BackgroundGastric cancer poses a major diagnostic and therapeutic challenge as surgical resection provides the only opportunity for a cure. Specific labeling of gastric cancer could distinguish resectable and nonresectable disease and facilitate an R0 resection, which could improve survival.MethodsTwo patient-derived gastric cancer lines, KG8 and KG10, were established from surgical specimens of two patients who underwent gastrectomy for gastric adenocarcinoma. Harvested tumor fragments were implanted into the greater curvature of the stomach to establish patient-derived orthotopic xenograft (PDOX) models. M5A (humanized anti-CEA antibody) or IgG control antibodies were conjugated with the near-infrared dye IRDye800CW. Mice received 50 µg of M5A-IR800 or 50 µg of IgG-IR800 intravenously and were imaged after 72 hr. Fluorescence imaging was performed by using the LI-COR Pearl Imaging System. A tumor-to-background ratio (TBR) was calculated by dividing the mean fluorescence intensity of the tumor versus adjacent stomach tissue.ResultsM5A-IR800 administration resulted in bright labeling of both KG8 and K10 tumors. In the KG8 PDOX models, the TBR for M5A-IR800 was 5.85 (SE ± 1.64) compared with IgG-IR800 at 0.70 (SE ± 0.17). The K10 PDOX models had a TBR of 3.71 (SE ± 0.73) for M5A-IR800 compared with 0.66 (SE ± 0.12) for IgG-IR800.ConclusionsHumanized anti-CEA (M5A) antibodies conjugated to fluorescent dyes provide bright and specific labeling of gastric cancer PDOX models. This tumor-specific fluorescent antibody is a promising potential clinical tool to detect the extent of disease for the determination of resectability as well as to visualize tumor margins during gastric cancer resection.

SIGNIFICANCE: Fluorescent organic dyes provide imaging capabilities at cellular and sub-cellular levels. However, a common problem associated with some of the existing dyes such as the US FDA-approved indocyanine green (ICG) is their weak fluorescence emission. Alternative dyes with greater emission characteristics would be useful in various imaging applications. Complementing optical imaging, magnetic resonance (MR) imaging enables deep tissue imaging. Nano-sized delivery systems containing dyes with greater fluorescence emission as well as MR contrast agents present a promising dual-mode platform with high optical sensitivity and deep tissue imaging for image-guided surgical applications. AIM: We have engineered a nano-sized platform, derived from erythrocyte ghosts (EGs), with dual near-infrared fluorescence and MR characteristics by co-encapsulation of a brominated carbocyanine dye and gadobenate dimeglumine (Gd-BOPTA). APPROACH: We have investigated the use of three brominated carbocyanine dyes (referred to as BrCy106, BrCy111, and BrCy112) with various degrees of bromination, structural symmetry, and acidic modifications for encapsulation by nano-sized EGs (nEGs) and compared their resulting optical characteristics with nEGs containing ICG. RESULTS: We find that asymmetric dyes (BrCy106 and BrCy112) with one dibromobenzene ring offer greater fluorescence emission characteristics. For example, the relative fluorescence quantum yield ( ϕ ) for nEGs fabricated using 100    μ M of BrCy112 is ∼ 41 -fold higher than nEGs fabricated using the same concentrations of ICG. The dual-mode nEGs containing BrCy112 and Gd-BOPTA show a nearly twofold increase in their ϕ as compared with their single optical mode counterpart. Cytotoxicity is not observed upon incubation of SKOV3 cells with nEGs containing BrCy112. CONCLUSIONS: Erythrocyte nano-ghosts with dual optical and MR characteristics may ultimately prove useful in various biomedical imaging applications such as image-guided tumor surgery where MR imaging can be used for tumor staging and mapping, and fluorescence imaging can help visualize small tumor nodules for resection.

SIGNIFICANCE: Spatial frequency domain imaging (SFDI) applies patterned near-infrared illumination to quantify the optical properties of subsurface tissue. The periocular region is unique due to its complex ocular adnexal anatomy. Although SFDI has been successfully applied to relatively flat in vivo tissues, regions that have significant height variations and curvature may result in optical property inaccuracies. AIM: We characterize the geometric impact of the periocular region on SFDI imaging reliability. APPROACH: SFDI was employed to measure the reduced scattering coefficient ( μ s ) and absorption coefficient ( μ a ) of the periocular region in a cast facial tissue-simulating phantom by capturing images along regions of interest (ROIs): inferior temporal quadrant (ITQ), inferior nasal quadrant (INQ), superior temporal quadrant (STQ), central eyelid margin (CEM), rostral lateral nasal bridge (RLNB), and forehead (FH). The phantom was placed on a chin rest and imaged nine times from an en face or side profile position, and the flat back of the phantom was measured 15 times. RESULTS: The measured μ a and μ s of a cast facial phantom are accurate when comparing the ITQ, INQ, STQ, and FH to its flat posterior surface. Paired t tests of ITQ, INQ, STQ, and FH μ a and μ s concluded that there is not enough evidence to suggest that imaging orientation impacted the measurement accuracy. Regions of extreme topographical variation, i.e., CEM and RLNB, did exhibit differences in measured optical properties. CONCLUSIONS: We are the first to evaluate the geometric implications of wide-field imaging along the periocular region using a solid tissue-simulating facial phantom. Results suggest that the ITQ, INQ, STQ, and FH of a generalized face have minimal impact on the SFDI measurement accuracy. Areas with heightened topographic variation exhibit measurement variability. Device and facial positioning do not appear to bias measurements. These findings confirm the need to carefully select ROIs when measuring optical properties along the periocular region.

SignificanceOver the past decade, machine learning (ML) algorithms have rapidly become much more widespread for numerous biomedical applications, including the diagnosis and categorization of disease and injury.AimHere, we seek to characterize the recent growth of ML techniques that use imaging data to classify burn wound severity and report on the accuracies of different approaches.ApproachTo this end, we present a comprehensive literature review of preclinical and clinical studies using ML techniques to classify the severity of burn wounds.ResultsThe majority of these reports used digital color photographs as input data to the classification algorithms, but recently there has been an increasing prevalence of the use of ML approaches using input data from more advanced optical imaging modalities (e.g., multispectral and hyperspectral imaging, optical coherence tomography), in addition to multimodal techniques. The classification accuracy of the different methods is reported; it typically ranges from ∼70% to 90% relative to the current gold standard of clinical judgment.ConclusionsThe field would benefit from systematic analysis of the effects of different input data modalities, training/testing sets, and ML classifiers on the reported accuracy. Despite this current limitation, ML-based algorithms show significant promise for assisting in objectively classifying burn wound severity.

