The utilization of waste and by-products as precursors in alkali-activated materials has great importance in the reduction of environmental pollution on slopes, landfills, and highways. Properties studies of alkali-activated mixtures using red ceramic waste (RCW) and blast furnace slag (BFS) have been scarcely investigated in the literature. This paper aims to evaluate the properties and behavior of alkali-activated pastes based on RCW and BFS. The influence of the following parameters was evaluated: silica modulus (Ms) between 1.0 and 1.3 and RCW content (25, 50, and 75 %). The compressive strength, workability, and reaction kinetics were analyzed. In addition, embodied environmental impacts and FTIR microstructural analyses were performed. Flow diameters in the range of 147.3 mm to 163 mm (Ms = 1.3) and 142.4 mm to 154.1 mm (Ms = 1.0) were observed and the RCW incorporation slightly reduced mixtures workability. An optimum RCW content in the range of 25 %-50 % was identified, which provided excellent mechanical and microstructural properties reaching values up to 68.68 MPa for the best mixture and values above 50 MPa for systems with 50 % RCW. Higher RCW contents also led to reaction delays according to the isothermal calorimetry test. The increase in Ms generated higher band intensities of the Si-O-T chemical bonds. The mixtures evaluated had significantly lower CO2 emissions and equivalent energy consumption than OPC-based binders, with reductions of up to 65.6 % (CO2 emission) and 75.2 % (energy consumption), indicating the environmental benefit provided by this type of mixture.

Immune cells are thought to be involved in a destructive cycle of sterile cerebral inflammatory responses in neurodegenerative diseases such as Parkinson’s Disease (PD). Despite their peripheral origin, immune cells may enter the CNS due to impaired blood–brain barrier function and may potentially contribute to neuronal damage. Hence, specific characteristics of peripherally activated immune cells could help in understanding neurodegeneration in PD and could potentially serve as accessible disease markers. To investigate immune cell activation status, the expression of receptors for cell surface molecules CD161, NKG2A, NKG2C and NKG2D as well as chemokine receptors CCR6, CXCR2, CXCR3 and CCR5 associated with neurodegenerative diseases was investigated. The frequencies of peripheral CD8+ T-cells expressing the inhibitory and activating receptors NKG2A and NKG2C, and the activating receptor NKG2D were higher in PD patients than in healthy matched controls. The frequencies of NKG2C+CD8− cells were also higher, whereas the frequencies of CD161+ cells were not significantly different. Of the chemokine receptor-expressing cells, only the proportion of CD4−CD56+CCR5+ T-cells was higher in PD patients than in the controls. These observations support the hypothesis that an imbalance in the activation state of T-cells plays a role in the pathological processes of PD and suggest that peripheral blood immune cell phenotypes could be specific early markers for inflammation in PD.

Abstract Background Organ transplantation represents the most effective and acceptable therapy for end-stage organ failure. However, its frequent practice often leads to a shortage of organs worldwide. To solve this dilemma, some countries, such as Portugal, have switched from an opt-in to an opt-out system, which has raised concerns about respect for individual autonomy. We aimed to evaluate whether young university students are aware of this opt-out system so that they can make informed, autonomous and conscious decisions, as well as to identify the factors that determine a positive attitude toward post-mortem organ donation. Methods An observational, cross-sectional study was developed and a questionnaire was administered to first-year students from six faculties of the University of Porto. Results Of the 841 participants, 60% were unaware that Portugal had adopted an opt-out system. Among the informed individuals, their main sources of information included social media, internet, and family. Furthermore, only 48% of all participants agreed with the current opt-out system. RESUMO Neste artigo, analisamos as metas de inflação, com foco no mecanismo de transmissão de juros e preços. Seus fundamentos teóricos são analisados com base na literatura convencional e alternativa. Discutimos o desempenho recente do setor de varejo no Brasil, destacando como a política monetária afeta os preços, direta e indiretamente. Concluímos a avaliação crítica da meta de inflação. Nossa principal hipótese é que as taxas de juros devem ser vistas não apenas como um instrumento para controlar o nível geral de preços e o nível de atividade, regulando indiretamente a demanda agregada. Também deve ser visto como um componente fundamental e direto dos preços formados no varejo brasileiro. O grande risco de uma meta de inflação, por um lado, é aumentar a deflação e, por outro, criar estagflação - desemprego e inflação.

