Zinc (Zn) and iron (Fe) are necessary for structural and metabolic functions of the body like, immune competence, physical growth, reproductive function, and neurobehavioral development. The deficiency of micronutrients is a severe public health distress in nearly all developing countries which leads to the syndrome of malnutrition mostly in children and women. The present study was conducted to compare the effect of different levels of Fe and Zn on feed intake (FI), feed conversion ratio (FCR), daily egg production, and Fe and Zn depositions in the egg yolk of laying hens. For this purpose, 80 commercial laying birds were allocated into four groups consisting of four replicates each (n = 5). The basal ration was provided to the control group (C) while three different levels (100, 200, and 300 mg/kg diet/element) of Fe and Zn (1:1) were provided to the FZ100, FZ200, and FZ300 groups, respectively. Overall FI, hen day egg production, and FCR were not affected (p > 0.05). The Fe and Zn content in egg yolk were significantly increased in the FZ300 group after 4 weeks of supplementation. The cost per egg produced in response to different levels of Fe and Zn supplementation was not affected however a numerical increase in cost/egg was found in the FZ300 group. In conclusion, Fe and Zn supplementation (300 mg/kg diet/element) significantly enhanced the egg Fe and Zn contents without adversely affecting production parameters including FI, egg production, and FCR of laying hens.

This study assessed the potential of a commercial probiotic mixture on performance, gut health and nutrient utilisation of broilers-fed poultry by-product meal (PBM)-based diet. Three-hundred-and-twenty Ross-308-day-old broiler chicks were obtained from a commercial poultry hatchery. The chicks were randomly assigned to four different treatment groups, each consisting of four replicates (n = 10) in a randomised complete design. The positive control group (CON) was fed commercial poultry ration, and the negative control group (N-CON) was fed PBM partially replacing soybean meal at a 65% level. The MSP1 and MSP2 groups contained the same diet as the N-CON group but supplemented with multi-strain probiotics (MSP) at 50 mg/kg and 100 mg/kg. BackgroundNeisseria gonorrhoeae is the bacterium responsible for gonorrhoea, one of the most common sexually transmitted infections (STIs) globally. In 2020, the World Health Organization (WHO) estimated 82.4 million new cases of Neisseria gonorrhoeae infections. Current treatments rely on antibiotics, but the emergence of multi drug resistance (MDR) strains poses a significant threat to public health. This research aims to use computational modeling of cyclotides as antimicrobial agents targeting the Neisseria gonorrhoeae PorB Porin protein to inhibit its pathogenicity.MethodologyThe PorB Porin protein was retrieved from the Protein Data Bank (PDB ID: 4AUI), cleaned, and visualized using Discovery Visual Studio. Physicochemical properties were predicted using ProtParam. Cyclotides were obtained from the CyBase database, with 3D models generated and refined via the Swiss Model for docking studies. HDOCK was used for molecular docking. Toxicity and allergenicity predictions were performed with ToxinPred and AlgPred. A heatmap of the peptide was created using Protein-Sol. Molecular dynamics (MD) simulations were conducted for 100,000 picoseconds using Desmond from Schrödinger LLC, while binding energy was analyzed using MMGBSA. Immune response simulations were done with C-ImmSim 10.1, and peptide simulation in water was performed via WebGro.ResultsThe protein’s GRAVY value is −0.539, indicating moderate hydrophilicity, and its isoelectric point is 9.14, suggesting a fundamental nature. Globa D had the highest docking score (−270.04 kcal/mol) and was deemed non-toxic and non-allergenic. MD simulations showed stable protein-ligand interactions, and MMGBSA revealed a low binding energy of −36.737 kcal/mol. Immune simulations indicated an effective immune response and peptide simulations demonstrated Globa D’s stability in water, making it a potential candidate for pharmaceutical applications.ConclusionGloba D proved the best drug candidate against Neisseria gonorrhoeae by inhibiting PorB Porin protein chain A. Further in vitro and in vivo studies are recommended to validate these findings and explore clinical applications.

Background: Blood transfusion, a prevalent practice in Pakistan with 3.5 million donors annually, carries a 1% risk of transfusion-transmitted infections, contributing to the transmission of infectious diseases. Syphilis, specifically, demonstrates an increasing seroprevalence. Objective: To assess the seroprevalence of syphilis among healthy volunteer blood donors in Rawalpindi. Study type, settings & duration: This prospective cross-sectional study was conducted at Railway General Hospital, Rawalpindi from July to December 2023. Methodology: After acquiring informed consent, blood samples were collected for screening using the immunochromatographic assay (ICT) to detect specific Treponema pallidum antibodies in human serum. The Healgen Syphilis detection Kits (GCSYP-402) were employed for this purpose, offering a rapid qualitative screening for syphilis. Results: A total of 1710 blood samples were screened using the immunochromatographic assay for detecting of Treponema pallidum antibodies. The syphilis seropositivity among donors revealed 16 (0.93%) reactive samples and 1,694 (99.07%) non-reactive samples. Males comprised 1,531 (89.54%) of the participants, with 179 (10.46%) being females. Among donors, 1,209 (70.71%) were single, and 587 (34.33%) had up to matric education. The majority of donors, 1,320 (77.20%), resided in urban areas. Statistical analysis showed no significant associations between syphilis seropositivity and age (p >0.902), gender (p >0.579), educational level (p >0.725), or residence (p >0.697). These findings provide valuable insights into the prevalence and demographic factors of syphilis among blood donors in the study. Conclusion: Enhanced donor evaluation and screening for blood donation can boost recipient safety and curb syphilis transmission.