BACKGROUND: Existing neuroimaging techniques, including magnetic resonance imaging, computed tomography, and high-resolution ultrasound, lack the capability to provide real-time intraoperative positioning images to surgeons. However, the clinical implementation of near-infrared fluorescence imaging technology has made it possible to directly visualize surgical target areas, offering a novel solution for real-time nerve identification during surgery. OBJECTIVE: To provide a summary and overview of the research progress in near-infrared fluorescence imaging technology for intraoperative neuroimaging. METHODS: The first author used the computer to search for the documents published from January 2010 to July 2023 in WanFang, CNKI, and PubMed with the key words of “near-infrared fluorescence imaging, optical imaging, nerve imaging” in Chinese and English. A few classic old documents were also included. Initial screening was performed by reading the titles and abstracts; duplicate, low-quality, and irrelevant content documents were excluded. A total of 69 articles were finally included for review. RESULTS AND CONCLUSION: Near-infrared fluorescence imaging guided by indocyanine green has been clinically used to identify and locate tubular organs such as blood vessels, ureters, and bile ducts, as well as various tumors during surgery. This technique is currently considered a well-established imaging method in precision surgery. In the field of intraoperative neurofluorescence imaging, indocyanine green is currently the only near-infrared fluorescent dye used in clinical research. The ideal neuroimaging agent should possess certain characteristics, including easy administration in the perioperative period, logD between 0.5 and 3 at pH=7.4, molecular mass below 500 Da, excitation and emission wavelengths within the near-infrared window, long-term retention in nerve tissue, high signal-to-background ratio, and high safety. In the future, the development of near-infrared neurofluorescence imaging agents should focus on synthesizing complexes of indocyanine green and neural-specific targets. This technology not only enables intraoperative neurofluorescence imaging, but also holds promising prospects for in-situ monitoring of nerve regeneration and diagnosis of neurological diseases. Integral imaging (InIm) 3D displays provide realistic 3D images with comfortable viewing experiences. However, existing approaches struggle to achieve high spatial resolution reproduction without relying on temporal or spatial multiplexing methods, which increases system complexity. Although precise control approaches for voxel formation have been explored to address this challenge, the existing methods lack systematic analysis theories for identifying the key factors affecting spatial resolution. In this study, a novel analysis framework is proposed that uses a point spread function (PSF) to characterize voxel formation. An analytical model based on the obtained PSF is established to achieve an aliasing‐free display. This framework identifies optimization directions for improving the spatial resolution, specifically by reducing the Gaussian blur caused by wavefront aberration in the lens unit. A targeted optimization of a compound lens array is conducted to achieve precise control of voxel formation. In these experiments, an InIm 3D display system prototype that substantially outperforms conventional InIm 3D displays is developed. The prototype achieves a 74.6% reduction in the system PSF and a 29.85% reduction in voxel size, enabling aliasing‐free voxels and a 2.82 times higher resolution in the InIm 3D display. Single-vesicle molecular profiling of cancer-associated extracellular vesicles (EVs) is increasingly being recognized as a powerful tool for cancer detection and monitoring. Mask and target dual imaging is a facile method to quantify the fraction of the molecularly targeted population of EVs in biofluids at the single-vesicle level. However, accurate and efficient dual imaging vesicle analysis has been challenging due to the interference of false signals on the mask images and the need to analyze a large number of images in clinical samples. In this work, we report a fully automatic dual imaging analysis method based on machine learning and use it with dual imaging single-vesicle technology (DISVT) to detect breast cancer at different stages. The convolutional neural network Resnet34 was used along with transfer learning to produce a suitable machine learning model that could accurately identify areas of interest in experimental data. A combination of experimental and synthetic data were used to train the model. Using DISVT and our machine learning-assisted image analysis platform, we determined the fractions of EpCAM-positive EVs and CD24-positive EVs over captured plasma EVs with CD81 marker in the blood plasma of pilot HER2-positive breast cancer patients and compared to those from healthy donors. The amount of both EpCAM-positive and CD24-positive EVs was found negligible for both healthy donors and Stage I patients. The amount of EpCAM-positive EVs (also CD81-positive) increased from 18% to 29% as the cancer progressed from Stage II to III. No significant increase was found with further progression to Stage IV. A similar trend was found for the CD24-positive EVs. Statistical analysis showed that both EpCAM and CD24 markers can detect HER2-positive breast cancer at Stages II, III, or IV. They can also differentiate individual cancer stages except those between Stage III and Stage IV. Due to the simplicity, high sensitivity, and high efficiency, the DISVT with the AI-assisted dual imaging analysis can be widely used for both basic research and clinical applications to quantitatively characterize molecularly targeted EV subtypes in biofluids. Background: Medical devices for fluorescence-guided surgery (FGS) are becoming available at a fast pace. The main challenge for surgeons lies in the lack of in-depth knowledge of optical imaging, different technical specifications and poor standardisation, and the selection of the best device based on clinical application. Methods: This manuscript aims to provide an up-to-date description of the commercially available fluorescence imaging platforms by comparing their mode of use, required settings, image types, compatible fluorophores, regulatory approval, and cost. We obtained this information by performing a broad literature search on PubMed and by contacting medical companies directly. The data for this review were collected up to November 2023. Results: Thirty-two devices made by 19 medical companies were identified. Ten systems are surgical microscopes, 5 can be used for both open and minimally invasive surgery (MIS), 6 can only be used for open surgery, and 10 only for MIS. One is a fluorescence system available for the Da Vinci robot. Nineteen devices can provide an overlay between fluorescence and white light image. All devices are compatible with Indocyanine Green, the most common fluorescence dye used intraoperatively. There is significant variability in the hardware and software of each device, which resulted in different sensitivity, fluorescence intensity, and image quality. All devices are CE-mark regulated, and 30 were FDA-approved. Conclusion: There is a prolific market of devices for FGS and healthcare professionals should have basic knowledge of their technical specifications to use it at best for each clinical indication. Photonic nanomaterials play a crucial role in facilitating the necessary signal for optical brain imaging, presenting a promising avenue for early diagnosis of brain‐related disorders. However, the blood‐brain barrier (BBB) presents a significant challenge, blocking the entry of most molecules or materials from the bloodstream into the brain. To overcome this, photonic nanocrystals in the form of gold clusters (LAuC) with size less than 3 nm, have been developed, with Levodopa conjugated to LAuC (Dop@LAuC) for targeted brain imaging. Dop@LAuC crosses the BBB and emits in the near‐infrared (NIR) wavelength, enabling real‐time optical brain imaging. An in vitro BBB model using brain endothelial cells showed that 50 % of Dop@LAuC crossed the barrier within 3 hours, compared to only 10 % of LAuC, highlighting the enhanced ability of L‐dopa‐conjugated gold clusters to penetrate the BBB. In vivo optical imaging in healthy mice further confirmed the material's efficacy to cross BBB without compromising the barrier integrity. [ABSTRACT FROM AUTHOR]

