Abstract

Difficulties in the therapy of infection, caused by S. aureus, depends on the resistance of staphylococci to antibiotics, proceeding to chronicity of the diseases and the development on that background the depression of innate immunity functions and decreasing of the host resistance to the infection. In spite of tremendous efforts of researchers, up to date there are no commercial antistaphylococcal vaccine the efficacy of which would be proved in the completed clinical trials.Aim: obtaining secreted protein-based substances of the S. aureus № 6 and investigation their immunogenicity.Material and methods. Secreted protein-based substances (SPS) were obtain as: «initial» – from the filtrates of the culture fluid of the S. aureus № 6, grown to the end of the exponential phase according to the technology described previously [9], and I SPS and II SPS – after the ion-exchange chromatography of the «initial» SPS on the columns with Q-Sepharose and DEAE-Sepharose. The level of specific IgG antibodies in sera of immunized rabbits and mice determined in ELISA, the immunogenic activity evaluated in experiments of active and passive protection from the challenge performed on BALB/с mice and also by the determination of the bacterial content in organs and in the test of abscesses formation in kidneys. Results. Investigated SBS possessed the antigenic activity (the level of specific IgG antibodies in sera of immunized animals increased 2.2 – 7.5 times compared to the control groups), that is favor of the activation of the adaptive immunity system and significant protective activity revealed in experiments of active (index of efficacy 2.63 – 4.28) and passive protection. The immunization of mice with the «initial» and II SPS led to significant decrease of the number of colonyforming units of S. aureus and formation of abscesses in kidneys of mice. It is evidently, that investigated SPS, influence on the severity of staphylococcal infection and possesses the therapeutic effect.Conclusion. The preformed complex analysis at the current stage allowed to reveal perspectives of the further study of «initial» and II SPS in pre-clinical trials, as candidates, possessing the high protective facilities, for including them in the drug composition for immune prophylaxis and immunotherapy of diseases, caused by S. aureus.

Abstract

Abstract. The effects of 'Profetal', a drug containing liophylized, dextran-stabilized human alpha-fetoprotein as main active component, were investigated on peripheral donor blood mononuclear leukocytes (ML), with respect to their functional properties and the potential to generate mature dendritic cells (DC).Addition of the drug to cell cultures at optimal doses was shown to cause significant increase in their proliferative capacity and ML blastic transformation levels, and enhanced cytotoxicity towards K562 tumor cells, as well as to induce maturation of antigen-presenting dendritic cells.On the basis of the data obtained, a conclusion is made that 'Profetal' is an active immunomodulatory factor, and it may be applied for generation of cytotoxic lymphocytes and mature DC, aiming to apply them for the biotherapy of oncological and infectious diseases.

Abstract

Morphology, topography, and immunohistochemical features of leukocyte infiltrates were studied in various sites of the liver samples from the patients with metastatic disease, been affected by hepatitis B and C viruses at different degree of activity. Liver of СВА mice with implanted САО-1 tumour was also under study. Histochemical, and functional features, as well as immune phenotype of these cells were investigated. It has been shown that the major fraction of leukocyte infiltrates, mostly associated with implanted tumours in experimental mice, and in the areas adjacent to the tumor in humans, like as on the peak of viral hepatitis activity, is composed of lymphocytes. They are presented by large numvers of activated proliferating and differentiating cells bearing specific antigens, as well as natural killers and T-lymphocytes, possessing high-level killer activity towards NK-sensitive, and autologous lines of cancer cells. Hence, the results of our study, generally, confirm the data from literature reporting on existence of a special lymphocyte subpopulation, NKT cells, in human or murine liver affected by hepatitis virus or malignant tumors. The data concerning functional properties of these cells may be used for development of immunotherapy methods of viral diseases and oncological conditions complicated by liver metastases.

Abstract

Immunomodulating ability of a bacterial preparation (Immunovak VP-4 vaccine), and a compound of eukariotic origin (profetal), the main active component of human α-fetoprotein were under investigation in present study. Culturing of mononuclear leukocytes with the immunomodulatory factors promotes quantitative increase of the cells that express surface antigens specific for cytotoxic lymphocytes, natural killers and natural killer T-cells. Both bio-active preparations possess an expressed anti-cancer effect in vitro, i.e., they induce activation of killing properties of human peripheral blood mononuclear leukocytes, which is accompanied by acquiring their characteristic phenotype.