This study investigated the effects of the β-mannanase enzyme and soyhulls on production performance, economics, egg quality, hematology and serum biochemistry, nutrient digestibility, gut morphology, digesta viscosity, and excreta consistency in laying hens during the late peak production phase (37 to 40 weeks of age). Golden brown hens (RIR × Fayoumi; n = 200) were fed a control diet (no soyhulls or enzymes) and diets containing four combinations, i.e., 3% soyhulls with 20 mg/kg β-mannanase (D1), 3% soyhulls with 30 mg/kg β-mannanase (D2), 9% soyhulls with 20 mg/kg β-mannanase (D3), and 9% soyhulls with 30 mg/kg β-mannanase (D4), for four weeks in four replicates of 10 birds each. Overall, a significantly higher (p < 0.05) feed intake, weight gain, feed conversion ratio, and water intake were calculated in the D2 group as compared to the control and remaining combinations of soyhulls and β-mannanase. No mortality was recorded during the entire experiment. Economically, the D1 and D2 groups showed the best results as compared to the D3 and D4 groups. Egg quality parameters like egg weight, shell weight and shell thickness, yolk weight, albumen weight and height, and the Haugh unit remained unchanged (p > 0.05). Similarly, the D2 group showed significantly lower total cholesterol, LDL, and VLDL levels and enhanced gut morphology with greater villus width, height, crypt depth, and surface area across intestinal segments. Crude protein (CP), crude fiber (CF), crude fat, and ash digestibility were higher (p < 0.05) in the D1 and D2 groups compared to the control. Digesta viscosity, excreta consistency, and other egg quality parameters remained unaffected. In conclusion, the dietary inclusion of a combination of 3% soyhulls and 30 mg/kg β-mannanase may have potential benefits for laying hens by improving some production performance and egg quality indicators and economics, lowering blood cholesterol, LDL, and VLDL levels, enhancing nutrient digestibility, and improving gut morphology without affecting egg quality.

Stingless bee (Hymenoptera, Apidae, and Trigona) honey is a remarkable “miracle liquid” with a wide range of medical benefits for conditions including gastroenteritis, cataracts, and wound healing. Our study aimed to isolate, identify, and characterize acid-resistant Lactobacillus spp. from sour honey distributed in Yunnan, China. To assess the safety of an entirely novel Lactobacillus pentosus strain, S4 (OM618128), based on probiotic property evaluation and whole-genome sequencing analysis. A 16S rRNA gene high-throughput sequencing analysis showed that Lactobacillus was abundant at the genus level in sour honey. Seven Lactobacillus strains (viz. S1–7) were isolated from sour honey using a multiple-anaerobic culture enrichment method. One potential acid-resistant isolate, Lactobacillus sp. S4, was obtained after screening the seven Lactobacillus isolates, and it had the highest lactic acid production (17.62 g/L), followed by Lactobacillus sp. S3 (17.07 g/L). Phylogenetic and comparative analyses of conserved sequence regions have shown that all seven strains are phylogenetically located in the Lactobacillus pentosus sub-cluster. In L. pentosus SYBC-MI, there is a circular chromosome (3288615 bps) and 11,466 bps plasmids. GC content is 44.03%. The number of predicted genes is 3,129, with 16 rRNAs and 74 tRNAs present. During the fermentation of foxtail millet by seven Lactobacillus pentosus (S1–7) strains isolated from sour honey, a potential tryptophan accumulating isolate, Lactobacillus pentosus S4, was obtained, which could reach a maximum tryptophan content of 238.43 mgL-1 that is 1.80 times the initial tryptophan content in the fermentation broth. This strain has strong acid tolerance, salt tolerance, and fermentation acid production abilities. This strain degrades nitrite at a rate of over 99%, and it has high probiotic potential as well. This project has established a solid foundation for further exploring the excellent lactic acid bacteria in sour honey. It is also investigating the key taxa and their role in the environment. According to the results of our studies, these LAB isolates provide a lot of potential for use in the future, as a source of probiotics for human, animals, and starter cultures for food applications.

Objectives: Disclosure of bad news is distressing for patients and family members. Our aim was to assess patients' perceptions and preferences regarding bad news in the health setting. Methods: Cross-sectional, multi-centered study supported by an external grant in 15 Government and Private Hospitals across Pakistan. A sample size of 1673 patients and family members was used. Ethics permission/consent was taken from each participating hospital and participant. Responses were compared across provinces, gender, age, education and income. Results: >80% patients preferred their relatives to know the diagnosis first and they wanted the news to be disclosed to them by doctors. Significant association between education level, income and preference for wanting to know the diagnosis was found. Reasons for wanting to know the diagnosis included treatment, prognosis and prevention options whereas reasons for not wanting to know included fear of emotions and God's will. Conclusion: The majority of Pakistani patients want to be informed and want the family to know first. Preferences for disclosure vary across, age, education and income level. Innovation: First countrywide study on this topic. Identifies need for culturally sensitive guidelines that include the family's role in disclosure of bad news.

