Background: After three years with no local transmission of malaria, an outbreak occurred in Iran in 2022. Key malaria control methods in Iran are including indoor residual spraying (IRS), long-lasting insecticide-treated nets (LLINs), and prompt diagnosis and treatment of malaria cases. Anopheles sacharovi is one of the main malaria vectors in Iran. This study aimed to determine the insecticides resistance status of An. sacharovi in northwestern Iran, to inform effective vector control programs in this region. Methods: Larval stages of An. sacharovi were collected from various larval habitats located in the villages along the Aras River. Adult susceptibility tests were performed on An. sacharovi using diagnostic doses of insecticides accordance to World Health Organization (WHO) guidelines. The study also evaluated agricultural insecticide and fertilizer usage alongside the presence of natural mosquito predators in breeding sites in the study area. Results: Alongside various chemicals such as silica, humic acid, superphosphate, sulfur, urea, and solupotasse at different dose levels, organophosphorus and pyrethroid insecticides are commonly used in rice fields and orchards. Anopheles sacharovi displayed diverse reactions to insecticides, demonstrating resistance to DDT but sensitivity to malathion, and showing similar reactions to carbamate and pyrethroid insecticides. Conclusions: These results provide significant insights into agricultural practices and the presence of mosquito larvae in the study area. The extensive use of a specific herbicide illustrates its popularity among farmers for weed control, while other agricultural products focus on enhancing soil fertility and productivity. The absence of mosquito larvae in habitats with predators indicates the usefulness of these predators in controlling the population of mosquitoes. The resistance of mosquitoes to certain insecticides highlights the need for careful selection and intermittent use of insecticides in vector control programs. These findings can inform the development of targeted strategies to reduce malaria transmission risks. Further research is essential for assessing the effectiveness of these interventions. The functional and structural features of microbial load in soil are influenced by the presence of insecticides. This study examined the impact of two organophosphorus insecticides, dimethoate and parathion on the microbial load of soil. The colony count of fungi, actinomycetes, bacteria, and nitrogen fixing bacteria was reduced after insecticides application at 7 and 14 days, but at 28 days the colony count began to increase. The growth of two white rot fungi including Pleurotus sajor-caju and Phanerochaete chrysosporium was affected by parathion, which was reflected by a decrease in colony radius to 1.85±0.05 and 0.75±0.06 cm, respectively, and by dimethoate, reflected by a decrease in colony radius to 3.33±0.12 and 1.85±0.05 cm, respectively at 40 mg/L compared to colony radius at control 7.90±0.12 and 7.50±0.06 cm, respectively. The applied low concentration (10 mg/L) encouraged P. sajor-caju and P. chrysosporium to remove up to 87.7% and 81.8% of dimethoate, respectively, and 69.20 and 68.30% of parathion, respectively compared with the decomposition at high dose (40 mg/L) at 28 days. Simple Summary: Carbamate insecticides like fenobucarb have been used in the Republic of Korea since 1981 to control brown planthopper (BPH). However, their excessive use has resulted in increased resistance in BPH Nilaparvata lugens. Therefore, in our current study, we aimed to investigate the main cause of higher resistance to fenobucarb in BPH19 compared to BPH80 and BPH15. We examined mutations in the acetylcholinesterase 1 gene, the target gene of carbamate, but did not find any previously reported mutations. However, through RNA-seq analysis, we discovered a strong correlation between the level of resistance development and the expression of the N. lugens carboxylesterase 1, Nl-EST1 gene in BPH19 compared to BPH80 and BPH15. This suggests that Nl-EST1 could be utilized as an expression marker for diagnosing resistance and integrated resistance management of N. lugens. Carbamate insecticides have been used for over four decades to control brown planthopper, Nilaparvata lugens, but resistance has been reported in many countries, including the Republic of Korea. The bioassay results on resistance to fenobucarb showed that the LC50 values were 3.08 for the susceptible strain, 10.06 for the 2015 strain, and 73.98 mg/L for the 2019 strain. Compared to the susceptible strain, the 2015 and 2019 strains exhibited resistance levels 3.27 and 24.02 times higher, respectively. To elucidate the reason for the varying levels of resistance to fenobucarb in these strains, mutations in the acetylcholinesterase 1 (ACE1) gene, the target gene of carbamate, were investigated, but no previously reported mutations were confirmed. Through RNA-seq analysis focusing on the expression of detoxification enzyme genes as an alternative resistance mechanism, it was found that the carboxylesterase gene Nl-EST1 was overexpressed 2.4 times in the 2015 strain and 4.7 times in the 2019 strain compared to the susceptible strain. This indicates a strong correlation between the level of resistance development in each strain and the expression level of Nl-EST1. Previously, Nl-EST1 was reported in an organophosphorus insecticide-resistant strain of Sri Lanka 2000. Thus, Nl-EST1 is crucial for developing resistance to organophosphorus and carbamate insecticides. Organophosphorus and carbamate Insectiside is common in Asia–Pacific region consisting of 63% of the Global death. Organophosphorus and Carbamate poisoning can lead into different complications in the respiratory,digestive,neurological aspects and maybe fatal in certain cases.Besides medical management of the same,early rehabilitation is also required to manage different kinds of neurological aspects caused due to Organophosphorus and carbamate poisoning. The study was done to introspect the early intervention in swallowing and speech therapy in organophosphorus and carbamate poisoning individual,as a part of management besides medical intervention. A 19 years old female reported to the hospital with history of consuming organophos and carbamate insecticide with the intention of self harming causing cardiac arrest and as diagnosed as Flaccid Dysarthria with Oro-Pharyngeal Dysphagia when assessed with diagnostic tools respectively. Early speech and swallowing intervention was provided and introspected using MASA which showed improvement in scores during the therapeutic intervention and was statistically analysed using linear regression analysis. The result showed the improvement in MASA scores (the slope of the best fit) and proved that MASA scores improved significantly (slope = 14.3, p < 0.05) over time as the therapy sessions proceeded. The regression model was also significant (p < 0.05). The motor-speech therapeutic intervention provided improvement in the kinematics of oro-motor skills along with improvement in intelligibility of speech. This study concludes that early intervention in managing speech and swallowing abilities in Dysarthria and Dysphagia is helpful besides medical intervention in such cases. [ABSTRACT FROM AUTHOR]

The use of some organophosphate insecticides is restricted or even banned in paddy fields due to their high toxicity to aquatic organisms. The aim of this study is to elucidate the main pathways and target organs of organophosphate insecticide toxicity to fish exposed via different routes by integrating histopathological and biochemical techniques. Using malathion as the model drug, when the dosage is 20–60 mg/L, the toxicity of whole body and head immersion drugs to zebrafish is much higher than that of trunk immersion drugs. A dose of 21.06–190.44 mg/kg of malathion feed was fed to adult zebrafish. Although the dosage was already high, no obvious toxicity was observed. Therefore, we believe that the drug mainly enters the fish body through the gills. When exposed to a drug solution of 20 mg/L and 60 mg/L, the fish showed significant neurological behavioral abnormalities, and the pathological damage to key organs and brain tissue was the most severe, showing obvious vacuolization and the highest residual amount (8.72–47.78 mg/L). The activity of acetylcholinesterase was the most inhibited (54.69–74.68%). Therefore, brain tissue is the key toxic target organ of malathion in fish. In addition, we compared the bioaccumulation effects of different water-soluble organophosphorus insecticides in fish and their toxic effects. Abstract Background Organic phosphorus insecticides (OPPs) are a class of environmental pollutants widely used worldwide with potential human health risks. We aimed to assess the association between exposure to OPPs and osteoarthritis (OA) particularly in participants with atherosclerotic cardiovascular disease (ASCVD). Methods Participants’ information was obtained from data in the National Health and Nutrition Examination (NHANES). Weighted logistic regression models were utilized to detect associations between OPPs metabolites and OA. Restricted cubic spline plots (RCS) were drawn to visualize the dose-response relationship between each metabolite and OA prevalence. Weighted quantile sum (WQS) regression and Bayesian kernel-machine regression (BKMR), were applied to investigate the joint effect of mixtures of OPPs on OA. Results A total of 6871 samples were included in our study, no significant associations between OPPs exposure and OA incidence were found in whole population. However, in a subset of 475 individuals with ASCVD, significant associations between DMP (odds ratio [OR] as a continuous variable = 1.22, 95% confidence interval [CI]: 1.07,1.28), DEP ((odds ratio [OR] of the highest tertile compared to the lowest = 2.43, 95% confidence interval [CI]: 1.21,4.86), and OA were observed. DMP and DEP showed an increasing dose-response relationship to the prevalence of OA, while DMTP, DETP, DMDTP and DEDTP showed a nonlinear relationship. Multi-contamination modeling revealed a 1.34-fold (95% confidence intervals:0.80, 2.26) higher prevalence of OA in participants with high co-exposure to OPPs compared to those with low co-exposure, with a preponderant weighting (0.87) for the dimethyl dialkyl phosphate metabolites (DMAPs). The BKMR also showed that co-exposure of mixed OPPs was associated with an increased prevalence of OA, with DMP showing a significant dose-response relationship. Conclusion High levels of urine dialkyl phosphate metabolites (DAP) of multiple OPPs are associated with an increased prevalence of OA in patients with ASCVD, suggesting the need to prevent exposure to OPPs in ASCVD patients to avoid triggering OA and further avoid the occurrence of cardiovascular events caused by OA.

