UnlabelledThe human commensal and opportunistic pathogen Candida albicans can switch between two distinct, heritable cell types, named "white" and "opaque," which differ in morphology, mating abilities, and metabolic preferences and in their interactions with the host immune system. Previous studies revealed a highly interconnected group of transcriptional regulators that control switching between the two cell types. Here, we identify Ssn6, the C. albicans functional homolog of the Saccharomyces cerevisiae transcriptional corepressor Cyc8, as a new regulator of white-opaque switching. In A: or α mating type strains, deletion of SSN6 results in mass switching from the white to the opaque cell type. Transcriptional profiling of ssn6 deletion mutant strains reveals that Ssn6 represses part of the opaque cell transcriptional program in white cells and the majority of the white cell transcriptional program in opaque cells. Genome-wide chromatin immunoprecipitation experiments demonstrate that Ssn6 is tightly integrated into the opaque cell regulatory circuit and that the positions to which it is bound across the genome strongly overlap those bound by Wor1 and Wor2, previously identified regulators of white-opaque switching. This work reveals the next layer in the white-opaque transcriptional circuitry by integrating a transcriptional regulator that does not bind DNA directly but instead associates with specific combinations of DNA-bound transcriptional regulators.ImportanceThe most common fungal pathogen of humans, C. albicans, undergoes several distinct morphological transitions during interactions with its host. One such transition, between cell types named "white" and "opaque," is regulated in an epigenetic manner, in the sense that changes in gene expression are heritably maintained without any modification of the primary genomic DNA sequence. Prior studies revealed a highly interconnected network of sequence-specific DNA-binding proteins that control this switch. We report the identification of Ssn6, which defines an additional layer of transcriptional regulation that is critical for this heritable switch. Ssn6 is necessary to maintain the white cell type and to properly express the opaque cell transcriptional program. Ssn6 does not bind DNA directly but rather associates with specific combinations of DNA-bound transcriptional regulators to control the switch. This work is significant because it reveals a new level of regulation of an important epigenetic switch in the predominant fungal pathogen of humans.

BackgroundAlthough telemedicine care has grown in recent years, telepsychiatry is growing at a slower pace than expected, because service providers often hamper the assimilation and expansion of telepsychiatry due their attitudes and perceptions. The unified theory of acceptance and use of technology (UTAUT) is a model that was developed to assess the factors influencing the assimilation of a new technology. We used the UTAUT model to examine the associations between the attitudes and perceptions of psychiatrists in Israel toward telepsychiatry and their intention to use it.MethodsAn online, close-ended questionnaire based on a modified UTAUT model was distributed among psychiatrists in Israel. Seventy-six questionnaires were completed and statistically analyzed.ResultsThe behavioral intention of Israeli psychiatrists to use telepsychiatry was relatively low, despite their perceptions of themselves as capable of high performance with low effort. Nonetheless, they were interested in using telepsychiatry voluntarily. Experience in telepsychiatry, and to a lesser extent, facilitating conditions, were found to be positively correlated with the intention to use telepsychiatry. Psychiatrists have a positive attitude toward treating patients by telepsychiatry and perceive its risk as moderate.DiscussionDespite high performance expectancy, low effort expectancy, low perceived risk, largely positive attitudes, high voluntariness, and the expectancy for facilitating conditions, the intention to use telepsychiatry was rather low. This result is explained by the low level of experience, which plays a pivotal role. We recommend promoting the facilitating conditions that affect the continued use of telepsychiatry when initiating its implementation, and conclude that it is critical to create a sense of success during the initial stages of experience.

14. Spatio-Temporal Low Count Processes with Application to Violent Crime Events

There is significant interest in being able to predict where crimes will happen, for example to aid in the efficient tasking of police and other protective measures. We aim to model both the temporal and spatial dependencies often exhibited by violent crimes in order to make such predictions. The temporal variation of crimes typically follows patterns familiar in time series analysis, but the spatial patterns are irregular and do not vary smoothly across the area. Instead we find that spatially disjoint regions exhibit correlated crime patterns. It is this indeterminate inter-region correlation structure along with the low-count, discrete nature of counts of serious crimes that motivates our proposed forecasting tool. In particular, we propose to model the crime counts in each region using an integer-valued first order autoregressive process. We take a Bayesian nonparametric approach to flexibly discover a clustering of these region-specific time series. We then describe how to account for covariates within this framework. Both approaches adjust for seasonality. We demonstrate our approach through an analysis of weekly reported violent crimes in Washington, D.C. between 2001-2008. Our forecasts outperform standard methods while additionally providing useful tools such as prediction intervals.