Brassica napus (B. napus) is susceptible to multiple abiotic stresses that can affect plant growth and development, ultimately reducing crop yields. In the past, many genes that provide tolerance to abiotic stresses have been identified and characterized. Peroxidase (POD) proteins, members of the oxidoreductase enzyme family, play a critical role in protecting plants against abiotic stresses. This study demonstrated a comprehensive investigation of the POD gene family in B. napus. As a result, a total of 109 POD genes were identified across the 19 chromosomes and classified into five distinct subgroups. Further, 44 duplicate events were identified; of these, two gene pairs were tandem and 42 were segmental. Synteny analysis revealed that segmental duplication was more prominent than tandem duplication among POD genes. Expression pattern analysis based on the RNA-seq data of B. napus indicated that BnPOD genes were expressed differently in various tissues; most of them were expressed in roots rather than in other tissues. To validate these findings, we performed RT-qPCR analysis on ten genes; these genes showed various expression levels under abiotic stresses. Our findings not only furnish valuable insights into the evolutionary dynamics of the BnPOD gene family but also serve as a foundation for subsequent investigations into the functional roles of POD genes in B. napus.

In this paper, we studied the effect of a magnetic field on the non-isothermal second-grade fluid confined in a vertically oscillating cylinder. The flow solution is magnetized using the perpendicular magnetic field. The resultant fluid flow is due to the oscillating boundary motion and buoyancy force. Here, the MHD flow is modeled using the Caputo-Fabrizio non-integer derivative approach. The exact solution of the governing continuity, momentum and energy equations is obtained by means of Laplace and finite Hankel transforms. The commercial simulation software, Mathematica is used for calculating the roots of the Bessel function. The effects of dimensionless parameters such as Grashof and Prandtl numbers, magnetic field and fractional parameters on the second-grade fluid flow are analyzed. Heat transfer is high at a small Prandtl number. Velocity correlates positively with Grashof number and magnetic field, and negatively with Prandtl number. The heat and mass transfer results obtained from both conventional and fractional models are compared as well.

Abstract The current work analyzes the effects of concentric ballooned catheterization and heat transfer on the hybrid nano blood flow through diseased arterial segment having both stenosis and aneurysm along its boundary. A fractional second-grade fluid model is considered which describes the non-Newtonian characteristics of the blood. Governing equations are linearized under mild stenosis and mild aneurysm assumptions. Precise articulations for various important flow characteristics such as heat transfer, hemodynamic velocity, wall shear stress, and resistance impedance are attained. Graphical portrayals for the impact of the significant parameters on the flow attributes have been devised. The streamlines of blood flow have been examined as well. The present finding is useful for drug conveyance system and biomedicines.

In recent years, the bacterial blight of cassava has caused substantial economic losses to the Chinese cassava industry. Chemical control methods have become the primary approach to control this disease; however, their widespread usage and harmful residues have raised concerns about environmental pollution. In order to avoid this, it is urgent to seek a green ecological method to prevent and control it. Biological control through the utilization of microorganisms not only effectively inhibits the disease, but also gives consideration to environmental friendliness. Therefore, investigating an endophytic biological control method for cassava bacterial blight is of great importance. In this study, cassava leaf tissues were used as test specimens in order to isolate endophytic bacteria by using dilution and separation methods. Bacillus ME9, derived from cassava endophytic bacteria, exhibits good antagonism against a diverse range of pathogens, including Xpm11. Its genome consists of a series of genes encoding antibacterial lipopeptides, which may be directly related to its antibacterial capabilities. Furthermore, inoculation resulted in a substantial change in the diversity of the endophytic bacterial community, characterized by improved diversity, and displayed an obvious inhibition of pathogenic bacterial growth, demonstrating successful colonization within plants. The results laid a foundation and provided theoretical support for the development and utilization of cassava endophytic bacterial diversity and endogenous disease control strategies.

In an urban and uncontrolled environment, the presence of mixed traffic of autonomous vehicles, classical vehicles, vulnerable road users, e.g., pedestrians, and unprecedented dynamic events makes it challenging for the classical autonomous vehicle to navigate the traffic safely. Therefore, the realization of collaborative autonomous driving has the potential to improve road safety and traffic efficiency. However, an obvious challenge in this regard is how to define, model, and simulate the environment that captures the dynamics of a complex and urban environment. Therefore, in this research, we first define the dynamics of the envisioned environment, where we capture the dynamics relevant to the complex urban environment, specifically, highlighting the challenges that are unaddressed and are within the scope of collaborative autonomous driving. To this end, we model the dynamic urban environment leveraging a probabilistic graphical model (PGM). To develop the proposed solution, a realistic simulation environment is required. There are a number of simulators—CARLA (Car Learning to Act), one of the prominent ones, provides rich features and environment; however, it still fails on a few fronts, for example, it cannot fully capture the complexity of an urban environment. Moreover, the classical CARLA mainly relies on manual code and multiple conditional statements, and it provides no pre-defined way to do things automatically based on the dynamic simulation environment. Hence, there is an urgent need to extend the off-the-shelf CARLA with more sophisticated settings that can model the required dynamics. In this regard, we comprehensively design, develop, and implement an extension of a classical CARLA referred to as CARLA+ for the complex environment by integrating the PGM framework. It provides a unified framework to automate the behavior of different actors leveraging PGMs. Instead of manually catering to each condition, CARLA+ enables the user to automate the modeling of different dynamics of the environment. Therefore, to validate the proposed CARLA+, experiments with different settings are designed and conducted. The experimental results demonstrate that CARLA+ is flexible enough to allow users to model various scenarios, ranging from simple controlled models to complex models learned directly from real-world data. In the future, we plan to extend CARLA+ by allowing for more configurable parameters and more flexibility on the type of probabilistic networks and models one can choose. The open-source code of CARLA+ is made publicly available for researchers.