Mining of organophosphorous (OPs)-degrading bacterial enzymes in collections of known bacterial strains and in natural biotopes are important research fields that lead to the isolation of novel OP-degrading enzymes. Then, implementation of strategies and methods of protein engineering and nanobiotechnology allow large-scale production of enzymes, displaying improved catalytic properties for medical uses and protection of the environment. For medical applications, the enzyme formulations must be stable in the bloodstream and upon storage and not susceptible to induce iatrogenic effects. This, in particular, includes the nanoencapsulation of bioscavengers of bacterial origin. In the application field of bioremediation, these enzymes play a crucial role in environmental cleanup by initiating the degradation of OPs, such as pesticides, in contaminated environments. In microbial cell configuration, these enzymes can break down chemical bonds of OPs and usually convert them into less toxic metabolites through a biotransformation process or contribute to their complete mineralization. In their purified state, they exhibit higher pollutant degradation efficiencies and the ability to operate under different environmental conditions. Thus, this review provides a clear overview of the current knowledge about applications of OP-reacting enzymes. It presents research works focusing on the use of these enzymes in various bioremediation strategies to mitigate environmental pollution and in medicine as alternative therapeutic means against OP poisoning. Introduction: Sarcopenia and diabetes mellitus (DM) have been shown to be related. It has been demonstrated that pesticides/insecticides are linked to various health issues, including DM. This study investigated the relationships between exposure to pesticides/insecticides and muscle strength among community-dwelling DM patients in a national sample of the United States (US). Methods: Data from the 2011–2012 and 2013–2014 U.S. National Health and Nutrition Examination Survey (NHANES) on people aged 20 years with diabetes were retrieved. A digital dynamometer was used to quantify handgrip strength, and urine pesticide concentrations were determined through laboratory testing. Regression models were used to investigate the relationship between pesticide/insecticide exposure and handgrip strength. Results: After weighting, the data from 412 NHANES participants represented 6,696,865 U.S. inhabitants. The mean age of the participants was 58.8 years. High para-nitrophenol levels (tertile 3 vs. tertile 1) were shown to be associated with lower handgrip strength in both males (aBeta = −7.25, 95% CI: −11.25, −3.25) and females (aBeta = −3.73, 95% CI: −6.89, −0.56). Further, females with elevated 2-isopropyl−4-methyl-pyrimidinol had decreased handgrip strength. Desethyl hydroxy N, N-diethyl-m-toluamide (DEET) was inversely related to handgrip strength in men aged ≥60 years. DEET acid and para-nitrophenol were inversely correlated to handgrip strength in women over 60 years. Conclusions: This study has linked certain pesticides/insecticides to decreased muscle strength in people with diabetes. Para-nitrophenol, in particular, is negatively related to muscular strength in both males and females, and 2-isopropyl-4-methyl-pyrimidinol is inversely related to muscle strength in females. Extensive use of pesticides in agricultural production has been causing serious health threats to humans and animals. Among them, phorate is a highly toxic organophosphorus insecticide that has been widely used in planting. Due to its harmful effects on human and animal health, it has been restricted for use in many countries. Analytical methods for the rapid and sensitive detection of phorate residues in agricultural products are urgently needed. In this study, a new method was developed by combining surface-enhanced Raman spectroscopy (SERS) and immunochromatography assay (ICA). Hybrid magnetic Fe3O4@Au@DTNB-Ab nanoprobes were prepared by modifying and growing Au nanoseeds on an Fe3O4 core. SERS activity of the nanoprobe was optimized by adjusting the concentration of the Au precursor. A rapid and sensitive assay was established by replacing the traditional colloidal gold-based ICA with hybrid SERS nanoprobes for SERS-ICA. After optimizing parameters including coating antibody concentrations and the composition and pH of the buffer solution, the limit of detection (LOD) for phorate could reach 1 ng/mL, with a linear range of 5~100 ng/mL. This LOD is remarkably lower than the maximum residue limit in vegetables and fruits set by the Chinese government. The feasibility of this method was further examined by conducting a spiking test with celery as the real sample. Sitophilus oryzae and Sitophilus zeamais are among the main pests of stored grains worldwide, they are very similar species and have great destructive potential, causing numerous damages to grain production. The control of these insects occurs, in general, through chemical insecticides such as the agrochemicals pirimiphos-methyl, an organophosphate, and bifenthrin, a pyrethroid, which are widely used to prevent infestation. Four strains of these two species were exposed to the two classes of agrochemicals mentioned. The polyacrylamide gel electrophoresis technique was used to identify the esterase patterns of the species and eight esterases were identified for S. zeamais, of which four cholinesterases, two carboxylesterases, one acetylesterase, and one arylesterase; and seven esterases for S. oryzae, being four cholinesterases, two carboxylesterases, and one acetylesterase. After exposure to agrochemicals, through dose-response bioassays, the LC50 was determined for each strain, with the SoB strain of S. oryzae being the most resistant to bifenthrin, the SzB strain of S. zeamais being the most resistant to pirimiphos-methyl and the SoA strain of S. oryzae was the most sensitive to both compounds. Analysis of the esterase patterns of insects exposed to these agrochemicals revealed the inhibition and probable involvement of carboxylesterases and cholinesterases in the detoxification processes of organophosphates and pyrethroids. Background Organophosphorus compounds, widely used in agriculture and industry, pose a serious threat to human health due to their acute neurotoxicity. Although traditional interventions for organophosphate poisoning are effective, they often come with significant side effects. Objective This paper aims to evaluate the potential of enzymes within biological organisms as organophosphorus bioclearing agents. It analyses the technical challenges in current enzyme research, such as substrate specificity, stereoselectivity, and immunogenicity, while exploring recent advancements in the field. Methods A comprehensive review of literature related to detoxifying enzymes or proteins was conducted. Existing studies on organophosphorus bioclearing agents were summarised, elucidating the biological detoxification mechanisms, with a particular focus on advancements in protein engineering and novel delivery methods. Results Current bioclearing agents can be categorised into stoichiometric and catalytic bioclearing agents, both of which have shown some success in preventing organophosphate poisoning. Technological advancements have significantly improved various properties of bioclearing agents, yet challenges remain, particularly in substrate specificity, stereoselectivity, and immunogenicity. Future research will focus on expanding the substrate spectrum, enhancing catalytic efficiency, prolonging in vivo half-life, and developing convenient administration methods. Conclusion With the progression of clinical trials, bioclearing agents are expected to become widely used as a new generation of therapeutic organophosphate detoxifiers. [ABSTRACT FROM AUTHOR]

