Orthodontic extrusion of canines, although challenging, can yield excellent results without tooth extraction. Temporary anchorage devices (TADs), involving mini-implants in the alveolar bone, offer stable anchorage via biomechanics. This case report details orthodontic extrusion of bilaterally impacted maxillary canines using TAD mini-implants and elastics. The patient had bimaxillary protrusion, moderate crowding, deep bite, and vertical growth pattern, necessitating careful treatment planning. The combination of TAD mini-implants with Class-II elastics simplified biomechanics. The treatment achieved efficient results, maintained adjacent teeth positioning, and improved patient compliance.

The present work aims towards the design and development of extended release formulation of freely water-soluble drug diltiazem hydrochloride (DLTZ) based on osmotic technology by using controlled porosity approach. DLTZ is an ideal candidate for a zero-order drug delivery system because it is freely water-soluble and has a short half-life (2-3 h). Sodium chloride (Osmogen) was added to the core tablet to alter the solubility of DLTZ in an aqueous medium. Cellulose acetate (CA) and sorbitol were used as semipermeable membrane and pore former, respectively. The effect of different formulation variables namely concentration of osmogen in the core tablet, % pore former, % weight gain, pH of the dissolution medium and agitation intensity on the in vitro release was studied. DLTZ release was directly proportional to % pore former and inversely proportional to % weight gain. The optimized formulation (F8) delivered DLTZ independent of pH and agitation intensity for 12 h at the upper level concentration of % pore former (25% w/w) and middle level concentration of % weight gain (6% w/w). The comparative study of elementary osmotic pump (EOP) and controlled porosity osmotic pump revealed that it superior than conventional EOP and also easier and cost effective to formulate.

Single individuals of the Greater Spotted Eagle (Clanga clanga), Indian Spotted Eagle (Clanga hastata), Tawny Eagle (Aquila rapax), Western Marsh Harrier (Circus aeruginosus), and two Pallid Harriers (Circus macrourus) were deployed with satellite transmitters in 2021 to study their home ranges, habitat associations, movement, and migration patterns. Data were collected for a combined number of 2291 days, providing 84,544 locations. Home ranges were calculated as kernel utilization distributions and expressed as 95% KDE and core areas as 50% KDE. Overall, eagles had larger home ranges (mean ± SD) of 942.70 ± 937.83 km2 compared to harriers, 43.84 ± 35.55 km2. Among eagles, the Greater Spotted Eagle had the largest home-range size of 2147.03 km2 calculated in Kazakhstan, while the female Pallid Harrier had the smallest home range of 5.74 km2 in Russia. Daily and monthly distances varied among eagles and harriers. The female Pallid Harrier covered the longest average monthly distance of 8585.43 ± 11,943.39 km, while the shortest monthly distance of 1338.22 ± 716.38 km was traveled by the Indian Spotted Eagle. All tagged birds migrated toward higher latitudes in the Northern hemisphere, except the Indian Spotted Eagle, which migrated to Pakistan. The male Western Marsh Harrier covered the longest migration distance in a shorter span of time, while the female Pallid Harrier took the longest to cover its migration distance. Overall, the daily distance covered during migration varied from 115.09 km traveled by the Indian Spotted Eagle to an overwhelming distance of 2035.85 km covered by the male Western Marsh Harrier. Scrubs, water bodies, croplands, and settlements were important habitat features associated with eagles, while croplands, open scrub, and built-up areas were associated with the female Pallid Harrier. The male Western Marsh Harrier was found to be primarily associated with saltpans and salt-affected areas having emergent vegetation. This study presents new insights into the movement and spatial ecology of long-distance migrant raptors that winter in Western India. We provide preliminary support for the use of the Western Circum–Himalayan Corridor as one of the important corridors of the Central Asian Flyway that warrants much appreciation among the current set of flyway corridors.

The purpose of this study is to investigate the molecular interactions and potential therapeutic uses of Eltrombopag (EPAG), a small molecule that activates the cMPL receptor. EPAG has been found to be effective in increasing platelet levels and alleviating thrombocytopenia. We utilized computational techniques to predict and confirm the complex formed by the ligand (EPAG) and the Thrombopoietin receptor (TPO-R) cMPL, elucidating the role of RAS, JAK-2, STAT-3, and other essential elements for downstream signaling. Molecular dynamics (MD) simulations were employed to evaluate the stability of the ligand across specific proteins, showing favorable characteristics. For the first time, we examined the presence of TPO-R in human umbilical cord mesenchymal stem cells (hUCMSC) and human gingival mesenchymal stem cells (hGMSC) proliferation. Furthermore, treatment with EPAG demonstrated angiogenesis and vasculature formation of endothelial lineage derived from both MSCs. It also indicated the activation of critical factors such as RUNX-1, GFI-1b, VEGF-A, MYB, GOF-1, and FLI-1. Additional experiments confirmed that EPAG could be an ideal molecule for protecting against UVB radiation damage, as gene expression (JAK-2, ERK-2, MCL-1, NFkB, and STAT-3) and protein CD90/cMPL analysis showed TPO-R activation in both hUCMSC and hGMSC. Overall, EPAG exhibits significant potential in treating radiation damage and mitigating the side effects of radiotherapy, warranting further clinical exploration.

