the Turkish population were calculated based on analytical results and coffee consumption data from the Turkish Nutrition and Health Survey 2019 (TNHS-2019) in three age categories (15–18, 19–64, and 65 + years). Nine of the samples contained more than 2.5 μg/kg OTA, with an average of 5.24 μg/kg. The OTA concentration in 3 samples exceeded the permissible maximum limit (5 μg/kg) established by Turkish legislation, and the mean concentration was 8.41 μg/kg. A margin of exposure (MOE) approach was used for risk characterization, considering both non-neoplastic and neoplastic consequences. There were no concerns about health risks because MOEs were more than 10,000 for all categories. Although the levels of OTA analyzed in Turkish coffee did not pose a risk to individuals in the three age categories, emphasis should be placed on minimizing and controlling OTA concentrations in Turkish coffee. The occurrence of Ochratoxin A (OTA) in wine is commonly known, but there is only limited information about its occurrence in grape mash and wines of German origin. Climate change has led to higher temperatures in the southern regions of Germany, which may increase the growth of fungi associated with the production of OTA and increase the content of this mycotoxin in grapes. A safe and rapid UHPLC-FLD method was developed and validated to assess the contamination of grape mash and wine with OTA. A total of 71 samples of grape mash and 30 wines from various wine producers in Baden-Württemberg, Germany, were analysed for OTA content. Simple Summary: Ochratoxin A (OTA) present in food products poses a serious threat to health due to its wide spectrum of effects, including genotoxicity, nephrotoxicity, neurotoxicity, embryotoxicity, teratogenicity, and immunosuppression. Moreover, it is associated with carcinogenesis, as a result of which it has been classified as a potential human carcinogen. In this review, literature reports on the correlations between carcinogenesis and OTA, with particular emphasis on certain organs such as the kidney or digestive system organs and the cancers associated with them, are gathered. Furthermore, we describe the potential mechanisms underlying OTA-induced carcinogenesis and discuss existing limitations. The obtained results indicate a strong correlation between OTA and renal cell carcinoma (RCC), with potential associations with other malignancies, including hepatocellular carcinoma (HCC), gallbladder cancer (GBC), and squamous cell carcinoma (SCC). OTA is implicated in oxidative stress, lipid peroxidation, apoptosis, DNA damage, adduct formation, miRNA deregulation, and distributions in the cell cycle, all of which may contribute to carcinogenesis. Conclusions: Despite significant research efforts, the topic remains inexhaustible and requires further investigation. The obtained results do not yield definitive conclusions, potentially due to species-specific differences in the animal models used and challenges in extrapolating these results to humans. Simple Summary: Mycotoxins are secondary metabolites of molds, primarily Penicillium, Fusarium, and Aspergillus species, that may be present in food and feed. Reports indicate that up to 25% of the world's cereal crops could be contaminated with these fungal toxins. Synbiotics, products that synergistically combine probiotics and prebiotics, can reduce harmful metabolites in the gastrointestinal tract by metabolizing or adsorbing toxic substances, including mycotoxins. In the present study, turkeys were fed diets contaminated with ochratoxin A and supplemented with probiotic or synbiotic preparations. The addition of probiotic and synbiotic preparations based on lactic acid bacteria strains, inulin, and Saccharomyces cerevisiae yeast to ochratoxin A-contaminated diets in commercial turkey farming may improve health status and reduce mycotoxin accumulation in poultry organs and tissues. The aim of this study was to evaluate carcass quality and analyze gastrointestinal functional status, ochratoxin A (OTA) accumulation in tissues and organs, and the health status of turkeys fed diets contaminated with OTA and supplemented with synbiotic preparations in comparison with commercial probiotic feed additives. The research involved 120 female BIG 6 turkeys, divided into six treatment groups (five replicates, four birds per replicate). Wheat naturally contaminated with OTA (662.03 μg/kg) was used in turkey diets. Turkeys in group 1 received an OTA-contaminated diet without additives. Groups 2 and 3 received 0.4 g/kg of probiotic preparation BioPlus 2B or Cylactin. Groups 4, 5, and 6 received 0.5 g/kg of synbiotics S1, S2, or S3. The following parameters were monitored: growth performance, carcass quality, gastrointestinal tract structure and digesta pH, health status, and concentrations of OTA in the blood and tissues of turkeys. The study found no significant differences in the growth performance and carcass quality of turkey. However, the introduction of probiotics or synbiotics into OTA-contaminated feed mixtures resulted in a reduced pH of the digesta in certain sections of the turkey digestive tract (p < 0.05). Additionally, the tested synbiotic additives significantly reduced liver weight in turkeys at weeks 6 and 15 (p < 0.05). Background: Ochratoxin A (OTA) is a toxin widely found in aquafeed ingredients, and hypoxia is a common problem in fish farming. In practice, aquatic animals tend to be more sensitive to hypoxia while feeds are contaminated with OTA, but no studies exist in this area. This research investigated the multiple biotoxicities of OTA and hypoxia combined on the liver of grass carp and explored the mitigating effect of curcumin (CUR). Methods: A total of 720 healthy juvenile grass carp (11.06 ± 0.05 g) were selected and assigned randomly to 4 experimental groups: control group (without OTA and CUR), 1.2 mg/kg OTA group, 400 mg/kg CUR group, and 1.2 mg/kg OTA + 400 mg/kg CUR group with three replicates each for 60 d. Subsequently, 32 fish were selected, divided into normoxia (18 fish) and hypoxia (18 fish) groups, and subjected to hypoxia stress for 96 h. Results: CUR can attenuate histopathological damage caused by coming to OTA and hypoxia by reducing vacuolation and nuclear excursion. The alleviation of this damage was associated with the attenuation of apoptosis in the mitochondrial pathway by decreasing the expression of the pro-apoptotic proteins Caspase 3, 8, 9, Bax, and Apaf1 while increasing the expression of the anti-apoptotic protein Bcl-2, and attenuation of endoplasmic reticulum stress (ERS) by reducing Grp78 expression and chop levels. This may be attributed to the fact that the addition of CUR increased the levels of catalase (CAT) and glutathione reductase (GSH), increased antioxidant capacity, and ensured the proper functioning of respiratory chain complexes I and II, which in turn reduced the high production of reactive