The emergence of Novichok agents, potent organophosphorus nerve agents, has spurred the demand for advanced analytical methods and toxicity assessments as a result of their involvement in high-profile incidents. This study focuses on the degradation products of Novichok agents, particularly their potential toxic effects on biological systems. Traditional in vivo methods for toxicity evaluation face ethical and practical constraints, prompting a shift toward in silico toxicology research. In this context, we conducted a comprehensive qualitative and quantitative analysis of acute oral toxicity (AOT) for Novichok degradation products, using various in silico methods, including TEST, CATMoS, ProTox-II, ADMETlab, ACD/Labs Percepta, and QSAR Toolbox. Adopting these methodologies aligns with the 3Rs principle, emphasising Replacement, Reduction, and Refinement in the realm of toxicological studies. Qualitative assessments with STopTox and admetSAR revealed toxic profiles for all degradation products, with predicted toxicophores highlighting structural features responsible for toxicity. Quantitative predictions yielded varied estimates of acute oral toxicity, with the most toxic degradation products being EOPAA, MOPGA, MOPAA, MPGA, EOPGA, and MPAA, respectively. Structural modifications common to all examined hydrolytic degradation products involve substituting the fluorine atom with a hydroxyl group, imparting consequential effects on toxicity. The need for sophisticated analytical techniques for identifying and quantifying Novichok degradation products is underscored due to their inherent reactivity. This study represents a crucial step in unravelling the complexities of Novichok toxicity, highlighting the ongoing need for research into its degradation processes to refine analytical methodologies and fortify readiness against potential threats. BACKGROUND: Chlorfenapyr is used to kill insects that are resistant to organophosphorus insecticides. Chlorfenapyr poisoning has a high mortality rate and is difficult to treat. This article aims to review the mechanisms, clinical presentations, and treatment strategies for chlorfenapyr poisoning. DATA RESOURCES: We conducted a review of the literature using PubMed, Web of Science, and SpringerLink from their beginnings to the end of October 2023. The inclusion criteria were systematic reviews, clinical guidelines, retrospective studies, and case reports on chlorfenapyr poisoning that focused on its mechanisms, clinical presentations, and treatment strategies. The references in the included studies were also examined to identify additional sources. RESULTS: We included 57 studies in this review. Chlorfenapyr can be degraded into tralopyril, which is more toxic and reduces energy production by inhibiting the conversion of adenosine diphosphate to adenosine triphosphate. High fever and altered mental status are characteristic clinical presentations of chlorfenapyr poisoning. Once it occurs, respiratory failure occurs immediately, ultimately leading to cardiac arrest and death. Chlorfenapyr poisoning is difficult to treat, and there is no specific antidote. CONCLUSION: Chlorfenapyr is a new pyrrole pesticide. Although it has been identified as a moderately toxic pesticide by the World Health Organization (WHO), the mortality rate of poisoned patients is extremely high. There is no specific antidote for chlorfenapyr poisoning. Therefore, based on the literature review, future efforts to explore rapid and effective detoxification methods, reconstitute intracellular oxidative phosphorylation couplings, identify early biomarkers of chlorfenapyr poisoning, and block the conversion of chlorfenapyr to tralopyril may be helpful for emergency physicians in the diagnosis and treatment of this disease. Intoxications with organophosphorus compounds (OPCs) effect a severe impairment of cholinergic neurotransmission that, as a result of overstimulation may lead to desensitization of nicotinic acetylcholine receptors (nAChRs) and finally to death due to respiratory paralysis. So far, therapeutics, that are capable to address and revert desensitized neuromuscular nAChRs into their resting, i.e. functional state are still missing. Still, among a class of compounds termed bispyridinium salts, which are characterized by the presence of two pyridinium subunits, constituents have been identified, that can counteract organophosphate poisoning by resensitizing desensitized nAChRs. According to comprehensive modeling studies this effect is mediated by an allosteric binding site at the nAChR termed MB327-PAM-1 site. For MB327, the most prominent representative of the bispyridinium salts and all other analogues studied so far, the affinity for the aforementioned binding site and the intrinsic activity measured in ex vivo and in in vivo experiments are distinctly too low, to meet the criteria to be fulfilled for therapeutic use. Hence, in order to identify new compounds with higher affinities for the MB327-PAM-1 binding site, as a basic requirement for an enhanced potency, two compound libraries, the ChemDiv library with 60 constituents and the Tocriscreen Plus library with 1280 members have been screened for hit compounds addressing the MB327-PAM-1 binding site, utilizing the [2H 6 ]MB327 MS Binding Assay recently developed by us. This led to the identification of a set of 10 chemically diverse compounds, all of which exhibit an IC 50 value of ≤ 10 µM (in the [2H 6 ]MB327 MS Binding Assay), which had been defined as selection criteria. The three most affine ligands, which besides a quinazoline scaffold share similarities with regard to the substitution pattern and the nature of the substituents, are UNC0638, UNC0642 and UNC0646. With binding affinities expressed as p K i values of 6.01 ± 0.10, 5.97 ± 0.05 and 6.23 ± 0.02, respectively, these compounds exceed the binding affinity of MB327 by more than one log unit. This renders them promising starting points for the development of drugs for the treatment of organophosphorus poisoning by addressing the MB327-PAM-1 binding site of the nAChR. • MS Binding Assays with [2H 6 ]MB327 as reporter ligand are used for library screening. • Library screening yields new ligands of the MB327-PAM-1 binding site of the nAChR. • Hit compounds with the highest binding affinities are quinazoline derivatives. Insecticides are widely used in agriculture, horticulture, medicine, and industry, as well as domestically. Increased insecticide application has been attributed to higher agricultural productivity and lower infield and postharvest crop loss in the twentieth century. Malathion is a very commonly used organophosphorus (OP) insecticide applied in over 100 food crops in agriculture and post-harvest storage. Different species of Aspergillus have shown their efficiency to degrade OP chemicals. A malathion tolerant Aspergillus niger MRU01, developed by prolonged malathion exposure, was tested for its efficiency to remove malathion as well as three other commonly used OP insecticides, viz., parathion, chlorpyrifos and dimethoate. The tolerant strain showed growth efficiency as well as enhanced production of phosphatases and esterase enzyme as compared to untreated and treated wild type (A. niger ATCC16404). The removal of malathion was 29% and 68%, in the wild type and the tolerant strain, respectively during five days. The tolerant strain was also able to remove dimethoate (63%) and chlorpyrifos (54%) with high efficiency while a low rate of removal of parathion (33%) was observed. The strain can be tested in the microcosms more specifically for bioremediation of other OP insecticides before its application in the field. The malathion tolerant Aspergillus niger MRU01 grew efficiently in the presence of 500 μM malathion. Treatment of the fungus with malathion and other OP insecticides – parathion, chlorpyrifos, and dimethoate – caused enhanced expression of phosphatases and esterases. The tolerant A. niger MRU01 strain had significantly lower mycelia concentrations of insecticides than of the wild type A. niger ATCC16404. Objective: This study aims to investigate the potential link between exposure to organophosphorus pesticides (OPPs) and suicidal ideation (SI) among adults. Methods: This study encompassed four cycles of the National Health and Nutrition Examination Survey (NHANES), involving 5244 participants aged 20 and above. SI was assessed using the Patient Health Questionnaire-9. The levels of exposure to OPPs were estimated by analyzing concentrations of OPP metabolites in urine samples. Multivariate logistic regression models were used to explore the association between exposure to each OPP and SI. Stratified analyses and interaction tests were conducted across various groups, including pairwise combinations of gender and age, as well as body mass index, smoking status, hypertension, and diabetes. Weighted Quantile Sum (WQS) regression and Bayesian Kernel Machine Regression (BKMR) models were applied to assess the cumulative impact of exposure to the four OPPs on SI, along with their respective contributions. Additionally, the potential interactions among these four OPPs were evaluated. Results: Multivariate logistic regression revealed that only dimethylthiophosphate (DMTP) among OPPs demonstrated a statistically significant positive association with SI [OR: 1.18; 95 % CI: 1.02–1.37]. Stratified analyses indicated that the influence of OPPs on SI was particularly pronounced in young and older men. The WQS regression analysis revealed a statistically significant association between the mixed metabolites of OPPs and SI [OR = 1.10, 95 % CI: 1.04–1.16], with DMTP (weighted 0.63) contributing the most. Furthermore, the BKMR model supported a positive trend in the overall impact of these OPP metabolites on SI, displaying notable individual exposure-response relationships for DMTP (PIP: 0.77). Conclusions: Our study suggests an association between exposure to DMTP and an increased risk of SI. Specifically, young adult males and older males appear particularly susceptible to the effects of OPP exposure.

