1. Laser-optical and numerical Research of the flow inside the lubricating gap of a journal bearing model

Abstract

The laser-optical research of the flow inside the lubricating gap of a journal bearing model is one important task in a larger overall project. The long-term objective is the development of an easy-to-work calculation tool which delivers information about the causes and consequences of cavitation processes in hydrodynamically lubricated journal bearings. Hence, it will be possible to find statements for advantageous and disadvantageous geometrical shapes of the bushings. In conclusion such a calculation tool can provide important insights for the construction and design of future journal bearings. Current design programs are based on a two-dimensional approach for the lubricating gap. The first dimension is the breath of the bearing and the second dimension is the circumferential direction of the bearing. The third dimension, the expansion of the gap in radial direction, will be neglected. Instead of an exact resolution of the flow pattern inside the gap, turbulence models are in use. Past studies on numerical and experimental field have shown that inside the lubricating gap clearly organized and predominantly laminar flow structures can be found. Thus, for a detailed analysis of the reasons and effects of cavitation bubbles, a three-dimensional resolution of the lubricating gap is inevitable. In addition to the qualitative evaluation of the flow with visualization experiments it is possible to perform angle-based velocity measurements inside the gap with the help of a triggered Laser-Doppler- Velocimeter (LDV). The results of these measurements are used to validate three-dimensional CFD flow simulations, and to optimize the numerical mesh structure and the boundary conditions. This paper will present the experimental setup of the bearing model, some exemplary results of the visualization experiments and LDV measurements as well as a comparison between experimental and numerical results.

Abstract

The tumour stroma, and in particular the extracellular matrix (ECM), is a salient feature of solid tumours that plays a crucial role in shaping their progression. Many desmoplastic tumours including breast cancer involve the significant accumulation of type I collagen. However, recently it has become clear that the precise distribution and organisation of matrix molecules such as collagen I is equally as important in the tumour as their abundance. Cancer-associated fibroblasts (CAFs) coexist within breast cancer tissues and play both pro- and anti-tumourigenic roles through remodelling the ECM. Here, using temporal proteomic profiling of decellularized tumours, we interrogate the evolving matrisome during breast cancer progression. We identify 4 key matrisomal clusters, and pinpoint collagen type XII as a critical component that regulates collagen type I organisation. Through combining our proteomics with single-cell transcriptomics, and genetic manipulation models, we show how CAF-secreted collagen XII alters collagen I organisation to create a pro-invasive microenvironment supporting metastatic dissemination. Finally, we show in patient cohorts that collagen XII may represent an indicator of breast cancer patients at high risk of metastatic relapse.

Abstract

Heterogeneous subtypes of cancer-associated fibroblasts (CAFs) coexist within pancreatic cancer tissues and can both promote and restrain disease progression. Here, we interrogate how cancer cells harboring distinct alterations in p53 manipulate CAFs. We reveal the existence of a p53-driven hierarchy, where cancer cells with a gain-of-function (GOF) mutant p53 educate a dominant population of CAFs that establish a pro-metastatic environment for GOF and null p53 cancer cells alike. We also demonstrate that CAFs educated by null p53 cancer cells may be reprogrammed by either GOF mutant p53 cells or their CAFs. We identify perlecan as a key component of this pro-metastatic environment. Using intravital imaging, we observe that these dominant CAFs delay cancer cell response to chemotherapy. Lastly, we reveal that depleting perlecan in the stroma combined with chemotherapy prolongs mouse survival, supporting it as a potential target for anti-stromal therapies in pancreatic cancer., Subtypes of cancer associated fibroblasts can both promote and suppress tumorigenesis. Here, the authors investigate how p53 status in pancreatic cancer cells affects their interaction with cancer associated fibroblasts, and report perlecan as a mediator of the pro-metastatic environment.

Abstract

Stipa splendens (syn. Achnatherum splendens) has been transferred to Neotrinia (Tzvelev) M. Nobis, P. Gudkovaet A. Nowak gen. nov.as N. splendens (Trin.) M. Nobis, P. Gudkova et A. Nowak, based on a comparison of the macromorphology and lemma micromorphology of the Asian representatives of the tribe Stipeae. Lemma epidermal patterns in the examined species are presented and discussed. Additionally, a new section Pennatherum M. Nobis comprising Achnatherum pelliotii, a species recently transferred to Achnatherum from Ptilagrostis is also proposed. This taxon clearly differs from Ptilagrostis by the lemma micromorphology. The species with typical maize-like lemma micromorphological pattern is apparent member of Achnatherum, however, because of having plumose awns and short, blind calluses, it was earlier included into Ptilagrostis. Achnatherum pelliotii is the only Old Word Achnatherum species, with long pilose awns, thus, we propose to place it in the new section., В статье приводится новый род Neotrinia (Tzvelev) M. Nobis, P. Abstract

