28. iTRAQ quantitative proteomic analysis differentially expressed proteins and signal pathways in henoch-schönlein purpura nephritis

Abstract

Henoch-Schönlein purpura nephritis (HSPN) has been considered as a major cause of chronic renal failure in children and a condition which can worsen clinical outcomes in adults. At present, the molecular mechanisms of HSPN are still unclear. In this study, iTRAQ quantitative proteomic analysis was performed on renal tissues collected from patients with HSPN and compared with those of patients after nephrectomy (controls). A total of 149 differentially expressed proteins (DEPs) were detected, of which, 97 being upregulated and 52 down-regulated. Protein functions and classifications were analyzed using Gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG). In addition, protein domains. expressive hierarchical clustering analysis and protein-protein interaction (PPI) analysis were also conducted for DEPs. The results of bioinformatics analysis indicated that DEPs were enriched in lipid metabolism and the adherens junction pathway. Among these proteins, CDC42 and CTNNB1 were identified as potential candidates involved in the pathogenesis of HSPN. Immunohistochemistry and real-time PCR further demonstrated that CDC42 and CTNNB1 were up-regulated in HSPN patients. These results provide new and important insights into some underlying molecular pathogenesis of HSPN.

29. Development and application of a rapid detection method for Clarireediaspp. using loop-mediated isothermal amplification (LAMP) assay

Abstract

Dollar spot, caused by the ascomycete fungus, Clarireediaspp., is a disease that seriously damages most grass species, especially those grown on golf courses. Early detection and intervention are essential for the effective management of this disease. In this study, Clarireediaspp. was detected in a faster, more efficient, and specific way than current conventional methods by using a loop-mediated isothermal amplification (LAMP) system that was developed based on the β-tubulingene. When hydroxynaphthol blue (HNB) was added, Clarireediaspp. DNA samples changed color from violet to sky blue after amplification, but those without DNA (negative control) did not. The LAMP assay was specific for Clarireediaspp. and did not amplify other common fungal pathogens or closely related species. The sensitivity of the LAMP method was 100 times greater than conventional PCR. In 219 potentially infected samples, 212 (96.80%) were confirmed positive by LAMP, 214 (97.72%) by tissue isolation, and 135 (61.64%) by PCR. All results validated that the rapid, sensitive, and reliable LAMP system could be conveniently applied for the detection of Clarireediaspp., as well as provide an expedient reference for the control and management of dollar spot.

Abstract

Carpetgrass (Axonopus compressus) is a creeping, stoloniferous, perennial warm-season grass that is adapted to humid tropical and subtropical climates. Recently, outbreaks of anthracnose disease of A. compressus caused by an unidentified Colletotrichum sp. were observed in the Hainan and Guangdong provinces in southern China. In late winter and early spring, the disease incidence reached 100moisture conditions, the crowns and oldest leaf sheaths of the majority of the plants became necrotic, which led to whole lawns turning reddish brown. Pathogenicity was confirmed by inoculating uninfected A. compressus plants with a conidial suspension of the Colletotrichum sp. isolated from diseased Axonopus plants. Phylogenetic analyses of the combined internal transcribed spacer, Sod2, Apn2, and Apn2/Mat1 sequences revealed the pathogen to be a novel species of the Colletotrichum graminicola species complex. Microscopic examination showed that the species was also morphologically distinct from related Colletotrichum species. As a result of the phylogenetic, morphological, and pathogenicity analyses, we propose the name Colletotrichum hainanense for this pathogen of A. compressus in southern China.

Abstract

Tidal waterways perform as the paths through which seawater invades. Their spatial-temporal variation with the shift in Yellow River's route requires the accurate measurement of their status. Visual interpretation for delineating tidal waterways is inefficient due to a substantial amount of work. Buffer analysis provides an alternative to visual interpretation. However, it is difficult to use the buffer distance threshold to identify tidal waterways from other freshwater bodies. In this study, we present a straightforward method for mapping tidal waterways. First, we adopted the normalized difference water body index (NDWI) to extract water bodies. Waterlines were subsequently delineated based on a morphological opening filter with a kernel of 9 × 9. Finally, tidal waterways were mapped based on the proposed topological relation in ArcGIS. We showed the utility of this approach by mapping tidal waterways in the Yellow River Delta, China. Abstract

Having a reliable ocean carbon flux (f(CO2)) retrieval model is essential to monitoring the global carbon cycle and to evaluating the climate change. Remote sensing techniques provide alternatives for f(CO2) retrieval with its advantages of wide area surveys and real-time monitoring. In the present study, a semianalytical f(CO2) estimation model was developed based on remote sensing data and in situ measurements in the Chinese Bohai Sea. The used model performed well (R2 = 0.84) in deriving f(CO2) based on the collected remotely sensed dataset, including sea surface temperature, estimated sea surface salinity, wind speed, Chl-a concentration. The results showed that the distribution of partial pressure of carbon dioxide (p(CO2)) and f(CO2) varied spatially and temporally during the 12 months in 2009. The spatial fluctuations of p(CO2) and f(CO2) in Bohai Sea in summer and autumn were more obvious than that in Spring and Winter. The highest values of p(CO2) and f(CO2) generally appeared in coastal regions. Moreover, the average f(CO2) value of the 12 months showed that the Bohai Sea performed as a weak carbon source in 2009. Abstract

The systemic fungal endophyte of the grass Achnatherum inebrians, Epichloë gansuensis,has important roles in enhancing resistance to biotic and abiotic stresses. In this work, we first evaluated the effects of E. gansuensison nitrogen metabolism, nitrogen use efficiency, and stoichiometry of A. inebriansunder varying nitrogen concentrations. The results demonstrated that E. gansuensissignificantly improved the growth of A. inebriansunder low nitrogen conditions. The fresh and dry weights, nitrogen reductase, nitrite reductase, and glutamine synthetase activity, NO3–, NH4+, N, and P content, and also the total N accumulation, N utilization efficiency, and N uptake efficiency were all higher in leaves of A. inebrianswith E. ganusensis(E+) plants than A. inebriansplants without this endophyte (E−) under low nitrogen availability. In conclusion, E. gansuensishas positive effects on improving the growth of A. inebriansunder low-nitrogen conditions by modulating the enzymes of nitrogen metabolism and enhancing nitrogen use efficiency.