oxygen species (ROS), thus alleviating apoptosis and ERS. Conclusions: In conclusion, our data demonstrate the effectiveness of CUR in attenuating liver injury caused by the combination of OTA and hypoxia. This study aimed to assess the presence of ochratoxin A (OTA) residues in the blood serum of slaughtered pigs in Greece. Samples were obtained from 1695 healthy slaughtered pigs originating from 113 different farms located in 21 geographic regional units in 8 different geographic regions of Greece and were analyzed using an immunosorbent assay (ELISA) and high-performance liquid chromatography with fluorescence detector (HPLC-FD). OTA contamination assessment showed that 782 (46.1%) and 1233 (72.7%) samples were OTA-positive, with a concentration range of 0.20–5.38 μg/L and 0.15–5.96 µg/L according to ELISA and HPLC-FD analysis, respectively. Also, 88 (77.9%) and 108 (95.6%) of farms were found to be OTA-positive by ELISA and HPLC-FD analysis, respectively. The highest OTA serum positivity rate (>98%) and toxin level (5.96 µg/L) determined by HPLC-FD were observed in the Thessaly region, whereas a high prevalence of up to 100% (range 75–100%) was found on farms in the Crete Island region. The detection of OTA in the serum of slaughtered pigs in different regions in Greece poses a risk for animal and human health and highlights the need for constant OTA monitoring in the swine industry and pork meat production facilities. 1. This trial investigated the effect on embryo injected with ochratoxin A (OTA) and the growth performance, jejunal morphology and barrier of ducklings to 21 d old. 2. Two hundred forty, fertilised eggs were individually weighed and randomly assigned to two groups, a control (CON) and the OTA treatment, according to average egg weight. On d 13 of embryonic development, the treatment group was injected with 8 ng OTA/g egg and the CON group was injected with NaHCO3 solution as a placebo. All newly hatched ducklings were assigned to the CON or OTA group based on the different treatments. Each treatment consisted of six replicates and each included 10 ducklings and the experiment lasted until 21 d of age. 3. The results showed that embryos injected with OTA affected the 21 d body weight (BW) and average daily gain (ADG) of ducklings (p < 0.05). OTA exposure increased the relative weights of the liver, pancreas, gizzard, proventriculus and jejunum (p < 0.05); and decreased the relative length of the jejunum of ducklings (p < 0.05). Moreover, jejunal crypt depth increased (p < 0.05) and the villus height-to-crypt depth ratio (Vh/Cd) decreased in the OTA-injected group (p < 0.05). Compared with those in the CON group, the mRNA expression of Zonula Occludens-1; (ZO-1) (p = 0.0582) and Occludin; (p = 0.0687) in the OTA treatment group was downregulated. 4. The findings demonstrated that a single low-dose injection of OTA increased body weight and daily gain in ducklings. Moreover, embryo exposure to OTA had negative effects with increased relative weight of organs and the jejunal crypt depth, decreased relative length of the intestine and mRNA expression of tight junctions (ZO-1, Occludin). Ochratoxin A (OTA), as one of the most important and harmful mycotoxins, is classed as possible human carcinogen (group 2B). As we all know, DNA damage may cause genomic instability, cell cycle disorder, activation of DNA damage pathway, and stimulation of DNA repair system. To explore the roles of DNA damage repair protein (hMLH1) on OTA-induced G 2 arrest, the DNA damage, chromosome aberration, cell cycle distribution and p53-p21 signaling pathway were evaluatd after different time OTA exposure (6, 12, 24, 48 h) in immortalized human gastric epithelial cells (GES-1). Our results demonstrated that OTA exposure could trigger genomic instability, DNA damage and G 2 phase arrest of GES-1 cells. At the same time, OTA treatment could increase the expression of hMLH1, and induce phosphorylation of the p53 protein, as well as p21, in response to DNA damage. Finally, inhibition of hMLH1 by siRNA effectively prevented the activation of p53-p21 signaling pathway and rescued the G 2 arrest elicited by OTA. This study demonstrated that hMLH1-p53-p21 signaling pathway played an important role in DNA damage and G 2 cell cycle arrest the mediated by OTA in GES-1 cells. • OTA treatment for different amounts of time in GES-1 cells could induce chromosomal aberrations, DNA damage and G 2 /M phase arrest. • OTA treatment for different amounts of time induced GES-1 cells to arrest at G 2 /M phase by regulating the expression of Cdc2, p-Cdc2 and CyclinB1. • The hMLH1-p53-p21 signaling pathway play an important role in OTA-caused G 2 phase arrest in GES-1 cells. In the current work, a covalent organic framework-reinforced hollow fiber-solid phase microextraction method has been introduced for the extraction of patulin and ochratoxin A from commercial apple juices before their determination with high-performance liquid chromatography-tandem mass spectrometry. This method benefited from the advantages of traditional hollow fiber-liquid phase microextraction and solid phase microextraction methods. Briefly, in this method, after preparation of the covalent organic framework-reinforced hollow fiber through a simple method, it was placed into the sample solution and the mixture was stirred to increase the contact surface. After stopping the stirring process, the hollow fiber was taken and placed into a vial containing choline chloride: dichloroacetic acid deep eutectic solvent as an elution solvent. In the following, the mixture was sonicated to accelerate the desorption process. Finally, the eluate containing the extracted analytes was taken and injected into separation system. Under optimized conditions, validation of the developed method was done and the outcomes indicated great precision (relative standard deviations ≤ 5.2%), good linearity (coefficient of determination ≥ 0.9974) in a wide range (77.3–1 × 106 ng L−1), and low limits of detection (23.2 and 15.6 ng L−1 for patulin and ochratoxin A, respectively) and quantification (77.3 and 52.1 ng L−1 for patulin and ochratoxin A, respectively). At the end, the suggested approach was employed for the analysis of patulin and ochratoxin A in thirty commercial apple juice samples. AbstractThe human immunodeficiency virus (HIV) heavily affects women from resource-limited settings who are vulnerable to potentially harmful mycotoxins including aflatoxin B1 (AFB1), fumonisin B1 (FB1) and ochratoxin A (OTA). We aimed to conduct biomonitoring and ascertain the determinants of maternal mycotoxin exposure in pregnancy, lactation and post-lactation periods. We conducted a retrospective longitudinal study in HIV-infected and HIV-uninfected women from Harare, Zimbabwe. 175 and 125 random urine samples in pregnancy and 24 months after delivery (post-lactation) respectively were analysed for aflatoxin M1 (AFM1) and FB1 by ELISA. 