Currently, the problem of providing protein nutrition for animals and reducing the cost of pork production is relevant. The choice of effective and, at the same time, cheap protein components for animal rations is one of the foundations of high-performance animal husbandry. The purpose of the work was to determine the effectiveness of using compound feed with the introduction of sunflower meal of increased feed value and a different structure of the grain group on the slaughter and meat quality of pigs. Our research has proven the preservation of the level of productivity of pigs and obtaining a sufficiently high level of profitability of pork production when feeding compound feed based on barley and sunflower meal of increased feed value with or without wheat. In the control group, which received barley-corn-wheat compound feed, the average daily gains during the scientific and economic experiment amounted to 683 g. The maximum deviation in the experimental groups was +2.8 %. The difference was statistically improbable. At the same time, a slight increase in feed consumption per 1 kg of growth was observed in the experimental group, which was fed combined feed based on barley and sunflower meal. A tendency to increase the slaughter yield of piglets of the research groups and a probable increase in the meat yield from the carcass in animals whose combined feed was injected with 15 % by mass of sunflower meal of increased fodder value was noted. Feeding young pigs with two-component combined feed with barley grit + 15 % sunflower meal of increased forage value contributes to a decrease in lard thickness by 2.5 %. The longest back muscle of these animals is characterized by a 1.0% higher content of adipose tissue, an 8.3 % intensity of muscle fiber staining, as well as a 6.1 % lower caloric content, an amount of calcium (P ≤ 0.05), active acidity (P < 0.01). At the same time, a tendency to a slight increase in the moisture content of lard by 0.9 % and a decrease in the melting temperature of lard by 1.5 °C was noted. The research results on the chemical composition of animal meat during the transition to low-component compound feed indicate that the dry matter content was practically at the same level. Protein was found the most in the meat of the second group of guinea pigs (20.64 %), which is 0.76 % higher than the control analogs. At the same time, the fat content in muscle tissue samples decreased in the second and third groups relative to the control by 0.17 % and 0.70 %, respectively. Specific changes in the chemical composition of animal meat in the third experimental group, compared to the control group, resulted in a 6.5 % decrease in its caloric content, coinciding with recent trends regarding the production of leaner pork. When switching to low-component compound feed with sunflower meal of increased fodder value, there is a tendency to an inevitable increase in losses during the culinary processing of pig meat, with a simultaneous decrease in its tenderness. The change in these indicators occurred due to the reduction in the moisture-retaining capacity of muscle tissue in the second and third experimental groups.

The work presents the results of the study of some biochemical indicators of blood serum, physicochemical properties, and chemical composition of muscle tissue of young pigs of the large white breed and the calculation of correlations between the main quantitative characteristics. The work was carried out in the agricultural formations of the Dnipropetrovsk region, the research center for biosafety and environmental control of agricultural resources of the Dnipro State Agrarian and Economic University, LLC “Globynskyi Myasokbinat” of the Poltava region, the zootechnical analysis laboratory of the Institute of Pig Breeding and Agro-industrial Production of the National Academy of Sciences and the animal husbandry laboratory of the state institution “Institute of Grain Crops of NAAS”. Studies of the fattening and meat qualities of young pigs of the large white breed show that the animals of the controlled population belong to the elite class by the age of reaching a live weight of 100 kg. This indicator in the animals of the experimental group ranges from 167 to 188 days. It was established that the number of samples of the longest back muscle of high quality according to the indicator “moisture retention capacity” is 12.0 %, “fat content” – 16.0%, “tenderness” – 12% and “color intensity” – 16%. A high level of phenotypic consolidation was established in the animals of the II experimental group (the age of reaching a live weight of 100 kg is 177–188 days) according to the content of total moisture (K1 = +0.458, K2 = +0.456), air-dry matter (K1 = +0.492, K2 = +0.500), ash (K1 = +0.527, K2 = +0.534), fat (K1 = +0.559, K2 = +0.518) and calcium (K1 = +0.385, K2 = +0.415). Reliable correlations were established between the following pairs of traits: moisture-holding capacity × protein content (r = -0.484, tr = 2.65), age at which live weight reached 100 kg × calcium (Ca) content (r = -0.392, tr = 2,15), age of reaching a live weight of 100 kg × moisture retention capacity (r = +0.447, tr = 2.40), age of reaching a live weight of 100 kg × loss during heat treatment (r = -0.390, tr = 2.14). This indicates the effectiveness of using the indicator “age of reaching 100 kg live weight, days” for early prediction of calcium (Ca), moisture retention capacity, and loss of muscle tissue during heat treatment in young pigs of the large white breed.