Summary: The mechanical environment generated through the adhesive interaction of endothelial cells (ECs) with the matrix controls nuclear tension, preventing aberrant gene synthesis and the transition from restrictive to leaky endothelium, a hallmark of acute lung injury (ALI). However, the mechanisms controlling tension transmission to the nucleus and EC-restrictive fate remain elusive. Here, we demonstrate that, in a kinase-independent manner, focal adhesion kinase (FAK) safeguards tension transmission to the nucleus to maintain EC-restrictive fate. In FAK-depleted ECs, robust activation of the RhoA-Rho-kinase pathway increased EC tension and phosphorylation of the nuclear envelope protein, emerin, activating DNMT3a. Activated DNMT3a methylates the KLF2 promoter, impairing the synthesis of KLF2 and its target S1PR1 to induce the leaky EC transcriptome. Repleting FAK (wild type or kinase dead) or inhibiting RhoA-emerin-DNMT3a activities in damaged lung ECs restored KLF2 transcription of the restrictive EC transcriptome. Thus, FAK sensing and control of tension transmission to the nucleus govern restrictive endothelium to maintain lung homeostasis.

Abstract The home range of a species is determined by a complex interplay of extrinsic and intrinsic factors, which can have profound impacts on the species’ resource use. Understanding these dynamics is especially important for conserving critically endangered species. In this study, we used satellite telemetry to investigate the home range of the critically endangered lesser florican (Sypheotides indicus) in Gujarat, India. We analysed GPS locations from 10 lesser floricans deployed with GPS/GSM transmitters between 2020 and 2022. The average home range size (95% KDE) was 10.73 ± 10.70 km2 (mean ± SD), while the average core area (50% KDE) was 1.95 ± 1.56 km2 (mean ± SD). The monthly and daily distances covered were 286.29 ± 599.42 km and 10.11 ± 19.78 km, respectively. Our analysis indicated that suitable habitats and movement patterns were the most important factors explaining the variation in home range size. Specifically, our results suggest that lesser floricans prefer multi-use agro-grassland habitat systems with heterogeneous structures to accommodate different life history requirements. This preference may reflect the depletion and degradation of grasslands across the species’ range. Therefore, managing grassland habitats amidst croplands should be one of the key conservation strategies for the lesser florican.

Using satellite telemetry, it is possible to track long-distance migrant birds with high accuracy and greater spatial coverage. However, prior to 2014, less than 1% of bird species in India had been monitored using this technology. Between January and February 2022, we deployed leg-mounted solar GPS/GSM satellite transmitters to Common Cranes and Demoiselle Cranes (two each) to study home ranges, movement, migration patterns, and habitat use. We used 95% kernels to define the total home range size and 50% kernels to delimit the core areas. The winter habitat use was assessed using Generalized Linear Models (GLM). The average home range of Common Cranes and Demoiselle Cranes was estimated as 161.22 ± 172.08 km2 and 971.40 ± 1023.57 km2, respectively. During migration, Common Crane-1 and Common Crane-2 traveled an average of 471.19 ± 442.42 and 176.97 ± 24.82 km per day, and Demoiselle Crane-1 and Demoiselle Crane-2 covered an average daily distance of 168.10 ± 203.77 and 192.97 ± 250.72 km, respectively. Water bodies and croplands were the most important habitat variables influencing crane presence positively. In recent years, the share of food grain crops within the study area has declined from 43% in 1994–1995 to 36% in 2014–2015, while the share of cotton crops has doubled from 11% to 20%, indicating a probable cause of concern in the near future.

