Silver nanoparticles (AgNPs) have been synthesized in the presence of Strawberry fruit extract (SBFE) at room temperature. The synthesized AgNPs was characterized by UV-vis spectroscopy, SEM, EDS, XRD, TEM and FTIR. The UV-vis spectra of the AgNPs show SPR band at 450 nm. TEM results indicate that AgNPs are spherical in shape and size range between 7–65 nm. Antibacterial activity of the synthesized AgNPs has been assessed against Pseudomonas aeruginosa and Bacillus licheniformis. The results show that AgNPs exhibit inhibitory effect and effect is a function of AgNPs concentration. The antibacterial activity of the prepared AgNPs has been compared with two antibiotics, amoxicillin and ciprofloxacin. It is found that the antibiotics perform better than AgNPs.

The Kingdom of Saudi Arabia (KSA) is the leading oil-exploring and -exploiting country in the world. As a result, contamination of the environment by petroleum products (mainly hydrocarbons) is common, necessitating strategies for their removal from the environment. Much work has been conducted on bacterial degradation of hydrocarbons in the KSA. This review comprehensively analyzed 43 research investigation articles on bacterial hydrocarbon degradation, mainly polyaromatic hydrocarbons (PAHs) within the KSA. More than 30 different bacterial genera were identified that were capable of degrading simple and complex PAHs, including benzo[a]pyrene and coronene. Different strategies for selecting and isolating these bacterial strains and their advantages and disadvantages were highlighted. The review also discussed the origins of sample inocula and the contributions of various research groups to this field. PAH metabolites produced by these bacteria were presented, and biochemical pathways of PAH degradation were proposed. More importantly, research gaps that could enrich our understanding of petroleum product biodegradation mechanisms were highlighted. Overall, the information presented in this paper will serve as a baseline for further research on optimizing bioremediation strategies in all petroleum-contaminated environments.

L’immensité des ressources minières de la RDC, en théorie, laisse croire à sa population que le pays est extrêmement riche à telle enseigne qu’il peut s’en développer. Or, dans la réalité, de toutes les nombreuses substances de plus de 110 espèces, 7% seulement sont exploitées et cela, de manière discontinue et irrégulière pour certains. La réflexion et le débat initiés dans cet article consistent à déconstruire les fausses idées de l’opinion publique sur l’appréhension qu’elle a des richesses minérales de la République Démocratique du Congo et d’en présenter la vraie réalité des potentialités et de l’exploitation minière quant à ce.

Polycyclic aromatic hydrocarbons (PAHs) are significant pollutants found in petroleum products. There is ample literature on the biodegradation of PAHs containing less than five rings, but little has been done on those with more than five rings. Coronene (CRN), a seven-ring-containing PAH, has only been shown to be degraded by one bacterial strain. In this study, a bacterial strain 10SCRN4D was isolated through enrichment in the presence of CRN and 10% NaCl (w/v). Analysis of the 16S rRNA gene identified the strain as Halomonas caseinilytica. The strain was able to degrade CRN in media containing 16.5–165 μM CRN with a doubling time of 9–16 hours and grew in a wide range of salinity (0.5–10%, w/v) and temperature (30–50°C) with optimum conditions of pH 7, salinity 0.5%–10% (w/v), and temperature 37°C. Over 20 days, almost 35% of 16.5 μM CRN was degraded, reaching 76% degradation after 80 days as measured by gas chromatography. The strain was also able to degrade smaller molecular weight PAHs such as benzo[a]pyrene, pyrene, and phenanthrene. This is the first report of Halomonas caseinilytica degrading CRN as the sole carbon source in high salinity, and thus highlights the potential of this strain in bioremediation.

Abstract The antifolate sulphadoxine-pyrimethamine (SP) has been used in the intermittent prevention of malaria in pregnancy (IPTp). SP is an ideal choice for IPTp, however, as resistance of Plasmodium falciparum to SP increases, data are accumulating that SP may no longer provide benefit in areas of high-level resistance. Probenecid was initially used as an adjunctive therapy to increase the blood concentration of penicillin; it has since been used to augment concentrations of other drugs, including antifolates. The addition of probenecid has been shown to increase the treatment efficacy of SP against malaria, suggesting that the combination of probenecid plus SP may prolong the useful lifespan of SP as an effective agent for IPTp. Here, the literature on the pharmacokinetics, adverse reactions, interactions and available data on the use of these drugs in pregnancy is reviewed, and the possible utility of an SP-probenecid combination is discussed. This article concludes by calling for further research into this potentially useful combination.

