Abstract

Background/Objectives: Hepatitis E virus (HEV) is an RNA virus recognized to be spread mainly by fecal-contaminated water. Its infection is known to be a serious threat to public health globally, mostly in developing countries, in which Africa is one of the regions sternly affected. An African-based vaccine is necessary to actively prevent HEV infection. Methods: This study developed an in silico epitope-based subunit vaccine, incorporating CTL, HTL, and BL epitopes with suitable linkers and adjuvants. Results: The in silico-designed vaccine construct proved immunogenic, non-allergenic, and non-toxic and displayed appropriate physicochemical properties with high solubility. The 3D structure was modeled and subjected to protein docking with Toll-like receptors 2, 3, 4, 6, 8, and 9, which showed a stable binding efficacy, and the dynamics simulation indicated steady interaction. Furthermore, the immune simulation predicted that the designed vaccine would instigate immune responses when administered to humans. Lastly, using a codon adaptation for the E. coli K12 bacterium produced optimum GC content and a high CAI value, which was followed by in silico integration into a pET28 b (+) cloning vector. Conclusions: Generally, these results propose that the design of an epitope-based subunit vaccine can function as an outstanding preventive vaccine candidate against HEV, although validation techniques via in vitro and in vivo approaches are required to justify this statement. Abstract

Predictive microbiology is a rapidly evolving field that has gained significant interest over the years due to its diverse application in food safety. Predictive models are widely used in food microbiology to estimate the growth of microorganisms in food products. These models represent the dynamic interactions between intrinsic and extrinsic food factors as mathematical equations and then apply these data to predict shelf life, spoilage, and microbial risk assessment. Due to their ability to predict the microbial risk, these tools are also integrated into hazard analysis critical control point (HACCP) protocols. However, like most new technologies, several limitations have been linked to their use. Predictive models have been found incapable of modeling the intricate microbial interactions in food colonized by different bacteria populations under dynamic environmental conditions. To address this issue, researchers are integrating several new technologies into predictive models to improve efficiency and accuracy. Increasingly, newer technologies such as whole genome sequencing (WGS), metagenomics, artificial intelligence, and machine learning are being rapidly adopted into newer-generation models. This has facilitated the development of devices based on robotics, the Internet of Things, and time-temperature indicators that are being incorporated into food processing both domestically and industrially globally. This study reviewed current research on predictive models, limitations, challenges, and newer technologies being integrated into developing more efficient models. Machine learning algorithms commonly employed in predictive modeling are discussed with emphasis on their application in research and industry and their advantages over traditional models. Abstract

The respiratory syncytial virus (RSV) is a common cause of respiratory tract infection. The illness typically begins with a low-grade fever, runny nose, cough and sometimes wheezing. Usage of immunostimulators to prevent infections such as RSV is of great importance. Immunostimulators such Trino-IB are drugs which stimulate immune system by inducing activation or increasing activity of any of its components. Thirty specific pathogen-free Wister rats were put into three groups (10 each). Group A was treated with Trino-IB before and after inoculation with RSV, Group B was inoculated with only RSV and Group C (control) was not given any treatment. Clinical signs, anal temperature were observed daily and rats were sacrificed at day 2, 4, 6, 8 and 10 for histological analysis. Rats in Group B showed more and severe clinical signs of RSV infection alongside histopathological changes of the lungs than the rats in Group A. The results of from the study showed that Trino-IB has effect on the immune system of the rats by suppressing the effect of RSV. The effect of Trino-IB on the infected rat explains the ability of the drug to act as an immune booster as well as to suppress the clinical effect of the virus.

Infection caused by Human parvovirus B19 (HPVB19) during pregnancy has been associated with adverse effects on fetus such as intrauterine fetal death but there is dearth information about the seroprevalence and risk factors among pregnant women. This study was carried out to determine the incidence of specific IgM antibodies to HPVB19 among pregnant women attending Bowen University Teaching Hospital, Ogbomoso, Oyo State. A total of 185 participants, in the age range 18-49 years, who consented voluntarily after thorough explanation of the purpose of the study were recruited for the study. Abstract

The free radical theory of aging suggests that aging is caused by accumulation of damage inflicted by reactive oxygen species (ROS). However, this concept has been very useful in defining the contribution of oxidative damage to the aging process, an increasing number of studies opposes it. The idea that oxidative damage represents only one of many causes of aging also has limitations, as it does not explain causal relationships and inevitability of damage accumulation. Here, it is discussed that heterogeneity, infidelity and imperfectness of each and every biological process may be responsible for the inevitable accumulation of by-products and other damage forms. Although ROS are prototypical byproducts, their contribution to aging is governed by the metabolic organization of the cell, its protective systems, and genotype. These factors are controlled by natural selection, dietary and genetic interventions that extend lifespan, change the composition of cumulative damage and the rates of accumulation of its various forms. Oxidative damage, like other specific damage types viewed in isolation or in combination, does not represent the cause of aging. Instead, biological imperfectness, which leads to inevitable accumulation of damage in the form of mildly deleterious molecular species, may help define the true root of aging. Free radical and other specialized damage theories served their purpose in the understanding of the aging process, but in the current form they limit further progress in this area. Abstract