The widespread presence of multidrug-resistant pathogenic microorganisms challenges the development of novel chemotype antimicrobials, insensitive to microbial tools of resistance. To date, various monoterpenoids have been shown as potential antimicrobials. Among many classes of molecules with antimicrobial activity, terpenes and terpenoids are an attractive basis for the design of antimicrobials because of their low toxicity and availability for various modifications. In this work, we report on the synthesis of sulfenimines from chiral trifluoromethylated and non-fluorinated pinane-type thiols. Final compounds were obtained with yields of up to 81%. Among the 13 sulfenimines obtained, 3 compounds were able to repress the growth of both bacteria (S. aureus, both MSSA and MRSA; P. aeruginosa) and fungi (C. albicans) with an MIC of 8–32 µg/mL. Although compounds exhibited relatively high cytotoxicity (the therapeutic index of 3), their chemotype can be used as a starter point for the development of disinfectants and antiseptics for targeting multidrug-resistant pathogens.

Introduction: Tuberculosis (TB) treatment typically involves a tailored combination of four antibiotics based on the drug resistance profile of the infecting strain. The increasing drug resistance of Mycobacterium tuberculosis ( Mtb ) requires the development of novel antibiotics to ensure effective treatment regimens. Gallium (Ga) is being explored as a repurposed drug against TB due to its ability to inhibit Mtb growth and disrupt iron metabolism. Given the potential interactions between Ga and established antibiotics, we investigated how a combination of Ga with levofloxacin (Lfx) or linezolid (Lzd) affects the growth and metabolome of a multidrug-resistant (MDR) Mtb clinical strain., Methods: Mtb was cultured using a BACTEC 960 system with concentrations of Ga ranging from 125 to 1,000 μM and with 250 to 500 μM of Ga combined with 0.125 mg/L of Lfx or Lzd. For metabolome analysis, the antibacterials were used at concentrations that inhibited the growth of bacteria without causing cell death. Metabolites were extracted from Mtb cells and analyzed using chromatography-mass spectrometry., Results: The MDR Mtb strain exhibited a dose-dependent response to Ga. Notably, the enhancement in growth inhibition was statistically significant for the Ga/Lfx combination compared to Ga alone, while no such significance was observed for Ga/Lzd. Moreover, exposure to Ga/Lfx or Ga/Lzd resulted in distinct metabolite profiles. Ga treatment increased the level of aconitate, fumarate, and glucose in the cells, suggesting the inhibition of iron-dependent aconitase and fumarate hydratase, as well as disruption of the pentose phosphate pathway. The levels of glucose, succinic acid, citric acid, and hexadecanoic acid followed a similar pattern in cells exposed to Ga and Ga/Lfx at 500 μM Ga but exhibited different trends at 250 μM Ga., Discussion: In the presence of Lfx, the Mtb metabolome changes induced by Ga are more pronounced compared to those observed with Lzd. Raman spectroscopy provides non-destructive, label-free quantitative studies of chemical compositions at the microscale as used on NASA's Perseverance rover on Mars. Such capabilities come at the cost of high requirements for instrumentation. Here we present a centimeter-scale miniaturization of a Raman spectrometer using cheap non-stabilized laser diodes, densely packed optics, and non-cooled small sensors. The performance is comparable with expensive bulky research-grade Raman systems. It has excellent sensitivity, low power consumption, perfect wavenumber, intensity calibration, and 7 cm -1 resolution within the 400-4000 cm -1 range using a built-in reference. High performance and versatility are demonstrated in use cases including quantification of methanol in