Abstract Background Previous investigations indicate that methotrexate, an old anticancer drug, could be used at low doses to treat malaria. A phase I evaluation was conducted to assess the safety and pharmacokinetic profile of this drug in healthy adult male Kenyan volunteers. Methods Twenty five healthy adult volunteers were recruited and admitted to receive a 5 mg dose of methotrexate/day/5 days. Pharmacokinetics blood sampling was carried out at 2, 4, 6, 12 and 24 hours following each dose. Nausea, vomiting, oral ulcers and other adverse events were solicited during follow up of 42 days. Results The mean age of participants was 23.9 ± 3.3 years. Adherence to protocol was 100%. No grade 3 solicited adverse events were observed. However, one case of transiently elevated liver enzymes, and one serious adverse event (not related to the product) were reported. The maximum concentration (Cmax) was 160-200 nM and after 6 hours, the effective concentration (Ceff) was Conclusion Low-dose methotraxate had an acceptable safety profile. However, methotrexate blood levels did not reach the desirable Ceff of 250-400-nM required to clear malaria infection in vivo. Further dose finding and safety studies are necessary to confirm suitability of this drug as an anti-malarial agent.

Abstract Background The spread of resistance to chloroquine (CQ) led to its withdrawal from use in most countries in sub-Saharan Africa in the 1990s. In Malawi, this withdrawal was followed by a rapid reduction in the frequency of resistance to the point where the drug is now considered to be effective once again, just nine years after its withdrawal. In this report, the polymorphisms of markers associated with CQ-resistance against Plasmodium falciparum isolates from coastal Kenya (Kilifi) were investigated, from 1993, prior to the withdrawal of CQ, to 2006, seven years after its withdrawal. Changes to those that occurred in the dihydrofolate reductase gene (dhfr) that confers resistance to the replacement drug, pyrimethamine/sulphadoxine were also compared. Methods Mutations associated with CQ resistance, at codons 76 of pfcrt, at 86 of pfmdr1, and at codons 51, 59 and 164 of dhfr were analysed using PCR-restriction enzyme methods. In total, 406, 240 and 323 isolates were genotyped for pfcrt-76, pfmdr1-86 and dhfr, respectively. Results From 1993 to 2006, the frequency of the pfcrt-76 mutant significantly decreased from around 95% to 60%, while the frequency of pfmdr1-86 did not decline, remaining around 75%. Though the frequency of dhfr mutants was already high (around 80%) at the start of the study, this frequency increased to above 95% during the study period. Mutation at codon 164 of dhfr was analysed in 2006 samples, and none of them had this mutation. Conclusion In accord with the study in Malawi, a reduction in resistance to CQ following official withdrawal in 1999 was found, but unlike Malawi, the decline of resistance to CQ in Kilifi was much slower. It is estimated that, at current rates of decline, it will take 13 more years for the clinical efficacy of CQ to be restored in Kilifi. In addition, CQ resistance was declining before the drug's official withdrawal, suggesting that, prior to the official ban, the use of CQ had decreased, probably due to its poor clinical effectiveness.

Abstract The selection of point mutation at codon 164 (from isoleucine to leucine) of the dihydrofolate reductase (DHFR) enzyme in Plasmodium falciparum is associated with high sulfadoxine /pyrimethamine (SP) resistance. Using the yeast expression system that allows the detection of dhfr allele present at low level, the presence of this mutation had previously been reported between 1998–1999 in Muheza, Tanzania, an area of high SP resistance. Eighty five P. falciparum isolates, obtained from the same area between 2002 and 2003, were analysed for the presence of Leu-164 mutation, using standard protocol based on PCR-RFLP. None of the isolates had the Leu-164 mutation.