Abstract

Abstract. We studied effects of Stimforte, an immunomodulatory drug of animal origin, upon mononuclear leukocytes (MLs) and morphology of lymphoid organs in the course of cyclophosphan-induced immunosuppression. It was demonstrated that Stimforte is able to correct the quantitative and subpopulational composition of splenic MLs, the structure of central and peripheral lymphopoietic tissues, as well as effector cell functions of innate immunity affected by the cytostatic drug. Abstract

An important parameter of immunotropic preparation effect is the influence on the production of cytokines that provide the interaction between the effectors of both congenital and adaptive immunity. Cytokine level was determined in the work while using the methods of mucosal immunization with Immunovac-VP-4 and compared with the subcutaneous introduction of this preparation. In case of intranasal and peroral methods of antigen application IL-5, IL-6, and IL-12 expression was found to be elevated, the same was true for IL-1β, IL-5, IL-6, IL-12, and IFN-γ under the subcutaneous immunization. Resulting mice cytokine profile confirmed that irrespective of the method of Immunovac introduction, the activation of immune effectors occurred that was manifested in the increase in proinflammatory and regulatory cytokine levels. It was concluded that the introduction of opportunistic microbial antigens initiated the activation of the cascade of immunologic reactions and under the influence of synthesized cytokines the polarization of immune response was involved that was predominantly of Th1 type.

Abstract

Immunophenotypic and morphological peculiarities of immune reactions under intranasal, peroral and subcutaneous introduction of multicomponent vaccine Immunovac VP-4 containing a group of opportunistic bacterial antigens were investigated. The investigated preparation was found to cause marked activation of congenital immunity effectors both in parenteral and mucosal immunization. It is manifested in the expression of differential, costimulatory, adhesive molecules on the surface of mononuclear leukocytes in proliferation of key mucosal immunity effectors (γδТ, В1, NK cells), and changes in structure, cellular composition of immunocompetent organs both regional and distant as to the site of introduction. Currently existing data on effects and mechanisms of vaccine and immunomodulator mucosal introduction allows considering the development of mucosal mono- and associated vaccines as the priority direction in modern vaccinology.

Abstract

The aim of the study was to investigate the dynamics of serum immunoglobulins from oncological patients in the perioperative period. As a result of investigations, it was established that already at the initial stage of the intraoperative period the IgM concentration was elevated in patients' sera, and persisted up to 3 days after surgical intervention, while exceeding the values of the control group by 30—80 %. At the same time, no significant changes were detected in IgA and IgG levels of examined patients in the perioperative period. The obtained data confirm the expediency of carrying out the monitoring of immunoglobulin levels in oncological patients both in intra-, and postoperative periods and can be used for the assessment of patient's immune status and in the choice of immunocorrecting therapy strategy.

Abstract

The study of morphological features of tissue organs of experimental animals after a single intragastric administration of an aqueous suspension nanodis-dispersed manganese (III, IV) at doses of 2000, 3500 and 5000 mg/kg showed complex of morphological changes in the form of circulatory disorders up to hemostasis and bleeding which lead to changes in the structure of organs, enhanced apoptosis, activation of macrophage system by proliferation and macrophage phagocytosis of degradation products of cells, hypertrophy of lymphoid tissue of immune system; development of histiocytic infiltrates in parenchymal organs and central nervous system. With the introduction of microsized analogues in the same doses of the morphological changes in the circulatory system, macrophage system and immune organs found predominantly in a dose of 5000 mg/kg. Changes were less significant and were presented as activation of macrophages in the liver in the form of activation of Kupffer cells, proliferation of lymphoid tissue in the spleen, lymph histiocytic infiltrates in the liver, kidney, medium size.