16. Workload forecasting for a call center: Methodology and a case study

Today's call center managers face multiple operational decision-making tasks. One of the most common is determining the weekly staffing levels to ensure customer satisfaction and meeting their needs while minimizing service costs. An initial step for producing the weekly schedule is forecasting the future system loads which involves predicting both arrival counts and average service times. We introduce an arrival count model which is based on a mixed Poisson process approach. The model is applied to data from an Israeli Telecom company call center. In our model, we also consider the effect of events such as billing on the arrival process and we demonstrate how to incorporate them as exogenous variables in the model. After obtaining the forecasted system load, in large call centers, a manager can choose to apply the QED (Quality-Efficiency Driven) regime's "square-root staffing" rule in order to balance the offered-load per server with the quality of service. Implementing this staffing rule requires that the forecasted values of the arrival counts and average service times maintain certain levels of precision. We develop different goodness of fit criteria that help determine our model's practical performance under the QED regime. The fundamental challenge in implementing inclusive schools is related to accepting students with special needs by other students. Lack of acceptance and respect for differences often trigger acts of bullying. There are insufficient studies describing that the perpetrator of bullying in school is a student with special needs. This study aims to measure the effectiveness of the educare program in improving students' pro-social attitudes to prevent bullying in inclusive schools. The Educare Program consisted of four lesson plan themes and became a treatment instrument. Single subject research (SSR) was used for the research method, implementing the baseline-intervention-maintenance (AB+M) phase with multiple baselines across subjects. Three students, including a student with a disability and a low pro-social attitude, were the subjects of this study. The data were collected through observation and analyzed through graphical visual inspection focusing on trend, level, latency, and percentage of all non-overlapping data (PAND). The results of the study presented in polygon graphs and PAND showed that the Educare Program is very effective in increasing students' pro-social attitudes. Almost all scores of pro-social attitudes obtained through trend with PAND reached 96.11%.

Sequence-specific DNA-binding proteins are among the most important classes of gene regulatory proteins, controlling changes in transcription that underlie many aspects of biology. In this work, we identify a transcriptional regulator from the human fungal pathogen Candida albicans that binds DNA specifically but has no detectable homology with any previously described DNA- or RNA-binding protein. This protein, named White-Opaque Regulator 3 (Wor3), regulates white-opaque switching, the ability of C. albicans to switch between two heritable cell types. We demonstrate that ectopic overexpression of WOR3 results in mass conversion of white cells to opaque cells and that deletion of WOR3 affects the stability of opaque cells at physiological temperatures. Genome-wide chromatin immunoprecipitation of Wor3 and gene expression profiling of a wor3 deletion mutant strain indicate that Wor3 is highly integrated into the previously described circuit regulating white-opaque switching and that it controls a subset of the opaque transcriptional program. We show by biochemical, genetic, and microfluidic experiments that Wor3 binds directly to DNA in a sequence-specific manner, and we identify the set of cis-regulatory sequences recognized by Wor3. Bioinformatic analyses indicate that the Wor3 family arose more recently in evolutionary time than most previously described DNA-binding domains; it is restricted to a small number of fungi that include the major fungal pathogens of humans. These observations show that new families of sequence-specific DNA-binding proteins may be restricted to small clades and suggest that current annotations--which rely on deep conservation--underestimate the fraction of genes coding for transcriptional regulators.

When evolutionary biology comes to mind, it is often accompanied by contemplating the vast amount of morphological or behavioral differences that exist between species. Whether Darwin’s finches or the pattern on an insect wing, we often reference these types of striking apparent visual differences in discussing the power of evolution. However, there are a growing number of examples that suggest that even core, fundamental processes like metabolism and mating are subject to evolutionary changes. In the face of these underlying evolutionary changes, the fundamental logic of these core processes must be conserved. A biosynthetic pathway must be upregulated in the face of nutrient starvation (Ch. 1), and a cellular identity must be conserved in order for mating to occur in yeast (Ch. 2), but how these logical outputs are maintained in spite of evolution acting on the molecules involved in these processes is the outstanding question explored in this work.