Autonomous vehicles and smart roads are not new concepts and the undergoing development to empower the vehicles for higher levels of automation has achieved initial milestones. However, the transportation industry and relevant research communities still require making considerable efforts to create smart and intelligent roads for autonomous driving. To achieve the results of such efforts, the CCAM infrastructure is a game changer and plays a key role in achieving higher levels of autonomous driving. In this paper, we present a smart infrastructure and autonomous driving capabilities enhanced by CCAM infrastructure. Meaning thereby, we lay down the technical requirements of the CCAM infrastructure: identify the right set of the sensory infrastructure, their interfacing, integration platform, and necessary communication interfaces to be interconnected with upstream and downstream solution components. Then, we parameterize the road and network infrastructures (and automated vehicles) to be advanced and evaluated during the research work, under the very distinct scenarios and conditions. For validation, we demonstrate the machine learning algorithms in mobility applications such as traffic flow and mobile communication demands. Consequently, we train multiple linear regression models and achieve accuracy of over 94% for predicting aforementioned demands on a daily basis. This research therefore equips the readers with relevant technical information required for enhancing CCAM infrastructure. It also encourages and guides the relevant research communities to implement the CCAM infrastructure towards creating smart and intelligent roads for autonomous driving.

Cellular vehicle-to-everything (C-V2X) is one of the enabling vehicular communication technologies gaining momentum from the standardization bodies, industry, and researchers aiming to realize fully autonomous driving and intelligent transportation systems. The 3rd Generation Partnership Project (3GPP) standardization body has actively been developing the standards evolving from 4G-V2X to 5G-V2X providing ultra-reliable low-latency communications and higher throughput to deliver the solutions for advanced C-V2X services. In this survey, we analyze the 3GPP standard documents relevant to V2X communication to present the complete vision of 3GPP-enabled C-V2X. To better equip the readers with knowledge of the topic, we describe the underlying concepts and an overview of the evolution of 3GPP C-V2X standardization. Furthermore, we provide the details of the enabling concepts for V2X support by 3GPP. In this connection, we carry out an exhaustive study of the 3GPP standard documents and provide a logical taxonomy of C-V2X related 3GPP standard documents divided into three categories: 4G, 4G & 5G, and 5G based V2X services. We provide a detailed analysis of these categories discussing the system architecture, network support, key issues, and potential solution approaches supported by the 3GPP. We also highlight the gap and the need for intelligence in the execution of different operations to enable the use-case scenarios of Level-5 autonomous driving. We believe, the paper will equip readers to comprehend the technological standards for the delivery of different ITS services of the higher level of autonomous driving.

Etiological patterns of uropathogens are different in different geographical regions due to the continuous evolution of bacteria, antibiotic sensitivity patterns and their misuse and overuse. Therefore, it is important to know the antibiotic susceptibility patterns for the prescription of suitable antibiotics. This study was conducted to determine the prevalence of uropathogens and their antimicrobial sensitivity patterns in the Kohat region of Pakistan. In this study, 100 samples were collected from both male and female subjects of all ages. Out of these 100 samples, 70 samples contained microbes. In 30 samples, no microbial growth was recorded. The percentages of positive culture from male and female subjects were 57% and 43%, respectively. Both Gram (+) and Gram (-) bacteria were found in UTI. Among them E.coli (34.21%) was predominant, followed by K. pneumoniae (10.52%), P. aeruginosa (9.21%), K. oxytoca (6.57%), C. albicans (5.26%), E. faecium (5.26%), E. faecalis (3.94%), S. aureus (3.94%), E. cloacea (2.63%), C. freundii (2.63%), P. mirabalis (2.63%) and A. baumannii (1.31%). Many of the isolates showed resistance to commonly used antibiotics. The sensitivity percentage of commonly used antibiotics against both Gram (+) and Gram (-) bacteria are as follows: Ampicillin (13%), Ceftriaxone (25%), Amikacin (77%), Gentamicin (41%), Augmentin (44.77%), Fosfomycin (64%), Cotrimoxazole (36%), Nitrofurantoin (68%), Ciprofloxacin (37%), Imipenem (78%), Meropenem (67%), Cefepime (25%) and Tetracycline (40%). The most effective antibiotics against both Gram (+) and Gram (-) bacteria were Fosfomycin, Imipenem, Meropenem Amikacin and Nitrofurantoin. In light of the findings of this study, it is strongly recommended to find new antimicrobial compounds. Moreover, it is imperative to evaluate the resistant patterns at genomic and proteomic levels to discover the genes responsible for antibiotic resistant patterns. The advancement in sensor technologies, mobile network technologies, and artificial intelligence has pushed the boundaries of different verticals, e.g., eHealth and autonomous driving. Statistics show that more than one million people are killed in traffic accidents yearly, where the vast majority of the accidents are caused by human negligence. Higher-level autonomous driving has great potential to enhance road safety and traffic efficiency. One of the most crucial links to building an autonomous system is the task of decision-making. The ability of a vehicle to make robust decisions on its own by anticipating and evaluating future outcomes is what makes it intelligent. Planning and decision-making technology in autonomous driving becomes even more challenging, due to the diversity of the dynamic environments the vehicle operates in, the uncertainty in the sensor information, and the complex interaction with other road participants. A significant amount of research has been carried out toward deploying autonomous vehicles to solve plenty of issues, however, how to deal with the high-level decision-making in a complex, uncertain, and urban environment is a comparatively less explored area. This paper provides an analysis of decision-making solutions approaches for autonomous driving. Various categories of approaches are analyzed with a comparison to classical decision-making approaches. Following, a crucial range of research gaps and open challenges have been highlighted that need to be addressed before higher-level autonomous vehicles hit the roads. We believe this survey will contribute to the research of decision-making methods for autonomous vehicles in the future by equipping the researchers with an overview of decision-making technology, its potential solution approaches, and challenges.

