Abstract

The objective of the paper is to determine the most suitable operational redundant assets and asset structure for a manufacturing enterprise. They are determined by the financial analysis of ratios, vertical and horizontal analysis, and the calculation of Altman Z-score. The recommended sizes of activity ratios are: the turnover of total assets should be equal to the value of 1, the time of inventory turnover should be as short as possible, the turnover of inventories should be as large as possible, the time of debt collection should be the shortest possible, the due date of fulfilling obligations should be equal to 2 or higher. The ratios are compared with the sector and the position of enterprise is assessed. The limit of the research is the incapability to determine the best asset structure, but only the best possible one, on the basis of comparing with the sector that sets the bar in a specific field. Every enterprise aspiring to be competitive should meet the sector standards, at least.

Abstract

Extraction of compounds with different physicochemical properties from a complex matrix usually involves several individual steps and requires large volumes of organic solvents. In this pioneering study, we propose a comprehensive two-step supercritical fluid extraction using carbon dioxide, ethanol, and water. This novel approach allows the extraction of non-polar and polar analytes within one run in two consecutive steps. Indeed, the first step with a dominant amount of CO2 with only 2% cosolvent allowed the selective extraction of non-polar volatile terpenes only in 20 min. The conditions were then automatically switched. Increasing the cosolvent volume in the extraction solvent up to 44% (v/v) resulted in the extraction of more polar compounds, including flavonoids and phenolic acids, in 60 min. Importantly, switching the supercritical fluid extraction (SFE) conditions does not require any manual intervention but results in two separate fractions containing target compounds with distinctly different physicochemical properties. The novel method was verified in terms of repeatability, accuracy, precision, and greenness. Two-step SFE was applied to seven plant species differing in volatile terpenes and phenolic profiles. The results proved that this concept is suitable for the analysis of complex plant samples. In addition, it enables a reduction in the toxic solvents consumption, extraction time, and manual intervention required for traditional extraction approaches when isolating different groups of metabolites. Abstract

The role of mass spectrometry (MS) has become more important in most application domains in recent years. Pharmaceutical analysis is specific due to its stringent regulation procedures, the need for good laboratory/manufacturing practices, and a large number of routine quality control analyses to be carried out. The role of MS is, therefore, very different throughout the whole drug development cycle. While it dominates within the drug discovery and development phase, in routine quality control, the role of MS is minor and indispensable only for selected applications. Moreover, its role is very different in the case of analysis of small molecule pharmaceuticals and biopharmaceuticals. Our review explains the role of current MS in the analysis of both small‐molecule chemical drugs and biopharmaceuticals. Important features of MS‐based technologies being implemented, method requirements, and related challenges are discussed. The differences in analytical procedures for small molecule pharmaceuticals and biopharmaceuticals are pointed out. While a single method or a small set of methods is usually sufficient for quality control in the case of small molecule pharmaceuticals and MS is often not indispensable, a large panel of methods including extensive use of MS must be used for quality control of biopharmaceuticals. Abstract

Bakalářská práce se věnuje problematice týkající se onemocnění karcinomem prostaty. Práce se skládá ze dvou částí, a to z části teoretické a praktické. Část teoretická se zaměřuje na rizikové faktory, histologii, prevenci a diagnostiku karcinomu prostaty, rovněž se zabývá klasifikací nádorů a vysvětluje pojmy gleasonovo score a prostatický specifický antigen (PSA) a jeho významu při včasné diagnostice tohoto závažného onemocnění. Rovněž jsou zde zmíněny současné i minulé možnosti léčby. Praktická část se věnuje popisné statistice souboru pacientů s tímto onemocněním, kteří byli léčeni různými metodami a výskytu možných poléčebných komplikací., This bachelor thesis deals with the topic of prostate cancer. The thesis consists of two parts namely the theoretical part and the practical part. The theoretical part focuses on risk factors, the histology, prevention, and diagnosis of prostate cancer, and in also concerned with the classification of tumors and explains the terms Gleason grading system and prostate - specific antigen (PSA) and its relevance to the diagnosis of this life - threatening illness. Additionally, both present and past treatment options are analyzed. Abstract

The systematic, phylogenetic, and biogeographic aspects of the rich squamate fauna of the Arabian Peninsula are becoming increasingly well understood. The Arabian members of the gecko genus Hemidactylus, the most diverse genus among Arabian squamates, have been the subject of several phylogenetic revisions in recent years. However, large parts of the peninsula lacked thorough sampling, for example, the coastal hyper-arid plains along the Red Sea and some offshore islands. In this study, we examine the genetic, morphological, and ecological diversification of a Hemidactylus clade that straddles the Red Sea and contains ten Arabian and three African species. We compiled a genetic dataset of seven markers (two mitochondrial and five nuclear) to reconstruct their phylogenetic relationships and collected morphological data to assess the level of interspecific morphological disparification. Our results support the existence of four yet undescribed species within the clade – two from Arabia and two from Ethiopia. We provide taxonomic descriptions of the two new Arabian species, one from the western Asir Mountains foothills and one from the Farasan Islands. The new species from the Asir Mountains foothills highlights the role of the southern Arabian coastal desert as an important yet often overlooked local biodiversity hotspot. The new species from the Farasan Islands represents the second vertebrate species endemic to the archipelago. Together with the descriptions of the diagnostic features of both species, we provide complete annotated mitochondrial genomes of both holotypes and of holotypes of two other species from the clade to characterize their mitogenomic composition and architecture.