6 weeks after delivery (lactation), 226 and 262 breast milk (BM) samples were analysed for AFM1 and OTA respectively by ELISA. The association of demographics and food consumption with mycotoxins was evaluated using multivariable logistic regression. In HIV-infected, urinary AFM1 was detected in 46/94 (Median: 0.05; Range: 0.04–0.46 ng mL−1) in pregnancy and 47/66 (Median: 0.05; Range: 0.04–1.01 ng mL−1) post-lactation. Urinary FB1 was detected in 86/94 (Median: 1.39; Range: 0.17–6.02 ng mL−1) in pregnancy and 56/66 (Median: 0.72; Range: 0.20–3.81 ng mL−1) post-lactation. BM AFM1 was detected in 28/110 (Median: 7.24; Range: 5.96–29.80 pg mL−1) and OTA in 11/129 (Median: 0.20; Range: 0.14–0.65 ng mL−1). In HIV-uninfected, urinary AFM1 was detected in 48/81 (Median: 0.05; Range: 0.04–1.06 ng mL−1) in pregnancy and 41/59 (Median: 0.05; Range: 0.04–0.52 ng mL−1) post-lactation. Urinary FB1 was detected in 74/81 (Median: 1.15; Range: 0.17–6.16 ng mL−1) in pregnancy and 55/59 (Median: 0.96; Range: 0.20–2.82 ng mL−1) post-lactation. BM AFM1 was detected in 38/116 (Median: 7.70; Range: 6.07–31.75 pg mL−1) and OTA in 4/133 (Median: 0.24; Range: 0.18–0.83 ng mL−1). Location, wealth, and peanut butter consumption were determinants of AFB1 exposure. HIV infection, BMI, location, rainy season, unemployment, and age were determinants of FB1 exposure. AbstractCoffee is one of the most commonly consumed beverages worldwide, so assessing its quality for potential health risks is essential. Therefore, this review aimed to determine the levels of ochratoxin A (OTA) in coffee worldwide and then estimate its human intake and health risks. The systematic search took place from June 1997 to April 2024 and 40 of 254 articles were selected based on the selection criteria. The results showed significant differences in average levels of OTA between countries, continents and coffee types (p < 0.001). Of 3256 samples, OTA was detected in 1778, accounting for 54.6% of the total, with the percentage of positive results varying between 7.5% and 100%. Only two studies reported OTA average levels in roasted coffee exceeding the maximum limit (ML) set by the European Commission (ML-EC = 5 μg/kg). The average OTA in soluble coffee was lower than ML-EC (10 μg/kg) in all studies, and in instant coffee, the level of OTA was higher than ML-EC (10 μg/kg) only in one study. The estimated daily intake (EDI) of OTA in all coffee types was lower than the provisional tolerable daily intake (PTDI) values set by joint FAO/WHO Expert Committee on Food Additives (JECFA) (14 ng/kg bw/day) and proposed by the European Food Safety Authority (EFSA) (17 ng/kg bw/day). Non-carcinogenic risk assessment through coffee consumption indicated that the hazard quotient (HQ) was below the acceptable level, HQ = 1. The Margin of Exposure (MoE) for neoplastic effects was acceptable and unacceptable for non-neoplastic effects (NNE) in 4.5% (one of 22 cases) of the roasted and soluble coffees, but acceptable for all instant coffees. In conclusion, the study shows that the OTA content of coffee is not toxic to consumers worldwide. In the current study, Ochratoxin A (OTA) levels and prevalence in beer were meta‐analyzed, and non‐carcinogenic risk was calculated using the target hazard quotient. Thirty papers with 70 data reports were included in our study. The pooled prevalence of OTA was 50.60%, 95% CI (confidence interval) (36.97–64.19). Five countries with the highest levels of OTA in beer were South Africa (1170.750 μg/L), Slovakia (31.300 μg/L), Portugal (3.140 μg/L), Tunisia (1.052 μg/L), and Greece (0.707 μg/L). The pooled levels of OTA were 0.089 μg/L, 95% CI (0.077–0.101 μg/L). Meta‐regression shows that OTA prevalence in beer decreased over time insignificantly (p value: .084). Except in South Africa and Slovakia, consumers in other countries are at an acceptable non‐carcinogenic risk due to OTA in beer. Summary In this research, the effects of washing treatments (potassium carbonate (PC), potassium hydroxide (PH), peracetic acid (PA), alkaline hydrogen peroxide (AHP)), ultrasound (US) and high‐intensity pulsed light (HIPL) technologies on the removal of ochratoxin A (OTA) residues in raisins were investigated. PC at 5 min (66.60%), PH at 10 min (65.25%), alkaline hydrogen peroxide at 5 min (63.30%) and HIPL at 12 J cm−2 (62.50%) were found to be the most successful applications in OTA degradation, respectively. Although the OTA degradation rate was high after chemical washing for 10 min, the raisins had a chemical odour. This study reviews global levels of ochratoxin A (OTA) in infant formula and cereal-based foods, using Monte Carlo simulation to assess risks. The review found 24 studies on global OTA levels in infant food and cereal-based products, using databases including PubMed, Scopus, Web of Science and Embase until March 2024. We estimated OTA exposure in infant food based on concentration, intake and body weight. The exposure and hazard quotient margin were calculated using BMDL10 and TDI values. Monte Carlo simulation evaluated human health risks from OTA in infant formula and cereal-based foods. A global study from 14 countries shows varying levels, surpassing EU limits in Tunisia, Ecuador, the USA, and generally in Africa, notably in infant cereals, which had higher levels than formula. Globally, OTA was present in 29.3% of the 3348 samples analyzed, with Lebanon at 95.2% and Brazil at 0%. Analysis indicates only non-carcinogenic risk for infants. Mycotoxin contamination in agri-food systems has been a serious global concern over the past few decades. Corn is easily contaminated by Ochratoxin A (OTA) and Zearalenone (ZEN), which seriously threaten the survival and health of humans and animals. Herein, a rapid and sensitive method of OTA and ZEN dual quantum dots (QDs) fluorescence immunochromatographic test strip was established in this study. OTA and ZEN fluorescence probes were prepared by carbodiimide. The OTA-OVA, ZEN-BSA and staphylococcal protein A were sprayed on the nitrocellulose filter membrane as the T1 line, T2 line and control line of the dual test strip, which exhibited high sensitivity, accuracy and specificity. For quantitative detection of OTA, the linear regression equation was y = 0.3222x + 0.3834 (R2 = 0.9687), LOD and IC50 were 0.132 ng/mL and 2.296 ng/mL, and the linear detection range was 0.269 ng/mL to 19.588 ng/mL. In addition, for quantitative detection of ZEN, its linear regression equation was y = 0.3077x + 0.3777 (R2 = 0.9648), LOD and IC50 were 0.125 ng/mL and 2.495 ng/mL, and the linear detection range was 0.264 ng/mL to 23.55 ng/mL. Conclusions In conclusion, our data demonstrate the effectiveness of CUR in attenuating liver injury caused by the combination of OTA and hypoxia. This study confirms the feasibility and efficacy of adding natural products to mitigate toxic damage to aquatic animals.