Dengue has become endemic in Pakistan with annual recurrence. A sudden increase in the dengue cases was reported from Rawalpindi in 2016, while an outbreak occurred for the first time in Peshawar in 2017. Therefore, a multi-center study was carried out to determine the circulating dengue virus (DENV) serotypes and Chikungunya virus (CHIKV) co-infection in Lahore, Rawalpindi, and Peshawar cities in 2016-18. A hospital-based cross-sectional study was carried out in Lahore and Rawalpindi in 2016-18, while a community-based study was carried out in Peshawar in 2017. The study participants were tested for dengue NS1 antigen using an immunochromatographic device while anti-dengue IgM/IgG antibodies were detected by indirect ELISA. All NS1 positive samples were used for DENV serotyping using multiplex real-time PCR assay. Additionally, dengue samples were tested for CHIKV co-infection using IgM/IgG ELISA. A total of 6291 samples were collected among which 8.11% were NS1 positive while 2.5% were PCR positive. DENV-2 was the most common serotype (75.5%) detected, followed by DENV-1 in 16.1%, DENV-3 in 3.9% and DENV-4 in 0.7% while DENV-1 and DENV-4 concurrent infections were detected in 3.9% samples. The present study explored chemical constituents of Scutellaria edelbergii essential oils (SEEO) for the first time, extracted through hydro-distillation, and screened them against the microbes and free radicals scavenging effect, pain-relieving, and anti-inflammatory potential employing standard techniques. The SEEO ingredients were noticed via Gas Chromatography-Mass-Spectrometry (GC-MS) analysis and presented fifty-two bioactive compounds contributed (89.52%) with dominant volatile constituent; 3-oxomanoyl oxide (10.09%), 24-norursa-3,12-diene (8.05%), and methyl 7-abieten-18-oate (7.02%). The MTT assay via 96 well-plate and agar-well diffusion techniques against various microbes was determined for minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC), IC50, and zone of inhibitions (ZOIs). The SEEO indicated considerable antimicrobial significance against tested bacterial strains viz. Escherichia coli, Pseudomonas aeruginosa, Klebsiella pneumoniae, and Enterococcus faecalis and the fungal strains Fusarium oxysporum and Candida albicans. The free radicals scavenging potential was noticed to be significant in 1,1-Diphenyl-2-picryl-hydrazyl (DPPH) as compared to 2,2′-azino-bis-3-ethylbenzotiazolin-6-sulfonic acid (ABTS) assays with IC50 = 125.0 ± 0.19 µg/mL and IC50 = 153.0 ± 0.31 µg/mL correspondingly; similarly, the antioxidant standard in the DPPH assay was found efficient as compared to ABTS assay. The SEEO also offered an appreciable analgesic significance and presented 54.71% in comparison with standard aspirin, 64.49% reduction in writhes, and an anti-inflammatory potential of 64.13%, as compared to the standard diclofenac sodium inhibition of 71.72%. The SEEO contain bioactive volatile ingredients with antimicrobial, free radical scavenging, pain, and inflammation relieving potentials. Computational analysis validated the anti-inflammatory potential of selected hit “methyl 7-abieten-18-oate” as a COX-2 enzyme inhibitor. Docking results were very good in terms of docked score (−7.8704 kcal/mol) and binding interactions with the functional residues; furthermore, MD simulation for 100 ns has presented a correlation with docking results with minor fluctuations. In silico, ADMET characteristics supported that methyl 7-abieten-18-oate could be recommended for further investigations in clinical tests and could prove its medicinal status as an anti-inflammatory drug.

The present analysis explores the chemical constituents and determines the in vitro antimicrobial, antidiabetic, and antioxidant significance of the essential oils (EOs) of the stem, leaves, and flowers of Ochradenus arabicus for the first time. The EOs of the flowers presented seventy-four constituents contributing to 81.46% of the total EOs, with the major compounds being 24-norursa-3,12-diene (13.06%), 24-norursa-3,12-dien-11-one (6.61%), and 24-noroleana-3,12-diene (6.25%). The stem EOs with sixty-one compounds contributed 95.95% of the total oil, whose main bioactive compounds were (+)-camphene (21.50%), eremophilene (5.87%), and δ-selinene (5.03%), while a minimum of fifty-one compounds in the leaves’ EOs (98.75%) were found, with the main constituents being n-hexadecanoic acid (12.32%), octacosane (8.62%), tetradecanoic acid (8.54%), and prehydro fersenyl acetone (7.27%). The antimicrobial activity of the EOs of O. arabicus stem, leaves, and flowers was assessed against two bacterial strains (Escherichia coli and Streptococcus aureus) and two fungal strains (Penicillium simplicissimum and Rhizoctonia solani) via the disc diffusion assay. However, the EOs extracted from the stem were found effective against one bacterial strain, E. coli, and one fungal strain, R. Solani, among the examined microbes in comparison to the standard and negative control. The tested EOs samples of the O. arabicus stem displayed a maximum potential to cure diabetes with an IC50 = 0.40 ± 0.10 µg/mL, followed by leaves and flowers with an IC50 = 0.71 ± 0.11 µg/mL and IC50 = 10.57 ± 0.18 µg/mL, respectively, as compared to the standard acarbose (IC50 = 377.26 ± 1.20 µg/mL). In addition, the EOs of O. arabicus flowers had the highest antioxidant activity (IC50 = 106.40 ± 0.19 µg/mL) as compared to the standard ascorbic acid (IC50 = 73.20 ± 0.17 µg/mL) using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay. In the ABTS assay, the EOs of the same sample (flower) depicted the utmost potential to scavenge the free radicals with an IC50 = 178.0 ± 0.14 µg/mL as compared with the ascorbic acid, having an IC50 of 87.34 ± 0.10 µg/mL the using 2,2-Azino-Bis-3-Ethylbenzothiazoline-6-Sulfonic acid (ABTS) assay. The EOs of all parts of O. arabicus have useful bioactive components due to which they present antidiabetic and antioxidant significance. Furthermore, additional investigations are considered necessary to expose the responsible components of the examined biological capabilities, which would be effective in the production of innovative drugs.