Background: In the oddball paradigm, the dorsolateral prefrontal cortex (DLPFC) is often associated with active cognitive responses, such as maintaining information in working memory or adapting response strategies. While some evidence points to the DLPFC's role in passive auditory deviance perception, a detailed understanding of the spatiotemporal neurodynamics involved remains unclear. Methods: In this study, event-related optical signals (EROS) and event-related potentials (ERPs) were simultaneously recorded for the first time over the prefrontal cortex using a 64-channel electroencephalography (EEG) system, during passive auditory deviance perception in 12 right-handed young adults (7 women and 5 men). In this oddball paradigm, deviant stimuli (a 1500 Hz pure tone) elicited a negative shift in the N1 ERP component, related to mismatch negativity (MMN), and a significant positive deflection associated with the P300, compared to standard stimuli (a 1000 Hz tone). Results: We hypothesize that the DLPFC not only participates in active tasks but also plays a critical role in processing deviant stimuli in passive conditions, shifting from pre-attentive to attentive processing. We detected enhanced neural activity in the left middle frontal gyrus (MFG), at the same timing of the MMN component, followed by later activation at the timing of the P3a ERP component in the right MFG. Conclusions: Understanding these dynamics will provide deeper insights into the DLPFC's role in evaluating the novelty or unexpectedness of the deviant stimulus, updating its cognitive value, and adjusting future predictions accordingly. However, the small number of subjects could limit the generalizability of the observations, in particular with respect to the effect of handedness, and additional studies with larger and more diverse samples are necessary to validate our conclusions. Background: The tumor microenvironment (TME) plays a critical role in cancer progression and response to immunotherapy. Immunotherapy targeting the immune system has emerged as a promising treatment modality, but challenges in understanding the TME limit its efficacy. Optical imaging strategies offer noninvasive, real‐time insights into the interactions between immune cells and the TME. Objective: This review assesses the progress of optical imaging technologies in monitoring immunotherapy within the TME and explores their potential applications in clinical trials and personalized cancer treatment. Methods: This is a comprehensive literature review based on the advances in optical imaging modalities including fluorescence imaging (FLI), bioluminescence imaging (BLI), and photoacoustic imaging (PAI). These modalities were analyzed for their capacity to provide high‐resolution, real‐time imaging of immune cell dynamics, tumor vasculature, and other critical components of the TME. Results: Optical imaging techniques have shown significant potential in tracking immune cell infiltration, assessing immune checkpoint inhibitors, and visualizing drug delivery within the TME. Technologies like FLI and BLI are pivotal in tracking immune responses in preclinical models, while PAI provides functional imaging with deeper tissue penetration. The integration of these modalities with immunotherapy holds promise for improving treatment monitoring and outcomes. Conclusion: Optical imaging is a powerful tool for understanding the complexities of the TME and optimizing immunotherapy. Further advancements in imaging technologies, combined with nanomaterial‐based approaches, could pave the way for enhanced diagnostic accuracy and therapeutic efficacy in cancer treatment. Objectives Traditional histopathology is a time-intensive and labor-intensive process involving tissue formalin fixation, paraffin embedding, and microtoming into thin sections for H&E staining. Frozen section analysis is a modality used during surgery to quickly evaluate tissue, but it has limitations, such as the size and number of the specimens that can be analyzed as well as difficulties with fatty and bony tissues. Our objective was to investigate the performance of nonlinear microscopy, a fluorescence microscopy technique, for the rapid examination of resected lung tumors. Methods In this proof-of-principle study, nonlinear microscopy imaging of resected lung tissue was performed on a total of 73 tissue specimens collected from 13 patients who underwent lobectomy, segmentectomy, or wedge resection for pulmonary nodules. Results Two pathologists reviewed the digital nonlinear microscopy images in comparison to the corresponding histopathologic H&E slides from a variety of pulmonary pathologies. Conclusions This study demonstrated that nonlinear microscopy readily replicates traditional H&E staining for both lung tumors and nonneoplastic pulmonary structures. Nonlinear microscopy provides many advantages over frozen section analysis and is an optical imaging platform that has the potential to augment rapid pathologic evaluation of resected tissues in the age of digital pathology. In the 2nd century AD, Galen argued that the failure to remove any single 'root' of a malignant tumor could result in a local relapse. After nearly 2 millennia, this problem appears to be even more challenging due to our increased understanding of the complexity of tumor formation and spread. Pathological analysis of tumor margins under a microscope remains the primary and only accepted method for confirming the complete tumor removal. However, this method is not an all-or-nothing test, and it can be compromised by various intrinsic and extrinsic limitations. Among the intrinsic limitations of pathological analysis we recall the pathologist handling, tissue shrinkage, the detection of minimal residual disease and the persistence of a precancerous field. Extrinsic limitations relate to surgical tools and their thermal damage, the different kinds of surgical resections and frozen sections collection. Surgeons, as well as oncologists and radiotherapists, should be well aware of and deeply understand these limitations to avoid misinterpretation of margin status, which can have serious consequences. Meanwhile, new technologies such as Narrow band imaging have shown promising results in assisting with the achievement of clear superficial resection margins. More recently, emerging techniques like Raman spectroscopy and near-infrared fluorescence have shown potential as real-time guides for surgical resection. The aim of this narrative review is to provide valuable insights into the complex process of margin analysis and underscore the importance of interdisciplinary collaboration between pathologists, surgeons, oncologists, and radiotherapists to optimize patient outcomes in oral cancer surgery. Geometric distortions in digital images, caused by factors such as lens defects and changes in camera angles, substantially influence the fidelity of the image by altering pixel positions and shapes. Current geometric distortion correction methods, focusing on specific types of distortions and relying on high computational resources, face limitations in universality and practicality across diverse real-world applications. We propose here a two-stage distortion correction method that integrates deep learning with traditional image registration algorithms for correcting multiple types of geometric distortion. Compared to state-of-the-art correction methods, our proposed method demonstrates flexibility, capable of addressing a wide range of geometric distortions and achieves superior correction results with fewer parameters. In addition, tests performed on synthetic datasets show an improvement of 10.39% for PSNR, 30.42% for SSIM, and 85% for processing speed, compared to the best performing methods to our knowledge. Finally, experiments with handheld medical endoscopic scanners confirm the applicability and robustness of our method in real-world scenarios. Our method offers a versatile and efficient solution for geometric distortion correction, suitable for various applications, including medical imaging and resource-limited embedded systems. Due to the continuous increase in the globalized outsourcing of printed circuit board (PCB) fabrication, PCB counterfeits have increased by a significant margin, necessitating rapid and advanced hardware assurance techniques. PCB image segmentation is the primary step in PCB assurance. Over the years, few PCB component segmentation methods have been proposed, and none of those has provided a definite performance benchmark. Besides, those methods have not discussed how the performance is correlated with underlying data or annotation quality. This work presents a PCB image segmentation benchmark. In addition, we explore how annotation quality affects component segmentation and present possible future research directions to work with coarse annotations to alleviate the human effort behind full data annotation tasks. We have analyzed the performance of the preferred deep neural network (DNN) architecture and Transformer architecture with the data annotation quality and presented the direction to leverage the outcome with limited quality annotations. Finally, we present the qualitative as well as the quantitative results to demonstrate the performance of our techniques and provide observations and future research directions on the overall task. Currently, one in three adults worldwide may be obese or overweight. Obesity is characterized by an excess of energy‐storing white fat. An ingenious and sought‐after strategy against obesity is to activate energy‐consuming beige fat, which is converted from white fat or brown fat. However, existing tools for assessing brown or beige fat activation in vivo have certain limitations, such as being cumbersome and expensive. Optical imaging is a relatively straightforward and economical imaging technique that utilizes light to peer into the structural–functional information of living organisms at multiple scales. Despite the availability of various optical imaging modalities for detecting brown or beige fat, there is a dearth of literature summarizing relevant studies. Accordingly, this review focuses on these optical modalities and elaborates on their imaging principles, characteristics, and recent research advances in the detection of brown or beige fat. Their imaging targets, advantages, and disadvantages are further concluded. As a methodological reference, this review can guide the selection of optimal optical modalities to noninvasively profile brown or beige fat activation from a specific biological perspective, maximizing the potential of optical imaging in anti‐obesity assessment. Postoperative bleeding is the most significant complication of tonsillectomy. Regular monitoring of post‐surgical wound healing in the pharynx is required. For this purpose, we propose endoscope‐based non‐invasive perfusion mapping and quantification. The combination of imaging photoplethysmography and image processing provides automated wound area selection and microcirculation characterization. In this feasibility study, we demonstrate the first results of the proposed approach to wound monitoring in clinical trial on eight patients after tonsillectomy. Combination of probe‐based optical system and image processing algorithms can provide the valuable and consistent data on perfusion distribution. The quantitative microcirculation data obtained 1, 4, and 7 days after surgery are in good agreement with existing monitoring protocols. [ABSTRACT FROM AUTHOR]