Profenofos (PF) and captan (CT) are among the most utilized organophosphorus insecticides and phthalimide fungicides, respectively. To elucidate the physicochemical and influential toxicokinetic factors, the mechanistic interactions of serum albumin and either PF or CT were carried out in the current study using a series of spectroscopy and computational analyses. Both PF and CT could bind to bovine serum albumin (BSA), a representative serum protein, with moderate binding constants in a range of 103–104 M−1. The bindings of PF and CT did not induce noticeable BSA's structural changes. Both pesticides bound preferentially to the site I pocket of BSA, where the hydrophobic interaction was the main binding mode of PF, and the electrostatic interaction drove the binding of CT. As a result, PF and CT may not only induce direct toxicity by themselves, but also compete with therapeutic drugs and essential substances to sit in the Sudlow site I of serum albumin, which may interfere with the pharmacokinetics and equilibrium of drugs and other substances causing consequent adverse effects. Dimethoate (DM) is one type of organophosphorus insecticide, that kills insects and mites both systemically and immediately upon contact and harms a range of non-target animals, including fish when it enters the aquatic habitat. This study aimed to determine the potentially harmful histopathological and biochemical impacts of Dimethoate on Nile tilapia fish. Sixty Oreochromis Niloticus freshwater fish, each weighing an average of 130 ± 10 g, were divided into two groups in triplicates. There were 6 groups, 3 control groups, and 3 experiment groups that received Dimethoate at a dose of 8 mg/l in water for 15 days. Blood samples were collected and used for biochemical indexes. For histological analysis, samples of the brain, gills, liver, and kidney were collected. The findings of the study demonstrated that fish treated with Dimethoate exhibited pale gills, anxious symptoms, as well as congestion and hemorrhages in many internal organs, comprising the brain, kidney, and liver. There was a significant increase in the blood levels of nitric oxide, lipid peroxide, and glutathione peroxidase and a significant fall in catalase levels in the Dimethoate group when compared to the control group. Diazinon (DNZ) is one of the most widely used organophosphorus poisons, which plays an important role in chemical pest control by controlling a wide range of chewing and sucking pests in gardens and fields. Diazinon causes death in living organisms by reducing cholinesterase and disrupting nerve cells. In this research, the effect of Rosa damascena extract against DNZ toxicity and oxidative damage induced by DZN in mice was studied. The mice toxified with DNZ (32.5 mg kg-1, intraperitoneally) and treated by pralidoxime (PM) (20 mg kg-1, intraperitoneally) or R. Damascena extract (50, 100, 200 mg kg-1, orally) daily for two weeks. In the end, the acetylcholinesterase (AchE), ferric-reducing antioxidant power (FRAP), Malondialdehyde (MDA), ALT, AST, ALP, and total bilirubin were assayed. DZN administration significantly lowered the AchE, and FRAP and increased the MDA (P < 0.05). However, R. Damascena extracts treatment caused a significant reduction in MDA level and restored the levels of AchE and FRAP as well as significantly prevented the DZN-induced increase in liver aminotransferases, ALP, and total bilirubin. Pesticide self-poisoning is one of the three most important global means of suicide, killing an estimated 110–168,000 people each year, mostly in poor rural Asian communities. Organophosphorus (OP) and carbamate anticholinesterase insecticides are responsible for about two-thirds of these deaths. Calcium channel blocking medicines (CCB) may reduce the effect of pesticides and prevent deaths. Two preclinical rodents' studies and eight clinical studies utilising nimodipine and magnesium sulphate (MgSO4), respectively, showed mixed results. We have established a multi-centre randomised controlled trial (RCT) of patients with OP or carbamate self-poisoning admitted to at least six major hospitals in Bangladesh. The study aims to recruit maximum 3,243 patients over four years. One-third of the patients selected at random will receive standard treatment, while one-third will be treated with additional nimodipine and one-third with additional MgSO4. The additional treatments will be given for 48 h. We will check mortality (currently an estimated 11% die with standard treatment) and need for intensive care for mechanical ventilation across the three groups. Organophosphorus agents, also known as nerve agents, are very dangerous chemicals that were used as chemical warfare agents. HI-6 is one of the most promising reactivators which is effective in reactivating AChE inhibited by many nerve agents. However, the fast in-vivo clearance of HI-6 became a large barrier for first aid use under some sophisticated circumstances. In this study, PEGylated liposomes loading HI-6 were prepared and evaluated in vitro and in vivo. For PEG-LP-HI-6, the optimal formulation's loading efficiency and encapsulation efficiency were 6.47 ± 0.10% and 71.2 ± 1.15%, respectively. According to the pharmacokinetic results, compared with free HI-6 and LP-HI-6, the intravenous injection of PEG-LP-HI-6 significantly extended t1/2 (1.47 ± 0.29 h), MRT (1.44 ± 0.07 h), and improved the AUC of HI-6 in vivo. Drug concentrations in the CNS also increased after the intravenous administration of PEG-LP-HI-6. For in vivo treatment study, twenty minutes after poison exposure, the survival rate of animals in saline, free HI-6, LP-HI-6 and PEG-LP-HI-6 groups were 0, 0, 30% and 70%, respectively. Compared with the non-PEGylated liposomes group and free HI-6, PEG-LP-HI-6 could prolong the survival time of experimental animals and alleviate the neurotoxic symptoms, which demonstrated great potential as a first-aid strategy for acute organophosphorus agent poisoning. Organophosphorus compounds (OPs) are phosphoric acid derivatives represented by the formula (R2XP=O/S), R as organic groups; however, they need not contain a direct carbonphosphorus bond. The organophosphorus compounds can be categorized into three classes, viz., organophosphates, carbamates nerve agents. The OPs having application as insecticides are generally phosphorothioates (i.e., containing P=S bond). These sulfur analogs are first bioactivated (in vivo) and converted to oxygen analogs responsible for exerting toxic action. These organophosphorus compounds are esters, fluorides, anhydrides, and amides of phosphoric, phosphorothioate, and phosphorodithioic acids. Organophosphorus poisoning kills individuals by causing central apnea; however, the underlying cause of death remains unclear. Following findings that the pre-Bötzinger complex impairment alone does not account for central apnea, we analyzed the effect of paraoxon on the brainstem-spinal cord preparation, spanning the lower medulla oblongata to phrenic nucleus. Respiratory bursts were recorded by connecting electrodes to the ventral 4th cervical nerve root of excised brainstem-spinal cord preparations obtained from 6-day-old Sprague–Dawley rats. We observed changes in respiratory bursts when paraoxon, neostigmine, atropine, and 2-pyridine aldoxime methiodide were administered via bath application. The percentage of burst extinction in the paraoxon-poisoning group was 50% compared with 0% and 18.2% in the atropine and 2-pyridine aldoxime methiodide treatment groups, respectively. Both treatments notably mitigated the paraoxon-induced reduction in respiratory bursts. In the neostigmine group, similar to paraoxon, bursts stopped in 66.7% of cases but were fully reversed by atropine. This indicates that the primary cause of central apnea is muscarinic receptor-mediated in response to acetylcholine excess. Paraoxon-induced central apnea is hypothesized to result from neural abnormalities within the inferior medulla oblongata to the phrenic nucleus, excluding pre-Bötzinger complex. Plant species and abiotic factors including season appear to be the most important variables influencing the frequency of visits by honeybees (Apis mellifera L.). In the present study, we evaluated the activity of honeybee workers visiting runner bean (Phaseolus coccineus L.) local cultivar 'Piękny Jaś'. The runner beans are widely cultivated in south-eastern Poland, and are an important forage plant for honeybees in agroecosystems. We aimed at a comprehensive monitoring of the health of colonies and symptoms in A. mellifera in response to acute exposure to pesticides. The most numerous visits of A. mellifera were observed at the highest flower opening of the runner bean. A very weak positive correlation was observed between the