Here we report the DNA methylation profile of 84 sporadic pancreatic neuroendocrine tumors (PanNETs) with associated clinical and genomic information. We identified three subgroups of PanNETs, termed T1, T2 and T3, with distinct patterns of methylation. The T1 subgroup was enriched for functional tumors and ATRX, DAXX and MEN1 wild-type genotypes. The T2 subgroup contained tumors with mutations in ATRX, DAXX and MEN1 and recurrent patterns of chromosomal losses in half of the genome with no association between regions with recurrent loss and methylation levels. T2 tumors were larger and had lower methylation in the MGMT gene body, which showed positive correlation with gene expression. The T3 subgroup harboured mutations in MEN1 with recurrent loss of chromosome 11, was enriched for grade G1 tumors and showed histological parameters associated with better prognosis. Our results suggest a role for methylation in both driving tumorigenesis and potentially stratifying prognosis in PanNETs.

Abstract

Pancreatic ductal adenocarcinoma (PDAC) is a highly metastatic, chemoresistant malignancy and is characterized by a dense, desmoplastic stroma that modulates PDAC progression. Here, we visualized transient manipulation of focal adhesion kinase (FAK), which integrates bidirectional cell-environment signaling, using intravital fluorescence lifetime imaging microscopy of the FAK-based Förster resonance energy transfer biosensor in mouse and patient-derived PDAC models. Parallel real-time quantification of the FUCCI cell cycle reporter guided us to improve PDAC response to standard-of-care chemotherapy at primary and secondary sites. Critically, micropatterned pillar plates and stiffness-tunable matrices were used to pinpoint the contribution of environmental cues to chemosensitization, while fluid flow–induced shear stress assessment, patient-derived matrices, and personalized in vivo models allowed us to deconstruct how FAK inhibition can reduce PDAC spread. Last, stratification of PDAC patient samples via Merlin status revealed a patient subset with poor prognosis that are likely to respond to FAK priming before chemotherapy.

Abstract

Aims: Understanding fine-grain diversity patterns across large spatial extents is fundamental for macroecological research and biodiversity conservation. Using the GrassPlot database, we provide benchmarks of fine-grain richness values of Palaearctic open habitats for vascular plants, bryophytes, lichens and complete vegetation (i.e., the sum of the former three groups). Location: Palaearctic biogeographic realm. Methods: We used 126,524 plots of eight standard grain sizes from the GrassPlot database: 0.0001, 0.001, 0.01, 0.1, 1, 10, 100 and 1,000 m2 and calculated the mean richness and standard deviations, as well as maximum, minimum, median, and first and third quartiles for each combination of grain size, taxonomic group, biome, region, vegetation type and phytosociological class. Results: Patterns of plant diversity in vegetation types and biomes differ across grain sizes and taxonomic groups. Overall, secondary (mostly semi-natural) grasslands and natural grasslands are the richest vegetation type. The open-access file ”GrassPlot Diversity Benchmarks” and the web tool “GrassPlot Diversity Explorer” are now available online (https://edgg.org/databases/GrasslandDiversityExplorer) and provide more insights into species richness patterns in the Palaearctic open habitats. Conclusions: The GrassPlot Diversity Benchmarks provide high-quality data on species richness in open habitat types across the Palaearctic. These benchmark data can be used in vegetation ecology, macroecology, biodiversity conservation and data quality checking. While the amount of data in the underlying GrassPlot database and their spatial coverage are smaller than in other extensive vegetation-plot databases, species recordings in GrassPlot are on average more complete, making it a valuable complementary data source in macroecology.

Abstract

Aims: Vegetation‐plot records provide information on the presence and cover or abundance of plants co‐occurring in the same community. Vegetation‐plot data are spread across research groups, environmental agencies and biodiversity research centers and, thus, are rarely accessible at continental or global scales. Here we present the sPlot database, which collates vegetation plots worldwide to allow for the exploration of global patterns in taxonomic, functional and phylogenetic diversity at the plant community level. Results: sPlot version 2.1 contains records from 1,121,244 vegetation plots, which comprise 23,586,216 records of plant species and their relative cover or abundance in plots collected worldwide between 1885 and 2015. We complemented the information for each plot by retrieving climate and soil conditions and the biogeographic context (e.g., biomes) from external sources, and by calculating community‐weighted means and variances of traits using gap‐filled data from the global plant trait database TRY. Moreover, we created a phylogenetic tree for 50,167 out of the 54,519 species identified in the plots. We present the first maps of global patterns of community richness and community‐weighted means of key traits. Conclusions: The availability of vegetation plot data in sPlot offers new avenues for vegetation analysis at the global scale.