Ochratoxin A (OTA) is one of the most common pollutants in aquatic feed. As a first line of defense, intestinal barriers could be utilized against OTA in order to prevent disorders. Natural product supplementation is one of the most popular strategies to alleviate toxicity induced by mycotoxins, but there is a lack of knowledge about how it functions in the teleost intestine. In this study, 720 juvenile grass carp of about 11 g were selected and four treatment groups (control group, OTA group, curcumin [Cur] group, and OTA + Cur group) were set up to conduct a 60-day growth test. After the test, the growth performance and intestinal health related indexes of grass carp were investigated. The addition of dietary Cur could have the following main results: (1) inhibit absorption and promote efflux transporters mRNA expression, reducing the residuals of OTA, (2) decrease oxidative stress by reducing oxidative damage and enhancing the expression of antioxidant enzymes, (3) promote mitochondrial fusion proteins to inhibit the expression of mitotic proteins and mitochondrial autophagy proteins and enhance mitochondrial function, (4) reduce necroptosis-related gene expression through inhibiting the tumor necrotic factor receptor-interacting protein kinase/mixed lineage kinase domain-like pathway, (5) reduce the expression of pro-inflammatory factors by inhibiting the Toll-like receptor 4/nuclear factor-κB signaling pathway to alleviate the intestinal inflammatory response. In summary, the results suggested that Cur could alleviate OTA-induced intestinal damage by enhancing antioxidant capacity and mitochondrial function as well as reducing necroptosis and inflammation in the grass carp intestine. This study provided a theoretical basis and production implications for dietary Cur that could improve growth performance and alleviate the intestinal damage induced by OTA in fish.

Background and objective: This study investigated the protective effects of kaempferol, a natural flavonoid known for its antioxidant properties, against ochratoxin A-induced renal damage. Ochratoxin A, a nephrotoxic mycotoxin found in contaminated food and feed, primarily induces renal damage through oxidative stress. The objective was to assess kaempferol’s potential in mitigating ochratoxin A -induced renal toxicity and its underlying molecular mechanisms. Methods: Male albino mice were divided into four groups: normal control, kaempferol control, ochratoxin A -induced renal toxicity, and ochratoxin A -induced renal toxicity treated with kaempferol. Ochratoxin A was administered twice a week for 8 weeks at 5 mg/kg body weight to induce renal toxicity. Kaempferol (50 mg/kg) was administered twice a week for 8 weeks after a 4-hour interval following ochratoxin A administration. Results: Results showed that kaempferol normalized levels of blood urea nitrogen (BUN) and creatinine, indicating protection against ochratoxin A-induced pathophysiology. Kaempferol also increased phosphorylation of PI3K and AKT, potentially activating antioxidative and anti-apoptotic signaling pathways regulated by Nrf2. Additionally, kaempferol enhanced Bcl-2 activation while suppressing caspase-3, suggesting anti-apoptotic effects. Histopathological analysis revealed reduced renal damage in kaempferol-treated mice compared to those exposed to ochratoxin A alone. Conclusion: Kaempferol demonstrated efficacy in preventing ochratoxin A -induced kidney injury through antioxidative and anti-apoptotic mechanisms. These findings suggest potential therapeutic applications of kaempferol in oxidative stress-induced renal damage.

A novel ratio colorimetric sensor based on magnetic nanoparticles (Fe3O4@Au) and silver nanoparticles (Ag NPs) was constructed for the detection of Ochratoxin A (OTA) by complementary DNA base pairing. The OTA aptamer coupled Fe3O4@Au was served as reference signal, and complementary DNA (cDNA) coupled Ag NPs was served as the response signal. A ratio nanoprobe (Fe3O4@Au-Ag) with two UV absorption peaks was prepared by regulating the ratio and DNA base complementarity. In absence of OTA, the absorbance peak ratio (A400/A580) of the ratio colorimetric sensor could maintain a constant value. The detection conditions (time, pH, and temperature) were optimized, and under optimized conditions, the sensor showed a good linear relationship to OTA detection, with a linear range of 0.01 ng·mL−1 to 1000 ng·mL−1 and a detection limit of 0.033 ng·mL−1. The sensor had good selectivity, reproducibility and high sensitivity, which was successfully applied to the detection of OTA in pea powder with a recovery rate of 93.9%~96.0%.

