Background: Adipose-derived mesenchymal stem cells (ADMSCs) have recently been studied for the treatment of knee osteoarthritis. The goal is pain reduction and improvement of joint function leading to superior health-related quality of life. Objectives: The aim of this study was to provide a comprehensive meta-analysis assessing the evidence on the use of ADMSCs in knee osteoarthritis. Design: This is a Meta-analysis of randomised controlled trials. Data Sources and Methods: PubMed/MEDLINE, Embase, and Cochrane Databases were searched for randomized controlled trials using ADMSCs to treat patients with knee osteoarthritis. Only trials comparing ADMSCs to placebo or conservative treatment were included. The outcomes studied were improvement in functional, pain, and quality of life scores along with radiographic findings. Results: A total of four trials were included, representing 138 patients with knee osteoarthritis. The hygrometric method is used to determine new thermodynamic data on water activity and saturated aqueous solution of the water/d-sucrose/ammonium dihydrogen phosphate (ADP) system in a wide range of NH4H2PO4 molality, ranging from 0.1 to 3 mol⋅kg−1, and for various d-sucrose contents from 0 to 4 mol⋅kg−1. Powder X-ray diffraction (XRD) and attenuated total reflection Fourier-transform infrared (ATR-FTIR) spectroscopy were used to characterize the solid state. The Pitzer–Simonson–Clegg model (PSC) is used to fit the experimental data of osmotic coefficient obtained from water activities data. The predicted saturated aqueous solutions, with the PSC model, are in good agreement with experimental data. For the concentration inferior to 1 mol⋅kg−1, the negative deviation from ideality was shown with increasing the ADP concentrations. The estimated values of the activity coefficient of d-sucrose, activity coefficient of ADP, and the Gibbs energy of transfer of ADP from water to mixture (water/d-sucrose) show that both ADP and d-sucrose exert significant salting-out effects on the aqueous solution. Regulatory T cells (Tregs) are key players in the regulation of inflammatory responses. In this study, two natural molecules, namely, sparteine sulfate (SS) and harpagoside (Harp), were investigated for their ability to induce Tregs in human peripheral blood mononuclear cells (PBMCs). PBMCs were isolated from healthy volunteers and grown in the presence or absence of ConA, with TGF-beta, SS or Harp. Expression of the mRNA of FoxP3, TGF-beta, IL-10 and GAPDH was assessed via q-PCR. The expression of Treg markers including CD4, CD25, CD127 and FoxP3 was measured via flow cytometry. The secretion of IL-10 and TGF-beta by cultured cells was assessed by ELISA. Furthermore, the suppressive role of SS and Harp on PBMCs in vitro was tested via allogeneic mixed lymphocyte reaction (MLR). Data obtained show that both compounds increased the expression of FoxP3, TGF-beta and IL-10 mRNA in resting PBMCs but to a lesser extent in activated cells. Moreover, they significantly increased the percent of CD4+CD25+FoxP3+CD127− Tregs in activated and naïve PBMCs. Functionally, both compounds caused a significant reduction in the stimulation index in allogeneic MLR. Together, our data demonstrate for the first time that SS and Harp can induce human Tregs in vitro and therefore have great potential as anti-inflammatory agents.