Metal nanoclusters, due to their unique photophysical properties, exhibit tremendous potential applications in the biomedical fields of biosensing, bioimaging, and photodynamic and photothermal therapies. This review systematically discusses the photoreactive behavior and application progress of metal nanoclusters in biomedical diagnostics and treatment, especially the electronic behavior after photosensitization treatment and their roles in various biomedical applications. Initially, the review introduces the electronic transition processes of metal nanoclusters after absorbing light energy, along with their applications in biosensing and imaging. Subsequently, the review analyzes the mechanisms of action of metal nanoclusters in photodynamic and photothermal therapies. Lastly, the review proposes the prospects and challenges of metal nanoclusters in biomedical applications, with the hope of providing a reference for further research and application of metal nanocluster materials. The human brain is a complex organ controlling daily activity. Present technique models have mostly focused on multi-layer brain tissues, which lack understanding of the propagation characteristics of various single brain tissues. To better understand the influence of different optical source types on individual brain tissues, we constructed single-layer brain models and simulated optical propagation using the Monte Carlo method. Based on the optical simulation results, sixteen optical source types had different optical energy distributions, and the distribution in cerebrospinal fluid had obvious characteristics. Five brain tissues (scalp, skull, cerebrospinal fluid, gray matter, and blood vessel) had the same set of the first three optical source types with maximum depth, while white matter had a different set of the first three optical source types with maximum depth. Each brain tissue had different optical source types with the maximum and minimum full width at half maximum. The study on single-layer brain tissues under different optical source types lays the foundation for constructing complex brain models with multiple tissue layers. The incorporation of gold nanoparticles (GNPs) into retinal imaging signifies a notable advancement in ophthalmology, offering improved accuracy in diagnosis and patient outcomes. This review explores the synthesis and unique properties of GNPs, highlighting their adjustable surface plasmon resonance, biocompatibility, and excellent optical absorption and scattering abilities. These features make GNPs advantageous contrast agents, enhancing the precision and quality of various imaging modalities, including photoacoustic imaging, optical coherence tomography, and fluorescence imaging. This paper analyzes the unique properties and corresponding mechanisms based on the morphological features of GNPs, highlighting the potential of GNPs in retinal disease diagnosis and management. Given the limitations currently encountered in clinical applications of GNPs, the approaches and strategies to overcome these limitations are also discussed. Introduction: Fibromyalgia syndrome (FMS) is a prevalent rheumatic disorder, and its pathogenesis includes genetic, neuroendocrine, and autonomic abnormalities, which may impact ocular structures. The aim was to conduct a comparative analysis of the ophthalmic vasculature and the retinal nerve fiber layer (RNFL) thickness between FMS and control groups using optical coherence tomography (OCT) and OCT angiography (OCTA). Methods: This cross-sectional comparative study included 43 FMS patients and 40 healthy controls recruited from a tertiary education and research hospital between January 2024 and May 2024. All patients satisfied the 2016 American College of Rheumatology criteria for FMS and consented. OCT and OCTA were used to assess the RNFL thickness and the retinal microvasculature structure. The Fibromyalgia Impact Questionnaire (FIQ) was performed to evaluate disease severity. Results: The study found significantly higher total retinal parafoveal thickness and foveal density in FMS patients (p = 0.017 and p = 0.044, respectively). Nevertheless, there were no significant differences among the groups concerning total retinal foveal thickness, foveal avascular zone characteristics, superficial and deep capillary plexus densities, choriocapillaris flow area, and outer retinal flow area values (p > 0.05). The RNFL thickness in all quadrants did not reveal significant differences between the groups (p > 0.05). Furthermore, there was no significant correlation between FIQ scores and OCTA parameters or RNFL thickness values (p > 0.05). Conclusion: The study revealed slight differences in retinal parafoveal thickness and foveal density in FMS patients, but no substantial vascular or neurodegenerative alterations were observed compared to healthy controls. Extracellular vesicles (EVs), nanosized lipid bilayer vesicles released by nearly all types of cells, play pivotal roles as intercellular signaling mediators with diverse biological activities. Their adaptability has attracted interest in exploring their role as disease biomarker theranostics. However, the in vivo biodistribution and pharmacokinetic profiles of EVs, particularly following administration into living subjects, remain unclear. Thus, in vivo imaging is vital to enhance our understanding of the homing and retention patterns, blood and tissue half‐life, and excretion pathways of exogenous EVs, thereby advancing real‐time monitoring within biological systems and their therapeutic applications. This review examines state‐of‐the‐art methods including EV labeling with various agents, including optical imaging, magnetic resonance imaging, and nuclear imaging. The strengths and weaknesses of each technique are comprehensively explored, emphasizing their clinical translation. Despite the potential of EVs as cancer theranostics, achieving a thorough understanding of their in vivo behavior is challenging. This review highlights the urgency of addressing current questions in the biology and therapeutic applications of EVs. It underscores the need for continued research to unravel the complexities surrounding EVs and their potential clinical implications. By identifying these challenges, this review contributes to ongoing efforts to optimize EV imaging techniques for clinical use. Toxic environmental pollutants pose a health risk for both humans and animals. Accumulation of industrial contaminants in freshwater fish may become a significant threat to biodiversity. Comprehensive monitoring of the impact of environmental stressors on fish functional systems is important and use of non‐invasive tools that can detect the presence of these toxicants in vivo is desirable. The blood circulatory system, by virtue of its sensitivity to the external stimuli, could be an informative indicator of chemical exposure. In this study, microscopic photoplethysmography‐based approach was used to investigate the cardiac activity in broad whitefish larvae (Coregonus nasus) under acute exposure to cadmium and phenol. We identified contamination‐induced abnormalities in the rhythms of the ventricle and atrium. Our results allow introducing additional endpoints to evaluate the cardiac dysfunction in fish larvae and contribute to the non‐invasive evaluation of the toxic effects of industrial pollutants on bioaccumulation and aquatic life. BACKGROUND: The obesity epidemic brought a need for accessible methods to monitor body composition, as excess adiposity has been associated with cardiovascular disease, metabolic disorders, and some cancers. Recent 3-dimensional optical (3DO) imaging advancements have provided opportunities for assessing body composition. However, the accuracy and precision of an overall 3DO body composition model in specific subgroups are unknown. OBJECTIVES: This study aimed to evaluate 3DOs accuracy and precision by subgroups of age, body mass index, and ethnicity. METHODS: A cross-sectional analysis was performed using data from the Shape Up! Adults study. Each participant received duplicate 3DO and dual-energy X-ray absorptiometry (DXA) scans. 