Totemisme adalah istilah menunjuk pada suatu kepercayaan atau agama yang hidup pada sebuah komunitas atau organisasi. Mereka mempercayai bahwa adanya daya atau sifat ilahi yang dikandung didalamnya merupakan pegangan hidup mereka. Pada temuan Durkheim tentang totemisme sebagai sistem agama yang paling sederhana dan primitif serta menjadi lambang organisasi sosial yakni oleh masyarakat tertentu. Orang Mentawai memiliki kepercayaan sendiri yang dianut oleh orang asli suku Mentawai yaitu Arat Sabulungan. Mereka percaya bahwa benda memiliki roh dan jiwa yaitu Seperti dalam pembangunan Uma, orang Mentawai memiliki ritual dalam pembangunan Uma. Jika ritual itu tidak dilaksanakan akan mengakibatkan orang yang membangun Uma celaka dan sakit. Sehingga dalam ritual pembangunan Uma orang Mentawai memaknai hal itu adalah sebagai keselamatan bagi mereka dan terhindar dari kesialan dan penyakit. Adapun tujuan penulis, ingin lebih menggali makna Arat Sabulungan dalam pembangunan Uma bagi orang Mentawai. Hasil penelitian diukur menggunakan metode kualitatif dengan teknik wawancara tertutup.

Tenaga pastoral perlu dasar ilmu pastoral, melalui perkuliahan pastoral dasar calon tenaga pastoral dapat memahami dasar ilmu pastoral, sehingga dapat siap berkarya di tengah masyarakat. Mata kuliah pastoral dasar di STP-IPI Malang menekankan pada praktik di lapangan seperti membaca kitab suci, membuat renungan, mengikuti peribadatan, dan lainnya Artikel ini akan membahas pelaksanaan pastoral dasar di STP-IPI Malang, faktor yang mendukung pelaksanaan pastoral dasar, serta faktor penghambat pelaksanaan pastoral dasar. Penelitian ini menggunakan rancangan penelitian kualitatif. Subyek penelitian adalah Mahasiswa tingkat II Program Studi Pendidikan Pengajaran Agama Katolik STP-IPI Malang. Metode pengumpulan data menggunakan cara wawancara dan observasi. Teknik analisa data menggunakan rumus Miles dan Huberman. Berdasarkan penelitian mahasiswa tidak sepenuhnya terlibat aktif dalam pelaksanaan pastoral dasar. Sebagian besar memiliki tujuan yang berbeda dengan tujuan umum yang ada di dalam diktat atau modul pastoral dasar.

- Abstract
Negara republik Indonesia adalah bangsa yang memiliki beragam budaya dengan sifat keanekaragaman meliputi budaya, agama, ras, bahasa, suku, tradisi dan sebagainya. Dalam bangsa yang pluralistik, sering kali terdapat krisis dan pertikaian yang terjadi antara kelompok-kelompok budaya dan membawa pengaruh pada kerukunan hidup. Tujuan penulisan ini adalah membahas keragaman budaya bangsa Indonesia, toleransi, moderisasi beragama dalam keragaman dan peran penyuluh agama dalam mewujudkan kedamaian bangsa Indonesia. Metode yang digunakan adalah penelitian pustaka. Kesimpulan kajian ini adalah bahwa dalam kehidupan multikultural diperlukan pemahaman dan kesadaran multibudaya yang menghargai perbedaan, kemajemukan dan kemauan berinteraksi dengan siapapun secara adil. Diperlukan sikap moderasi beragama berupa pengakuan atas keberadaan pihak lain, memiliki sikap toleran, penghormatan atas perbedaan pendapat dan tidak memaksakan kehendak dengan cara kekerasan. Diperlukan peran pemerintah, tokoh masyarakat, dan para penyuluh agama untuk mensosialisasikan, menumbuhkembangkan sikap toleransi dan moderasi beragama kepada masyarakat sehingga terbentuknya kehidupan yang tentram dan nyaman.

Orang Muda Katolik merupakan agen perubahan dalam Gereja yang diharapkan dapat memberi kesaksian hidup ditengah dunia modern saat ini yang didasarkan pada sabda Yesus kepada para Rasul. Sedangkan dalam kehidupan sehari-hari, orang muda katolik memiliki kebiasaan suka mabuk-mabukan, bermain judi dan jarang terlibat dalam kegiatan menggereja. Maka pembinaan dalam kegiatan Gereja diharapkan dapat mendorong semangat Orang Muda Katolik untuk hadir dan terlibat aktif dalam kegiatan Gereja. Tujuan penelitian ini adalah untuk mengetahui sejauh mana Keterlibatan Orang Muda Katolik Dalam Kegiatan Menggereja. Penelitian ini menggunakan rancangan penelitian dengan pendekatan kuantitatif. Populasi data sebanyak 40 orang, dengan sampel data sebanyak 40 orang responden yang merupakan anggota dari Orang Muda Katolik. Metode pengumpulan data yang digunakan dalam penelitian ini adalah menyebarkan angket. Teknik analisis data yang digunakan yaitu menggunakan rumus Skoring. Hasil dari penelitian ini diketahui dari pengolahan data yaitu teknik skoring. Berdasarkan hasil pengolahan data secara keseluruhan menggunakan teknik skoring diperoleh skor 1.71 artinya Orang Muda Katolik terlibat aktif Dalam Kegiatan Menggereja.