number of honeybees on P. coccineus, the number of visited flowers, the time spent per flower and air temperature. The visitation rates of honeybees were more frequent at mid-day and decreased after 15:00. Signs of poisoning were detected in two out of seven apiaries monitored for acute pesticide exposure symptoms on runner bean plantations. The analysis of dead honeybee samples revealed the acute exposure of honeybees to the imidacloprid (neonicotinoid) and chlorpyrifos (organophosphorus) insecticides, which are highly toxic and banned in the European Union. Hazard quotient (HQ) screening showed an elevated burden of imidacloprid and chlorpyrifos corresponding to 7.1% and 10% of the LDD50, respectively, most likely indicating bee poisoning due to chronic exposure to these substances with contaminated food. Background: The susceptibility of pests and natural enemies to conventional insecticides is a critical element in judging the success of integrated pest management programs implementing biological control as a tactic. In this study, the susceptibility of the cotton leafworm Spodoptera littoralis (Boisd.) (Lepidoptera: Noctuidae) to an organophosphorus insecticide (chlorpyrifos) and the entomopathogenic fungus Beauveria bassiana was evaluated through field applications during two successive sugar beet seasons. The populations of the coccinellid, chrysopid, and formicid predators were estimated to indicate pesticide risk assessment strategy. The root and sugar yields were compared during the whole experiment to emphasize the impact of both control strategies on the pre-harvest loss in this strategic crop. Results: Results indicated that over the course of the two seasons of the investigation, chlorpyrifos significantly reduced the population density of S. littoralis and its associated predators. It showed a detrimental effect on S. littoralis larval population than untreated areas, causing an overall reduction of 97 and 92% during the 1st and 2nd seasons, respectively. The biopesticide based on entomopathogenic fungus Beauveria bassiana caused an overall reduction of S. littoralis, reaching 96 and 65% during the 1st and 2nd seasons, respectively. Meanwhile, B. bassiana slightly affects the population of predators compared to insecticide-free areas, so it was recommended for application in sugar beet fields without providing considerable risk to the associated insect predators. The sugar beetroot production and its sugar content increased significantly in both treatments during the two growing seasons compared to non-treated plots. Conclusion: Chlorpyrifos is not recommended for pest control; instead, B. bassiana is considered an effective biopesticide in the Egyptian sugar beet fields infested with S. littoralis as part of an integrated pest management program. Their effect was direct through reducing pest population and indirectly increasing the sugar beetroot production and increasing its total sugar content. Hence, both the sugar beetroot quality and quantity were improved favoring the microbial agent over the chemical insecticide in application for mitigating the chemical hazards toward the associated predators. Methyl parathion (MP), an organophosphorus insecticide, is commonly used in agricultural products for food preservation and pest control. Due to the severe threat it poses to food safety and the environment, monitoring MP residues has attracted much attention. Traditional spectroscopic and chromatographic methods have been used successfully to analyze MP in a wide range of samples; however, these approaches have several drawbacks, such as requiring specialized equipment, trained technicians, and extensive sample preparation time. Due to these restrictions, there is a growing demand for analysis methods that can reliably and quickly detect MP at trace quantities while also being quick, sensitive, and selective. Electrochemical sensors have emerged over the past few decades as a viable alternative to more time-consuming and laborious analysis methods for detecting MP. However, the performance of electrochemical sensors has been dramatically improved thanks to recent breakthroughs in nanoscience. This study offers an overview of the creation and operation of carbon nanomaterial-based electrochemical sensors (including carbon nanotubes (CNTs), graphene (Gr), and other carbon nanomaterials) to identify MP residues in waters, fruits, and vegetables. Organophosphorus insecticides (OPs), acting as serine phosphorylating agents in acetylcholinesterase (AChE), are highly effective neurotoxic insecticides. In our previous research, we found that six herbivorous pests and four ladybirds howed significantly higher AChE LC50 values than seven parasitoids and a predator (Epistrophe balteate), and that there was a significant correlation with the corresponding bimolecular rate constant (Ki) value. The Ki value of pests was much smaller than that of natural enemies and had a higher LC50 value.Then, we speculated that the low sensitivity of the pest AChE to OPs may be associated with its higher recovery and lower aging ability. In this work, the I50 and I90 were calculated, to determine the sensibility of AChE in ten representative species, including Plutella xylostella, Prodenia litura, Musca domestica, and Cavia porcellus, to paraoxon and malaoxon. The enzyme activities were measured at various time points, and kinetic calculations were used to obtain their spontaneous reactivation (Ks) and aging (Ka) constants, which were comprehensively compared. We conclude that the Ka and Ks of the AChE inhibited by OPs showed primarily species-specific correlations, and little correlation with the sensitivity to OPs. The differences in the AChE sensitivity to paraoxon among the ten species were much greater than in the sensitivity to malaoxon. Compared to paraoxon, malaoxon was more selective for Cavia porcellus. Coleoptera insects showed a stronger dephosphorylation ability than other insect groups. The recovery ability of phospho-AChE was stronger in mammals than in insects, which could be related to the low sensitivity of the AChE site of action to OPs. The Ka of the AChE inhibited by malaoxon was larger than that inhibited by paraoxon with the corresponding biomaterials, indicating that the OP type had a substantial relationship with the Ka of the AChE. We further discovered that, when insects were inhibited by OP, the tendency of AChE to undergo aging was greater than that of dephosphorylation. Overall, the study provides valuable information on the action mechanism of various OPs on AChE in several species, which could be used to further research into AChE and the potential dangers that organophosphates pose to animals. In modern agricultural practices, organophosphorus pesticides or insecticides (OPs) are regularly used to restrain pests. Their limits are closely monitored since their residual hinders the capability of acetylcholinesterase (AChE) and brings out a threatening accumulation of the neurotransmitter acetylcholine (ACh), which affects human well-being. Therefore, spotting OPs in food and the environment is compulsory to prevent human health. Several techniques are available to identify OPs but encounter shortcomings like time-consuming, operating costs, and slow results achievement, which calls for further solutions. Herein, we present a rapid colorimetric sensor for quantifying OPs in foods using TMB as a substrate, a multi-enzyme cascade system, and the synergistic property of core-shell Palladinum@Platinum (Pd@Pt) nanoparticles. The multi-enzyme cascade response framework is a straightforward and effective strategy for OPs recognition and can resolve the previously mentioned concerns. Numerous OPs, including Carbofuran, Malathion, Parathion, Phoxim, Rojor, and Phosmet, were successfully quantified at different concentrations. The cascade method established using Pd@Pt had a simple and easy operation, a lower detection limit range of (1–2.5 ng/mL), and a short detection time of about 50 min. With an R2 value of over 0.93, OPs showed a linear range of 10–200 ng/mL, portraying its achievement in quantifying pesticide residue. Background: With the restriction of organophosphorus and other insecticides, pyrethroids are currently the second most-used group of insecticides worldwide due to their advantages such as effectiveness and low toxicity for mammalian. Animal studies and clinical case reports have documented associations between adverse health outcomesand exposure to pyrethroids. At present, the association between chronic pyrethroid exposure and osteoarthritis (OA) remains elusive. Methods: Cross-sectional data from the National Health and Nutrition Examination Survey 1999–2002 and 2007–2014 were used to explore the associations of