beverages, in-vivo Raman measurements of human skin, fermentation monitoring, chemical Raman mapping at sub-micrometer resolution, quantitative SERS mapping of the anti-cancer drug methotrexate and in-vitro bacteria identification. We foresee that the miniaturization will allow realization of super-compact Raman spectrometers for integration in smartphones and medical devices, democratizing Raman technology., (© 2024. MoS 2 nanoribbons have attracted increased interest due to their properties, which can be tailored by tuning their dimensions. Herein, the growth of MoS 2 nanoribbons and triangular crystals formed by the reaction between films of MoOx (22 nanoribbons arising from distinct contributions from the single-layer edges and multilayer core. Nanoscale imaging reveals a blue-shifted exciton emission of the monolayer edge compared to the isolated MoS 2 monolayers due to built-in local strain and disorder. We further report on an ultrasensitive photodetector made of a single MoS 2 nanoribbon with a responsivity of 8.72 × 10 2  A W -1 at 532 nm, among the highest reported up-to-date for single-nanoribbon photodetectors. We have recorded ATR-IR spectra of binary mixtures in the (quasi-)ideal systems Benzene-Toluene, Benzene-Carbon tetrachloride and Benzene-Cyclohexane and performed classical least squares, inverse least squares and principal component regression on the resulting spectra. In contrast to the general expectation, the spectra of ideal mixtures follow only roughly Beer's approximation, in particular stronger bands show shifts and increased intensities for intermediary compositions since the polarization of matter by light cannot be neglected. As a consequence, these conventional regression techniques lead to principle and unavoidable errors, even though some of the classical regression techniques are assumed to be able to cope with nonlinearities. In particular in the system Benzene-Carbon tetrachloride large errors result and the relative average error of the volume fraction determination is about 10 % for all three methods. Especially remarkable is that the multivariate regression methods do not perform better than the classical univariate calibration if for the latter a peak due to an oscillator with comparably low oscillator strength is selected, since for such bands polarization effects are weak and Beer's approximation holds comparably well., (Copyright © 2022 Elsevier B.V. We have recorded attenuated total reflection infrared spectra of binary mixtures in the (quasi-)ideal systems benzene-toluene, benzene-carbon tetrachloride, and benzene-cyclohexane. We used two-dimensional correlation spectroscopy, principal component analysis, and multivariate curve resolution to analyze the data. The 2D correlation proves nonlinearities, also in spectral ranges with no obvious deviations from Beer's approximation. The number of principal components is much higher than two and multivariate curve resolution carried out under the assumption of the presence of a third component, results in spectra which only show bands of the original components. The results negate the presence of third components, since any complex should have lower symmetry than the individual molecules and thus more and/or different infrared-active bands in the spectra. Based on Lorentz-Lorenz theory and literature values of the optical constants, we show that the nonlinearities and additional principal components are consequences of local field effects and the polarization of matter by light. Lorentz-Lorenz theory is, however, not able to explain, for example, the different blueshifts of the strong A 2u band of benzene in the three mixtures. Obviously, infrared spectroscopy is sensitive to the short-range order around the molecules, which changes with content, their shapes, and their anisotropy.