Abstract

The count of stromal precursor cells in bone marrow transplants from CBA mice, transplanted to animals immunized with killed type 5 group A streptococcus vaccine, decreased 4.5-6.5 times (depending on the transplant age) in comparison with the grafts transplanted to normal recipients. The counts of stromal precursor cells in 1.5-3-month bone marrow transplants from animals immunized with killed streptococcal vaccine transplanted to normal mice were virtually the same, while in 7-month transplants they decreased 2-fold in comparison with their counts in bone marrow transplants from normal CBA mice transplanted to normal animals. The content of stromal precursor cells in the femoral bone marrow of animals immunized with killed streptococcal vaccine was appreciably (3.5 times) higher than in the bone marrow of normal mice. The results attest to an appreciable effect of streptococcal antigens on the bone marrow stromal tissue and suggest that not all stromal precursor cells, whose count increases after injection of antigens, are responsible for transplantability of the stromal tissue in case of its heterotopic transplantation.

Abstract

Addition of irradiated bone marrow feeder from guinea pigs and rabbits to cultures of passed rabbit bone marrow fibroblasts considerably decreases the efficiency of colony-formation and the total amount of fibroblasts in cultures. The presence of irradiated rabbit blood platelets sharply increases these parameters. The addition of irradiated bone marrow feeder to rabbit bone marrow fibroblasts cultured under conditions promoting cell differentiation also decreases the total content of fibroblasts in cultures. Our results suggest that cultured descendants of rabbit bone marrow stromal precursors preserve both the sensitivity to inhibitory factors produced by bone marrow cells coexisting with stromal precursors in cultures (typical of stromal precursors of these animals) and high sensitivity to growth-stimulating platelet-derived factors (typical of stromal precursors from rabbits and other animal species).

Abstract

Immunization of CBA mice with killed group A streptococcus (type 5) vaccine changed the counts of stromal precursor cells (CFC-F) in bone marrow transplants at different donorrecipient combinations (normal, N, or immune, I). CFC-F counts in bone marrow transplants from normal mice transplanted to immunized animals decreased 4-6-fold depending on the transplant age in comparison with similar transplants in normal recipients. The percentage of CFC-F colonies with alkaline phosphatase (osteogenesis marker) activity decreased more than 2-fold. Similarly, the count of CFC-F in the transplants was 2-fold lower during delayed (7 months) period after bone marrow transplantation from immunized donors (8–12 days after the end of immunization) to intact recipients, while 2 months after transplantation it was 3-fold lower. The mean optical density of the bone capsule in preparations stained for glycogen and alkaline phosphatase was 1.5-3 times lower in the N → I and I → N experiments in comparison with the control (N → N). On the other hand, CFC-F count in the femoral bone marrow of immunized animals was significantly (3.5-2.5 times) higher during the period from 8 days to 8 months after the end of immunization compared to CFC-F count in the femoral bone marrow of intact mice. These results attest to a significant prolonged effect of streptococcal antigens on the bone marrow stromal tissue. These data also indicate that not all CFC-F, the counts of which increased in response to antigens, are responsible for transplantability of the stromal tissue in heterotopic transplantation. Immunization by streptococcal antigens seemed to suppress transplantability and osteogenic activity of stromal stem cells. The efficiency of CFC-F cloning in mouse bone marrow cultures increased significantly (2-3-fold) in the presence of sera from immune mice. The levels of TNF-α and IFN-γ were low in this serum (2.7 and 6 times lower, respectively) in comparison with normal serum. Presumably, the effects of streptococcal antigens on stromal tissue were mediated through serum cytokines.

Abstract

The efficiency of cloning of stromal precursor cell increased more than 2-fold in splenic cultures and more than 3-fold in bone marrow cultures 24 h after injection of Profetal preparation to mice in vivo. The number of nucleated cells did not change in the bone marrow and slightly increased in the spleen. Addition of Profetal in vitro 2-fold decreased the efficiency of stromal precursor cells colony formation in mouse splenic cultures and dose-dependently decreased this process in bone marrow cultures derived from these animals, the maximum (5-fold) inhibitory effect was observed in a dose of 50 microg/ml. Addition of Profetal to cultured human bone marrow fibroblasts did not change the content of stromal fibroblasts in cultures. These data indicate the possibility of indirect effect of alpha-fetoprotein on the number of stromal precursor cell in hemopoietic and lymphoid organs.