Tradisi temu manten ialah tradisi yang dilakukan masyarakat jawa. Tradisi tersebut biasa digelar ketika seseorang memiliki hajat untuk menikahkan putra-putrinya. Temu manten juga disebut dengan upacara Panggih, artinya upacara pertemuan kedua belah pihak pengantin laki-laki dan pengantin perempuan di kediaman mempelai perempuan. Adapun nilai-nilai Kristiani yang terkandung adalah : kesabaran, kesetiaan, kelemahlembutan, cinta kasih, pelayanan, kesetaraan, dan tidak mencari keuntungan. Tujuan dari penulisan ini adalah untuk mengetahui nilai-nilai Kristiani yang terkandung dalam prosesi Temu Manten dalam pernikahan adat jawa khususnya di Paroki Maria Annunciata Lohdalem Keuskupan Malang yang pada dasarnya masyarakat atau umat kurang memahami arti dan makna nilai-nilai Kristiani yang terkandung didalam prosesi Temu manten dalam pernikahan adat jawa. Penelitian ini menggunakan Studi Pustaka dengan cara mempelajari atau menganalisa tentang upacara Temu Manten pada perkawinan adat Jawa. Hasil penelitian menunjukkan bahwa, pelaksanaan tradisi Temu Manten atau panggih tidak terlepas juga dari nilai-nilai Kristiani yang ada khususnya pada Agama Katolik.

Agama merupakan bentuk keyakinan manusia terhadap sesuatu yang bersifat adikodrati yang berkaitan dengan Tuhan Sang Maha Kuasa yang menyertai seluruh ruang lingkup kehidupan manusia baik individu maupun masyarakat. Dengan mengetahui fungsi yang terkandung di dalam agama, maka orang yang beragama dapat merasakan nilai-nilai yang luhur terutama dalam berperilaku ditengah masyarakat. Agama memiliki fungsi yang harus dijaga dan dikembangkan dalam kehidupan bermasyarakat antara lain: Fungsi Edukatif, fungsi penyelamat, fungsi sebagai pendamaian, fungsi kontrol sosial, fungsi pemupuk rasa solidaritas, fungsi transformatif, fungsi kreatif, fungsi sublimatif. Kesimpulannya Agama berfungsi sebagai sarana dan lambang keagamaan dalam masyarakat, kesakralan bersumber pada kekuatan yang dinyatakan berlaku oleh masyarakat secara keseluruhan bagi setiap anggotanya, dan fungsinya adalah mempertahankan dan memperkuat rasa solidaritas, kewajiban serta perilaku sosial. Penelitian ini menggunakan metode kualitatif deskriptif melalui studi pustaka. Peneliti mendeskripsikan dan menguraikan fungsi agama terhadap perilaku sosial masyarakat. Data tersebut diolah melalui beberapa artikel, jurnal dan berita.

Children with autism are children who have a nervous system disorder, caused by heredity factor. Autism is not a symptom of disease but it is a syndrome (a collection of symptoms) in which social deviations, language abilities and concerns are concerned. With regard to language skills, parents of children with autism often seek ways to allow children to speak fluently as a child in general, regardless of the child's abilities. This is the obstacle in interacting. The purpose of this study was to determine the expressive communication ability of children with autism using PECS (Picture Exchanges Communication System) method. This research uses Pre-Experimental Design method in the form of one-group pretest-posttest design. This form is used because researchers want to know the value generated by each subject after treatment is done. Method of data collection using observation method. The results showed an increase in expressive communication of children with autism after using the PECS method. The highest score was obtained by MM and LV subjects, with an increase of expressive communication value of 12 each. While KF subjects obtained an increase in value of 10 and the subject of VR 8. Thus, PECS Method could be one of the reference to improve expressive communication of children with autism.