3DO meshes were digitally registered and reposed using Meshcapade. Principal component analysis was performed on 3DO meshes. The resulting principal components estimated DXA whole-body and regional body composition using stepwise forward linear regression with 5-fold cross-validation. Duplicate 3DO and DXA scans were used for test-retest precision. Students t tests were performed between 3DO and DXA by subgroup to determine significant differences. RESULTS: Six hundred thirty-four participants (females = 346) had completed the study at the time of the analysis. 3DO total fat mass in the entire sample achieved R2 of 0.94 with root mean squared error (RMSE) of 2.91 kg compared to DXA in females and similarly in males. 3DO total fat mass achieved a % coefficient of variation (RMSE) of 1.76% (0.44 kg), whereas DXA was 0.98% (0.24 kg) in females and similarly in males. There were no mean differences for total fat, fat-free, percent fat, or visceral adipose tissue by age group (P > 0.068). However, there were mean differences for underweight, Asian, and Black females as well as Native Hawaiian or other Pacific Islanders (P < 0.038). CONCLUSIONS: A single 3DO body composition model produced accurate and precise body composition estimates that can be used on diverse populations. However, adjustments to specific subgroups may be warranted to improve the accuracy in those that had significant differences. Neurofeedback allows individuals to monitor and self-regulate their brain activity, potentially improving human brain function. Beyond the traditional electrophysiological approach using primarily electroencephalography, brain haemodynamics measured with functional magnetic resonance imaging (fMRI) and more recently, functional near-infrared spectroscopy (fNIRS) have been used (haemodynamic-based neurofeedback), particularly to improve the spatial specificity of neurofeedback. Over recent years, especially fNIRS has attracted great attention because it offers several advantages over fMRI such as increased user accessibility, cost-effectiveness and mobility—the latter being the most distinct feature of fNIRS. The next logical step would be to transfer haemodynamic-based neurofeedback protocols that have already been proven and validated by fMRI to mobile fNIRS. However, this undertaking is not always easy, especially since fNIRS novices may miss important fNIRS-specific methodological challenges. This review is aimed at researchers from different fields who seek to exploit the unique capabilities of fNIRS for neurofeedback. It carefully addresses fNIRS-specific challenges and offers suggestions for possible solutions. If the challenges raised are addressed and further developed, fNIRS could emerge as a useful neurofeedback technique with its own unique application potential—the targeted training of brain activity in real-world environments, thereby significantly expanding the scope and scalability of haemodynamic-based neurofeedback applications. It works by the application of direct fluorescence and the loss of fluorescence in visible and non visible high-risk oral lesions. Numerous publications have offered insights on the possible use of VELscope® for identifying oral mucosal lesions. By analysing patterns in publication outputs, citation metrics and collaborative networks, researchers can gain insights into the development, influence and key contributors in the field of VELscope®, guiding future research directions and policy decisions. Aim: To track down and analyse all studies published on the application of VELscope® in the diagnosis of oral mucosal lesions and to assist future researchers by highlighting the most cited earlier publications. Materials and Methods: This bibliometric analysis was conducted using articles published on the applications of VELscope® in detecting oral mucosal lesions between 2006 and 2022. In December 2023, a comprehensive search was performed on PubMed and Scopus databases using medical subject heading terms like “VELscope®,” “Autofluorescence imaging system,” and “Oral” in several combinations. Bibliographic data regarding the top 100 most cited articles were extracted and collated. Results: The analysis showed that the most contributions came from the journals “Oral Oncology,” “Oral Diseases,” and “Photodiagnosis and Photodynamic Therapy,” with significant contributions from researchers in the USA and India. The highest number of publications occurred in 2015, with a notable peak in the decade 2011-2020. The articles were cumulatively cited 4,559 times. Conclusion: The current bibliographic analysis provided valuable insights into the development and impact of research on the applications of VELscope® in dentistry by highlighting publication trends, influential works, leading authors and collaborative networks. These insights can guide future research directions, inform policy decisions and enhance understanding of the evolution and scholarly influence.

Abstract Background Tooth segmentation on intraoral scanned (IOS) data is a prerequisite for clinical applications in digital workflows. Current state-of-the-art methods lack the robustness to handle variability in dental conditions. This study aims to propose and evaluate the performance of a convolutional neural network (CNN) model for automatic tooth segmentation on IOS images. Methods A dataset of 761 IOS images (380 upper jaws, 381 lower jaws) was acquired using an intraoral scanner. The inclusion criteria included a full set of permanent teeth, teeth with orthodontic brackets, and partially edentulous dentition. A multi-step 3D U-Net pipeline was designed for automated tooth segmentation on IOS images. The model’s performance was assessed in terms of time and accuracy. Additionally, the model was deployed on an online cloud-based platform, where a separate subsample of 18 IOS images was used to test the clinical applicability of the model by comparing three modes of segmentation: automated artificial intelligence-driven (A-AI), refined (R-AI), and semi-automatic (SA) segmentation. Results The average time for automated segmentation was 31.7 ± 8.1 s per jaw. The CNN model achieved an Intersection over Union (IoU) score of 91%, with the full set of teeth achieving the highest performance and the partially edentulous group scoring the lowest. In terms of clinical applicability, SA took an average of 860.4 s per case, whereas R-AI showed a 2.6-fold decrease in time (328.5 s). Furthermore, R-AI offered higher performance and reliability compared to SA, regardless of the dentition group. Conclusions The 3D U-Net pipeline was accurate, efficient, and consistent for automatic tooth segmentation on IOS images. The online cloud-based platform could serve as a viable alternative for IOS segmentation.

The visualization of drugs in living systems has become key techniques in modern therapeutics. Recent advancements in optical imaging technologies and molecular design strategies have revolutionized drug visualization. At the subcellular level, super-resolution microscopy has allowed exploration of the molecular landscape within individual cells and the cellular response to drugs. Moving beyond subcellular imaging, researchers have integrated multiple modes, like optical near-infrared II imaging, to study the complex spatiotemporal interactions between drugs and their surroundings. By combining these visualization approaches, researchers gain supplementary information on physiological parameters, metabolic activity, and tissue composition, leading to a comprehensive understanding of drug behavior. This review focuses on cutting-edge technologies in drug visualization, particularly fluorescence imaging, and the main types of fluorescent molecules used. Additionally, we discuss current challenges and prospects in targeted drug research, emphasizing the importance of multidisciplinary cooperation in advancing drug visualization. With the integration of advanced imaging technology and molecular design, drug visualization has the potential to redefine our understanding of pharmacology, enabling the analysis of drug micro-dynamics in subcellular environments from new perspectives and deepening pharmacological research to the levels of the cell and organelles.

Optical imaging is crucial for cell examination, but its instruments' bulky optics limit their flexibility, and a single wavelength provides limited sample information. This paper presents a fibre cantilever-based endoscopic scanner for multiwavelength reflectance confocal microscopy. The scanner utilises the dual-core fibre cantilever to produce Lissajous scans while accommodating two distinct wavelengths. Coloured surfaces were imaged at wavelengths of 520 nm, 635 nm and 850 nm, and verified with a colour chart and reflectance spectroscopy. Blue ink-stained plant stem cross-section samples were also imaged and compared. Furthermore, the scanner performed structure morphology imaging on cellular samples. The 520 nm and 635 nm wavelengths create low- and high-contrast images respectively, while the 850 nm wavelength delivers a sharper image while also capturing the surface profile of the target sample. The findings demonstrated the endoscopic scanner’s capability to extract information based on different wavelengths, providing a more diverse view of the sample and enhancing the surface profile. Using 635 nm and 850 nm, the reconstructed images have a contrast enhancement of 67.9% and 90.8% respectively from 520 nm. The smallest feature size measured on the cellular sample is 5 µm. The endoscopic scanner has the potential to study microfluidic and cell morphology.