The in vivo fertilization process occurs in the presence of follicular fluid (FF). The aim of this study was to evaluate the effect of in vitro fertilization medium supplementation with 5% or 10% bovine follicular fluid (BFF) on the production of in vitro bovine embryos. FF was collected from ovarian follicles with a diameter of 8–10 mm, and cumulus–oocyte complexes (COCs) were co‐incubated with sperm for 24 h in the commercial medium BotuFIV® (BotuPharma©), being distributed among the experimental groups: oocytes fertilized in a control medium; oocytes fertilized in a medium supplemented with 5% BFF; and oocytes fertilized in a medium supplemented with 10% BFF. After fertilization, the zygotes were cultured in vitro for 8 days. Embryo development was assessed through cleavage rates (day 2) and blastocyst formation rates (day 8). The relative expression of the genes OCT4, IFNT2, BAX, HSP70 and SOD2 was measured using the real‐time polymerase chain reaction method. There was no difference (p >.05) among the different experimental groups in terms of cleavage rates and blastocyst formation rates. Regarding the gene expression results, only the blastocysts from oocytes fertilized with 10% BFF showed significantly lower expression of IFNT2 (p =.003) and SOD2 (p =.01) genes compared to blastocysts from oocytes fertilized in control medium alone, while there was no difference between blastocyst from oocytes fertilized in control medium and the ones from oocytes fertilized with 5% BFF. In addition to this, the blastocysts from oocytes fertilized with 5% BFF showed significantly reduced levels of expression of the heat shock protein HSP70 (p <.001) and the pro‐apoptotic protein BAX (p =.015) compared to blastocysts from oocytes fertilized with control medium. This may indicate that lower supplementation of BFF to the IVF medium creates a more suitable environment for fertilization and is less stressful for the zygote. [ABSTRACT FROM AUTHOR]

Abstract Papaya (Carica papaya L.) is a fruit tree of great economic and food interest worldwide. Its propagation can occur both sexually and asexually. Traditionally, it is propagated by seeds; however, vegetative propagation has been often used with the use of in vitro and ex vitro culture techniques. In vegetative propagation, the cutting method has been extensively explored. Research has also been developed using grafting. In recent years, papaya cultivation has also become a model for other fruit plants in the use of biotechnological techniques aiming applications ranging from germplasm conservation to genetic transformation of plants. Since the beginning of the 1970s, the culture of papaya tree tissues has been intensely studied, allowing greater control of in vitro development, especially for micropropagation, organogenesis and somatic embryogenesis techniques.

Therapeutic success in treating patients with systemic lupus erythematosus (SLE) is limited by the multivariate disease etiology, multi-organ presentation, systemic involvement, and complex immunopathogenesis. Agents targeting B-cell differentiation and survival are not efficacious for all patients, indicating a need to target other inflammatory mediators. One such target is the type I interferon pathway. Type I interferons upregulate interferon gene signatures and mediate critical antiviral responses. Dysregulated type I interferon signaling is detectable in many patients with SLE and other autoimmune diseases, and the extent of this dysregulation is associated with disease severity, making type I interferons therapeutically tangible targets. The recent approval of the type I interferon-blocking antibody, anifrolumab, by the US Food and Drug Administration for the treatment of patients with SLE demonstrates the value of targeting this pathway. Nevertheless, the interferon pathway has pleiotropic biology, with multiple cellular targets and signaling components that are incompletely understood. Deconvoluting the complexity of the type I interferon pathway and its intersection with lupus disease pathology will be valuable for further development of targeted SLE therapeutics. This review summarizes the immune mediators of the interferon pathway, its association with disease pathogenesis, and therapeutic modalities targeting the dysregulated interferon pathway.