Sebagian masyarakat Indonesia, belum tangguh dalam menghadapi pengalaman krisis seperti sakit penyakit, bencana, penderitaan, kematian dan pengalaman krisis lainnya dalam hidup meski mereka berkuasa dan kaya raya. Karena sadar bahwa, dirinya merasa tidak mampu melewati pengalaman- pengalaman krisis, ia lebih memilih untuk percaya kepada kekuatan-kekuatan di luar iman yang dianutnya. Masyarakat Jawa sendiri memiliki tradisi budaya yaitu sesajen. Tradisi sesajen merupakan sebuah tindakan rasa syukur masyarakat terhadap apa yang terjadi dalam kehidupan. Tindakan inilah merupakan salah satu cara bagi masyarakat Jawa menanggulangi krisis-krisis dalam hidup. Tujuan penelitian ini adalah 1) Menganalisis pengalaman krisis manusia 2) mendeskripsikan kebudayaan sesajen sebagai tanda ucapan syukur atas kehidupan masyarakat Jawa, 3) mendeskripsikan sosiologi agama dalam tradisi sesajen. Dalam penulisan ini penulis menggunakan metode kualitatif melalui studi pustaka dengan menggunakan tiga teknik SLR. Hasil dari penulisan ini mau menunjukan bahwa, masyarakat Jawa masih mempercayai budaya tradisi sesajen sebagai ucapan syukur atas kehidupan yang diperoleh.

Tor-tor merupakan salah satu tarian yang berasal dari suku Batak. Penggunaan Tor-tor sering dijumpai pada acara pernikahan, kematian, dan pesta Batak. Selain itu tari Tor-tor menjadi sebuah ekspresi gerakan estetis serta artistik di mana setiap tarian yang ditampilkan memiliki makna dari setiap gerakannya. Dalam Gereja Katolik penggunaan tari Tor-tor digunakan pada saat perarakan dan persembahan misa Inkulturasi. Yang menjadi permasalahan umat kurang memahami makna penggunaan tari Tor-tor dalam perayaan Ekaristi dan sebagian umat menyampaikan bahwa penggunaan tari Tor-tor dalam perayaan Ekaristi justru mengurangi kesakralan Misa Ekaristi itu sendiri. Penelitian ini bertujuan untuk mengetahui pemahaman umat mengenai makna serta tujuan tari Tor-tor dalam perayaan Ekaristi. Metode yang digunakan dalam penelitian ini adalah metode deskriptif. Teknik pengumpulan data adalah teknik observasi dan wawancara. Pengolahan data dilakukan dengan menggunakan analisis kualitatif. Adapun hasilnya bahwa tari Tor-tor tidak memiliki makna khusus dalam perayaan Ekaristi, penggunaannya hanya sebagai hiburan misa inkulturasi.