In the present study, the suitability of groundwater for drinking and irrigation purpose were analyzed. A total of 65 groundwater samples were collected and analyzed for relevant water quality parameters from Sirdala block of Nawada district Bihar (India). To estimate the WQI for the drinking purpose, various parameters such as pH, TDS, TH, AS and major ions (Ca2+, Mg2+, Na+, K+, Cl−, F−, HCO3−, NO3−, SO42−) were considered. The WQI for irrigation, various irrigation indices namely soluble sodium percent (Na%), sodium adsorption ratio (SAR), residual sodium bicarbonate (RSCB), permeability index (PI), magnesium hazards ratio (MHR), Kelly's ratio (KR), potential salinity (PS), Cl−: HCO3−, Mg(2+): Ca(2+) and Na+: Ca(2+) were employed and discussed in addition with Wilcox and USSL diagram. Plots, namely Gibbs ratio and Piper diagram, were used to understand chemical characteristics of groundwater. The WQI for drinking purpose was observed between 57.67 and 929.90 in which most samples (70.76%) were unsuitable for drinking. However, the WQI for irrigation purpose varied from 87.88 to 434.67 in which 50% of samples were suitable. Gibb's plots determined in this study reveal that evaporation-precipitation was the dominant factor that influences hydrochemistry. Piper plots suggest that Na-HCO3− water type was major hydrochemical facies in groundwater. HIGHLIGHTS The Piper diagram is applied to explore the dominancy of hydrochemical facies.; Water quality parameters except for EC, TDS, , and are within the acceptable limits for drinking water quality.; WQID shows a predominance of poor water quality in the area.; Indices PI, MHR, KR, Na%, and PS are within the acceptable limit for irrigation purposes.; WQII shows a predominance of no restriction to slight restriction classes.;

Background: Nasal morphometry is a technique which includes the study of a part, size, and shape of the human nose. The nasal index (NI), nostril shape, and philtrum shape are the parameters commonly studied. These may show variations according to regional, ethnic, and climatic differences. Hence, a study was done to calculate the NI among a subgroup and classify the nose type based on it. Also to assess the gender variations and to observe the nostril and philtrum shapes. Methodology: This was a cross-sectional study consisted of 210 undergraduate students at a medical university. The nasal height and nasal breadth were measured in centimeters with a Digital Vernier Caliper. Subsequently, the NI was calculated. Based on these nasal indices, the nose was classified into the following: leptorrhine: 84.9. The nostril shapes (rectangular/triangular/teardrop/round) and philtrum shapes (triangular/parallel/flat/concave) were also observed. Results: The mean NI was 76.06 ± 8.614 with a range from 54.7 to 110. Mesorrhine variety was the most common type. The predominant shape of the nostrils found was rectangular, whereas the most common shape of the philtrum found was triangular. Conclusions: Nasal anthropometric measurements are a simple, reliable, and cost-effective method to study the facial characteristics among a population. These measurements provide guidelines that may play a crucial role in forensics and facial reconstructive procedures.

Triple-negative breast cancer (TNBC) is one of the deadliest subtypes of breast cancer (BC) for its high aggressiveness, heterogeneity, and hypoxic nature. Based on biological and clinical observations the TNBC related mortality is very high worldwide. Emerging studies have clearly demonstrated that hypoxia regulates the critical metabolic, developmental, and survival pathways in TNBC, which include glycolysis and angiogenesis. Alterations to these pathways accelerate the cancer stem cells (CSCs) enrichment and immune escape, which further lead to tumor invasion, migration, and metastasis. Beside this, hypoxia also manipulates the epigenetic plasticity and DNA damage response (DDR) to syndicate TNBC survival and its progression. Hypoxia fundamentally creates the low oxygen condition responsible for the alteration in Hypoxia-Inducible Factor-1alpha (HIF-1α) signaling within the tumor microenvironment, allowing tumors to survive and making them resistant to various therapies. Therefore, there is an urgent need for society to establish target-based therapies that overcome the resistance and limitations of the current treatment plan for TNBC. In this review article, we have thoroughly discussed the plausible significance of HIF-1α as a target in various therapeutic regimens such as chemotherapy, radiotherapy, immunotherapy, anti-angiogenic therapy, adjuvant therapy photodynamic therapy, adoptive cell therapy, combination therapies, antibody drug conjugates and cancer vaccines. Further, we also reviewed here the intrinsic mechanism and existing issues in targeting HIF-1α while improvising the current therapeutic strategies. This review highlights and discusses the future perspectives and the major alternatives to overcome TNBC resistance by targeting hypoxia-induced signaling.

The present study aimed to assess the population density, structure, and population change of nine wild prey species in the semi-arid landscape of Saurashtra, Gujarat, India. A total of eight sites, representing a gradient from highly protected woodlands and grasslands to unreserved grasslands, were selected for sampling. We employed the road transect methodology under a distance sampling framework to achieve our objectives. We evaluated the realized growth rate of the Gir ungulate population through linear regression analysis. Our findings revealed that deer species exhibited higher density and biomass in woodlands compared to grasslands and coastal forests. On the other hand, antelopes showed higher density and biomass in grasslands and coastal forests compared to woodlands. The density gradient of wild prey species was influenced by various factors, including habitat structure, social organization, grouping tendencies, and topography. Over the last four decades, the population of wild prey species in Gir showed minimal changes. Our study provides a comprehensive understanding of wild prey species' density and biomass patterns at the landscape level. The inclusion of findings from ecologically significant and unique areas, such as coastal forests, further enhances the importance of this study. The implications of this study extend beyond the conservation of wild prey species alone; they also contribute to the conservation of the large carnivore guild in the Saurashtra landscape.