pyrethroid exposure and OA. Urinary level of 3-phenoxybenzoic acid (3-PBA) in urine samples was used to evaluate the exposure of pyrethroid, and OA was determined on the basis of self-reported physician diagnoses. Multivariable logistic regression models were used to investigate the association between pyrethroid exposure and OA. Results: Among the 6528 participants, 650 had OA. The weighted geometric mean of urinary volume-based 3-PBA concentration were 0.45 µg/L. With adjustments for major confounders, compared to participants in the lowest quartile of urinary volume-based 3-PBA, those in the highest quartilehad higher odds of OA (odds ratio, 1.39; 95% confidence interval: 1.01, 1.92). There was no nonlinear relationship between urinary volume-based 3-PBA and OA (P for non-linearity = 0.89). Cowpea beetle, Callosobruchus maculatus (Fabricius) (Coleoptera: Chrysomelidae) is one of the most dangerous insect pests in field and storage. It causes a high damage to the legumes by decreased dietary values, germination and seed weights. Using synthetic insecticides for controlling stored grain pests have a lot of problems for humans and environment. Therefore, the mixing seeds laboratory experiments were conducted to study the efficacy of certain alternative safety compounds against C. maculatus such as Beauveria bassiana, Metarhizium anisopliae, ascorbic acid, boric acid, talc powder and kaolin compared with an organophosphorus nervous insecticide, malathion 1% dust, that recommended in Egypt for controlling stored grain pests. The present results showed that the malathion was the highest toxicity effect against cowpea beetle adults with LC50(3.89 ppm) after 3-day post treatment. However, the boric acid was the lowest toxicity effect with LC50 (72855 ppm) after 6 days post treatment. The other tested materials showed moderate effects. These results indicate that malathion is still having high effect against cowpea beetle. The toxicity of bio-insecticides (M. anisopliae and B. bassiana) after 6days were more than toxicity malathion 1%dust after 1day.Wheraes, the toxicity of bio-insecticides (M. anisopliae and B. bassiana) increased with increase of period of exposure from 2 to 6 days high rates about 28 times, 31 times, respectively. The findings showed that the tested compounds have a promising insecticidal activity against C. maculatus and can be used as possible alternatives to synthetic chemical insecticides such as malathion for the control of stored product insects. Introduction: Pendimethalin is an herbicide that is used worldwide to attack certain wide leaf and grassy weeds in a variety of agricultural and non-crop regions, with a claimed very low toxicity to human. There have been some few reported cases of acute pendimethalin toxicity to date, around the world. We report a case of pendimethalin toxicity resembling and managed initially as organophosphorus poisoning. Case Description: A 42-year-old agricultural worker was found unresponsive in his field with profuse oral secretions. The patient presented with altered mental status, where Glasgow comma scale was 10, oxygen saturation at room air was 88 %, Blood pressure measured 125/75, pulse rate was 95 beats per minute, and temperature was 37 °C. The patient's pupils were constricted, equal, regular, and reactive. Chest examination revealed scattered respiratory crepitations. The patient also had a distinct garlic odor very similar to that of Organophosphate compounds. The results of his laboratory investigations revealed only hypoxemia. The patient received atropine and supportive management, which prompted the chest secretions to clear up and allow him to restore consciousness without experiencing any neurological problems. Conclusion: Pendimethalin toxicity can present with manifestations mimicking acute organophosphorus poisoning. Proper supportive care is needed for the management of acute pendimethalin toxicity. Organophosphate insecticides have been widely used in agriculture to control insect pests and raised growing problems including insecticide resistance. According to recent studies, microbial symbionts play important roles in developing insecticide resistance. The bean bug Riptortus pedestris is a notorious pest of leguminous crops, harbouring diverse microorganisms in their insect midguts. However, the potential biological roles of many gut‐associated bacteria are largely unknown. In this study, we isolated a bacterial strain from the midgut of R. pedestris and identified it as Serratia marcescens based on its morphological characteristics and the phylogenetic analysis of the 16S rRNA gene sequence. A halo of phosphorus‐solubilizing bacteria was observed on a medium containing lecithin and an organophosphorus insecticide dimethoate. Subsequently, the microbial degradation of dimethoate was determined using Gas Chromatography with Mass Spectroscopy (GC–MS). Further analysis indicated that the S. marcescens contained organophosphorus‐degrading MBL‐fold metallo‐hydrolase gene. Additionally, we discovered that the bacterium could colonize the insect midgut stably and enhance the host survivorship when exposed to dimethoate. Our results revealed that an insect gut‐associated bacterium could degrade organophosphorus compounds, thereby reducing related insecticide toxicity to the insect hosts. Different regions may have different profiles depending on location and standards of living. To know the profile and outcome of acute poisoning and envenomation among children at our region this study was undertaken. Methods: The ambispective study from year 2014 -2021 reviewed all maximum possible cases of pediatric poisoning and envenomation by retrieving files from the medical record department and pertinent information collected. Results: Out of 188 analyzed data, 97 (51.3%) were males. Median (Q1, Q3) age in years was 9.5 (2.7, 15.3). Total cases of acute poisoning were 91(48.4%), 80(42.6%) snakebite/scorpion/other bites whereas 17(9%) drowning & hanging. In outcome, deaths were in 7(3.7%) whereas 167(88.8%) discharged, 12(6.3%) LAMA and referral 2(1.1%). Accidental and suicidal poisoning cases were 123(65.4%) & 39(20.8%) respectively. Among substance poisoning, the top 2 causes were organophosphorus poisoning 37(41.1%) & kerosene 13(14.4%). Ventilator support, vasoactive drug support, debridement and dialysis were required in 58(30.9%), 21(11.2%), 14(7.5%) & 3(1.6%) cases respectively. Metabolic acidosis and hyponatremia in 25(13.3) & 16(8.5%) cases whereas mortality was significantly associated with the need for vasoactive support and aluminum phosphide poisoning. Conclusion: In children, acute accidental poisoning was commonest followed by envenomation at our tertiary care canter. For a half-century, the commercial wild silkworm, Antheraea pernyi, has been protected by coumaphos, which is an internal organophosphorus insecticide used to kill the potential parasitic fly larvae inside. Knowledge about the detoxification genes of A. pernyi as well as the detoxification mechanism for this species remains severely limited. In this study, we identified 281 detoxification genes (32 GSTs, 48 ABCs, 104 CYPs, and 97 COEs) in the genome of this insect, which are unevenly distributed over 46 chromosomes. When compared to the domesticated silkworm, Bombyx mori, a lepidopteran model species, A. pernyi has a similar number of ABCs, but a greater number of GSTs, CYPs, and COEs. By transcriptome-based expression analysis, we found that coumaphos at a safe concentration level significantly changed the pathways related to ATPase complex function and the transporter complex in A. pernyi. KEGG functional enrichment analysis indicated that protein processing in the endoplasmic reticulum was the most affected pathway after coumaphos treatment. Finally, we identified four significantly up-regulated detoxification genes (ABCB1, ABCB3, ABCG11, and ae43) and one significantly down-regulated detoxification gene (CYP6AE9) in response to coumaphos treatment, suggesting that these five genes may contribute to detoxification of coumaphos in A. pernyi. Our study provides the first set of detoxification genes for wild silkworms from Saturniidae and highlights the importance of detoxification gene repertoire in insect pesticide tolerance. BACKGROUND: Hormesis is a common phenomenon in toxicology described as low‐dose stimulation due to a toxin which causes inhibition at a high dose. Pesticide hormesis in plants has attracted considerable research interest in recent years; however, the specific mechanism has not yet been clarified. Acephate is an organophosphorus insecticide that is used worldwide. Here, hormesis in tomato (Solanum lycopersicum L.) plant growth and photosynthesis after acephate exposure is confirmed, as