Background/Objectives: Three-dimensional (3D) analysis of maxillofacial structures in dysmorphic patients offers clinical advantages over 2D analysis due to its high accuracy and precision in measuring many morphological parameters. Currently, no reliable gold standard exists for calculating 3D volumetric measurements of maxillofacial structures when captured by 3D surface imaging techniques. The aim of this scoping review is to provide an overview of the scientific literature related to 3D surface imaging methods used for volumetric analysis of the dysmorphic maxillofacial structures of patients affected by CL/P or other syndromes and to provide an update on the existing protocols, methods, and, when available, reference data. Methods: A total of 17 papers selected according to strict inclusion and exclusion criteria were reviewed for the qualitative analysis out of more than 4500 articles published between 2002 and 2024 that were retrieved from the main electronic scientific databases according to the PRISMA-ScR guidelines. A qualitative synthesis of the protocols used for the selection of the anatomical areas of interest and details on the methods used for the calculation of their volume was completed. Results: The results suggest a great degree of heterogeneity between the reviewed studies in all the aspects analysed (patient population, anatomical structure, area selection, and volume calculation), which prevents any chance of direct comparison between the reported volumetric data. Conclusions: Our qualitative analysis revealed dissimilarities in the procedures specified in the studies, highlighting the need to develop uniform methods and protocols and the need for comparative studies to verify the validity of methods in order to achieve high levels of scientific evidence, homogeneity of volumetric data, and clinical consensus on the methods to use for 3D volumetric surface-based analysis. Photodynamic therapy (PDT) is a light-based anticancer therapy that can induce tumor necrosis and/or apoptosis. Two important factors contributing to the efficacy of PDT are the concentration of the photosensitizer in the tumor tissue and its preferential accumulation in the tumor tissue compared to that in normal tissues. In this study, we investigated the use of optical imaging for monitoring whole-body bio-distribution of the fluorescent (660 nm) photosensitizer Bremachlorin in vivo, in a murine pancreatic ductal adenocarcinoma (PDAC) model. Moreover, we non-invasively, examined the induction of tumor necrosis after PDT treatment using near-infrared fluorescent imaging of the necrosis avid cyanine dye IRDye®-800CW Carboxylate. Using whole-body fluorescence imaging, we observed that Bremachlorin preferentially accumulated in pancreatic tumors. Furthermore, in a longitudinal study we showed that 3 hours after Bremachlorin administration, the fluorescent tumor signal reached its maximum. In addition, the tumor-to-background ratio at all-time points was approximately 1.4. Ex vivo, at 6 hours after Bremachlorin administration, the tumor-to-muscle or -normal pancreas ratio exhibited a greater difference than it did at 24 hours, suggesting that, in terms of efficacy, 6 hours after Bremachlorin administration was an effective time point for PDT treatment of PDAC. In vivo administration of the near infrared fluorescence agent IRDye®-800CW Carboxylate showed that PDT, 6 hours after administration of Bremachlorin, selectively induced necrosis in the tumor tissues, which was subsequently confirmed histologically. In conclusion, by using in vivo fluorescence imaging, we could non-invasively and longitudinally monitor, the whole-body distribution of Bremachlorin. Furthermore, we successfully used IRDye®-800CW Carboxylate, a near-infrared fluorescent necrosis avid agent, to image PDT-induced necrotic cell death as a measure of therapeutic efficacy. Purpose: Prostate-specific membrane antigen (PSMA) is increasingly used to image prostate cancer in clinical practice. We sought to develop and test a humanised PSMA minibody IAB2M conjugated to the fluorophore IRDye 800CW-NHS ester in men undergoing robot-assisted laparoscopic radical prostatectomy (RARP) to image prostate cancer cells during surgery. Methods: The minibody was evaluated pre-clinically using PSMA positive/negative xenograft models, following which 23 men undergoing RARP between 2018 and 2020 received between 2.5 mg and 20 mg of IR800-IAB2M intravenously, at intervals between 24 h and 17 days prior to surgery. At every step of the procedure, the prostate, pelvic lymph node chains and extra-prostatic surrounding tissue were imaged with a dual Near-infrared (NIR) and white light optical platform for fluorescence in vivo and ex vivo. Histopathological evaluation of intraoperative and postoperative microscopic fluorescence imaging was undertaken for verification. Results: Twenty-three patients were evaluated to optimise both the dose of the reagent and the interval between injection and surgery and secure the best possible specificity of fluorescence images. Six cases are presented in detail as exemplars. Overall sensitivity and specificity in detecting non-lymph-node extra-prostatic cancer tissue were 100% and 65%, and 64% and 64% respectively for lymph node positivity. There were no side-effects associated with administration of the reagent. Conclusion: Intraoperative imaging of prostate cancer tissue is feasible and safe using IR800-IAB2M. Further evaluation is underway to assess the benefit of using the technique in improving completion of surgical excision during RARP. TFEB and TFE3 belong to the MiT/TFE family of transcription factors that bind identical DNA responsive elements in the regulatory regions of target genes. They are involved in regulating lysosomal biogenesis, function, exocytosis, autophagy, and lipid catabolism. Precise control of TFEB and TFE3 activity is crucial for processes such as senescence, stress response, energy metabolism, and cellular catabolism. Dysregulation of these factors is implicated in various diseases, thus researchers have explored pharmacological approaches to modulate MiT/TFE activity, considering these transcription factors as potential therapeutic targets. However, the physiological complexity of their functions and the lack of suitable in vivo tools have limited the development of selective MiT/TFE modulating agents. Here, we have created a reporter-based biosensor, named CLEARoptimized, facilitating the pharmacological profiling of TFEB- and TFE3-mediated transcription. This innovative tool enables the measurement of TFEB and TFE3 activity in living cells and mice through imaging and biochemical techniques. CLEARoptimized consists of a promoter with six coordinated lysosomal expression and regulation motifs identified through an in-depth bioinformatic analysis of the promoters of 128 TFEB-target genes. The biosensor drives the expression of luciferase and tdTomato reporter genes, allowing the quantification of TFEB and TFE3 activity in cells and in animals through optical imaging and biochemical assays. The biosensor's validity was confirmed by modulating MiT/TFE activity in both cell culture and reporter mice using physiological and pharmacological stimuli. Overall, this study introduces an innovative tool for studying autophagy and lysosomal pathway modulation at various biological levels, from individual cells to the entire organism. Surface reconstruction determines the fate of catalytic sites on the near-surface during the oxygen evolution reaction. However, deciphering the conversion mechanism of various intermediate-states during surface reconstruction remains a challenge. Herein, we employed an optical imaging technique to draw the landscape of dynamic surface reconstruction on individual Co3O4 nanoparticles. By regulating the surface states of Co3O4 nanoparticles, we explored dynamic growth of the CoOx(OH)y sublayer on single Co3O4 nanoparticles and directly identified the conversion between two dynamics. Rich oxygen vacancies induced more active sites on the surface and prolonged surface reconstruction, which enhanced electrochemical redox and oxygen evolution. These results were further verified by in situ electrochemical extinction spectroscopy of single Co3O4 nanoparticles. We elucidate the heterogeneous evolution of surface reconstruction on individual Co3O4 nanoparticles and present a unique perspective to understand the fate of catalytic species on the nanosurface, which is of enduring significance for investigating the heterogeneity of multielectron-transfer events. Objectives Methods Results Conclusions To systematically assess the current available literature concerning advanced optical imaging methods for the detection and diagnosis of bladder cancer (BCa), focusing particularly on the sensitivity and specificity of these techniques.First a scoping search was performed to identify all available optical techniques for BCa detection and diagnosis. The optical imaging techniques used for detecting BCa are: the Storz professional image enhancement system (IMAGE1 S), narrow‐band imaging (NBI), photoacoustic imaging (PAI), autofluorescence imaging (AFI), photodynamic diagnosis (PDD), and scanning fibre endoscopy (SFE). The staging and grading techniques for BCa are: optical coherence tomography (OCT), confocal laser endomicroscopy (CLE), Raman spectroscopy, endocytoscopy, and non‐linear optical microscopy (NLO). Then a systematic literature search was conducted using MEDLINE, EMBASE and Web of Science from inception to 21 November 2023. Articles were screened and selected by two independent reviewers. Inclusion criteria were: reporting on both the sensitivity and specificity of a particular technique and comparison to histopathology, and in the case of a detection technique