Abstract

Crosslinking mass spectrometry (Crosslinking MS) has substantially matured as a method over the last two decades through parallel development in multiple labs, demonstrating its applicability for protein structure determination, conformation analysis and mapping protein interactions in complex mixtures. Crosslinking MS has become a much-appreciated and routinely applied tool especially in structural biology. Therefore, it is timely that the community commits to the development of methodological and reporting standards. Abstract

Model-driven engineering (MDE) provides tools and methods for the manipulation of formal models. In this letter, we leverage MDE for the transformation of production system models into flat files that are understood by general purpose planning tools and that enable the computation of plans, i.e., sequences of production steps that are required to reach certain production goals. These plans are then merged back into the production system model, thus enriching the formalized production system knowledge.

Abstract

The number of publications in the field of chemical cross-linking combined with mass spectrometry (XL-MS) to derive constraints for protein three-dimensional structure modeling and to probe protein-protein interactions has increased during the last years. As the technique is now becoming routine for in vitro and in vivo applications in proteomics and structural biology there is a pressing need to define protocols as well as data analysis and reporting formats. Such consensus formats should become accepted in the field and be shown to lead to reproducible results. This first, community-based harmonization study on XL-MS is based on the results of 32 groups participating worldwide. The aim of this paper is to summarize the status quo of XL-MS and to compare and evaluate existing cross-linking strategies. Our study therefore builds the framework for establishing best practice guidelines to conduct cross-linking experiments, perform data analysis, and define reporting formats with the ultimate goal of assisting scientists to generate accurate and reproducible XL-MS results.

Abstract

Russian wheat aphid (RWA) ( Kurdjumov) is a serious invasive pest of small-grain cereals and many grass species. An efficient strategy to defy aphid attacks is to identify sources of natural resistance and transfer resistance genes into susceptible crop cultivars. Revealing the genes helps understand plant defense mechanisms and engineer plants with durable resistance to the pest. To date, more than 15 RWA resistance genes have been identified in wheat ( L.) but none of them has been cloned. Previously, we genetically mapped the RWA resistance gene into an interval of 0.83 cM on the short arm of chromosome 7D and spanned it with five bacterial artificial chromosome (BAC) clones. Here, we used a targeted strategy combining traditional approaches toward gene cloning (genetic mapping and sequencing of BAC clones) with novel technologies, including optical mapping and long-read nanopore sequencing. The latter, with reads spanning the entire length of a BAC insert, enabled us to assemble the whole region, a task that was not achievable with short reads. Long-read optical mapping validated the DNA sequence in the interval and revealed a difference in the locus organization between resistant and susceptible genotypes. The complete and accurate sequence of the region facilitated the identification of new markers and precise annotation of the interval, revealing six high-confidence genes. Identification of as the most likely candidate opens an avenue for its validation through functional genomics approaches.

32. Radical and incremental innovations performance in Eastern European SMEs: An empirical study of developed and emerging economies

Abstract

Assessing the effectiveness of radical and incremental innovation in SMEs in Eastern European countries is a significant issue in regional economics. In contrast, radical innovation entails the creation of wholly novel goods or organization processes that establish new markets. Progressive enhancements to current goods and processes are referred to as incremental innovation.   This study explains the influence of incremental and radical innovation on performance of small and medium-sized enterprises in Eastern Europe. Employing EUROSTAT data from 2012 to 2021 using regression analysis and a graphical model, we examined basic, radical, and incremental innovation and total researcher performance in SMEs, which could enormously affect business performance in Eastern European countries. Our findings indicated that radical, incremental innovation and total research are all favourably related to the performance of SMEs and an approach to studying various innovations and their impact on SMEs. The particular performance effects of radical and incremental innovations in Eastern European SMEs might vary considerably. To give more specific knowledge and suggestions for SMEs, empirical investigations and comprehensive research are required. Our results can contribute to SMEs both theoretically and practically, providing fresh theoretical perspectives for scholars and helpful management consequences for administrators of SMEs in developing countries.

38. Extending UTAUT with competitive pressure for SMEs digitalization adoption in two European nations: a multi-group analysis.

Abstract

Purpose: In this article, the authors draw-upon an extended unified theory of acceptance and use of technology (UTAUT) and propose a research model involving performance expectancy (PE), effort expectancy (EE), facilitating conditions (FC) and competitive pressure (CP) as potential salient factors explaining the adoption of digitalization in European SMEs. The authors also postulate that there may be cross-cultural differences, thereby leading us to include the country as a moderator in the model. Design/methodology/approach: The authors validate this model with a cross-cultural sample involving 188 owner-managers from the Czech Republic and Slovakia and through the partial least square structural equation modeling (PLS-SEM) techniques as well as multi-group analysis. Findings: The results using the study's global dataset indicate that PE, FC and CP significantly affect owner-managers intentions toward digitalization in SMEs. The authors' application of the multi-group analysis also suggests that although the two countries differ in digitalization adoption intention, the differences are statistically insignificant. In the conclusion, the authors highlight several implications these findings have for theory and practice. Practical implications: The authors recommend that the providers of emerging digital technologies should improve on the performance features of those technologies and ensure they are relevant to the SMEs. By doing so, the adoption of digitalization will grow, because owner-managers of SMEs will have the confidence that adopting such technologies will improve their operations. Second, SMEs are required to provide adequate organizational and technical infrastructure to support digitalization adoption. Originality/value: Aside from being among the few attempts to extend the explanatory power of UTAUT with PE, EE, FC and CP in investigating digitalization adoption in SMEs context, this study also validates its model with rigorous methodological approach as well as three datasets (global, Czech Republic and Slovakia) thereby strengthening the validity of the results. Abstract

Undesirable translocation of soil particles in the direction of movement of machines can occur during soil tillage. The result can be a reduction in soil fertility, especially when the cultivation is deeper. A field experiment was based on sandy-loam cambisoil, where white limestone grit was used as tracers (fraction 10–16 mm). A Kromexim blade cultivator was used for tillage, the other machine was an Akpil disc cultivator. After loosening the soil to a depth of 0.10 m, the displacement of the tracers was eva¬luated. A significant influence of the field slope and the movement direction of the machines on the displacement of the tracers was revealed. With the blade cultivator, the movement of tracers was detected up to a distance of 2.4 m when driving uphill, 3.6 m when driving in the contour direction and 4.2 m when moving down the slope. With the disc cultivator, significantly less displacement of the tracers was found – at all levels of inclination up to a distance of 1 m. The results of the field experiment can contribute to extend the knowledge of one of the negative effects on the fertility of arable soil. [ABSTRACT FROM AUTHOR]