Summary: Background: Screening for Barrett's oesophagus relies on endoscopy, which is invasive and few who undergo the procedure are found to have the condition. We aimed to use machine learning techniques to develop and externally validate a simple risk prediction panel to screen individuals for Barrett's oesophagus. Methods: In this prospective study, machine learning risk prediction in Barrett's oesophagus (MARK-BE), we used data from two case-control studies, BEST2 and BOOST, to compile training and validation datasets. From the BEST2 study, we analysed questionnaires from 1299 patients, of whom 880 (67·7%) had Barrett's oesophagus, including 40 with invasive oesophageal adenocarcinoma, and 419 (32·3%) were controls. We randomly split (6:4) the cohort using a computer algorithm into a training dataset of 776 patients and a testing dataset of 523 patients. We compiled an external validation cohort from the BOOST study, which included 398 patients, comprising 198 patients with Barrett's oesophagus (23 with oesophageal adenocarcinoma) and 200 controls. We identified independently important diagnostic features of Barrett's oesophagus using the machine learning techniques information gain and correlation-based feature selection. We assessed multiple classification tools to create a multivariable risk prediction model. Internal validation of the model using the BEST2 testing dataset was followed by external validation using the BOOST external validation dataset. From these data we created a prediction panel to identify at-risk individuals. Findings: The BEST2 study included 40 diagnostic features. Of these, 19 added information gain but after correlation-based feature selection only eight showed independent diagnostic value including age, sex, cigarette smoking, waist circumference, frequency of stomach pain, duration of heartburn and acidic taste, and taking antireflux medication, of which all were associated with increased risk of Barrett's oesophagus, except frequency of stomach pain, with was inversely associated in a case-control population. Logistic regression offered the highest prediction quality with an area under the receiver-operator curve (AUC) of 0·87 (95% CI 0·84–0·90; sensitivity set at 90%; specificity of 68%). In the testing dataset, AUC was 0·86 (0·83–0·89; sensitivity set at 90%; specificity of 65%). In the external validation dataset, the AUC was 0·81 (0·74–0·84; sensitivity set at 90%; specificity of 58%). Interpretation: Our diagnostic model offers valid predictions of diagnosis of Barrett's oesophagus in patients with symptomatic gastro-oesophageal reflux disease, assisting in identifying who should go forward to invasive confirmatory testing. Our predictive panel suggests that overweight men who have been taking antireflux medication for a long time might merit particular consideration for further testing. Our risk prediction panel is quick and simple to administer but will need further calibration and validation in a prospective study in primary care. Background Inappropriate antibiotic use in acute respiratory infections (ARIs) is a major public health concern, however data for people living with HIV (PLHIV) are limited. Methods The HIV Virtual Cohort Study is a retrospective cohort of adult Department of Defense beneficiaries. Male PLHIV cases (n=2,413) were matched 1:2 to controls without HIV (n=4,826) by age, gender, race/ethnicity, and beneficiary status. ARI encounters between 2016-2020 and corresponding antibiotic prescriptions were characterized as always, sometimes, or never appropriate based on ICD-10 coding. Incidence of ARI encounters and antibiotic appropriateness were compared between PLHIV and controls. Subgroup analyses were assessed by CD4 count and viral load suppression on antiretroviral therapy (ART). Results Mean rates of ARI encounters were similar for PLHIV (1,066 per 1,000 person-years) and controls (1,010 per 1,000 person-years), however the rate was double among PLHIV without VL suppression (2,018 per 1,000 person-years). Antibiotics were prescribed in 26% of encounters among PLHIV compared to 34% for controls (p= Discussion ARI encounters were similar for PLHIV and those without HIV, however, PLHIV with lower CD4 counts and/or non-suppressed VL had more frequent ARI visits. Inappropriate antibiotic use for ARIs was high in both populations and focused interventions to improve antibiotic appropriateness for prescribers caring for PLHIV should be pursued.

Baik dari segi tempat, waktu, benda, alat, ataupun orang-orang yang menjalankan upacara tersebut. Adapun makna dari upacara kematian dan hewan kurban ini ialah bahwa upacara kematian merupakan penghormatan terakhir bagi yang meninggal, sedangkan hewan kurban dimaknai sebagai penjalin hubungan yang harmonis dengan arwah leluhur, dan untuk memperbaiki hubungan yang telah rusak akibat adanya anggota kabihu (marga/klan) yang melanggar adat istiadat. Tujuan Penelitian ini adalah untuk mengetahui makna dari upacara kematian dalam keagamaan Marapu. Jenis Penelitian menggunakan metode kualitatif deskriptif tentang makna upacara kematian dan hewan kurban dalam keagamaan Marapu. Teknik pengumpulan data yaitu melalui pengumpulan data dengan studi pustaka. Adapun upaya yang dilakukan ialah menggali makna upacara kematian dan hewan kurban dalam keagamaan Marapu dari berbagai sumber.

Gereja memandang perkawinan sebagai panggilan hidup yang tertinggi dan suci karena Allah sendiri yang memimpin dan memberkati melalui sakramen perkawinan sebagai hidup berkeluarga. Sakramen perkawinan sendiri memiliki perjanjian yaitu sekali seumur hidup atau tidak terceraikan. Meski dikatakan tidak terceraikan tidak sedikit juga pasangan Katolik yang tidak bisa mempertahankan kesetiaannya dalam berumah tangga. Penelitian ini menggunakan metode studi pustaka. Studi pustaka merupakan suatu kajian yang berdasarkan khazanah ilmu yang bersumber pada buku-buku kepustakaan yang sesuai dengan pokok permasalahan. Melihat situasi sekarang ini yang maraknya Perceraian dikarenakan kurangnya kesetiaan satu pasangan dengan contoh perselingkuhan, hal ini menunjukkan bahwa kesetiaan adalah problem bagi pasangan suami istri yang menyepelekan keskralan janji perkawinan. Berdasarkan latar belakang tersebut, penulis melakukan studi lapangan terhadap pasangan katolik yang aktif dalam kegiatan gereja dan memberikan komitmen yang baik terhadap pernikahannya juga memotivasi bagi pasangan Katolik yang tidak aktif dalam kegiatan menggereja maupun kerohanian dan mendorong pasangan Katolik agar hidup sesuai Ajaran Gereja.