Simple Summary: Ochratoxin A (OTA), a nephrotoxic mycotoxin categorized as a possible carcinogenic agent, is a current problem in the cheese manufacturing industry. The main OTA producers are Penicillium nordicum and Penicillium verrucosum. Therefore, we aimed to investigate different cheese ripening phases to hinder OTA production. We elaborated cheese analogues, inoculated both fungal species, and performed two different ripening phases, at 10 and 15 °C. Despite the fact that both were able to grow, they did not express genes related to OTA production, and no OTA was detected during two weeks of ripening. Therefore, we show that, when the first phases of ripening are maintained at <15 °C, the possibility of OTA production is dramatically minimized. This information is of utmost importance for industries to enhance the safety of their cheese products, which positively impacts human health. A lack of control of the technological abiotic parameters apparent during cheese manufacture, including temperature and relative humidity, results in this dairy product being prone to mold contamination. Sometimes, inoculant molds are used to obtain the characteristic sensory properties of this type of product. However, during the maturation process, some unwanted molds can colonize the ripening cheese and produce mycotoxins. Mycotoxigenic molds such as Penicillium nordicum and Penicillium verrucosum can colonize ripened cheeses, contaminating them with ochratoxin A (OTA), a nephrotoxic 2B toxin. Thus, the presence of OTA in cheeses could represent a hazard to consumers' health. This study has evaluated the growth and OTA production of P. nordicum and P. verrucosum on a cheese analogue under simulated ripening conditions of 10 and 15 °C and 0.96 water activity (aw). Ecophysiological, molecular, and analytical tools assessed the mold growth, gene expression, and OTA production under these environmental conditions. Both species were able to effectively colonize the cheese under these ripening conditions. However, neither species expressed the otapks and otanps biosynthetic genes or produced phenotypic OTA. Therefore, these results suggest a relatively low risk of exposure to OTA for consumers of this type of cheese product. The conditions used were thus appropriate for cheese ripening to minimize the potential for contamination with such mycotoxins. Ochratoxin A (OTA) is a mycotoxin mainly produced by Aspergillus section Circumdati and section Nigri across the coffee chain. OTA is nephrotoxic and is a threat to human health. This review summarizes current knowledge on how to reduce OTA concentration in coffee from farm to cup. After a brief introduction to the OTA occurrence in coffee, current good management practices are introduced. The core of this review focuses on biocontrol and microbial decontamination by lactic acid bacteria, yeasts and fungi, and their associated enzymes currently reported in the literature. Special attention is given to publications closest to in vivo applications of biocontrol agents and microbial OTA adsorption or degradation agents. A novel electrochemical aptasensor was prepared for the simultaneous determination of aflatoxin B1 (AFB1) and ochratoxin A (OTA). Composites of Au nanoparticles and polyethyleneimine-reduced graphene oxide (AuNPs/PEI-RGO) with good electrical conductivity and high specific surface area were employed as the supporting substrate, demonstrating the ability to provide more binding sites for aptamers and accelerate the electron transfer. Aptamers were immobilized on a AuNPs/PEI-RGO surface to specifically recognize AFB1 and OTA. A metal–organic framework of UiO-66-NH2 served as the signal carrier to load metal ions of Cu2+ and Pb2+, which facilitated the generation of independent current peaks and effectively improved the electrochemical signals. The prepared aptasensor exhibited sensitive current responses for AFB1 and OTA with a linear range of 0.01 to 1000 ng/mL, with detection limits of 6.2 ng/L for AFB1 and 3.7 ng/L for OTA, respectively. The aptasensor was applied to detect AFB1 and OTA in cereal samples, achieving results comparable with HPLC-MS, with recovery results from 92.5% to 104.1%. Mycotoxins in animal feed pose health risks and economic losses, but using various adsorbent types could potentially protect animals from mycotoxicosis. The study aimed to assess the effect of OTA on the health of broiler chickens and to envisage the ameliorative potential of clay adsorbents. The objectives of this in vivo study were to investigate the effects of OTA on productivity, biochemical parameters, fecal residues, and the preventive effects of indigenous and commercial clay of Balochistan as adsorbents to alleviate the adverse effects of exposure. Male broiler chickens (n = 160) were treated with 400 μg/kg OTA and 0.5 g/kg clay adsorbent for 42 days, with feed and water available in an ad libitum manner. The amount of OTA in diet and fecal residues was assessed through HPLC. The administration of OTA in the diet, resulted in a significant (p < 0.05) decrease in the average daily gain (ADG) and average daily feed intake (ADFI) while increasing the feed conversion ratio (FCR) as compared to the control group. Furthermore, no significant (p > 0.05) differences were found between the weight gain of broiler chickens fed without OTA (positive control) and that of chickens fed adsorbent. The group given a diet containing OTA without adsorbents as compared to the control and adsorbent-supplemented group has shown a significant (p < 0.05) increase in the relative weight of the liver, kidney, gizzard, and proventriculus while decreasing the relative weight of the spleen and bursa of Fabricius. Alterations in the levels of serum total protein (TP), cholesterol (CHL), serum urea (SU), enzymatic activity (aspartate aminotransferase (AST) and alanine transaminase (ALT)), and creatinine were observed in the OTA-intoxicated and adsorbent-supplemented groups as compared to the control group. Adsorbent supplementation resulted in a significantly (p < 0.05) higher OTA content in the faeces. It can be concluded from the results of this study, that OTA intoxication negatively affects the health of broiler chickens, and the clay of Balochistan has shown effective adsorption potential against OTA. Ochratoxin A (OTA) is a widespread food toxin produced by Aspergillus ochraceus and other molds. In this study, we developed and established acute OTA toxicity conditions in mice, which received daily oral doses of OTA between 0.5 up to 8 mg/kg body weight up to 7 days and were subjected to histological and biochemical analysis to characterize renal and hepatic damage. Oral administration of OTA for 7 days resulted in loss of body weight in a dose-dependent manner and increased the levels of serum biomarkers of hepatic and renal damage. The kidney was more sensitive to OTA-induced damage than the liver. In addition to necrosis, OTA induced hepatic and renal apoptosis in dose- and time-dependent manners. Especially, a high dose of OTA (8 mg/kg body weight) administered for 7 days led to necroptosis in both liver and kidney tissues. OTA dose-dependently increased the oxidative stress levels, including lipid peroxidation, in the liver and kidneys. OTA disrupted mitochondrial dynamics and structure in hepatic and renal cells, leading to the dysregulation of mitochondrial homeostasis. OTA increased transferrin receptor 1 and decreased glutathione peroxidase 4 levels in a dose- and time-dependent manner. These results suggest the induction of ferroptosis. Collectively, this study highlighted the characteristics of acute OTA-induced hepatic and renal toxicity in mice in terms of oxidative stress, mitochondrial damage, and multiple cell death mechanisms, including necroptosis and ferroptosis. Antimicrobial peptides (AMPs) function to extensively suppress various problematic factors and are considered a new