Abstract

Degeneration and adaptation are two competing sides of the same coin called resilience in the progressive processes of brain aging or diseases. Degeneration accumulates during brain aging and other cerebral activities, causing structural atrophy and dysfunction. At the same time, adaptation allows brain network reorganize to compensate for structural loss to maintain cognition function. Although hidden resilience mechanism is critical and fundamental to uncover the brain aging law, due to the lack of datasets and appropriate methodology, it remains essentially unknown how these two processes interact dynamically across brain networks. To quantitatively investigate this complex process, we analyze aging brains based on 6-year follow-up multimodal neuroimaging database from 63 persons. We reveal the critical mechanism of network resilience that various perturbation may cause fast brain structural atrophy, and then brain can reorganize its functional layout to lower its operational efficiency, which helps to slow down the structural atrophy and finally recover its functional efficiency equilibrium. This empirical finding could be explained by our theoretical model, suggesting one universal resilience dynamical function. This resilience is achieved in the brain functional network with evolving percolation and rich-club features. 29. Ultrasound Stimulation Modulates Microglia M1/M2 Polarization and Affects Hippocampal Proteomic Changes in a Mouse Model of Alzheimer's Disease.

Abstract

Background: The effectiveness of ultrasound stimulation in treating Alzheimer's disease (AD) has been reported in previous studies, but the underlying mechanisms remain unclear. This study investigated the effects of ultrasound stimulation on the proportion and function of microglia of different phenotypes, as well as on the levels of inflammatory factors. Additionally, it revealed the alterations in proteomic molecules in the mouse hippocampus following ultrasound stimulation treatment, aiming to uncover potential new molecular mechanisms. Methods: Ultrasound stimulation was used to stimulate the hippocampus for 30 min per day for 5 days in the ultrasound stimulation‐treated group. Amyloid plaque deposition was measured using immunofluorescence staining. M1 and M2 type microglia were labeled using immunofluorescent double staining, and the ratio was calculated. The levels of Aβ42, IL‐10, and TNF‐α were determined using ELISA kits. The quantitative proteomics method was employed to explore molecular changes in hippocampal proteins. Results: Ultrasound stimulation treatment reduced the average fluorescence intensity of amyloid plaques and the concentration of Aβ42. Compared to the AD group, ultrasound stimulation resulted in a 14% reduction in the proportion of M1 microglia and a 12% increase in the proportion of M2 microglia. The concentration of the anti‐inflammatory factor IL‐10 was significantly increased in the ultrasound stimulation‐treated group. Proteomics analysis revealed 753 differentially expressed proteins between the ultrasound stimulation‐treated and AD groups, with most being enriched in the oxidative phosphorylation pathway of mitochondria. Additionally, the activity of cytochrome c oxidase, involved in oxidative phosphorylation, was increased after ultrasound stimulation treatment. Conclusions: Ultrasound stimulation affects microglial polarization, reduces amyloid plaque load, and enhances levels of anti‐inflammatory factors in APP/PS1 mice. Proteomics analysis reveals molecular changes in hippocampal proteins after ultrasound stimulation treatment. The mechanism behind ultrasound stimulation‐induced modulation of microglial polarization may be related to changes in mitochondrial oxidative phosphorylation. [ABSTRACT FROM AUTHOR]

42. Mandarin Electrolaryngeal Speech Recognition Based on WaveNet-CTC

Abstract

Purpose: The application of Chinese Mandarin electrolaryngeal (EL) speech for laryngectomees has been limited by its drawbacks such as single fundamental frequency, mechanical sound, and large radiation noise. To improve the intelligibility of Chinese Mandarin EL speech, a new perspective using the automatic speech recognition (ASR) system was proposed, which can convert EL speech into healthy speech, if combined with text-to-speech. Method: An ASR system was designed to recognize EL speech based on a deep learning model WaveNet and the connectionist temporal classification (WaveNet-CTC). This system mainly consists of 3 parts: the acoustic model, the language model, and the decoding model. The acoustic features are extracted during speech preprocessing, and 3,230 utterances of EL speech mixed with 10,000 utterances of healthy speech are used to train the ASR system. Comparative experiment was designed to evaluate the performance of the proposed method. Results: The results show that the proposed ASR system has higher stability and generalizability compared with the traditional methods, manifesting superiority in terms of Chinese characters, Chinese words, short sentences, and long sentences. Phoneme confusion occurs more easily in the stop and affricate of EL speech than the healthy speech. However, the highest accuracy of the ASR could reach 83.24% when 3,230 utterances of EL speech were used to train the ASR system. Conclusions: This study indicates that EL speech could be recognized effectively by the ASR based on WaveNet-CTC. This proposed method has a higher generalization performance and better stability than the traditional methods. A higher accuracy of the ASR system based on WaveNet-CTC can be obtained, which means that EL speech can be converted into healthy speech.