stimulation occurred at low stress levels, whereas inhibition occurred after exposure to high concentrations. RESULTS: We found that low acephate concentration (5‐fold lower than recommended application dosage) could enhance chlorophyll biosynthesis and stimulate photosynthesis effects, and thus improve S. lycopersicum growth. A high level of acephate (5‐fold higher than recommended application dosage) stress inhibited chlorophyll accumulation, decreased photosystem II efficiency and blocked antioxidant reactions in leaves, increasing reactive oxygen species levels and damaging plant growth. Transcriptomic analysis and quantitative real‐time PCR results revealed that the photosynthesis – antenna proteins pathway played a crucial role in the hormesis effect, and that LHCB7 as well as LHCP from the pathway were the most sensitive to acephate hormesis. CONCLUSION: Our results showed that acephate could induce hormesis in tomato plant growth and photosynthesis, and that photosystem II and the photosynthesis – antenna proteins pathway played important roles in hormesis. Diazinon (DZN) is an insecticide extensively used to control pests in crops and animals. However, its indicriminated use may lead to liver damage in animals and humans. This study aimed to evaluate the toxicity of DZN (25–150 µM) on human hepatoblastoma (HepG2) cells after 24 and 48 h of exposure and the role of its biotransformation on the toxicological potential. We also tested the protective effect of tetrahydrocurcumin (THC), an antioxidant agent, in the DZN-induced citotoxicity. DZN caused cytotoxicity in the HepG2 cells, inhibiting cell proliferation and reducing cell viability in a dose- and time-dependent manner. The pre-incubation of HepG2 cells with chemical inducers of cytochrome P450 monooxygenase 3-methylcholanthrene and phenobarbital resulted in a further decrease of cell viability associated with DZN exposure. In addition, the metabolite diazoxon was more toxic than DZN. Our results also revealed that THC alleviated DZN-induced cytotoxicity and reactive oxygen and nitrogen species (RONS) generation in HepG2 cells. In conclusion, our data provide novel insights into the involvement of biotransformation in the mechanisms of DZN-induced cytotoxicity and suggest that amelioration of RONS accumulation might be involved in the protective effect of THC on DZN-induced liver injury.

Simple Summary: We used the geometric morphometrics method to explore whether the exposure of the parental females to pirimiphos-methyl at short exposure times may impact the wings of the F1 generation of Rhyzopertha dominica, Prostephanus truncatus, and Tenebrio molitor. For this purpose, adult females of the three coleopterans were exposed to treated concrete at the label dose for 30 min, 3, 5, 8, 16, 24, and 36 h. Their offspring responded differently to pirimiphos-methyl. The most noticeable deformations were observed in the elytra and hindwings of T. molitor, and males experienced deeper morphological changes than females. The hindwings of P. truncatus exhibited morphological deformations after 36 h of exposure to pirimiphos-methyl. Rhyzopertha dominica proved the least susceptible species since pirimiphos-methyl did not have any effect on both elytra and hindwings. Tenebrio molitor L. (Coleoptera: Tenebrionidae), Prostephanus truncatus (Horn), and Rhyzopertha dominica (F.) (Coleoptera: Bostrychidae) are noxious insect pests of grains in storages. Pirimiphos-methyl is widely used to protect grains at the post-harvest stage. However, the sub-lethal impact of this active ingredient on the offspring of all three coleopterans remains unknown. Thus, mated females of each species were exposed separately to pirimiphos-methyl at short exposures (30 min, 3, 5, 8, 16, 24, and 36 h), where the elytra and hindwings of the adult progeny were analyzed with the geometric morphometrics method. Males and females of all species were incorporated into the analysis. The results revealed variability among species. Tenebrio molitor was the most sensitive among three species, displaying significant deformations in the elytra and hindwings. Males had more conspicuous morphological changes than females. Prostephanus truncatus hindwings exhibited deformities after 36 h of exposure to pirimiphos-methyl. In contrast, R. dominica offspring were not affected by pirimiphos-methyl. In light of our findings, organophosphorus insecticides may cause variable sub-lethal effects to stored-product insects. Pesticide use is an integral part in global agricultural practice, however, these chemicals have recently been identified as emerging pollutants that impact on ecosystem health and sometimes require remediative intervention. This study assessed the biodegradative ability of species of Bacillus on the organochlorine herbicide, paraquat and the organophosphorus insecticide, dichlorvos. The Bacillus isolates were identified based on their genomic, biochemical and morphological characteristics while the ability of the isolates to degrade the pesticides was determined by monitoring changes in optical density, pH, total viable count and pesticide concentration in in vitro systems over an 18-day period using pesticide-modified mineral salts broth. The two Bacilli isolates were identified as Bacillus crassostreae and Bacillus niabensis. The organochlorine herbicide was more readily utilised by the axenic Bacillus isolates and their mixed culture than the organophosphorus insecticide as evidenced by variations in turbidity and counts. Biodegradation was more effective with the mixed culture than with the axenic cultures. Pesticide degradation levels of 64.26 % - 93.70 % and 70.43 % - 98.20 % were observed for the insecticide and herbicide respectively for both the bacilli and their mixed culture as revealed by gas chromatography analysis. The degradation efficiencies of the two bacilli for both dichlorvos and paraquat showed significant differences from each other and from the mixed culture at 95% confidence interval. The study established the potential of Bacillus crassostreae, Bacillus niabensis and their mixed culture to be employed as inocula in the detoxification of pesticide-contaminated environmental media. Organophosphates (OP) poisoning continues to be a major cause of deliberate self-harm and an important cause of morbidity and mortality in India. We aim to study the clinico-epidemiological features in patients presenting with OP poisoning. This prospective observational study involved 400 patients above 18 years of age admitted to the emergency department of a tertiary care institution in Northern India. The study cohort was graded into mild, moderate & severe based on the Peradeniya Organophosphorus Poisoning (POP) scale and degree of cholinesterase inhibition. The majority of patients (40%) in our study population were aged between 18-25 years. There was a significant female preponderance (82% vs. 18%; P=0.000). Suicide was the most common motive (93%) and ingestion was the most common mode of poisoning (99%). The muscarinic and nicotinic symptoms were common at the presentation. Seizures were present in 11% of patients. On the POP severity scale, mild, moderate and severe poisoning was seen at 50%, 35% and 15%, respectively. Cholinesterase inhibition was mild (>2 kU/L) in 31%, moderate (1-2 kU/L) in 46% and severe (<1 kU/L) in 23%. A total of 48% of patients had complications. Ventilator support was needed in 43% of the patients. An overall mortality of 18% was observed in the study. Introduction: Organophosphorus pesticide self-poisoning is estimated to kill around 200000 people yearly. Early recognition is life-saving as the mortality rates are high following OP Poisoning. Acute Physiology and Chronic Health Evaluation II Score (APACHE II) Sequential Organ Failure Assessment Score (SOFA), and Poisoning Severity Score (PSS) with Lactate, are used to evaluate the prognosis. Materials and Methods: This prospective observational study included 236 individuals with acute OP poisoning who presented to the emergency department and were hospitalized in the critical care unit and step-down ICU. PSS, SOFA, and APACHE II scores were calculated at Emergency Department and followed up with the patient until discharge or death. Results and Discussion: Patients had a mean age of 32.8 ± 13.4 years and were 49.2% female and 50.8% male. Out of 236 patients, 22 patients died and 214 were discharged. The best cut-off values for predicting mortality with PSS, SOFA, and APACHE II scores were 2, 3, and 15, with AUROC of 0.929, 0.970, and 0.984. In addition to Lactate to the above scores, the AUROC increased to 0.981, 0.993, and 0.992, respectively. Cupriavidus nantongensis X1T is a type strain of the genus Cupriavidus, that can degrade eight