comparison to white light cystoscopy (WLC).Out of 6707 articles, 189 underwent full‐text review, resulting in 52 inclusions. No articles met criteria for IMAGE1 S, PAI, SFE, Raman spectroscopy, and endocytoscopy. All detection techniques showed higher sensitivity than WLC, with NBI leading (87.8–100%). Overall, detection technique specificity was comparable to WLC, with PDD being most specific (23.3–100%). CLE and OCT varied in sensitivity and specificity, with OCT showing higher specificity for BCa diagnosis, notably for carcinoma in situ (97–99%) compared to CLE (62.5–81.3%). NLO demonstrated high sensitivity and specificity (90–97% and 77–100%, respectively) based on limited data from two small ex vivo studies.Optical techniques with the most potential are PDD for detecting and OCT for staging and grading BCa. Methods: A dataset of 761 IOS images (380 upper jaws, 381 lower jaws) was acquired using an intraoral scanner. The inclusion criteria included a full set of permanent teeth, teeth with orthodontic brackets, and partially edentulous dentition. A multi-step 3D U-Net pipeline was designed for automated tooth segmentation on IOS images. The model's performance was assessed in terms of time and accuracy. Additionally, the model was deployed on an online cloud-based platform, where a separate subsample of 18 IOS images was used to test the clinical applicability of the model by comparing three modes of segmentation: automated artificial intelligence-driven (A-AI), refined (R-AI), and semi-automatic (SA) segmentation. Results: The average time for automated segmentation was 31.7 ± 8.1 s per jaw. The CNN model achieved an Intersection over Union (IoU) score of 91%, with the full set of teeth achieving the highest performance and the partially edentulous group scoring the lowest. In terms of clinical applicability, SA took an average of 860.4 s per case, whereas R-AI showed a 2.6-fold decrease in time (328.5 s). Furthermore, R-AI offered higher performance and reliability compared to SA, regardless of the dentition group. Conclusions: The 3D U-Net pipeline was accurate, efficient, and consistent for automatic tooth segmentation on IOS images. Skin diseases are a thorny and worrying matter, due to the special nature of the diseases and their similarity in the distinctive features of each disease. This makes doctors face difficulty in arriving at the correct diagnosis. There are two ways to determine the type of skin disease: the surgical method, which is a painful and expensive method and requires special preparations, and the second method is a non-invasive imaging the lesion at an acceptable magnification rate that preserves the important features of the disease and applies one of the diagnostic rules. The non-invasive skin scanning method, which makes it possible to detect microscopically small features that are invisible to the unaided eyes is optical imaging of skin disorders. For efficiently recognizing physical structure and Stages of development of skin disease and its response to therapy, non-invasive, and less painful scanning methods, such as confocal laser microscopy (CLM), multi-photon microscopy (MP), Raman microscopy (RM), photo-acoustic microscopy (PAM), optical coherence microscopy (OCT), and dermatoscopy (DMY), have been enhanced and implemented for skin scanning. Optical imaging is a type of skin surface imaging that offers precise visualization of skin features. The DMY is a non-intrusive human body tool for evaluating skin disorders. The device is portable and auto-illumination, which imaging skin structure that is often invisible to the naked eye. DMY produces digital images and records for re-examination for later use. There are 3-types of DMYs, traditional non-polarized light contact DMY (TNPC), polarized light contact DMY (PC), and polarized light contactless DMY (PCS). TNPC-DMY needs to touch the covered skin with an interfacing liquid. PC provides better penetration than TNPC and interfacing liquid is not required. OCT is a light-reflecting imaging system. It produces direct scanning with a resolution of around 10 µm plus a depth into the skin of up to 2*10−3 m, without needing to print or store. It has been utilized frequently in numerous disorders as well as skin scanning. The technology produces efficient visualization of changes in normal skin tissue. OCT results and histological structure show significant agreement in studies of skin tumors, and inflammatory disorders. OCT is available in a variety of designs, including time-variants (TV-OCT), Fourier-domain (FD-OCT), line field confocal (LC-OCT), spectral domain (SD-OCT), and high-resolution (HR-OCT). Therefore, this study, presents an overview of fundamental concept, design, for many various types of each technique, evaluation, and weaknesses according to practical tests and searches. This is very useful for researchers in the field of skin diseases detection, particularly in the field of automated diagnosis by artificial intelligence where the researcher is intimately familiar with the stages of development of diagnostic mechanisms and how traditional methods have contributed development in computer aided diagnosis. The two majors widely used optical imaging methods for skin are dermatoscopy and OCT, which are improving dermatoscopy diagnostic efficacy by five to thirty percent. The outcomes of this research are summarized in the next points, mentioned techniques rely on the kind of lesion being examined and the dermatologist's skill. Significance: Perturbations in the microcirculatory system have been observed in neurological conditions, such as Alzheimer's disease or systemic inflammation. However, changes occurring at the level of the capillary are difficult to translate to biomarkers that could be measured macroscopically. Aim: We aim to evaluate whether transit time changes reflect capillary stalling and to what degree. Approach: We employ a combined spectral optical coherence tomography (OCT) and fluorescence optical imaging (FOI) system to investigate the relation between capillary stalling and transit time in a mouse model of systemic inflammation induced by intraperitoneal injection of lipopolysaccharide. Angiograms are obtained using OCT, and fluorescence signal images are acquired by the FOI system upon intravenous injection of fluorescein isothiocyanate via a catheter inserted into the tail vein. Results: Our findings reveal that lipopolysaccharide (LPS) administration significantly increases both the percentage and duration of capillary stalling compared to mice receiving a 0.9% saline injection. Moreover, LPS-induced mice exhibit significantly prolonged arteriovenous transit time compared to control mice. Conclusions: These observations suggest that capillary stalling, induced by inflammation, modulates cerebral mean transit time, a measure that has translational potential. [ABSTRACT FROM AUTHOR]

The brain enables adaptive behavior via the dynamic coordination of diverse neuronal signals across spatial and temporal scales: from fast action potential patterns in microcircuits to slower patterns of distributed activity in brain-wide networks. Understanding principles of multiscale dynamics requires simultaneous monitoring of signals in multiple, distributed network nodes. Combining optical and electrical recordings of brain activity is promising for collecting data across multiple scales and can reveal aspects of coordinated dynamics invisible to standard, singlemodality approaches. We review recent progress in combining opto- and electrophysiology, focusing on mouse studies that shed new light on the function of single neurons by embedding their activity in the context of brain-wide activity patterns. Optical and electrical readouts can be tailored to desired scales to tackle specific questions. For example, fast dynamics in single cells or local populations recorded with multi-electrode arrays can be related to simultaneously acquired optical signals that report activity in specified subpopulations of neurons, in non-neuronal cells, or in neuromodulatory pathways. Conversely, two-photon imaging can be used to densely monitor activity in local circuits while sampling electrical activity in distant brain areas at the same time. Genetically encoded voltage indicators (GEVIs) are protein-based optical sensors that allow for measurements from genetically defined populations of neurons. Although in vivo imaging in the mammalian brain with early generation GEVIs was difficult due to poor membrane expression and low signal-to-noise ratio, newer and more sensitive GEVIs have begun to make them useful for answering fundamental questions in neuroscience. We discuss principles of imaging using GEVIs and genetically encoded calcium indicators, both useful tools for in vivo imaging of neuronal activity, and review some of the recent mechanistic advances that have led to GEVI improvements. We provide an overview of the mouse olfactory bulb (OB) and discuss recent studies using the GEVI ArcLight to study different cell types within the bulb using both widefield and two-photon microscopy. Specific emphasis is placed on using GEVIs to begin to study the principles of concentration coding in the OB, how to interpret the optical signals from population measurements in the in vivo brain, and future developments that will push the field forward. The numerical calculation of partially coherent imaging involves a fourfold integral, which is numerically complex and impracticable to be calculated directly. The use of coherent-mode decomposition (CMD) can make this problem more manageable but finding the coherent-modes for complicated partial coherent fields (without already-known coherent-mode expansion) are rather computationally intensive. In this letter, a modified modal expansion method is