Streptococcus pneumoniae , a major cause of pneumonia, sepsis, and meningitis worldwide, has the nasopharynges of small children as its main ecological niche. Depletion of pneumococci from this niche would reduce the disease burden and could be achieved using small molecules with narrow-spectrum antibacterial activity. We identified the alkylated dicyclohexyl carboxylic acid 2CCA-1 as a potent inducer of autolysin-mediated lysis of S. pneumoniae , while having low activity against Staphylococcus aureus 2CCA-1-resistant strains were found to have inactivating mutations in fakB3 , known to be required for uptake of host polyunsaturated fatty acids, as well as through inactivation of the transcriptional regulator gene fabT , vital for endogenous, de novo fatty acid synthesis regulation. Structure activity relationship exploration revealed that, besides the central dicyclohexyl group, the fatty acid-like structural features of 2CCA-1 were essential for its activity. The lysis-inducing activity of 2CCA-1 was considerably more potent than that of free fatty acids and required growing bacteria, suggesting that 2CCA-1 needs to be metabolized to exert its antimicrobial activity. Total lipid analysis of 2CCA-1 treated bacteria identified unique masses that were modeled to 2CCA-1 containing lysophosphatidic and phosphatidic acid in wild-type but not in fakB3 mutant bacteria. This suggests that 2CCA-1 is metabolized as a fatty acid via FakB3 and utilized as a phospholipid building block, leading to accumulation of toxic phospholipid species. Analysis of FabT-mediated fakB3 expression elucidates how the pneumococcus could ensure membrane homeostasis and concurrent economic use of host-derived fatty acids. IMPORTANCE Fatty acid biosynthesis is an attractive antibiotic target, as it affects the supply of membrane phospholipid building blocks. In Streptococcus pneumoniae , it is not sufficient to target only the endogenous fatty acid synthesis machinery, as uptake of host fatty acids may bypass this inhibition. Here, we describe a small-molecule compound, 2CCA-1, with potent bactericidal activity that upon interactions with the fatty acid binding protein FakB3, which is present in a limited number of Gram-positive species, becomes metabolized and incorporated as a toxic phospholipid species. Resistance to 2CCA-1 developed specifically in fakB3 and the regulatory gene fabT These mutants reveal a regulatory connection between the extracellular polyunsaturated fatty acid metabolism and endogenous fatty acid synthesis in S. pneumoniae , which could ensure balance between efficient scavenging of host polyunsaturated fatty acids and membrane homeostasis. The wide and ever-increasing applications of thermoplasmonics demand the need for sensitive and reliable tools to probe optical absorptions of individual nanoparticles. However, most of the currently available techniques focus only on measuring the surface temperature of nanostructures in a particular medium and are either invasive or suffer from low sensitivity, lengthy calibration, or the inability to probe single structures with nanogaps. Here, we present for the first time the use of micromechanical SiN string resonators for quantifying optical absorption cross sections of individual plasmonic nanostructures. Monomers and dimers of nanospheres, nanostars, shell-isolated nanoparticles, and nanocubes are probed. A reliable data treatment method is developed to obtain the absorption cross sections as a function of responsivity across a string. The presented method exhibits an excellent sensitivity of ∼89 Hz/K. This allows quantification of optical absorption cross sections of individual plasmonic structures even when their plasmon resonance wavelengths are far from the laser excitation wavelength. The experimentally obtained optical absorption cross sections agree well with the simulations. Influencing factors including polarization, surface morphology, and nanogap size are discussed. The developed method and the obtained optical absorption profiles facilitate future development and optimization of thermoplasmonic applications.

Non-destructive orientation mapping is an important characterization tool in materials science and geoscience for understanding and/or improving material properties based on their grain structure. Confocal Raman microscopy is a powerful non-destructive technique for chemical mapping of organic and inorganic materials. Here we demonstrate orientation mapping by means of Polarized Raman Microscopy (PRM). While the concept that PRM is sensitive to orientation changes is known, to our knowledge, an actual quantitative orientation mapping has never been presented before. Using a concept of ambiguity-free orientation determination analysis, we present fast and quantitative single-acquisition Raman-based orientation mapping by simultaneous registration of multiple Raman scattering spectra obtained at different polarizations. We demonstrate applications of this approach for two- and three-dimensional orientation mapping of a multigrain semiconductor, a pharmaceutical tablet formulation and a polycrystalline sapphire sample. This technique can potentially move traditional X-ray and electron diffraction type experiments into conventional optical laboratories.

To enhance oral bioavailability of poorly soluble drugs, microfabricated devices can be utilized. One example of such devices is microcontainers. These are cylindrical in shape with an inner cavity for drug loading and with only the top side open for release. Supercritical CO 2 (scCO 2 ) impregnation is an interesting technique for loading drugs into polymeric matrices in, for example, microcontainers since it avoids the use of organic solvents and is cheap. One of the main drawbacks of this technique is the unknown three-dimensional drug distribution in the polymer matrix. The aim of this study was to investigate the loading of two poorly soluble drugs, naproxen and ketoprofen, by scCO 2 impregnation into confined polymer matrices of different sizes. Three different sizes of microcontainers (small, medium, and large) and, thereby, different surface areas accessible for impregnation were compared. From in vitro studies, the amount of naproxen and ketoprofen loaded into the different microcontainers and their corresponding release profiles were seen to be similar. A custom-made Raman microscope facilitated volumetric Raman maps of an entire microcontainer filled with polyvinylpyrrolidone (PVP) and scCO 2 impregnated with either naproxen or ketoprofen. In all microcontainer sizes, the drugs were only detected in the top layer of the polymer matrix, explaining the observed similar release profiles. Using X-ray powder diffraction and Raman spectroscopy, the solid state form of the drugs was evaluated, showing that ketoprofen was amorphous in all microcontainer sizes. Naproxen was found not to be crystalline nor amorphous but in a less ordered configuration than the crystalline state. In conclusion, volumetric Raman mapping is a powerful technology for imaging drug distribution and drug crystallinity in polymers and allowed us to conclude that (i) scCO 2 impregnation depth does not depend on surface area and (ii) impregnated drugs are noncrystalline.