The wastewaters from pharmaceutical manufacturing units, hospitals, and domestic sewage contaminated with excretal matters of medicine users are the prime sources of pharmaceutical pollutants (PPs) in natural water bodies. In the present study, PPs have been considered one of the emerging pollutants (EPs) and a cause of concern in river health assessment. Beyond the reported increase in antibiotic-resistant bacteria (ABRB), PPs have been found adversely affecting the biotic diversity in such water environments. Considering Algae, Macroinvertebrates, and Fishes as three distinct trophic level indicators, the present study puts forward a framework for showing River Health Condition (RHC) based on the calculation of a River Health Index (RHI). The RHI is calculated using six Indicator Group Scores (IGS) which individually reflect river health in a defined category of water quality characteristics. While Dissolved Oxygen Related Parameters (DORP), Nutrients (NT), and PPs are taken as causative agents affecting RHCs, scores of Algal-Bacterial (AB) symbiosis, Macroinvertebrates (MI), and Fishes (F) are considered as an effect of such environmental conditions. Current wastewater treatment technologies are also not very effective in the removal of PPs. The objective of the present study is to review the harmful effects of PPs on the aquatic environment, particularly on the chemical and biotic indicators of river health. Based on predicted no-effect concentrations (PNEC) for algae, macroinvertebrates, and fishes in the aquatic environment and measured environmental concentration (MEC) in the river, the estimated risk quotient (RQ) for norfloxacin in the Isakavagu-Nakkavagu stream of river Godavari, Hyderabad is found 293 for algae, 39 for MI, and 335 for fish. Among PPs, in Indian rivers, the presence of caffeine is the most frequent, with algae at the highest level of risk (RQmax= 24.5).Broadly six PPs, including azithromycin, caffeine, diclofenac, naproxen, norfloxacin, and sulfamethoxazole are found above PNEC values in Indian rivers. The application of IGS and RHI in understanding and presenting the river health condition (RHC) through colored hexagons has been demonstrated for the river Ganga near Varanasi (India) as an example. Identification of critical indicator groups, based on IGS provides a scientific basis for planned intervention for river health restoration to achieve an acceptable category.

Abstract: Introduction: The genetic testing to confirm or rule out an acute promyelocytic leukemia (APL) typically takes a minimum of 24‐72 hours. Flow cytometric immunophenotyping (FCI) on the other hand provides rapid and objective information to differentiate APL from non‐APL. Methods: FCI features, with single‐tube 8‐color combination using CD45, CD34, HAL‐DR, CD11b, CD13, CD33, and CD117 and CD64, were compared for the 30 consecutive APL and 30 non‐APL acute myeloid leukemia (AML) cases which morphologically mimicked an APL. The diagnosis was confirmed by cytogenetic or molecular genetic testing in the form of t (15:17) (q22; q21)/variant translocations or PML‐RARA fusion transcript analysis. Results: The APL cells lacked CD34, HLA‐DR, and CD11b in 90%, 90%, and 93.3% cases, respectively. Myeloid antigens such as CD33, CD13, CD117, and CD64 were expressed in 96.7%, 96.7%, 76.7%, and 70% cases, respectively. The dual negative profiles, CD34‐/HLA‐DR‐ or HLA‐DR‐/CD11b‐, were noted in 90% and 93.3% cases. The triple‐negative (CD34‐/HLA‐DR‐/CD11b‐) profile was noted in 90% of the cases. The sensitivity, specificity, and positive predictive value (PPV) of CD34‐/HLA‐DR‐ and HLA‐DR‐/CD11b‐ profiles for the diagnosis of APL were found to be 90%, 80% & 81.1% and 93.3%, 86.7%& 87.5%, respectively. Combining the above two profiles resulted in a triple‐negative profile (CD34‐, HLA‐DR‐ and CD11b‐), which had a better specificity (93.3%) and positive predictive value (93.1%), with similar sensitivity. Conclusion: FCI is a rapid and reliable modality for the diagnosis of an APL. The triple‐negative profile (CD34‐/HLA‐DR‐/CD11b‐) rapidly and specifically identifies an APL case. [ABSTRACT FROM AUTHOR]