Abstract

Chronischer beruflicher Stress hat eine hohe Prävalenz und stellt einen Risikofaktor für verschiedenste Erkrankungen dar. Neben den gesundheitlichen Beschwerden für den Arbeitnehmer sind insbesondere damit verbundener Absentismus und Präsentismus eine Belastung für den Arbeitgeber. Vor diesem Hintergrund wird Online-Gesundheitstrainings das Potenzial zugeschrieben, eine zeitgemäße und wirksame Erweiterung im Betrieblichen Gesundheitsmanagement zu sein. Sie eröffnen die Möglichkeit, evidenzbasierte Angebote niedrigschwellig und für eine große Anzahl von Berufstätigen zu Verfügung zu stellen. Arbeitnehmer können diese Interventionen entsprechend ihrer zeitlichen Möglichkeiten und Bedürfnisse flexibel nutzen und sind nicht örtlich gebunden. Herausforderungen betreffen den Datenschutz und die Adhärenz. Internationale Studien zu Online-Gesundheitstrainings belegen, dass Depressivität, Ängste oder Schlafstörungen substanziell und nachhaltig reduziert werden können. Die ersten Studien aus Deutschland bestätigen diese positiven Effekte. Allerdings liegen erst wenige Studien aus dem Bereich der Prävention vor, die sich speziell auf gestresste Arbeitnehmer beziehen. Dass solche Angebote auch für deutsche Arbeitnehmer wirksam sein können, zeigt ein an der Leuphana Universität Lüneburg entwickeltes und in randomisiert-kontrollierten Studien evaluiertes onlinebasiertes und mobilunterstütztes Stressbewältigungstraining. Möglichkeiten, Grenzen und Entwicklungsperspektiven von Onlinetrainings im Rahmen des Betrieblichen Gesundheitsmanagements werden diskutiert.

Abstract

Application of advanced intravital imaging facilitates dynamic monitoring of pathway activity upon therapeutic inhibition. Here, we assess resistance to therapeutic inhibition of the PI3K pathway within the hypoxic microenvironment of pancreatic ductal adenocarcinoma (PDAC) and identify a phenomenon whereby pronounced hypoxia-induced resistance is observed for three clinically relevant inhibitors. To address this clinical problem, we have mapped tumor hypoxia by both immunofluorescence and phosphorescence lifetime imaging of oxygen-sensitive nanoparticles and demonstrate that these hypoxic regions move transiently around the tumor. To overlay this microenvironmental information with drug response, we applied a FRET biosensor for Akt activity, which is a key effector of the PI3K pathway. Performing dual intravital imaging of drug response in different tumor compartments, we demonstrate an improved drug response to a combination therapy using the dual mTORC1/2 inhibitor AZD2014 with the hypoxia-activated pro-drug TH-302. Intravital imaging facilitates the real-time tracking and targeting of moving hypoxic regions within pancreatic ductal adenocarcinoma. Using this approach, Conway et al. alleviate hypoxia-induced resistance to a dual mTORC1/2 inhibitor AZD2014, improving PI3K pathway inhibition and demonstrating a powerful dual imaging modality applicable to targeting other pathways and cancers.

Abstract

The emerging standard of care for patients with inoperable pancreatic cancer is a combination of cytotoxic drugs gemcitabine and Abraxane, but patient response remains moderate. Pancreatic cancer development and metastasis occur in complex settings, with reciprocal feedback from microenvironmental cues influencing both disease progression and drug response. Little is known about how sequential dual targeting of tumor tissue tension and vasculature before chemotherapy can affect tumor response. We used intravital imaging to assess how transient manipulation of the tumor tissue, or "priming," using the pharmaceutical Rho kinase inhibitor Fasudil affects response to chemotherapy. Intravital Förster resonance energy transfer imaging of a cyclin-dependent kinase 1 biosensor to monitor the efficacy of cytotoxic drugs revealed that priming improves pancreatic cancer response to gemcitabine/Abraxane at both primary and secondary sites. Transient priming also sensitized cells to shear stress and impaired colonization efficiency and fibrotic niche remodeling within the liver, three important features of cancer spread. Last, we demonstrate a graded response to priming in stratified patient-derived tumors, indicating that fine-tuned tissue manipulation before chemotherapy may offer opportunities in both primary and metastatic targeting of pancreatic cancer.