Scutellaria (Lamiaceae) comprises over 360 species. Based on its morphological structure of calyx, also known as Skullcap, it is herbaceous by habit and cosmopolitan by habitat. The species of Scutellaria are widely used in local communities as a natural remedy. The genus contributed over three hundred bioactive compounds mainly represented by flavonoids and phenols, chemical ingredients which serve as potential candidates for the therapy of various biological activities. Thus, the current review is an attempt to highlight the biological significance and its correlation to various isolated bioactive ingredients including flavonoids, terpenoids, phenols, alkaloids, and steroids. However, flavonoids were the dominant group observed. The findings of the Scutellaria reveal that due to its affluent basis of numerous chemical ingredients it has a diverse range of pharmacological potentials, such as antimicrobial, antioxidant, antifeedant, enzyme inhibition, anti-inflammatory, and analgesic significance. Currently, various bioactive ingredients have been investigated for various biological activities from the genus Scutellaria in vitro and in vivo. Furthermore, these data help us to highlight its biomedical application and to isolate the responsible compounds to produce innovative medications as an alternative to synthetic drugs.

A new series of (S)-flurbiprofen derivatives 4a–4p and 5a–5n were synthesized with different aromatic or aliphatic aldehydes and ketones to produce Schiff’s bases and their structures were confirmed through HR-ESI-MS, 1H, and 13C-NMR spectroscopy. The α-glucosidase inhibitory activities of the newly synthesized compounds were scrutinized, in which six compounds 5k, 4h, 5h, 4d, 4b, and 5i showed potent inhibition in the range of 0.93 to 10.26 µM, respectively, whereas fifteen compounds 4c, 4g, 4i, 4j, 4l, 4m, 4o, 4p, 5c, 5d, 5j, 5l, 5m, 5n and 1 exhibited significant inhibitory activity with IC50 in range of = 11.42 to 48.39 µM. In addition, compounds 5g, 5f, 4k, 4n, and 4f displayed moderate-to-low activities. The modes of binding of all the active compounds were determined through the molecular docking approach, which revealed that two residues, specifically Glu277 and His351 are important in the stabilization of the active compounds in the active site of α-glucosidase. Furthermore, these compounds block the active site with high binding energies (−7.51 to −3.36 kcal/mol) thereby inhibiting the function of the enzyme.

The recent study investigated the in vitro anti-diabetic impact of the crude extract (MeOH) and subfractions ethyl acetate (EtOAc); chloroform; n-butanol; n-hexane; and aqueous fraction of S. edelbergii and processed the active EtOAc fraction for the identification of chemical constituents for the first time via ESI-LC-MS analysis through positive ionization mode (PIM) and negative ionization mode (NIM); the identified compounds were further validated through computational analysis via standard approaches. The crude extract and subfractions presented appreciable activity against the α-glucosidase inhibitory assay. However, the EtOAc fraction with IC50 = 0.14 ± 0.06 µg/mL revealed the maximum potential among the fractions used, followed by the MeOH and n-hexane extract with IC50 = 1.47 ± 0.14 and 2.18 ± 0.30 µg/mL, respectively. Moreover, the acarbose showed an IC50 = 377.26 ± 1.20 µg/ mL whereas the least inhibition was observed for the chloroform fraction, with an IC50 = 23.97 ± 0.14 µg/mL. Due to the significance of the EtOAc fraction, when profiled for its chemical constituents, it presented 16 compounds among which the flavonoid class was dominant, and offered eight compounds, of which six were identified in NIM, and two compounds in PIM. Moreover, five terpenoids were identified—three and two in NIM and PIM, respectively—as well as two alkaloids, both of which were detected in PIM. The EtOAc fraction also contained one phenol that was noticed in PIM. The detected flavonoids, terpenoids, alkaloids, and phenols are well-known for their diverse biomedical applications. The potent EtOAc fraction was submitted to computational analysis for further validation of α-glucosidase significance to profile the responsible compounds. The pharmacokinetic estimations and protein-ligand molecular docking results with the support of molecular dynamic simulation trajectories at 100 ns suggested that two bioactive compounds—dihydrocatalpol and leucosceptoside A—from the EtOAc fraction presented excellent drug-like properties and stable conformations; hence, these bioactive compounds could be potential inhibitors of alpha-glucosidase enzyme based on intermolecular interactions with significant residues, docking score, and binding free energy estimation. The stated findings reflect that S. edelbergii is a rich source of bioactive compounds offering potential cures for diabetes mellitus; in particular, dihydrocatalpol and leucosceptoside A could be excellent therapeutic options for the progress of novel drugs to overcome diabetes mellitus.