Background: Arboviruses are an emerging threat of significant impact on human health and well-being. With increasing proportion of the world living in urban environments, inadvertently, there is the creation of better habitats for vector species. Methodology: Ninety-three plasma samples were collected from subjects in the age range of 1–75 years and screened for Immunoglobulin (IgM) to dengue fever virus (DENV), West Nile virus (WNV), and yellow fever virus (YFV) using the third generation enzyme linked immunosorbent assay. Results: An overall prevalence of 52.7% (49/93) was recorded from the recruited individuals. IgM antibodies to DENV, YFV, and WNV were found in 16/49 (17.2%), 16/49 (17.2%), and 17/49 (18.3%), respectively. High prevalence was recorded in the age groups 16–30, 31–45, and 61–75 years. Gender analysis of the positive samples showed higher prevalence among females than males. The result also showed high prevalence in urban settings than rural settings for DENV and WNV, however, for YFV, higher prevalence was found in the rural area. The prevalence of dual and trio arboviral infection showed 17.2% and 11.8%, respectively. Conclusions: This study confirms the circulation of DENV, YFV, and WNV in Ogbomoso and therefore, suggests the need for public awareness on vector control.

Abstract

An active biosurfactant-producer from an agronomic environment majorly from the rhizosphere of a wheat plant in western Nigeria was utilized for the production of rhamnolipid. The isolated bacteria were selected using different methods such as oil displacement test, emulsification activity, and surface tension respectively. Based on partial sequenced 16S rDNA analysis of isolate, C1501 was identified as Pseudomonas aeruginosa with an accession number KF976394 having 100% similarity to P. aeruginosa LMG 1242T. The results showed that Pseudomonas aeruginosa has the ability to grow and reduce surface tension under a wide range of pH and carbon source. It was observed that strain C1501 reduced the surface tension of glycerol than glucose at the various concentration tested. The surface tension of 61.2 dynes/cm was also reduced to 30 dynes/cm at 12 h after the inoculation. The biosurfactant obtained from strain C1501 was purified and characterized using TLC, Liquid Chromatography/Mass Spectrometry (LC–MS) and nuclear magnetic resonance spectroscopy (NMR). The gravimetric analysis showed that strain C1501 efficiently biodegrade the tested crude oil with the following degradation percentage of 33%, 71.3% and 96% on 5th, 10th and 20th day respectively. The biosurfactant did not exhibit an inhibitory effect on the seed of economic crops tested, but demonstrated some broad antimicrobial activities against gram positive, gram negative and rot-inducing fungus when compared to the synthetic surfactant from Tween 20. New isomers and congeners rhamnolipids were detected from the Liquid Chromatography/Mass Spectrometry (LCMS). The predicted structure of the rhamnolipid was found to be L-rhamnosyl-L-rhamnosyl-3-b-hydroxydodecenoate. The study suggests application of the strain C1501 biosurfactant as an appropriate candidate for many applications such as biomedical, food industries, agriculture, and bioremediation

A nutraceutical product may be defined as a substance, which have physiological benefits nor gives protection against chronic diseases. Abstract

Hepatitis C virus is one of the emerging infectious diseases that can be transmitted through blood-to-blood contact. This study was carried out to determine the prevalence of anti-HCV antibodies among potential blood donors and pregnant women attending Bowen University Teaching Hospital (BUTH), Ogbomoso, Oyo State. This hospital-based study was conducted from December 2014 to September 2015. The study group (N = 279) included potential blood donors and pregnant women. Data on socio-demographic characteristics and potential risk factors were collected using a structured questionnaire. The presence of anti-HCV antibodies in serum samples of the studied subjects were detected using third-generation Enzyme Linked Immunosorbent Assay (ELISA) (WKEA Med Supplies Corp, China). Chisquare test was utilized to assess the association between the socio-demographic variables and HCV status. Logistic regression was done to determine the strength of association between risk factors and HCV status. Statistical significance was set at P ˂ 0.05. Overall seroprevalence of hepatitis C virus infection was found to be 1.79% consisting 0.36% of pregnant women and 1.43% of blood donors. None of the socio-demographic characteristics and potential risk factors among the study groups were significantly associated with hepatitis C virus infection. This study found a seroprevalence of anti-HCV antibody to be 1.79%, thus, screening of pregnant women and blood donors for HCV infections with the use of ELISA is recommended because of its important role in detecting the presence of anti-HCV antibody with utmost specificity and sensitivity.