Abstract

The small GTPase RhoA is involved in a variety of fundamental processes in normal tissue. Spatiotemporal control of RhoA is thought to govern mechanosensing, growth, and motility of cells, while its deregulation is associated with disease development. Here, we describe the generation of a RhoA-fluorescence resonance energy transfer (FRET) biosensor mouse and its utility for monitoring real-time activity of RhoA in a variety of native tissues in vivo. We assess changes in RhoA activity during mechanosensing of osteocytes within the bone and during neutrophil migration. We also demonstrate spatiotemporal order of RhoA activity within crypt cells of the small intestine and during different stages of mammary gestation. Subsequently, we reveal co-option of RhoA activity in both invasive breast and pancreatic cancers, and we assess drug targeting in these disease settings, illustrating the potential for utilizing this mouse to study RhoA activity in vivo in real time. Nobis et al. generated a RhoA-FRET biosensor mouse to characterize and quantify the spatiotemporal distribution of RhoA activity in native mammalian tissues in vivo during development and disease progression. They show that RhoA activity is tightly regulated during various normal biological processes and is co-opted in disease settings, such as invasive breast and pancreatic cancers.

29. Distribution patterns of segetal weeds of cereal crops in Tajikistan

Abstract

Using the literature data and field research conducted in 2009-2013 the distribution patterns, habitat conditions, phytogeographical characterisation and endangerment of weeds occurring in cereal crops in Tajikistan were analysed. We found out that Tajik weed flora of cereal crops counts 686 taxa. The most species rich families include Asteraceae, Poaceae and Fabaceae. The highest number of cereal weeds were noted in large river valleys of Syr-Daria, Amu-Daria and their tributaries in south-western and northern Tajikistan. This subregions have the warmest climate conditions and extensive arable lands. The greatest weed species richness was observed in submontane and montane elevations between approx. 700 and 1,900 m a.s.l. Cereal weeds occur frequently outside segetal communities in Tajikistan. They were noted usually in screes, wastelands, xerothermophilous grasslands, river gravel beds and in steppes habitats. The assessment of threat status reveals that ca. 33% of total cereal weed flora in Tajikistan are disappearing or occur very rarely. According to the chorological data we find that in the cereals of Tajikistan, 35 endemic and 14 subendemic species occur. The most numerous chorological elements of threatened weed flora of Tajikistan are Irano-Turanian (55%), pluriregional (16%), cosmopolitan (14,5%), Mediterranean (9%) and Eurosiberian (5%) species. Further research is suggested to explore the distribution patterns of all weed species in Tajikistan as it should be useful for economy and effectiveness of crop production as well as conservation of most valuable species.

Abstract

E-cadherin-mediated cell-cell junctions play a prominent role in maintaining the epithelial architecture. The disruption or deregulation of these adhesions in cancer can lead to the collapse of tumor epithelia that precedes invasion and subsequent metastasis. Here we generated an E-cadherin-GFP mouse that enables intravital photobleaching and quantification of E-cadherin mobility in live tissue without affecting normal biology. We demonstrate the broad applications of this mouse by examining E-cadherin regulation in multiple tissues, including mammary, brain, liver, and kidney tissue, while specifically monitoring E-cadherin mobility during disease progression in the pancreas. We assess E-cadherin stability in native pancreatic tissue upon genetic manipulation involving Kras and p53 or in response to anti-invasive drug treatment and gain insights into the dynamic remodeling of E-cadherin during in situ cancer progression. FRAP in the E-cadherin-GFP mouse, therefore, promises to be a valuable tool to fundamentally expand our understanding of E-cadherin-mediated events in native microenvironments. Erami et al. generate an E-cadherin-GFP mouse to demonstrate real-time quantification of E-cadherin mobility using intravital photobleaching in a range of tissue types. They show that changes in E-cadherin mobility correlate with changes in cell junction integrity and invasiveness while demonstrating applications of the mouse for future drug discovery studies.

Hydrodynamic journal bearings are commonly used in many technical applications because they provide low friction and minimal wear. In general, flow simulation during the engineering design process is carried out by means of the Reynolds equation. The Reynolds equation is a non-linear two-dimensional differential equation, which is based on the pressure and the gap between shaft and bushing in relation to bearing clearance, eccentricity, bushing deformation and load. However, due its two-dimensional nature it is inaccurate where the lubricant flow inside the bearing becomes three-dimensional e.g., in the vicinity of feed holes or grooves. The work on hand presents the numerical approach and the cavitation model based on the Rayleigh-Plesset equation. Moreover, a bearing flow experiment was designed and constructed with the goal to validate numerical results. Finally, the validated 3D simulation model is applied on a real bearing, which was subject to an experimental investigation targeting cavitation. The numerical results include images of complex three-dimensional flow structures, vortices and vapor distributions. In comparison of 2D and 3D simulation, the two-dimensional approach gives wrong information in 2 out of 6 critical regions pertaining cavitation failing in both, over- and under-prediction of cavitation. In summary, a new numerical model expands the scope for the numerical simulation of the lubricant flow in hydrodynamic journal bearings and improves the prediction of cavitation.