Pterostilbene (PTS) in blueberries is a phytoalexin with antioxidant properties. PTS exerts strong cytoprotective effects on various cells via Nuclear Factor Erythroid 2 like 2 (NFE2L2) pathway. We evaluated the antioxidant PTS treatment in mouse preimplantation embryos. In vitro culture media were supplemented with different concentrations of PTS. Treatment of zygotes with 0.25 μM PTS improved the development of day 4 blastocysts (P < 0.05). Moreover, H2O2 treatment significantly increased the reactive oxygen species level and reduced the glutathione level in mouse blastocyst, whereas PTS treatment counteracted these effects. The fluorescence intensity of apoptotic positive cell was higher in the H2O2 group than in the PTS group. Furthermore, PTS-treated embryos significantly increased the protein expression of NFE2L2 in the nucleus and decreased Kelch-like ECH-associated protein1 (KEAP1). PTS treatment significantly increased the expression of downstream target genes involved in the NFE2L2 pathway, such as catalase (CAT), heme oxygenase1 (HMOX1), glutathione peroxidase (GPX), and superoxide dismutase (SOD); these genes confer cellular protection. In addition, PTS treatment significantly increased the expression of anti-apoptotic B-cell lymphoma 2 (BCL2), with a concomitant reduction in the apoptotic Bcl-2-associated X protein (BAX) and Caspase-3 genes in the embryo. PTS treatment also increased the protein expression of BCL2 and reduced the protein expression of BAX in the mouse embryo. In conclusion, PTS activated NFE2L2 signaling pathway in the development of mouse embryos by altering downstream expression of genes involved in the antioxidant mechanisms and apoptosis.

The current study aimed to rationally develop and characterize pH-sensitive controlled release hydrogels by graft polymerization of gelatin (Gel) and hydroxypropyl methyl cellulose (HPMC) in the presence of glutaraldehyde (GA) using quetiapine fumarate for the treatment of schizophrenia. The prepared hydrogels discs were subjected to various physicochemical studies including: swelling, diffusion, porosity, sol-gel analysis, Fourier transform infrared spectroscopy, differential scanning calorimetry, and scanning electron microscopy. Three different pH values (1.2, 6.8 and 7.4) were used to determine shape, transition, and controlled release behavior of prepared hydrogels. The current study aimed to explore the crude oils obtained from the n-hexane fraction of Scutellaria edelbergii and further analyzed, for the first time, for their chemical composition, in vitro antibacterial, antifungal, antioxidant, antidiabetic, and in vivo anti-inflammatory, and analgesic activities. For the phytochemical composition, the oils proceeded to gas chromatography-mass spectrometry (GC-MS) analysis and from the resultant chromatogram, 42 bioactive constituents were identified. Among them, the major components were linoleic acid ethyl ester (19.67%) followed by ethyl oleate (18.45%), linolenic acid methyl ester (11.67%), and palmitic acid ethyl ester (11.01%). Tetrazolium 96-well plate MTT assay and agar-well diffusion methods were used to evaluate the isolated oil for its minimum inhibitory concentrations (MIC), minimum bactericidal concentration (MBC), half-maximal inhibitory concentrations (IC50), and zone of inhibitions that could determine the potential antimicrobial efficacy’s. Substantial antibacterial activities were observed against the clinical isolates comprising of three Gram-negative bacteria, viz., Escherichia coli, Klebsiella pneumoniae, and Pseudomonas aeruginosa, and one Gram-positive bacterial strain, Enterococcus faecalis. The oils were also effective against Candida albicans and Fusarium oxysporum when evaluated for their antifungal potential. Moreover, significant antioxidant potential with IC50 values of 136.4 and 161.5 µg/mL for extracted oil was evaluated through DPPH (1,1-Diphenyl-2-picryl-hydrazyl) and ABTS assays compared with standard ascorbic acid where the IC50 values were 44.49 and 67.78 µg/mL, respectively, against the tested free radicals. The oils was also potent, inhibiting the α-glucosidase (IC50 5.45 ± 0.42 µg/mL) enzyme compared to the standard. Anti-glucosidase potential was visualized through molecular docking simulations where ten compounds of the oil were found to be the leading inhibitors of the selected enzyme based on interactions, binding energy, and binding affinity. The oil was found to be an effective anti-inflammatory (61%) agent compared with diclofenac sodium (70.92%) via the carrageenan-induced assay. An appreciable (48.28%) analgesic activity in correlation with the standard aspirin was observed through the acetic acid-induced writhing bioassay. The oil from the n-hexane fraction of S. edelbergii contained valuable bioactive constituents that can act as in vitro biological and in vivo pharmacological agents. However, further studies are needed to uncover individual responsible compounds of the observed biological potentials which would be helpful in devising novel drugs.