The role of nonlinear stiffness in the performance of the piezoelectric vibrational energy harvester (pVEH) was discussed. Harmonic balance and numerical methods are applied to characterize the electromechanical response of pVEHs based on Duffing oscillator at a deterministic harmonic excitation of fundamental vibration characteristics (2 Hz, 1 m·s−2), which corresponds to human walking. Then, the response to a vibration with two harmonic waves, which has a fixed fundamental frequency (2 Hz, 1 m·s−2) and a frequency varied from 1.5 to 2.5 Hz. Chimerism is a widespread phenomenon across the tree of life. It is defined as a multicellular organism composed of cells from other genetically distinct entities. This ability to 'tolerate' non-self cells may be linked to susceptibility to diseases like cancer. Here we test whether chimerism is associated with cancers across obligately multicellular organisms in the tree of life. We classified 12 obligately multicellular taxa from lowest to highest chimerism levels based on the existing literature on the presence of chimerism in these species. We then tested for associations of chimerism with tumour invasiveness, neoplasia (benign or malignant) prevalence and malignancy prevalence in 11 terrestrial mammalian species. We found that taxa with higher levels of chimerism have higher tumour invasiveness, though there was no association between malignancy or neoplasia and chimerism among mammals. This suggests that there may be an important biological relationship between chimerism and susceptibility to tissue invasion by cancerous cells. Background: Psoriasis is a chronic inflammatory immune mediated disease arising from interaction between genetic risk variants and the environment. Maternally expressed gene3 (MEG3) is a long noncoding RNA (lncRNA) known for gene transcription regulation and inhibiting proliferation. MEG3 competes with microRNA (miRNA-21) influencing cell proliferation and apoptosis balance. Endoplasmic reticulum (ER) stress proteins promote cell survival via unfolded protein response (UPR) influenced by MEG3. We aimed to detect the possible role of MEG3, miRNA-21 and ER stress proteins in pathogenesis of psoriasis vulgaris., Methods: Human GRP78, ATF6, caspase3 tissue levels were assayed by Enzyme Linked Immunosorbent Assay (ELISA). Assessment of long non-coding MEG3 and miRNA 21 expressions was done by quantitative real time polymerase chain reaction (qRT-PCR)., Results: Expression of MEG3 was significantly downregulated, while miRNA-21 was remarkably upregulated, ER stress proteins GRP78, ATF6, and caspase 3 all showed low levels in homogenized psoriatic lesions when compared to normal skin. miRNA 21 and MEG3 were identified as possible diagnostic markers for psoriasis vulgaris., Discussion: MEG3 is barely expressed in psoriatic lesions while miRNA-21 expression is remarkably elevated but when correlated to each other there was unexpected positive correlation. Background: Sepsis is one of the most common causes of multiorgan failure. Sepsis requires the presence of infection with a resultant systemic inflammatory state. Organ dysfunction occurs from the combination of the two processes. Sepsis is the main cause of mortality at intensive care units, with 30-50% mortality rate for all septic episodes. MicroRNA (miRNA) profile data could be beneficial as a specific diagnostic biomarker for sepsis and systemic inflammatory response syndrome (SIRS)., Methods: Expression of miRNAs -122, -181b, -223 and -146a levels were assayed by quantitative real time polymerase chain reaction (qRT-PCR) in a prospective case control study, where forty septic cases were compared to 40 healthy controls of matched age and gender., Results: miRNAs -122 and -181b were significantly upregulated during early septic conditions, indicating that they could be sensitive and specific biomarkers for diagnosing sepsis. miRNA-223 and miRNA-146a could also represent highly specific and sensitive diagnostic biomarkers, as they were found to be significantly down-regulated. Serum levels of miRNA-223 could be used to predict poor prognosis with 70% sensitivity and 75% specificity, whereas the other three miRNAs could not predict prognosis., Conclusion: Our study shows that all tested miRNAs can be used for early detection of sepsis, with miRNA-223 being predictive of mortality, hence preventing multi-organ failure and reducing mortality, and predicting poor outcomes, thereby assisting in early categorization of ICU patients for rapid appropriate treatment and medico legal aspects.

Boron heteroatom distributions are shown to be significantly different in two closely related layered borosilicates synthesized with subtly different alkylammonium surfactant species. The complicated order and disorder near framework boron sites in both borosilicates were characterized at the molecular level by using a combination of multi-dimensional solid-state nuclear magnetic resonance (NMR) spectroscopy techniques and first-principles calculations. Specifically, two-dimensional (2D) solid-state J-mediated (through-bond) (11)B{(29)Si} NMR analyses provide direct and local information on framework boron sites that are covalently bonded to silicon sites through bridging oxygen atoms. The resolution and identification of correlated signals from distinct (11)B-O-(29)Si site pairs reveal distinct distributions of boron heteroatoms in layered borosilicate frameworks synthesized with the different C16H33N(+)Me3 and C16H33N(+)Me2Et structure-directing surfactant species. The analyses establish that boron atoms are distributed non-selectively among different types of silicon sites in the layered C16H33N(+)Me3-directed borosilicate framework, whereas boron atoms are preferentially incorporated into incompletely condensed Q(3)-type sites in the C16H33N(+)Me2Et-directed borosilicate material. Interestingly, framework boron species appear to induce framework condensation of their next-nearest-neighbor silicon sites in the C16H33N(+)Me3-directed borosilicate. By comparison, the incorporation of boron atoms is found to preserve the topology of the C16H33N(+)Me2Et-directed borosilicate frameworks. The differences in boron site distributions and local boron-induced structural transformations for the two surfactant-directed borosilicates appear to be due to different extents of cross-linking of the siliceous frameworks. The molecular-level insights are supported by density functional theory (DFT) calculations, which show the distinct influences of boron atoms on the C16H33N(+)Me3- and C16H33N(+)Me2Et-directed borosilicate frameworks, consistent with the experimental observations.