Context: (-)-α-Bisabolol (BISA) is a sesquiterpene alcohol widely used as scent in cosmetic preparations, perfumes, shampoos, toilet soaps and other toiletries with potential for use in the pharmaceutical area. Objective: To evaluate the corneal antinociceptive efficacy of BISA and to analyze the best solubilizing agent. Materials and methods: Acute corneal nociception was induced by the local application of hypertonic saline (5 M NaCl; 20 μL) to the corneal surface of Swiss mice (n = 8/group) 60 min after topical treatment with solutions or ointment containing BISA (50–200 mg/mL). The number of eye wipes performed with the ipsilateral forepaw was counted for a period of 30 s. Control groups (vehicles) were included. Background: Plaque-associated gingivitis is a prevalent disease and research in its treatment using herbal agents must be encouraged to verify which would be a useful addition to the current range or chemotherapeutic treatment options. Aims: The aim of this study was to evaluate the clinical effect of a mouth rinse containing 10% Anacardium occidentale (AO) Linn., a typical plant commonly found in the Northeast Region of Brazil, on the reduction of plaque and gingivitis in comparison to a gold-standard chemotherapeutic agent. Materials and Methods: Thirty normosystemic adult volunteers of both genders, who had a minimum of twenty natural teeth, aging between 18 and 32 years, were enrolled in this crossover, controlled, examiner-blind clinical study. They were randomly allocated into three groups: 10% AO Linn. (n = 10); 0.12% chlorhexidine digluconate (CLX, n = 10); or placebo (PB, n = 10). All volunteers were instructed to brush their teeth with a fluoridated dentifrice two times a day (12/12 h) and to rinse for 1 min with one of the mouthwashes (AO, CLX, or PB) 30 min after tooth brushing for 1 month. Plaque index (PLI) and gingival bleeding index (BLI) were recorded on days 0 and 30. Nonparametric Kruskal-Wallis and Wilcoxon tests (α = 0.05) were performed to evaluate statistical differences among groups. Results: There was a significant reduction (P < 0.05) on plaque and gingivitis at day 30 just in CLX ([PLI = 0.47 ± 0.16; -30%]; [BLI = 0.15 ± 0.09; -55.8%]) and AO ([PLI = 0.49 ± 0.21; -31%]; [BLI = 0.13 ± 0.10; -56.6%]) groups, but no statistically significant difference was observed among them (P > 0.05). Conclusion: Mouthwash containing 10% AO was effective as an antiplaque and antigingivitis agent, in a similar manner that 0.12% CLX.

Parkinson's disease (PD) shows high heterogeneity with regard to the underlying molecular pathogenesis involving multiple pathways and mechanisms. Diagnosis is still challenging and rests entirely on clinical features. Thus, there is an urgent need for robust diagnostic biofluid markers. Untargeted metabolomics allows establishing low-molecular compound biomarkers in a wide range of complex diseases by the measurement of various molecular classes in biofluids such as blood plasma, serum, and cerebrospinal fluid (CSF). Here, we applied untargeted high-resolution mass spectrometry to determine plasma and CSF metabolite profiles. We semiquantitatively determined small-molecule levels (≤1.5 kDa) in the plasma and CSF from early PD patients (disease duration 0–4 years; n = 80 and 40, respectively), and sex- and age-matched controls (n = 76 and 38, respectively). We performed statistical analyses utilizing partial least square and random forest analysis with a 70/30 training and testing split approach, leading to the identification of 20 promising plasma and 14 CSF metabolites. These metabolites differentiated the test set with an AUC of 0.8 (plasma) and 0.9 (CSF). Characteristics of the metabolites indicate perturbations in the glycerophospholipid, sphingolipid, and amino acid metabolism in PD, which underscores the high power of metabolomic approaches. Further studies will enable to develop a potential metabolite-based biomarker panel specific for PD.

