This study investigated the effect of carvacrol stress on the physiological characteristics and transcriptome of Saccharomyces cerevisiae, a common spoilage yeast in fruit and vegetable products. The results showed that the minimum inhibitory concentration (MIC) of carvacrol on S. cerevisiae was 160 mg/mL. With the increase in temperature (20–40 ℃), the inhibitory effect of carvacrol (160 mg/mL) on yeast growth gradually increased. Environmental pH (3–4.5) had no significant impact on the inhibitory effect of carvacrol. However, the addition of fructose (20–80 g/L) significantly reduced the antifungal activity. Carvacrol treatment damaged the structure of the cell envelope of S. cerevisiae, increased the plasma membrane permeability and caused plasma membrane depolarization, resulting in leakage of intracellular substances, a decrease in mitochondrial membrane potential, an imbalance in intracellular Ca2＋ homeostasis, and a decrease in intracellular ATP content. The transcriptomic analysis revealed that carvacrol stress inhibited the biosynthesis of mitochondrial respiratory chain complexes, ATP synthase and mitochondrial ribosome (MR) proteins, which could block electron transport along the respiratory chain and interfere with life activities related to MR.

Spores are the main dormant forms of some bacteria under harsh environment conditions, which are widely distributed in nature. Bacterial spores have a rigid structure, which helps resist to environmental stresses such as heat and ultraviolet radiation. At present, it is difficult to inactivate bacterial spores by the conventional sterilization methods used in the food industry. Therefore, bacterial spores are one of the most important factors influencing the food safety. In recent years, the application of non-thermal processing technologies in killing spores has received great deal of attention. Cold plasma is a novel non-thermal physical sterilization technology, which has a broad application prospect in the food industry due to its various advantages such as high bactericidal efficiency, low energy consumption, and secondary pollution-free. In this paper, the application of cold plasma in the inactivation of bacterial spores in foods is reviewed, and the underlying mechanisms are also summarized in detail. Moreover, the effects of diverse factors on cold plasma-induced spores inactivation are analyzed, and the future research direction is also prospected. In recent decades, food safety has emerged as a worldwide public health issue with economic and political implications. Pesticide residues, mycotoxins, allergens, and antinutritional factors are the primary concerns associated with food products due to their potential adverse health effects. Although various conventional processing methods (such as washing, peeling, and cooking) have been used to reduce or eliminate these hazards from agricultural food materials, the results obtained are not quite satisfactory. Recently, atmospheric cold plasma (ACP), an emerging low -temperature and green processing technology, has shown great potential for mitigating food hazards. However, detailed descriptions of the effects of ACP treatment on food hazards are still not available. Thus, the current review aims to highlight recent studies on the efficacy and application of ACP in the reduction or elimination of pesticide residues, mycotoxins, allergens, and antinutritional factors in various food products. The possible working mechanisms of ACP and its effect on food quality, and the toxicity of degradation products are emphatically discussed. In addition, multiple factors affecting the efficacy of ACP are summarized in detail. At the same time, the major technical challenges for practical application and future development prospects of this emerging technology are also highlighted. [ABSTRACT FROM AUTHOR]

This study investigated the effects of the combined treatment with cinnamon oil (CIN) and petit-high pressure CO2 (p-HPCO2) against Saccharomyces cerevisiae. The results showed that CIN and p-HPCO2 exhibited a synergistic antifungal effect against S. cerevisiae. After being treated by CIN at a final concentration of 0.02% and p-HPCO2 under 1.3 MPa at 25 °C for 2 h, the S. cerevisiae population decreased by 3.35 log10 CFU/mL, which was significantly (p < 0.05) higher than that of CIN (1.11 log10 CFU/mL) or p-HPCO2 (0.31 log10 CFU/mL). Through scanning electron microscopy, fluorescence staining, and other approaches, a disorder of the structure and function of the cell membrane was observed after the CIN + p-HPCO2 treatment, such as severe morphological changes, increased membrane permeability, decreased cell membrane potential, and loss of membrane integrity. CIN + p-HPCO2 also induced mitochondrial membrane depolarization in S. cerevisiae cells, which could be associated with the decrease in intracellular ATP observed in this study. Moreover, the expression of genes involved in ergosterol synthesis in S. cerevisiae was up-regulated after exposure to CIN + p-HPCO2, which might be an adaptive response to membrane damage. This paper aimed to investigate the bactericidal effect of petit-high pressure carbon dioxide (p-HPCD) against Pseudomonas deceptionensis CM2 isolated from chicken as a function of CO2 pressure or process time. The results showed that the bactericidal effect of p-HPCD on P. deceptionensis CM2 gradually enhanced with increasing CO2 pressure or prolonging process time. After p-HPCD treatment under 1. 3 MPa at 25 T for 2 h, 4 h and 6 h, the populations of P. deceptionensis CM2 were decreased by 1.48 log10 CFU/mL, 3. 11 log10 CFU/mL and 3. 55 log10 CFU/mL, respectively. p-HPCD treatment under 1. 3 MPa at 25 T for 4 h resulted in obvious shrinkage and cracks on the surface of P. deceptionensis CM2 cells. Moreover, p-HPCD treatment caused the leakage of nucleic acids and proteins from P. deceptionensis CM2 cells. Cell membrane permeability and membrane potential also significantly changed after p-HPCD treatment. [ABSTRACT FROM AUTHOR]