alternative for improving livestock health and enhancing immunomodulation. In this study, we explored whether AMP regulation has positive influences on Ochratoxin A (OTA) exposure using a porcine intestinal epithelial cell line (IPEC-J2 cells). We constructed a beta-defensin 1 (DEFB1) expression vector and used it to transfection IPEC-J2 cells to construct AMP overexpression cell lines. The results showed that OTA induced cytotoxicity, decreased cell migration, and increased inflammatory markers mRNA in IPEC-J2 cells. In DEFB1 overexpressing cell lines, OTA-induced reduced cell migration and increased inflammatory markers mRNA were alleviated. Additionally, a natural product capable of inducing DEFB1 expression, which was selected through high-throughput screening, showed significant alleviation of cytotoxicity, cell migration, and inflammatory markers compared to OTA-treated IPEC-J2 cells. This study investigated the prevalence of the mycotoxin Ochratoxin A in poultry feed samples from Eastern Algeria, an important poultry production region. Ochratoxin A, produced by Aspergillus and Penicillium fungi, can contaminate feed, leading to toxicity issues for poultry and human beings consuming contaminated products. Analysis of 42 feed samples using the Enzyme-Linked Immunosorbent Assay (ELISA), found 62% contained detectable ochratoxin A levels exceeding the concentration of 1.50 ng g−1, varying between 2.23 ng g−1 and 3.23 ng g−1 with an average of 2.59 ng g−1. Although the widespread prevalence is below current regulatory limits, there is a need for continued monitoring to prevent contamination and toxin transfer to poultry meat and eggs. The study recommends expanding testing to more regions and implementing Ochratoxin A surveillance. MD-2 pineapple fruits are susceptible to phytopathogenic fungi producing mycotoxins, which can cause up to 40% of postharvest losses. This study aimed to isolate and identify fungi associated with postharvest diseases on pineapple fruits, and mycotoxins produced. Thirty-seven fungal strains were isolated from the pineapple fruit at the full maturation stage. About 51.37% of the fungal strains identified were belonging to Aspergillus. Those fungi that caused diseases in pineapple fruits were Aspergillus sclerotioniger, A. tubingensis, Penicillium citrinum, Fusarium equiseti, Trichoderma asperelliodes and T. asperellum. Aspergillus niger produced the highest ochratoxin A (OTA), scoring 85.51 µg/kg. On the other hand, P. citrinum scored 94.06 µg/kg, the highest quantity of total aflatoxins. Ochratoxin A (OTA) is a toxic fungal metabolite that is commonly found in cereals and animal feed. It is economically damaging and potentially hazardous to human health. Herein, we propose an electrochemical immunosensor for the rapid detection of OTA using anti-OTA antibodies and diazonium-functionalized, screen-printed electrodes. We attached 4-aminobenzoic acid to an electrode surface, activated the carboxyl groups on the surface with carbodiimide, and attached an antibody to the diazo layer. Subsequently, we used bovine serum protein as a blocker to prevent non-specific antigens from binding to the antibody. We evaluated the performance of the sensor by cyclic voltammetry, electrochemical impedance spectroscopy, and differential pulse voltammetry. The sensor is highly specific and sensitive, has good linear responses in the range 20–200 ng/mL, a limit of detection of 0.5 ng/mL, and good recoveries of 90.5%–100.9% in spiked samples. Viticulture has been an important economic sector for centuries. In recent decades, global wine production has fluctuated between 250 and almost 300 million hectoliters, and in 2022, the value of wine exports reached EUR 37.6 billion. Climate change and the associated higher temperatures could favor the occurrence of ochratoxin A (OTA) in wine. OTA is a mycotoxin produced by some species of the genera Aspergillus and Penicillium and has nephrotoxic, immunotoxic, teratogenic, hepatotoxic, and carcinogenic effects on animals and humans. The presence of this toxin in wine is related to the type of wine—red wines are more frequently contaminated with OTA—and the geographical location of the vineyard. In Europe, the lower the latitude, the greater the risk of OTA contamination in wine. However, climate change could increase the risk of OTA contamination in wine in other regions. Due to their toxic effects, the development of effective and environmentally friendly methods to prevent, decontaminate, and degrade OTA is essential. Contamination of food with mycotoxins can pose harmful effects on the health of consumers in the long term. Coffee contamination with mycotoxins has become a global concern. This study attempted to meta-analyze the concentration and prevalence of ochratoxin A (OTA) in coffee products and estimate consumers' health risks. The search was conducted among international databases, including Scopus, PubMed, Embase, and Web of Science, for 1 January 2010 to 1 May 2022. The concentration and prevalence of OTA in coffee products were meta-analyzed according to country subgroups. Health risk assessment was conducted based on Margin of Exposures (MOEs) using the Monte Carlo simulation (MCS) technique. The three countries that had the highest Pooled concentration of OTA in coffee were observed in Chile (100.00%), Kuwait (100.00%), and France (100.00%). The overall prevalence of OTA in coffee products was 58.01%, 95% CI (48.37–67.39). The three countries that had the highest concentration of OTA were Philippines (39.55 μg/kg) > Turkey (39.32 μg/kg) > and Panama (21.33 μg/kg). The mean of MOEs in the adult consumers in Panama (9,526) and the Philippines (8,873) was lower than 10,000, while the mean of MOEs in other countries was higher than 10,000. The current trial aimed to investigate the ameliorative effect of pumpkin seeds extract (PSE, Cucurbita maxima)-supplemented diets against ochratoxin A (OTA) toxicity in terms of growth performance, serum biochemical parameters, antioxidative enzymes, immunity parameters and intestine and liver histopathology of Nile tilapia (Oreochromis niloticus). Two hundred randomly distributed healthy fish (initial weight = 16.84 ± 0.061 g) were divided into four diverse groups. Each group of fish was stocked into five aquaria, each containing 10 fish. The first group was served as control and fed a basal contaminated diet (600 μg OTA/kg diet) while the other three groups were fed OTA contaminated diets and supplemented with graded amounts of PSE (0.25, 0.5 or 1 mg/kg). The treatments used were as follows: OTA + PSE (0.25), OTA + PSE (0.5), and OTA + PSE (1). The results showed that fish administered high levels of PSE significantly boosted growth and feed efficiency parameters. Additionally, supplementing OTA-contaminated diets with 1 mg PSE/kg diet reduced the deleterious effects of OTA on liver enzyme activities (aspartate aminotransferase, and alanine amniotransferase). Meanwhile, the protein constituents were significantly improved in all PSE-supplemented groups. In the same context, the antioxidant characteristics (superoxide dismutase, glutathione peroxidase, total antioxidant activity, and malonaldehyde), proinflammatory cytokines (interleukin-1, IL-1), and immune responses (lysozyme and immunoglobulin M and D) were substantially improved by supplemented feed diets with 1 mg PSE/kg diet, however, tumor necrosis factor was suppressed. Particularly, the OTA levels in the muscle were not detected in the fish group fed OTA-contaminated diets and supplemented with 1 mg PSE/kg diet. Considerably, supplementing OTA-contaminated diets with 1 mg PSE/kg diet considerably improved hepatocytes and hepatoportal pancreas structure, with just a few fatty damaged cells present, and revealed healthy intestinal morphologies. Finally, diets supplemented with 1 mg PSE/kg diet could mitigate OTA's adverse impacts on Nile tilapia growth and health.