- Abstract
Pembina iman anak yang ada di Paroki Santo Andreas Tidar berasal dari lima lingkungan dan OMK. Dalam penelitian ini penulis mengemukakan pokok permasalahan yaitu: Sejauh mana peran pembina dalam pelaksanaan bina iman anak usia dini dan apakah ada perbedaan peran antar pembina yang satu dengan pembina yang lain dalam pelaksanaan bina iman anak usia dini di Paroki Santo Andreas Tidar. Penelitian ini menggunakan penelitian deskriptif kuantitatif. Pengumpulan data menggunakan angket tertutup kepada 30 orang dan teknik analisa data menggunakan rumus Skoring dan Uji Binomial. Hasil pengolahan data dengan rumus skoring diperoleh skor rata-rata 2,51 artinya pembina menjalankan perannya dengan baik. Pengolahan data menggunakan uji binomial diperoleh nilai z= 1,3401 nilai tersebut lebih tinggi dari taraf signifikan 0,05 artinya tidak ada perbedaan yang signifikan antara peran pembina yang satu dengan pembina yang lain. Dari hasil pengolahan data menggunakan rumus binomial tersebut maka H(0) diterima dan H(a) ditolak. Oleh karena itu, para pembina diharapkan agar tetap semangat dalam menjalankan perannya dalam dalam pelaksanaan bina iman anak usia dini serta dala menerapkan hal-hal baru yang relevan di kalangan anak usia dini.

- Abstract
Peristiwa memprihatinkan selalu terjadi dalam hidup bermasyarakat, seperti pertengkaran, memukul, saling mendiamkan, bahkan membunuh sesama diakibatkan karena sakit hati dan dendam yang berkepanjangan. Hal ini terjadi karena tidak ada pengampunan. Tujuan penelitian ini untuk mengungkapkan dan mendeskripsikan konsep pengampunan yang ada di dalam Injil Matius dan upaya umat Kristiani agar mendorong para pelaku penyimpangan pengampunan untuk kembali ke jalan Tuhan sehingga memperoleh ampunan dari Tuhan dan sesama. Jenis penelitian ini menggunakan metode kualitatif deskriptif tentang pengampunan menurut Injil Matius serta upaya umat Kristiani memperoleh pengampunan dari Tuhan dan sesama. Teknik pengumpulan data menggunakan wawancara mendalam kepada sejumlah informan yang terlibat di dalam proses pengampunan umat Kristiani dan penelusuran dokumen terkait pengampunan di media massa daring. Hasil penelitian disimpulkan bahwa umat Kristiani tidak terlepas dari salah dan dosa sepanjang hidup. Ada upaya untuk kembali ke jalan Tuhan, yaitu mengaku dosa, sharing, memaafkan, diskusi, dan saling mendoakan demi memperoleh kedamaian dalam hidup.

- Abstract
Kitab Suci Perjanjian Baru menyampaikan dengan jelas makna luhur dari perkawinan berdasarkan ajaran Yesus. Yesus sangat menghargai kesetiaan dan menolak perceraian. Katekese adalah pembinaan iman kepada jemaat, khususnya tentang ajaran Kristen, yang diberikan secara organisatoris dan sistematis, agar umat memasuki kepenuhan hidup Kristen. Kepada jemaat di Tesalonika Paulus menasihati agar umat Kristen menghayati dan menghargai perkawinan dengan menjauhkan diri dari percabulan dan seks bebas (1Tes 4:3-8). Kepada jemaat di Korintuspun ia mengecam percabulan dan segala bentuk penyelewengan seksual karena bertentangan dengan etika Kristen dan kekudusan perkawinan (1Kor 5-7). Dalam Surat Pastoralnya ia juga melawan aliran sesat yang bertendensi anti perkawinan, emansipatif dan tidak menghargai anak-anak (1Tim 3:4; 5:14; Tit 2:4-5). Karenanya, Paulus memberi tata tertib dalam hidup berumah tangga yang mengatur hubungan suami-isteri dan orang tua-anak (Ef 5:22-6:4 bdk. 1Ptr 3:1-7). Yesus menegaskan bahwa hubungan suami-isteri yang telah dipersatukan oleh Allah itu tidak boleh diceraikan manusia (Mrk 10:2-12; Mat 19:3-12).