Cerebrospinal fluid (CSF) has often been used as the source of choice for biomarker discovery with the goal to support the diagnosis of neurodegenerative diseases. For this study, we selected 15 CSF protein markers which were identified in previously published clinical investigations and proposed as potential biomarkers for PD diagnosis. We aimed at investigating and confirming their suitability for early stage diagnosis of the disease. The current study was performed in a two-fold confirmatory approach. Firstly, the CSF protein markers were analysed in confirmatory cohort I comprising 80 controls and 80 early clinical PD patients. Through univariate analysis we found significant changes of six potential biomarkers (α-syn, DJ-1, Aβ42, S100β, p-Tau and t-Tau). In order to increase robustness of the observations for potential patient differentiation, we developed-based on a machine learning approach-an algorithm which enabled identifying a panel of markers which would improve clinical diagnosis. Based on that model, a panel comprised of α-syn, S100β and UCHL1 were suggested as promising candidates. Secondly, we aimed at replicating our observations in an independent cohort (confirmatory cohort II) comprising 30 controls and 30 PD patients. The univariate analysis demonstrated Aβ42 as the only reproducible potential biomarker. Taking into account both technical and clinical aspects, these observations suggest that the large majority of the investigated CSF proteins currently proposed as potential biomarkers lack robustness and reproducibility in supporting diagnosis in the early clinical stages of PD.

Abstract: This study proposes improving the understanding of the main aspects involved in the design of warehouses by the construction of a framework that reveals the state-of-art. The initial research bibliography generated a framework, which was structured in three dimensions: inputs; design and implementation; and outputs. The validation of framework was accomplished through a systematic review of the literature, covering 68 articles published in the period 1999- 2015. This study covered the main aspects highlighted in the academic literature that influence the design of warehouses. Additionally, an overview of the publications based on a theoretical/empirical and a quantitative/qualitative approach was pointed out. This paper aims to contribute to both industry and academic. On the one hand, the framework aggregates value for professionals by permitting the rapid identification of variables, which must be considered in warehouse design. On the other hand, by systematizing the warehouse design area, researchers are able to identify gaps that may generate future studies.

O objetivo do tratamento de esgoto anaeróbio é maximizar a fração de material orgânico no afluente, que é transformada em metano, minimizando, assim, as frações de DQO que são descarregadas com o efluente ou como lodo de excesso. Os dados experimentais deste artigo mostram que, no caso de aplicação de um reator UASB para o tratamento de esgoto, o projeto do separador de fases tem influência importante no desempenho do reator. Um separador de fases eficiente leva à retenção de uma maior massa de lodo, o que resulta em aumento do tempo médio de retenção de sólidos ou da idade do lodo. Os dados mostram que a idade do lodo é o parâmetro operacional fundamental que determina a eficiência do tratamento anaeróbio. Uma maneira simples de melhorar o desempenho do separador de fases é a aplicação de placas paralelas na zona de sedimentação do reator UASB, acima do separador de fases convencional de prismas triangulares com uma base aberta.

Hepatocellular carcinoma (HCC) is the second most common cause of cancer death worldwide, but is still lacking sensitive and specific biomarkers for early diagnosis and prognosis. In this study, we applied targeted massively parallel semiconductor sequencing to assess methylation on a panel of genes (FBLN1, HINT2, LAMC1, LTBP1, LTBP2, PSMA2, PSMA7, PXDN, TGFB1, UBE2L3, VIM and YWHAZ) in plasma circulating cell-free DNA (cfDNA) and to evaluate the potential of these genes as HCC biomarkers in two different series, one from France (42 HCC cases and 42 controls) and one from Thailand (42 HCC cases, 26 chronic liver disease cases and 42 controls). We also analyzed a set of HCC and adjacent tissues and liver cell lines to further compare with 'The Cancer Genome Atlas' (TCGA) data. The methylation in cfDNA was detected for FBLN1, PSMA7, PXDN and VIM, with differences in methylation patterns between cases and controls for FBLN1 and VIM. The average methylation level across analyzed CpG-sites was associated with higher odds of HCC for VIM (1.48 [1.02, 2.16] for French cases and 2.18 [1.28, 3.72] for Thai cases), and lower odds of HCC for FBLN1 (0.89 [0.76, 1.03] for French cases and 0.75 [0.63, 0.88] for Thai cases). In conclusion, our study provides evidence that changes in VIM and FBLN1 methylation levels in cfDNA are associated with HCC and could represent useful plasma-based biomarkers. Also, the potential to investigate methylation patterns in cfDNA could bring new strategies for HCC detection and monitoring high-risk groups and response to treatment.