In present article, hydromagnetic transport of dust particles in gas flow over an inclined plane is examined under the influence of thermal radiation and viscous dissipation. The Rosseland diffusion flux model is utilized for thermal radiation effects. The resulting non-linear ordinary differential equations are converted into similar forms via suitable similarity transforms and then tackled numerically by bvp4c built in code of Matlab software. The influences of numerous emergent parameters on flow features like velocities, temperature distributions and Nusselt number are depicted graphically and discussed in detail. Among the diverse clinical manifestations of Disorders of Sex Development (DSD), the ambiguous look of genitalia is the most typical disorder. In this condition, the sex of the newborn cannot be readily differentiated because of atypical appearance of the external genitalia. Thus, the treatment of such a child is composite from both sociocultural and medical viewpoints. Decisions that affect the future sexual function and sex assignment of the child are made on behalf of the child by their parents and healthcare professionals. Such decisions are rarely simple and straightforward. The outcome data resulting from such decisions is limited and conflicting, and cannot offer clear guidelines to the community. This case presents a 20-year-old patient who presented with primary amenorrhea and delayed pubertal features, and was diagnosed as 46, XY typical male genotype. As she had presented with a female phenotype at birth, she had been raised as a female and developed a consistently female sex identity. She underwent surgery, during which remnant testes were removed, and bilateral breast augmentation and clitoroplasty were performed. This decision to be rehabilitated as a girl was made by the patient herself. In this part of the world, sexual variation that blurs the line between male and female is stigmatized. The birth of a child with DSD is emotionally traumatizing and considered a social stigma and taboo. Therefore, social familiarity regarding this treatable disorder is currently needed in Pakistan irrespective of the cultural, social, and religious beliefs, and to protect the society from the ambiguous obscurity of the unthinkable of what was earlier called “intersex disorders.”

In this study, the transport of Al 2 O 3 nanoparticles in ethylene glycol conventional fluid over a linearly stretching cylinder is investigated. The current research employs a convective surface boundary condition for heat transfer exploration. Flux model proposed by Rosseland is employed to examine effect of thermal radiations. The governing flow problem comprises highly nonlinear ordinary differential equations. Similarity transformations are used to reduce the equations in similar forms, which are then solved by Runge–Kutta–Fehlberg fourth-fifth order numerical scheme with shooting algorithm in MATLAB software. In order to authenticate the accuracy of our results, we have contrasted results with those obtained by Ishak et al., Wang, and Pandey and Kumar and found that they are in better concord, as revealed in Table 2. The impact of numerous emerging parameters on velocity distribution and heat transfer distribution are argued in all aspects and depicted through graphs.

BACKGROUND:Drug resistant tuberculosis (DR-TB) is a major public health problem in developing countries such as Pakistan. OBJECTIVE:The current study was conducted to assess the frequency of drug resistant tuberculosis including multi drug resistance (MDR- TB) as well as risk factors for development of DR-TB, in Punjab, Pakistan. METHODOLOGY:Drug susceptibility testing (DST) was performed, using proportion method, for 2367 culture positive Mycobacterium tuberculosis (MTB) cases that were enrolled from January 2012 to December 2013 in the province of Punjab, Pakistan, against first-line anti-tuberculosis drugs. The data was analyzed using statistical software; SPSS version 18. RESULTS:Out of 2367 isolates, 273 (11.5%) were resistant to at least one anti-TB drug, while 221 (9.3%) showed MDR- TB. Risk factors for development of MDR-TB were early age (ranges between 10-25 years) and previously treated TB patients. CONCLUSION:DR-TB is a considerable problem in Pakistan. Major risk factors are previous history of TB treatment and younger age group. It emphasizes the need for effective TB control Program in the country.

Objectives: To determine the frequency of different types of cleft lip and palate, geographical distribution and its association with consanguinity, family history and other syndromes in the Northern Pakistani population. Study design: Descriptive cross-sectional study. Settings: This study was performed in Plastic and Reconstructive units of Hayat Medical Complex Peshawar Pakistan, Aman Hospital Peshawar Pakistan and Abasin Hospital Peshawar, Pakistan from November 2010 to December 2011. Material and Methods: All the patients, irrespective of their gender, presenting with cleft lip/palate deformities were included in the study. Previously operated cases and those who did not agree with the study protocol were excluded from the study. The data were collected through questionnaires as well as history and physical examination. The data were organized and analyzed through Statistical Package for Social Sciences version 17 (SPSS 17). The results were expressed in the form of tables and figures. Results: A total of 159 patients of cleft lip and cleft palate deformities were included in the study, having a mean age of 3.5+6.59 years and containing 59.1% males and 40.9% females with a ratio of 1.4:1. A cleft lip with palate, cleft palate and cleft lip were found in 51.6%, 31.4% and 17% of cases, respectively. Left-sided clefts were most common in the cleft lip with palate and the isolated cleft lip deformity. A cleft lip with palate was a male dominant variety (62.8% of cases), while in the cleft palate variety, the dominant gender was female. In 61.6% of cases, the parent had a consanguineous relationship. In 21.4% of cases, family history was positive for the cleft lip/palate. Other congenital anomalies were associated in 10.7% of cases. Conclusion: Cleft deformities of the lip and palate affect the male population more than females with cleft lips, in association with a cleft palate being the most common anomaly. Females are mainly affected by an isolated cleft palate. The high prevalence of these deformities in consanguineous marriages emphasizes educating people. This paper looks at the effects of radiative heat transfer on the peristaltic transport of a Sisko fluid in an asymmetric channel with nonuniform wall temperatures. Adopting the lubrication theory, highly nonlinear coupled governing equations involving power law index as an exponent have been linearized and perturbation solutions are obtained about the Sisko fluid parameter. Analytical solutions for the stream function, axial pressure gradient, axial velocity, skin friction, and Nusselt number are derived for three different cases (i.e., shear thinning fluid, viscous fluid, and shear thickening fluid). The effects of Grashof number, radiation parameter, and other configuration parameters on pumping, trapping, temperature, Nusselt number, and skin friction have been examined in detail. A good agreement has been found for the case of viscous fluid with existing results.