ABSTRACT: Phytic acid (PA) is a natural antioxidant with various biological activities, providing protective effects in multiple animals. Ochratoxin A (OTA) is a mold toxin commonly found in feed, which induces multi-organ damage, with kidney being the target organ of its toxicity. This study investigates the protective effects of PA on OTA-induced renal damage and its potential mechanisms in chicks. The results demonstrates that PA treatment restores OTA-induced renal pathological injuries, reverses the diminished activities of antioxidant enzymes, reduces the accumulation of malondialdehyde, and normalizes the expression of pro-inflammatory cytokines, which confirms that PA can alleviate OTA-induced renal damage. Further investigations reveal that OTA-induced renal injury accompanied by an increase in tissue iron content and the transcription levels of ferroptosis-related genes (TFR, ACSL4, and HO-1), and a decrease in the levels of SLC7A11 and GPX4. PA treatment reverses all these effects, indicating that PA mitigates OTA-induced renal ferroptosis. Moreover, PA supplementation improves intestinal morphology and mucosal function, corrects OTA-induced changes in the intestinal microbiota. Besides, PA microbiota transplantation alleviates renal inflammation and oxidative stress caused by OTA. In conclusion, PA plays a protective role against renal damage through the regulation of ferroptosis and the intestinal microbiota, possibly providing novel insights into the control and prevention of OTA-related nephrotoxicity.

The production of raisins, a method of grape preservation since antiquity, has evolved with various drying techniques that significantly impact the quality and safety of the final product. This study evaluates the efficacy of a solar indirect dryer compared to traditional sun-drying methods for drying Centennial Seedless and Sultanina grape cultivars in Crete, Greece. Key parameters assessed include environmental conditions, drying time, grape color, fungal contamination, and Ochratoxin A (OTA) levels. Grapes were dried in a controlled solar chamber and under open sun conditions. The solar chamber maintained higher average temperatures (34 °C) and lower relative humidity (39.7%) than outside conditions (24.2 °C and 58.7%, respectively), significantly reducing the drying time from 12 to 7 days. Raisins dried in the solar chamber exhibited improved color quality, with higher Lightness (L*), Hue Angle (h), and Chroma (C*) values, attributed to minimized enzymatic and nonenzymatic browning. Mycological analysis revealed a substantial reduction in Aspergillus section Nigri contamination in chamber-dried raisins, with mean colony-forming units per gram significantly lower than those of sun-dried raisins. Consequently, OTA levels were also significantly reduced in chamber-dried raisins, with Centennial Seedless showing a mean concentration of 1.01 µg/kg compared to 2.66 µg/kg in sun-dried samples, and Sultanina showing 0.70 µg/kg versus 2.05 µg/kg, respectively. These findings underscore the advantages of using solar indirect dryers to enhance drying efficiency, improve color quality, and reduce fungal and OTA contamination, highlighting the importance of adopting controlled drying technologies for safer, higher-quality raisins.

Ochratoxin A (OTA) in samples was captured and concentrated by immunoaffinity chromatography (IAC) and detected by enzyme-linked immunosorbent assay (ELISA). The anti-OTA monoclonal antibodies used in IAC and ELISA came from different clone strains, belonging to different idiotypes, and there were differences between their selectivity for binding to OTA targets. The combined use of IAC and ELISA could effectively filter out the interference from OTA analogues, thus improving the specificity and sensitivity of immunological analysis. The limit of detection (LOD) for OTA in spiked samples was 0.2 ng/g, the quantification limit was 0.4 ng/g, and the average recovery was 75.9%. Although the recovery of the IAC-ELISA method was slightly lower than that of ELISA, the sensitivity was increased by 60 times. For 49 real samples, the proportion of samples that tested positive by this combined method was 100% consistent with the result of the national standard method, with a missed detection rate of 0%. Therefore, the developed method exhibited significant advantages in accuracy. As a comprehensive immunoassay, this method does not require any large-scale instruments or equipment, and has no strict requirement on the operating environment, thereby making it easy for grassroot laboratories to analyze OTA in samples.