Situasi sosial yang terjadi di Desa Sidodadi dan Gajahrejo yang disebut kampung moderasi beragama adalah masyarakat belum mengenyam pendidikan formal, terjadi perkawinan sedarah, agama minoritas kesulitan membangun rumah ibadat sekaligus kehilangan kebebasan untuk memilih agama, dan masyarakat tidak memiliki tokoh sebagai penggerak untuk hidup dalam toleransi. Metode penelitian yang digunakan adalah kualitatif fenomenologi melalui wawancara mendalam, observasi, dan dokumentasi. Analisis data menggunakan teknik fenomenologi, yakni verbatim, horizonalization, cluster of meaning, dan kesimpulan. Hasil dan Pembahasan; pertama, Pandangan masyarakat mengenai toleransi. Masyarakat kampung moderasi beragama memiliki pandangan menerima, menghargai, dan menghormati perbedaan keyakinan yang ada dalam masyarakat. Perbedaan membawa keindahan dalam hidup bersama. Kedua; sikap masyarakat, memberi kebebasan kepada sesama untuk memilih keyakinan, menjalankan ibadah, dan membangun rumah ibadat bagi minoritas. Ketiga, upaya menanamkan toleransi dalam kehidupan masyarakat melalui pendidikan formal dan informal. Pendidikan formal ditempuh melalui Taman Kanak-Kanak dan Sekolah Dasar. Sedangkan pendidikan informal diupayakan melalui berbagai kegiatan yang dapat menciptakan kekerabatan dan harmoni dalam masyarakat. Kegiatan kemasyarakatan menjadi wadah untuk membangun dan memupuk toleransi. Simpulan: toleransi telah menjadi bagian integral dalam masyarakat di kampung moderasi beragama. Implementasi toleransi dilakukan dengan mengejawantahkan esensi ajaran agama yang melindungi martabat manusia serta membangun kemaslahatan berdasarkan prinsip adil, berimbang, harmoni yang berlandaskan Pancasila.

Perkembangan dunia dewasa ini sangat maju ditandai kehadiran ilmu pengetahuan dan teknologi yang canggih. Perkembangan dunia yang sangat maju ini menuntut manusia untuk discerment diri secara baik terhadap perkembangan yang terjadi, terlebih kaum muda. Kaum muda perlu membagi waktu dan discernment dengan baik antara kehidupan sosialnya dan kehidupan rohaninya. Menurut pengalaman penulis, di Stasi Santa Theresia Nusa Jaya, kegiatan Orang Muda Katolik (OMK) masih berjalan dengan baik dan lancar namun itu tidak menjamin perilaku sosial mereka di masyarakat. Maka tujuan penelitian ini adalah untuk mengetahui sejauh mana kegiatan rohani dilaksanakan oleh Kaum Muda Katolik di Stasi Santa Maria Nusa Jaya. Jenis penelitian ini adalah penelitian kuantitatif, yang merupakan subjek penelitiannya adalah Kaum Muda yang berusia 13-35 tahun, yang berjumlah 32 orang. Pengumpulan data menggunakan angket tertutup, sedangkan teknis analisa data menggunakan rumus F porsen dan table scoring. Berdasarkan data diperoleh hasil yaitu, ada pengaruh antara kegiatan rohani dan perilaku sosial.

During translation initiation, eIF4G1 dynamically interacts with eIF4E and eIF1. While the role of eIF4E–eIF4G1 is well established, the regulatory functions of eIF4G1–eIF1 are poorly understood. Here, we report the identification of the eIF4G1–eIF1 inhibitors i14G1-10 and i14G1-12. i14G1s directly bind eIF4G1 and inhibit translation in vitro and in the cell, and their effects on translation are dependent on eIF4G1 levels. Translatome analyses revealed that i14G1s mimic eIF1 and eIF4G1 perturbations on the stringency of start codon selection and the opposing roles of eIF1–eIF4G1 in scanning-dependent and scanning-independent short 5′ untranslated region (UTR) translation. Remarkably, i14G1s activate ER/unfolded protein response (UPR) stress-response genes via enhanced ribosome loading, elevated 5′UTR translation at near-cognate AUGs, and unexpected concomitant up-regulation of coding-region translation. These effects are, at least in part, independent of eIF2α-phosphorylation. Interestingly, eIF4G1–eIF1 interaction itself is negatively regulated by ER stress and mTOR inhibition. Thus, i14G1s uncover an unknown mechanism of ER/UPR translational stress response and are valuable research tools and potential drugs against diseases exhibiting dysregulated translation.