Crystalline solids can incorporate water molecules into their crystal lattice causing a dramatic impact on their properties. This explains the increasing interest in understanding the dehydration pathways of these solids. However, the classical thermal analytical techniques cannot spatially resolve the dehydration pathway of organic hydrates at the single particle level. We have developed a new method for imaging the dehydration of organic hydrates using Raman line-focus microscopy during heating of a particle. Based on this approach, we propose a new metastable intermediate of theophylline monohydrate during the three-step dehydration process of this system and further, we visualize the complex nature of the three-step dehydration pathway of nitrofurantoin monohydrate to its stable anhydrous form. A Raman line-focus mapping option was applied for fast simultaneous mapping of differently sized and shaped particles of nitrofurantoin monohydrate, revealing the appearance of multiple solid-state forms and the non-uniformity of this particle system during the complex dehydration process. This method provides an in-depth understanding of phase transformations and can be used to explain practical industrial challenges related to variations in the quality of particulate materials.

Given the commercial importance of the compounds produced by genetically modified organisms, there is a need for screening methods which facilitate the evaluation of newly developed strains, especially during the phase of proof-of-concept development. We report a time-efficient analysis method for the screening of bacterial strains, which enables the detection of two structurally similar secondary bacterial metabolites. By combining liquid-liquid extraction and surface-enhanced Raman scattering we were able to quantify p-coumaric acid and cinnamic acid, produced by genetically modified E. coli from tyrosine and phenylalanine, respectively. With the simple sample pre-treatment method, and by applying a partial least squares data analysis method, we simultaneously detected the analytes from four E. coli strains cultured in the presence or absence of tyrosine and phenylalanine.

An application of 2D-BN nanoparticles as a spectroscopic marker, weak luminescent marker and anticancer drug (doxorubicin, DOX) delivery system with protection properties was studied for the LNCaP strains of cancer cells using FTIR and Raman spectroscopy for analysing the cancer cells, cells with BN, the cancer cells with DOX, and the cancer cells with BN nanoparticles loaded by DOX. Study of IR absorption and Raman spectra of the LNCaP strains of cancer cells incubated with 2D-BN nanoparticles for 1 hour showed that the 2D-BN nanoparticles could pass through the cell membrane and localize inside the membrane or close to the membrane in the cytoplasm of the cells. We registered the spectra of the disturbed lipids during the DOX-2D-BN passing through the membrane. After incubation for 2 hours and more, spectral changes in other structural components of the cell (nuclei, cytoplasm, mitochondria) can be registered. Confocal microscopy showed that a gold nanostructured support enhances the fluorescence of the cancer cells with 2D-BN as well as that with DOX, however the double action of 2D-BN and DOX on the cancer cells aggravates the emission property of the studied system. An MTT test showed that the toxicity of DOX on the 2D-BN nanoparticles is less than that on the reference cells, and at the same time the efficiency of the DOX action on the cancer cells does not change., Competing Interests: There are no conflicts of interest to declare., (This journal is © The Royal Society of Chemistry.)

We present the development of an automated centrifugal microfluidic platform with integrated sample pre-treatment (filtration and liquid-liquid extraction) and detection (SERS-based sensing). The platform consists of eight calibration and four assay modules, fabricated with polypropylene using injection molding and bonded with ultrasonic welding. The platform was used for detection of a secondary bacterial metabolite (p-coumaric acid) from bacterial supernatant. The obtained extraction efficiency was comparable to values obtained in batch experiments and the SERS-based sensing showed a good correlation with HPLC analysis.