Polycyclic aromatic hydrocarbons (PAHs) are chemicals that are released into the environment during activities of the petroleum industry. The bioaccumulation, carcinogenic and mutagenic potential of PAHs necessitates the bioremediation of these contaminants. However, bioremediation of PAHs has a number of limitations including the inability of a single microbe to degrade all of the PAH fraction’s environmental constituents. Therefore, a different paradigm, employing microalgal-bacterial consortium (MBC), may be used to effectively remove PAHs contaminants. In this type of interaction, the microalgae and bacteria species in the consortium work together in a way that enhances the overall performance of the MBC. Bacterial species in the consortium provide essential nutrients or growth factors by degrading toxic substances and provide these to microalgae, while the microalgae species provide organic carbon for the bacterial species to grow. For the first time, the ability of Gonium pectorale (G. pectorale) microalgae to break down phenanthrene (PHE) and anthracene (ANT) was investigated. Phenanthrene was shown to be more effectively degraded by G. pectorale (98%) as compared to Bacillus licheniformis (B. licheniformis) 19%. Similarly, G. pectorale has effectively degrade anthracene (98%) as compared with B. licheniformis (45%). The consortia of G. pectorale and B. licheniformis has shown a slight increase in the degradation of PHE (96%) and ANT (99%). Our findings show that B. licheniformis did not inhibit the growth of G. pectorale and in the consortia has effectively eliminated the PAHs from the media. Therefore G. pectorale has a tremendous potential to remove PAHs from the polluted environment. Future research will be conducted to assess Gonium’s capacity to eliminate PAHs that exhibit high molar masses than that of PHE and ANT.

We describe the MalariaGEN Pf7 data resource, the seventh release of Plasmodium falciparum genome variation data from the MalariaGEN network. It comprises over 20,000 samples from 82 partner studies in 33 countries, including several malaria endemic regions that were previously underrepresented. For the first time we include dried blood spot samples that were sequenced after selective whole genome amplification, necessitating new methods to genotype copy number variations. We identify a large number of newly emerging crt mutations in parts of Southeast Asia, and show examples of heterogeneities in patterns of drug resistance within Africa and within the Indian subcontinent. We describe the profile of variations in the C-terminal of the csp gene and relate this to the sequence used in the RTS,S and R21 malaria vaccines. Pf7 provides high-quality data on genotype calls for 6 million SNPs and short indels, analysis of large deletions that cause failure of rapid diagnostic tests, and systematic characterisation of six major drug resistance loci, all of which can be freely downloaded from the MalariaGEN website.

Hydrogen (H2) fuel is assessed to be a major component of sustainable energy systems in the net‐zero world. However, hydrogen storage is challenging and requires safe and environmentally friendly solutions like H2 geo‐sequestration. This study evaluates the effects of sulphate‐reducing bacteria (SRB) on H2 geological storage potential in the basalt rock. Fourier‐transform infrared spectroscopy (FTIR) findings show the presence of significant components, that is, O‐Si‐O and organic functional groups, that is, aromatics, amine salts, alkane, and cyclohexane in the basalt rock immersed in the nutrient solution without SRB. However, we found that C‐H stretching modes of organics with peaks at 1,465 cm−1 were observed. Consequently, amine salt (N‐H) (850–750 cm−1), solvent impurities (C‐H), and alkane spectrums are components of nutrient solutions and can be results of metabolic microbial activity that can influence on the surface of the basalt rock. Hence, these changes indicate the presence of microbial activity which might affect the surface chemistry of the rock leading to wettability alteration. We observed that the contact angle (θ) of brine‐H2 on the rock surface slightly changed from 500 to 4,000 psi pressure after the effect of bacteria at 50 °C. The wettability changed the surface of the rock from strong water‐wet to weak or intermediate water‐wet condition (i.e., θ < 75°) at 4,000 psi and temperatures 25 and 50 °C after the bacteria effect. The affiliation of brine water reduces on the rock surface with increasing temperatures and pressures, even without microbial influence. Additionally, we investigated interfacial tension and capillary pressure on SRB bacteria treated basalt which is not yet reported in the published work. Interfacial tension (IFT) and Pc of H2 were reduced by 19% and 65%, respectively at 50 °C and 4,000 psi after the bacteria effect. Hence, the above findings could help to answer the key factors of the reservoir rock including wettability, capillary pressure, and interfacial tension to plan a field‐scale H2 geo‐sequestration strategy under the influence of biotic life. © 2024 Society of Chemical Industry and John Wiley & Sons, Ltd.