Abstract Background Ochratoxin A (OTA), a globally abundant and extremely hazardous pollutant, is a significant source of contamination in aquafeeds and is responsible for severe food pollution. The developmental toxicity of OTA and the potential relieving strategy of natural products remain unclear. This study screened the substance curcumin (Cur), which had the best effect in alleviating OTA inhibition of myoblast proliferation, from 96 natural products and investigated its effect and mechanism in reducing OTA myotoxicity in vivo and in vitro. Methods A total of 720 healthy juvenile grass carp, with an initial average body weight of 11.06 ± 0.05 g, were randomly assigned into 4 groups: the control group (without OTA and Cur), 1.2 mg/kg OTA group, 400 mg/kg Cur group, and 1.2 mg/kg OTA + 400 mg/kg Cur group. Each treatment consisted of 3 replicates (180 fish) for 60 d. Results Firstly, we cultured, purified, and identified myoblasts using the tissue block culture method. Through preliminary screening and re-screening of 96 substances, we examined cell proliferation-related indicators such as cell viability and ultimately found that Cur had the best effect. Secondly, Cur could alleviate OTA-inhibited myoblast differentiation and myofibrillar development-related proteins (MyoG and MYHC) in vivo and in vitro and improve the growth performance of grass carp. Then, Cur could also promote the expression of OTA-inhibited protein synthesis-related proteins (S6K1 and TOR), which was related to the activation of the AKT/TOR signaling pathway. Finally, Cur could downregulate the expression of OTA-enhanced protein degradation-related genes (murf1, foxo3a, and ub), which was related to the inhibition of the FoxO3a signaling pathway. Conclusions In summary, our data demonstrated the effectiveness of Cur in alleviating OTA myotoxicity in vivo and in vitro. This study confirms the rapidity, feasibility, and effectiveness of establishing a natural product screening method targeting myoblasts to alleviate fungal toxin toxicity. Graphical Abstract

Introduction. Aflatoxins and ochratoxin A are secondary metabolites of microscopic mold fungi. They seriously threaten human and animal health. These toxins are carcinogenic, teratogenic, hepatotoxic, nephrotoxic substances. International Agency for Research on Cancer (IARC) has classified them as class IA and IIB carcinogens. The maximum permissible concentrations of aflatoxins and ochratoxin A in the Russian Federation are currently regulated only in food products according to the Technical Regulation of the Customs Union 021 "On Food Safety". The content of aflatoxins and ochratoxin A in medicinal plant raw materials is not regulated in any way. Harmful environmental and biological conditions such as temperature, humidity, air quality, insects and others during post-harvest handling can lead to contamination of medicinal raw materials with aflatoxins and ochratoxin A. Therefore, the detection of these toxins is one of the problems of raw materials quality control.Text. The present review summarizes the following methods of sample purification, used at the sample preparation stage: solid-phase extraction, immunoaffin column and QuEChERS. This review also summarizes the following modern analysis methods for the identification of aflatoxin and ochratoxin A in medicinal plant raw materials: thin-layer chromatography, liquid chromatography, gas chromatography and screening methods.Conclusion. The QuEChERS method has been identified as the most popular method for preparing samples for analysis. It is based on solid phase extraction technology. This method combines the variability of approaches to sample purification and allows people to study a wider range of toxins. The high-performance liquid chromatography method has been identified as the most popular method for qualitative and quantitative analysis. It has high selectivity, multicomponent analysis and low detection limits.

Objective: This study was conducted to evaluate the prevalence of total aflatoxin (AF) and ochratoxin A (OTA) in poultry feed ingredients under different environmental conditions during the summer and winter seasons, while the hygiene quality of the feed ingredient was assessed through viable fungal count (VFC). Materials and Methods: A total of 288 poultry feed ingredients (n = 96 each) samples were collected from different poultry shops, which were initially analyzed for the presence of AF and OTA through thin layer chromatography (TLC) and then confirmed the contamination concentration through the enzyme-linked immunosorbent assay method. Results: The results of the current study confirmed the incidence of contamination with AF and OTA by TLC and ELISA methods. The contamination level of AF ranged from 26.09 to 50.56 (mean = 41.22 ± 9.45) μg/kg, whereas the contamination level of OTA ranged from 50.13 to 6.21 (mean 42.60 ± 6.21) μg/kg. The contamination level of AF was found to be above the permissible level set by the Food and Drug Administration (20 μg/kg), whereas the contamination level of OTA was below the permissible limits. Moreover, the VFC values were also below the recommended level. The results showed that the association between AF, OTA, and moisture content was significant (p < 0.05). Conclusion: Mycotoxin contamination was significantly (p < 0.05) highest in the winter season. These findings suggested that continuous monitoring regimes might prevent mycotoxin contamination in poultry feed ingredients. [J Adv Vet Anim Res 2024; 11(1.000): 107-113]

In this study, the effect of ochratoxin A (OTA) concentration and discharge voltage on the degradation of OTA in artificially contaminated raisins by low-temperature discharge plasma was explore. The results indicated that OTA in raisins contaminated with 50 μg/mL OTA was completely degraded by plasma treatment for 10 minutes at a discharge voltage of 75 kV. The degraded products of OTA were detected by high performance liquid chromatography-quadruple-electrostatic field orbitrap high resolution mass spectrometry. Based on the primary and secondary mass spectra, two major degraded products with m/z 426.071 5 (B) and m/z 158.154 0 (C) and the possible degradation pathway of OTA were speculated. There was no significant change in the physicochemical quality of raisins before and after plasma treatment, and the relative contents of most acids, aldehydes, alcohols and ketones did not significantly change, except for a significant decrease in the contents of 3-methylbutyric acid, acetic acid and caprylic acid among acids, and a significant increase in the contents of formic acid, valeric acid, and 2-ethylhexanoic acid. The research results suggest that low temperature discharge plasma can effectively degrade OTA while having no significant effect on raisin quality, which can provide a reference for OTA degradation in contaminated foods.

Coffee is one of the most important agricultural export commodities being relied upon to generate revenue of foreign currency. To this end, it has to be of high quality to satisfy expectation and standards of international coffee markets. However, several mycoflora are naturally associated with coffee, some of which may produce fungal toxins such as Ochratoxin A (OTA) unless the safety of the products is carefully managed. Therefore, this research was initiated to identify the prevalent mycotoxigenic fungi and evaluate the presence of OTA in coffee beans collected from the soil surface and from wet and dry processing coffee mills. A total of 77 coffee samples were collected from Gera, Gomma, Limu Kosa and Mana districts of Jimma Zone, Oromia Regional State, Ethiopia. Malt Extract Agar (MEA) was used for the isolation and identification of fungi using macro and microscopic characteristics of the isolates, and High Performance Liquid Chromatography (HPLC) was used to detect and quantify OTA in green coffee beans using the AOAC method. Different fungal genera, including Aspergillus (84.74%), Fusarium (8.75%), Penicillium (5.49%), and Rhizopus (1.02%), were found associated with coffee beans collected from different coffee processes. Mean OTA values of 6.24 µg/kg, 2.05 µg/kg and 1.2 µg/kg were detected in coffee bean samples collected from soil surface, dry, and wet processed samples, respectively. In the present study, coffee beans collected from soil surfaces were more contaminated with various filamentous molds and their toxins than the dry and wet processed coffee beans with relatively better safety status.