В статье представлены результаты исследования анатомического строения листовой пластинки 15 видов сибирских ковылей: S. capillata, S. grandis, S. krylovii, S. baicalensis, S. praecapillata, S. consanguinea, S. orientalis, S. lessingiana, S. kirghisorum, S. dasyphylla, S. pennata, S. pulcherrima, S. zalesskii, S. glareosa, S. klemenzii. У исследованных видов были выявлены следующие диагностические признаки: относительная высота и степень выраженности ребер, их форма, количество и тип жилок, степень развития и расположение склеренхимы, количество и тип трихом на адаксиальной поверхности листьев. Несмотря на однородность этих признаков у многих видов в пределах секции, на секционном уровне они проявляют отличительные морфологические закономерности и имеют высокую таксономическую значимость., Results of studies of the anatomical structure of leaf blade of Siberian feather grasses are presented. A cross sections of leave blades were studied in 15 species: S. capillata, S. grandis, S. krylovii, S. baicalensis, S. praecapillata, S. consanguinea, S. orientalis, S. lessingiana, S. kirghisorum, S. dasyphylla, S. pennata, S. pulcherrima, S. zalesskii, S. glareosa, S. klemenzii. The following diagnostic characters were identified: relative height and degree of expressiveness of edges, quantity and type of veins, extent of development and arrangement of sclerenchyma as well as number and type of trichomes on the adaxial surface of leaves. Despite the uniformity of these features in many species within sections, they exhibit distinctive morphological patterns and have been proved to be taxonomically useful.

Hydrodynamic journal bearings are widely used in technical and industrial applications due to their favourable wearing quality and operating characteristics. In the recent years, various experimental and numerical analyses were carried out on the design layout, the load capacity and the durability of the bearing. For typical applications the two-dimensional Reynolds differential equation is solved numerically to calculate the pressure distribution in the oil film, which is essential to simulate the dynamic behavior of the bearing. This approach however, does not allow any detailed predictions of the local three-dimensional flow structures. To understand the mechanisms, which are driven by local flow phenomena, it is necessary to solve the full Navier-Stokes-Equations in 3D together with the conservation of mass. An accurate computation of a three-dimensional flow field requires a careful discretisation of the model. Moreover, only a deliberately chosen meshing based on the optimum number of cells across the gap achieves a sufficient numerical accuracy with acceptable computational effort. This work presents variations of the mesh generation of small gaps in journal bearing models and the computed flow fields, respectively. The threedimensional calculations are validated with measured experimental data done by Laser-Doppler-Velocimetry (LDV). In conclusion of this process the comparison of the velocity profiles of the flow field across the gap yield the necessary numerical discretisation limit applicable to the computation of the flow in journal bearings.

The conference was organised by Brunel University and supported by the Institution of Mechanical Engineers, IPEM, the Italian Union of Thermofluid dynamics, the Process Intensification Network, HEXAG - the Heat Exchange Action Group and the Institute of Mathematics and its Applications. In general, the fluid flow in journal bearings can be described by the Navier-Stokes Equations and the conservation of mass. The application of the small gap criterion allows a simplification of these equations yielding the Reynolds Equation, which links the local gap size with the pressure gradient resulting in a powerful tool for the designing process of journal bearings. Typically, the Reynolds Equation is used in EHD-design software based on FE-methods, which is used to compute pressure distributions, forces, deformations and many more parameters needed for the selection of the right bearing geometry. However, there are regions in the journal bearing where the Reynolds Equation must fail, because either the small gap criterion or the Couette flow assumption is violated. There are pockets, grooves and holes, which are necessary to distribute the oil supply across the gap. Moreover, the oil feed represents a cross flow perpendicular to the circumferential main flow. In these regions three dimensional flow structures replace the undisturbed Couette flow, which are strongly affected by vortices, but are non-turbulent due to the Re-scale. This work presents experimental data obtained from a cylinder apparatus with moderate gap sizes, which features independently rotating cylinders and a cross flow through a hole in the sidewall. LDV-measurements of velocity profiles and visualization methods to animate the three dimensional nature of the flow are presented. The experimental data are used to validate 3D-CFD calculations, which are expanded towards smaller gap sizes in the range of typical journal bearings in automotive applications.