kinds of organophosphorus insecticides (OPs). Conventional genetic manipulations in Cupriavidus species are time-consuming, difficult, and hard to control. The clustered regularly interspaced short palindromic repeat (CRISPR)/associated protein 9 (Cas9) system has emerged as a powerful tool for genome editing applied in prokaryotes and eukaryotes due to its simplicity, efficiency, and accuracy. Here, we combined CRISPR/Cas9 with the Red system to perform seamless genetic manipulation in the X1T strain. Two plasmids, pACasN and pDCRH were constructed. The pACasN plasmid contained Cas9 nuclease and Red recombinase, and the pDCRH plasmid contained the dual single-guide RNA (sgRNA) of organophosphorus hydrolase (OpdB) in the X1T strain. For gene editing, two plasmids were transferred to the X1T strain and a mutant strain in which genetic recombination had taken place, resulting in the targeted deletion of opdB. The incidence of homologous recombination was over 30%. Biodegradation experiments suggested that the opdB gene was responsible for the catabolism of organophosphorus insecticides. This study was the first to use the CRISPR/Cas9 system for gene targeting in the genus Cupriavidus, and it furthered our understanding of the process of degradation of organophosphorus insecticides in the X1T strain. Objective: The aim of this study was to evaluate the association between organophosphorus pesticides (OPPs) exposure and sleep problems. Methods: In this study, data from 6295 participants aged 18 years or older were collected from the National Health and Nutrition Examination Survey (NHANES). The dialkyl phosphate compounds (DAPs) metabolites, OPPs exposure biomarker, were examined using solid phase extraction-high coupled with isotope dilution-ultrahigh performance liquid chromatography-tandem mass spectrometry. Data on short sleep duration (SSD), self-reported trouble sleeping and self-reported sleep disorder were collected from the database. Weighted generalized logistic model, weighted quantile sum (WQS) model, and quantile-based g calculation (QGC) methods were used for analyzing the collected data. Results: The prevalence of SSD, self-reported trouble sleeping and self-reported sleep disorder in this study were 28.91 % (1814/6274), 25.31 % (1593/6294), and 9.05 % (289/3195), respectively. The wide use of the insecticide profenofos in crop production has led to serious ecological water problems in agricultural fields. With the increasing global production of nuts, a large amount of nutshell waste has a serious impact on the environment. Turning nutshell waste into biochar to remove high levels of profenofos in water is a cost-effective treatment method. In this study, biochars made from nutshell waste are investigated for the adsorption of aromatic organophosphorus insecticide profenofos. The adsorption amount of nutshell biochar was 13-fold higher than crop stalk biochar in removing profenofos from water. The results indicated that the adsorption of profenofos by nutshell biochar was specific. Scanning electron microscope (SEM) and Brunauer–Emmett–Teller (BET) analysis showed that nutshell biochars had a larger specific surface area and more microporous structures. Meanwhile, nutshell biochars could exhibit a stable adsorption capacity at different initial concentrations of profenofos (10–40 mg/L), temperature (298–318 K), and pH (3–7). Desorption and reuse experiments showed that profenofos was firmly bound to nutshell biochars in water and could be extracted from the biochars with acetonitrile. Within 10 times of recycling, nutshell biochar had a stable and strong adsorption capacity for profenofos. The adsorption process of profenofos by nutshell biochar was pore diffusion and surface adsorption, which is consistent with the pseudo second-order kinetic model and the Freundlich isotherm model. Elemental and Fourier transform infrared spectroscopy (FTIR) analyses showed that the adsorption mechanism of profenofos on nutshell biochar was mainly through π-π and hydrophobic interactions. Nutshell biochar also showed strong adsorption capacity for other aromatic organophosphorus pesticides, and the adsorption rates of methyl parathion, isocarbophos and 2-chloro-4-bromophenol were 85%, 73% and 73%, respectively. The dynamic trajectory of metabolites and gene expression related to phosphorus absorption and utilization in soybean seedling roots were determined under short- and long-term phosphorus deficiency stress. The metabolome results showed that TCA and GS/GOGAT cycles were enhanced after 2 days of phosphorus deficiency stress; however, they were inhibited after 15 days. GC-TOF-MS showed that phosphorus deficiency increased the accumulation of amino acids significantly after 2 days, whereas organic acids and lipid substances increased significantly after 15 days. Quantitative reverse transcription-polymerase chain reaction (RT-PCR) showed that transcriptional levels of five key genes related to phosphorus activation and phosphorus starvation signal transduction increased continuously with phosphorus deficiency. The expression of GmPHT1 and GmSPX triggered the phosphorus starvation signal pathway and induced the expression of the GmPS and GmPAP genes to enhance the synthesis and secretion of organophosphorus hydrolase and organic acid in soybean roots under phosphorus deficiency. The phospholipid metabolism was enhanced significantly after 15 days of stress and when GmSQD , a crucial enzyme in lipid biosynthesis, was up-regulated. Chlorpyrifos (CP), a broad-spectrum organophosphorus insecticide, is known for deleterious effects on soil enzymatic activities. Hence, the present study aims to examine the resilience effect of biochar (BC) aided Pelargonium graveolens L. plantation on enzymatic activities of chlorpyrifos contaminated soil. The two chlorpyrifos contaminated agriculture soils (with concentrations: S1: 46.1 and S2: 95.5 mg kg−1) were taken for the pot experiment. The plant biomass, plant growth parameters, soil microbial biomass, and enzymatic activities such as alkaline phosphatase, N-acetyl glucosaminidase, aryl sulphatase, cellulase, β-glucosidase, dehydrogenase, phenoloxidase, and peroxidase enzymes were examined. Ecoenzyme activities and their stoichiometry were used to enumerate the different indices including geometric mean, weighted mean, biochemical activity indices, integrated biological response, treated-soil quality index, and vector analysis in all treatments. The results of the study demonstrated that the biochar incorporation enhanced the tolerance of P. graveolens (from 42–45% to 55–67%) in chlorpyrifos contaminated soil and reduced the CP accumulation in plants. A reduction in the inhibitory effect of chlorpyrifos on soil enzymatic activities and plant growth by BC incorporation was observed along with an increase in the activities of ecoenzymes (16.7–18.6%) in soil. The investigation indicated more microbial investments in C and P than that in N acquisition under CP stress. The BC amendment catalyzed the activities of lignin and cellulose-degrading enzymes and enhanced nutrition acquisition. The CP contamination and BC amendment have no significant effect on the oil quality of P. graveolens. During storage, soybean kernels can be attacked by insects, which are preventively controlled with insecticides. Information on the dissipation of insecticide residues is crucial to know their final concentrations in food and establish the waiting periods necessary for consumption without health risks, and to determine the minimum waiting period necessary to comply with national and international standards. The aims of this study were to quantify the residue levels of organophosphorus insecticides (dichlorvos, chlorpyrifos-methyl and pirimiphos-methyl) in stored soybean, establish the effect during the storage period, and model the dissipation dynamics. Insecticide residues in soybeans were analyzed at 2, 30, 60, 90 and 120 days after application. An analytical method based on QuEChERS extraction followed by gas chromatography mass spectrometry (GC-MS/MS) determination was validated, with mean recoveries of 82-105%, depending on the spiking levels. Residues decreased below 80% of the initial concentration at 60 days after application and below quantifiable levels at 120 days. Residues followed a pseudo-first-order dissipation dynamics [Ct = C0×exp(-k.t)], with the dissipation constant (k) and half-lives being 0.538 and 1.3 days for dichlorvos, 0.018 and 38.8 days for chlorpyrifos-methyl, and 0.023 and 30.1 days for pirimiphos-methyl, respectively. These results allow concluding that, at the recommended dosage, these insecticides are safe for use on soybean grains stored under standard conditions commonly nowadays used in Argentina.