proposed, which significantly reduces the requirement of computational resources. The propagation of partial coherence in imaging systems with extremely large sampling number could be handled by an ordinary computer. A comparison between the new method and the traditional method in terms of memory resource requirements and computational time consumption is also detailed in this article. Optical imaging is crucial for cell examination, but its instruments' bulky optics limit their flexibility, and a single wavelength provides limited sample information. This paper presents a fibre cantilever-based endoscopic scanner for multiwavelength reflectance confocal microscopy. The scanner utilises the dual-core fibre cantilever to produce Lissajous scans while accommodating two distinct wavelengths. Coloured surfaces were imaged at wavelengths of 520 nm, 635 nm and 850 nm, and verified with a colour chart and reflectance spectroscopy. Blue ink-stained plant stem cross-section samples were also imaged and compared. Furthermore, the scanner performed structure morphology imaging on cellular samples. The 520 nm and 635 nm wavelengths create low- and high-contrast images respectively, while the 850 nm wavelength delivers a sharper image while also capturing the surface profile of the target sample. The findings demonstrated the endoscopic scanner's capability to extract information based on different wavelengths, providing a more diverse view of the sample and enhancing the surface profile. Using 635 nm and 850 nm, the reconstructed images have a contrast enhancement of 67.9% and 90.8% respectively from 520 nm. The smallest feature size measured on the cellular sample is 5 µm. Mild cognitive impairment (MCI) is common in people with chronic kidney disease (CKD), and its prevalence increases with progressive loss of kidney function. MCI is characterized by a decline in cognitive performance greater than expected for an individual age and education level but with minimal impairment of instrumental activities of daily living. Deterioration can affect one or several cognitive domains (attention, memory, executive functions, language, and perceptual motor or social cognition). Given the increasing prevalence of kidney disease, more and more people with CKD will also develop MCI causing an enormous disease burden for these individuals, their relatives, and society. However, the underlying pathomechanisms are poorly understood, and current therapies mostly aim at supporting patients in their daily lives. This illustrates the urgent need to elucidate the pathogenesis and potential therapeutic targets and test novel therapies in appropriate preclinical models. Here, we will outline the necessary criteria for experimental modeling of cognitive disorders in CKD. We discuss the use of mice, rats, and zebrafish as model systems and present valuable techniques through which kidney function and cognitive impairment can be assessed in this setting. AbstractThe rise of infectious diseases as a public health concern has necessitated the development of rapid and precise diagnostic methods. Imaging techniques like nuclear and optical imaging provide the ability to diagnose infectious diseases within the body, eliminating delays caused by sampling and pre-enrichments of clinical samples and offering spatial information that can aid in a more informed diagnosis. Traditional molecular probes are typically created to image infected tissue without accurately identifying the pathogen. In contrast, oligonucleotides can be tailored to target specific RNA sequences, allowing for the identification of pathogens, and even generating antibiotic susceptibility profiles by focusing on drug resistance genes. Despite the benefits that nucleic acid mimics (NAMs) have provided in terms of stabilizing oligonucleotides, the inadequate delivery of these relatively large molecules into the cytoplasm of bacteria remains a challenge for widespread use of this technology. This review summarizes the key advancements in the field of oligonucleotide probes for in vivo imaging, highlighting the most promising delivery systems described in the literature for developing optical imaging through in vivo hybridization. The visualization of drugs in living systems has become key techniques in modern therapeutics. Recent advancements in optical imaging technologies and molecular design strategies have revolutionized drug visualization. At the subcellular level, super-resolution microscopy has allowed exploration of the molecular landscape within individual cells and the cellular response to drugs. Moving beyond subcellular imaging, researchers have integrated multiple modes, like optical near-infrared II imaging, to study the complex spatiotemporal interactions between drugs and their surroundings. By combining these visualization approaches, researchers gain supplementary information on physiological parameters, metabolic activity, and tissue composition, leading to a comprehensive understanding of drug behavior. This review focuses on cutting-edge technologies in drug visualization, particularly fluorescence imaging, and the main types of fluorescent molecules used. Additionally, we discuss current challenges and prospects in targeted drug research, emphasizing the importance of multidisciplinary cooperation in advancing drug visualization. With the integration of advanced imaging technology and molecular design, drug visualization has the potential to redefine our understanding of pharmacology, enabling the analysis of drug micro-dynamics in subcellular environments from new perspectives and deepening pharmacological research to the levels of the cell and organelles. INTRODUCTION: Delirium is associated with mortality and new onset dementia, yet the underlying pathophysiology remains poorly understood. Development of imaging biomarkers has been difficult given the challenging nature of imaging delirious patients. Diffuse optical tomography (DOT) offers a promising approach for investigating delirium given its portability and three‐dimensional capabilities. METHODS: Twenty‐five delirious and matched non‐delirious patients (n = 50) were examined using DOT, comparing cerebral oxygenation and functional connectivity in the prefrontal cortex during and after an episode of delirium. RESULTS: Total hemoglobin values were significantly decreased in the delirium group, even after delirium resolution. Functional connectivity between the dorsolateral prefrontal cortex and dorsomedial prefrontal cortex was strengthened post‐resolution compared to during an episode; however, this relationship was still significantly weaker compared to controls. DISCUSSION: These findings highlight DOT's potential as an imaging biomarker to measure impaired cerebral oxygenation and functional dysconnectivity during and after delirium. Future studies should focus on the role of cerebral oxygenation in delirium pathogenesis and exploring the etiological link between delirium and dementias. The 3D imaging-based model reconstruction technology is being applied in various aspects of fruit planting management, picking, transportation, and promoting fruit industry automation development. The fruit model based on 3D imaging technology is closer to the external characteristics of the fruit itself. More accurate data is obtained when the model is imported into a computer for simulation and analysis for growth prediction, quality grading, and mechanical damage. The reconstructed fruit models can also be used to calibrate the parameters of the robot's vision system, optimize the grasping strategy of the picking machinery, and improve the identification sorting and picking accuracy. Some articles have been published on the principles of 3D imaging technology, and there are also numerous studies on the practical application of fruit models. However, no review article exists on applying 3D reconstruction and modeling of fruits using 3D imaging techniques. This paper will focus on the acquiring and processing of 3D data and analyzing the application of imaging principles and technologies such as triangulation, time-of-flight(TOF), interferometry, electromagnetic radiation(EMR), and ultrasound imaging(US). Also, this paper summarizes the application of fruit models for quality assessment, mechanical property study, and picking machinery. Salinomycin (Sal) has been recently discovered as a novel chemotherapeutic agent against various cancers including prostate cancer which is one of the most commonly diagnosed cancers affecting male populations worldwide. Herein we designed salinomycin nanocarrier (Sal-NPs) to extend its systemic circulation and to increase its anticancer potential. Prepared nanoform showed high encapsulation and sustained release profile for salinomycin. The present study elucidated the cytotoxicity and mechanism of apoptotic cell death of Sal-NPs against prostate cancer both in vitro and in vivo. At all measured concentrations, Sal-NPs showed more significant cytotoxicity to DU145 and PC3 cells than Sal alone. This effect was mediated by apoptosis, as confirmed by ROS generation, loss of MMP and cell cycle arrest at the G1 phase in both cells. Sal-NPs efficiently inhibited migration of PC3 and DU145 cells via effectively downregulating the epithelial mesenchymal transition. Also, the results confirmed that Sal-NPs can effectively inhibit the induction of Prostate adenocarcinoma in male Wistar rats. Sal-NPs treatment exhibited a decrease in tumour sizes, a reduction in prostate weight, and an increase in body weight, which suggests that Sal-NPs is more effective than salinomycin alone. Our results suggest that the molecular mechanism underlying the Sal-NPs anticancer effect may lead to the development of a potential therapeutic strategy for treating prostate adenocarcinoma.
